A Meta-Analysis of fMRI Studies of Youth Cannabis Use: Alterations in Executive Control, Social Cognition/Emotion Processing, and Reward Processing in Cannabis Using Youth.

Background: Adolescent cannabis use (CU) is associated with adverse health outcomes and may be increasing in response to changing cannabis laws. Recent imaging studies have identified differences in brain activity between adult CU and controls that are more prominent in early onset users. Whether these differences are present in adolescent CU and relate to age/developmental stage, sex, or cannabis exposure is unknown. Methods: A systematic review and subsequent effect-size seed-based d mapping (SDM) meta-analysis were conducted to examine differences in blood-oxygen-level-dependent (BOLD) response during fMRI studies between CU and non-using typically developing (TD) youth. Supplemental analyses investigated differences in BOLD signal in CU and TD youth as a function of sex, psychiatric comorbidity, and the dose and severity of cannabis exposure. Results: From 1371 citations, 45 fMRI studies were identified for inclusion in the SDM meta-analysis. These studies compared BOLD response contrasts in 1216 CU and 1486 non-using TD participants. In primary meta-analyses stratified by cognitive paradigms, CU (compared to TD) youth showed greater activation in the rostral medial prefrontal cortex (rmPFC) and decreased activation in the dorsal mPFC (dmPFC) and dorsal anterior cingulate cortex (dACC) during executive control and social cognition/emotion processing, respectively. In meta-regression analyses and subgroup meta-analyses, sex, cannabis use disorder (CUD) severity, and psychiatric comorbidity were correlated with brain activation differences between CU and TD youth in mPFC and insular cortical regions. Activation differences in the caudate, thalamus, insula, dmPFC/dACC, and precentral and postcentral gyri varied as a function of the length of abstinence. Conclusions: Using an SDM meta-analytic approach, this report identified differences in neuronal response between CU and TD youth during executive control, emotion processing, and reward processing in cortical and subcortical brain regions that varied as a function of sex, CUD severity, psychiatric comorbidity, and length of abstinence. Whether aberrant brain function in CU youth is attributable to common predispositional factors, cannabis-induced neuroadaptive changes, or both warrants further investigation.

Distinctive roles of mPFC subregions in forming impressions and guiding social interaction based on others' social behaviour.

People can quickly form impressions of others from their social behaviour, which can guide their future social interactions. This study investigated how the type and timing of others' social decisions affect the impression formation and social interactions. In each trial, participants watched a responder's decision in an ultimatum game, decided whether to choose the responder as their next partner for proposer or responder and reported the perceived warmth, competence and likability of the responder. Participants preferred responders who accepted (i.e. accepters) unfair offers for the responder and those who rejected (i.e. rejecters) unfair offers for the proposer in their next ultimatum game, and the rostral medial prefrontal cortex (mPFC) activity encoded such a strategic context-dependent valuation when choosing partners. Slow rejecters were perceived as warmer than fast rejecters, which was mirrored by the anterior mid-cingulate cortex activity when watching others' decisions, possibly detecting and resolving conflicting impressions. Finally, those who perceived accepters vs rejecters as warmer showed higher ventral mPFC responses to accepters vs rejecters when choosing a partner, regardless of the context. The present study suggests that distinctive subregions of the mPFC may be differentially involved in forming impressions and guiding social interactions with others based on their social behaviours.

Intolerance of uncertainty is associated with heightened responding in the prefrontal cortex during cue-signalled uncertainty of threat

Heightened responding to uncertain threat is considered a hallmark of anxiety disorder pathology. We sought to determine whether individual differences in self-reported intolerance of uncertainty (IU), a key transdiagnostic dimension in anxiety-related pathology, underlies differential recruitment of neural circuitry during cue-signalled uncertainty of threat (n = 42). In an instructed threat of shock task, cues signalled uncertain threat of shock (50%) or certain safety from shock. Ratings of arousal and valence, skin conductance response (SCR), and functional magnetic resonance imaging were acquired. Overall, participants displayed greater ratings of arousal and negative valence, SCR, and amygdala activation to uncertain threat versus safe cues. IU was not associated with greater arousal ratings, SCR, or amygdala activation to uncertain threat versus safe cues. However, we found that high IU was associated with greater ratings of negative valence and greater activity in the medial prefrontal cortex and dorsomedial rostral prefrontal cortex to uncertain threat versus safe cues. These findings suggest that during cue-signalled uncertainty of threat, individuals high in IU rate uncertain threat as aversive and engage prefrontal cortical regions known to be involved in safety-signalling and conscious threat appraisal. Taken together, these findings highlight the potential of IU in modulating safety-signalling and conscious appraisal mechanisms in situations with cue-signalled uncertainty of threat, which may be relevant to models of anxiety-related pathology.

