The usability of MCP-1, fetuin-A, TAS, and TOS levels in the diagnosis of acute myocardial infarction.

Our aims were to determine whether the levels of plasma monocyte chemotactic protein-1 (MCP-1), fetuin-A, serum total antioxidant status (TAS), and serum total oxidant status (TOS) are cardiac biomarkers and to clarify their relationship with each other in acute myocardial infarction (AMI). The study included 90 participants: 60 patients with AMI [30 with and 30 without ST-segment elevation myocardial infarction (STEMI)] and 30 cardiac patients without AMI. The diagnostic values of serum Hs-cTnT, MCP-1, fetuin-A, TAS, and TOS levels in predicting AMI were evaluated statistically. Median levels of MCP-1 [120.10 ng/L (interquartile range: 76.94-230.54 ng/L)] and TOS [2.89 U/MI (IQR: 2.31-3.94 U/Ml)] were statistically higher, and median levels of fetuin-A [433.52 mg/L (IQR: 387.89-584.49 mg/L)] and TAS (3.10 ± 0.86 U/mL) were lower in patients with AMI than in controls. The parameter with the area under the curve (0.815), sensitivity (73.3%), and specificity (66.7%) closest to those of Hs-cTnT was fetuin-A, followed by MCP-1, TOS, and TAS, respectively. A one-unit increase in MCP-1 levels increased the probability of AMI by 1.023 times (p = 0.002). A one-unit increase in fetuin-A levels decreased the probability of AMI by 0.995 times (p = 0.003). A one-unit increase in serum TOS levels was 1.29 times more characteristic of STEMI than of NSTEMI (p = 0.044). MCP-1, oxidative stress parameters, and fetuin-A might support Hs-cTnT levels in the early diagnosis of AMI. Fetuin-A and MCP-1 levels may be independent risk factors for AMI, whereas TOS could be used to distinguish STEMI from NSTEMI.

Determination of Serum Trace Elements and Oxidative Stress in Bitches with Transmissible Venereal Tumor

Background: This study was conducted to compare the changes in oxidative stress levels with some trace elements in blood serum in healthy bitches and bitches with transmissible venereal tumor (TVT). Methods: A total of 40 bitches were used in the study. According to the genital organ examinations, healthy bitches constituted the control group (n=20) and those with venereal tumors constituted the TVT group (n=20). Total oxidant status (TOS), total antioxidant status (TAS) and oxidative stress index (OSI) were determined spectrophotometrically. In addition, some serum trace element levels such as magnesium (Mg), manganese (Mn), iron (Fe), copper (Cu), zinc (Zn) and selenium (Se) were measured using an ICP-MS analysis device. Result: The serum Se, Mg and Zn levels were lower in the TVT group compared to the control group (p less than 0.01) and serum Fe and Cu levels were higher in the TVT group than in the control group (p less than 0.01). Serum TOS and OSI levels were higher (p less than 0.001) and serum TAS levels were lower (p less than 0.001) in the TVT group compared to the control group. The study results showed that oxidative stress is high in bitches with TVT, and in parallel with this, changes occur in the amount of certain trace elements that are associated with oxidative stress.

Serum total oxidant and anti-oxidant status in children with cancer.

Oxidative stress has a potential role in carcinogenesis. Anti-oxidant enzymes have a neutralizing effect on both cancer initiation and progression. We aimed to assess the oxidant and anti-oxidant levels of pediatric cancer patients and to compare the levels in healthy controls. The study involved 105 pediatric cancer patients (40 undergoing chemotherapy, 65 survivors) and 40 healthy children. The serum total oxidant status (TOS) and total anti-oxidant status (TAS) were measured. The oxidative stress index was significantly lower in pediatric cancer patients compared to the levels in the controls (0.20 ± 0.07 vs. 0.26 ± 0.10; P = 0.001). The mean serum TAS level was significantly higher in patient groups compared to the level in the control (1.87 ± 0.48 vs. 1.63 ± 0.32 mmol/L, P = 0.001). The TAS level of children with cancer in survivors was also found to be significantly higher compared to the levels in the control group (1.85 ± 0.45 vs. 1.63 ± 0.32 mmol/L, P = 0.005). Radiotherapy, surgery, relapsed disease, presence of metastases, and receiving enteral nutritional support caused no change in the TAS/TOS level. It has been revealed for the first time that the serum total anti-oxidant level was high in children undergoing chemotherapy and the survivor group as well. Moreover, the oxidative stress index was low in children with cancer. Longitudinal prospective studies are needed to reveal the alterations in oxidant status among children with cancer.

