Stimulation of regenerative processes in the gums after curettage of the gums with diamond burs through the use of local wound-healing preparations

Relevance. Correcting periodontal and aesthetic problems associated with irrational previous fixed prosthetics is a difficulty faced by orthopedic dentists all over the world. One of the solutions at the moment is to use the idea of vertical preparation with further rational temporary and permanent prostheticsObjective. Assessment of symptoms in patients with vertical preparation, with mandatory curettage of the gums with a flame-shaped bur, receiving local therapy, including the use of the domestic drug Superlimf adhesive plates.Materials and methods. During the clinical study, a comparative analysis of the symptoms of gums traumatized during preparation was carried out in groups of patients with different treatment options. A total of 36 people took part in the study. The observation was carried out for 28 days.Results. It was found that groups of patients treated with Superlimf or Solcoseryl showed the best results in terms of increasing gum volume.Conclusion. Our data allows us to conclude that the use of additional medications such as Superlimf or Solcoseryl Dental can speed up the gum healing process in some patients, which is why it is possible to move to the stage of permanent prosthetics earlier.

Effects of transplanted mesenchymal stem cells on repair of the lung tissue of rats with experimental pulmonary fibrosis

Pulmonary fibrosis is one of the commonest forms of interstitial lung diseases with poorly studied methods of its treatment in both human and veterinary medicines. Therefore, this paper focused on seeking alternative methods of its diagnostics and treatment. The article provides the results of the study of bronchoalveolar lavage fluid of rats with experimental lung fibrosis and influence of transplanted allogeneic mesenchymal stem cells of the bone marrow on stimulation of regenerative processes in damaged lung tissues. The studies were conducted on female Wistar rats with pulmonary fibrosis modeled using single transthoracic injection of solution of bleomycin hydrochloride. For the purpose of treatment, we used allogeneic mesenchymal stem cells introduced by various methods and the traditional treatment. We determined that best normalization of the parameters of the studied brochoalveolar lavage occurred in animals that received mesenchymal stem cells. The most active repair processes were in the experimental group that received the mesenchymal stem cells directly to the lung tissue. The animals that received intravenous injection of mesenchymal stemm cells were observed to have lower clinical parameters of the brochoalveolar lavage, but still better than such in the group treated traditionally. The lowest parameters were in animals that received the traditional treatment; they were greater than the phisological parameters, but significantly exceeded them in animals of the control group, indicating presence of inflammatory process in the lung tissue. The conducted cytological assays of the samples of the brochoalveolar lavage revealed that experimental animals with experimental pulmonary fibrosis had development of macrophage and lymphocytic reactions under the influence of transplanted mesenchymal stemm cells. We observed no atypical cells in all the experimental groups. This allows us to draw a conclusion that using stem cells by various methods of transplantation does not stimulate the onset of negative reactons (formation of atypical cells, metastatic processes, etc). Thus, the results of the study of the influence of transplanted mesenchymal stem cells demonstrate that in the conditions of experimental pulmonary fibrosis, the activity of regenerative processes in pathologically altered lung tissue may be an effective method of treatment of animals with this kind of pathology.

Вплив препаратів кріоконсервованої плаценти на перебіг неспецифічного гонартриту в експерименті

