Metabolomic profiling of barley extracts obtained via different solvents and evaluation of their anti-inflammatory efficacy

Barley (Hordeum vulgare L.) stands out for its special nutritional value compared to other cereals, garnering significant attention in recent years. In this study, a comprehensive chemical and biological evaluation of different barley cultivars extracted with different solvents was performed. Ultra-high performance liquid chromatography coupled with electrospray ionization quadrupole time-of-flight tandem mass spectrometry (UHPLC-ESI-QTOF-MS/MS) was used in the phytochemical study. The molecular docking with absorption, distribution, metabolism, excretion, and toxicity (ADMET) study and multivariate analysis alongside the quantification of total phenolic contents (TPC) were performed. Furthermore, the anti-inflammatory action was in vitro assessed by measuring COX-1, COX-2, and 5-LOX inhibition. This study seeks to establish a suitable solvent for extracting anti-inflammatory properties from diverse barley cultivars. The methanol extract obtained from the G.131 cultivar, followed by the 80% methanol extract from the G.136 cultivar, exhibited the most favorable IC50 values in comparison to the reference drugs indomethacin and celecoxib. The phytochemical profile revealed more than 60 compounds that belonged to nitrogen-containing compounds, phenolic compounds, organic acids, flavonoids, and their glycosides, hordatines, and other classes. The TPC and COX-2 inhibition was positively correlated with isoscoparin, coumaroyle–OH–eagmatine, isoorientin, isovitexin 7-O-rhamnosylglucoside, isoscoparin 7-O-glucoside, and isoorientin-7-O-[6-feruloyl]-glucoside. These findings along with their pharmacodynamics study suggested a potential synergistic and significant contribution of these compounds with the anti-inflammatory activity.

Recovery Optimization of Plant Derived Antioxidants And Their Incorporation in Cosmetic Creams to Enhance Antioxidant Potential

Plants waste is enriched with valuable antioxidants. Extraction optimization, quantification of Total phenolic content (TPC) and evaluation of their anti-aging potential was the prime goal of this research study. Lime, Neem, Radish leaves, and carrot pulp were extracted with two types of solvents under various extraction conditions. The highest TPC were extracted through boiling water (2-3min) for all plant materials as follow: Neem leaves (112.3mg GAE/g), Lime leaves (108.5mg GAE/g), Radish leaves (76.02mg GAE/g) and Carrot pulp (65.84mg GAE/g); Highest free radical scavenging activities were achieved in Neem Leaves (92.82%), Lime leaves (92.34%), Radish leaves (87.4%) and Carrot Pulp (81.22%) under different extraction conditions. Water as extraction solvent yielded greater TPC values than ethanol. Upon the addition of phytoextracts, a substantial increase in the antioxidant activity of the cream samples was detected. Upon loading 2% lime extract, one cream sample exhibited a rise in TPC content from 1.8 to 54.05mg GAE/g and antioxidant activity from 19.34% to 95.35%. Cream samples infused with phytoextracts also showed notable antimicrobial activities. Conclusively, waste derived phytoextracts can be cost-effectively utilized in formulation of skin anti-aging creams.

In vitro antioxidant activity and total phenolic content of Digera muricata leaves

In the present study quantification of total phenolic content and evaluation of in vitro antioxidant capability of five different extracts of Digera muricata was carried out by different assays. DPPH radical, superoxide radical, hydroxyl radical scavenging activity and metal chelating activity were calculated and compared with standard antioxidants. Reducing power of the extract was also determined. All extracts showed good free radical scavenging ability which gets enhanced with increasing concentration. Methanolic extract possessed highest scavenging ability and it has maximum phenolic content (41.87 mg/g GAE). Significant correlation was observed between antioxidant assays and total phenolic content indicating that phenolics may be contributing towards antioxidant activity. The results conclude that D. muricata extracts are a potential source of natural antioxidants and could show promise as therapeutic agent in preventing the progression of oxidative stress related degenerative disorders.

Comparative Methods to Evaluate the Antioxidant Capacity of Propolis: An Attempt to Explain the Differences.

