Impact of prescribed fire on soil microbial communities in a Southern Appalachian Forest clear-cut.

Escalating wildfire frequency and severity, exacerbated by shifting climate patterns, pose significant ecological and economic challenges. Prescribed burns, a common forest management tool, aim to mitigate wildfire risks and protect biodiversity. Nevertheless, understanding the impact of prescribed burns on soil and microbial communities in temperate mixed forests, considering temporal dynamics and slash fuel types, remains crucial. Our study, conducted at the University of Tennessee Forest Resources AgResearch and Education Center in Oak Ridge, TN, employed controlled burns across various treatments, and the findings indicate that low-intensity prescribed burns have none or minimal short-term effects on soil parameters but may alter soil nutrient concentrations, as evidenced by significant changes in porewater acetate, formate, and nitrate concentrations. These burns also induce shifts in microbial community structure and diversity, with Proteobacteria and Acidobacteria increasing significantly post-fire, possibly aiding soil recovery. In contrast, Verrucomicrobia showed a notable decrease over time, and other specific microbial taxa correlated with soil pH, porewater nitrate, ammonium, and phosphate concentrations. Our research contributes to understanding the intricate relationships between prescribed fire, soil dynamics, and microbial responses in temperate mixed forests in the Southern Appalachian Region, which is valuable for informed land management practices in the face of evolving environmental challenges.

Targeted remodeling of the human gut microbiome using Juemingzi (Senna seed extracts).

The genus Senna contains globally distributed plant species of which the leaves, roots, and seeds have multiple traditional medicinal and nutritional uses. Notable chemical compounds derived from Senna spp. include sennosides and emodin which have been tested for antimicrobial effects in addition to their known laxative functions. However, studies of the effects of the combined chemical components on intact human gut microbiome communities are lacking. This study evaluated the effects of Juemingzi (Senna sp.) extract on the human gut microbiome using SIFR® (Systemic Intestinal Fermentation Research) technology. After a 48-hour human fecal incubation, we measured total bacterial cell density and fermentation products including pH, gas production and concentrations of short chain fatty acids (SCFAs). The initial and post-incubation microbial community structure and functional potential were characterized using shotgun metagenomic sequencing. Juemingzi (Senna seed) extracts displayed strong, taxon-specific anti-microbial effects as indicated by significant reductions in cell density (40%) and intra-sample community diversity. Members of the Bacteroidota were nearly eliminated over the 48-hour incubation. While generally part of a healthy gut microbiome, specific species of Bacteroides can be pathogenic. The active persistence of the members of the Enterobacteriaceae and selected Actinomycetota despite the reduction in overall cell numbers was demonstrated by increased fermentative outputs including high concentrations of gas and acetate with correspondingly reduced pH. These large-scale shifts in microbial community structure indicate the need for further evaluation of dosages and potential administration with prebiotic or synbiotic supplements. Overall, the very specific effects of these extracts may offer the potential for targeted antimicrobial uses or as a tool in the targeted remodeling of the gut microbiome.

Carbon sequestration potential of biochar in soil from the perspective of organic carbon structural modification

Application of biochar derived from biomass resources in soil is an encouraging method to decelerate global warming via carbon sequestration. While there has been extensive research on the priming effect of biochar on the mineralization of native soil organic carbon (nSOC), its correlation with soil organic carbon (SOC) structural variations remains poorly understood. A series of incubation experiments with soils amended with biochar prepared at 300 °C, 450 °C, and 600 °C were performed to explore the shifts in nSOC structure and reveal the interconnections among soil properties, SOC structural variation, and the priming effect induced by biochar. Biochar exhibited an “inhibitory concentration” on soil microorganisms, where 1 % biochar enhanced bacterial and fungal richness and diversity, while 3 % biochar reduced bacterial richness and diversity although soil nutrient conditions were improved. Biochar-treated soil had higher humic acid- and humin-like fractions (HAL and HML) but lower fulvic acid-like components (FAL). Moreover, bulk SOC, FAL, HAL, and HML of soils with 300 °C and 450 °C biochar contained more aromatic-type species (e.g., lignin). Such changes were related to improvement of the soil nutrient condition and shifts in microbial community structures, in addition to the “accumulation of biochar-derived OC in the nSOC pool”. Stable carbon isotopes indicated that the inhibitory effect on SOC mineralization (−94.5 to −8.0 mg CO2-C kg−1 soil) induced by biochar was significantly explained by the change in FAL and HAL contents. The findings improved the understanding of the carbon sequestration potential of biochar in soils for SOC structural modification.