Neural Concomitants of Remote Memory in a Comedian with Exceptional Verbal Memory

Most studies exploring how remote memory is represented in the brain are based on strong episodic self-related components. Because of methodological reasons, much less is known about how the information concerning the semantic part of autobiographical memory is retrieved, and whether the brain correlates differ according to the autobiographical moment of the memory formation. In the present study, we explored the neural concomitants of the retrieval of texts learnt at different periods of life, in a comedian with exceptional verbal memory skills. This 49-year-old comedian was instructed to recite aloud a total of 30 texts he learnt during three different epochs: before the age of 15 years (E1), between the age of 15 and 25 years (E2), and after the age of 25 years (E3). The most salient activation was observed for memory from the farthest period, with a preponderance of the medial rostral prefrontal cortex (PFC) and of the precuneus. There was no hippocampal activation during text retrieval by comparison to a control condition, whatever the learning period. This study supports the assumption that the recall of remote semantic memories can occur without hippocampal activation. We discussed the activation of the rostral PFC during retrieval of the oldest (and best consolidated) memories as the possible involvement of control meta-memory processes rather than memory processes per se.

The neural network underpinning social feedback contingent upon one's action: An fMRI study

Praise enhances motor performance; however, the underlying feedback pathway is unknown. Here, we hypothesized that the social evaluation feedback to the motor system is modified by the top-down effect of the social contingency valuation system, such as the anterior rostral medial prefrontal cortex (arMPFC). We developed a pseudo-interactive task that simplified a conversational student-teacher interaction and conducted a functional magnetic resonance imaging study with 33 participants (13 men, 20 women; mean age = 21.7 years; standard deviation = 2.0 years). The participant inside the scanner uttered the pseudo-English word to the English teacher outside the scanner. The teacher provided feedback of acceptance or rejection by either gestures or words, through video. As a control condition, the pseudo-word was read aloud by a computer. Approval from the teacher enhanced the participants’ pleasure rate. Feedback to the participants’ utterance, either rejection or acceptance, activated the arMPFC. Irrespective of the preceding utterance by self or computer, acceptance compared with rejection activated the right primary visual cortex (V1), and the reverse activated the left V1. This valence-dependent laterality of V1 activation indicates that the effect is not the domain-general modulation of visual processing. Instead, the early visual cortices are part of the valence-specific representation of the social signal. Physio-physiological interaction analysis with the seed regions in the right and left V1 and the modulator region in the arMPFC showed enhanced connectivity with the bilateral primary motor cortex. These findings indicate that the socially contingent, self-relevant signals from others act as feedback to the motor control system, and this process is mediated by the early visual cortex.

Metabolite abnormalities in psychosis risk: A meta-analysis of proton magnetic resonance spectroscopy studies.

Accumulating evidence implicates that individuals at high-risk of psychosis have already exhibited pathophysiological changes in brain metabolites including glutamate, gamma-Aminobutyric Acid (GABA), N-Acetylaspartate (NAA), creatine (Cr), myo-inositol (MI) and choline (Cho). These changes may contribute to the development of schizophrenia and associate with psychotic genes. However, specific metabolic changes of brain sub-regions in individuals at risk have still been controversial. Thus, the current study aimed to investigate the brain metabolic changes including glutamate, Glx, GABA, GABA/Glx, NAA, Cr, MI and Cho levels in individuals at risk by conducting a case-control meta-analysis and meta-regression of proton magnetic resonance spectroscopy studies. Primary outcomes revealed that individuals at risk exhibited increased Cr levels at the rostral medial prefrontal cortex (rmPFC), decreased NAA and Cr levels at the thalamus, and increased MI levels at the dorsolateral prefrontal cortex. Sub-group analyses further indicated that individuals with clinical high-risk (CHR) exhibited increased Cr levels at the medial prefrontal cortex (mPFC) and decreased Glx levels at the thalamus, while individuals with genetic risk (siblings of psychiatric patients) exhibited significant increased Glx and MI levels at the mPFC. However, GABA, GABA/Glx and Cho levels showed no significant result. These findings suggest that the dysfunctional metabolites at the mPFC and the thalamus may be an essential neurobiological basis at the early stage of psychosis.