Актуальність. Лікування патології суглобів як дегенеративного, так і травматичного генезу — процес тривалий, багатокомпонентний, завжди вимагає спільних зусиль лікаря та пацієнта. Вибір оптимальних засобів терапії складний та неоднозначний. Біологічно активні речовини — одні з потужних засобів боротьби із запальним процесом, що є постійним супутником пошкодження. Завдяки своїм антиоксидантним, імуномодулюючим та регенеративним властивостям фрагменти плаценти визнані перспективним засобом лікування артропатій різної етіології. Мета дослідження — визначення морфологічних характеристик складових структур колінних суглобів щурів у нормі в умовах експериментального артриту та після введення кріоконсервованої плаценти. Матеріали та методи. Експериментальне дослідження проводилось на 115 білих щурах. З інтактною групою (10 тварин) порівнювались 3 контрольні групи по 35 тварин кожна, яким виконувались: 1) моделювання каргінаніндукованого асептичного запалення колінного суглоба; 2) підшкірна імплантація фрагмента плаценти; 3) підшкірна імплантація фрагмента плаценти на фоні асептичного запалення колінного суглоба. Гістологічні препарати готували за загальними методиками із забарвленням гематоксиліном та еозином та за Ван Гізоном. Результати. При створенні запального вогнища зміни в хрящовій тканині визначались із третьої доби у вигляді лакунарності, каріопікнозу як етап некробіозу хондроцитів, що був особливо виражений із 10-ї до 14-ї доби. Морфологічні зміни кісткової тканини щурів, яким вводився ƛ-карагінан, відмічались на 7-му добу дослідження у вигляді резорбції та деструкції кісткових балок. У подальшому (10-та доба) запальний процес призводив до картини фрагментованої архітектоніки кісткової тканини, що спостерігалась до 21-ї доби. Із боку синовіальної оболонки спостерігаються гіперплазія ворсин, лімфоїдна інфільтрація, поява додаткових шарів синовіїцитів як результат проліферативної реакції. Найбільш виражені зміни спостерігались у покривному шарі синовіальної оболонки. При однократному введенні кріоконсервованої плаценти ознаки запалення вірогідно зменшувались: обмежувалась площа деструкції хряща та кістки, у синовіальній оболонці знижувались ознаки мукоїдного набухання. Процеси функціонального кровонаповнення мікроциркуляторного русла переважали над ішемічними. Стимуляція регенеративних процесів забезпечувала структурне відновлення на 21-шу добу спостереження. Висновки. Препарати кріоконсервованої плаценти впливають на перебіг запального процесу в умовах експериментального остеоартриту. Найбільш вірогідний терапевтичний ефект визначається з боку синовіальної оболонки.

Exosomes: Natural Nanoparticles with Therapeutic Potential

Due to their natural origin, small size, and stability in biological fluids, exosomes can reach target cells, accumulate in tissues, cross the blood–brain barrier, and induce therapeutic effects. This review discusses the properties of exosomes, the methods of their isolation, characterization, and storage, and the prospects of their therapeutic applications, first, for stimulation of regenerative processes, and second, for drug delivery.

Features of the course of regeneration processes in myometry under the influence of general magnetotherapy and selective pulsed electrical stimulation with low frequency currents in patients after myomectomy

The aim of the study was to identify the features of the course of repair processes in myometry under the influence of selective pulsed electrostimulation as a monomethod and in combination with system-wide magnetotherapy according to ultrasound data in patients after myomectomy. The study included 105 patients aged 28 to 59 years after myomectomy. All patients were divided into 3 groups of 35 people: the control group did not receive physical therapy. Patients of the main group with 2 days after surgery was carried out, the total magnetic therapy with the duration of 15 minutes, at the rate of 10 daily procedures in combination with the selective pulsed electrical stimulation currents of low frequency with a duration of 25 minutes, at the rate of 10 daily procedures; patients in the comparison group from the 2nd day after surgery was conducted by selective pulsed electrical stimulation currents of low frequency, duration 25 minutes, at the rate of 10 daily procedures. In the study it was found that the most pronounced stimulation of regenerative processes and proper formation of postoperative scar in patients in the early postoperative period after myomectomy was noted under the influence of complex application of General magnetic therapy in combination with selective pulsed electrical stimulation currents of low frequencies through tristimulous and regenerative eff ects are somewhat less significant results were obtained when monophospate selective pulsed electrical stimulation currents of low frequency. The obtained results indicate the prospects of including the developed complex in the programs of early rehabilitation of patients after myomectomy to improve the flow of regeneration processes in the myometrium and prevent the development of suture failure.

Эффективность экстрактов <i>Saussurea controversa</i> и <i>Fillipendula ulmaria</i> на фоне антибиотикотерапии остеомиелита в эксперименте

Курсовое применение экстрактов Saussurea controversa DC (S.c.) и Fillipendula ulmaria (L). Maxim. (F.u.), содержащих широкий набор флавонолгликозидов, при экспериментальном остеомиелите приводит к стимуляции гранулопоэза в 2,8 и 2,2 раза соответственно (p = 0,04) и регенерации костной ткани (плотность распределения остеобластов возрастает в 5 раз, p = 0,02) в зоне поражения у крыс. В результате исследования миелограмм костного мозга отмечены активация митотической активности гранулоцитов, увеличение количества миелобластов в 5,6 и 4,2 раза (p = 0,02) и зрелых форм гранулоцитов (в 2,5 и 1,5 раза, p = 0,04), измененных в результате антибиотикотерапии. После применения экстракта S.c. в костном мозге крыс с остеомиелитом возрастает общее количество ядросодержащих клеток в 2,7 (p = 0,04) раза. Улучшение морфологической картины костной ткани (активация клеток эндоста, периоста, появление остеобластических «почек», формирование грануляционной ткани) и формирование зрелой костной ткани более выражено (на 12 %, p = 0,02) при применении экстракта S.c., по сравнению с экстрактом F.u.

