Unveiling the variability in cadmium accumulation and tolerance characteristics: a comparative study of Basma and Yunyan 87 tobacco varieties

ABSTRACT Tobacco (Nicotiana tabacum L.) shows promise for remediating Cd-contaminated soil due to its significant Cd accumulation capabilities. Although various tobacco varieties exhibit distinct Cd bioaccumulation capacities, a comprehensive understanding of the underlying mechanisms is lacking. This study, conducted using hydroponics, explores differences in Cd accumulation and tolerance mechanisms between two tobacco varieties, Basma and Yunyan 87. The results showed that Cd stress reduced the dry weight, tolerance index, and root morphology for both varieties. Basma exhibited a relatively smaller decline in these indices compared to Yunyan 87. Moreover, Basma demonstrated a higher Cd bioconcentration factor (BCF), concentration, and accumulated content, signifying its superior tolerance and bioaccumulation capacity to Cd compared to Yunyan 87. The Carbonyl Cyanide3-ChloroPhenylhydrazone (CCCP) addition resulted in reduced Cd accumulation and BCFs in both tobacco species. This effect was more pronounced in Basma, suggesting that Basma relies more on an active transport process than Yunyan 87. This could potentially explain its enhanced bioaccumulation ability. Subcellular Cd distribution analysis revealed Basma's preference for distributing Cd in soluble fractions, while Yunyan 87 favoured the cell wall fractions. Transmission electron microscope showed that Basma's organelles were less damaged than Yunyan 87's under Cd stress, possibly contributing to the superior tolerance of Basma. Therefore, these results provided a theoretical foundation for development of Cd-contaminated soil tobacco remediation technology.

Biomonitoring of Potentially Toxic Elements in an Abandoned Mining Region Using Taraxacum officinale: A Case Study on the “Tsar Asen” Mine in Bulgaria

Mining is a significant industrial and economic activity, but it can also result in ore waste that contains several potentially toxic elements (PTEs). Furthermore, the wide range of observed transfer and accumulation factors suggests that not only the soil element concentrations but also the soil characteristics are important for uptake, which is recognized as a serious problem with potential impacts on human health and ecosystems. The aim of this study was to perform a pollution risk assessment of a region near an abandoned mine using suitable biomonitoring followed by multivariate statistical treatment of the obtained results. The content of PTEs in the soil and plant samples was determined to exceed the background concentrations typical for Bulgaria. The observed concentrations of copper in this study’s soil samples exceeded 2 to 16 times the maximum permissible values. The Taraxacum officinale concentrations for PTEs, particularly for Cd (5.13 mg/kg), were higher than the background levels. The obtained results for PTEs show significant TFs for Cd and Sn, while the AFs outline a significant accumulation of Cd and Sb. The applied multivariate statistical approaches revealed differences between the sampling locations and relationships between the elemental transfer/accumulation factors and soil cation exchange capacity. The outcomes from the statistical analysis confirm that the usage of both element factors and soil properties in biomonitoring studies are essential for reliable risk assessment.

Application of two- and multiway chemometric strategies for describing elementomic changes in pepper plants exposed to cadmium stress by multielement determination

The objective of this work was to evaluate elemental changes in pepper exposed to Cd stress through different chemometric tools. For this purpose, pepper plants were grown under five different treatments with different Cd concentrations in the nutrient solution. Considering the hypothesis that pepper plants exposed to Cd stress during growth undergo changes in the macro- and microelemental distribution in leaves, stems, and roots, principal component analysis (PCA) and parallel factor (PARAFAC) analysis were applied to compare bidirectional and multivariate chemometric strategies to assess elemental changes in pepper plants. Since the number of variables and the data generated were large and complex, the application of chemometric tools was justified to facilitate the visualization and interpretation of results. The mineral composition, namely the Ca, Cd, Cu, Fe, K, Mg, Mn, N, and P contents, was assessed in 180 samples of leaves, stems, and roots of the cultivated peppers. Then, PCA and PARAFAC analysis were applied to compare bidirectional and multivariate chemometric strategies to assess elemental changes throughout pepper plants. The visualization of the trend on each sample and their intrinsic relationship with the variables were possible with the application of PCA. The use of PARAFAC analysis permitted the simultaneous study of all samples in a straightforward representation of the information that facilitated a quick and comprehensive understanding of the spatial distribution of elements in plants. Thus, macroelements (Ca, K, Mg, N, and P) that were found in higher concentrations in leaves did not present significant differences in the distribution along the plants under different treatment conditions. In contrast, a significant impact on the microelement (Cu, Fe, and Mn) distribution was produced between uncontaminated and contaminated samples. This analysis revealed a significant accumulation of Cd in roots and adverse effects on normal plant growth, demonstrating their level of phytotoxicity to pepper.