Enhancing anaerobic digestion performance of oxytetracycline-laden wastewater through micro-nano bubble ozonation pretreatment

This study investigated the potential of micro-nano bubble (MNB) ozonation pretreatment to eliminate oxytetracycline (OTC) from wastewater and improve subsequent anaerobic digestion (AD) performance. The findings revealed that MNB ozonation achieved efficient OTC oxidation (>99 % in 60 min), and significantly enhanced methane production by 51 % compared to conventional ozonation (under 30 min of pretreatment). Additionally, MNB ozonation resulted in a decrease in the soluble chemical oxygen demand and reduced volatile fatty acid accumulation compared to conventional ozonation. Furthermore, the study sheds light on the profound impact of OTC and its oxidation by-products on the sludge microbiome. Exposure to OTC and its oxidation by-products resulted in alterations in extracellular polymeric substances composition and led to significant shifts in microbial community structure. This study highlights the promise of MNB ozonation as an effective approach for pharmaceutical pollutant removal and the optimization of AD performance in wastewater treatment, with implications for improved environmental sustainability.

Comparative study of high-solid anaerobic digestion at laboratory and industrial scale – Process performance and microbial community structure

High-solid anaerobic digestion (HSD) for biogas production, compared with wet digestion, is attracting interest due to advantages such as reduced fresh water usage, improved digestate quality and potential for high organic loading rates. However, the underlying processes are not well described and evaluated for HSD. In this study, two laboratory-scale reactors (46 L) of plug-flow type were designed to simulate an industrial-scale HSD process co-digesting food waste, agricultural waste and garden residues under thermophilic conditions. Performance of the laboratory-scale HSD process under stable and disturbed conditions was compared with that in industrial-scale reactors. The results showed that the laboratory- and industrial-scale processes had similar efficiency (93 %) and VS-reduction (43 % and 41 %, respectively) and relatively similar specific methane production (339 and 366 NL CH4/kg VS, respectively). Results from tracer studies combined with chemical analyses showed no phase-separation or plug-flow behaviour along the horizontal axis in either laboratory- or industrial-scale reactors, indicating a need for further process optimisation. Analyses of microbial community structure showed high similarity between laboratory- and industrial-scale, but with some differences caused by downscaling. During the experiment, the laboratory- and industrial-scale processes both showed signs of disturbance, i.e. VFA accumulation at NH4+-N levels > 4 g/L, accompanied by a shift in microbial community structure at both scales, with significant increases in relative abundance of e.g. genera Defluviitoga and Methanothermobacter. In conclusion, this study confirmed the validity of simulating HSD at laboratory scale, thus providing valuable insights into biogas production from high-solid substrates, both in laboratory- and industrial-scale processes.

Survivability and proliferation of microorganisms in bentonite with implication to radioactive waste geological disposal: strong effect of temperature and negligible effect of pressure

As bentonite hosts a diverse spectrum of indigenous microorganisms with the potential to influence the long-term stability of deep geological repositories, it is essential to understand the factors influencing microbial activity under repository conditions. Here, we focus on two factors, i.e., temperature and swelling pressure, using a suspension of Cerny Vrch bentonite to boost microbial activity and evaluate microbial response. Suspensions were exposed either to different pressures (10, 12 and 15 MPa; to simulate the effect of swelling pressure) or elevated temperatures (60, 70, 80 and 90 °C; to simulate the effect of cannister heating) for four weeks. Each treatment was followed by a period of anaerobic incubation at atmospheric pressure/laboratory temperature to assess microbial recovery after treatment. Microbial load and community structure were then estimated using molecular-genetic methods, with presence of living cells confirmed through microscopic analysis. Our study demonstrated that discrete application of pressure did not influence on overall microbial activity or proliferation, implying that pressure evolution during bentonite swelling is not the critical factor responsible for microbial suppression in saturated bentonites. However, pressure treatment caused significant shifts in microbial community structure. We also demonstrated that microbial activity decreased with increasing temperature, and that heat treatment strongly influenced bentonite microbial community structure, with several thermophilic taxa identified. A temperature of 90 °C proved to be limiting for microbial activity and proliferation in all bentonite suspensions. Our study emphasizes the crucial role of a deep understanding of microbial activity under repository-relevant conditions in identifying possible strategies to mitigate the microbial potential within the deep geological repository and increase its long-term stability and safety.

