Enzyme-induced degradation of natural and artificial linear polyanions

Synthetic and natural polymers are widely used for constructing drug delivery systems. Biocompatibility, water solubility and non-toxicity make polymers a convenient matrix for encapsulation, delivery and release of bioactive compounds [1–6]. Coupling of a drug with a biodegraded polymer matrix is a promising way for a controlled drug delivery. Along this line, the degradation of the four polymers in the presence of two enzymes in aqueous solutions was investigated. The following polymers were used: natural polysaccharides, sodium alginate and sodium hyaluronate, artificial (modified) sodium carboxymethylcellulose and synthetic sodium polyacrylate (control); their degradation was caused by the addition of alginate lyase and hyaluronidase. The first enzyme only cleaved the specific alginate substrate and left three other intact. Contrastingly, the second enzyme degraded all three polysaccharides, including artificial carboxymethylcellulose, but did not degrade synthetic polyacrylate. The biodegradation of polymers was accompanied by decreasing the size of polymer particles in solution from 100-200 nm down to 20-30 nm; the latter are capable of removing from the body through the kidneys. The initial polysaccharides showed the negative surface charge in aqueous solution, which changed but retained negative after biodegradation. The initial and biodegraded polysaccharides demonstrated negligible cytotoxicity during long exposure period. The obtained results are valuable for the development of polymer carriers for drug encapsulation and delivery.

Bioactive compounds delivery and bioavailability in structured edible oils systems.

The health benefits of bioactive compounds are dependent on the amount of intake as well as on the amount of these compounds that become bioavailable and bioaccessible. Various systems have been developed to deliver and increase the bioaccessibility of bioactive compounds. This review explores the impact of gelled (oleogels, bigels, emulgels, emulsions, hydrogels, and hydrogel beads), micro-(gels, particles, spheres, capsules, emulsions, and solid lipid microparticles) and nanoencapsulated systems (nanoparticles, solid lipid nanoparticles, nanostructured lipid carriers, nanoemulsions, liposomes, and nanoliposomes) on the digestibility and bioavailability of lipophilic and hydrophilic bioactives. Structurant molecules, the oil type, antioxidants, emulsifiers, and coatings in delivery systems with promising potential in food applications are critically discussed. The release and bio-accessibility of bioactive compounds in gelled systems are influenced by various factors, such as the type and concentration of gelators, the gelator-to-oil ratio, the type of antioxidant, the network of the system, and its hydrophobicity. The stability, bioaccessibility, and controlled release of bioactives were improved in structured emulsions. Several variables, including wall material, oil/water ratios, encapsulation process, and pH conditions, can affect the bioactives release in microencapsulated systems. Factors like coating type and core-to-wall ratio impact the stability and release of core components. The encapsulating material, the encapsulation technology, and the nature of the nanomaterials all have an impact on the bioaccessibility of nanoencapsulated systems. Nanoliposomes provide enhanced stability and absorption. In general, all encapsulated systems have shown great potential in improving the distribution and availability of bioactive compounds.

A Systematic Review of Nano-Encapsulation for Improving the Bioavailability of Dietary Supplements and Nutraceuticals

Nanoencapsulation is a growing technique in the field of nutraceuticals and dietary supplements, offering several advantages over non-encapsulated forms. This systematic review assesses the impact of nanoencapsulation on key factors such as bioavailability, stability, tolerance, and encapsulation efficiency. The selected studies demonstrate that nanoencapsulation enhances the protection and controlled release of bioactive compounds, leading to improved efficacy and therapeutic potential. Furthermore, it increases the bioavailability of nutrients and active ingredients while maintaining their stability under different conditions. The review also highlights the positive effects on product safety and consumer tolerance, making nanoencapsulation a promising strategy for optimizing the formulation of dietary supplements. These findings suggest that nanoencapsulation could play a critical role in advancing the effectiveness of nutraceutical products. Alginate Spheres: Influence of Agar and Xanthan Gum Incorporation on Membrane Stability and Permeability

