Ximenia americana L.: Chemical Characterization and Gastroprotective Effect

Ximenia americana L., popularly known in Brazil as “ameixa do-mato, ameixa-brava, and ameixa-do-sertão,” is widely used in folk medicine to treat several intestinal disorders. The present study assessed the potential mechanisms of action underlying the gastroprotective activity of the hydroethanolic extract of Ximenia americana L. (EHXA) stem bark. The acute toxicity of EHXA was estimated, and later, the gastroprotective effect in mice was assessed through acute models of gastric lesions induced by acidified or absolute ethanol and indomethacin, where the following mechanisms were pharmacologically analyzed: the involvement of prostaglandins (PG), histamine (H2) receptors, ATP-dependent potassium channels, sulfhydryl groups (SH), α2 adrenergic receptors, nitric oxide (NO), myeloperoxidase (MPO), gastric mucus production, and acetylcholine-mediated intestinal motility. Regarding toxicity, EHXA did not cause deaths or signs of toxicity (LD50 greater than or equal to 2000 mg/kg/p.o.). When the gastroprotective effect was assessed, EHXA (50, 100, and 200 mg/kg/p.o.) reduced the rate of lesions induced by acidified ethanol by 65.63; 53.66, and 58.02% in absolute ethanol at 88.91, 78.82, and 74.68%, respectively, when compared to the negative control group. In the indomethacin-induced gastric injury model, the reductions were 84.69, 55.99, 55.99, and 42.50%, respectively. The study revealed that EHXA might stimulate mucus production and reduce intestinal motility through SH groups, NO production, and activation of α2 adrenergic receptors. The results indicated that EHXA had significant gastroprotective activity in the evaluated models. However, further investigation is required to elucidate the cellular and molecular events underlying the action of EHXA components and to correlate them with the modulation of the signaling pathways, as demonstrated by the current pharmacological approach. Therefore, the results demonstrated in the present study, as well as previously reported findings, support the recommendation of using this species in traditional communities in Brazil.

Anti-ulcer Activity of Polyherbal Formulation from Indian Medicinal Plants Cumin Cyminum, Trigonella Foenum Graecum, Piper Nigrum, Curcuma Longa

Aim: The objective of this study was to assess the Anti-ulcer Activity of the Polyherbal Formulation made from the seeds of Cumin Cyminum, Trigonella Foenum Graecum, Piper Nigrum L. and rhizomes of Curcuma Longa in Wistar albino rats. Methods: Male Albino Rat of Wistar strain were used to evaluate the Polyherbal Formulation’s anti-ulcer properties utilising an ethanol-induced gastric ulcer model. The effectiveness of PHF was assessed by determining, gastric pH and gastric mucus, GSH, malondialdehyde (MDA), and nitric oxide (NO) levels in stomach tissues. Results: The administration of the 400 mg/kg dosage of polyherbal formulation significantly exhibits the gastroprotective effects, when compared to the ethanol group. Conclusion: PHF, containing seeds extracts of Cumin Cyminum, Trigonella Foenum Graecum, Piper Nigrum L. and rhizomes of Curcuma Longa was found to possess antiulcer properties in ethanol induced experimental animal models of gastric ulcers, and these findings suggest that the significant gastroprotective activity could be mediated by its antioxidant activity.

Critical Analysis of Formulation andProbable Mode of Action of Avipattikara churna: A Comprehensive Review

Background: The mode of action of a compound Ayurvedic formulation is a critical and essential issue to be concentrated in assuring the therapeutic efficacy and safety based on its composition. Avipattikara Churna is one of the commonly used formulations containing herbs-mineral drugs. Objective: To emphasize the rationality of Avipattikara Churna and its mode of action by classical and contemporary review. Data Source: Classical texts like Vangasena Samhita, Bhaishajya Ratnavali, Rasendra Sara sangraha, Rasendra Chintamani, and published research works. Material and Methods: In the present study a critical analysis based on ingredients and probable mode of action of Avipattikara Churna in different indicated clinical conditions like agnimandya (digestive impairment), vibandha (constipation), amlapitta (hyperacidity), arsha (piles), mutraghata (retention of urine) and prameha (diabetes mellitus) were done based on classical and contemporary research works. Result: Data from the critical review of classical and contemporary research works. Conclusion: Avipattikara Churna is a versatile formulation used in a wide range of gastrointestinal disorders. By this review, it can be emphasized that Avipattikara Churna, which is in clinical practice for Amlapitta (Hyperacidity) and vibandha (constipation) is a tailored formula by its ingredients. In-vivo studies not only justify the effectiveness of the formulation but provide more evidence of the safety and efficacy of the formulation. Clinical research works substantiate its significant gastroprotective activity.