Oxidative stress and antioxidant status in threatened preterm labor.

The purpose of this study was the evaluation of total oxidant status (TOS), total antioxidant status (TAS), oxidative stress index (OSI) and superoxide dismutase (SOD) levels in women with threatened preterm labor (TPL) and also to compare the levels of these oxidative stress biomarkers of TPL pregnancies that had preterm and term deliveries. This case-control study was conducted on 46 patients diagnosed with TPL and 47 healthy pregnant women matched for gestational age. Patients with threatened preterm labor were divided into two groups: true preterm birth (TPB) group (n = 16) and false preterm birth (FPB) group (n = 30) groups. Maternal serum SOD, TOS and TAS levels were measured by a spectrophotometric method using a commertial kit. OSI level for each patient was calculated by using the formula: (TOS (μmol·H2O2·equiv/L) × 100)/(TAS (μmol·Trolox·equiv/L)). The mean TAS levels of the TPB and FPB groups were significantly lower than those of the control group (0.96 ± 0.3 vs 1.36 ± 0.34, p1 < 0.001; 0.97 ± 0.22 vs 1.36 ± 0.34, p2 < 0.001, respectively). The mean SOD, TOS and OSI levels of the TPB and FPB groups were significantly higher than those of the control group (p < 0.001). There was no significant difference between the TPB and FPB groups for any oxidative stress biomarkers. The maternal serum oxidative stress biomarkers are increased in pregnancies with TPL. However, these are not effective in predicting preterm birth in pregnancies with TPL.

Evaluation of Oxidative Stress Levels and Dynamic Thiol-disulfide Balance in Patients with Retinopathy of Prematurity

Purpose The aim of this study is to evaluate both dynamic thiol-disulfide homeostasis and oxidative stress (OS) levels in patients with retinopathy of prematurity (ROP). Methods A total of 129 infants of <34 weeks gestational age were enrolled in the present study. The thiol-disulfide homeostasis was determined by using the new, cost-effective and fully automated colorimetric method. Total antioxidant status (TAS), Total oxidant status (TOS) and Oxidative stress index (OSI) levels were evaluated. Results We found serum TAS levels were lower while serum TOS and OSI levels were significantly higher in patients with ROP compare to the without ROP group (p < .05). However, native, total and disulfide values were not statistically significant between the groups (p > .05). In addition, we also evaluated the native, total and disulfide levels in patients with ROP according to grades and no statistically significant results were found (p > .05). Low birth weight (p = .001), gestational age (p = .001) and 5-min Apgar score were significantly lower in the ROP group. Conclusion This study revealed that dynamic thiol-disulfide homeostasis was changed in patients with ROP. Increased TOS and decreased TAS levels may be associated with functional reduction of the antioxidant system due to increased OS. This indicate that ROP patients are highly sensitive to OS. The dynamic thiol-disulfide homeostasis may conduce to the pathophysiological mechanism and disease follow-up in patients with ROP. The results of this study show that ROP patients are highly sensitive to oxidative stress.

SERUM TOTAL OXIDANT AND ANTIOXIDANT STATUS IN CHILDREN WITH CANCER

Oxidative stress has a potential role in carcinogenesis. Antioxidant enzymes have neutralizing effect both on cancer initiation, and progression. We aimed to assess the oxidant and antioxidant levels of pediatric cancer patients and to compare the levels in healthy controls. The study involved 105 pediatric cancer patients (40 undergoing chemotherapy, 65 survivors) and 40 healthy children. The serum total oxidant status (TOS) and total antioxidant status (TAS) were measured. The TOS and oxidative stress index were lower in pediatric cancer patients compared to the levels in the controls (3.73±1.35 vs. 4.21±1.72 µmol/L; p=0.08; 0.20±0.07 vs. 0.26±0.10; p=0.001, respectively). The mean serum TAS level was higher in patient groups compared to the level in the control (1.87±0.48 vs. 1.63±0.32 mmol/L, p=0.001). The TAS level of children with cancer in survivors was still found to be significantly higher compared to the levels in the control group (1.85±0.45 vs. 1.63±0.32 mmol/L, p=0.005). Radiotherapy, surgery, relapsed disease, presence of metastases and receiving enteral nutritional support caused no change in the TAS/TOS level. It has been revealed for the first time that the serum total antioxidant level increased during cancer treatment and didn't normalize after cessation of therapy for a long time.