Calcium alginate spheres with a volume of about 5 mL can be used for important purposes. Those that incorporate oolong tea give, in addition to the recreational aspect, the possibility of drinking small quantities of this tea, because oolong tea can be used as a compound with antioxidant properties. This incorporation can be achieved by reverse spherification (5 mL). Six types of spheres have been made, all of them with calcium alginate and the presence or absence of agar-agar and xanthan gum—XG—in two concentrations. The weight loss of the spheres, the release of bioactive compounds over time (a total of 48 h), the surface (internal and external) of the membranes, and the physical characteristics of these membranes have been analyzed. The data obtained indicate that the presence of XG prevents the formation of precipitates inside the spheres and slows down weight loss. It also provides opacity to the spheres. However, the incorporation of agar-agar does not have a significant influence on the different parameters analyzed. The release of catechins reaches a maximum of 80% of what could be achieved under ideal conditions, and it reaches 90% in the first 3 h. The incorporation of XG increases the shelf life of the spheres, slows down the release of chelate, and decreases weight loss over time, allowing for a new perspective on the spherification process.

Thermal stability and in vitro digestive behavior of Pickering emulsion stabilized by high-amylose starch nanocrystals

In this study, high-amylose starch (HAS) was processed using sulfuric acid-ultrasonic cross-linking to produce high-amylose starch nanocrystals (HASNC). These nanocrystals were used to stabilize Pickering emulsions and assess their effectiveness in encapsulating β-carotene. Normal starch nanocrystals (NSNC) were prepared similarly for comparison. The HASNC retained key HAS properties, such as heat and enzyme resistance, providing several advantages to HASNC-stabilized emulsions. First, after exposure to 100 °C heat and in vitro tests simulating the mouth and stomach, the HASNC-stabilized emulsions demonstrated significantly greater stability and higher β-carotene retention compared to the NSNC-stabilized emulsions. This enhanced stability is attributed to the lower gelatinization degree and increased resistance to α-amylase hydrolysis of HASNC, which provides stronger steric stabilization of the oil droplets. Second, during in vitro small intestine tests, the greater enzyme resistance of HASNC allowed for the formation of a denser barrier around the oil droplets, effectively preventing lipase and bile salts from contacting the oil droplets. This led to a reduced rate and extent of lipid digestion and facilitated a sustained-release effect. Consequently, HASNC, as a starch-based emulsifier, show great potential as an effective delivery system for the sustained release of bioactive compounds.

1α,25-Dihydroxyvitamin D Downregulates Adipocyte Impact on Breast Cancer Cell Migration and Adipokine Release.

Excess adiposity is associated with a higher risk of breast cancer metastasis and mortality. Evidence suggests that dietary vitamin D inhibits breast cancer metastasis. However, the mechanistic link between vitamin D's regulation of adipocyte metabolism and metastasis has not been previously investigated. Therefore, the purpose of these experiments was to examine the effect of the active form of vitamin D, 1α,25-dihydroxyvitamin D (1,25(OH)2D), on adipocyte release of bioactive compounds and whether the impact on adipocytes leads to inhibition of breast cancer cell migration, an important step of metastasis. Differentiated 3T3-L1 adipocytes were treated with 1,25(OH)2D for two days, followed by either harvesting the adipocytes or collecting adipocyte-conditioned media without 1,25(OH)2D. A transwell migration assay was conducted with vehicle- or 1,25(OH)2D-conditioned media. In order to explore the mechanism underlying effects on breast cancer metastatic capability, the mRNA expression of leptin, adiponectin, insulin-like growth factor (IGF-1), interleukin-6 (IL-6), and monocyte chemoattractant protein-1 (MCP-1) was measured in adipocytes following either vehicle or 1,25(OH)2D treatment. Conditioned media from 1,25(OH)2D-treated adipocytes inhibited the migration of metastatic MDA-MB-231 breast cancer cells compared to conditioned media from vehicle-treated adipocytes. Treatment of adipocytes with 1,25(OH)2D decreased mRNA expression of leptin, adiponectin, IGF-1, IL-6, and MCP-1. Consistent with mRNA expression, concentrations of leptin, adiponectin, IGF-1, and IL-6 in adipocyte-conditioned media were decreased with 1,25(OH)2D treatment, although MCP-1 remained unchanged. In summary, these results suggest that 1,25(OH)2D alters adipocyte secretions to prevent breast cancer metastasis.

