Umbelliprenin attenuates comorbid behavioral disorders in acetic acid-induced colitis in mice: mechanistic insights into hippocampal oxidative stress and neuroinflammation.

Inflammatory bowel disease (IBD) is often accompanied by psychiatric disorders. Emerging evidence suggests that neuroinflammation and oxidative stress contribute to the psychiatric symptoms associated with IBD. Umbelliprenin (UMB) possesses several pharmacological properties, including anti-inflammatory and antioxidant effects. This study aimed to investigate the protective effects of UMB on comorbid behavioral disorders in a mouse model of experimental colitis, focusing on its potential anti-neuroinflammatory and antioxidant activities. After inducing colitis with acetic acid, male NMRI mice were treated for 7 consecutive days with UMB, saline, or dexamethasone. Behavioral assessments included the forced swimming test (FST), splash test, open field test (OFT), and elevated plus maze (EPM). Histopathological changes in the colon were evaluated, and total antioxidant capacity (TAC), malondialdehyde (MDA) levels, and the expression of inflammatory genes (TNFα, IL1β, and TLR4) were measured in the hippocampus. Colitis was associated with increased immobility time in the FST, reduced entries and time spent in the open arms of the EPM, decreased grooming behavior in the splash test, and reduced time spent in the central zone of the OFT. Colitis also resulted in a reduction in TAC and an increase in MDA levels and inflammatory gene expression in the hippocampus. UMB treatment mitigated the behavioral disorders associated with colitis, reduced neuroinflammation and oxidative stress in the hippocampus, and alleviated histopathological alterations in the colon. In conclusion, UMB may reduce behavioral disorders induced by colitis by decreasing oxidative stress and neuroinflammation in the hippocampus.

Anticancer Action of Silver Nanoparticles in SKBR3 Breast Cancer Cells through Promotion of Oxidative Stress and Apoptosis.

Silver nanoparticles (AgNPs) are known as one of the highly utilized NPs owing to their unique characteristics in the field of cancer research. The goal of this research was to explore the oxidative stress, apoptosis, and angiogenesis in SKBR3 breast cancer cells after exposure to AgNPs. The survival rate of SKBR3 cancer cells and MCF-10A normal breast cells was assessed under the effects of different concentrations (0, 32, 64, 128, and 250 μg/ml) by MTT method. The oxidative condition was assessed by measuring reactive oxygen species (ROS) production, total oxidant status (TOS), total antioxidant capacity (TAC), malondialdehyde (MDA), and antioxidant enzyme activity (CAT, GPx, and CAT) using colorimetric-based kits. Flow cytometry and Hoechst 33258 staining were performed to investigate the induction of apoptosis. Furthermore, the expression of Bcl-2-associated X protein (Bax), B-cell lymphoma 2 (Bcl-2), and caspase 3 and 7 activity was measured. The cell migration and vascular endothelial growth factor-A (VEGF-A) gene expression, protein kinase B (AKT), phosphatidylinositol 3-kinase (PI3K) were also studied. The MTT results indicated that AgNPs inhibit the SKBR3 cells' viability in a concentration-dependent way. Besides, AgNPs markedly induced oxidative stress via increasing TOS content, MDA production, reduction of TAC, and regulation of antioxidant enzyme level. Additionally, AgNPs promoted apoptosis as revealed by an enhancement in Bax/Bcl-2 expression ratio. Findings also indicated that AgNPs suppress the expression of genes (VEGF-A, AKT, and PI3K) involved in angiogenesis. Altogether, our data revealed that AgNPs initiate oxidative stress and apoptosis in SKBR3 breast cancer cells, dose dependently.

The effect of melatonin on capecitabine-induced hepatic and renal toxicity in rats.

Capecitabine (CAPE), an antimetabolite chemotherapy, can induce hepatic and renal toxicity. Melatonin (MEL), a neurohormone, possesses antioxidant, anti-apoptotic and anti-inflammatory effects. This study investigated the impact of MEL on capecitabine-induced hepatic and renal toxicity. Twenty-five male Wistar rats were categorized into five groups for the study. The groups included a control group, MEL10 group (rats receiving daily intraperitoneal injections of 5mg/kg MEL), CAPE 500 group (rats receiving weekly intraperitoneal injections of 500mg/kg CAPE), CAPE + MEL five group, and CAPE + MEL 10 group. All groups were treated for a duration of 6 weeks. Various hematological, serological, biochemical, and histopathological assessments were conducted to evaluate the objective of the study. The administration of CAPE led to significant liver and kidney toxicity, as evidenced by elevated levels of malondialdehyde (MDA), myeloperoxidase (MPO), nitric oxide (NO), as well as serological markers including AST, ALT, ALP, BUN, and creatinine. CAPE exposure also resulted in a reduction in total antioxidant capacity (TAC) and glutathione peroxidase (GPx) levels. Histological examination revealed hyperemia in both liver and kidney tissues exposed to CAPE. However, treatment with MEL demonstrated positive effects. MEL administration alleviated oxidative stress, reduced levels of liver enzymes, BUN, and creatinine, and ameliorated histopathological degenerations. MEL also increased GPx and TAC levels. Moreover, MEL treatment aided in restoring the body weight that was lost due to CAPE exposure. Our findings indicated that the administration of MEL in rats significantly enhanced the hepatic and renal toxicity induced by CAPE.

