Exogenous Melatonin Improves Drought Tolerance in Maize Seedlings by Regulating Photosynthesis and the Ascorbate–Glutathione Cycle

Melatonin is known to exert protective effects in maize against drought stress, but the knowledge regarding interaction among the melatonin, photosynthetic efficiency, ascorbate–glutathione cycle still remains elusive. Two contrasting maize (Zea mays L.) genotypes, SD609 (drought-tolerant) and SD902 (drought-sensitive), were subjected to moderate and severe drought stress along with 100 μM melatonin treatment. Exogenous melatonin increased fluorescence curve levels for both genotypes under drought stress, and higher in SD609 than SD902. In both genotypes, melatonin application also significantly increased the photochemical efficiency of photosystem II (PSII), the effective quantum yield of PSII and photosystem I (PSI), and the electron transport rate between PSII and PSI under drought stress, but it decreased the quantum yield of energy dissipation and the donor side and acceptor side impairment of PSI under drought stress, regardless of stress severity. Exogenous melatonin also contributed to the reduction of membrane damage under drought stress, as reflected by significantly decreased levels of MDA, superoxide anions and H2O2 of both genotypes exposed to drought. Furthermore, melatonin treatment increased enzyme activity in AsA–GSH cycle, and upregulated the genes expression of related enzymes in AsA–GSH cycle. The effects of melatonin were more pronounced in SD609 than SD902. Collectively, these results indicated that exogenous melatonin application increased photosynthetic electron transport rate and accelerated the AsA–GSH cycle, which are factors that play an important role in drought tolerance in maize. The beneficial effect of melatonin on the drought-tolerance SD609 was more obvious.

Biological effects of the free and embedded metribuzin and tribenuron-methyl herbicides on various cultivated weed species

The present study addresses the herbicidal activity and biological effects of the metribuzin (MET) and tribenuron-methyl (TBM) herbicides used to control various weed species (Amaranthus retroflexus, Sinapis arvensis, and Leucanthemum maximum). The effects of the free herbicides and the herbicides embedded in granules of degradable polymer poly-3-hydroxybutyrate [P(3HB)] blended with birch wood flour were compared. Metribuzin, regardless of the form, caused 100% mortality of the three weeds by day 21. The herbicidal activity of tribenuron-methyl was lower than that of metribuzin, but the embedded TBM was superior to the free herbicide in the length and strength of its action on the weeds. Both metribuzin forms dramatically decreased the main parameters of fluorescence: maximum quantum yield of photosystem-II [Y(II)max], maximum quantum yield of non-photochemical quenching [Y(NPQ)max], and maximum rate of non-cyclic electron transport [ETRmax] and concentrations of chlorophyll a and b. The effect of the embedded TBM on the photosynthetic activity of the weeds was lower in the first two weeks of the growth of herbicide-treated plants but lasted longer than the effect of the free TBM and increased over time. Embedding of metribuzin in the matrix of degradable blend did not decrease its herbicidal activity.

Photosynthetic and ultrastructural adaptability of Anemone shikokiana leaves to heterogeneous habitats

Anemone shikokiana (Makina) Makina is distributed in two heterogeneous habitats, including mountaintop shrubland and conifer and broad-leaf mixed forest. To better understand the mechanisms used by A. shikokiana in adapting to these different environments, the photosynthetic efficiency, chlorophyll fluorescence and ultrastructure of the leaves were investigated. The findings revealed that, under the same optical quantum flux density, the net photosynthetic rate in the leaves of mountaintop shrubland was significantly higher than that found in the mixed forest, but the stomatal conductance, intercellular CO2 concentrations and transpiration rate were lower. The effective quantum yield of photosystem II (PSII), photochemical quenching coefficient, non-photochemical quenching coefficient and electron transfer rate in the conifer and broad-leaf mixed forest were significantly lower than those values determined for plants in mountaintop shrubland. However, the maximal quantum yield of PSII exhibited no significant difference between the two habitats. Transmission electron micrographs revealed that the numbers of chloroplasts and mitochondria per mesophyll cell and starch grains per chloroplast in the mountaintop shrubs were higher than those found in the conifer and broad-leaf mixed forest samples. Moreover, they showed an increasing trend month by month in April, May and June. It indicated that elevated temperature increased their numbers. These findings illustrate that A. shikokiana efficiently uses environmentally limited resources to adapt to different living environments. The study reveals that the mechanisms that underlie the response of A. shikokiana to heterogeneous habitats involve photosynthetic and ultrastructural variations, thus providing a theoretical basis for future study.