Anti-Ulcerogenic and Cytoprotective Effects of Saharian (Sidr) Honey from Algeria.

This study aimed at investigating the gastro-protective effects of Algerian Sahara (Sidr) honey from Apis mellifera intermissa against HCl/Ethanol-induced gastric ulcers in rats. Total phenolic content, antioxidant activity and phenolic compounds were determined. Then, three groups of rats (control, HCl/ Ethanol-induced ulcer, and orally administered honey) were used for the determination of gastro-protective effect of Sidr honey. Total phenolic content, total flavonoid content, and DPPH activity of the honey sample were determined as 47.35±3.35 mg GAE/ 100 g, 2.13±0.17 mg QE/ 100 g, and 229.24±0.02 mg/mL, respectively. Oral pretreatment of rats with honey (1.2 g/Kg body weight orally at an interval of 2 days) protected gastric mucosa against HCl/Ethanol-induced damage by decreasing ulcer score, the volume and acidity of gastric juice and increasing pH. These results were confirmed by the histological assessment, which demonstrated a significant gastro-protective activity of Saharian (Sidr) honey against HCl/Ethanol-induced stomach ulcer. Plasma tumor necrosis factor-α, IL-6 and PGE2 were also measured. Sahara honey significantly decreased the plasma TNF-α, PGE2, and IL-6 concentrations.

UPLC-qTOF-MS Phytochemical Profile and Antiulcer Potential of Cyperus conglomeratus Rottb. Alcoholic Extract.

Cyperus has been commonly used as a multi-use medicinal plant in folk medicine worldwide. The objectives of our study were to determine the different metabolites in the Cyperus conglomeratus Rottb. methanol extract, and to assess its in vivo gastroprotective effect in ethanol-induced gastric ulcer model in rats. Serum levels of galactin-3 and TNF-α were employed as biochemical markers. To pinpoint for active agents, comprehensive metabolites profiling of extract via UPLC-qTOF-MS/MS was employed. A total of 77 chromatographic peaks were detected, of which 70 were annotated. The detected metabolites were categorized into phenolic acids and their derivatives, flavonoids, stilbenes, aurones, quinones, terpenes, and steroids. Rats were divided into six groups; healthy control, ulcer control, standard drug group, and 25, 50, 100 mg/kg of C. conglomeratus treated rats. Pre-treatment with C. conglomeratus alcohol extract significantly reduced galactin-3, and TNF-α in ethanol-induced ulcer model at 25, 50, and 100 mg/kg. Further histopathological and histochemical studies revealed moderate erosion of superficial epithelium, few infiltrated inflammatory cells, and depletion of gastric tissue glycoprotein in the ulcer group. Treatment with the extract protected the gastric epithelial cells in a dose-dependent manner. It could be concluded that C. conglomeratus extract provides significant gastroprotective activity in ethanol-induced gastric ulcer and ought to be included in nutraceuticals in the future for ulcer treatment.

Evaluation of Antiulcerogenic Activity of Clerodendron Infortunatum Extract on Albino Rat Gastric Ulceration