Increased Total Antioxidant Capacity Value Improves Survival of Fat Grafts in Rat Model.

Although studies aiming to increase fat graft survival continue, in this study, the authors aimed to investigate the effects of different antioxidants on total antioxidant capacity (TAC) and their effect on graft survival. Thirty-two male Wistar rats were divided into four equally sized groups, including a control group and three antioxidant groups receiving either melatonin (10 mg/kg), zinc (2 mg/kg), or vitamins E and C (100 mg/kg). Autologous fat grafts (1.7 ± 0.4 g) were transferred to the dorsal subcutaneous region, and total antioxidant capacity was measured on days 0 and 1, week 1, and monthly until the third month. Transferred graft volume and mass (1.3 ± 0.4 g) were measured using the liquid overflow method and precision scales at the end of the study. Routine hematoxylin and eosin staining and immunohistochemistry against perilipin were performed for semiqualitative analysis and H-score for viable adipose cells, respectively. Collected fat grafts measured significantly less in weight and volume and the survival rate was lower in the control group ( P < 0.01). The control group exhibited a reduction in TAC, whereas all groups receiving antioxidants had an increase in TAC during the first week ( P = 0.02, P = 0.008, and P = 0.004 for melatonin, zinc, and vitamins, respectively). Immunohistochemistry of the antioxidant group demonstrated a statistically significant excess and reactivity of cells staining with perilipin antibodies. In this animal study, the beneficial effect of antioxidants on fat graft survival can be related to the significant increase in TAC following the first week of their administration. Antioxidants improve fat graft survival and quality in this rat model, by increasing total antioxidant capacity in the first week of administration.

STUDY SOME PHYSIOLOGICAL ASPECTS AND HISTOPATHOLOGICAL CHANGES IN MALE RABBITS INTOXICATED BY CHLORPYRIFOS

The current experience on chlorpyrifos was undertaken to investigate the potential alterations in Physiological Criteria and in liver and kidney tissues subsequent oral exposure of chlorpyrifos in male rabbits . twelve of male rabbits were employed in this experience, distributed into two groups randomly , group one received only corn oil and served as control, whereas, while group two received CPF 33.3 mg/kg dissolved with( 1ml ) corn oil daily, the exposure continued for 28 consecutive days orally. outcomes of this work revealed that sub-acute oral administration of chlorpyrifos cause to oxidative stress evidenced by pronounced elevation value of oxidative biomarker malondialdehyde ( MDA) concomitantly with elevated in values of inflammatory biomarkers ( TNF-α and CRP) and significantly reduction in total antioxidant capacity (TAC) value, as well as this findings exhibited that chlorpyrifos administration cause to marked alteration of blood indices values as well as histopathological changes were observed in liver and kidney which as represented degenerative of hepatocyte, dilatation and congestion in central vein, While tubular epithelial degenerations, vacuolations and severe dilation in renal cortex tubules. Conclusion: we concluded from findings in this study the oral exposure to CPF evoked inflammation response and oxidative damage as represented by pronounced elevation in levels of CRP,TNF-alpha and MDA biomarkers as well as pathohistological alterations were observably in liver and kidney that indicating that chlorpyrifos elicits numerous adverse effects noteworthy making it a major public health issue.

Effects of sesamin on Aβ1-42-induced oxidative stress and LTP impairment in a rat model of Alzheimer's disease.