The Anti-Inflammatory and Antioxidant Effects of Propofol and Sevoflurane in Children With Cyanotic Congenital Heart Disease

The authors aimed to compare the anti-inflammatory and antioxidant effects of propofol and sevoflurane in children with cyanotic congenital heart disease (CCHD) undergoing cardiac surgery with cardiopulmonary bypass. Prospective, randomized, double-blind study. Single center, university hospital. Children ages 1-10 years with CCHD undergoing elective cardiac surgery with cardiopulmonary bypass. Children were randomized to receive general anesthesia with either sevoflurane (group S) or propofol (group P). Systemic inflammatory response syndrome (SIRS) occurrence was assessed at the end of the surgery and at the sixth, 12th, and 24th postoperative hours. Blood samples were obtained at 4 times: after anesthesia induction (T0), after release of the aortic cross-clamp (T1), at the end of the surgery (T2), and at the postoperative 24th hour (T3). The serum levels of interleukin 6 and tumor necrosis factor alpha, and the total antioxidant status (TAS) and total oxidant status, were analyzed. SIRS was more common in group S than in group P at all times (p=0.020, p=0.036, p=0.004, p=0.008). There were no significant differences between the groups in the mean tumor necrosis factor alpha and interleukin 6 levels at any time. The TAS level at T2 was higher in group P than group S (p=0.036). The serum TAS level increased at T2 compared with T0 in group P, but it decreased in group S (p=0.041). The results showed that propofol provided a greater antioxidant effect and reduced SIRS postoperatively more than sevoflurane in children with CCHD undergoing cardiac surgery.

Relationship between Selected Micronutrient Concentrations, Total Antioxidant Status, Pain Severity, and the Image of 1H MR Spectroscopy in Degenerative Spine Disease: A Case-Control Study.

Osteoarthritis of the spine is a common disease and constitutes a significant problem in highly developed countries. Due to the aging of the population, the number of patients with advanced degenerative changes continues to grow. Trace elements and antioxidant status may play an active role in the pathogenesis of osteoarthritis of the spine. The aim of this study was to assess the relationship between total antioxidant status (TAS), the concentration of selected elements (Zn, Cu, Se, Mn) in the serum, nutritional and environmental factors, the severity of pain, and images of degenerative changes in the spine demonstrated in proton magnetic resonance spectroscopy (1H MRS). The study included 90 patients with degenerative spine disease and 40 healthy people. Serum Zn, Cu, Se, and Mn concentrations were determined by atomic absorption spectrometry. The Cu/Zn molar ratio was calculated. TAS was determined spectrophotometrically using a ready-made Randox kit. The severity of spine pain was assessed using the visual analogue scale VAS. In the 1H MRS study, the fat/water ratio was determined in L1 and L5 vertebral bodies and in the L4/5 intervertebral disc. In patients with osteoarthritis of the spine, there was a reduced concentration of Zn and Cu in the serum (0.77 ± 0.22 mg/L, 1.1 ± 0.35 mg/L) compared to the control group (0.83 ± 0.13 mg/L, 1.25 ± 0.41 mg/L, respectively). In the study group, we also observed a significantly lower Cu concentration in smokers (1.07 ± 0.35 mg/L) compared to non-smokers (1.17 ± 0.34 mg/L). A relationship between the female sex and a decreased concentration of TAS in the serum, both in people with degenerative spine disease and in healthy people, has been demonstrated. In patients with serum TAS levels below the reference values, the fat/water ratio was higher in the L5 spine shaft, while in those with elevated Mn levels, the value was higher in the L4/L5 intervertebral disc, which indicates a greater degree of degeneration in both cases. Patients with normal serum Cu concentration experienced lower pain intensity assessed in the VAS scale. The study showed some abnormalities in serum trace element levels and TAS in patients with osteoarthritis of the spine.

The therapeutic effects of distinct exercise types on metabolic syndrome-induced reproductive system impairment in male rats: Potential contribution of mitochondria-related genes.