Clerodendron infortunatum Linn. (Verbenaceae) is an important and widely used medicinal plant, reported to contain active bitter substance like clerodin, has been widely used as tonic and anthelmintic agent in the country sides of North India. Though, variously used in Ayurveda, Unani system of medicine and Homeopathy in case of ailments like diarrhoea, skin disorders, venereal and scrofulous complaints, wounds, post-natal complications, as vermifuge, laxative and cholagogue, for the removal of ascarids in anus, as external applications on tumours, etc., Background: The present study reports the antiulcer properties of 50% ethanolic extract of Clerodendron infortunatum Linn. (Verbenaceae) leaves have assessed in different acute and chronic gastric ulcer models in rats. Methods: EECI (100, 200 and 400 mg/kg body weight) was administered orally, twice daily for 5 days for prevention from ethanol (EtOH), Cold-restraint stress (CRS), Pylorus-ligation (PL) and 10 days for prevention of acetic acid induced ulcers. Results: The EECI showed significant gastric ulcer protective effect in doses of 400 mg/kg, when given twice daily for 5 days against gastric ulcers induced by ethanol (EtOH), cold restraint stress (CRS) and pyloric ligation (PL). EECI showed dose dependent decrease in ulcer index (UI) against ulcers induced by: (i) Ethanol (control UI: 29.52±2.4 mm2/rat, EECI decrease 21.12±2.3 – 5.3±1.0 (ii) Cold restraint stress (CRS) (control UI: 32.13±2.1, EECI decrease 24.02±2.2 – 4.7±1.1 (iii) pylorus ligation (control UI: 29.25±2.4 mm2/rat, EECI decrease 22.32±2.7 – 5.3±1.0, EECI 400 mg/kg significantly healed ulcers induced by 50% acetic acid after 5 (control UI: 32.30 ± 2.9, EECI decrease 26.27±1.9 – 8.56±0.98 and after 10 days treatment (control UI: 29.02 ± 2.6, healing 21.28±2.1 – 5.78±1.2. EECI prevents the oxidative damage of gastric mucosa by blocking lipid peroxidation and by significant decrease in superoxide dismutase, and increase in catalase activity. Conclusions: Our results show that Clerodendron infortunatum Linn. (Verbenaceae) possess significant gastro-protective activity which might be due to gastric defence factors and clerodin might be the main constituents responsible for this activity.

Carrageenans-Sulfated Polysaccharides from Red Seaweeds as Matrices for the Inclusion of Echinochrome.

The possibility of using different types of carrageenans (CRG) as matrixes for incorporating of echinochrome A (Ech) was investigated. Ech interacts with carrageenans and is incorporated into the macromolecular structure of the polysaccharide. The inclusion of Ech in carrageenan matrices decreased its oxidative degradation and improved its solubility. The changing in the charge and morphology of CRGs during binding with Ech was observed. The rate of Ech release from CRG matrices depended on the structure of the used polysaccharide and the presence of specific ions. The gastroprotective effect of CRG/Ech complexes was investigated on the model of stomach ulcers induced by indomethacin in rats. Complexes of CRG/Ech exhibited significant gastroprotective activity that exceeded the activity of the reference drug Phosphalugel. The gastroprotective effect of the complexes can be associated with their protective layer on the surface of the mucous membrane of a stomach.

Protective effect of oleane-12-en-3β-ol-28-oic acid 3β-D-glucopyranoside in ethanol induced gastric ulcer by enhancing the prostaglandin E2 level

Ethnopharmacological relevanceLantana camara is a popular invasive weed utilized in the management of ulcer in different part of world. Study of specific compound present in this plant responsible for their antulcer activity is main topic of concern. Current study designed for evaluation of the antiulcer activity of oleane-12-en-3β-ol-28-oic acid 3β-D-glucopyranoside (OAG) from Lantana camara L. Materials and methodsAntiulcer activity was carried out on NSAID’s (Aspirin) and ethanol induced ulcer model. The efficacy of the OAG on ulcer index, percentage protection and gastric acid secretion were evaluated. ResultsUlcer protection percentage (38.37%) was significant (P < 0.001) higher in the groups treated with the higher OAG dose (50mg/kg), it also recover the mucosa with no redness, no inflammation, mild dilation of blood vessels. OAG significantly (P < 0.01 and P < 0.001) reduce acidity, free acidity and gastric acid volume. It also significantly (P < 0.01) increases the pH of stomach. ConclusionOn the basis of results, it can be concluded that OAG shows significant gastroprotective activity by gastric acid secretion inhibition and afford protection against gastric mucosal damage. Further increase of prostaglandin E2 level establishes the mechanism of antiulcer activity of OAG.

SCREENING OF WILD FRUIT TREES WITH GASTROPROTECTIVE ACTIVITY IN DIFFERENT EXPERIMENTAL MODELS.