Core-shell electrospun polycaprolactone nanofibers, loaded with rifampicin and coated with silver nanoparticles, for tissue engineering applications

In the field of tissue engineering, the use of core-shell fibers represents an advantageous approach to protect and finely tune the release of bioactive compounds with the aim to regulate their efficacy. In this work, core-shell electrospun polycaprolactone nanofiber-based membranes, loaded with rifampicin and coated with silver nanoparticles, were developed and characterized. The membranes are composed by randomly oriented nanofibers with a homogeneous diameter, as demonstrated by scanning electron microscopy (SEM). An air-plasma treatment was applied to increase the hydrophilicity of the membranes as confirmed by contact angle measurements. The rifampicin release from untreated and air-plasma treated membranes, evaluated by UV spectrophotometry, displayed a similar and constant over-time release profile, demonstrating that the air-plasma treatment does not degrade the rifampicin, loaded in the core region of the nanofibers. The presence and the distribution of silver nanoparticles on the nanofiber surface were investigated by SEM and Energy Dispersive Spectroscopy. Moreover, SEM imaging demonstrated that the produced membranes possess a good stability over time, in terms of structure maintenance. The developed membranes showed a good biocompatibility towards murine fibroblasts, human osteosarcoma cells and urotheliocytes, reveling the absence of cytotoxic effects. Moreover, doble-functionalized membranes inhibit the growth of E. coli and S. aureus. Thanks to the possibilities offered by the coaxial electrospinning, the membranes here proposed are promising for several tissue engineering applications.

Microencapsulation of Betalains Extracted from Garambullo (Myrtillocactus geometrizans) to Produce Active Chitosan-Polyvinyl Alcohol Films with Delayed Release of Bioactive Compounds.

Garambullo is a plant with little industrial application. However, garambullo contains betalains, photosensitive phytochemical compounds, which through microencapsulation can be used in chitosan-polyvinyl alcohol (PVOH) films for application in tomato coatings. These biopackages were characterized by physical tests, water vapor permeability, puncture tests, extension, color, differential scanning calorimetry (DCS), Fourier transform infrared (FTIR) spectroscopy, and antioxidant and antimicrobial activity analyses. The influence of the biopackages on the tomato coatings was measured using parameters such as minimum weight loss close to 2% at day 9, pH of 4.6, Brix of 5.5, titratable acidity of 1 g acid/100 mL sample, and shelf life of up to 18 days. The biopackages containing betalain microcapsules had a water vapor permeability of 2 × 10-14 g/h·m·Pa and an elongation of 5 ± 0.5%, indicating that the package did not stretch. The deformation at the breaking point for the package without and with microcapsules was 0.569 and 1.620, respectively. With respect to color, adding white microcapsules and betalains can cause the material to darken, resulting in a yellowish color. Furthermore, the phenolic content was greater for the biopackages with betalains, while there was no significant difference in the antioxidant activity since the active compounds were not released. According to the in vitro results, the inhibition of B. cinerea was achieved on the eighth day when the active compounds were released from the microcapsules. The tomato with betalains lost 2% of its weight, and B. cinerea was inhibited, extending its shelf life to 18 days. The proposed biopackages have good properties as biopolymers and inhibit the presence of B. cinerea.

Microwave pre-treatment of niger (Guizotia abyssinica) seeds: Impact on stability, physicochemical characteristics, bioactive and fatty acid profiles of extracted oil

The study explored the impact of microwave pre-treatment (MPT) at different powers (360, 540 and 720 W) and exposure times (2, 4, 6 and 8 min) on the qualitative characteristics of niger seed oil (NgO). The changes in physicochemical characteristics, oxidative stability index (OSI), phenolic constituents, Maillard products, tocopherols, pigments and phytosterols in NgO were elucidated after MPT of seeds. The NgO yield increased from 33.19 % to 38.76 % and OSI augmented from 2.69 to 3.93 h upon MPT at 540 W and 720 W for 8 min, respectively. The tocopherols and phytosterols levels augmented upon MPT at 540 W for 4 and 6 min, respectively while declined by further elevating power (720 W) and exposure time (8 min). The total hydroxycinnamic (27.41 mg/kg) and hydroxybenzoic (1.48 mg/kg) acids and total flavonoids (115.83 mg/kg) were highest upon MPT at 720 W for 6 min while viscosity (56.71 mPa.s), chlorophylls (0.12 mg/kg), carotenoids (0.85 mg/kg) and 5-hydroxymethylfurfural (1.38 mg/kg) were highest upon MPT at 720 W for 8 min. In conclusion, the MPT promoted the release of bioactive compounds and lowered the generation of harmful oxidative by-products. The results highlight the potential of MPT in enhancing quality attributes of NgO for use in the food, cosmetics, and pharmaceutical industries.

