Quantitative Determination of Incarvillateine in Incarvillea sinensis by Solid Phase Extraction and High Performance Liquid Chromatography

Method for rapid quantitative analysis of incarvillateine in Incarvillea sinensis by high-performance liquid chromatography (HPLC) has been developed. The sample preparation involves solid phase extraction (SPE) with a mixed-mode reversed-phase and cation-exchange cartridge. The linear calibration range for incarvillateine was 0.002-0.5 mg/ml. The limit of detection was 0.35 microg/ml (S/N=3). Intra- and interday precisions were less than 0.36% (n=6) and 1.61% (n=18), respectively. The recovery of incarvillateine was 97.61-102.44% with the relative standard deviation (RSD) ranging from 0.63 to 1.93% (n=3). This method was proposed as a simple, rapid and accurate method for quantitative determination of incarvillateine content in various samples of Incarvillea sinensis collected from different areas of China.

Quantitative determination of short‐chain branching content and distribution in commercial polyethylenes by thermally fractionated differential scanning calorimetry

AbstractA method for rapid quantitative analysis of the content and distribution of short chain branching (SCB) for α‐olefin/ethylene copolymers based on thermally fractionated DSC is presented. Eight commercial polyethylenes, four made with conventional Ziegler‐Natta catalysts and four made with metallocene catalysts, were analyzed by differential scanning calorimetry (DSC), after having been thermally segregated by successive nucleation annealing (SNA). The polyethylenes were also analyzed by temperature rising elution fractionation (TREF) and carbon‐13 nuclear magnetic resonance (13C‐NMR). The SNA‐DSC procedure segregates polyethylenes according to methylene sequence lengths (MSL). The relationship between DSC melting temperature and SCB content was obtained by calibration with linear hydrocarbons; TREF results were not used in the SNA‐DSC calibration. Deconvolution of the SNA‐DSC endotherms yielded estimates of the average SCB contents and SCB distributions. The SCB contents obtained from the SNA‐DSC for linear low density polyethylenes agreed very well with the SCB contents obtained by 13C‐NMR and TREF, and the SCB distributions measured by SNA‐DSC were very similar to those obtained by TREF. The SCB contents obtained by SNA‐DSC for ultra‐low density polyethylenes, made with metallocene catalysts, were about 20% lower than the values obtained by 13C‐NMR; the values obtained by TREF were even lower.

Rapid quantification of murine endothelin-1 and vasoactive intestinal contractor gene expression levels by a real-time PCR system

A rapid quantitative analysis method for murine endothelin-1 (ET-1) and vasoactive intestinal contractor (VIC) gene expression levels was established using a real-time polymerase chain reaction (PCR). We designed primer pairs and TaqMan probes specific for murine prepro-ET-1 (PPET-1) and prepro-VIC (PPVIC) genes, based on the cDNA sequence region common to both mouse and rat. The dynamic range for detection in this system spanned 100 000-fold of the starting molecule. The gene expression levels of PPET-1 and PPVIC were estimated as gene expression rates normalized by the expression of the house-keeping gene, glyceraldehyde-3-phosphate dehydrogenase. To examine the reproducibility of this assay system, we calculated the intra-assay and interassay coefficients of variation of the gene expression rate, which ranged from 16.2 to 55.0% and from 24.2 to 56.5%, respectively. Using this system, we examined gene expression levels of PPET-1 and PPVIC in mouse tissues. PPET-1 gene expression was found in all tissues at relatively high levels, whereas high levels of PPVIC gene expression were observed only in stomach, intestine, uterus, and ovary. The gene expression patterns agreed well with those determined by RNase protection assay and conventional PCR. These results show that this new rapid method is accurate and reproducible.

A Simple Rapid Method To Estimate Hyaluronic Acid Concentrations in Rooster Comb and Wattle Using Cellulose Acetate Electrophoresis

A simple rapid method of quantitative analysis of hyaluronic acid (HA) from comb and wattle tissues is described. The technique involves short-time acetone drying and proteolysis of tissues, cellulose acetate electrophoresis of the digests without deproteinization, and measurement of absorbance of the solution of Alcian Blue extracted from the HA band. With this method, the determination of HA, which may take 1 week or longer with other methods, could be made within a day.

Analysis of phospholipids in brain tissue by 31P NMR at different compositions of the solvent system chloroform-methanol-water.

