Background: Antioxidants are compounds that prevent an oxidation reaction from free radical compounds and important role in capturing free radicals in the human body. Excessive free radicals in the body can cause tissue damage and degenerative diseases. Pandanus amarillyfolius are known to act as natural antioxidant that have the potential to capture free radical compounds, because of their presence of flavonoid compounds. Objective: This study aims to determine the free radical scavenging activity of DPPH against the leaf extract of Pandanus amaryllifolius. Method: Pandanus amaryllifolius extraction was carried out using the maceration method with methanol as solvent (1:10). Concentration was carried out using a rotary evaporator to obtain a thick extract. Phytochemical screening and TLC test were carried out qualitatively using the mobile phase n-butanol: acetic acid: water (6:2:2) and phytochemical testing including flavonoids, alkaloids, saponins, tannins, steroids or terpenoids to determine the presence of flavonoid compounds as antioxidants. Free radical scavenging activity was tested using DPPH free radical scavenger with concentrations of vitamin C 2, 4, 5, 6, 8 ppm and pandanus fragrance extract concentrations of 10, 15, 20, 25 ppm and measured using a UV-Vis spectrophotometer to calculate the IC50 value. Result: The results of the phytochemical screening test of fragrant pandan indicate the presence of flavonoid compounds. The Rf value of the TLC test obtained 0.765 quarcetin and 0.787 extract and the resulting yellow spots, the Rf value of pandan extract which was almost the same as the Rf value of quercetin indicated that the pandan extract may contain the same compounds as quercetin, namely flavonoid compounds. The results of the free radical scavenging activity of methanol extract of pandan leaves were 86,861 g/ml and for comparison, vitamin C was 39,103 g/ml. Based on the statistical analysis of the T-Test, it is known that there is a difference between the sample and the standard with a significant value (p <0.05), namely 0.001 and 0.023. Conclusion: The Pandanus amaryllifolius extract test using the DPPH method is in the strong category (IC50 <50 -100 g/ml) and the comparison of Vitamin C is in the very strong category (IC50 <50 g/ml).

The Impact of Tadbir as a Pre-Application Treatment of Medicinal Plants on Their Chemical Profiles and Biological Activities in Traditional Persian Pharmacy

Based on basic traditional medicine practice, many plants undergo primary treatments to improve their pharmacological characteristics or to attenuate unwanted or unfavorable features of drugs before incorporating them into drug formulations. These treatments are called “Tadbir” in Traditional Persian Medicine (TPM). The purpose of these processes includes but is not limited to eliminating unnecessary compounds, excluding harmful properties such as toxicity and poignancy, and improving their overall natural properties and effectiveness. Here, the effect of vinegar and acetic acid treatment on three herbal specimens, including Carum carvi L. fruits (CC), Trachyspermum ammi (L.) Sprague fruits (TA), and Nigella sativa L. seeds (NS) were investigated. The treated and non-treated samples were subjected to essential oil and methanol extraction. Further, to assess the alterations in the essential oil constituents caused by Tadbir, samples were analyzed by Gas Chromatography/Mass Spectrometry (GC/MS) and HPTLC fingerprinting techniques. Total phenol and flavonoid content and DPPH free radical scavenging activity of methanol extracts were evaluated. As a result of the treatments, TA extracts showed significant rise in phenol and flavonoid contents. Total phenol content increased from 98.50±1.01 in non-treated increased to 181.20±0.27 mg GAE/g Ext. in the vinegar-treated TA fruit extract and total flavonoid showed a rise from 8.97±1.12 to 12.89±0.41 mg QE/g Ext. This may be the reason behind its lower IC50 values in DPPH free radical scavenging assay. Interestingly, Tadbir treatment of TA fruits with 4% acetic acid, lowered the IC50 value from 1019.42±75.65µg/mL in non-treated control to 274.2±17.22 µg/mL; while vinegar caused a lower degree of reduction in IC50 value (369.4±5.54 µg/mL) in DPPH free radical scavenging assay. However, CC fruit extracts, showed a decrease in phenolic content; while demonstrating an increase in flavonoids. Interestingly, phenol and flavonoid contents were significantly enhanced in treated NS seed extracts. The results of all extracts were found significantly different (p<0,05) from each other and the non-treated control. The conclusive results of the present study may partly justify the pre-application of Tadbir treatments of medicinal plants in traditional pharmacy.