ZnO nanoparticles impact on the photosynthetic activity of Vicia faba: Effect of particle size and concentration

The present study evaluated the impact of ZnONPs on the photosynthetic activity of Vicia faba plants. Time evolution of chlorophyll a fluorescence parameters (Fv/Fm, NPQ, Fq′/Fm′, Fq′/Fv′, and Fv′/Fm′), leaf temperature, and stomatal conductance (gs) were monitored during 144 h for leaves subjected to ZnONPs (25 and 70 nm diameter) at 50, 100, 200, and 300 mg L−1 as well as for control leaves. The ZnONPs morphology and size were characterized by transmission electron microscopy. Their capacity to generate reactive oxygen species (ROS) and release Zn ions were assessed by fluorescence and dialysis experiments, respectively. The results revealed that ZnONPs internalized in the leaves, impacting the photosynthetic performance of Vicia faba, reducing the quantum efficiency of photosystem II (PSII), steady-state PSII quantum yield, coefficient of photochemical quenching in steady-state, and stomatal conductance. The particle size played a key role as the photosynthetic functioning of the plant was more affected by ZnONPs of 25 nm diameter than those of 70 nm. A similar rate constant ROS production (kROS) of about 4.6 10−1 M−1 s−1 was found for both ZnONPs. In contrast, a rate constant of Zn2+ dissolution (KdZn+2) of 1.064 10−3 and 0.743 10−3 h−1 was determined for ZnONPs of 25 and 70 nm diameter, respectively. Thus, our findings indicate that Zn2+ ions played the major role on the size-dependent phytotoxicity because ZnONPs of 25 and 70 nm presented similar ability of ROS generation by themselves while 25 nm ZnONPs released 30% more Zn2+ than 70-nm.

Eco-Physiological Screening of Different Tomato Genotypes in Response to High Temperatures: A Combined Field-to-Laboratory Approach.

High temperatures represent a limitation for growth and development of many crop species. Several studies have demonstrated that the yield reduction of tomato under high temperatures and drought is mainly due to a photosynthetic decline. In this paper, a set of 15 tomato genotypes were screened for tolerance to elevated temperatures by cultivating plants under plastic walk-in tunnels. To assess the potential tolerance of tomato genotypes to high temperatures, measurements of chlorophyll fluorescence, pigments content and leaf functional traits have been carried out together with the evaluation of the final yields. Based on the greenhouse trials, a group of eight putative heat-sensitive and heat-tolerant tomato genotypes was selected for laboratory experiments aimed at investigating the effects of short-term high temperatures treatments in controlled conditions. The chlorophyll fluorescence induction kinetics were recorded on detached leaves treated for 60 min at 35 °C or at 45 °C. The last treatment significantly affected the photosystem II (PSII) photochemical efficiency (namely maximum PSII quantum efficiency, Fv/Fm, and quantum yield of PSII electron transport, ΦPSII) and the non-photochemical quenching (NPQ) in the majority of genotypes. The short-term heat shock treatments also led to significant differences in the shape of the slow Kautsky kinetics and its significant time points (chlorophyll fluorescence levels minimum O, peak P, semi-steady state S, maximum M, terminal steady state T) compared to the control, demonstrating heat shock-induced changes in PSII functionality. Genotypes potentially tolerant to high temperatures have been identified. Our findings support the idea that chlorophyll fluorescence parameters (i.e., ΦPSII or NPQ) and some leaf functional traits may be used as a tool to detect high temperatures-tolerant tomato cultivars.