Mechanisms of total phosphorus removal and reduction of β-lactam antibiotic resistance genes by exogenous fungal combination activated sludge

This study utilized Trichoderma and activated sludge to construct combined activated sludge (TAS). The metagenomic approach was employed to examine the shifts in microbial community structure and function of TAS under amoxicillin stress and investigate the mechanism underlying the reduction of β-lactam antibiotic resistance genes (β-ARGs). The findings demonstrated that the elevated aundance of glpa, glpd, ugpq, glpq, and glpb were primarily responsible for the reduction in total phosphorus (TP) removal by TAS. The increased abundance of Proteobacteria and Verrucomicrobia led to enhanced expression of ugpb, phnd, and phne, thereby improving the TP removal of TAS. Furthermore, antibiotic inactivation has gradually become the primary antibiotic resistance mechanism in TAS. Specifically, an increase in the abundance of OXA-309 in TAS will decrease the probability of amoxicillin accumulation in TAS. A decrease in β-ARGs diversity confirmed this. This study presents a novel approach to reducing antibiotic and ARG accumulation in sludge.

Effect of HRT on nitrogen removal from low carbon source wastewater enhanced by slurry and its mechanism

Insufficient carbon sources in low-carbon wastewater have consistently hindered nitrogen removal performance. This work utilized slurry after anaerobic digestion as an additional carbon source, and the optimal hydraulic retention time (HRT) parameters were obtained through adjustment. The findings revealed that, under HRT conditions of 24–12 h, the discharge standards of wastewater treatment plants (WWTPs) could be met. It had been proven that HRT 12 h was the more cost-effective parameter for application in WWTPs, and the removal efficiencies for COD, NH4+-N, NO3–-N, and TN reached impressive levels of 82.43%, 95.24%, 77.49%, and 86.70%, respectively. However, the HRT 6 h struggled to achieve efficient wastewater treatment. Mechanistic analysis demonstrated that shortening the HRT resulted in considerably higher microbial richness, diversity, and evenness, causing notable shifts in microbial community structure. Additionally, a shortened HRT reduced the enrichment abundance of key bacterial phyla (Proteobacteria, Bacteroidetes) and genera (Hydrogenophaga, Filimonas, Meganema) that played an important role in nitrogen and organic matter removal. Furthermore, a shortened HRT increased hydraulic shear forces, impeding the enrichment of carbon and nitrogen metabolism pathways, and reducing the abundance of essential genes associated with glycolysis (HK, pfkA, FBA, GAPDH) and nitrogen metabolism processes (narG, narH, narI, norB, nirS, nosZ). It was noteworthy that the microbial community under the HRT 6 h condition carried the highest risk of microbial pathogenicity. This work revealed the optimal parameters, and carbon and nitrogen metabolism mechanism of wastewater treatment systems with slurry as a carbon source for the first time, and provided novel ideas and insights for low-carbon wastewater treatment.

Nitrogen deposition raises temperature sensitivity of soil organic matter decomposition in subtropical forest