Solvent effects on terpenoid compositions and antioxidant activities of Cinnamomum camphora (L.) J. Presl extracts and the main antioxidant agent evaluation through in vitro and in vivo assay

Plant secondary metabolites can protect organisms against oxidative stress caused by adverse environmental conditions. Cinnamomum camphora (L.) J. Presl contains plentiful terpenoids and is subdivided into 5 chemotypes. To develop natural antioxidants using the plant terpenoids, the terpenoid composition and antioxidant abilities of methanol, ethyl acetate, n-hexane and petroleum ether extracts from linalool and eucalyptol chemotypes were investigated, and the solvent effects on in vitro antioxidant activity of 8 main terpenoids were analyzed. Meanwhile, the in vivo effects of two strong antioxidant terpenoids were evaluated. For the two chemotypes, the 4 solvents exhibited the same extracting effects on the terpenoid types, but methanol extracts contained the highest content of terpenoids, which should contribute to their strongest scavenging activity against 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) free radicals. In linalool and eucalyptol chemotype extracts, linalool, eucalyptol, ocimene, α-pinene, D-limonene, terpinene, β-pinene and longifolene were the 8 main terpenoids. Among the 4 solvents, the 8 terpenoids showed the strongest free radical-scavenging activity with methanol as the reaction medium, which might result from strong-polarity methanol easily activating C = C unsaturated bonds in these compounds. This might also contribute to the strongest scavenging activity of methanol extracts against free radicals. Among these main components, ocimene and longifolene separately showed the strongest activity in scavenging DPPH and ABTS free radical. In in vivo assay, the two compounds significantly lowered the reactive oxygen species (ROS) levels in Chlamydomonas reinhardtii, a single-celled model organism, under H2O2 stress, and the lowering effects gradually enhanced with increasing the compound concentration, resulting in the corresponding promoting effect on the algal growth. At the same concentration, longifolene showed the strongest effect on protecting the algal cells against oxidative stress. Therefore, methanol was suitable for extracting terpenoids in natural antioxidant development, and ocimene and longifolene were two strong antioxidant terpenoids without cell toxicity, with the latter having stronger in vivo antioxidant activity.Graphical

Evaluation of Antioxidant Potential and Free Radical Scavenging Activity of Methanol Extract from Scrophularia striata

Objectives: The effectiveness of Scrophularia striata in controlling infections and promoting wound healing has been reported. This study aimed to investigate the antioxidant properties of the methanol extract from Scrophularia striata.
 Methods: Scrophularia striata, a perennial wild plant found in various temperate and tropical areas of Iran, underwent a methanol extraction process to obtain its active compounds. The antioxidant property of the methanol extract of Scrophularia striata was evaluated by quantifying the total antioxidant level, determining the total phenol content, and conducting DPPH radical scavenging assays.
 Results: As the extraction concentrations of Scrophularia striata increase, both the total antioxidant level and total phenol content rise dramatically. With the progression of time and increase in plant extract concentrations, the efficacy of DPPH radical scavenging also shows a corresponding enhancement. Moreover, the IC50% value of Scrophularia striata for DPPH radical scavenging consistently decreases over the observation period.
 Conclusion: The data suggest that Scrophularia striata possesses antioxidant properties. The presence of flavonoids and phenolic compounds in Scrophularia striata highlights its potential to alleviate various disorders by modulating oxidative stress levels.