Leaf chlorophyll estimates of temperate deciduous shrubs during autumn senescence using a SPAD-502 meter and calibration with extracted chlorophyll

The soil plant analysis development (SPAD) meter proved effective at estimating leaf Chl content of native and non-native temperate deciduous shrubs. However, due to the change in the relationship between SPAD values and Chl content during the course of autumn senescence, it may be necessary to establish separate calibration equations to cover a range of leaf Chl concentrations. Non-destructive estimates of leaf chlorophyll (Chl) content by hand-held chlorophyll meters such as the Minolta SPAD-502 provide an effective time-efficient method of collecting field data on senescence. To establish useable species-specific calibration equations of relationships between chlorophyll meter readings and leaf Chl content. During autumn, we collected SPAD values and quantum yield of photosystem-II (ΦPSII) over a 10-week period from 1044 leaf samples representing native and non-native temperate deciduous shrub species growing in the wild. Subsequently, Chl was extracted from leaf discs (1 cm diameter), incubated at 65 °C for 2 h in dimethyl sulfoxide, and quantified by spectroscopy. Relationships between extracted Chl and SPAD values were established using linear, quadratic, and exponential equations. Quadratic functions proved the most reliable fit for the data. Interestingly, non-native species tended to have higher leaf Chl content and SPAD values but also exhibited higher variability than native species. The strength (r2) of the SPAD-Chl relationships was weaker than those reported for agricultural or greenhouse-grown plants, but strengthened over time as Chl, ΦPSII, and SPAD values declined. The SPAD meter proved effective at estimating leaf Chl content of native and non-native temperate deciduous shrubs. These estimates may be used in future to validate remote sensing (satellite and near-surface) vegetation indices in late autumn when trees are leafless.

Manganese Repairs the Oxygen-Evolving Complex (OEC) in Maize (Zea mays L.) Damage During Seawater Vulnerability

This study was aimed to investigate how exogenous manganese (Mn) would limit damage in the oxygen-evolving complex (OEC) and photosynthetic apparatus of maize seedlings caused during seawater vulnerability. In this study, seawater was applied in 2-week-old maize (Zea mays L.) seedling, and the degree of damage of photosynthetic pigment pool, the OEC, and net electron transport rate were observed. Mn supplement was also added in maize seedlings to limit the damage of the OEC and photosynthetic apparatus caused during salinity. Leaf relative water content (RWC), fresh weight (FW), and photosynthetic pigment pool (chlorophyll a, chlorophyll b, and carotenoids) sharply declined after 7 days of treatment; however, Mn supplement increased these values. Chlorophyll fluorescence induction (OJIP) transients showed low Fv/Fo, an additional K step, enhanced variable fluorescence (VK) and degree of damage to the OEC (WK) during salinity, and indicates damage of OEC at electron donor side of photosystem II (PSII). The OEC intact within PSII was a primary damage center during salinity which inhibited electron transport process that resulted in a huge loss of maximum quantum yield of PSII (Fv/Fm), but a significant recovery in photosynthetic apparatus was observed after exogenous application of manganese. Structural and functional integrity of the photosynthetic apparatus was recovered up to a certain extent after exogenous application of Mn. Findings from this study should help to understand the basic knowledge of photosynthetic apparatus efficiency in response to damage caused by exposure to seawater. Outcomes of this study will be used to mitigate salinity problem with Mn supplement for growth and development of crops.

Comparisons of Chlorophyll Fluorescence and Physiological Characteristics of Wheat Seedlings Influenced by Iso-Osmotic Stresses from Polyethylene Glycol and Sodium Chloride