Medical Rehabilitation after Hydronephrosis Surgical Treatment in Children

The article presents modern technologies of medical rehabilitation of children operated for hydronephrosis. The importance of medical rehabilitation of children with hydronephrosis is associated with high incidence, possibility of developing chronic kidney disease, and early disability of these patients. After surgery on the ureter it is important to use medical rehabilitation technologies aimed at preventing urodynamics disorders of upper and lower urinary tract, and at stimulation of regenerative processes. In recent years, particular attention has been paid to high-intensity pulsed electromagnetic field therapy, which has a more significant and long-term stimulating effect compared to various types of electrical stimulation. Clinical studies were conducted in 80 children operated for hydronephrosis, aged 3 to 15. The main group consisted of 40 children who received high-intensity pulsed magnetic therapy, and 40 children were included in the control group (without physiotherapy). Basic treatment included uroseptics and exercise therapy. Effectiveness of the method was assessed using clinical, laboratory and ultrasound data, and Doppler ultrasound of the renal blood flow. The results of the study demonstrated the rationale of including high-intensity pulsed electromagnetic field therapy in medical rehabilitation of children operated for hydronephrosis. The positive effect of high-intensity pulsed electromagnetic therapy on the urodynamics of the urinary tract (reduction in the size of renal pelvis and calyx) and renal blood flow (restoration of blood flow to the renal cortex) were revealed. This method helped to reduce inflammation in the renal parenchyma (reduction of proteinuria, leukocyturia) in children operated on for hydronephrosis. A significantly higher effectiveness of medical rehabilitation with the use of high-intensity pulsed magnetic therapy was proved (87.5%).

Efficacy and safety of micronized purified flavonoid fraction (MPFF) in treatment of venous diseases in geriatric patients

Analysed herein is the experience with micronized purified flavonoid fraction (MPFF) in treatment of geriatric patients with decompensated forms of chronic venous diseases. A total of 218 geriatric patients were examined and treated. Of these, 132 patients had trophic disorders (C4-C6 class), including 40 patients found to have venous trophic ulcers (VTU). Therapeutic policy consisted in wound cleansing, application of topical antiseptics, hydroalginate dressing, stimulation of regenerative processes by therapy with MPFF at a dose of 1,000 mg daily. The microcirculatory blood flow was examined by means of laser Doppler flowmetry. By the end of a 4-week course of comprehensive treatment, 31 of 40 patients were found to have elevated microcirculation index to 16.9±2.1 perfusion units, suggesting improved perfusion of the lower-limb skin. Venous trophic ulcers completely epithelialized in 9 patients and decreased by half of their initial area in 22 patients. Repeat assessment of cardiac activity showed a decreased functional class of chronic heart failure in 9 of 17 patients with healed VTU. Assessing safety of phlebotrophic therapy revealed neither adverse reactions nor decompensation of accompanying diseases during the whole follow-up period.

Antimicrobial and bone-forming activity of a copper coated implant in a rabbit model.

Current strategies in implant technology are directed to generate bioactive implants that are capable to activate the regenerative potential of the surrounding tissue. On the other hand, implant-related infections are a common problem in orthopaedic trauma patients. To meet both challenges, i.e. to generate a bone implant with regenerative and antimicrobial characteristics, we tested the use of copper coated nails for surgical fixation in a rabbit model. Copper acetate was galvanically deposited with a copper load of 1 µg/mm2 onto a porous oxide layer of Ti6Al4V nails, which were used for the fixation of a tibia fracture, inoculated with bacteria. After implantation of the nail the concentration of copper ions did not increase in blood which indicates that copper released from the implant was locally restricted to the fracture site. After four weeks, analyses of the extracted implants revealed a distinct antimicrobial effect of copper, because copper completely prevented both a weak adhesion and firm attachment of biofilm-forming bacteria on the titanium implant. To evaluate fracture healing, radiographic examination demonstrated an increased callus index in animals with copper coated nails. This result indicates a stimulated bone formation by releasing copper ions. We conclude that the use of implants with a defined load of copper ions enables both prevention of bacterial infection and the stimulation of regenerative processes.