Modulating the Release Kinetics of Natural Product Actinomycin from Bacterial Nanocellulose Films and Their Antimicrobial Activity.

The present study aimed to create a more sustainable and controlled delivery system based on natural biopolymer bacterial nanocellulose (BNC) and bacterial natural product actinomycin (Act), with the applicative potential in the biomedical field. In order to provide improved interaction between BNC and the active compound, and thus to modulate the release kinetics, the TEMPO oxidation of BNC support was carried out. A mix of actinomycins from bacterial fermentation (ActX) were used as natural antimicrobial agents with an established bioactivity profile and clinical use. BNC and TEMPO-oxidized BNC films with incorporated active compounds were obtained and analyzed by FTIR, SEM, XPS, and XRD. The ActX release profiles were determined in phosphate-buffer solution, PBS, at 37 °C over time. FTIR analysis confirmed the improved incorporation and efficiency of ActX adsorption on oxidized BNC due to the availability of more active sites provided by oxidation. SEM analysis indicated the incorporation of ActX into the less-dense morphology of the TEMPO-oxidized BNC in comparison to pure BNC. The release kinetics of ActX were significantly affected by the BNC structure, and the activated BNC sample indicated the sustained release of active compounds over time, corresponding to the Fickian diffusion mechanism. Antimicrobial tests using Staphylococcus aureus NCTC 6571 confirmed the potency of this BNC-based system for biomedical applications, taking advantage of the capacity of modified BNC to control and modulate the release of bioactive compounds.

In Vitro Antioxidant Activity of Liposomal Formulations of Sea Buckthorn and Grape Pomace.

This study evaluated the impact of the encapsulation of sea buckthorn and grape pomace extracts in liposomal formulations on the retention and release of bioactive compounds and their antioxidant activity. The profile and composition of lipophilic extracts of sea buckthorn and hydrophilic extracts of grape pomace were analyzed. Encapsulation efficiency, retention rate, and the content of bioactive compounds encapsulated in liposomal formulations prepared in two media-water and ethanol-were evaluated. The encapsulation efficiency varied between 84 and 90%, indicating the superior encapsulation of the bioactive compounds. The retention rate varied between 79 and 86%, which indicated the stability of the liposome-encapsulated compounds over time. The antioxidant activity of the encapsulated samples was determined in vitro, under the conditions of gastric (pH 1.8) and intestinal (pH 8.2) digestion, in relation to the non-encapsulated extracts. The antioxidant activity of both liposomal formulations was higher than that of the nonencapsulated extracts during gastric digestion. Moreover, an increase over time in the antioxidant activity, expressed as % DPPH inhibition, was observed for all samples, with around 90% DPPH inhibition for non-encapsulated extracts and 92% for the encapsulated extracts, demonstrating the stability of bioactive compounds in acidic pH. Oppositely, when exposed to intestinal simulated digestion (alkaline pH), the antioxidant activity decreased over time to around 24% DPPH inhibition for both encapsulated and nonencapsulated extracts. These results provide a foundation for the further development and application of liposomal delivery systems in functional foods.