Wheat (Triticum aestivum) cultivar Taichung SEL.2 (TCS2) is a salt-tolerance variety, but the mechanism involved remains unclear. This study aims to distinguish between the non-ionic osmotic and salt-mediated physiological effects on TCS2. Osmotic agents polyethylene glycol (PEG) and sodium chloride (NaCl) were applied at three iso-osmotic levels, level 1 containing 24% (w/v) PEG and 200 mM NaCl, level 2 containing 26.5% (w/v) PEG and 250 mM NaCl), and level 3 containing 29% (w/v) PEG and 300 mM NaCl, respectively. According to the investigation of chlorophyll fluorescence in the better NaCl-treated seedlings, maximal quantum yield of photosystem II (PSII) (Fv/Fm) and significant higher effective quantum yield of PSII (ΦPSII) at level 3 were observed. Meanwhile, the non-photochemical quenching of PSII (NPQ) and the quantum yield of regulated energy dissipation of PSII [Y(NPQ)] were significantly higher in the NaCl-treated seedlings, and the quantum yield of non-regulated energy dissipation of PSII [Y(NO)] in the NaCl-treated seedlings was lower than the PEG-treated ones at level 2 and level 3. Furthermore, the less extensive degradation of photosynthetic pigments, the better ascorbate peroxidase (APX) activity and the less accumulation of malondialdehyde (MDA) were also observed in NaCl-treated seedlings. In the morphological traits, shoot elongation in NaCl-treated seedlings was also preserved. These results suggest that TCS2 is more resistant to NaCl-induced osmotic stress than to the PEG-induced stress. This study contributes to plant breeder interest in drought- and/or salt-tolerant wheat varieties.

Genetic architecture of photosynthesis energy partitioning as revealed by a genome-wide association approach.

The photosynthesis process is determined by the intensity level and spectral quality of the light; therefore, leaves need to adapt to a changing environment. The incident energy absorbed can exceed the sink capability of the photosystems, and, in this context, photoinhibition may occur in both photosystem II (PSII) and photosystem I (PSI). Quantum yield parameters analyses reveal how the energy is managed. These parameters are genotype-dependent, and this genotypic variability is a good opportunity to apply mapping association strategies to identify genomic regions associated with photosynthesis energy partitioning. An experimental and mathematical approach is proposed for the determination of an index which estimates the energy per photon flux for each spectral bandwidth (Δλ) of the light incident (QI index). Based on the QI, the spectral quality of the plant growth, environmental lighting, and the actinic light of PAM were quantitatively very similar which allowed an accurate phenotyping strategy of a rice population. A total of 143 genomic single regions associated with at least one trait of chlorophyll fluorescence were identified. Moreover, chromosome 5 gathers most of these regions indicating the importance of this chromosome in the genetic regulation of the photochemistry process. Through a GWAS strategy, 32 genes of rice genome associated with the main parameters of the photochemistry process of photosynthesis in rice were identified. Association between light-harvesting complexes and the potential quantum yield of PSII, as well as the relationship between coding regions for PSI-linked proteins in energy distribution during the photochemical process of photosynthesis is analyzed.

Photosynthetic characterization of transgenic Synechocystis expressing a plant thiol/disulfide-modulating protein

ABSTRACT A previous study showed that introducing an Arabidopsis thaliana thiol/disulfide-modulating protein, Low Quantum Yield of Photosystem II 1 (LQY1), into the cyanobacterium Synechocystis sp. PCC6803 increased the efficiency of Photosystem II (PSII) photochemistry. In the present study, the authors provided additional evidence for the role of AtLQY1 in improving PSII photochemical efficiency and cell growth. Light response curve analysis showed that AtLQY1-expressing Synechocystis grown at a moderate growth light intensity (50 µmol photons m−2 s−1) had higher minimal, maximal, and variable fluorescence than the empty-vector control, under a wide range of actinic light intensities. Light induction and dark recovery curves demonstrated that AtLQY1-expressing Synechocystis grown at the moderate growth light intensity had higher effective PSII quantum yield, higher photochemical quenching, lower regulated heat dissipation (non-photochemical quenching), low amounts of reduced plastoquinone, and higher amounts of oxidized plastoquinone than the empty-vector control. Furthermore, growth curve analysis indicated that AtLQY1-expressing Synechocystis grew faster than the empty-vector control at the moderate growth light intensity. These results suggest that transgenic expression of AtLQY1 in Synechocystis significantly improves PSII photochemical efficiency and overall cell growth.