The study of EDTA enhanced Cd accumulation and formation in Napier grass using synchrotron μX-ray fluorescence imaging and X-ray absorption spectroscopy

In this study, Pennisetum purpureum, a fast growing and highly tolerant plant, was used as a Cadmium (Cd) hyperaccumulator associated with Ethylenediaminetetraacetic acid (EDTA). It was found that P. purpureum's shoots and roots grown in the presence of EDTA accumulated significantly higher Cd than those grown in the absence of EDTA. Synchrotron μX-ray fluorescence imaging demonstrated the significant Cd accumulation in the internal tissues of root: endodermis, xylem and phloem, while at stem, it was found at the external tissues: epidermis and phloem. Synchrotron X-ray Absorption Spectroscopy was used to characterize the Cd speciation in plant cells. P. purpureum was discovered to accumulate Cd (II) form. In order to determine the possibilities of Cd bonding in chemical compounds, linear combination fitting of Cd K-edge XANES spectra was carried out. The EDTA-free condition as a control revealed mainly Cd–N/O bonding ligand, while the EDTA-treated condition mostly showed Cd–N/O and Cd–S bonding. The overall results suggested that EDTA is a potential substance to enhance the efficiency of Cd absorption in P. purpureum.

Combining Contamination Indices and Multivariate Statistical Analysis for Metal Pollution Evaluation during the Last Century in Lacustrine Sediments of Lacu Sărat Lake, Romania.

Integrated study of both water and sediment in lakes provides important information regarding the human impact on the environment. The current work is focused on the correlation between age, source, composition, and degree of human intervention over the last 178 years and health impact of sediments from Lacu Sărat Lake (Romania), one of the most important balneo-climateric resorts in the country. The novelty relies on the fact that this is the first time the temporal patterns of metal contamination and the human health effects associated with the metal exposure from sediment core samples have been assessed. The sediment contamination status was determined by evaluating several indices, such as the enrichment factor, geo-accumulation index, metal pollution index, and potential ecological risk index, etc. Results showed a significant accumulation of Cd, Cr, As and Ni and a major contribution of Pb, Zn, Cd, Hg, Cr as well as Cu to the potential acute toxicity. The sediment quality guidelines emphasized a risk concerning the life and proper development of benthic organisms in Lacu Sărat Lake. Moreover, the incidental ingestion lifetime carcinogenic risk values for As and Cr suggest a potential risk of developing cancer. A strong human impact was observed especially between 1950 and 1990, which can be attributed to the rapid economic growth and intensive industrial development strategies pursued by the communist political regime in Romania.

Nano-selenium alleviates cadmium-induced cerebellar injury by activating metal regulatory transcription factor 1 mediated metal response