Impact of ohmic heating extraction on the bioactive components of parsley: comparison with conventional and green extraction techniques

The extraction process is an important step that is widely used in food, agriculture, cosmetics, pharmaceuticals, modern and alternative medicine, etc. Because of plants contain low amount of beneficial phytochemicals, obtaining high amounts of the desired compound is important in terms of extraction efficiency. In recent years, modern and green technologies have become more important; they also bring advantages such as high selectivity, less solvent, time, and energy requirements. Apigenin (4′, 5, 7-trihydroxyflavone), a naturally occurring flavonoid, possesses a variety of pharmaceutical properties such as anticancer, anti-inflammatory, and antioxidant properties which are very important for human health. Parsley is one of the most common herbs used in cuisines and is known as the best source of apigenin. Therefore, this study aimed to investigate the effects of various extraction methods on the extraction of biologically active compounds from parsley leaves. Conventional (maceration), ultrasound-assisted extraction (UAE), and ohmic heating extraction (OHE) methods were compared in terms of yield, total phenolic content (TPC), total flavonoid content (TFC), antioxidant activities, and apigenin quantities. The results showed that the UAE and OHE techniques had higher extraction efficiency than the maceration method. UAE achieved satisfactory extraction efficiency, shorter processing time, high energy savings, and retention of heat-labile compounds. OHE, as an advanced heating technology, increased the permeability of cell membranes, facilitating the release of bioactive compounds. The results revealed that the OHE technique was the most effective method for apigenin extraction (159.63 ppb) and antioxidant activity.

Evaluation of functional extrudates enriched with essential oils for enhanced stability

This study evaluates the performance and storage stability of functional extrudates enriched with natural additives, utilizing advanced food engineering and processing techniques. Essential oils from oregano, rosemary, hypericum, and chamomile were incorporated in both free and encapsulated forms using electrohydrodynamic processing, spray drying, and freeze drying, and added to the food matrix through extrusion processing. The encapsulated additives demonstrated superior retention and controlled release of bioactive compounds, with spray-dried agents achieving the highest total phenolic content (1.66 mg GAE/g) post-extrusion. During storage, these extrudates exhibited enhanced stability with low degradation rate constants (1.90×10⁻³ days⁻¹ at aw=0.33, 25°C, over 240 days). The bioactive extrudates maintained remarkable stability in terms of hardness and colour change, with hardness inversely correlated to moisture content. These findings highlight the potential of using encapsulation and extrusion processing to create stable, naturally bioactive food products, reducing dependence on synthetic additives. The research underscores the integration of emerging technologies in food science to enhance the stability and delivery of essential oils, supporting the development of sustainable and health-promoting food systems.

Preparation and characterization of hydrophobically-modified sodium alginate derivatives as carriers for fucoxanthin

Micelles are nano-architectured structures capable of encapsulating hydrophobic substances in aqueous solutions, thereby improving their stability, solubility, and bioavailability to control the release of bioactive compounds. In this study, the sodium alginate backbone was modified via a hydrophobic modification scheme where octyl chains were covalently attached to the alginate chains via esterification reactions. Fourier-transformed infrared spectrometry (FTIR), 1H nuclear magnetic resonance (1H NMR), X-ray diffraction (XRD), and thermal gravimetric analysis (TGA) were used to characterize the structure of the sodium alginate derivatives of varying molar mass (ALG-C8). Fluorescence spectroscopy, surface tension measurements, and dynamic light scattering (DLS) were employed to assess the self-assembly performance of ALG-C8. The three methods produced consistent results, indicating that self-assembly decreased with higher molar mass. The self-assembled ALG-C8 micelles were utilized to encapsulate fucoxanthin. The loading capacity (LC) and encapsulation efficiency (EE) were determined using UV–Vis spectrophotometry. The molar mass of ALG-C8 has a key influence on fucoxanthin loading and release behavior. The ALG-C8 molecules with the lowest molar mass produced micelles with the smallest hydration diameter, resulting in the highest LC and EE. Furthermore, the release of fucoxanthin is significantly influenced by the pH and ionic strength of the medium. ALG-C8 micellar-like aggregates exhibit resistance to low pH and high ionic strength environments, releasing encapsulated material at the target site under alkaline conditions. Therefore, synthesized ALG-C8 is a potential candidate for preparing pH-responsive self-assembled micellar-like aggregates, enabling targeted delivery and gradual release of hydrophobic functional food components.