Effect of Hexavalent Chromium [Cr(VI)] on Phytoremediation Potential and Biochemical Response of Hybrid Napier Grass with and without EDTA Application.

Hexavalent chromium [Cr(VI)] contamination has become an emergent concern in China. Previous field investigations have found that hybrid Napier grass is widely distributed in Cr(VI) contaminated areas. This study investigated the phytoremediation potential and biochemical response of hybrid Napier grass (Pennisetum americanus L. × Pennisetum purpureum Schumach) grown in soil contaminated with Cr(VI) (0, 20, 40, and 60 mg kg−1) with and without Ethylene diamine tetra acetic acid (EDTA) (4 mM) application. The results indicated that root length, shoot height, dry weight, leaf area, chlorophyll, and photosystem II (PSII) parameters viz.; apparent electron transport rate (ETR), effective quantum yield of PSII (ΦPSⅡ), maximal PSII photochemical efficiency (Fv/Fm), potential activity of PSII (Fv/Fo), photochemical quenching (qP), and non-photochemical quenching (qN) decreased with the increasing Cr(VI) concentration. EDTA application further aggravated reduction of dry biomass and photosystem II. The concentration and the accumulation of Cr in shoot and root, and both the bioaccumulation factor (BAF) and transfer factor (TF) increased with increasing Cr(VI) concentrations and further enhanced with EDTA application. Though the Cr(VI) and Ethylene diamine tetra acetic acid (EDTA) stress reduced tolerance, but, even at highest Cr(VI) concentration, plant could exhibited strong resistance, as evidenced by increase in superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) activities. Hybrid Napier grass, due to its BAF > 1 and a TF < 1, would be applicable for Cr phytostabilization. Moreover, limiting metal transport to aerial parts of plant would prevent animal’s ingestion, restrict soil mobility, and consequently reduce transmission across the food chain.

Introducing an Arabidopsis thaliana Thylakoid Thiol/Disulfide-Modulating Protein Into Synechocystis Increases the Efficiency of Photosystem II Photochemistry.

Photosynthetic species are subjected to a variety of environmental stresses, including suboptimal irradiance. In oxygenic photosynthetic organisms, a major effect of high light exposure is damage to the Photosystem II (PSII) reaction-center protein D1. This process even happens under low or moderate light. To cope with photodamage to D1, photosynthetic organisms evolved an intricate PSII repair and reassembly cycle, which requires the participation of different auxiliary proteins, including thiol/disulfide-modulating proteins. Most of these auxiliary proteins exist ubiquitously in oxygenic photosynthetic organisms. Due to differences in mobility and environmental conditions, land plants are subject to more extensive high light stress than algae and cyanobacteria. Therefore, land plants evolved additional thiol/disulfide-modulating proteins, such as Low Quantum Yield of PSII 1 (LQY1), to aid in the repair and reassembly cycle of PSII. In this study, we introduced an Arabidopsis thaliana homolog of LQY1 (AtLQY1) into the cyanobacterium Synechocystis sp. PCC6803 and performed a series of biochemical and physiological assays on AtLQY1-expressing Synechocystis. At a moderate growth light intensity (50 µmol photons m-2 s-1), AtLQY1-expressing Synechocystis was found to have significantly higher F v /F m, and lower nonphotochemical quenching and reactive oxygen species levels than the empty-vector control, which is opposite from the loss-of-function Atlqy1 mutant phenotype. Light response curve analysis of PSII operating efficiency and electron transport rate showed that AtLQY1-expressing Synechocystis also outperform the empty-vector control under higher light intensities. The increases in F v /F m, PSII operating efficiency, and PSII electron transport rate in AtLQY1-expressing Synechocystis under such growth conditions most likely come from an increased amount of PSII, because the level of D1 protein was found to be higher in AtLQY1-expressing Synechocystis. These results suggest that introducing AtLQY1 is beneficial to Synechocystis.