Experience in the Use of Hydrogel Wound Dressings to Stimulate Regenerative Processes of Healing of Venous Ulcers in Patients with Chronic Venous Insufficiency

This article presents the experience of complex treatment of trophic ulcers of venous etiology using of the adsorption bacteriostatic wound coverings in 54 patients. The features of the application of these dressings with their modification as well as the stimulation of regenerative processes using the hydrogel dressings with hyaluronic acid are described. These results suggest that the use of adsorption bacteriostatic wound coverings promotes faster regression of clinical symptoms and closure of venous trophic ulcers and reduces the duration of hospital stay compared to standard treatment. In turn the use of hydrogel wound coverings can reduce pain associated with dressing change and improve the patient’s quality of life.

Stimulation of regenerative processes of qualified athletes specializing in the high jump with a running start

Цель. Изучить влияние восстановительных средств на специальную подготовленность квалифицированных спортсменок, которые специализируются в прыжках в высоту с разбега. Результаты. Важною целевой установкой проведения серии экспериментальных исследований было определения влияния восстановительных средств на активизацию пусковых механизмов специальной подготовленности и условия, которые обеспечивают более высокий уровень проявления специальной выносливости на протяжении соревновательной деятельности. Выводы . Использование в исследованиях комплекса восстановительных средств было частью системы средств неспецифических влияний и формирования на этой основе мобилизационных возможностей спортсменок в разных условиях тренировочной и соревновательной деятельности. Важным при этом было использование неспецифических средств естественного характера при минимальной затрате времени.

β-Amyloid 1-42 Oligomers Impair Function of Human Embryonic Stem Cell-Derived Forebrain Cholinergic Neurons

Cognitive impairment in Alzheimer's disease (AD) patients is associated with a decline in the levels of growth factors, impairment of axonal transport and marked degeneration of basal forebrain cholinergic neurons (BFCNs). Neurogenesis persists in the adult human brain, and the stimulation of regenerative processes in the CNS is an attractive prospect for neuroreplacement therapy in neurodegenerative diseases such as AD. Currently, it is still not clear how the pathophysiological environment in the AD brain affects stem cell biology. Previous studies investigating the effects of the β-amyloid (Aβ) peptide on neurogenesis have been inconclusive, since both neurogenic and neurotoxic effects on progenitor cell populations have been reported. In this study, we treated pluripotent human embryonic stem (hES) cells with nerve growth factor (NGF) as well as with fibrillar and oligomeric Aβ1-40 and Aβ1-42 (nM-µM concentrations) and thereafter studied the differentiation in vitro during 28-35 days. The process applied real time quantitative PCR, immunocytochemistry as well as functional studies of intracellular calcium signaling. Treatment with NGF promoted the differentiation into functionally mature BFCNs. In comparison to untreated cells, oligomeric Aβ1–40 increased the number of functional neurons, whereas oligomeric Aβ1–42 suppressed the number of functional neurons. Interestingly, oligomeric Aβ exposure did not influence the number of hES cell-derived neurons compared with untreated cells, while in contrast fibrillar Aβ1–40 and Aβ1–42 induced gliogenesis. These findings indicate that Aβ1–42 oligomers may impair the function of stem cell-derived neurons. We propose that it may be possible for future AD therapies to promote the maturation of functional stem cell-derived neurons by altering the brain microenvironment with trophic support and by targeting different aggregation forms of Aβ.

EFFICIENCY OF BIOLOGICALLY ACTIVE COMPLEXES OF SOME PLANTS AT INFLUENCE PHYSICAL AND CHEMICAL POLLUTANTS AN ENVIRONMENT

Biologically active substances of the polyphenolic nature from leaves Begonia erytrophylla, from elevated weight Laserpitium latifolium and Vaccinium macrocarpon Ait are isolated. The biological estimation of the isolated complexes in experiments on Tetrahymena pyriformis has revealed the expressed protective effect at individual and combined influence of lead, sodium nitrite and ultraviolet radiation. The protective effect was showed in stimulation of regenerative processes in the irradiated cells and prolongation of life cycle of a population.

Electrical stimulation in the treatment of trophic ulcers of the lower extremities

The most common cause of the development of trophic ulcers of the lower extremities is chronic venous insufficiency, which accounts for 85 - 92% of lesions [1, 4]. In the treatment of patients with trophic ulcers, the surgeon focuses on two key points: correction of perverted venous blood flow in the limb and stimulation of regenerative processes in the ulcer area. If in matters of surgical correction of venous circulation, thanks to the achievements of phlebology, there are fewer and fewer controversial and unresolved issues every year, then the problem of local treatment of ulcers is still far from final resolution. In this regard, the search for new ways of treating trophic ulcers is justified.