Propolis is a natural product produced by bees that contains a complex mixture of compounds, including phenolic compounds and flavonoids. These compounds contribute to its biological activities, such as antioxidant capacity. This study analysed the pollen profile, total phenolic content (TPC), antioxidant properties, and phenolic compound profile of four propolis samples from Portugal. The total phenolic compounds in the samples were determined by six different techniques: four different Folin-Ciocalteu (F-C) methods, spectrophotometry (SPECT), and voltammetry (SWV). Of the six methods, SPECT allowed the highest quantification, while SWV achieved the lowest. The mean TPC values for these methods were 422 ± 98 and 47 ± 11 mg GAE/g sample, respectively. Antioxidant capacity was determined by four different methods: DPPH, FRAP, original ferrocyanide (OFec), and modified ferrocyanide (MFec). The MFec method gave the highest antioxidant capacity for all samples, followed by the DPPH method. The study also investigated the correlation between TPC and antioxidant capacity with the presence of hydroxybenzoic acid (HBA), hydroxycinnamic acid (HCA), and flavonoids (FLAV) in propolis samples. The results showed that the concentrations of specific compounds in propolis samples can significantly impact their antioxidant capacity and TPC quantification. Analysis of the profile of phenolic compounds by the UHPLC-DAD-ESI-MS technique identified chrysin, caffeic acid isoprenyl ester, pinocembrin, galangin, pinobanksin-3-O-acetate, and caffeic acid phenyl ester as the major compounds in the four propolis samples. In conclusion, this study shows the importance of the choice of method for determining TPC and antioxidant activity in samples and the contribution of HBA and HCA content to their quantification.

Persuasive and Strategic Approaches for Preliminary Phytochemical Screening, Quantitative Analysis and Antioxidant Activity of Crude Plant Extracts from Clematis napaulensis DC Indigenous to Nagaland, India

The present study is designed to extract and investigate the bioactive phytoconstituents, their secondary metabolites, and antioxidant potential from the leaf extracts of Clematis napaulensis belonging to the family Ranunculaceae. Extract was investigated for its preliminary phytoconstituents by chemical analysis method. Quantification of total phenolic content by Folin-Ciocalteau assay; Total Flavonoid Content and Total Triterpenoid Content (TTC) were quantified by colorimetric method. Antioxidant potential was evaluated in vitro using Phosphomolybdenum assay, Hydroxyl radical scavenging activity, Reducing power assay, and the DPPH method. The preliminary phytochemical screening results suggested that a wide variety of pharmacologically active phytoconstituents such as phenolic compounds, alkaloids, glycosides, flavonoids, saponins, tannins, triterpenoids, gums, proteins, and reducing sugars were present whereas non-reducing sugar was absent. The total phenolic content in the methanolic extract was 0.8308 μg/mL of gallic acid equivalent; the total flavonoid content of the plant was 1.3850 μg/mL of quercetin equivalent and the total triterpenoid content was found to be 1.414 μg/mL of oleanolic acid equivalent. In vitro antioxidant studies revealed that Clematis napaulensis possess good antioxidant potential. This is the first report of Clematis napaulensis, the phytochemical screening results suggested that the extract would be a potential alternative also it can be explored to discover and development of new chemical moieties for treating various diseases.

Organic solvent extraction and spectrophotometric quantification of total phenolic content of soil

Phenolic compounds are regarded as the most abundant plant metabolites that are known to decompose progressively into soils, likened to other soil organic materials. Once assimilated into soils, they can control soil processes, including organic matter decomposition and nutrient cycling. Established that phenolic compounds can influence nutrients availability and soil quality, it becomes crucial to investigate into soil phenolics through the application of appropriate extraction technique and quantification of ​total phenolic content ​in soils. This study therefore aimed at utilizing ethanol, hexane and diethyl ether organic solvents to extract and quantify total phenolic content of soil, sampled from a vegetable growing area. Conventional organic solvent extraction method was employed to extract phenolics, while spectrophotometric technique was utilized to quantify total phenolic content. The highest extraction yield of 34.52% was achieved with ethanol followed by diethyl ether (28.23%) and hexane (25.47%). Interestingly, hexane, which had the least extraction yield, rather recorded the highest phenolics concentration of 5.50 ± 0.02 mgGAE/g, with ethanol producing a concentration of 2.04 ± 0.05 mgGAE/g and 3.82 ± 0.01 mgGAE/g for diethyl ether. The percent recovery, limit of detection (LOD) and limit of quantification (LOQ) of phenolic compounds were found to be 102%, 0.8 mg/g and 1.5 mg/g for ethanol; 96%, 0.6 mg/g and 1.2 mg/g for diethyl ether and 94%, 0.4 mg/g and 1.0 mg/g for hexane respectively. These results indicate that for an extraction efficiency and greater yield, the use of ethanol as solvent is preferred whereas extraction using hexane is suitable for total phenolics quantification. The findings of this study have provided a vital insight regarding the influence of organic solvents on the extractability and quantification of total phenolic content of soil.