The Mitochondrial Respiratory Chain Maintains the Photosynthetic Electron Flow in Arabidopsis thaliana Leaves under High-Light Stress

The plant respiratory chain includes the ATP-coupling cytochrome pathway (CP) and ATP-uncoupling alternative oxidase (AOX). Under high-light (HL) conditions, plants experience photoinhibition, leading to a damaged photosystem II (PSII). The respiratory chain is considered to affect PSII maintenance and photosynthetic electron transport under HL conditions. However, the underlying details remain unclear. In this study, we investigated the respiratory chain functions related to PSII maintenance and photosynthetic electron transport in plants exposed to HL stress. We measured the HL-induced decrease in the maximum quantum yield of PSII in the leaves of wild-type and AOX1a-knockout (aox1a) Arabidopsis thaliana plants in which CP was partially inhibited by a complex-III inhibitor. We also calculated PSII photodamage and repair rate constants. Both rate constants changed when CP was partially inhibited in aox1a plants, suggesting that the respiratory chain is related to both processes. Before HL stress, photosynthetic linear electron flow (LEF) decreased when CP was partially inhibited. After HL stress, aox1a in the presence of the CP inhibitor showed significantly decreased rates of LEF. The electron flow downstream from PSII and on the donor side of photosystem I may have been suppressed. The function of respiratory chain is required to maintain the optimal LEF as well as PSII maintenance especially under the HL stress.

Drought‐Net rainfall shelters did not cause nondrought effects on photosynthesis for California central coast plants

AbstractRainfall interception shelters are frequently used to study the ecological consequences of drought. One common shelter design employs V‐shaped plastic troughs spaced on a supporting frame to intercept rainfall. Shading, reflection, and infrared radiation may alter the radiative environment under shelters in ways independent of their intended effect on soil moisture. We measured microclimate and several photosynthetic variables for watered, potted plants under rain‐out shelters and in open‐air, unsheltered plots. We tested whether the shelter infrastructure altered aboveground micrometeorology and photosynthesis for watered, potted plants of native Californian species: Elymus glaucus, Eriogonum latifolium, Mimulus aurantiacus, and Morella californica. We quantified the effects on photosynthesis in terms of light harvesting by photosystem II (PSII) and leaf‐level gas exchange on open‐air and shelter plots, the quantum yield of PSII for darkened leaves, dark respiration, and nocturnal stomatal conductance. The rain‐out shelter reduced daily integrated photosynthetically active radiation by 20%. Air temperature, leaf temperature, and leaf‐to‐air vapour pressure difference were not different under shelters compared with controls during the day. Likewise, there were no effects of shelters on net CO2 assimilation, stomatal conductance to water vapour (gs), internal leaf (CO2), or electron transport rate through PSII during the daytime. At night, Tair was 0.6°C higher under shelters, but there were no effects on dark respiration or stomatal conductance. Despite some differences in micrometeorology under rain‐out shelters compared with open‐air plots, there were little or no aboveground nondrought effects of the shelters on leaf‐level photosynthesis for watered, potted plants of these California native plant species.

Chlorophyll Fluorescence Imaging Analysis for Elucidating the Mechanism of Photosystem II Acclimation to Cadmium Exposure in the Hyperaccumulating Plant Noccaea caerulescens.

We provide new data on the mechanism of Noccaea caerulescens acclimation to Cd exposure by elucidating the process of photosystem II (PSII) acclimation by chlorophyll fluorescence imaging analysis. Seeds from the metallophyte N. caerulescens were grown in hydroponic culture for 12 weeks before exposure to 40 and 120 μM Cd for 3 and 4 days. At the beginning of exposure to 40 μM Cd, we observed a spatial leaf heterogeneity of decreased PSII photochemistry, that later recovered completely. This acclimation was achieved possibly through the reduced plastoquinone (PQ) pool signaling. Exposure to 120 μM Cd under the growth light did not affect PSII photochemistry, while under high light due to a photoprotective mechanism (regulated heat dissipation for protection) that down-regulated PSII quantum yield, the quantum yield of non-regulated energy loss in PSII (ΦNO) decreased even more than control values. Thus, N. caerulescens plants exposed to 120 μM Cd for 4 days exhibited lower reactive oxygen species (ROS) production as singlet oxygen (1O2). The response of N. caerulescens to Cd exposure fits the ‘Threshold for Tolerance Model’, with a lag time of 4 d and a threshold concentration of 40 μM Cd required for the induction of the acclimation mechanism.