0315 Gray Matter Volume of the Rostral Medial Prefrontal Cortex is Associated with Resilience to Mood Decline During Overnight Sleep Deprivation

Abstract Introduction Vulnerability to sleep deprivation (SD) has been attributed to inter-individual trait-like differences in the ability to sustain vigilance and subjective alertness, which may have distinct neurobiological substrates. We have previously shown that reduced suppression of the Default Mode Network (DMN) during a cognitive task was predictive of global vulnerability to SD. However, little is known about vulnerability to mood decrements during SD and the underlying neurobiological mechanisms. Using voxel-based morphometry (VBM), we assessed structural differences in gray matter volume (GMV) of a region of the anterior DMN, the medial prefrontal cortex and its association with self-reported mood during 29 hours of SD. Methods 45 healthy participants (23 male; Ages 20-43) underwent 3T structural magnetic resonance imaging (MRI). Within 4 days, participants underwent an overnight SD session (29 hours awake total) which included hourly mood assessments with several visual analog mood scales (VAMS) assessing positive and negative affect. Hourly VAMS data were converted into a comparative metric of percent worsening of mood scores from 19:00 until noon the next day. These scores were averaged to determine a “mood resilience” score, with higher scores indicating greater mood sustainment. Using SPM12, the mean mood resilience scores were correlated with whole-brain gray matter volume, restricted to the medial prefrontal cortex, p<.05, FWE corrected, with a cluster threshold of 137 voxels. Results Overnight mood resilience was significantly correlated with greater grey matter volume in right rostral medial prefrontal cortex (p<.05, corrected; k=137). Conclusion Individuals with greater gray matter volume within a circumscribed region of the right medial prefrontal cortex demonstrated greater resilience to mood degradation over 29 hours of continuous wakefulness. This same region of the brain has been shown to be critical for the passive maintenance of emotions. We speculate that greater GMV could protect against mood decline by better sustaining emotional state during SD. Attenuated activation of the anterior rostral medial prefrontal cortex on self-relevant social reward processing in individuals with autism spectrum disorder.

The social motivation hypothesis posits that people with autism spectrum disorder (ASD) find social stimuli less rewarding and are therefore less motivated towards social interaction than people with neuro-typical development (TD). However, the less rewarding social stimuli characteristics during social interaction for people with ASD are largely unknown. The contingent positive responsiveness of others relevant to self-action motivates the early development of social interaction, thus representing a social reward. As individuals with ASD often exhibit atypical responses to self-relevant stimuli in their early life, we hypothesized that the self-relevant responses of others are less rewarding for individuals with ASD. To test this hypothesis, we conducted a functional magnetic resonance imaging study using a social contingency task. During the task, the participants attempted to make the audience laugh by telling funny jokes and thus activating the anterior rostral medial prefrontal cortex (arMPFC) of TD individuals (Sumiya etal., 2017). We explicitly predicted that the atypical activation of the arMPFC is related to the reduced reward value of self-relevant responses to others in individuals with ASD. Thirty-one adults with ASD and 24 age- and intelligence quotient-matched TD adults participated in the study. Participants with ASD reported significantly lower pleasure after the audience's responses to their own actions than those in the TD group. Correspondingly, the self-related activation of the arMPFC, defined by the results of our previous study, was attenuated in the ASD group compared to the TD group. The present findings indicate that weak self-relevant outcome processing mediated by the arMPFC of individuals with ASD dampens the rewarding nature of social interaction.