Antihyperglycemic effect and phytochemical investigation of Rubia cordifolia (Indian Madder) leaves extract

Abstract Medicinal plants are used as an important source of medicines in pharmaceutical industry. Rubia cordifolia is widely used to cure diabetes mellitus. Present study was aimed to investigate the antihyperglycemic effects of different fractions of R. cordifolia leaves and to analyze its antioxidant effect and phytochemical composition. Male albino mice were randomly distributed into seven groups (n = 7). Group-I was normal control, group-II was Alloxan (100 mg/kg)-induced diabetic control, and group-III was standard drug (Glibenclamide 0.5 mg/kg)-treated group. Animals in groups IV–VII were treated with n-hexane fraction, ethyl acetate fraction, n-butanol fraction and aqueous fraction of R. cordifolia, orally administered (100 mg/kg) once daily up to 28 days after Alloxan induction, respectively. Methanolic extract (ME) and fractions of R. cordifilia were analyzed for antioxidant activity and quantification of total phenolic content and total flavonoid content. HPLC of ME and most active fractions were performed. The results showed that RCEF (G-V) and RCBF (G-VI) have significantly (P < 0.05) reduced the increased level of glucose as compared to toxicant control group. It was further revealed that EF and BF have higher antioxidant activity (having IC50 34.9, 36.86 (µg/mL)) owing to phenolic and flavonoid identified by HPLC.

GC-MS Analysis and <i>In Vitro</i> Antioxidant Activity of Crude Extracts and Different Fractions of Moringa Oleifera Pods

Moringa oleifera (M. oleifera) has been known as a medicinal plant. The objectives of this study is to identify the chemical compositions of M. oleifera pods extracts and to evaluate its total phenolic content (TPC) and antioxidant activity. The methodology of this study began with the collection of the pod's sample, drying and grinding the pods into powdered form. M. oleifera pods extracts were prepared by using methanol (MeOH), hexane, dichloromethane (DCM), and ethyl acetate (EtOAc) solvent which yielded respective residues. All extracts were subjected to Gas Chromatography-Mass Spectrometry (GC-MS) to identify its chemical compositions. Major compound found in hexane, DCM, EtOAc, and MeOH extracts were nonanoic acid (30.94 %), diisooctyl phthalate (89.45 %), benzoic acid, 4-hydroxy- (37.57 %) and octanoic acid (19.19 %), respectively. The TPC quantification was done via Folin-Ciocalteu method showed that EtOAc extract has the highest phenols content with 500.05 mg GAE/g. Generally, all extract contain moderate phenolic content. M. oleifera pods extracts were screened for the antioxidant activity via 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical. Hexane, DCM and EtOAc extracts exhibited good free radical scavenging activity with IC50 of 65.79 μg/mL, 74.93 μg/mL and 82.83 μg/mL, respectively. MeOH showed the lowest free radical scavenging activity with IC50 529.6 μg/mL. However, antioxidant activity of all extracts was low compared to standard, ascorbic acid (AA) with IC50 of 19.57 μg/mL. The presence of various bioactive constituents in the M. oleifera pods extracts might contributed to the antioxidant activity of the plant.

Study of Alcoholic Extract of Acacia Farnesiana (L.) Willd Pods

Identification of bioactive compound from alcoholic extract of Acacia farnesiana leguminosae pods by using preliminary phytochemical test or thin layer chromatography and the quantification of total phenolic content by folin-ciocalteu reagent method. shade dried grounded powder of Acacia farnesiana pods was successively extracted with petroleum ether, chloroform and alcohol in soxhlet apparatus, alcoholic compound obtained in more amount as compare to other two extract so alcoholic extract was used for identification of bioactive compound. The alcoholic extract of leguminosae pods indicates the presence of major bioactive compound. Analysis of the alcoholic extract by TLC have identified naringenin from extract, and the total phenolic content in alcoholic extract was found to be 22%(w/w). Therefore the present study deals with qualitative analysis of alcoholic extract of legumae pericarp (pod wall) of Acacia farnesiana L. In which we analyze various phytochemical which are useful for contoling various diseases TLC method used for identification of the content of naringenin from active extract of Acacia farnesiana pods. It is concluded that, legume pods contain maximum phytoconstituents or phenolic content.