Determination of In-vitro antioxidant activity of Ougeinia oojeinensis leaves

In this investigation, the In-vitro antioxidant Activity of extracts of leaves of Ougeinia oojeinensis was evaluated. The antioxidant activity in vitro was measured by means DPPH, Reducing power and hydroxyl radical scavenging activity. DPPH radical scavenging activity of ethyl acetate extract of Ougeinia oojeinensis exhibited percent inhibition 66.774% and its IC50 value was found to be 43.503μg/ml. DPPH radical scavenging activity of methanol extract of Ougeinia oojeinensis exhibited percent inhibition 76.267% and its IC50 value was found to be 25.722μg/ml. Ascorbic acid was used as a reference compound which exhibited percent inhibition 89.428% and showed IC50 value of 15.615 μg/ml. Similarly hydroxyl radical scavenging activity of ethyl acetate extract of Ougeinia oojeinensis exhibited percent inhibition 62.183% and its IC50 value was found to be 69.341 μg/ml. Similary, hydroxyl radical scavenging activity of methanol extract of Ougeinia oojeinensis exhibited percent inhibition 75.84% and its IC50 value was found to be 33.360μg/ml. Ascorbic acid was used as a reference compound which exhibited percent inhibition 87.749% and showed IC50 value of 9.904 μg/ml. The methanolic extract showed strong reducing capacity as compared to ethyl acetate extract of Ougeinia oojeinensis.

Phytochemical screening and in-vitro antibacterial and DPPH free radical scavenging activities of methanol extract of root of Combretum album Pers.

The objective of the study was to investigate in-vitro antibacterial and antioxidant activities of methanolic extract of Combretum album Pers. root. In brief antibacterial efficacies of methanol extract and its petroleum ether fraction (MePET), chloroform fraction (MeCH), ethyl acetate fraction (MeEA) and water fraction (MeAQ) were determined by agar well diffusion assay along with Minimum Inhibitory Concentrations (MICs). In-vitro antioxidant efficacies were evaluated by DPPH radical scavenging method. Preliminary phytochemical assay, Fourier Transform Infrared Spectroscopy and Gas Chromatography-Mass Spectroscopy analyses were employed to detect the plausible active ingredients. Methanol extract showed broad spectrum antibacterial activity having highest inhibition zone against Staphylococcus aureus MTCC 2940 (21.67±0.58 mm) and MICs ranged from100 µg/ml to 250 µg/ml. The scavenging activity of methanol extract of root of C. album was concentration dependant and IC50 value was 136.08 µg/ml. The lowest MIC (5 µg/ml) was noted with MeCH against B. subtilis (MTCC 441). MeCH showed highest antioxidant activity with an IC50 value of 12.98 ?g/mL and MePET, MeEA, MeAQ and Ascorbic acid presented antioxidant potential with IC50 values of 16.10, 15.07, 17.44 and 13.40??g/mL respectively. Preliminary phytochemical tests and FT-IR analysis revealed presence of various phytochemicals and functional groups like hydroxyl, carboxylic acids, amidines, amines, aromatics and esters. Three compounds were elucidated from bioactive TLC fraction of MeCH in GC-MS analysis supported by presence of various functional groups in MeCH as detected in FT-IR analysis. It is concluded that methanolic extract of C. album root is a potential antibacterial and antioxidant agent where presence of N-[5-[4-pyridinyl)-1H-1,2,4-triazol-3-yl]-benzamide (compound 1), pyruvic acid (compound 2) and methyloacetone (compound 3) are responsible as possible active ingredients.

Antioxidant activities of pomegranate peel methanolic and water extracts by in vitro methods

In this study, the antioxidant activities of pomegranate (Punica granatum) peel extracts were investigated using different antioxidant activity methods. The antioxidant activities of extracts prepared at different concentrations (100-1000 μg/ml) were compared with α-tocopherol, ascorbic acid, BHT, and BHA standards. The methanol extract of peels had the highest yield (45.5%). Chelating activities (Fe2+) of methanol extract at 100-1000 μg/ml concentrations ranged between 16.63 and 46.45%. However, the chelating activities of all test extracts were lower than the standards. The reducing power of all extracts increased concentration-dependent. DPPH˙˙ radical scavenging activity of methanol extract varied in the range of 18.53 to 88.75%. The highest percentage inhibition of linoleic acid oxidation was found in 250 μg/ml concentrations. Trolox equivalent (TEs) antioxidant capacity values of methanol extract ranged from 3.09 to 15.27 µM TEs 10 g-1. It was determined that methanol extract showed a dose-dependent inhibition of the antioxidant activities.