The present study examined the protective effect of sesamin (Ses) on β-amyloid (Aβ)-induced long-term potentiation (LTP) impairment at the PP-DG synapses in male rats. Wistar rats were randomly assigned to seven groups: control, sham, Aβ; ICV Aβ1-42 microinjection, Ses, Aβ + Ses; first, ICV Aβ injections and then receiving Ses, Ses + Aβ: four weeks of pretreatment with Ses and then Aβ injection, and Ses + Aβ + Ses: pre (four weeks) and post (four weeks) treatment with Ses. Ses-treated groups received 30mg/kg of Ses once a day by oral gavage for four weeks. After the treatment period, the animals were positioned in a stereotaxic device for surgery and field potential recording. The population spike (PS) amplitude and slope of excitatory postsynaptic potentials (EPSP) were evaluated in the DG region. Serum oxidative stress biomarkers (total oxidant status (TOS) and total antioxidant capacity (TAC)) were measured. Aβ impaired LTP induction at the PP-DG synapses evidenced by a decrease in EPSP slope and PS amplitude of LTP. In Aβ rats, Ses increased EPSP slope and PS amplitude of LTP in the DG granular cells. Also, an increase in TOS and a reduction in TAC caused by Aβ were significantly corrected by Ses. Ses could prevent Aβ-induced LTP impairment at the PP-DG synapses in male rats, which can be due to its preventive effects on oxidative stress.

Nicorandil and Bone Marrow-derived Mesenchymal Stem Cells Therapeutic Effect after Ureteral Obstruction in Adult Male Albino Rats.

Chronic kidney disease is a global health problem for which renal fibrogenesis is the final treatment target. In our work, we have highlighted two new strategies, nicorandil and Bone marrow-derived mesenchymal stem cells (BM-MSCs), as effective in reversing renal fibrosis induced by partial unilateral ureteral obstruction (PUUO). The current study included 96 male albino rats randomly divided into four groups, with 24 rats per group; Group I, the control group; Group II, PUUO, where two-thirds of the left ureter was entrenched in the psoas muscle; Group III, same surgical procedure as in Group II for 7 days, and then the rats received 15 mg/kg/day nicorandil once daily for 21 days; and Group IV, same surgical procedure as in Group II for 7 days, and then rats were given 3 × 106 of labeled MSCs injected intravenous, and left for 21 days. Blood and kidney tissues were collected for biochemical, histological, and molecular analyses. Both the nicorandil and BM-MSCs treatment groups could ameliorate kidney damage evidenced by inhibition of MDA elevation and total antioxidant capacity reduction caused by PUUO. Also, there was a significant reduction observed in TNF, TGF, IL6, collagen I, and α-SMA in addition to improvement in histological examination. However, a significant difference was found between the BM-MSCs and nicorandil-treated groups. Our results suggest that BM-MSCs and nicorandil improved renal fibrosis progression through their antiapoptotic, anti-inflammatory, and antifibrotic effects in male albino rats subjected to PUUO, with BM-MSCs being more effective compared to nicorandil.

Effects of adenosine and dipyridamole on serum levels of IL-6, TNF - , and some antioxidants in rabbits

Nearly 30 years of research findings stated that adenosine adjust ischemia. Although, adenosine use for treatment has not been accepted clinically in wide manner, also there have no many clinical trials. Aims: study the effects of adenosine and dipyridamole on total Antioxidant Capacity (TAC), Adenosine Deaminase (ADA), GSH (Glutathione), TNF - a (tumor necrosis factor Alpha) and IL-6 (Interleukin 6) in rabbits. Material and methods: Thirty five male rabbits were included in the study. The animals were divided into 3 groups: Group one(5 animals): injected (i.p) with 2 ml of distilled water/day (control group). Group two (15 animals): were treated by intraperitoneal injection of adenosine, they were divided into 3 sub groups (5 animals) according to adenosine dose:1 mg/kg, 2mg/kg and 4 mg/kg.Group 3 (15 animals): were treated by dipyridamole orally, they were divided into 3 sub groups (5 animals) according to dipyridamole dose:4 mg/kg, 8 mg/kg and 12 mg/kg. Total Antioxidant Capacity Assay kit, Adenosine Deaminase (ADA) colorimetric Assay kit, Rabbit IL-6 (Interleukin 6) ELIISA kit, Rabbit TNF - a (tumor necrosis factor Alpha), GSH (Glutathione) ELISA kits were used. Results: ANOVA Test and Duncan’s Multiple Range Test were applied between study groups. In comparison among adenosine, dipyridamole and control groups, highly significant differences were found among them in TAC,ADA and GSH levels while no significant differences were found in TNF -a and IL-6 levels among all groups. Conclusion: both adenosine, dipyridamole cause reduction in total Antioxidant Capacity together with increase in glutathione levels and Adenosine Deaminase.

Does age pay off? Effects of three-generational experiments of nanodiamond exposure and withdrawal in wild and longevity-selected model animals.