A sedentary lifestyle and high fructose dietary habits cause diseases, such as metabolic syndrome (MS). The study was aimed to investigate the potential ameliorative effects of different exercise interventions on high fructose-induced MS-mediated reproductive system disruption of male rats. Rats were divided into four groups (n=6): Control, MS, MS+aerobic exercise (AE) and MS+anaerobic exercise (ANE). MS was induced by using tap water containing 30% fructose for 8weeks. After the induction of MS, AE/ANE implementations were started for 6weeks. Testis tissue and serum samples of rats were stored for biochemical and molecular analyses. Serum total antioxidant status level increased in the MS+AE group compared to all groups. Also, serum total oxidant status level, which increased by MS, decreased with AE, but not altered with ANE. Moreover, MS markedly decreased serum luteinizing hormone, but not changed the follicle-stimulating hormone. However, serum hormone levels were similar to the control group in both MS+AE and MS+ANE groups. MS upregulated mitochondria-related genes' mRNA expressions (MFN2, PGC1A, PPARG, PARP2 and TXNL4B). These increases, except for PPARG, were normalized with both exercise types. These results revealed that mitochondria-related genes may have a crucial role in MS-mediated male reproductive impairment and therapeutic effects of exercises.

İdiyopatik Konjenital Kataraktı Olan Pediatrik Hastalarda Sistemik ve Oküler Oksidatif Stres Arasındaki İlişkinin İncelenmesi: Kesitsel Çalışma

Objective: To investigate the systemic and ocular oxidant-antioxidant status of pediatric patients with idiopathic congenital cataracts. Material and Methods: This study investigated the serum and aqueous humor samples of 18 cases with idiopathic congenital cataract and the serum samples of 20 age-and sex-matched healthy cases. Total antioxidant status (TAS), total oxidant status (TOS), oxidative stress index (OSI), total thiol (T-SH), and thiobarbituric acid reactive substances (TBARS) levels in serum and aqueous humor samples were evaluated using spectrophotometric and spectrofluorometric methods. The weight, height, and body mass index parameters were recorded. Results: Study groups had statistically similar serum TAS, TOS, OSI, T-SH, and TBARS levels (p˃0.05). Statistically significant correlations between oxidant-antioxidant status in serum and aqueous humor were not detected in the patient group (p˃0.05). There was a statistically significant positive correlation was observed between the axial length and aqueous humor T-SH levels (r=0.651, p=0.042). Additionally, statistically significant positive correlation was observed between the age and serum TBARS levels in the patient group (r=0.422, p=0.008). Conclusion: In terms of oxidant-antioxidant status, this study did not find a correlation between serum and aqueous humor samples in pediatric patients with idiopathic congenital cataract and no difference in serum samples between study groups. This study suggests that oxidative stress may not be a part of idiopathic cataract development in the pediatric population.

Oxidative and nitrosative stress in patients with meningitis

Introduction and aim. Meningitis is an acute inflammation of the protective membranes covering the brain and spinal cord, known as the meninges. In this study, oxidative and nitrosative stress were evaluated in cerebrospinal fluid (CSF) and blood samples that were taken from patients with meningitis. Our goal was to identify a fast and a reliable biomarker using these parameters in order to the early diagnose of bacterial meningitis. Material and methods. In this study, 37 bacterial meningitis, 30 tuberculous meningitis and 30 viral meningitis cases were included. Serum/CSF total oxidant status (TAS) and total antioxidant status (TOS) were measured by the Erel method. Nitrotyrosine concentrations were quantified by using ELISA in both serum and CSF Results. Serum nitrotyrosine, CSF TAS and TOS levels were not significantly different in three groups (p&gt;0.05). CSF nitrotyrosine levels were significantly higher in bacterial meningitis than tuberculous meningitis group (p&lt;0.05). Viral meningitis patients had higher serum TOS and TAS concentrations than tuberculous meningitis group (p&lt;0.05). Conclusion. As a result, we can say that the oxidative and nitrosative stress markers studied are not a rapid and reliable biomarker in bacterial meningitis’s diagnosis.