Brain phospholipids can be quantitated by high-resolution 31P NMR of crude brain tissue extracts in the solvent system chloroform-methanol-water (+EDTA) which was introduced recently (P. Meneses and T. Glonek, J. Lipid Res. 29, 679 (1988)). Phospholipid resonance positions depend on the type of tissue extract and on solvent composition. The effects of systematic variation of the solvent system on phospholipid NMR profiles are presented. Resolution can be optimized by adjustment of the solvent composition. Virtually all phospholipid classes can be resolved, and the major phospholipids in brain: sphingomyelin and phosphatidyl-choline, -serine, -inositol, -ethanolamine, and -ethanolamine plasmalogen can be quantitated easily. Additional resonances have been assigned to phosphatidylcholine plasmalogen, alkylacyl-phosphatidylcholine and phosphatidylinositolbisphosphate. NMR offers a rapid method for quantitative analysis of the phospholipid composition in brain tissue which requires minimum sample handling.

Simple and Rapid Method for Quantitative Analysis of Boric Acid in Agars

A method was developed for the rapid and simple analysis of boric acid in agar and seaweed.Hard glass beakers and flasks were used to dissolve the samples in diluted sulfuric acid (0.1-1.8N) by boiling. The optimum boiling times were 3min for agar and 10min for seaweed. These sample solutions did not gel even if the temperature fell to below 20°C. The concentration of boric acid in the solution was immediately determined by the spectrophotometric curcumic method after extraction with 2-ethyl-1, 3-hexanediol in chloroform.A time of only 1.5 to 2.0 hours was required to analyze several samples simultaneously. The duplicate analysis showed good precision, and the relative standard deviation was 5.4-6.8% for boric acid at levels corresponding to 1, 000mg/kg in dry samples.The value obtained by the present method was about 20 to 25% higher than the value obtained by the official method based on titration.

A simple and rapid method of quantitative analysis of phosphoamino acids by high-performance liquid chromatography

A high-performance liquid chromatographic system for the separation of nonradiolabeled phosphoamino acids and orthophosphate by ion-pair reverse-phase chromatography has been developed. By the use of low-ionic-strength phthalate buffers at pH 6.3, the phosphoamino acids can be visualized by virtue of this uv-active eluant. The technique is sensitive to 200 pmol of phosphoamino acid and has been shown to be directly applicable to the analysis of isolated phosphoproteins.

Quantitative Analytical Technique Using Holography in Studing Structural Characteristics of Machine Tools

A new rapid quantitative analysis method of 3-D displacements of large complex objects, such as machine tools by using holography is described emphatically in this paper. Full-field 3-D displacements of such a machine can be evaluated rapidly by the microcomputer and only a few, geometric parameters need to be measured. A prime system for automatically identifying the hologram fringe patterns of large objects has been developed as well. Once holograms put is the system, the data of full-field 3-D displacements will be obtained from the output end of the system. The method of identifying displacement signs with the real-time holography is presented in this paper too. Then, it is expounded that the rapid 3-D quantitative analysis of holography in machinery industry is realiable.

Measurement of urine porphyrins and porphyrinogens

A simple and rapid method for quantitative analysis of urine porphyrins is described. After acidification of urine to 1 n with HCl, the first derivative of the absorption spectrum is recorded in the region of the Soret band maximum. Nonspecific interference is not significant, and the point of intersection of the derivative of the Soret band maximum with the baseline provides an estimate of the ratio of uro- to coproporphyrins. Using this approach, which avoids solvent extraction prior to absorptiometry, we have defined pH-dependent differences in porphyrin and porphyrinogen partition between ethyl acetate and aqueous solvent or urine. These marked differences have been used to define conditions for separation of porphyrin and porphyrinogen prior to measurement. Using the methods described, recovery of uro- and coproporphyrinogens added to rat urine ranged from 87 to 100%.

Rapid Determination of Cellulose in Viscose

A new method for rapid quantitative analysis of cellulose in viscose has been developed. The method is to regenerate cellulose in viscose by use of ammonium chloride and hydrochloric acid, the regenerated cellulose is separated by centrifuging, taken up in sulfuric acid, oxidized with an excess of potassium dichromate solution and the excess of the dichromate is back titrated with the standard ferrous ammonium sulfate solution with the use of barium salt of diphenylamine as an internal indicator. The time required for the analysis by this method is about one hour while it requires about hours by usual quantitative method.