This study aims to investigate the role of metal regulatory transcription factor 1 (MTF1)-mediated metal response in cadmium (Cd)-induced cerebellar injury, and to evaluate the antagonistic effects of nano-selenium (Nano-Se) against Cd toxicity. A total of 80 chicks (1 d old, male, Hy-Line Variety White) were randomly allocated to 4 treatment groups for 3 months: the control group (fed with a basic diet, n=20), the Nano-Se group (basic diet with 1 mg/kg nano-Se 1mg/kg Nano-Se in basic diet, n=20), the Nano-Se+Cd group (basic diet with1mg/kg Nano-Se and 140mg/kg CdCl2, n=20) and the Cd group (basic diet with140mg/kg CdCl2 , n=20). The results of the experiment showed that the Purkinje cells were significantly decreased with their degradation and indistinct nucleoli after Cd exposure. Moreover, exposure to Cd caused a significant accumulation of Cd and cupper. However, the contents of Se, iron, and zinc were decreased, thereby disturbing the metal homeostasis in the cerebellum. The Cd exposure also resulted in high levels of malondialdehyde (MDA) and down regulation of selenoprotein transcriptome. Furthermore, the expressions of MTF1, metallothionein 1 (MT1), MT2, zinc transporter 3 (ZNT3), ZNT5, ZNT10, zrt, irt-like protein 8 (ZIP8), ZIP10, transferrin (TF), ferroportin 1 (FPN1), ATPase copper transporting beta (ATP7B), and copper uptake protein 1 (CTR1) were inhibited by Cd exposure. However, all these changes were significantly alleviated by the supplementation of Nano-Se. This study proved that Cd could disorder metal homeostasis and induce oxidative stress, whereas Nano-Se could relieve all these negative effects caused by Cd via activating the MTF1-mediated metal response in the cerebellum of chicken.

Impact of Cadmium Stress on Growth and Physio-Biochemical Attributes of Eruca sativa Mill.

Plants may experience adverse effects from Cadmium (Cd). As a result of its toxicity and mobility within the soil-plant continuum, it is attracting the attention of soil scientists and plant nutritionists. In this study, we subjected young Eruca sativa Mill. seedlings to different levels of Cd applications (0, 1.5, 6 and 30 µmol/L) via pot experiment to explore its morpho-physio-biochemical adaptations. Our results revealed a significant Cd accumulation in leaves at high Cd stress. It was also demonstrated that Cd stress inhibited photosynthetic rate and pigment levels, ascorbate peroxidase (APX), guaiacol peroxidase (GPX), catalase (CAT), and superoxide dismutase (SOD) enzyme activities, and increased malondialdehyde (MDA) levels. Conversely, the concentration of total ascorbate (TAS) increased at all levels of Cd application, whereas that of ascorbic acid (ASA), and dehydroascorbate (DHA) increased at 1.5 (non-significant), 6, 30 and 6 µmol/L (significant), though their concentrations decreased non-significantly at 30 µmol/L application. In conclusion, Cd-subjected E. sativa seedlings diverted much energy from growth towards the synthesis of anti-oxidant metabolites and osmolytes. However, they did not seem to have protected the E. sativa seedlings from Cd-induced oxidative stress, causing a decrease in osmotic adjustment, and an increase in oxidative damage, which resulted in a reduction in photosynthesis and growth. Accordingly, we recommend that the cultivation of E. sativa should be avoided on soil with Cd contamination.

Heavy Metal Pollution and Health Risk Assessment of Vegetable-Soil Systems of Facilities Irrigated with Wastewater in Northern China.

Soil pollution by heavy metals is a major concern in China and has received much attention in recent years. Aiming to investigate the status of heavy metal pollution and the safety of vegetables in the soil of wastewater-irrigated facilities, this study investigated the distribution and migration characteristics of heavy metals in vegetable–soil systems of facilities in a typical sewage irrigation area of the Xi River, Shenyang City, northern China. Health risks due to the fact of exposure to heavy metals in the vegetable soil of facilities and ingrown vegetables through different exposure pathways were evaluated. Spatial interpolation and a potential ecological risk assessment were applied to evaluate the soil quality. Bioaccumulation factors (BCFs) were used to analyze the absorption and transportation capacity of Cd, Cu, Pb, and Zn by different parts of different vegetables. The results showed that the average concentration of Cd exceeded the standard values by 1.82 times and accumulated by 11 times, suggesting that Cd poses the most severe pollution among the four metals in the soil of facilities in the Xi River sewage irrigation area. In the city, a significant accumulation of Cd in the soil was identified with different spatial distributions. Cd also contributed the most in terms of the estimated potential ecological risk index, while the impacts of the other three metals were relatively small. The concentrations of heavy metals were mostly lower than the limit set by the corresponding Chinese standards. Various BCFs were observed for the four metals in the order Cd > Zn > Cu > Pb. Vegetables also demonstrated different BCFs in the order of leaf vegetables > Rhizome vegetable > Solanaceae vegetable. The magnitude of the noncarcinogenic risk for all four heavy metals was less than one for all three exposure routes and did not cause significant noncarcinogenic health effects in humans. However, the carcinogenic risk of Cd from some vegetables via dietary intake was considered higher. Protection measures should be taken to implement better pollution control and land use planning.