Given the increase of people with gastrointestinal disorders, the search for alternative treatments with fewer side effects is vital, as well as the demand for food or plants that can help protect the stomach. The aim of this study was to evaluate the gastroprotective action of the extracts of wild fruit trees of Myrcianthes pungens (guabiju); Inga vera Willd. (ingá-banana) and Marlierea tomentosa Cambess. (guarapuruna) in in vivo pharmacological models. The different parts of the fruits were separately subjected to a process of extraction by methanol. Two experimental pharmacological models were conducted in mice; the gastric ulcer model induced by non-steroidal anti-inflammatory (indomethacin), and the gastric ulcer model induced by ethanol/HCl, which allowed us to evaluate the gastroprotective activity of the extracts at a dose of 250 mg/kg. Subsequently, the total lesion area (mm2) and relative lesion area (%) were determined. The results showed significant gastroprotective activity against the aggressive agents used - ethanol and indomethacin - for all the extracts tested. It is assumed that the fruits have bioactive compounds such as antioxidant substances that act on the prostaglandin levels, protecting them from the damage caused by ethanol and indomethacin. These results prompt further studies to isolate and identify the active properties.

Gastroprotective effect and mechanism of action of Croton rhamnifolioides essential oil in mice.

Croton rhamnifolioides Pax is a plant species that have been used in the folk medicine to treat ulcers, inflammations and hypertension. However, despite the relevant data obtained from ethnopharmacological studies, the pharmacological properties endorsing the efficacy of this plant to treat ulcer remain to be elucidated. The present study aimed to characterize the chemical profile and evaluate the gastroprotective activity of the essential oil obtained from C. rhamnifolioides Pax (OECC) in mice. The essential oil of Croton rhamnifolioides was obtained by hydrodistillation and analyzed by gas-phase chromatography coupled to mass spectrometry (GC/MS). The median lethal dose was determined employing an acute toxicity test. The gastroprotective activity of the OECC was investigated using animal models of gastric ulcer induced by the administration of absolute ethanol, acidified ethanol or indomethacin. Mechanisms of action were investigated using the physical barrier test and by in vivo evaluation of the involvement of the following molecular pathways: nitric oxide, ATP - dependent potassium channels, α2 - noradrenergic receptors, capsaicin - sensitive afferent neurons and opioid receptor. We identified the presence of 21 compounds in OECC, including spathulenol and 1,8 - cineole as major constituents. In orally administered mice, OECC caused no significant toxicity. OECC significantly prevented gastric lesions in all mice models. The barrier test demonstrated that the gastroprotective activity of OECC occurs in a systemic dimension. Our results demonstrated that the gastroprotective effect of OECC involves mechanisms that are related to modulation of opioid receptors and nitric oxide. In conclusion, OECC demonstrated significant gastroprotective activity associated with low toxicity, providing scientific evidences that C. rhamnifolioides have the potential for the development of new antiulcer drugs.

Study of the Gastroprotective Effect of Extracts and Semipurified Fractions of Chresta martii DC. and Identification of Its Principal Compounds.

Chresta martii (Asteraceae) is a species widely used by the population of the Xingu region of Sergipe, Brazil, in the form of a decoction (aerial parts) for the treatment of gastrointestinal diseases. The study aims to assess the gastroprotective activity of organic extracts and semipurified fractions and identify the principal compounds present in C. martii responsible for such activity. The organic extracts (cyclohexane: ECCm, ethyl acetate: EACm, and ethanol: EECm) were obtained from the dried aerial parts (500 g) of C. martii. For evaluation of the gastroprotective activity of extracts (50, 100, or 200 mg/kg; p.o.), male Swiss Webster mice (25–30 g) were used which had gastric ulcers induced by indomethacin (40 mg/kg, s.c.) or ethanol (0.2 mL/animal; p.o.). Among the extracts evaluated, EACm exhibited significant (P < 0.05) gastroprotective activity in the models used. The fractionation of EACm was performed in a silica gel column 60 eluted with the following compounds: [chloroform—F1 yield (10%)], [chloroform/ethyl acetate (1/1)—F2 yield (6%)], [ethyl acetate—F3 yield (8%)], and [ethyl/methanol acetate (1/1)—F4 yield (5%)]. Of the fractions described above, the F1 (25 mg/kg; p.o.) had greater gastroprotective activity (P < 0.05) than that displayed by ranitidine (80 mg/kg; p.o.) in the ethanol-induced ulcer model. The refractionation of F1 produced 23 subfractions and from these two yellow amorphous compounds were obtained by recrystallization, Rf: 0.46 and 0.31 (ethyl acetate : chloroform 5 : 5). The compounds isolated were characterized by nuclear magnetic resonance spectroscopy (1H-NMR and 13C-NMR) and identified as flavones: chrysoeriol (yield: 0.43%) and 3′,4′-dimethoxyluteolin (yield: 0.58%). Conclusion. Flavone 3′,4′-dimethoxyluteolin is the principal compound present in the species C. martii and is probably responsible for gastroprotective activity observed in this species.