Subtropical ecosystems are strongly affected by nitrogen (N) deposition, impacting soil organic matter (SOM) availability and stocks. Here we aimed to reveal the effects of N deposition on i) the structure and functioning of microbial communities and ii) the temperature sensitivity (Q10) of SOM decomposition. Phosphorus (P) limited evergreen forest in Guangdong Province, southeastern China, was selected, and N deposition (factor level: N (100 kg N ha−1 y−1 (NH4NO3)) and control (water), arranged into randomized complete block design (n = 3)) was performed during 2.5 y. After that soils from 0 to 20 cm were collected, analyzed for the set of parameters and incubated at 15, and 25, and 35 °C for 112 days. N deposition increased the microbial biomass N and the content of fungal and Gram-positive bacterial biomarkers; activities of beta-glucosidase (BG) and acid phosphatase (ACP) also increased showing the intensification of SOM decomposition. The Q10 of SOM decomposition under N deposition was 1.66 and increased by 1.4 times than under control. Xylosidase (BX), BG, and ACP activities increased with temperature under N but decreased with the incubation duration, indicating either low production and/or decomposition of enzymes. Activities of polyphenol-(PPO) and peroxidases (POD) were higher under N than in the control soil and were constant during the incubation showing the intensification of recalcitrant SOM decomposition. At the early incubation stage (10 days), the increase of Q10 of CO2 efflux was explained by the activities of BX, BQ, ACP, and POD and the quality of the available dissolved organic matter pool. At the later incubation stages (112 days), the drop of Q10 of CO2 efflux was due to the depletion of the labile organic substances and the shift of microbial community structure to K-strategists. Thus, N deposition decoupled the effects of extracellular enzyme activities from microbial community structure on Q10 of SOM decomposition in the subtropical forest soil.

A change in substance and microbial community structure during the co-composting of kitchen waste anaerobic digestion effluent, sewage sludge and Chinese medicine residue

Anaerobic digestion is a resource recovery method for organic waste, gaining attention due to carbon reduction. Disposing of anaerobic digestion effluent (ADE) is crucial for developing anaerobic digestion, but conventional wastewater treatment fails to effectively recover nutrients contained in the ADE. In the present study, the ADE without solid-liquid separation was mixed with sewage sludge and Chinese medicine residue for the composting, where the ADE could be recovered at high temperature through humification. Besides, the nitrogen balance, humification process, and microbial dynamics during the composting process were studied. The results showed that the group supplemented with ADE could increase the nitrogen retention efficiency by 2.21 % compared to the control group. High ammonia nitrogen content and salinity did not negatively affect the maturity and phytotoxicity of compost products and even increase the humification degree of compost products. Moreover, additional ADE may not alter microbial community structure, which could contribute to microbial succession. This is the first time to investigate the substance transformation and shift in microbial community structure while applying composting process for ADE treatment, in which the anaerobic-aerobic collaborative disposal process provides an alternative solution for the recovery of ADE.

Compost, plants and endophytes versus metal contamination: choice of a restoration strategy steers the microbiome in polymetallic mine waste

Finding solutions for the remediation and restoration of abandoned mining areas is of great environmental importance as they pose a risk to ecosystem health. In this study, our aim was to determine how remediation strategies with (i) compost amendment, (ii) planting a metal-tolerant grass Bouteloua curtipendula, and (iii) its inoculation with beneficial endophytes influenced the microbiome of metal-contaminated tailings originating from the abandoned Blue Nose Mine, SE Arizona, near Patagonia (USA). We conducted an indoor microcosm experiment followed by a metataxonomic analysis of the mine tailings, compost, and root samples. Our results showed that each remediation strategy promoted a distinct pattern of microbial community structure in the mine tailings, which correlated with changes in their chemical properties. The combination of compost amendment and endophyte inoculation led to the highest prokaryotic diversity and total nitrogen and organic carbon, but also induced shifts in microbial community structure that significantly correlated with an enhanced potential for mobilization of Cu and Sb. Our findings show that soil health metrics (total nitrogen, organic carbon and pH) improved, and microbial community changed, due to organic matter input and endophyte inoculation, which enhanced metal leaching from the mine waste and potentially increased environmental risks posed by Cu and Sb. We further emphasize that because the initial choice of remediation strategy can significantly impact trace element mobility via modulation of both soil chemistry and microbial communities, site specific, bench-scale preliminary tests, as reported here, can help determine the potential risk of a chosen strategy.