Toxicological responses of juvenile Chinese shrimp Fenneropenaeus chinensis and swimming crab Portunus trituberculatus exposed to cadmium

Cadmium (Cd) is one of the typical metal pollutants in the Bohai Sea. To evaluate the acute toxicological effects of Cd on marine crustaceans, juvenile Fenneropenaeus chinensis and Portunus trituberculatus were exposed to Cd at environmentally relevant concentrations (5 and 50 μg/L) for 96 h. Cd accumulation, antioxidants and metabolite profiles were characterized to elucidate the responses of juvenile crustaceans to Cd stress. Significant Cd accumulation was observed in both juvenile crustaceans in 50 μg/L Cd-treated group. Results showed that Cd exposure induced hormesis based on the alterations of GSH, SOD and CAT activities (i.e. increased levels in the low concentration of Cd treatment and recovered levels in the high concentration of Cd treatment) in juvenile P. trituberculatus. Similarly, the responses of GSH contents presented hormesis pattern in Cd-treated juvenile F. chinensis. Na+-K+-ATPase contents were significantly elevated in 50 μg/L Cd-treated group. In addition, untargeted NMR-based metabolomics indicated Cd caused the disturbance in osmotic regulation and energy consumption in both juvenile F. chinensis and P. trituberculatus via different pathways. The immunotoxicity and movement disorder were uniquely demonstrated in juvenile P. trituberculatus after Cd exposure.

Screening of Xanthium strumarium (IAPS) Growing on Abandoned Habitats in Khyber Pakhtunkhwa, Pakistan: Perspectives for Phytoremediation

The ecological impacts of invasive alien plant species (IAPS) are well-documented, but a dearth of studies exist concerning its economic, livelihood, biotechnological, and health risk assessment perspectives. IAPS management is currently expanding to ecological indicator and biotechnological aspects. Hence, this study aimed to investigate the phytoremediation potential, biomedical, and bio-safety applications of X. strumarium growing in different abandoned habitats in Khyber Pakhtunkhwa, Pakistan. In this study, 45 plants and soil samples were gathered from 15 abandoned sites and analyzed for Pb, Cd, Cu, and Zn concentrations; bioaccumulation (BA); and translocation factor (TF). The assayed Pb and Cd concentration was higher and above threshold in both soil–plant samples. BAF was found higher in roots than intact plants despite having a significant accumulation of Cd, Pb, and Zn, which shows high metals tolerance of this IAPS. PCA-ordination explained a high cumulative variance (98.9%) and separated roads and densely populated sites with comparatively high metals concentration, indicating the pseudometallophyte nature of X. strumarium. Soil, sand, and plant biomass were shown to be the major determinants affecting the heavy metals concentration and its phytoremediation significantly, which may be due to the soil’s metalliferous nature in the study area. This IAPS exhibited strong translocation and hyperaccumulation capacity in different functional traits with comparatively high Pb, Cd, and Zn (≥1 TF) mobility and, hence, can effectively be used for Pb phytoextraction and phytostabilization of Cd, Cu, and Zn, respectively. Likewise, several other non-spontaneous IAPS growing on such abandoned habitats might be promising for developing a reasonable strategic framework for heavy metals mitigation and health risk implications in this region.