Antiulcer and antiadipogenic acitities of carotenoid and anthocyanin extracts from color-fleshed sweet potato

Sweet potato (Ipomoea batatas Lam.) is an important industrial crop and source of food that contains useful dietary fiber and vitamins. Recently, orange- and purple- fleshed varieties have been shed light on owing to their healthful components, carotenoids and anthocyanins, respectively. In this study, we examined the protective effect on the HCl/EtOH-induced gastric ulcer mice model and anti-oxidant activities. Each carotenoid and anthocyanin extract from orange- and purple- fleshed sweet potatoes was evaluated for gastro-protection in mice using a HCl/EtOH-induced ulcer model. Efficacy was assessed by determination of mean ulcer size, and protection rate. Oral administration of the carotenoid and anthocyanin extract (100 mg/kg) protected against gastric lesions by 78.1% and 60.7%, as compared to a positive control, sucralfate (77.5%) in this ulcer model, respectively. This finding suggests that the significant gastroprotective activity could be mediated by its antioxidant activity which was evaluated by using different antioxidant assays including DPPH, ABTS, and FRAP. In addition, anti-adipogenic activity of these extracts was also evaluated. As a result, carotenoid extract suppressed the differentiation of 3T3-L1 preadipocytes to adipocytes. Gastroprotective effect of standardized leaf extract from Careya arborea on experimental gastric ulcers in rats

Context: The leaf of Careya arborea Roxb. (Lecthidaceae) has been advocated in Ayurveda for the treatment of various disorders, including ulcers, healing of wounds and several skin diseases.Objective: The 70% ethanol (EtOH) extract of C. arborea leaves (CALE) was investigated for its gastroprotective effect in different gastric ulcer models.Materials and methods: CALE (100, 200, and 400 mg/kg body weight) was administered orally, twice daily for 5 d, for preventing aspirin (ASP)-, EtOH-, pylorus ligation (PL)-, and cold restraint stress (CRS)-induced ulcer in rats. The status of the antioxidant enzymes in CRS-induced ulcers, H+K+ATPase activity, gastric wall mucous in EtOH-induced ulcer, and gastric secretion parameters were estimated in the PL-induced ulcer model.Results: CALE exhibited significant (p < 0.01) dose-dependent inhibition of ulcer index in ASP 12.90–51.61%, EtOH 11.97–40.35%, PL 28.63–63.92%, and CRS 38.30–66.37%, respectively. A significant (p < 0.001) decrease occurred in the level of H+K+ATPase, volume of gastric juice, and acid output. Simultaneously, the level of gastric wall mucus was increased significantly (p < 0.05). The antioxidant enzyme levels of LPO and SOD were decreased with concomitant increase in catalase activity in CRS-induced ulcers. High-performance thin-layer chromatography (HPTLC) showed the presence of quercetin, ellagic acid, and gallic acid (0.31%, 0.24%, and 0.71% w/w, respectively) in CALE.Conclusions: Our results show that C. arborea possesses significant gastro-protective activity, probably due to its free radical scavenging activity, and validate the folklore claim.

Gastroprotective effects (in rodents) of a flavonoid rich fraction obtained from Syngonanthus macrolepsis

Syngonanthus macrolepis, popularly known in Brazil as 'sempre-vivas', is a plant from the family Eriocaulaceae, it is found in the states of Minas Gerais and Bahia. The species contains a variety of constituents, including flavonoids with gastroprotective effect. In this work, a flavonoid-rich fraction (Sm-FRF) obtained from scapes of S. macrolepis was investigated for preventing gastric ulceration in mice and rats. The activity was evaluated in models of induced gastric ulcer (absolute ethanol, stress, non-steroidal anti-inflammatory drugs and pylorus ligation). The cytoprotective mechanisms of the Sm-FRF in relation to sulfhydryl (SH) groups, nitric oxide (NO) and antioxidant enzymes were also evaluated. The Sm-FRF (100 mg/kg, p.o.) significantly reduced gastric injury in all models, and did not alter gastric juice parameters after pylorus ligation. The results indicate significant gastroprotective activity for the Sm-FRF, which probably involves the participation of both SH groups and the antioxidant system. Both are integral parts of the gastrointestinal mucosa's cytoprotective mechanisms against aggressive factors.