Spike sorting based on shape, phase, and distribution features, and K-TOPS clustering with validity and error indices

Spike sorting is one of the most important data analysis problems in neurophysiology. The precision in all steps of the spike-sorting procedure critically affects the accuracy of all subsequent analyses. After data preprocessing and spike detection have been carried out properly, both feature extraction and spike clustering are the most critical subsequent steps of the spike-sorting procedure. The proposed spike sorting approach comprised a new feature extraction method based on shape, phase, and distribution features of each spike (hereinafter SS-SPDF method), which reveal significant information of the neural events under study. In addition, we applied an efficient clustering algorithm based on K-means and template optimization in phase space (hereinafter K-TOPS) that included two integrative clustering measures (validity and error indices) to verify the cohesion-dispersion among spike events during classification and the misclassification of clustering, respectively. The proposed method/algorithm was tested on both simulated data and real neural recordings. The results obtained for these datasets suggest that our spike sorting approach provides an efficient way for sorting both single-unit spikes and overlapping waveforms. By analyzing raw extracellular recordings collected from the rostral-medial prefrontal cortex (rmPFC) of behaving rabbits during classical eyeblink conditioning, we have demonstrated that the present method/algorithm performs better at classifying spikes and neurons and at assessing their modulating properties than other methods currently used in neurophysiology.

Self-efficacy modulates the neural correlates of craving in male smokers and ex-smokers: an fMRI study.

The regulation of cue-induced craving for cigarettes is a key factor in smoking cessation. Outcomes of smoking cessation have been linked to self-efficacy, faith in one's own ability, in smokers. However, no study has examined the neural basis of self-efficacy during the control of craving. We examined whether self-efficacy can affect the neural response to smoking cues in smokers and ex-smokers using functional magnetic resonance imaging. During scanning, participants were instructed (1) to view smoking-related images passively, (2) to view the smoking-related images with a strategy focused on self-efficacy to control cue-induced craving or (3) to view neutral images. In smokers, the self-efficacy strategy significantly reduced self-reported craving. This strategy was related to increased activation in the rostral medial prefrontal cortex (rmPFC) and the pregenual anterior cingulate cortex in smokers compared with ex-smokers. Furthermore, smokers showed increased effective connectivity between rmPFC and hippocampus and between pregenual anterior cingulate cortex and parahippocampus gyrus when employing the self-efficacy strategy compared with ex-smokers. The magnitude of the rmPFC-hippocampus connectivity was positively correlated with self-reported self-efficacy. Our findings suggest that in smokers, self-efficacy is related to activation and connectivity in brain regions involved in regulating craving and self-assessment. The current study provides evidence for understanding the vunderlying cognitive and neurobiological mechanisms involved in the control of craving to smoke cigarettes.

Youthful Brains in Older Adults: Preserved Neuroanatomy in the Default Mode and Salience Networks Contributes to Youthful Memory in Superaging.

Decline in cognitive skills, especially in memory, is often viewed as part of "normal" aging. Yet some individuals "age better" than others. Building on prior research showing that cortical thickness in one brain region, the anterior midcingulate cortex, is preserved in older adults with memory performance abilities equal to or better than those of people 20-30 years younger (i.e., "superagers"), we examined the structural integrity of two large-scale intrinsic brain networks in superaging: the default mode network, typically engaged during memory encoding and retrieval tasks, and the salience network, typically engaged during attention, motivation, and executive function tasks. We predicted that superagers would have preserved cortical thickness in critical nodes in these networks. We defined superagers (60-80 years old) based on their performance compared to young adults (18-32 years old) on the California Verbal Learning Test Long Delay Free Recall test. We found regions within the networks of interest where the cerebral cortex of superagers was thicker than that of typical older adults, and where superagers were anatomically indistinguishable from young adults; hippocampal volume was also preserved in superagers. Within the full group of older adults, thickness of a number of regions, including the anterior temporal cortex, rostral medial prefrontal cortex, and anterior midcingulate cortex, correlated with memory performance, as did the volume of the hippocampus. These results indicate older adults with youthful memory abilities have youthful brain regions in key paralimbic and limbic nodes of the default mode and salience networks that support attentional, executive, and mnemonic processes subserving memory function. Memory performance typically declines with age, as does cortical structural integrity, yet some older adults maintain youthful memory. We tested the hypothesis that superagers (older individuals with youthful memory performance) would exhibit preserved neuroanatomy in key brain networks subserving memory. We found that superagers not only perform similarly to young adults on memory testing, they also do not show the typical patterns of brain atrophy in certain regions. These regions are contained largely within two major intrinsic brain networks: the default mode network, implicated in memory encoding, storage, and retrieval, and the salience network, associated with attention and executive processes involved in encoding and retrieval. Preserved neuroanatomical integrity in these networks is associated with better memory performance among older adults.