Nanodiamonds (NDs) are considered a material with low toxicity. However, no studies describe the effects of ND withdrawal after multigenerational exposure. The aim was to evaluate ND exposure (in the 1st and 2nd generations) effects at low concentrations (0.2 or 2mgkg-1) and withdrawal (in the 3rd generation) in the wild (H) and longevity-selected (D) model insect Acheta domesticus. We measured selected oxidative stress parameters, immunity, types of cell death, and DNA damage. Most of the results obtained in the 1st generation, e.g., catalase (CAT), total antioxidant capacity (TAC), heat shock proteins (HSP70), defensins, or apoptosis level, confirmed no significant toxicity of low doses of NDs. Interestingly, strain-specific differences were observed. D-strain crickets reduced autophagy, the number of ROS+cells, and DNA damage. The effect can be a symptom of mobilization of the organism and stimulation of physiological defense mechanisms in long-living organisms. The 2nd-generation D-strain insects fed ND-spiked food at higher concentrations manifested a reduction in CAT, TAC, early apoptosis, and DNA damage, together with an increase in HSP70 and defensins. ROS+cells and cells with reduced membrane potential and autophagy did not differ significantly from the control. H-strain insects revealed a higher number of ROS+cells and cells with reduced membrane potential, decreased CAT activity, and early apoptosis. Elimination of NDs from the diet in the 3rd generation did not cause full recovery of the measured parameters. We noticed an increase in the concentration of HSP70 and defensins (H-strain) and a decrease in apoptosis (D-strain). However, the most visible increase was a significant increase in DNA damage, especially in H-strain individuals. The results suggest prolonged adverse effects of NDs on cellular functions, reaching beyond "contact time" with these particles. Unintentional and/or uncontrolled ND pollution of the environment poses a new challenge for all organisms inhabiting it, particularly during multigenerational exposure.

Understanding the Behaviour of Human Cell Types under Simulated Microgravity Conditions: The Case of Erythrocytes.

Erythrocytes are highly specialized cells in human body, and their main function is to ensure the gas exchanges, O2 and CO2, within the body. The exposure to microgravity environment leads to several health risks such as those affecting red blood cells. In this work, we investigated the changes that occur in the structure and function of red blood cells under simulated microgravity, compared to terrestrial conditions, at different time points using biochemical and biophysical techniques. Erythrocytes exposed to simulated microgravity showed morphological changes, a constant increase in reactive oxygen species (ROS), a significant reduction in total antioxidant capacity (TAC), a remarkable and constant decrease in total glutathione (GSH) concentration, and an augmentation in malondialdehyde (MDA) at increasing times. Moreover, experiments were performed to evaluate the lipid profile of erythrocyte membranes which showed an upregulation in the following membrane phosphocholines (PC): PC16:0_16:0, PC 33:5, PC18:2_18:2, PC 15:1_20:4 and SM d42:1. Thus, remarkable changes in erythrocyte cytoskeletal architecture and membrane stiffness due to oxidative damage have been found under microgravity conditions, in addition to factors that contribute to the plasticity of the red blood cells (RBCs) including shape, size, cell viscosity and membrane rigidity. This study represents our first investigation into the effects of microgravity on erythrocytes and will be followed by other experiments towards understanding the behaviour of different human cell types in microgravity.

Citrullus colocynthis Seed Ameliorates Layer Performance and Immune Response under Acute Oxidative Stress Induced by Paraquat Injection.

