Environmental factors and particle size shape the community structure of airborne total and pathogenic bacteria in a university campus.

Given the dense population on university campuses, indoor and outdoor airborne bacterial contamination may lead to the rapid spread of diseases in a university environment. However, there are few studies of the characteristics of airborne and pathogenic bacterial communities in different sites on a university campus. In this study, we collected particulate matter samples from indoor and outdoor locations at a university in Bengbu City, Anhui Province, China, and analyzed the community characteristics of airborne and pathogenic bacteria using a high-throughput sequencing technique. The results showed that the composition of the dominant airborne and pathogenic bacterial communities was consistent among sites at the phylum and genus levels, with differences in their relative abundance. There were significant differences in the structure of the airborne and pathogenic bacterial communities between indoor and outdoor sites (p < 0.05). An analysis of similarities (ANOSIM) indicated that the structure of airborne bacterial communities in indoor sites was influenced by the room occupancy rate, ventilation conditions, and the extent of indoor furnishing (p < 0.05), while the structure of pathogenic bacterial communities was influenced by the number of individuals and spatial dimensions (p < 0.05). The impact of particle size on the structure of airborne and pathogenic bacterial communities was relatively minor. A total of 194 suspected pathogenic bacterial species were identified, accounting for 0.0001-1.3923% of the total airborne bacteria, all of which were conditional pathogens. Among them, Saccharopolyspora rectivirgula, Acinetobacter johnsonii, and Moraxella osloensis exhibited relatively high relative abundance, accounting for 24.40, 16.22, and 8.66% of the total pathogenic bacteria, respectively. Moreover, 18 emerging or re-emerging pathogenic bacterial species with significant implications for human health were identified, although their relative abundance was relatively low (0.5098%). The relative abundance of pathogenic bacteria in indoor environments was significantly higher than outdoors, with the laboratory and dormitory having the highest levels. The findings of this study provide valuable guidance for the prevention and control of airborne bacterial contamination and the associated health risks in both a campus environment and other public spaces with high occupancy rates.

Overlooked Significant Impact of Trace Metals on the Bacterial Community of PM2.5 in High‐Time Resolution

AbstractBioaerosol has been increasingly recognized as a hazardous factor, possibly leading to environmental issues and health risks. However, the driving factors were rarely investigated, which impeded comprehensive assessment of environmental quality. Herein ambient trace metals and bacterial community in PM2.5 and their interactions during severe pollution were characterized in high time resolution. The results showed that the mass concentrations of total trace elements in PM2.5 were 1.4–9.4 μg m−3, with As and Cr exceeding the national standard, which might pose health risks to humans, especially children with frequently hand‐to‐mouth activity. The airborne bacterial community structure responded to the changing environmental and meteorological variables including PM2.5, related water‐soluble ions, SO2, O3, temperature, relative humidity, and wind speed. Our results suggested that the trace metal was an overlooked factor in shaping the bacterial community compared to PM2.5, related water‐soluble inorganic ions, trace gases, and meteorological variables based on the aggregated boosted tree. Cd, Ga, and Nb were main trace metals factors affecting the bacterial community. The better knowledge of airborne bacterial diversity in high‐time resolution were essential to assess the environmental quality and help to explore the interactions of bioaerosols and atmospheric pollutants in PM2.5. The synergistic toxicity effects for the combination of trace metals and bioaerosol should be considered in future studies when evaluating their health effects.