Antiulcer activity of Muntingia calabura leaves involves the modulation of endogenous nitric oxide and nonprotein sulfhydryl compounds

Context: Muntingia calabura L. (Muntingiaceae) is a native plant species of the American continent and is widely cultivated in warm areas in Asia, including Malaysia. The plant is traditionally used to relieve pain from gastric ulcers.Objective: This study was designed to determine the antiulcer activity of a methanol extract of M. calabura leaves (MEMC) and the possible mechanisms of action involved.Materials and methods: An acute toxicity study was conducted using a single oral dose of 2000 mg/kg MEMC. The antiulcer activity of MEMC was evaluated in absolute ethanol- and indomethacin-induced gastric ulcer rat models. MEMC was administered orally (dose range 25–500 mg/kg) to rats fasted for 24 h. The animals were pretreated with NG-nitro-l-arginine methyl esters (l-NAME) or N-ethylmaleimide (NEM) prior to MEMC treatment to assess the possible involvement of endogenous nitric oxide (NO) and nonprotein sulfhydryl (NP-SH) compounds in the gastroprotective effect of MEMC.Results: As the administered dose did not cause toxicity in the rats, the oral median lethal dose (LD50) of MEMC was >2000 mg/kg in rats. MEMC exerted significant (p < 0.001) gastroprotective activity in the ethanol- and indomethacin-induced ulcer models dose-dependently. Histological evaluation supported the observed antiulcer activity of MEMC. l-NAME and NEM pretreatment significantly (p < 0.05) reversed and abolished the gastroprotective effect of MEMC, respectively.Discussion and conclusion: The results obtained indicate that MEMC has significant antiulcer activity that might involve the participation of endogenous NO and NP-SH compounds. These findings provide new pharmacological information regarding the potential use of M. calabura.

Antiulcer activity of a polyherbal formulation (PHF) from Indian medicinal plants

AimThe present study was aimed at evaluating the antiulcer activity of the polyherbal formulation (PHF) containing the leaf extracts of Moringa oleifera, Raphinus sativus, and Amaranthus tricolor in rats. MethodsThe antiulcer activity of the polyherbal formulation (PHF) was evaluated using different models of gastric ulcers: ethanol-induced, indomethacin-induced and ischemia reperfusion-induced gastric ulcers. Efficacy was assessed by determining the ulcer index. ResultsAdministration of the polyherbal formulation (150 mg·kg−1, p.o.) offered significant protection against indomethacin-induced, ethanol-induced, and ischemic reperfusion-induced ulcer models when compared to the control group. ConclusionPHF, containing leaf extracts of Moringa oleifera, Raphinus sativus, and Amaranthus tricolor, was found to possess antiulcer properties in three experimental animal models of gastric ulcers, and these findings suggest that the significant gastroprotective activity could be mediated by its antioxidant activity.

Protective effect of standardized extract of Cleome viscosa against experimentally induced gastric lesions in the rat

Context: Cleome viscosa Linn. (Capparidaceae) is used traditionally in the Indian system of medicine as a carminative, anthelmintic, and diuretic, and used for healing wounds, ulcers and diarrhea.Objective: A 70% ethanol (EtOH) extract of the aerial parts of Cleome viscosa extract (CVE) was investigated for gastroprotective activity in different gastric ulcer models in order to validate ethnobotanical claims regarding the plant use in ulcers.Materials and methods: CVE (100, 200 and 400 mg/kg body weight) was administered orally, twice daily for 5 d, for prevention from EtOH, pylorus ligation (PL) and cold restraint stress (CRS)-induced ulcers in rats. Estimation of H+K+ATPase activity and gastric wall mucous were performed in EtOH-induced ulcer, antioxidant enzyme activities in supernatant mitochondrial fraction of CRS-induced ulcer, and gastric secretion parameters were estimated in PL-induced ulcer modelResults: CVE showed significant (p < 0.01) dose-dependent inhibition of lesion index in EtOH 15.93–42.30%, PL 26.34–59.28% and CRS 22.58–54.03%, respectively. CVE prevents the oxidative damage of gastric mucosa by blocking lipid peroxidation and by a significant (p < 0.001) decrease in superoxide dismutase, and an increase in catalase activity. A significant (p < 0.01) decrease occurred in the level of H+K+ATPase, volume of gastric juice and total acidity. Simultaneously, the level of gastric wall mucus and pH were increased significantly (p < 0.05). High performance thin layer chromatography analysis showed the presence of quercetin and gallic acid (0.3% and 0.25% w/w, respectively) in CVE.Conclusions: Results of our study showed that C. viscosa possesses significant gastroprotective activity, probably due to free radical scavenging activity, and validates the folklore claim.