Evaluation of Antioxidant Activity of Sour Cherry Stalk Extracts by in Vitro Methods

In this study, the antioxidant activities of sour cherry (Prunus cerasus L.) stalk extracts were investigated using β-caroten/linoleic acid, metal chelating, reducing power, DPPH radical scavenging, ABTS method and TEAC method. For this purpose the stalks were extracted separately by using distilled water, methanol, ethyl acetate and hexane solvents following dried and ground to fine powder. The antioxidant activities of extracts prepared at the different concentration (100-1000 μg/mL) were compared with α-tocopherol, ascorbic acid, BHT and BHA standards. The methanol extract of stalk had the highest yield (22%). Chelating activities (Fe2+) of methanol extract at 100-1000 μg/mL concentrations ranged between 18.45 and 67.75%. However, chelating activities of all test extracts were found lower than standards. The reducing power of all extracts except hexane increased depending upon the increase in concentration. DPPH radical scavenging activity of methanol extract varied in the range of 14.57 to 85.78%. The lowest EC50 value was determined as 0.494 µg/mL for methanol extract. But, the highest inhibition percentage of linoleic acid oxidation was found in hexane extract (57.22%). TEAC values of methanol extract ranged from 15.43 to 20.54 µM Trolox equivalent/10 g. It was determined that methanol extract showed a dose-dependent inhibition on the antioxidant activities.

In-vitro antioxidant, antidiabetic and toxic effect of Ageratum houstonianum from Chitwan district Nepal

Medicinal plants are safe and the oldest natural products used for many years to conserve food, to treat health disorders and to prevent diseases. The active chemical compounds formed during secondary vegetal metabolism is usually responsible for the biological properties of some plant species used throughout the world for various purposes including treatment of diabetes, cancer, infectious diseases etc. The present study was undertaken to analyze the phytochemicals by colour differentiation method, to evaluate the toxic effect by phytotoxic assay, antidiabetic activity by α amylase enzyme inhibition and antioxidant potential by DPPH (2,2-diphenyl-1-picrylhydrazyl) scavenging activity of methanolic extract of Ageratum houstonianum. Treatment of problem in carbohydrate uptake needed the inhibition of α-amylase plays a role in the digestion of polysaccharide and glycogen, is made a strategy for controlling diabetes. For this study whole plant was collected, dried and the powder was made. The extraction was carried out by cold percolation in which methanol was used as a solvent. The methanolic extract was subjected to In-vitro phytotoxic activity by adopting the standard protocol. The α-amylase enzyme inhibition activity of plant extract was carried out by using starch as substrate, pancreatic α amylase as the enzyme, and acarbose as standard. The result of in-vitro phytotoxic bioassay revealed that the plant extract showed moderate activity with percentage growth regulation 80 and 25 percent in a concentration-dependent manner. The α-amylase enzyme inhibition was 74.13 to 99.39 percent in a dose-dependent manner. The antioxidant potential of Ageratum houstonianum extract showed mild activity with IC50 123.67 μg/ml as compared to the standard ascorbic acid IC50 5.38 μg/ ml. It is concluded from the present study that Ageratum houstonianum could be used as a natural source to isolate antioxidant, anti-hyperglycemic agent, herbicide and weedicide as it shows a good α amylase inhibition, radical scavenging and phytotoxic activity respectively.

In vitro antioxidant and anticancer activity of methanolic extract of Alangium salvifolium subsp. hexapetalum (Wangerin)

The present study was implemented for revealing the antioxidant and anticancer effect of the aerial parts of methanolic extract of Alangium salvifolium subspecie hexapetalum (Wangerin). Preliminary screening of the methanolic extract was carried out to determine the presence of plant secondary metabolites such as alkaloids, glycosides, flavonoids, terpenoids, steroids, and sterols. Free radical scavenging activity was evaluated using DPPH radical scavenging assay, ABTS radical scavenging assay and reducing power assay. Invert phase contrast microscope and MTT assay method was used to analyse the number of viable cells using HT29 cell lines treated with different concentrations of the aerial parts of methanolic extract of Alangium salvifolium subspecie hexapetalum (Wangerin). The preliminary phytochemical analysis of the methanolic extract of Alangium salvifolium subspecie hexapetalum (Wangerin) unveil the presence of alkaloids, glycosides, flavonoids, tannins, phenolic compounds, carbohydrates and proteins as secondary metabolites. The free radical scavenging activity also revealed the presence of good antioxidant activity in the extract. A dose dependent growth inhibition of HT29 cell lines were seen when treated with methanolic extract of aerial parts of Alangium salvifolium subspecie hexapetalum (Wangerin). As the concentration of extract increases the percentage of viable cells decreases. The IC50 value for radical scavenging activity of methanolic extract was found to be 175μg/ml (DPPH radical scavenging assay) and 40μg/ml (ABTS scavenging assay). The IC50 value for anticancer activity was found to be 30.22μg/ml. The investigational outcome proves that the methanolic extract of aerial parts of Alangium salvifolium subspecie hexapetalum (Wangerin) showed promising antioxidant and cytotoxic activities.