Effects of aerosol pollution on PM2.5-associated bacteria in typical inland and coastal cities of northern China during the winter heating season

Frequent heavy aerosol pollution occurs during the winter heating season in northern China. Here, we characterized the airborne bacterial community structure and concentration, during typical pollution episodes that occurred during the winter heating season of 2017–2018 in Jinan and Weihai. During this heating season, five and four heavy pollution episodes were observed in Jinan and Weihai, respectively. Compared with December and January, pollution episodes in March were significantly affected by sand dust events. Higher Bacillales were identified in the March samples from Jinan, indicating that sand dust influences bacterial communities. During similar pollution episodes, air pollution in the coastal city of Weihai was lower than the inland city of Jinan. The predominant bacteria included Staphylococcus, Cyanobacteria, Lactobacillus, Deinococcus, Enbydrobacter, Ralstonia, Bacillus, Comamonas, and Sphingomonas. These predominant bacteria are mainly from Proteobacteria, Firmicutes, Cyanobacteria, Actinobacteria, and Bacteroidetes phyla. Bacterial concentration showed significant variation with increased airborne pollutants. The highest concentration appeared during moderate pollution (up to 106 cells/m3), whereas bacterial concentration decreased during heavy and severe pollution (105 cells/m3), which may be related to toxic effects of high pollutant concentrations during heavy or severe pollution. Community structure variation indicated that Cyanobacterial genera were dominant in clean or slight pollution. With increased PM2.5, Staphylococcus increased and became the most abundant bacteria in moderate pollution (up to 40%). During heavy or severe pollution, bacteria that are adaptable to harsh or extreme environments predominate, such as Deinococcus and Bacillus. In the assessment of health risks from air pollution, the bioaerosols risks must consider. Additionally, although most microbial genera are similar between the two cities, there are important differences associated with pollution level. During air pollution regulation in different regions with varied geographical and climatic conditions, bioaerosol pollution difference is an unignored factor.

Structural Characteristics and Functional Implications of PM2.5 Bacterial Communities During Fall in Beijing and Shanghai, China.

Air pollution characterized by fine particulate matter (PM2.5) frequently has occurred in China, and has posed threats to human health. The physiochemical characteristics of airborne PM2.5 have been extensively studied, but its bacterial structures and functions have not yet been well studied. Herein, we focused on the structural characteristics and functional implications of airborne bacteria under different pollution levels in Beijing and Shanghai. The α- and β-diversities showed no obvious difference in two cities (p > 0.05). The dominant phyla Proteobacteria, Firmicutes, and Actinobacteria with total abundance of over 92% were found in all PM2.5 samples. The results of weighted unifrac non-metric multidimensional scaling (NMDS) suggested that air pollution was no obviously correlated with bacterial community but dispersed disorderly. Furthermore, canonical correlation analysis (CCA) and permutation test indicated that , , and wind speed were the key factors that associated with airborne bacterial community structure. Chemical components of particulate matter played more important role in structuring bacterial community than meteorological conditions based on the result of partial CCA. In addition, the annotation of metabolic pathway suggested that the predominant genus Pseudomonas was obviously correlated with disease infections. Several dominant species might contribute to organic degradation, nitrogen cycles, and ice-nuclei activities in environments. Overall, this work enhanced our understanding of functions of airborne bacteria and highlighted their potential role in atmospheric chemical progresses.

Spatiotemporal Controls on the Urban Aerobiome

Greater exposure to environmental microorganisms has been hypothesized to reduce the likelihood of developing autoimmune disorders, and vegetation is known to be a source of diverse microbiota to the air. However, the spatiotemporal dynamics of airborne microbial communities in urban environments with varying amounts and types of vegetation are poorly understood. In this study we used high-throughput sequencing of the bacterial 16S rRNA gene to assess whether fine-scale variation in urban vegetation influences the diversity, composition, or structure of airborne bacterial communities over time. We used passive settling dishes to collect airborne bacteria from 36 sites representing three urban land cover types (forest, grassland, paved) over a 3-month period in Eugene-Springfield, Oregon, USA. We used remote sensing data (aerial 4-band orthoimagery and LiDAR) and geographic information systems (GIS) to assess detailed site characteristics (e.g., total vegetation cover and structural diversity) for each site. Our initial analysis indicated that site was the most important factor explaining variation in bacterial community structure (R2 = 0.32, p < 0.001), followed by sampling date (R2 = 0.24, p < 0.001), while land cover type was a significant but weak predictor (R2 = 0.06, p < 0.001) and other vegetation metrics were even less predictive. However, when samples were analyzed separately by date, the explanatory power of land cover type increased substantially; six of nine dates showed significant effects (p < 0.05) with R2 ranging from 0.16–0.31, indicating that land cover type had a marked influence on bacterial community structure that was obscured by the effects of site and sampling date. Despite the importance of site as a predictor of bacterial community structure, Mantel tests for spatial correlation were insignificant for most sampling dates, suggesting that localized site characteristics were driving this relationship. We use our results to propose a space-time conceptual model of the interactions between site-scale environmental features (e.g., vegetation characteristics) and regional-scale temporal processes and events (e.g., agricultural harvesting) to understand and perhaps manage intraurban airborne bacterial communities.