Gastroprotective Effects of Ascaridole on Gastric Ulcer in Rats

ObjectiveTo evaluate the gastroprotective activity of ascaridole. MethodsThe gastroprotective effect of ascaridole was evaluated on ulcer healing in rats with acetic acid-induced chronic gastric ulcer, pylorus ligation- and Aspirin-induced gastric ulcer. Ascaridole was ig administered with the dosages of 10 and 20 mg/kg once daily for 7 d. ResultsAscaridole showed the significant anti-ulcer effects. In acetic acid-induced gastric ulcer rats, the ulcer areas after 10 and 20 mg/kg of ascaridole treatment were (65.1 ± 20.0) and (50.6 ± 11.0) mm2, respectively, which were significant lower (P < 0.01) than that of the control group [(116.7 ± 35.8) mm2]. For pylorus ligation model, ascaridole showed a gastric ulcer healing effect in a dose-dependent manner. Ascaridole at the dose of 20 mg/kg showed 50% ulcer protection and had a significant (P < 0.05) gastroprotective activity since it decreased the total acidity and pepsin activity. Compared to the control group, the two dosages of ascaridole showed the significant reduction (P < 0.05) in the ulcer index on Aspirin-induced ulcer. ConclusionThis study provides evidence that ascaridole shows potential efficacy on the healing of gastric ulcers induced by acetic acid, Aspirin, and pylorus ligation.

Gastroprotective mechanisms of the chloroform and ethyl acetate phases of Praxelis clematidea (Griseb.) R.M.King &amp; H.Robinson (Asteraceae)

Flavonoid-rich Praxelis clematidea (Griseb.) R.M.King & H.Robinson (Asteraceae) is a native plant of South America. This study evaluates the gastroprotective activity and possible mechanisms for both the chloroform (CHCl3P) and ethyl acetate phases (AcOEtP) obtained from aerial parts of the plant. The activity was investigated using acute models of gastric ulcer. Gastric secretion biochemical parameters were determined after pylorus ligature. The participation of cytoprotective factors such as mucus, nitric oxide (NO), sulfhydryl (SH) groups, prostaglandin E2 (PGE2), reduced glutathione (GSH), superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione reductase (GR), reduction of lipid peroxidation (malondialdehyde level), and polymorphonuclear infiltration (myeloperoxidase activity), was also investigated. CHCl3P (125, 250, and 500 mg/kg) and AcOEtP (62.5, 125, and 250 mg/kg) showed significant gastroprotective activity, reducing the ulcerative index by 75, 83, 88% and 66, 66, 81% for ethanol; 67, 67, 56% and 56, 53, 58% for a non-steroidal anti-inflammatory drug (NSAID); and 74, 58, 59% and 64, 65, 61% for stress-induced gastric ulcer, respectively. CHCl3P (125 mg/kg) and AcOEtP (62.5 mg/kg) significantly reduced the ulcerative area by 78 and 83%, respectively, for the ischemia-reperfusion model. They also did not alter the biochemical parameters of gastric secretion, the GSH level or the activities of SOD, GPx or GR. They increased the quantity of gastric mucus, not dependent on NO, yet dependent on SH groups, and maintained PGE2 levels. The P. clematidea phases demonstrated gastroprotective activity related to cytoprotective factors.

Gastroprotective effect of Achyranthes aspera Linn. leaf on rats

ObjectiveThe study was aimed at evaluating the antiulcer activity of ethanolic extract of Achyranthes aspera (EEAA) leaf. MethodsThe anti-ulcer assays were performed on pylorus ligation and chronic ethanol induced ulcer model. The effects of the EEAA on gastric content volume, pH, free acidity, total acidity and ulcer index were evaluated. ResultsThe percentage of ulcer protection (59.55% and 35.58%) was significantly (P < 0.001) higher in the groups treated with the high dose of EEAA (600 mg/kg), it also reduced the volume of gastric juice and total acidity whereas, gastric pH was increased significantly. ConclusionsThe results of this study show significant gastroprotective activity of EEAA may be due to presence of phyto-constituents like flavanoids, saponins and tannins.
