Thiazaborolidines and BH3 adducts derived from thioephedrines

The synthesis of 1,3,2-thiazaborolidine and 2,3-dihydro-2,1,3-borathiazolidines from reaction of ephedrine disulfide and 3,4-dimethyl-5-phenyl-1,3-thiazolidine-2-thione with BH 3 -THF is reported. The two reactions afforded optically active borohydride heterocycles. The X-ray diffraction structure of (1 R ,2 R )-(−)-chlorodeoxy- pseudo -ephedrine hydrochloride, (1 R ,2 R )-(−)-thiosulfonic d deoxy- pseudo -ephedrine and (3 S ,4 R ,5 R )-(+)-dihydro-3,4-dimethyl-5-phenyl-2,1,3-borathiazolidine are reported. Synthesis of three optically active borohydride heterocycles 1 (X= CH 3 , H) and 2 and the X-ray diffraction structure of (1R, 2R)-(−)-chlorodeoxy- pseudo -ephedrine hydrochloride and (1R,2R)-(−)-thiosulfonic deoxy- pseudo -ephedrine and (3S,4R,5R)-(+)-2,3-dihydro-3,4-dimethyl-5-phenyl-2,1,3-borathiazolidine are reported.

Carbon disulfide added to the active methylene group in1 in basicDMF at room temperature to yield the non-isolable 1:1 adduct2.In-situ cyclization of the latter with some α-bromo compounds afforded the corresponding polyfunctionally substituted thiophenes5. The reactivity of5 towards a variety of reagents was studied. Chemical and spectroscopic evidences for the structure of the new compounds are provided.

Reaction of Carbon Disulfide with Active Methylenes: Novel Synthesis of Thiophene, Thieno[2,3-b]thiophene, Thieno[3,2-c]pyrazole and Thieno[3,2-b]pyridine Derivatives

The active methylene compounds - derivatives of malonic or 4-oxobutanoic acid - reacted with carbon disulfide in dimethyl formamide containing potassium hydroxide to yield the non-isolable dipotassium disulfide intermediates. The latter were cyclized by α-haloketones to give thiophene, thieno[2,3-b]thiophene, thieno[3,2-c]pyrazole and thieno[3,2-b]pyridine derivatives. The reactivity of the products toward various reagents was studied.

Lanthanoid metal reacts with diaryl and -alkyl disulfides in the presence of a catalytic amount of benzophenone to generate lanthanoid (III) thiolates. Lanthanoid thiolates thus formed in situ, react with enones to give Michael adducts in good yields.

Thietane, tetrahydrothiophene and tetrahydrothiopyran formation in reaction of methylene-interrupted dienoates with dimethyl disulfide

The reaction of dimethyl disulfide in I 2 with a series of methylene-interrupted methyl dienoates was examined. In all cases ring-closure occurs and thietane formation competes favorably with tetrahydrothiophene formation. When the I 2 concentration was increased, tetrahydrothiopyran formation was observed. These results are explained in terms of two distinct reaction pathways.

A study of the fluorine atom + dimethyl disulfide reaction by photoelectron spectroscopy, supported by ab initio molecular orbital calculations

The F+CH 3 SSCH 3 reaction has been studied by He I photoelectron spectroscopy and is seen to proceed via two competing routes, a hydrogen atom abstraction channel to generate the CH 3 SSCH 2 radical, and an adduct channel, where the adduct unimolecularly decomposes to generate CH 3 SF and CH 3 S. A weak broad band with adiabatic and vertical ionization energies of 7.00±0.07 eV and 7.45± 0.03 eV, respectively, appearing at short reaction times, has been assigned to the first photoelectron band of CH 3 SSCH 2

Reactivity of 42 disulfides with thiol group of human haemoglobin and human serum albumin

The reactivities of disulfides of different compound families towards thiol groups of human haemoglobin and human serum albumin were determined at physiological pH 7.4 by anion-exchange liquid chromatography. The apparent second-order kinetic rate constants, K 1, were calculated for the reaction of these disulfides with each protein. The results show that the studied heterocyclic disulfides are the most reactive compounds with both proteins. The lipophilic properties of these disulfides were evaluated by reversed-phase high performance liquid chromatography, using the percentage of acetonitrile (PAC) required for eluting each compound of the chromatographic column in a water-acetonitrile gradient. The structure-reactivity correlations between log K 1 and log PAC are stated for each protein and compared. They fit a parabolic curve which permits one to define a lipophilic domain corresponding to a quantitative reaction of disulfides towards these proteins. The studied disulfides present a similar optimum of reactivity for both proteins.

Novel transition metal-catalyzed reactions of organic chalcogen compounds with acetylenes are described. In the presence of transition metal-catalysts such as Pd (PPh3)4, the addition of diaryl disulfides and diselenides to terminal acetylenes takes place stereoselectively to give high yields of (Z) -1, 2-bis (arylthio) -1-alkenes and (Z) -1, 2-bis (arylseleno) -1-alkenes, respectively. Similarly, many transition metal-complexes catalyze the addition of thiols (and selenols) to acetylenes, providing vinylic sulfides (and selenides) regioselectively. These reactions may involve vinylic metals as the intermediate. Thus, when the reaction of diaryl disulfides and diselenides with terminal acetylenes is performed under the pressure of carbon monoxide, the carbonylative addition occurs and carbon monoxide is regioselectively incorporated on the side of the terminal carbon of the acetylenes. The obtained carbonylation products, i.e., α, β-unsaturated thiolesters and selenolesters, can be reduced chemoselectively to corresponding α, β-unsaturated aldehydes by treatment with n-Bu3SnH in the presence of Pd (PPh3)4. We believe that this study will open up a new field of transition metal chemistry in combination with sulfur and selenium chemistry.