Trace elements and microbial community composition associated with airborne PM2.5 in wetlands: A case study in Tablas de Daimiel National Park

Tablas de Daimiel National Park (TDNP) is one of the most important wetlands in the Iberian Peninsula. Due to its location near various cities and new industries focused on agricultural waste revalorization, we investigated concurrently the concentrations of particulate matter 2.5 (PM2.5) mass, trace element composition, and associated microbial communities (bacteria and fungi) during a year-long study. The goal of this study was to explore the dependencies among these physicochemical and microbiological parameters on a seasonal time scale. Additionally, we assessed meteorological conditions and back trajectories to shed light on atmospheric mechanisms and sources related to these elements. We found the variability of PM2.5 to be influenced by local meteorological parameters. Through the analysis of crustal enrichment factors (EFs), bivariate correlations, and air mass patterns, we determined that soil resuspension was the primary contributor to elevated metal concentrations in PM2.5 within the park, followed by other minor sources, such as traffic emissions and Sahara dust intrusions. The measured metal levels were used to calculate the ecological risk in the area, resulting in a low ecological risk index (RI) of 52. Shifts in microbial community structure were observed to be mainly driven by changes in air temperature and Cu concentration. The results from this study contribute to a better understanding of the environmental dynamics in TDNP. Taken together, our findings will aid in the development of effective strategies for its conservation and management.

Antibiotics and their associations with antibiotic resistance genes and microbial communities in estuarine and coastal sediment of Quanzhou Bay, Southeast China

The antibiotic concentrations spanned from 11.2 to 173.8 ng/g, with quinolones and tetracyclines being observed to be prevalent. The amount of microbial biomass as determined by Phospholipid fatty acid (PLFA) ranged from 2.92 to 10.99 mg kg−1, with G- bacteria dominating. A total of 254 distinct ARGs and 10 MEGs were identified, with multidrug ARGs having the highest relative abundance (1.18 × 10−2 to 3.00 × 10−1 copies/16S rRNA gene copies), while vancomycin and sulfonamide resistance genes were the least abundant. Results from canonical-correlation analyses combined with redundancy analysis indicated that macrolides were significantly related to the shifts of microbial community structure in sediments, particularly in G+ bacteria that were more sensitive to antibiotic residues. It was observed that sulfonamide ARGs had a greater correlation with residual antibiotics than other ARGs. This study provided a field evidence that multiple residual antibiotics from coastal sites could cause fundamental shifts in microbial community and their associated ARGs.

Impact of a natural disturbance on the performance and microbial communities in a full-scale constructed wetland for industrial wastewater treatment

Constructed Wetlands (CWs) are a cost-effective, versatile and sustainable choice for wastewater treatment. In these environments, microbial communities play a significant role in pollutant removal. However, little is known about how microbial communities in full-scale CWs contribute to maintaining water quality or how their dynamics change in response to pulse disturbances such as fire or freezes. Furthermore, few studies have examined the relationship between CW microbial community structure and performance in full-scale industrial operations. We characterized the water-column and leaf-litter layer microbial communities in a 110-acre free water surface CW that provides tertiary wastewater treatment to a plastics manufacturing plant. The CW’s sampling campaign was conducted over a 12-month period that included Winter Storm Uri, a 100-year freeze event. Analysis of 16S rRNA gene amplicon sequences revealed that the bacterial communities experienced a temporal shift. There was also a shift in microbial community structure between the influent and the first segment of the CW. However, no differences in microbial community structure were observed in the second segment of the CW. There was a negative association between microbial community diversity and chlorophyll a, as well as microbial community diversity and total suspended solids (TSS); demonstrating an increase in microbial biodiversity as water quality improved throughout the CW. Six months after the freeze, CW performance in terms of removal of water quality constituents began to return to former removal trends. Yet, there was still a significant difference in microbial community structure within the CW relative to the previous year. This suggests CW functional resilience despite a shift in microbial community structure in the wetland.

Facilitative interaction networks in experimental microbial community dynamics.

Facilitative interactions between microbial species are ubiquitous in various types of ecosystems on the Earth. Therefore, inferring how entangled webs of interspecific interactions shift through time in microbial ecosystems is an essential step for understanding ecological processes driving microbiome dynamics. By compiling shotgun metagenomic sequencing data of an experimental microbial community, we examined how the architectural features of facilitative interaction networks could change through time. A metabolic modeling approach for estimating dependence between microbial genomes (species) allowed us to infer the network structure of potential facilitative interactions at 13 time points through the 110-day monitoring of experimental microbiomes. We then found that positive feedback loops, which were theoretically predicted to promote cascade breakdown of ecological communities, existed within the inferred networks of metabolic interactions prior to the drastic community-compositional shift observed in the microbiome time-series. We further applied "directed-graph" analyses to pinpoint potential keystone species located at the "upper stream" positions of such feedback loops. These analyses on facilitative interactions will help us understand key mechanisms causing catastrophic shifts in microbial community structure.