Colocynth, i.e., Citrullus colocynthis, is an herbaceous plant known to have antioxidant properties. Employing laying hens, this study investigated the potency of dietary colocynth seed supplementation to reduce the deleterious effects of acute oxidative stress induced by paraquat injection. The results demonstrated that supplementing layers’ diets with colocynth seed at 0.1% alleviated oxidative stress responses and significantly improved egg production performance. Furthermore, the immunological responses of the acute-oxidative-stressed layers were enhanced with colocynth seed supplementation. Thus, the inclusion of colocynth seed in layer chickens’ diets can improve egg production performance, restore the redox balance, and enhance immunological responses when they are reared under acute oxidative stress conditions.Oxidative stress is a detrimental physiological state that threatens birds’ productivity and general health. Colocynth is an herbal plant known for its bioactive properties, and it is mainly known for its antioxidant effects. This study’s purpose was to investigate how effective colocynth seed is at lowering the detrimental impact of acute oxidative stress caused by paraquat (PQ) injection in laying hens. A total of 360 Hy-Line Brown chickens, aged 39 weeks, were gathered and divided into four equal groups (10 hens × 9 replicates) in a 2 × 2 factorial design. The experimental groups were given either a basal diet or the basal diet supplemented with colocynth seed (1% of diet). Starting from week 40 of age and for 7 successive days, the experimental groups were either injected daily with paraquat (5 mg/kg body weight) or with saline (0.5 mL, 0.9% NaCl). Egg production performance with selected stress biomarkers and immunological response parameters were investigated at the end of week 40 of age. Our data revealed a significant reduction in egg production with an increase in blood stress biomarkers (i.e., HSP-70, corticosterone, and H/L ratio) in PQ-injected groups compared with non-stressed groups. Furthermore, an unbalanced redox state was detected in acute oxidative stress groups, with a significant rise in lipid peroxidation level, a reduction in total antioxidant capacity (TAC), and a drop in superoxide dismutase (SOD) and catalase enzyme activity. Supplementing PQ-injected hens with colocynth seed reduced the deleterious effects of acute oxidative stress. There was a significant drop in stress biomarkers with a significant rise in antioxidant enzyme activity and TAC observed in the PQ-injected group provided with colocynth seed supplementation. Remarkably, supplementation of colocynth in the non-stressed group resulted in a significant 27% increase in TAC concentration and 17% higher SOD activity when compared with the non-stressed control group. Colocynth supplementation in the PQ-injected group elevated the total white blood cell count by 25% and improved the B-lymphocyte proliferation index (a 1.3-fold increase) compared with the PQ-injected group that did not receive supplementation. Moreover, the non-stressed colocynth-supplemented group had significantly higher cell-mediated and humoral immune responses than the non-stressed control group. This study demonstrated that colocynth seed supplementation in birds exposed to acute oxidative stress may effectively alleviate its negative impacts on production performance, immunological responses, and redox status. We also inferred that, under normal conditions, colocynth seed can be added to laying hens’ diets to stimulate production and ameliorate immune responses.

Mortality, energy reserves, and oxidative stress responses of three native freshwater mussels to temperature as an indicator of potential impacts of climate change: A laboratory experimental approach.

Due to rapid global climate change, the temperature of streams and rivers is predicted to be increased, influencing aquatic organisms, particularly bivalve species. To better understand their response to elevated temperature, three native species of freshwater mussels namely, Coelatura aegyptiaca, Mutela rostrata and Chambardia rubens were exposed for 21 days to four temperature treatments (18 - 25-30 and 35 °C). Mortality was recorded on a daily basis over the experiment. A variety of responses including biomarkers at the cellular level and whole organism level were examined in surviving organisms after 21 days. In response to exposure to thermal stress, three mussel species exposed to 35 °C showed 100% mortality after 21 days. However, exposure to 25 °C and 30 °C induced significant decreases in adenosine triphosphate (ATP) production and energy reserves (Protein, lipid and glycogen) compared to the control group maintained at 18 °C. Temperature-induced oxidative stress, indicated by the greatest reduction in total antioxidant capacity (TAC) and elevated levels of lipid peroxidation products. As energy generation decreased and oxidative stress increased, trends were for condition indexes (CI) and survival to decrease. In comparison, C.rubens has a higher thermal tolerance than C.aegyptiaca, and M.rostrata. The responses reported herein, along with the predicted rates of climate warming, indicated that freshwater mussels in the Nile River, Egypt will be negatively impacted by climate change and may incur significant losses in the coming years.

Crayfish (Procambarus clarkii) TRPA1 is required for the defense against Aeromonas hydrophila infection under high temperature conditions and contributes to heat sensing.

Temperature is an important environmental factor influencing immune responses of crayfish. However, the mechanism underlying how temperature affects immune responses remains unclear. Here, we identified an ortholog of the transient receptor potential ankyrin subtype 1 (TRPA1), a temperature sensor of Drosophila, from Procambarus clarkii (PcTRPA1-1). Its expression was induced by high temperature and challenge with heat-killed A. hydrophila at high temperature, but not at lower temperature. PcTRPA1-1 silencing led to increased mortality of crayfish challenged with live A. hydrophila at high temperature (32°C), but had no statistically significant effect on crayfish mortality at 24°C. This suggests that PcTRPA1-1 is involved in the immune responses of crayfish at high temperature as a potential temperature sensor. Further assay exhibited that PcTRPA1-1 silencing affected immune responses of crayfish, including increase of lipid peroxidation, reduction of total antioxidant capacity, decreased phenoloxidase activity and disruption of circadian rhythm of total hemocyte count entrained by temperature cycles. PcTRPA1-1 silencing also decreased the expression of PcHSP70 and PcHSP90 which are responsive to heat stimuli and bacterial challenge. The results collectively indicate that TRPA1 contributes to heat sensing of crayfish and is required for crayfish defense against bacterial infection.