Leguminous ferritin, a natural protein for iron supplementation, Pickering emulsion formation and encapsulation of bioactive molecules

AbstractFerritin is a naturally occurring iron storage protein. Leguminous ferritins exhibit unique structural features, including diverse subunit composition and an extension peptide, which contribute to superior thermal stability compared to animal ferritins. The high iron content, remarkable effectiveness, low risk of oxidative damage and thermal stability make the leguminous ferritin an attractive candidate for iron supplementation. Moreover, apoferritin is an excellent nanosized carrier for encapsulating bioactive compounds due to its inherent inner cavity, water solubility, biocompatibility, and reversible self‐assembly behavior. However, the harsh condition during encapsulation by unmodified ferritins may cause damage to sensitive bioactive compounds. Thus, different processing methods are employed to alter the leguminous ferritin structures, including chemical, enzymatic, mild heat treatments, and nonthermal processing to achieve gentler encapsulation conditions for a wide range of bioactive compounds. Another challenge is to improve the stability of leguminous ferritin to withstand gastric digestion. The degradation of ferritin by proteases may lead to premature release of bioactive compounds. Recent works demonstrated that certain phenolic compounds such as proanthocyanidin‐induced protein association, thereby enhancing digestive stability of ferritins, leading to a sustained release and a potentially greater bioavailability of bioactive compounds. Leguminous ferritin also has the potential to serve as a stabilizer for the Pickering emulsion, where the hydrophilic and hydrophobic compounds can be encapsulated in the ferritin nanocages and oil phase, respectively. The release and absorption of bioactive compounds in encapsulates and emulsions will need to be further demonstrated through in vivo studies.

Evaluation of Solid-State Fermentation Conditions from Pineapple Peel Waste for Release of Bioactive Compounds by Aspergillus niger spp.

Currently, agroindustrial waste can be used to obtain bioactive compounds. The solid-state fermentation is an alternative for the valorization of these waste and to be able to release bioactive compounds that may be of interest to different industrial sectors. The aim of this study was to evaluate solid-state fermentation conditions using pineapple peel waste as the substrate with Aspergillus niger spp., to release bioactive compounds using a Plackett–Burman exploratory design. Temperature, humidity, inoculum, NaNO3, MgSO4, KCl, and KH2PO4 conditions in the fermentation process were evaluated. The antioxidant capacity was determined, and the main compounds of the fermentation extracts were identified. The results revealed that the Aspergillus niger HT3 strain reached a hydrolyzable tannin release of 10.00 mg/g, While Aspergillus niger Aa20 reached a condensed tannin release of 82.59 mg/g. The KH2PO4 affects the release of condensed tannins with A. niger Aa20, and MgSO4 affects the release of hydrolyzable tannins with A. niger HT3. In addition, a positive antioxidant activity was demonstrated for the DPPH, ABTS, and FRAP technique. The main compounds in the fermented pineapple peel were 3-feruloylquinic acid, caffeic acid, lariciresinol, and 3-hydroxyphloretin 2′-O-xylosyl-glucoside, among others. The solid-state fermentation process is a biotechnological alternative for the release of bioactive compounds.

Development and evaluation of guar gum-coated nano-nutriosomes for cyanidin-3-O-glucoside encapsulation

Cyanidin-3-O-glucoside (C3G) is a type of water-soluble flavonoid compound that is abundantly found in fruits and vegetables. C3G possesses numerous biological activities, however, it is prone to breakdown under environmental conditions. To overcome these issues, we developed nano-nutriosome (NS) carriers created by vortex-mixing and probe-sonication techniques for C3G encapsulation in which the phospholipid and Nutriose® FB06 were chosen as carrier material, and guar gum (GG) as a coating material to formulate a unilamellar and multicompartment structure. This study aimed to develop and evaluate C3G-loaded nano-nutriosomes coated by GG (GG-C3G-NS) for improving physicochemical stability, antioxidant activity, cellular uptake, and controlled release properties. The C3G-NS and GG-C3G-NS are nanosized (143.47 to 154.13 nm), with high encapsulation efficiency (>93.31 %). The NS carriers successfully encapsulated C3G which was confirmed by transmission electron microscopy, differential scanning calorimetry, and Fourier transform infrared spectroscopy. C3G showed more stability in storage, thermal, pH, ionic, and oxidative conditions. Furthermore, the NS exhibited a better-controlled release of C3G in different food stimulant conditions and in vitro release study. Additionally, NS systems enhanced cellular uptake and showed no cytotoxicity. Overall, GG-NS could be a promising nanocarrier for improving the stability, controlled release, and antioxidant activity of bioactive compounds.