Microbiomes of a disease-resistant genotype of Acropora cervicornis are resistant to acute, but not chronic, nutrient enrichment

Chronically high levels of inorganic nutrients have been documented in Florida’s coral reefs and are linked to increased prevalence and severity of coral bleaching and disease. Naturally disease-resistant genotypes of the staghorn coral Acropora cervicornis are rare, and it is unknown whether prolonged exposure to acute or chronic high nutrient levels will reduce the disease tolerance of these genotypes. Recently, the relative abundance of the bacterial genus Aquarickettsia was identified as a significant indicator of disease susceptibility in A. cervicornis, and the abundance of this bacterial species was previously found to increase under chronic and acute nutrient enrichment. We therefore examined the impact of common constituents of nutrient pollution (phosphate, nitrate, and ammonium) on microbial community structure in a disease-resistant genotype with naturally low abundances of Aquarickettsia. We found that although this putative parasite responded positively to nutrient enrichment in a disease-resistant host, relative abundances remained low (< 0.5%). Further, while microbial diversity was not altered significantly after 3 weeks of nutrient enrichment, 6 weeks of enrichment was sufficient to shift microbiome diversity and composition. Coral growth rates were also reduced by 6 weeks of nitrate treatment compared to untreated conditions. Together these data suggest that the microbiomes of disease-resistant A. cervicornis may be initially resistant to shifts in microbial community structure, but succumb to compositional and diversity alterations after more sustained environmental pressure. As the maintenance of disease-resistant genotypes is critical for coral population management and restoration, a complete understanding of how these genotypes respond to environmental stressors is necessary to predict their longevity.

Assessment of a novel continuous cleaning device using metatranscriptomics in diverse hospital environments.

Despite routine implementation of cleaning and disinfection practices in clinical healthcare settings, high-touch environmental surfaces and contaminated equipment often serve as reservoirs for the transmission of pathogens associated with healthcare-associated infections (HAIs). The current study involved the analysis of high-touch surface swabs using a metatranscriptomic sequencing workflow (CSI-Dx™) to assess the efficacy of cleanSURFACES® technology in decreasing microbial burden by limiting re-contamination. This is a non-human single center study conducted in the Emergency Department (ED) and on an inpatient Oncology Ward of Walter Reed National Military Medical Center that have followed hygienic practices during the COVID-19 pandemic environment. Although there was no difference in observed microbial richness (two-tailed Wilcoxon test with Holm correction, P > 0.05), beta diversity findings identified shifts in microbial community structure between surfaces from baseline and post-intervention timepoints (Day 1, Day 7, Day 14, and Day 28). Biomarker and regression analyses identified significant reductions in annotated transcripts for various clinically relevant microorganisms' post-intervention, coagulase-negative staphylococci and Malassezia restricta, at ED and Oncology ward, respectively. Additionally, post-intervention samples predominantly consisted of Proteobacteria and to a lesser extent skin commensals and endogenous environmental microorganisms in both departments. Findings support the value of cleanSURFACES®, when coupled with routine disinfection practices, to effectively impact on the composition of active microbial communities found on high-touch surfaces in two different patient care areas of the hospital (one outpatient and one inpatient) with unique demands and patient-centered practices.

Microbial communities associated with kelp detritus in temperate and subantarctic intertidal sediments.