Comparison of chemical composition and airborne bacterial community structure in PM2.5 during haze and non-haze days in the winter in Guilin, China

Haze is a typical type of air pollution and frequently has occurred in the winter in Guilin city recently, but information relating to chemical compositions and airborne bacterial community structure of atmospheric particulate matter (PM2.5) during haze days remains very limited. In this study, the PM2.5 concentration, chemical components including water-soluble ions and metal elements, and bacterial community compositions of PM2.5 on both haze and non-haze days were characterized, and the relationships between them were studied. The results showed that levels of the secondary aerosol particles of SO42−, NO3−, and NH4+ in haze PM2.5 associated with local fuel combustion and vehicle emissions were significantly higher than non-haze samples. Similar trends were observed for the detected trace metal elements. High-throughput sequencing results indicated that higher PM2.5 concentration caused lower bacterial richness and diversity. However, the airborne bacterial community structure remained stable, and no significant difference was found between haze and non-haze PM2.5. Firmicutes, Proteobacteria, and Bacteroidetes were the most abundant phyla accounting for a total of 97.2% on average. In contrast to non-haze PM2.5, the bacterial community structure of haze PM2.5 was mostly positively correlated with SO42−, NO3−, NH4+, K+, and Cl−, suggesting these secondary aerosol particles were probably of great importance on haze pollution and variability of the bacterial compositions of PM2.5.

Influence of seasonality, air mass origin and particulate matter chemical composition on airborne bacterial community structure in the Po Valley, Italy

The integration of chemical and biological data in aerosol studies represents a new challenge in atmospheric science. In this perspective it will be possible to gain a clearer and deeper comprehension of biogeochemical cycles in the atmosphere.In this view, this study aimed to investigate the relationships occurring between bacterial populations and PM chemical composition in one of the most polluted and urbanized areas in Europe: the Po Valley (Italy). Moreover, seasonality, long- and short-range transports were also evaluated to investigate the influence on airborne bacterial communities.PM samples were collected in two cities of the Po Valley (Milan and Venice) characterized by different meteorological conditions and atmospheric pollutant sources. Samples were analysed for water-soluble inorganic ions (WSIIs) and bacterial community structure. Chemical and biological data were jointly processed by using redundancy discriminate analysis (RDA), while the influence of atmospheric circulation was evaluated by using wind ground data and back-trajectories analysis.Results showed strong seasonal shifts of bacterial community structure in both cities, while a different behaviour was observed for air mass circulation at Milan ad Venice sites: long-range transport significantly affected bacterial populations in Milan whereas local ground wind had more influence in the Venice area. Moreover, difference in taxonomic composition can be mostly addressed to the characteristics of sampling sites. This evidence could suggest that, while PM composition is influenced by long-range transport, bacterial populations are affected, besides transport, by other factors (i.e., season and sampling site location). This perspective allow to better understand and explain airborne bacterial community behaviour.