Combined effects of polystyrene microplastics and cadmium on oxidative stress, apoptosis, and GH/IGF axis in zebrafish early life stages

The toxicological interactions of microplastics (MPs) and heavy metals have been paid much attention in aquatic organism. The mechanisms are not fully clear, particularly in fish early life stages. To the end, zebrafish embryos were exposed to 500 μg/L MPs, 5 μg/L cadmium (Cd), and their combination for 30 days. Body weight, adsorption characteristics of Cd onto MPs, Cd accumulation, oxidative stress, apoptosis, and growth hormone/insulin-like growth factor-I (GH/IGF) axis were examined. Exposure to MPs and Cd alone reduced body weight, which was aggravated by co-exposure. An increase in reactive oxygen species (ROS) levels was observed in larvae exposed to Cd or MPs + Cd, suggesting an induction of oxidative stress. Lipid peroxidation levels were not affected by exposure to MPs and Cd alone but dramatically enhanced by co-exposure, which may be explained by the reduction of total antioxidant capacity (TAOC) and activity levels of Mn-superoxide dismutase (Mn-SOD) and catalase (CAT) after co-exposure. Increased apoptotic cells were observed in the vertebral body of larvae exposed to Cd, the esophagus of larvae exposed to MPs, and both organs of larvae exposed to MPs + Cd, which was further confirmed by changes in the activities of Caspase-3, Caspase-8 and Caspase-9. PCR array on the transcription of genes related to growth, oxidative stress and apoptosis was examined, showing that the combined exposure resulted in greater magnitude of changes than MPs and Cd alone. The results indicate that MPs can enhance the negative effects of Cd on growth, oxidative damage and apoptosis in early life stages of zebrafish. However, the adsorption of Cd onto MPs was not observed and the combined exposure did not increase the Cd content in larvae compared to the single Cd exposure, implying that vector role of MPs in Cd uptake is negligible.

Oxidative Stress in Dairy Cows: Insights into the Mechanistic Mode of Actions and Mitigating Strategies.

This review examines several molecular mechanisms underpinning oxidative stress in ruminants and their effects on blood and milk oxidative traits. We also investigate strategies to alleviate or repair oxidative damages by improving animal immune functions using novel feed additives. Microbial pathogenic cells, feeding management, and body condition score were some of the studied factors, inducing oxidative stress in ruminants. The predominance of Streptococcus spp. (24.22%), Acinetobacter spp. (21.37%), Romboutsia spp. (4.99%), Turicibacter spp., (2.64%), Stenotrophomonas spp. (2.33%), and Enterococcus spp. (1.86%) was found in the microbiome of mastitis cows with a decrease of d-mannose and increase of xanthine:guanine ratio when Streptococcus increased. Diversity of energy sources favoring the growth of Fusobacterium make it a keystone taxon contributing to metritis. Ruminal volatile fatty acids rose with high-concentrate diets that decreased the ruminal pH, causing a lysis of rumen microbes and release of endotoxins. Moreover, lipopolysaccharide (LPS) concentration, malondialdehyde (MDA), and superoxide dismutase (SOD) activities increased in high concentrate cows accompanied by a reduction of total antioxidant capacity (T-AOC), glutathione peroxidase (GPx), and catalase (CAT) activity. In addition, albumin and paraoxonase concentrations were inversely related to oxidative stress and contributed to the protection of low-density and high-density lipoproteins against lipid peroxidation, protein carbonyl, and lactoperoxidase. High concentrate diets increased the expression of MAPK pro-inflammatory genes and decreased the expression of antioxidant genes and proteins in mammary epithelial tissues. The expression levels of NrF2, NQO1, MT1E, UGT1A1, MGST3, and MT1A were downregulated, whereas NF-kB was upregulated with a high-grain or high concentrate diet. Amino-acids, vitamins, trace elements, and plant extracts have shown promising results through enhancing immune functions and repairing damaged cells exposed to oxidative stress. Further studies comparing the long-term effect of synthetic feed additives and natural plant additives on animal health and physiology remain to be investigated.

Are there any relationships between personality type, salivary total antioxidant level and academic stress?