The Low Molecular Mass Photosystem II Protein PsbTn Is Important for Light Acclimation.

Photosystem II (PSII) is a supramolecular complex containing over 30 protein subunits and a large set of cofactors, including various pigments and quinones as well as Mn, Ca, Cl, and Fe ions. Eukaryotic PSII complexes contain many subunits not found in their bacterial counterparts, including the proteins PsbP (PSII), PsbQ, PsbS, and PsbW, as well as the highly homologous, low-molecular-mass subunits PsbTn1 and PsbTn2 whose function is currently unknown. To determine the function of PsbTn1 and PsbTn2, we generated single and double psbTn1 and psbTn2 knockout mutants in Arabidopsis (Arabidopsis thaliana). Cross linking and reciprocal coimmunoprecipitation experiments revealed that PsbTn is a lumenal PSII protein situated next to the cytochrome b 559 subunit PsbE. The removal of the PsbTn proteins decreased the oxygen evolution rate and PSII core phosphorylation level but increased the susceptibility of PSII to photoinhibition and the production of reactive oxygen species. The assembly and stability of PSII were unaffected, indicating that the deficiencies of the psbTn1 psbTn2 double mutants are due to structural changes. Double mutants exhibited a higher rate of nonphotochemical quenching of excited states than the wild type and single mutants, as well as slower state transition kinetics and a lower quantum yield of PSII when grown in the field. Based on these results, we propose that the main function of the PsbTn proteins is to enable PSII to acclimate to light shifts or intense illumination.

Far-red light enhances photochemical efficiency in a wavelength-dependent manner.

Linear electron transport depends on balanced excitation of photosystem I and II. Far-red light preferentially excites photosystem I (PSI) and can enhance the photosynthetic efficiency when combined with light that over-excites photosystem II (PSII). The efficiency of different wavelengths of far-red light exciting PSI was quantified by measuring the change in quantum yield of PSII (ΦPSII ) of lettuce (Lactuca sativa) under red/blue light with narrowband far-red light added (from 678 to 752 nm, obtained using laser diodes). The ΦPSII of lettuce increased with increasing wavelengths of added light from 678 to 703 nm, indicating longer wavelengths within this region are increasingly used more efficiently by PSI than by PSII. Adding 721 nm light resulted in similar ΦPSII as adding 703 nm light, but ΦPSII tended to decrease as wavelength increased from 721 to 731 nm, likely due to decreasing absorptance and low photon energy. Adding 752 nm light did not affect ΦPSII . Leaf chlorophyll fluorescence light response measurements showed lettuce had higher ΦPSII under halogen light (rich in far-red) than under red/blue light (which over-excites PSII). Far-red light is more photosynthetically active than commonly believed, because of its synergistic interaction with light of shorter wavelengths.

Response of natural phytoplankton communities exposed to crude oil and chemical dispersants during a mesocosm experiment