Variations in the structure of airborne bacterial communities in Tsogt-Ovoo of Gobi desert area during dust events

Asian dust events transport the airborne bacteria in Chinese desert regions as well as mineral particles and influence downwind area varying biological ecosystems and climate changes. However, the airborne bacterial dynamics were rarely investigated in the Gobi desert area, where dust events are highly frequent. In this study, air samplings were sequentially performed at a 2-m high above the ground at the sampling site located in desert area (Tsogt-Ovoo of Gobi desert; Mongolia 44.2304°N, 105.1700°E). During the dust event days, the bacterial cells and mineral particles increased to more than tenfold of concentrations. MiSeq sequencing targeting 16S ribosomal DNA revealed that the airborne bacteria in desert area mainly belonged to the classes Acidobacteria, Actinobacteria, Bacteroidetes, Chloroflexi, Bacilli, Alpha-proteobacteria, Beta-proteobacteria, and Gamma-proteobacteria. The bacterial community structures were different between dust events and non-dust events. The air samples collected at the dust events indicated high abundance rates of Alpha-proteobacteria, which were reported to dominate on the leaf surfaces of plants or in the saline lake environments. After the dust events, the members of Firmicutes (Bacilli) and Bacteroidetes, which are known to form endospore and attach with coarse particles, respectively, increased their relative abundances in the air samples. Presumably, the bacterial compositions and diversities in atmosphere significantly vary during dust events, which carry some particles from grassland (phyllo-sphere), dry lake, and sand surfaces, as well as some bacterial populations such as Firmicutes and Bacteroidetes maintain in the atmosphere for longer time.

Variations in the structure of airborne bacterial communities in a downwind area during an Asian dust (Kosa) event

Asian dust (Kosa) events transport airborne microorganisms that significantly impact biological ecosystems, human health, and ice-cloud formation in downwind areas. However, the composition and population dynamics of airborne bacteria have rarely been investigated in downwind areas during Kosa events. In this study, air samplings were sequentially performed at the top of a 10-m high building within the Kosa event arrival area (Kanazawa City, Japan) from May 1 to May 7, 2011, during a Kosa event. The particle concentrations of bacterial cells and mineral particles were ten-fold higher during the Kosa event than on non-Kosa event days. A 16S ribosomal DNA clone library prepared from the air samples primarily contained sequences from three phyla: Cyanobacteria, Firmicutes, and Alphaproteobacteria. The clones from Cyanobacteria were mainly from a marine type of Synechococcus species that was dominant during the first phase of the Kosa event and was continuously detected throughout the Kosa event. The clones from Alphaproteobacteria were mainly detected at the initial and final periods of the Kosa event, and phylogenetic analysis showed that their sequences clustered with those from a marine bacterial clade (the SAR clade) and Sphingomonas spp. During the middle of the Kosa event, the Firmicutes species Bacillus subtilis and Bacillus pumilus were predominant; these species are known to be predominant in the atmosphere above the Chinese desert, which is the source of the dust during Kosa events. The clones obtained after the Kosa event had finished were mainly from Bacillus megaterium, which is thought to originate from local terrestrial areas. Our results suggest that airborne bacterial communities at the ground level in areas affected by Kosa events change their species compositions during a Kosa event toward those containing terrestrial and pelagic bacteria transported from the Sea of Japan and the continental area of China by the Kosa event.

Temporal variability and effect of environmental variables on airborne bacterial communities in an urban area of Northern Italy

Despite airborne microorganisms representing a relevant fraction of atmospheric suspended particles, only a small amount of information is currently available on their abundance and diversity and very few studies have investigated the environmental factors influencing the structure of airborne bacterial communities. In this work, we used quantitative PCR and Illumina technology to provide a thorough description of airborne bacterial communities in the urban area of Milan (Italy). Forty samples were collected in 10-day sampling sessions, with one session per season. The mean bacterial abundance was about 10⁴ ribosomal operons per m³ of air and was lower in winter than in the other seasons. Communities were dominated by Actinobacteridae, Clostridiales, Sphingobacteriales and few proteobacterial orders (Burkholderiales, Rhizobiales, Sphingomonadales and Pseudomonadales). Chloroplasts were abundant in all samples. A higher abundance of Actinobacteridae, which are typical soil-inhabiting bacteria, and a lower abundance of chloroplasts in samples collected on cold days were observed. The variation in community composition observed within seasons was comparable to that observed between seasons, thus suggesting that airborne bacterial communities show large temporal variability, even between consecutive days. The structure of airborne bacterial communities therefore suggests that soil and plants are the sources which contribute most to the airborne communities of Milan atmosphere, but the structure of the bacterial community seems to depend mainly on the source of bacteria that predominates in a given period of time.