Stress is one of the most important determinants of total antioxidant capacity (TAC). This study aimed to assess alterations in salivary TAC following academic stress according to the personality type. This descriptive study evaluated 53 dental students at Shahid Beheshti University of Medical Sciences, Tehran, Iran, who were divided into type A (n = 25) and type B (n = 28) personality groups using the Bortner questionnaire. Saliva samples were collected during the 1st week of the semester (a low-stress period) and during the 1st week of the final exams (a high-stress period). Salivary TAC was measured using a specific kit. The data was analyzed using the repeated measures analysis of variance (ANOVA), the analysis of covariance (ANCOVA), the χ2 tests, the independent t tests, and the Bonferroni adjustments. Overall, salivary TAC in the high-stress period was significantly lower than that in the low-stress period (0.27 vs 0.31 mM) (p = 0.016). The comparison of salivary TAC between the type A and type B personality groups in the low-stress period showed no significant difference (p = 0.450). During the highstress period, a reduction in salivary TAC was recorded in both groups, which was borderline significant in the type A personality group (p = 0.050), but non-significant in the type B personality group (p = 0.140). The comparison of salivary TAC between the type A and B personality groups in the high-stress period also revealed no significant difference (p = 0.780). Academic stress can decrease salivary TAC, and the personality type has no significant effect on this relationship.

Gum Arabic Edible Coating Reduces Postharvest Decay and Alleviates Nutritional Quality Deterioration of Ponkan Fruit During Cold Storage.

The storability recession during storage limits the postharvest storage life of Ponkan (Citrus reticulata Blanco cv. Ponkan) fruit and its nutritional value, which potentially lead to huge losses. To develop an effective technique to reduce Ponkan fruit postharvest decay and to maintain the nutritional quality, the preservation effect of 9, 12, and 15% postharvest gum arabic (GA) coatings on Ponkan fruit was investigated. The 12 and 15% GA coatings effectively reduced fruit decay as well as weight loss, retained higher total soluble solids (TSS) content, suppressed titratable acidity (TA) degradation, and postponed the rise in ripening index (RI). Moreover, the 12% GA-coated fruit exhibited a lower respiration rate, electrical conductivity, and malondialdehyde (MDA) accumulation than the uncoated (control) fruit. The 12% GA coating treatment decreased the loss of ascorbic acid (AsA), total phenols, and total flavonoids and maintained higher amounts of non-enzymatic antioxidants. Furthermore, the 12% GA coating treatment increased antioxidant enzymes' activities as well as delayed the reduction of total antioxidant capacity (TAC). These results suggest that, with the cold storage increasing time, the 12% GA-coated fruit exhibited better postharvest storability and higher nutritional quality than the control fruit. The GA coating treatment could be used as a commercial wax to improve postharvest storability, extend its storage life, and maintain the nutritional value of Ponkan fruit up to 120 days of cold storage.

The Therapeutic Effect of Myrrh (Commiphora molmol) and Doxorubicin on Diethylnitrosamine Induced Hepatocarcinogenesis in Male Albino Rats

Background:This study was conducted to assess the therapeutic effect of Myrrh on Diethylnitrosamine (DEN)-induced hepatocarcinogenesis (HCC) in male albino rats. Methods:Fifty male albino rats were divided into five groups (10 rats each). Group 1 (control group) received distilled water. Group 2 (positive control) was injected intraperitoneally with DEN (55 mg/kg b.w) twice a week for two weeks, while group 3 (DOX) received doxorubicin i.p (10 mg/ kg b.w) after concomitant with DEN twice a week for four weeks. Groups 4 and 5 received a low dose of Myrrh (250 mg/kg b.w) and a high dose of Myrrh (500 mg/kg b.w) respectively daily for four weeks after the induction with DEN. The sera were used to estimate the liver enzymes (ALT, AST, and ALP), Alpha-fetoprotein (AFP), Total antioxidant capacity (TAC), and Tumor necrosis factor-ἁ (TNF-ἁ). Also, the liver tissues were collected to determine the oxidative stress markers in addition to the histopathological and immunohistochemical investigations. Results:The results showed that the induction of DEN causes a significant increase in the level of liver enzymes (ALT, AST, and ALP), AFP and TNF-ἁ as well as produce oxidative stress indicated by increasing of malondialdehyde (MDA) with the reduction in TAC and glutathione (GSH). Meanwhile, there are noticeable histopathological lesions with loss of hepatic architecture. This was accompanied by a significant increase of immunohistochemical markers; Caspase-3, vascular endothelial growth factor (VEGF), transforming growth factor β1(TGF- β1), and carcinoembryonic antigen (CEA) percentage area. The treatment of DEN rats with DOX reduced the alterations in most parameters. A marked amelioration of all parameters in a dose-dependent manner of Myrrh to the values almost near to those of the control group. Conclusion:Our data revealed that Water extract of Myrrh (C. molmol) has a potential therapeutic effect in attenuation of HCC induced DEN.