SERUM LEVELS OF OXIDATIVE STRESS MARKERS IN SUBCLINICAL AND OVERT HYPOTHYROIDISM VERSUS CONTROL GROUP IN POPULATION OF KUTAHYA CITY, TURKEY

Background: Overproduction of oxygen-related free radicals and inadequate antioxidant defense are critical in hypothyroidism, as they might lead to future life-threatening diseases. The objectives of this study were to compare serum levels of oxidative stress markers; total oxidant status (TOS), total antioxidant status (TAS), oxidative stress index (OSI), paraoxonase-1 (PON-1), and arylesterase (ARYL) in patients with subclinical hypothyroidism (SH) and overt hypothyroidism (OH) versus healthy controls in population of Kutahya city, Turkey.Materials Methods: This cross-sectional study was conducted at Department of Biochemistry, Faculty of Medicine, Kutahya University of Health Sciences, Kutahya, Turkey, from July to December, 2016. Three study groups of 45 each were formed as healthy controls, patients with SH and OH. Serum levels of TOS, TAS, OSI, PON-1, and ARYL were five research variables with ratio scale, but skewed, so described by median and IQR with 95% Cl. Kruskal-Wallis and multiple comparison tests were used for hypotheses testing.Results: Serum levels of TOS and OSI were higher in SH group (p-values 0.032; .0001; respectively) and OH group (p-values .0001; .0001, respectively) than healthy controls, while serum levels of TAS, PON-1 and ARYL were lower in SH group (p-values .0001; .0001; .0001, respectively) and OH group (p-values .0001; .0001; .0001, respectively) than control group.Conclusion: Our study showed that serum levels of TOS and OSI were higher in subclinical and overt hypothyroidism than in healthy controls; while serum levels of TAS, PON-1, and ARYL were lower in subclinical and overt hypothyroidism than in healthy controls.

Serum level of Total Antioxidant Status and Malondialdehyde in the Patients with Polycystic Ovarian Syndrome: a Comparative Study

Background: Polycystic ovary syndrome (PCOS) is a complex endocrine disorder in women of reproductive age and a major cause of infertility. Anti- Mullerian Hormone (AMH), a valid marker of ovarian function, is used for the diagnosis of PCOS. The aim of this study is to assess the serum level of total antioxidant status (TAS) and malondialdehyde (MDA) in the premenopausal women with PCOS. &#x0D; Methods: A cross-sectional analytical study was conducted at Padmashree Diagnostics, Department of Gynecology, India from April 2016 to March 2017. Census method was used to select the samples. The oxidative stress was measured by measuring the level of TAS and MDA. The data was recorded in structured proforma and and enter in SPSS version 16.0. The data were expressed in frequency, percentage, mean and standard deviation and the results obtained were analyzed using pearson’s correlation and student’s t-test. The p-value less than 0.05 was taken as statistically significant.&#x0D; Results: The measured serum AMH level among the patients with PCOS was within normal range in 67.5% (27) and above normal range in 32.5% (13). The serum TAS level in normal AMH group was 814.0 + 291 µMol/L and in above normal range group was 720 + 240 µMol/L, (p &lt;0.001). Similarly, the serum level of MDA in normal AMH group was 6.64 + 0.21 µMol/L, and in above normal range group was 8.78 + 0.22 µMol/L (p = 0.01). The measured level of AMH was significantly associated with the increased level of MDA (p&lt;0.001) and decreased level of TAS (p&lt;0.001). The measured MDA level in high AMH group did show statistically significant than the normal AMH group.&#x0D; Conclusions: Serum level of TAS and MDA along with AMH are valuable diagnostic biomarker for the diagnosis of PCOS in the resource poor settings where ultrasound facility is unavailable.

Oxidative Stress Level in Patients with Subarachnoid Hemorrhage.

One of the antioxidant mechanisms is the dynamic balance between thiol and disulfide, which, in subarachnoid hemorrhage and other chronic diseases, is disrupted in favor of the latter. The two most commonly used oxidative stress (OS) biochemical markers are the oxidative stress index (OSI) value, which indicates the total oxidant status (TOS) and total antioxidant status (TAS) balance, and the thiol-disulfide (TDS) value, which indicates the total thiol (TT) and native thiol (NT) balance. High OS levels require further investigations. We aimed to investigate the OS level in aneurysmal SAH (aSAH) patients. In this clinical prospective study, blood samples were collected from 50 consecutively treated patients with aSAH and 50 volunteers. Serum TOS, TAS, TT, and NT levels were measured using Erel's method via a spectrophotometer. The Glasgow Coma Scale (GCS) scores, Fisher grades, length of hospital stay (LOS), and the Glasgow Outcome Scale (GOS) scores were recorded. Consequently, the OSI and TDS values were calculated in all participants. A statistically significant difference was observed in the TAS, TOS, OSI, and TDS values between the aSAH patients and the controls. The TT and NT values were significantly lower in aSAH patients than in the controls. A correlation was identified between the OSI values and the GCS scores. Although a correlation was observed between the TDS values and the LOS, no correlation was found between the OSI and the TDS values. The OSI and TDS, which are OS indicators, might serve as the additional objective nominal data to evaluate the treatment efficacy and follow-up for SAH patients. Moreover, decreasing the OSI values and increasing the TT values can be used as improvement indicators in the treated aSAH patients. If we can reduce the OS at the early stage of SAH, it could improve the prognosis by reducing both the morbidity and mortality rates. Further randomized investigations are required to prove the findings in this prospective study.