Dissociation of the rostral and dorsolateral prefrontal cortex during sequence learning in saccades: a TMS investigation.

This experiment sought to find whether differences exist between the dorsolateral prefrontal cortex (DLPFC) and the medial rostral prefrontal cortex (MRPFC) for performing stimulus-independent and stimulus-oriented tasks, respectively. To find a causal relationship in these areas, we employed the use of trans-cranial magnetic stimulation (TMS). Prefrontal areas were stimulated whilst participants performed random or predictable sequence learning tasks at stimulus onset (1st presentation of the sequence only for both Random and Predictable), or during the inter-sequence interval. Overall, we found that during the predictable task a significant decrease in saccade latency, gain and duration was found when compared to the randomised conditions, as expected and observed previously. However, TMS stimulation in DLPFC during the delay in the predictive sequence learning task reduced this predictive ability by delaying the saccadic onset and generating abnormal reductions in saccadic gains during prediction. In contrast, we found that stimulation during a delay in MRPFC reversed the normal effects on peak velocity of the task with the predictive task revealing higher peak velocity than the randomised task. These findings provide causal evidence for independent functions of DLPFC and MRPFC in performing stimulus-independent processing during sequence learning in saccades.

A Variable Oscillator Underlies the Measurement of Time Intervals in the Rostral Medial Prefrontal Cortex during Classical Eyeblink Conditioning in Rabbits.

The way in which brains generate working memories (those used for the transient processing and storage of newly acquired information) is still an intriguing question. Here, we report that the firing activities of neurons located in the rostromedial prefrontal cortex recorded in alert behaving rabbits are controlled by a dynamic oscillator. This oscillator generated firing frequencies in a variable band of 3-12 Hz depending on the conditioned stimulus-unconditioned stimulus intervals (1 s, 500 ms, 250 ms) selected for classical eyeblink conditioning of behaving rabbits. Shorter (50 ms) and longer (2 s) intervals failed to activate the oscillator and prevented the acquisition of conditioned eyelid responses. This is an unexpected mechanism to generate sustained firing activities in neural circuits generating working memories.

When gaze opens the channel for communication: Integrative role of IFG and MPFC

Recent advances in the field of cognitive neuroscience have revealed that direct gaze modulates activity in cortical and subcortical key regions of the ‘social brain network’, including the inferior frontal gyrus (IFG) and the anterior rostral medial prefrontal cortex (arMPFC). However, very little is known about how direct gaze is processed during live interaction with a real partner. Here, for the first time we used an experimental setup allowing the participant inside an MRI scanner to interact face-to-face with a partner located in the scanner room. Depending on condition, the participant and the partner were instructed either to look at each other in the eyes or to direct their gaze away from the other. As control conditions, participants gazed at their own eyes, reflected in a mirror, or gazed at a picture of the partner's eyes. Results revealed that direct gaze by the partner was associated with activity in areas involved in production and comprehension of language and action, including the IFG, the premotor cortex (PM), and the supplementary motor area (SMA). Activations in these areas were observed regardless of the participant's gaze behavior. In contrast, increased activity in arMPFC, an area involved in inference of other mental states during social interaction and communication, was only observed when the participant reciprocated the partner's direct gaze so as to establish mutual gaze. Psychophysiological interaction (PPI) analysis revealed effective connectivity between the IFG and the arMPFC during mutual gaze. This suggests that, within a larger network concerned with the processing of social gaze, mutual gaze with a real partner is established by an increased coupling between areas involved in the detection of communicative intentions, language, and social interaction.

Neural mechanisms of economic commitment in the human medial prefrontal cortex.