Architectural design influences the diversity and structure of the built environment microbiome.

Buildings are complex ecosystems that house trillions of microorganisms interacting with each other, with humans and with their environment. Understanding the ecological and evolutionary processes that determine the diversity and composition of the built environment microbiome—the community of microorganisms that live indoors—is important for understanding the relationship between building design, biodiversity and human health. In this study, we used high-throughput sequencing of the bacterial 16S rRNA gene to quantify relationships between building attributes and airborne bacterial communities at a health-care facility. We quantified airborne bacterial community structure and environmental conditions in patient rooms exposed to mechanical or window ventilation and in outdoor air. The phylogenetic diversity of airborne bacterial communities was lower indoors than outdoors, and mechanically ventilated rooms contained less diverse microbial communities than did window-ventilated rooms. Bacterial communities in indoor environments contained many taxa that are absent or rare outdoors, including taxa closely related to potential human pathogens. Building attributes, specifically the source of ventilation air, airflow rates, relative humidity and temperature, were correlated with the diversity and composition of indoor bacterial communities. The relative abundance of bacteria closely related to human pathogens was higher indoors than outdoors, and higher in rooms with lower airflow rates and lower relative humidity. The observed relationship between building design and airborne bacterial diversity suggests that we can manage indoor environments, altering through building design and operation the community of microbial species that potentially colonize the human microbiome during our time indoors.

Temporal variability of airborne bacterial community structure in an urban area

Temporal airborne bacterial genetic community structure and meteorological factors were analysed above an urban area in the northwest of France from December 2003 to April 2004 with a sampling strategy considering different time intervals (from an hour to a month). Principal component analysis (PCA) of B-ARISA (Bacterial-Automated Ribosomal Intergenic Spacer Analysis) profiles revealed a hierarchy in the temporal variability of bacterial community: daily<weekly<seasonal. Co-inertia analysis between B-ARISA data and meteorological factors demonstrated the correlation between the seasonal variability in the bacterial community and climatic conditions such as temperature and relative humidity, whereas daily and weekly variability seemed likely to be managed by other factors such as anthropogenic activities.

Assessing genetic structure and diversity of airborne bacterial communities by DNA fingerprinting and 16S rDNA clone library

The density, genetic structure and diversity of airborne bacterial communities were assessed in the outdoor atmosphere. Two air samples were collected on the same location (north of France) at two dates (March 2003 (sample1) and May 2003 (sample 2)). Molecular culture-independent methods were used to characterise airborne bacterial communities regardless of the cell culturability. The automated-ribosomal intergenic spacer analysis (A-RISA) was performed to characterise the community structure in each sample. For both sampling dates, complex A-RISA patterns were observed suggesting a highly diverse community structure, comparable to those found in soil, water or sediment environments. Furthermore, differences in the genetic structure of airborne bacterial communities were observed between samples 1 and 2 suggesting an important variability in time. A clone library of 16S rDNA directly amplified from air DNA of sample 1 was constructed and sequenced to analyse the community composition and diversity. The Proteobacteria group had the greatest representation (60%), with bacteria belonging to the different subdivisions α- (19%), β-(21%), γ-(12%) and δ-(8%). Firmicute and Actinobacteria were also well represented with 14% and 12%, respectively. Most of the identified bacteria are known to be commonly associated with soil or plant environments suggesting that the atmosphere is mainly colonised transiently by microorganisms from local sources, depending on air fluxes.