Evaluation of the Effects of Nitrilotriacetic Acid as a Chelating Agent on the Biochemical Toxicity of Lead in Oreochromis niloticus.

In the present research, the effects of sublethal lead (Pb) concentrations on total antioxidant status (TAS), total oxidant status (TOS), oxidative stress index (OSI) levels, enzyme activities (aspartate transaminase, AST; alanine transaminase, ALT; lactate dehydrogenase, LDH), ion levels (magnesium, Mg; sodium, Na; potassium, K; chlorine, Cl; calcium, Ca), and some metabolite levels (cholesterol, triglyceride, HDL, LDL, albumin, total protein) in the blood serum of Oreochromis niloticus and the protective function of nitrilotriacetic acid (NTA) due to its chelating characteristic were investigated. O. niloticus, which has an important position in the food chain and is often preferred in toxicological studies, was exposed to 0.1ppm Pb, 0.1ppm Pb + 0.3ppm NTA, 1ppm Pb, and 1ppm Pb + 3ppm NTA concentrations for 7 and 21days. At the end of the duration, serum TAS and TOS levels were measured spectrophotometrically with Rel Assay Diagnostics; other enzyme activities, ion levels, and metabolite parameters were done by an autoanalyzer using commercial kits. Depending on the exposure periods and concentrations, the changes in the parameters were determined. It is determined that, under the influence of high ambient concentration of lead, TOS, OSI, AST, ALT, LDH, LDL, triglyceride, and Mg levels increased, while TAS, albumin, and K levels decreased after 21days. These increases/decreases in all serum biochemical parameters were generally higher in fish treated with Pb alone compared to fish treated with a mixture of Pb + NTA. This study shows that these changes in serum parameters could be used as an indicator to assess on metal toxicity.

Relationship between diet, oxidative stress, and inflammation in ankylosing spondylitis.

Objectives This study aims to investigate the relationship between disease activity, dietary phytochemical index (DPI), and serum total oxidant status (TOS) and total antioxidant status (TAS) in patients with ankylosing spondylitis (AS).Patients and methods Between August 2020 and January 2021, a total of 37 patients (23 males, 14 females; mean age: 39.3±9.4 years; range, 21 to 61 years) with AS and 36 age-, sex-, and body mass index-matched healthy individuals (24 males, 12 females; mean age: 37.9±8.9 years; range, 20 to 60 years) were included. Serum TAS (μmoLTroloxEq/L) and TOS (μmoL H2O2Eq/L) measurements were performed and the oxidative stress index (OSI) was calculated. Dietary evaluation was made from a one-day dietary record and DPI was calculated.Results Serum TAS level in AS patients was significantly lower than the healthy group (p=0.003). Serum TOS level was similar in both groups. The OSI of patients was significantly higher than the controls (p=0.035). The mean DPI, polyunsaturated fatty acid, n-3 fatty acid, and vitamin C intake of patients were significantly lower than controls (p=0.042, p=0.033, and p=0.022, respectively). A moderate positive correlation was found between the TAS level and DPI of the control group (r=0.352, p=0.035). According to medications, no significant difference was seen between the groups in terms of patients’ characteristics, DPI, and laboratory tests and there was no correlation between DPI, TAS, TOS, and OSI.Conclusion Lower DPI and lower n-3 fatty acid and vitamin C intake in patient group demonstrated that patients with AS should pay more attention to their diet to increase serum antioxidant status.

Effects of amantadine and topiramate on neuronal damage in rats with experimental cerebral ischemia-reperfusion.