Neurobiologists have studied decisions by offering successive, independent choices between goods or gambles. However, choices often have lasting consequences, as when investing in a house or choosing a partner. Here, humans decided whether to commit (by acceptance or rejection) to prospects that provided sustained financial return. BOLD signals in the rostral medial prefrontal cortex (rmPFC) encoded stimulus value only when acceptance or rejection was deferred into the future, suggesting a role in integrating value signals over time. By contrast, the dorsal anterior cingulate cortex (dACC) encoded stimulus value only when participants rejected (or deferred accepting) a prospect. dACC BOLD signals reflected two decision biases-to defer commitments to later, and to weight potential losses more heavily than gains-that (paradoxically) maximised reward in this task. These findings offer fresh insights into the pressures that shape economic decisions, and the computation of value in the medial prefrontal cortex.

Creating external reminders for delayed intentions: Dissociable influence on “task-positive” and “task-negative” brain networks

Studies of prospective memory and other paradigms requiring participants to remember delayed intentions typically reveal a distinction between lateral and medial rostral prefrontal cortex, whereby the experimental condition yields increased signal in the former region and decreased signal in the latter. These regions comprise nodes of larger “task-positive” and “task-negative” networks that often show opposite patterns of signal change in response to diverse cognitive demands. However, it is not clear to what extent activity in these networks is A) inverse but equivalent, or B) functionally dissociable. In order to address this question, participants performed an “intention-offloading” task while undergoing fMRI. On each trial they remembered a delayed intention, which they had the opportunity to fulfill after a brief filled delay. In one condition they were required to set an external reminder of this intention, while in the other they acted without any external memory aid. Results indicated a clear functional dissociation between the two networks. Compared with a control task with no delayed intention, there was a highly significant reduction in task-negative deactivation when participants used an external memory aid. However, there was no reduction in task-positive activation. These results are consistent with previous evidence that medial rostral prefrontal cortex plays a prominent role in representing the content of delayed intentions, accompanied by a reduction in BOLD signal and potentially increased theta-band oscillatory activity. This role is no longer required once an external reminder has been created. By contrast, lateral rostral prefrontal cortex may play a content-free role, unaffected by the offloading of content into the external environment.

Remote effects of hypothalamic lesions in the prefrontal cortex of craniopharygioma patients

Albeit histologically low grade (WHO Io) brain tumors, craniopharyngiomas and/or their surgical removal frequently affect the hypothalamus, amongst other brain regions at risk. Due to rich hypothalamic connections with prefrontal and limbic regions, hypothalamic injury may adversely affect neural substrates of emotion processing and higher cognitive control, including memory and executive functions. The current study is the first to investigate the consequences of hypothalamic involvement on neural substrates of emotional and cognitive functioning.Ten patients with childhood craniopharyngioma and known hypothalamic involvement and fifteen age- and intelligence matched control subjects (median age: 17.8 and 17.3 yrs.) were studied with functional magnetic resonance imaging and an emotional face recognition task. During encoding, participants were asked to classify neutral and emotional faces. In a subsequent recognition phase, participants had to recognize these old faces within a set of new faces.Behavioral performance was comparable between patients and controls. Neural activity revealed, however, differential recruitment of fronto-limbic brain regions during recognition. Patients exhibited an abnormal pattern of task-induced activation and deactivation in the anterior and posterior rostral medial prefrontal cortex and a higher functional coupling between anterior rostral medial prefrontal cortex and the thalamus. Additionally, we found a higher reactivity in the patients’ amygdala to emotional relative to neutral faces when compared to healthy controls.Our data provide first evidence that hypothalamic damage impacts neural correlates of memory retrieval in medial prefrontal cortex, indicating a less efficient use of an area involved in executive control processes. We propose that the deactivation failure in the patients’ anterior rostral medial prefrontal cortex is related to an increased coupling with the thalamus and reflects a reduced efficiency to flexibly adapt to task demands.