Free radical scavenging activity of methanolic extract of temurui (Murraya koenigii L. Spreng) collected from Langsa, Aceh

Free Radicals are substances that derive from incompletely oxidized compounds that can damage cells and cause inflammatory diseases, atherosclerosis, cancer, and aging. Plants are known as a source of medicines that are being developed by researchers, such as temurui (Murraya koenigii L. Spreng) which collected from Langsa, Aceh. Temurui or curry leaves, a plant that is usually used as a cooking spice by Aceh people, were extracted using maceration method with n-hexane, ethyl acetate, and methanol, respectively. Methanolic extract of temurui showed very strong DPPH scavenging activity with the IC50 value of 77.818 ppm. This activity was contributed by secondary metabolites contained in that plant, such as alkaloids, terpenoids, saponins, flavonoids, and tannins. This plant have great potent to develop as new drugs, especially new antioxidant drugs.

DPPH radical scavenging activity of methanol extract of Indonesian Etlingera elatior flower and leave

This research goal was to investigate the antioxidant activity of methanol extract of kecombrang (Etlingera elatior) flowers and leaves. Extraction processes by using maceration technique. Dried samples were extracted by using commercial methanol for 48 h. Antioxidant activity was measured as DPPH radical scavenging activity. The results showed that IC50 of the methanol extract of kecombrang shoot leaves was 14.53 ppm; the methanol extract of kecombrang mature leaves was 28.97 ppm; and the methanol extract of kecombrang flowers was 105.90 ppm. It is indicated that the extracts can be used as an agent to scavenging free radicals.

SCREENING OF PHYTOCHEMICAL, ANTIOXIDANT ACTIVITY AND ANTI-BACTERIAL ACTIVITY OF MARINE SEAWEEDS

Objective: Phytochemical is naturally present in the seaweeds which biologically play a significant role. The intention of this study was designed to screen the phytochemical constituents and antimicrobial potential of selected seaweed collected from Rameshwaram and Tuticorin Southern coast of India.Methods: The present study investigated the presence of phytochemical constituents and also total phenol, total carbohydrate and total protein quantity of the brown seaweed. Dictyopteris delicatula, Padina gymnospora, Acanthophora spicifera, Portieria hornemannii and Ulva faciata were extracted with solvents having different polarities like methanol, ethanol, chloroform and water and screened for the phytochemical constituents, total phenol, total carbohydrate, total protein and DPPH with standard procedure. The antibacterial activities of the seaweeds were examined by agar well diffusion method.Results: Among the five seaweeds, U. faciata showed the maximum number of active constituents in the methanol extract likewise P. gymnospora was found to have a number of diligent compounds in ethanol extract. A. spicifera showed minimum compounds in ethanol as well as chloroform extract. Moreover A. spicifera, P. hornemannii have shown the superior quantity of protein and carbohydrate when compared to other species. The scavenging activity of methanol extracts at 5 mg/ml concentration P. hornemannii shows 18.2% and A. spicifera possess 17.1%. In the antibacterial activity, methanol extracts of all the seaweed showed a potential inhibitory activity against B. cereus and P. aeruginosa compared to other pathogens.Conclusion: The crude extract of seaweed manifest preferable antimicrobial and antioxidant activities, hence in the future, it would be good if it is further taken for treatment of human diseases or as new antimicrobial agents to replace synthetic antimicrobial agents.