EFFECT OF OXYMATRINE ON TRICLOSAN-INDUCED OVARIAN TOXICITY IN FEMALE RATS.

Introduction: Triclosan [TCS] is a broad-spectrum antibacterial xenoestrogencommonly used in cosmetics, soap and various products. Exposure to Triclosancreates a disruption between reactive oxygen species (ROS) and antioxidant defenses.Oxymatrine [OMT] has potent anti-cancer, anti-fibrosis, and anti-oxidant effects. Aimof work: To study the effect of Oxymatrine on Triclosan-induced ovarian toxicity.Material and Methods: The studied rats have been divided into five groups formeasurement of total antioxidant capacity, Caspase-3, the levels of Tumor NecrosisFactor –alpha[TNF- α], Estrogen and Progesterone levels , and Liposaccharide BindingProtein [LBP] gene expression in response to Triclosan-induced ovarian toxicity by realtimePolymerase Chain Reaction [PCR]. Results: Triclolsan [TCS] caused statisticallysignificant reduction in total antioxidant capacity, with statistically significant elevationin TNF-α and Caspase-3 activity compared to the control group. Oxymatrine inducedstatistically significant elevation in total antioxidant capacity, statistically significantdecrease in the level of TNF-α, Caspase-3 activity with increased levels of Estrogenand Progesterone compared to the Triclosan group. Histopathological and electronmicroscope examination revealed vacuolar degeneration and atretic follicles in ratstreated with Triclosan, and significant improvement after Oxymatrine intake. In theTriclolsan + Oxymatrine [TCS+OMT] group, gene expression levels of TNF‑α,Caspase 3, Mitogen-activated protein kinase [MAPK], Nuclear Factor-kappa [NF-KB], and Liposaccharide Binding Protein [LBP] were statistically significant reduced comparedto the Triclolsan group. Conclusion: The use of Oxymatrine [OMT] as anti-oxidant leadto decrease in genetic expression of Tumor Necrosis Factor –alpha [TNF‑α] , Caspase 3,p38-MAPK, [NF-KB], and LBP, in Triclosan-induced ovarian injuries.

Bone marrow mononuclear cells boosts anti-cytogentical aberration effect of N-acetylcysteine and α-lipoic acid in rat’s liver and bone marrow: implication of oxidative and inflammatory pathways

This study investigates the hepatoprotective effect of bone marrow mononuclear cells (BM-MNCs) transplantation, N-acetylcysteine (NAC) and α-lipoic acid (ALA). Rats were administrated carbon tetrachloride (CCl4) (1 mg/kg, i.p.) twice/week for 8 weeks for the induction of hepatotoxicity. 7 groups of rats were used as follows: Normal control, CCl4, CCl4 co-administered with BM-MNCs (1 × 106 in 0.1 ml PBS, i.v.), or NAC (300 mg/kg, p.o) or ALA (100 mg/kg, p.o) single or combination. Liver function was tested by measuring serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) and albumin as well as interleukin-6 (IL-6), interleukin-10 (IL-10), tumor necrosis factor-α (TNF-α), malondialdehyde (MDA), total antioxidant capacity (TAC), glutathione peroxidase (Gpx), superoxide dismutase (SOD) and catalase (CAT) activities in liver homogenates. Besides that, estimation of DNA damage was performed. In addition to Micronucleus test and histopathological investigation. CCl4 treated rats showed elevation in ALT, AST, TNF-α, IL-6 and MDA accompanied by reduction in ALB, IL-10, SOD, CAT, GPx and TAC and increased the number of DNA breaks in liver tissue, showed many micronucleated polychromatic erythrocytes (MnPCEs) in bone marrow. NAC, ALA, BM-MNCs and their combination caused a reduction of ALT, AST, while, increase albumin, CAT, TAC, GPx, SOD as compared to CCl4 treated groups. Also decrease in MDA, IL-6 and TNF-α concurrently with an increase in IL-10. Moreover, BM-MNCs, NAC, ALA, and their combination decreased DNA tail %, and the count of MnPCEs. BM-MNCs combination with NAC or ALA exerted significant antioxidant, anti-inflammatory and anti-cytogenetical aberrations effect compared to each of them alone. Highlights CCl4 elevated ALT, AST, TNF-α, IL-6 and MDA CCl4 reduced ALB, IL-10, SOD, CAT, GPx and TAC CCl4 increased the number of DNA breaks in liver NAC, ALA and BM-MNCs reduced ALT, AST, while, increase albumin, CAT, TAC, GPx, SOD NAC, ALA and BM-MNCs decreased in MDA, IL-6 and TNF-α and increased IL-10