Cd Pollution and Safe Planting Zoning in Paddy Soils: A Case Study in a District of Chongqing

Rice has a strong ability to accumulate Cd in soil, and it is of great significance to study Cd pollution and safe planting zoning in paddy soils. In this work, 300 sets of paddy soil-rice samples were simultaneously collected in 22 towns in a District of Chongqing, and soil pH, soil total and available Cd contents, and brown rice Cd contents were determined. Soil Cd pollution was assessed using the geoaccumulation index, bioconcentration factor, and the single-factor pollution index. Based on the Cd pollution indices of soil and brown rice, safe planting zoning for rice was determined. The results showed that the paddy soils were generally acidic, and total Cd contents ranged from 0.09 mg·kg-1 to 1.60 mg·kg-1, with 35.0% of sites exceeding the risk screening value. The Cd contents of the brown rice ranged from 0.002 mg·kg-1 to 0.808 mg·kg-1 and exceeded the food safety limit in 13.7% of cases. Pearson correlation analysis showed that the Cd content of brown rice was significantly positively correlated with soil total and available Cd (P<0.01). The pollution evaluation indicated that significant Cd accumulation occurred in the paddy soils, with some areas showing light-to-moderate pollution levels. The enrichment coefficients of rice to soil Cd ranged from 0.004 to 1.72. Overall, the paddy soils in the studied area were considered generally safe with respect to Cd pollution, with low-risk areas distributed in the south, west, and east, whereas some medium-high risk areas were detected in eight towns.

Factors affecting cadmium accumulation in the soil profiles in an urban agricultural area

Soil Cd pollution is a serious environmental issue associated with human activities. However, the factors determining exogenous Cd dynamics in the soil profile in a complex environment are not well understood. Based on regional observations from 169 soil profiles across the Chengdu Plain, this study explored the key factors controlling Cd accumulation in the soil profile under actual field conditions. Results showed that total soil Cd contents decreased from 0.377 to 0.196 mg kg−1 with increasing soil depth. The effects of phosphate fertilizer rates, road density and precipitation on the difference in total soil Cd content were only observed in topsoil, while agricultural land-use type and topography had no impact. In contrast, significant differences in the total soil Cd content among different parent material types were found in the 0–20, 40–60 and 60–100 cm soil depths. One sample t-tests showed that significant Cd accumulation occurred in the whole soil profile in soils formed from Q4 (Quaternary Holocene) grey alluvium, while soils formed from Q3 (Quaternary Pleistocene) old alluvium and Q4 grey-brown alluvium showed significant Cd accumulation only in the 0–40 cm soil layers. In the topsoil, acid soluble Cd accounted for the largest proportion of the total Cd in soils formed from Q4 grey alluvium, reducible Cd was the main fraction in soils formed from Q4 grey-brown alluvium, while reducible Cd and residual Cd contributed the largest proportion of the total soil Cd in soils formed from Q3 old alluvium. The above results indicated that parent material was the decisive factor determining the magnitudes and depths of exogenous Cd accumulation in the soil profile due to its impacts on the Cd fraction distributions. These findings suggested that the parent material-induced Cd fraction distributions and accumulation should be considered for effectively exploring targeted remediation strategies for Cd pollution.

Geogenic enrichment of potentially toxic metals in agricultural soils derived from black shale in northwest Zhejiang, China: Pathways to and risks from associated crops