Antibacterial nanofibrous film encapsulated with 4-terpineol/β-cyclodextrin inclusion complexes: Relative humidity-triggered release and shrimps preservation application

Antibacterial active packaging enables extensive biological effects to improve food safety. However, it still remains difficult to achieve controlled and precise release of bioactive compounds to the specific target location with required quantity in food packaging applications. Here, a novel relative humidity (RH) - responsive polyvinyl alcohol/chitosan (PVA/CS) composite nanofibrous film incorporated with 4-terpineol/β-cyclodextrin inclusion complexes (4-TA@β-CD ICs) was engineered by electrospinning that can be exploited as a functional packaging materials. The characterization results showed the thermal stability of the films was enhanced after the incorporation due to the hydrogen bonds between ICs and biopolymers. Besides, the 4 wt% 4-TA@β-CD ICs/PVA/CS film exhibited optimized morphology, crystallinity, mechanical properties, and water vapor permeability. The incorporation of ICs dramatically promoted the antioxidant and inhibition activity of the film especially towards Shewanella putrefaciens. Moreover, the release behavior of 4-TA indicated that high RH accelerated the releasing rate, and the film obtained a sustained-release effect. Finally, the film with long-term antibacterial effectiveness succeeded in extending the shelf life of Litopenaeus vannamei shrimps by an additional 2 days at 4 °C. This work developed a RH-responsive release packaging material, which may serve as an effective active food packaging.

An effective potential Bifidobacterium animalis F1-7 delivery strategy: Supramolecular hydrogel - sodium alginate/tryptophan-Sulfobutylether-β-cyclodextrin (Alg/Trp-SBE-β-CD)

Sodium alginate exhibits poor stability when applied as a single gel network, which undermines its efficacy in probiotic encapsulation and protection. This study aimed to incorporate alginate (Alg) with cyclodextrin to prepare a supramolecular hydrogel to enhance the encapsulation and viability of probiotics. Specifically, l-tryptophan (L-Trp) and Sulfobutylether-β-cyclodextrin (SBE-β-CD) were employed to self-assemble into an inclusion complex (IC). Alg/Trp-SBE-β-CD hydrogels were formed using calcium ion-mediated cross-linking and IC addition. Experimental data revealed that the Alg/Trp-SBE-β-CD gels with 1 wt% CaCl2 exhibited good overall performance. The hydrogels significantly improved the stability and release properties of loaded Bifidobacterium animalis F1-7 compared to single Alg gels. Notably, the survival rate was 95.5% after 2 h in simulated gastric fluid (SGF), and the release rate reached 97.7% after continuous release in simulated intestinal fluid (SIF) for 6 h. Probiotic viable counts were maintained at 7–8 log CFU/g after 4 weeks of storage. This study indicates that a hybrid wall material of Alg and Trp-SBE-β-CD IC can enhance the stability and controlled release of bioactive compounds including synbiotics (prebiotics + probiotic preparations).

Insights into multi-component digestion and antioxidant release from plant protein-seaweed-based Maillard conjugate-MRP systems

Maillard conjugation to modify protein functionality results in the formation of a protein-polysaccharide complex. The resultant conjugate mixture is rich in different Maillard reaction products (MRP) and unreacted components. The investigation pertaining to the digestion and intestinal fate of these components is challenging due to the multitude of effects and interactions between different components. This study attempts to understand gastrointestinal behaviour and diffusion/release patterns of different components from a complex system made of the amaranth-Gracilaria seaweed system. Data acquired from in-vitro INFOGEST protocol and intestinal dialysis absorption model were used to investigate the amino acid release and antioxidant diffusion during different phases of digestion. The analysis suggests a significant improvement in the radical scavenging capacity of overall antioxidants released from the conjugate-MRP system compared to that of amaranth protein alone based on different antioxidant assays (DPPH, FRAP, ICred) during each stage of digestion. The permeability and overall intestinal absorption ratio of antioxidants through a dialysis membrane shows an improvement of up to 55 % compared to that of the plant protein. Moreover, amino acid and phyto-compounds release patterns of the conjugate-MRP system exhibit selective shielding effects towards the gastric phase, which can potentially be investigated for targeted release of bioactive compounds.