Kelp forests, among the most productive ecosystems on Earth, cover large areas of the South Atlantic coast. Sediment heterotrophic bacteria have a pivotal role in the degradation of kelp biomass, however, the response of sediment microbial communities to periodic kelp biomass inputs is mostly unknown. Here, we show that kelp biomass induced rapid changes in overlying water chemistry and shifts in sediment microbial communities, which differed in the experimental systems containing Macrocystis pyrifera (M) and Undaria pinnatifida (U) with sediments of the respective regions. We observed results compatible with the degradation of labile, high molecular weight compounds into smaller and more refractory compounds towards the end of the incubations. The capability of microbial communities to degrade alginate, the major component of kelp cell walls, significantly increased with respect to controls after kelp biomass addition (Absorbance at 235 nm 1.2 ± 0.3 and 1.0 ± 0.2 for M and U, respectively, controls <0.2, t = 4 days). Shifts in microbial community structure (based on 16S rRNA gene amplicon sequencing) were tightly related to the kelp treatment and, to a lesser extent, to the sediment provenance (Principal Coordinates Analysis, 80 % of variation explained in the first two axes). Dissolved oxygen, pH, salinity, alginolytic potential, Absorbance at 235 and 600 nm, total N, total C, and SUVA index correlated significantly with community structure. Differentially abundant populations between kelp-amended treatments and controls included members of the Flavobacteriia class (Algibacter and Polaribacter), and Gammaproteobacteria (Psychromonas and Marinomonas), among others. Metagenomes of M and U-amended sediments contained sequences from 18 of the 19 enzyme families related to alginate or fucoidan degradation. Specific taxonomic groups were associated with enzyme classes targeting different substrates, suggesting niche differentiation. This work expands our knowledge on the patterns of microbial assemblages from intertidal sediments in response to kelp biomass inputs.

Coupled mixotrophic denitrification and utilization of refractory organics driven by Mn redox circulation for significantly enhanced nitrogen removal

Coupled mixotrophic denitrification and degradation of organics driven by redox transition of Mn for nitrogen removal has attracted much attention. Herein, this study explored the removal performance and mechanisms for nitrogen and refractory organics from secondary effluent in up-flow MnOx biofilter. Results showed that the removal of organics and nitrate was significantly enhanced by the synergistic process of heterotrophic denitrification and Mn(II)-driven autotrophic denitrification (MnAD), which were originated from the facilitation of Mn circulation. But nitrate removal was closely related to the types of carbon source and Mn(II) concentration. Single small molecular carbon source (glucose) performed better than mixed carbon source (humic acid and glucose) in nitrate removal process (74.8% in stage 1–2 vs. 54.1% in stage 3–5). And raising external Mn(II) concentration increased the contribution of MnAD (60.2% in stage 5 vs. 46.5% in stage 3) to nitrate removal. Furthermore, the relationship between Mn/N transformation and microbial community structure shifts revealed that the redox transition between Mn(II) and Mn(IV) promoted the enrichment of denitrogenation bacteria and functional genes, thus contributing to pollutants removal. Our studies expand the knowledge of MnOx-mediated pollutants removal processes and support the potential application of MnOx for removal of residual refractory organics and nitrogen.

Co-substrate promoting the biodegradation of refractory DOM in semi-coking wastewater: DOM evolution and microbial community

Semi-coking wastewater treatment has become a difficult issue because of its complexity, heterogeneity and toxicity. In this study, co-metabolism with ethanol or sodium acetate as co-substrates was applied to improve the biological degradation of refractory dissolved organic matter (DOM) in semi-coking wastewater. Kinetic model simulation results indicated that both co-substrates addition improved COD removal efficiency and biodegradation rate. Excitation-emission matrix (EEM) spectroscopy and size exclusion chromatography with organic carbon detector (SEC-OCD) analysis were applied to evaluate the DOM biodegradation in terms of fluorescence and molecular weight (MW). Although co-substrates resulted in a comparable enhanced removal efficiency of protein-like and humic-like fluorophores, there are distinct differences in their specific protein-like fluorescent peaks (Peak C1&C2). Ethanol addition was more conducive to the removal of humic substance with apparent molecular weight (AMW) ranging from 1.1 kDa to 20 kDa. Acetate addition resulted in higher removal of building blocks of humic substances (BB, AMW 0.45–1.1 kDa) and proteinaceous biopolymers (BP, AMW >20 kDa). Co-metabolism triggered the shift of microbial community structure and metabolic functions. Using co-occurrence network analysis, nine keystone genera were identified, which maintained the functional stability of bacterial community responding to organic toxicants stress. LefSe analysis elucidated that the potential major genera involved in refractory organics biodegradation with acetate addition were Stenotrophomonas (40.3%) and Pseudomonas (21.2%), while Pseudoxanthomonas (10.4%) with ethanol addition. Phylogenetic investigation of communities by reconstruction of unobserved states (PICRUSt2) analysis showed that benzoate, styrene, xylene and naphthalene degradation pathway were enhanced by acetate addition as compared to ethanol.