Synthesis of Novel Naphthoquinone Fused 1, 3-Dithioles

Abstract 2, 3-Dichloro-1, 4-naphthoquinone 3 reacts with 1, 1 -dithiolates 2, prepared from the reaction of CH-acids and carbon disulfide, to give 2-methylidene-4, 10-dioxonaphtho [2, 3-d]-1, 3-dithioles 4. The similar reaction of 3 and dithiocarbamates 6 affords 2-imino-4, 10-dioxonaphtho [2, 3-d] dithioles 7.

Elevation of Glutathione Levels in Bovine Pulmonary Artery Endothelial Cells by N-Acetylcysteine

N-Acetylcysteine (NAC), a cysteine derivative with chemoprotective and radioprotective effects, was found to elevate bovine pulmonary artery endothelial cell (EC) glutathione after in vitro incubation. The elevation in glutathione was associated with enhanced uptake of radioactivity of cystine from the medium. Because cystine in medium was converted rapidly to cysteine and cysteinyl-NAC in the presence of NAC and given that cysteine has a higher affinity for uptake by EC than cystine, we conclude that the enhanced uptake of radioactivity was in the form of cysteine and at least part of the stimulatory effect of NAC on EC glutathione was due to a formation of cysteine by a mixed disulfide reaction of NAC with cystine similar to that previously reported for Chinese hamster ovarian cells (R. D. Issels et al. 1988. Biochem. Pharmacol. 37:881-888). However, NAC was more effective than cysteine in elevating cellular glutathione at equimolar concentrations, and at higher concentrations of NAC an elevation of EC glutathione occurred even in the absence of cystine in the medium through a currently unknown mechanism. Thus, at least two mechanisms are operative in the elevation of endothelial cellular glutathione by NAC. NAC may be a useful compound for elevating glutathione of the pulmonary vasculature for protection against oxidant stress.

Scanning tunneling microscopy of DNA: The chemical modification of gold surfaces for immobilization of DNA

We have investigated the coulostatic immobilization of DNA to a chemically modified gold surface. The chemical modification involves the covalent attachment of chemically polarizable groups by reaction of thiols and disulfides with clean gold surfaces. We have determined that chemically modified surfaces possessing pendant cationic groups bind DNA. In this paper we report the reliable and reproducible scanning tunneling microscopy (STM) imaging of DNA using a gold surface chemically modified with N,N,-dimethyl-2-mercaptoethylamine and present the first STM images of an entire genetically functional DNA molecule, the circular plasmid pBS+.

Reactions of chloro(diethylenetriamine)platinum(1+) chloride and chloro(terpyridine)platinum(1+) chloride with thiols, thioethers, and dialkyl disulfides: a platinum-195 NMR study

The reactions of monofunctional platinum compounds [Pt(dien)Cl]Cl (dien = 1,5-diamino-3-azapentane) and [Pt(trpy)Cl]Cl (trpy = 2,2[prime],2[double prime]-terpyridine) with thiols (RSH), thioethers (RSR), and disulfides (RSSR) have been investigated by [sup 1]H, [sup 13]C, and [sup 195]Pt NMR spectroscopy. Empirical trends in [sup 195]Pt NMR chemical shifts are noted in terms of the classes sulfur functional groups coordinated to platinum. It appears that neither the steric bulk nor the electronic character of the alkyl group makes a significant contribution to the [sup 195]Pt nuclear shielding. Reactions of dialkyl disulfides with [Pt(dien)Cl]Cl initially lead to a monodentate complex [Pt(dien)(RSSR)][sup 2+] which can be isolated as the ClO[sub 4][sup [minus]] or PF[sub 6][sup [minus]] salts. In the presence of [Pt(dien)Cl]Cl, the monodentate disulfido complex undergoes further reaction to cleave the S-S bond in the disulfide ligand, via a redox process.

Reactions of Sulfenic and Sulfoxylic Acid Derivatives with Olefins in the Presence of Sulfur Trioxide and its Complexes

Abstract The present review deals with the application of sulfenic [RS-OH] and sulfoxylic [HO-S-OH] acid derivatives in electrophilic addition reactions. We consider a new method for generation of reactive electrophilic sulfenylating species from sulfenic/sulfoxylic acid derivatives and sulfur trioxide (or its complexes). This autoreview covers our results in the systematic study of SO3-mediated addition to olefins and of sulfenylation of olefins with these novel electrophiles. The reactions of disulfides, sulfenyl chlorides, sulfenamides, alkyl sulfenates, aminosulfenyl halides and thiobisamines with a wide range of unsaturated compounds are discussed. A special chapter concerns the acid-catalyzed nucleophilic substitution of the products obtained.