PHYTOCHEMICAL ANALYSIS AND CONSTITUENTS OF HEXANE EXTRACT OF MELASTOMA MALABATHRICUM L

Hexane, dichloromethane, ethyl acetate, methanol and 50 % aqueous methanol extracts of leaf, flower and fruit of Melastoma malabathricum were screened for the presence of different classes of phytochemicals. In phytochemical screening, ethyl acetate, methanol and 50 % aqueous methanol extracts showed the presence of phenolics and flavonoids. Glycosides were found only in the 50 % methanol extract of fruit. Free radical scavenging activity of methanol extract of leaf, flower and fruit was determined using 2,2-Diphenyl-1-picrylhydrazyl (DPPH) free radical. Total phenolic content was measured spectrophotometrically using Folin-Ciocalteu, total flavonoids content by using aluminum chloride and total sugar content by using anthrone reagent. Gallic acid was used as a standard for the calibration of phenolics, catechin for flavonoids and glucose for carbohydrates. The highest amount of phenolic was found in the 50 % methanol extract of leaf (242.88 ± 3.52 mg GAE/g extract), the highest amount of flavonoids was found in the ethyl acetate extract of leaf (127.41 ± 6.33 mg CE/g extract). Total sugar in 50 % methanol extract of fruit was found to be 151.12 ± 8.79 mg GE/g extract. Methanol extracts of flower showed the strongest antioxidant activity with IC50 value of 17.23 μg/ml. Silyl derivative of hexane extracts were prepared by using N-methyl-N-(trimetylsilyl) trifluoroacetamide for gas chromatography-mass spectroscopic (GC-MS) analysis. The GC-MS analysis of hexane extract allowed the identification of 17 phyto-constituents by comparing the fragmentation pattern of each component in mass spectrum with the standard NIST mass spectral database.

IN VITRO ANTIOXIDANT POTENTIAL AND HEPATOPROTECTIVE ACTIVITY OF TAXUS WALLICHIANA

Objectives: Taxus wallichiana, the member of family Taxaceae is used for the cure of various ailments by a large ethnic population native to the forests in Kashmir valley. The aim of this study is to evaluate antioxidant potential and hepatoprotective action of T. wallichiana as claimed by the ethnic population segment. Methods: Antioxidant potential of different T. wallichiana extracts was evaluated by estimating the total phenolics, 1,1-diphenyl -2-picrylhydrazyl (DPPH), reducing power, microsomal lipid peroxidation (LPO), and hydroxyl radical scavenging activity through standard protocols. Hepatoprotective activity of T. wallichiana was determined by evaluation of liver marker enzyme (aspartate transaminase, alanine transaminase, and alkaline phosphatase) levels using standard protocols and by histological analysis of rat liver tissues of different animal groups against CCl4 induced liver damage. Results: Phenolic content expressed as gallic acid equivalents (GAE/g) was found maximum in case of ethanolic extract (153.34 mg GAE/g), followed by extracts of methanol (149.24 mg GAE/g), hexane (134 mg GAE/g), ethyl acetate (128.9 mg GAE/g), chloroform (121.37 mg GAE/g), and aqueous (113.00 mg GAE/g). At concentration of 700 μg/ml, DPPH radical scavenging activity of methanol extract was 88.29% with IC50 (212.00 μg/ml), aqueous (82.47%) with IC50 (258.29 μg/ml), and ethyl acetate (79.57%) with IC50 (301.80%μg/ml). The reducing power of the extracts increased in a concentration dependent manner. At concentration of 700 μg/ml, 79%, 68%, and 56% inhibition was observed with extracts of methanol, aqueous, and ethyl acetate on microsomal LPO with IC50 values of 126.09, 168.83, and151.96 μg/ml, respectively. Superoxide radical scavenging activity of T. wallichiana extracts increased in a dose-dependent manner with IC50 values 170.30 μg/ml (methanol), 257.00 μg/ml (aqueous), and 297.55 μg/ml ethyl acetate extract. T. wallichiana extracts exhibited antioxidant effects on calf thymus DNA damage. The results further depict the hepatoprotective action of T. wallichiana extract at doses of 100 and 300 mg/kg and were comparable with that of standard treatment comprising 50 mg/kg-day of Vitamin C, a known hepatoprotective agent. The data were substantiated with histopathological studies of rat liver sections. Conclusion: These results comprehensively depict that T. wallichiana extracts are endowed with the free radical sequestering potential and be employed as potential antioxidant and hepatoprotective arsenal against many oxidative stress linked diseases.