Agricultural soils derived from black shale are typically enriched in potentially toxic metals. This is a serious problem, both in terms of the ecological environment and human health. To assess the levels of potentially toxic metals, 90 paired soil-crops samples were collected from the Anji Country, western Zhejiang province, a typical exposed black shale area in China. Concentrations and bioavailability of potentially toxic metals in the soil-crops system were measured, and the associated potential risks were further evaluated. Results showed the enrichment of potentially toxic metals (i.e. Cd, Pb, Cu, Zn and Ni) in the soil and crop samples, especially a significant accumulation of Cd. Sequential extraction data indicated that Cd in soils derived from black shale was the second most dominant element in the exchangeable fraction (mean at 33.42%) and possessed high bioavailability, whereas Pb was mostly retained in the residual fraction (mean at 76.34%) and exhibited low mobility. The total concentration as well as mobility and bioavailability of Cd were the highest in the sampled soils. This resulted in a high potential ecological risk in areas with agricultural soils derived from black shale, which could eventually jeopardize the health of local residents through various exposure pathways. Overall, our findings provide a scientific basis for developing suitable management strategies to mitigate the exposure to potentially toxic metals in high risk areas.

Global characterization of dose-dependent effects of cadmium in clam Ruditapes philippinarum.

Cadmium (Cd) is being frequently detected in marine organisms. However, dose-dependent effects of Cd challenged unraveling the toxicological mechanisms of Cd to marine organisms and developing biomarkers. Here, the dose-dependent effects of Cd on clams Ruditapes philippinarum following exposure to 5 doses of Cd (3, 9, 27, 81, 243μg/L) were investigated using benchmark dose (BMD) method. By model fitting, calculation of BMD values was performed on transcriptomic profiles, metals concentrations, and antioxidant indices. Cd exposure induced not only significant Cd accumulation in clams, but also marked alterations of essential metals such as Ca, Cu, Zn, Mn, and Fe. Gene regulation posed little influence on essential metal homeostasis, indicated by poor enrichment of differentially expressed genes (DEGs) associated with metal binding and metal transport in lower concentrations of Cd-treated groups. BMD analysis on biological processes and pathways showed that peptide cross-linking was the most sensitive biological process to Cd exposure, followed by focal adhesion, ubiquitin mediated proteolysis, and apoptosis. Occurrence of apoptosis was also confirmed by TUENL-positive staining in gills and hepatopancreas of clams treated with Cd. Furthermore, many DEGs, such as transglutaminases (TGs), metallothionein (MT), STEAP2-like and laccase, which presented linear or monotonic curves and relatively low BMD values, were potentially preferable biomarkers in clams to Cd. Overall, BMD analysis on transcriptomic profiles, metals concentrations and biochemical endpoints unraveled the sensitiveness of key events in response to Cd treatments, which provided new insights in exploring the toxicological mechanisms of Cd in clams as well as biomarker selection.

Cadmium Tolerance in Alfalfa is Related to the Up-regulation of Iron and Sulfur Transporter Genes along with Phytochelatin Accumulation

Cadmium (Cd) toxicity is a serious limitation for agricultural production. In this study, we explored tolerance mechanism associated with Cd toxicity tolerance in alfalfa plants. We used three distinct alfalfa cultivars M. sativa cv. Vernal, M. sativa cv. Zhung Mu, and M. sativa cv. Xing Jiang Daye in this study. Cd showed declined chlorophyll score in Xing Jiang Daye compared with Zhung Mu and Vernal. No significant change observed among the cultivars for root and shoot length. Atomic absorption spectroscopy analysis demonstrated a significant accumulation of Cd, Fe, S and PC in distinct alfalfa cultivars. However, Zhung Mu and Xing Jiang Daye declined Cd accumulation in root, where Fe, S and PC incremented only in Zhung Mu. It suggests that excess Cd in Zhung Mu possibly inhibited in root by the increased accumulation of Fe, S and PC. This was further confirmed by the response of Fe (MsIRT1) and S transporters (MsSULTR1;2 and MsSULTR1;3), and MsPCS1 genes associated with Fe, S and PC availability and translocation in roots and shoots. It suggests that specially the transcript signal inducing the responses to adjust Cd especially in Zhung Mu. This finding provides the essential background for further molecular breeding program for forage crops.

Uptake of Trace Elements in the Water Fern Azolla filiculoides after Short-Term Application of Chestnut Wood Distillate (Pyroligneous Acid).