Ischemia-reperfusion models are used to evaluate treatment options that may minimize cellular damage after ischemia. To investigate the effects of amantadine and topiramate on apoptosis and cellular oxidative damage. This experiment was performed using 30 male Wistar albino rats. The right internal carotid artery was identified and clamped with an aneurysm clip under general anesthesia, except for animals in the control group. After 10 min of occlusion, the aneurysm clip was removed, allowing reperfusion. After reperfusion and a waiting period of 12 h, the test and control groups were intraperitoneally administered the following solutions: the sham group received 10 mg/kg of isotonic solution, the amantadine group received 20 mg/kg of amantadine, the topiramate group received 40 mg/kg of topiramate, and the amantadine-topiramate group received 20 mg/kg of amantadine and 40 mg/kg of topiramate. After 24 h, the rats were euthanized. Apoptosis was evaluated using the TUNEL method. Total antioxidant status (TAS), total oxidant status (TOS), total thiol, and ischemia-modified albumin (IMA) levels were measured in both brain tissue and serum samples. The rate of apoptosis in the sham and amantadine groups increased significantly compared to the control group and the non-ischemic counter hemisphere. In the amantadine-topiramate group, both serum TAS and tissue thiol levels decreased. Tissue TOS levels were significantly higher in the topiramate group compared to all other test groups. Tissue TAS levels were significantly higher in the amantadine group compared to all other test groups. This experimental ischemia-reperfusion model revealed that topiramate reduces apoptosis in the early period after ischemia and that its combination with amantadine does not provide additional benefits against cell death. However, topiramate did not have an inhibitory effect on the oxidative stress biomarkers used in our study (TAS, TOS, IMA, and thiol). Studies that reveal the neuroprotective mechanism of action and long-term effects of topiramate are needed to complement this study.

Assessment of Oxidative Stress Index in Sub-clinical Hypothyroidism

Objective: Subclinical hypothyroidism(SCH)is a mild form of hypothyroidism where the TSH levels are slightly above the normal rangeand T3 &amp; T4 values are within the normal limits.Oxidative stress caused by reactive oxygen species (ROS) causing an imbalance in the antioxidant/oxidant status of an individual is well recognized in hyperthyroidism &amp; overt hypothyroidism but not in (SCH). Recently much attention is focussed on SCH, as it exhibits the same cardiovascular consequences as overt hypothyroidism. The current study evaluates the oxidative stress index (OSI) and the correlation of OSI with thyroid profile and Paraoxonase -1 (PON- 1), the antioxidant enzyme associated with HDL in subjects with subclinical hypothyroidism. OSI was calculated after measuring the total oxidant status (TOS) and total antioxidant status (TAS). This cross-sectional study included 64 subjects with sub-clinical hypothyroidism and 128 subjects with normal thyroid profile. Age of the subjects ranged between 25 and 75 years. Materials and methods: Serum levels of TAS, TOS and PON-1 was measured in the subjects whose thyroid profile was determined. OSI was calculated and differences between the variables in the two groups were analysed by the non-parametric tests. Correlation analysis was done by the spearman’s test. The diagnostic ability of OSI was assessed by ROC Curve. Results: Significant increase in the OSI and TOS was observed in subjects with SCH. There was no change in the antioxidant status as well as PON-1 activity.Conclusion: OSI can be considered as an efficient marker in the evaluation of oxidative stress.

Does oxidative stress correlate with disease activity and severity in alopecia areata? An analytical study.

Oxidative stress (OS) has been implicated as a contributory factor in the multifactorial etiopathogenesis of alopecia areata (AA). But with the existing data, it is unclear whether OS is a cause or effect of the disease state in Alopecia areata. To compare the OS parameters viz.malon-di-aldehyde (MDA), superoxide dismutase (SOD), total antioxidant status (TAS) in serum of patients with alopecia areata versus age and sex matched controls, and assess their correlation with the severity of the disease. Forty clinically diagnosed patients of alopecia areata and forty (n=40) age and sex-matched healthy controls were recruited. ELISA was used for the evaluation of MDA, and spectrophotometric method was used to evaluate serum TAS and whole blood SOD. Mean serum TAS and whole blood SOD levels of cases were significantly lower than controls (p=0.005 and p=0.002, respectively). Mean serum MDA level of patients was significantly higher compared to controls (p=0.001). While levels of serum TAS and whole blood SOD were found to decrease from mild to severe grades of disease (p=0.003, p < 0.001 respectively), levels of MDA increased with increasing disease severity (p<0.001). The OS parameters were deranged in all subsets of AA, with the greatest derangement seen with whole blood SOD levels.