During the 2010 Deepwater Horizon oil spill, the chemical dispersant Corexit was applied over vast areas of the Gulf of Mexico. Marine phytoplankton play a key role in aggregate formation through the production of extracellular polymeric materials (EPS), an important step in the biological carbon pump. This study examined the impacts of oil and dispersants on the composition and physiology of natural marine phytoplankton communities from the Gulf of Mexico during a 72-hour mesocosm experiment and consequences to carbon export. The communities were treated using the water accommodated fraction (WAF) of oil, which was produced by adding Macondo surrogate oil to natural seawater and mixed for 24 h in the dark. A chemically enhanced WAF (CEWAF) was made in a similar manner, but using a mixture of oil and the dispersant Corexit in a 20:1 ratio as well as a diluted CEWAF (DCEWAF). Phytoplankton communities exposed to WAF showed no significant changes in PSII quantum yield (Fv/Fm) or electron transfer rates (ETRmax) compared to Control communities. In contrast, both Fv/Fm and ETRmax declined rapidly in communities treated with either CEWAF or DCEWAF. Analysis of other photophysiological parameters showed that photosystem II (PSII) antenna size and PSII connectivity factor were not altered by exposure to DCEWAF, suggesting that processes downstream of PSII were affected. The eukaryote community composition in each experimental tank was characterized at the end of the 72 h exposure time using 18S rRNA sequencing. Diatoms dominated the communities in both the control and WAF treatments (52 and 56% relative abundance respectively), while in CEWAF and DCEWAF treatments were dominated by heterotrophic Euglenozoa (51 and 84% respectively). Diatoms made up the largest relative contribution to the autotrophic eukaryote community in all treatments. EPS concentration was four times higher in CEWAF tanks compared to other treatments. Changes in particle size distributions (a proxy for aggregates) over time indicated that a higher degree of particle aggregation occurred in both the CEWAF and DCEWAF treatments than the WAF or Controls. Our results demonstrate that chemically dispersed oil has more negative impacts on photophysiology, phytoplankton community structure and aggregation dynamics than oil alone, with potential implications for export processes that affect the distribution and turnover of carbon and oil in the water column.

Influence of the magnesium content in apple leaves photosynthetic capacity

Fruit trees photosynthesis is determined by abiotic factors affecting crop productivity. The aim of this study was to evaluate the influence of magnesium nutritional status and exposure to solar radiation in the photosynthetic capacity of leaves of Malus domestica 'Cripp's Pink'. A two-factor experiment was conducted: high (+Mg) and low (-Mg) foliar Mg content (0.26 and 0.19%, respectively) in leaves were exposed to high (HR) and low (LR) solar radiation (1600 and 600 μmol m-2 s-1, respectively). Chlorophyll concentration, maximum quantum yield of photosystem-II (Fv/Fm) and net photosynthesis rate was determined in leaves. HR and LR -Mg leaves had 11.5 and 1.8% less chlorophyll concentration respectively than HR and LR +Mg leaves; no effects were observed in chlorophyll concentration by exposure. Fv/Fm was affected by sun exposition but not by Mg content. In HR leaves Fv/Fm was 5-6% lower than in LR leaves. Photosynthesis was affected by magnesium content when leaves where exposed to high solar radiation. Maximum net photosynthesis in HR +Mg leaves was 20.0 μmol m-2 s-1, while in HR -Mg was 11.1 μmol m-2 s-1. Net photosynthesis in LR leaves was not affected by Mg content. These results show that apple leaves photosynthetic capacity could be affected by lower Mg content when exposed to high solar radiation.

Effects of the heavy metal cadmium on photosynthetic activity and the xanthophyll cycle in Phaeodactylum tricornutum

Cadmium (Cd) is one of the most common and widespread heavy metals in the environment. Cd has adverse effects on photosynthesis that are countered by photosystem I (PSI) and photosystem II (PSII); however, the protective responses of these photosystems to heavy metal stress remain unclear. Using the model diatom Phaeodactylum tricornutum, a biological indicator that is widely used to assess the impact of environmental toxins, we simultaneously measured the effects of Cd on PSI and PSII and examined the levels of pigments in response to high light treatments before and after Cd exposure. Cd significantly reduced the quantum yield and electron transport rates of PSI and PSII. The quantum yield of non-photochemical energy dissipation in PSI due to donor side limitation increased faster than the quantum yield due to acceptor side limitation. The Cd treatment activated the P. tricornutum xanthophyll cycle under artificial light conditions, as indicated by an increased diatoxanthin content. Xanthophyll is important for photoprotection; therefore, the accumulation of diatoxanthin may down-regulate PSII activities to reduce oxidative damage. Together, our results suggest that the rapid reduction in PSII activities following Cd exposure is an adaptive response to heavy metal stress that reflects the variable exposure to external stressors in the native P. tricornutum environment.