Treatments of crops with additives to increase their productivity may pose environmental risks and induce negative effects also on non-target organisms. In this study, we investigated the potential effect of chestnut wood distillate (pyroligneous acid) used in agriculture, on the accumulation of trace elements in aquatic plants. As a model species, the common water fern Azolla filiculoides Lam. was selected, being often used also in phytoremediation processes. The content of selected elements of toxicological concern (As, Ba, Cd, Cu, Fe, Mn, Ni, Pb, Zn) was assessed in the fern after short-term treatments (1–3 days) over a range of wood distillate concentrations 1:300 (3.33 mL/L), 1:500 (2.00 mL/L), 1:700 (1.43 mL/L). A statistically significant accumulation of Cd, Cu, Mn, Pb, Zn (1:700) and Pb (1:300) was recorded after three days of incubation, despite the concentrations remained overall low. Using treatment vs. control ratios, a trend of increasing temporal uptake was detected for As, Ba, Fe, Mn, Pb (1:700); Mn, Pb (1:500), and only Pb at 1:300. The results suggested that, under the experimental conditions, element uptake is positively influenced by time and negatively by increasing concentrations of wood distillate, likely due to the acidification of the medium. On the whole, the element concentrations measured in A. filiculoides were low and did not pose any toxicological concern.

Augmentation of cadmium-induced oxidative cytotoxicity by pioglitazone in renal tubular epithelial cells.

The aim of this study was to examine whether a peroxisome proliferator-activated receptor (PPAR)-γ agonist could affect cadmium (Cd)-induced cytotoxicity via the increased expression of megalin, one of the uptake pathways, using renal epithelial LLC-PK1 cells. The treatment with 1 µM Cd for 24 h was not cytotoxic; however, when the cells were pretreated with 0.1 µM pioglitazone for 12 h and then exposed to 1 µM Cd for 24 h, significant accumulation of Cd and cytotoxicity were detected, with an increase in megalin mRNA expression. In addition, pretreatment with pioglitazone significantly increased the Cd-induced generation of hydrogen peroxide and cell apoptosis. The augmented Cd-induced cytotoxicity and apoptosis on preincubation with pioglitazone were inhibited by prior treatment with GW 9662 (PPAR-γ antagonist). These findings suggest that a PPAR-γ agonist could augment Cd-induced oxidative injury and cell apoptosis, possibly dependent on the expression level of the uptake pathway.

Impact of Salicylic Acid and PGPR on the Drought Tolerance and Phytoremediation Potential of Helianthus annus.

The present study was aimed to isolate and characterize plant growth promoting rhizobacteria (PGPR) from the rhizosphere of rainfed area (Karak) in Pakistan. The influence of isolated rhizobacteria, in association with salicylic acid (SA), physiological attributes, drought tolerance potential, and phytoremediation in drought-stressed sunflower exposed was investigated. The isolated bacteria were named P1 and P2 and characterized on the basis of colony morphology and biochemical traits. Both PGPR P1 and P2 were identified on the basis of 16S-rRNA gene sequencing as Planomicrobium chinense strain P1 (Accession No. MF616408) and Bacillus cereus strain P2 (Accession No. MF616406). The fresh cultures (24 h old) of isolates were used to soak the seeds pre-sowing. SA was foliar applied at three-leaf-stage. Likewise, the 30-days-old seedlings (three leaf stage) were exposed to drought stress. Drought stress was imposed to 30-days-old plants (three-leaf stage) by withholding water supply for the next 15 days until the soil water content reached 10%. The PGPR and/or SA treatment resulted in significant accumulation of Cd (84%), Pb (66%), and Ni (65%) in the rhizosphere. PGPR also induced accumulation of Cd and Ni in plant shoot. Combined treatment of PGPR and SA increased the Cu (21%), Co (11%), and Zn (8%) accumulation but decreased (12%) the Fe accumulation as compared to coinoculation of PGPR P1 and P2. Inoculation of plants with PGPR significantly increased shoot length (60%), root length (68%), root fresh (61%), and dry (63%) biomass under water stress. The inoculated plants had increased chlorophyll (67%), carotenoid (70%), leaf protein (64%), sugar (64%), and phenolic (62%) contents while lower leaf proline (62%) content, malondialdehyde (MDA) (64%), and antioxidant enzymes (67%) which suggest their role in drought tolerance. It is concluded that integrative use of PGPR in combination with SA found to be an efficacious strategy to improve the phytoremediation of heavy metals and plant growth under stressed conditions particularly under water-deficient conditions.