How do we process event-based and time-based intentions in the brain? an fMRI study of prospective memory in healthy individuals

Prospective memory (PM) refers to the ability to remember to do something in the future, either in response to an event (event-based) or after a certain amount of time has elapsed (time-based). While the distinction between event- and time-based PM is widely acknowledged in the literature, little is known about the processes they share and those they do not. This is particularly true concerning their brain substrates, as almost all neuroimaging studies so far have focused on event-based PM. We proposed a functional magnetic resonance imaging paradigm assessing both event-based and time-based PM to 20 healthy young individuals. Analyses revealed that event- and time-based PM both induced activation in the posterior frontal and parietal cortices, and deactivation in the medial rostral prefrontal cortex. In addition, activation more specific to each condition, which may underlie differences in strategic monitoring, was highlighted. Thus, occipital areas were more activated during event-based PM, probably reflecting target-checking, while a network comprising the dorsolateral prefrontal cortex, the cuneus/precuneus and, to a lesser extent, the inferior parietal lobule, superior temporal gyrus, and the cerebellum, was more activated in time-based PM, which may reflect the involvement of time-estimation processes. These results confirm the allocation of attentional resources to the maintenance of intention for event-based and time-based PM, as well as the engagement of distinct mechanisms reflecting the monitoring strategies specific to each condition.

Respiratory, metabolic and cardiac functions are altered by disinhibition of subregions of the medial prefrontal cortex

The prefrontal cortex (PFC) is referred to as the visceral motor cortex; however, little is known about whether this region influences respiratory or metabolic outflows. The aim of this study was to describe simultaneous changes in respiratory, metabolic and cardiovascular functions evoked by disinhibition of the medial PFC (mPFC) and adjacent lateral septal nucleus (LSN). In urethane-anaesthetized rats, bicuculline methiodide was microinjected (2 mm; GABA-A receptor antagonist) into 90 sites in the mPFC at 0.72-4.00 mm from bregma. Phrenic nerve amplitude and frequency, arterial pressure, heart rate, splanchnic and lumbar sympathetic nerve activities (SNA), expired CO2, and core and brown adipose tissue temperatures were measured. Novel findings included disturbances to respiratory rhythm evoked from all subregions of the mPFC. Injections into the cingulate cortex evoked reductions in central respiratory function exclusively, whereas in ventral sites, particularly the infralimbic region, increases in respiratory drive and frequency, and metabolic and cardiac outflows were evoked. Disinhibition of sites in surrounding regions revealed that the LSN could evoke cardiovascular changes accompanied by distinct oscillations in SNA, as well as increases in respiratory amplitude. We show that activation of neurons within the mPFC and LSN influence respiratory, metabolic and cardiac outflows in a site-dependent manner. This study has implications with respect to the altered PFC neuronal activity seen in stress-related and mental health disorders, and suggests how basic physiological systems may be affected.

Altered Striatal Functional Connectivity in Subjects With an At-Risk Mental State for Psychosis

Recent functional imaging work in individuals experiencing an at-risk mental state (ARMS) for psychosis has implicated dorsal striatal abnormalities in the emergence of psychotic symptoms, contrasting with earlier findings implicating the ventral striatum. Our aims here were to characterize putative dorsal and ventral striatal circuit-level abnormalities in ARMS individuals using resting-state functional magnetic resonance imaging (fMRI) and to investigate their relationship to positive psychotic symptoms. Resting-state fMRI was acquired in 74 ARMS subjects and 35 matched healthy controls. An established method for mapping ventral and dorsal striatal functional connectivity was used to examine corticostriatal functional integrity. Positive psychotic symptoms were assessed using the Comprehensive Assessment of At-Risk Mental State and the Positive and Negative Syndrome Scale. Compared with healthy controls, ARMS subjects showed reductions in functional connectivity between the dorsal caudate and right dorsolateral prefrontal cortex, left rostral medial prefrontal cortex, and thalamus, and between the dorsal putamen and left thalamic and lenticular nuclei. ARMS subjects also showed increased functional connectivity between the ventral putamen and the insula, frontal operculum, and superior temporal gyrus bilaterally. No differences in ventral striatal (ie, nucleus accumbens) functional connectivity were found. Altered functional connectivity in corticostriatal circuits were significantly correlated with positive psychotic symptoms. Together, these results suggest that risk for psychosis is mediated by a complex interplay of alterations in both dorsal and ventral corticostriatal systems.