Cadmium induced oxidative stress and biochemical responses in cyanobacterium Nostoc muscorum

The present study deals with the growth, photosynthesis, oxidative stress and heavy metal accumulation ability of Nostoc muscorum exposed to different levels (2, 4, 8, 16, 20 μM) of cadmium (Cd) concentrations. Growth and photosynthetic pigments i.e., chlorophyll a, carotenoids and phycocyanin were significantly affected by cadmium exposure and inhibition was found to be dose dependent. 14C-fixation appeared to be more sensitive to Cd than whole cell oxygen evolution. Significant accumulation of Cd in the cells of N. muscorum was noticed after 1 and 2 h of exposure and the accumulation rate was dose and time dependent. Furthermore, the levels of superoxide radicals and hydrogen peroxide (H2O2) were found significantly increased by cadmium exposure which in turn accelerated the formation of malondialdehyde (MDA) content, and protein and DNA damage. The selected dose of Cd (20 μM) showed the induction of new polypeptide of ~23.24 kD and the loss of ~37.84 kD and ~69.63 kD whereas the remaining bands were inhibited as compared to control. Significant DNA fragmentation which is a hallmark of programmed cell death (PCD) was also observed in the cells treated with 20 μM of Cd for 48 h. The decrease in proline and total phenol content at 8 and 16 μM suggest that the cells of N. muscorum were not able to mitigate the oxidative stress induced by cadmium exposure. Similarly, the decreased activities of antioxidant enzymes i.e., superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD) also indicates the failure of the antioxidant defense system of N. muscorum to survive at higher concentration (8 and 16 μM) of cadmium.

The effects of chronic cadmium exposure on repeat swimming performance and anaerobic metabolism in brown trout (Salmo trutta) and lake whitefish (Coregonus clupeaformis)

This study investigates the effect of chronic Cd exposure on the ability to perform repeat swim challenges in brown trout (Salmo trutta) and lake whitefish (Coregonus clupeaformis). Fish were exposed to waterborne Cd (18nM) in moderately hard water (120mgL−1 CaCO3) for 30 days. This level of exposure has been shown to cause sublethal physiological disruption and acclimation responses but no impairment of sustained swimming capacity (Ucrit) in single swim challenges. Swim trials were done over the course of the exposure and each one consisted of an initial swim to 85% of the Ucrit of control fish, a 30min recovery period and finally a second swim challenge to determine Ucrit. Plasma and tissue samples were collected before and after each of the swim periods. As expected from previous studies, Cd exposure resulted in significant accumulation of Cd in gills, liver and kidney but not in white muscle. Exposure also induced a loss of plasma Ca followed by subsequent recovery (in lake whitefish but not brown trout) with few mortalities (100% survival for lake whitefish and 93% for brown trout). Both control and exposed fish swam to 85% of the single swim Ucrit and no differences in performance were seen. The Ucrit of unexposed controls in the second swim challenges were not different from the single swim Ucrit. However, second swim performance was significantly reduced in Cd exposed fish, particularly after a week of exposure where 31% and 38% reductions were observed for brown trout and lake whitefish respectively. Swimming to 85% Ucrit resulted in metabolic expenditure with little recovery after 30min. Few differences were observed between control and Cd exposed fish with the exception of a reduction in resting white muscle ATP stores of Cd exposed fish after 1 week of exposure. The results show that chronic sublethal Cd exposure results in an impairment of swimming ability in repeat swim challenges but this impairment is generally not related to metabolic processes in white muscle.