Induction, Multiplication, and Evaluation of Antioxidant Activity of Polyalthia bullata Callus, a Woody Medicinal Plant.

Polyalthia bullata is an endangered medicinal plant species. Hence, establishment of P. bullata callus culture is hoped to assist in mass production of secondary metabolites. Leaf and midrib were explants for callus induction. Both of them were cultured on Murashige and Skoog (MS) and Woody Plant Medium (WPM) containing different types and concentrations of auxins (2,4-dichlorophenoxyacetic acid (2,4-D), α-naphthaleneacetic acid (NAA), picloram, and dicamba). The callus produced was further multiplied on MS and WPM supplemented with different concentrations of 2,4-D, NAA, picloram, dicamba, indole-3-acetic acid (IAA), and indole-3-butyric acid (IBA) media. The quantification of total phenolic content (TPC), total flavonoid content (TFC) and antioxidant capacity was further carried out on P. bullata callus, and the results were subjected to correlation analysis. Among the media, the WPM + 16.56 µM picloram (53.33 ± 22.06%) was the best for callus induction while MS + 30 µM dicamba was the best for callus multiplication. The TPC, TFC, and EC50 of DPPH scavenging activity were determined at 0.657 ± 0.07 mg GAE/g FW, 0.491 ± 0.03 mg QE/g, and 85.59 ± 6.09 µg/mL in P. bullata callus, respectively. The positive correlation between DPPH scavenging activity with TPC was determined at r = 0.869, and that of TFC was at r = 0.904. Hence, the P. bullata callus has an ability to accumulate antioxidants. It therefore can be a medium for secondary metabolites production.

Phytochemical screening, polyphenols, flavonoids and tannin content, antioxidant activities and FTIR characterization of Marrubium vulgare L. from 2 different localities of Northeast of Morocco

Chemical compositions, biological and antioxidant activities of plants are widely affected by several parameters and conditions, such as geographical and climatic conditions, type of extract (aqueous or organic), as well as the polarity of the extracting solvent. Therefore the present study was the first one designed to study the phytochemical composition, the content of polyphenols, tannins and flavonoids, the antioxidant activities and the chemical composition analysis by FTIR spectroscopic of organic (ethanol, methanol, ethyl Acetate, petroleum ether) and aqueous extracts of Marrubium vulgare L. leaves, collected from two different sampling localities in the North-East of Morocco: Oulad Daoud Zkhanine and the Cape Three Forks. A phytochemical screening was carried out by specific coloring and precipitation reactions. The colorimetric method Folin- Ciocalteu was used for the quantification of total phenolic content. The method of aluminum chloride was employed for the quantification of total flavonoid content and the method of vanillin for the determination of tannins. The antioxidant power was evaluated by the DPPH and ABTS methods. The chemical composition of the organic extracts was analyzed by the FTIR spectroscopy method. Depending on the sampling location of M.vulgare L., the type of extract (aqueous or organic), the polarity of the extracting solvent, and the phytochemical screening revealed the presence of the following secondary metabolites: catechic tannins, terpenoids, polyphenols and flavonoids. The total concentrations of total polyphenols, flavonoids and tannins varied respectively between 0.27 ± 0.1 and 86.91 ± 1.22 μg gallic acid equivalents/mg,6.08 ± 0.17 and 33.82 ± 0.90 μg quercetin equivalents/mg and 2.73 ± 1.15 and 252.68 ± 4.50 μg catechin equivalents/mg. The antioxidant activity that was evaluated by DPPH and ABTS method showed that ethanol extract, methanol and ethyl acetate extract had the highest percentages of inhibition, unlike petroleum ether extract. The inhibitory concentrations (IC50) ranged from 324.55 ± 0.66 to 980 ± 0.62 μg/ml for DPPH and from 107.85 ± 0.19 to 890.74 ± 0.17 μg/ml for ABTS. FTIR spectroscopic analysis has revealed different characteristic peak values with various functional groups in the extracts such as amide, alcohol, phenol compounds. In general, the organic and aqueous extracts of M.vulgare L. that were harvested from Oulad Daoud Zkhanin were richer in secondary metabolites, and showed higher concentrations of polyphenol, flavonoids and tannins. In addition, they revealed a higher antioxidant capacity than the extracts of M. vulgare L.from the Cape Three Forks.Overall this study highlighted the potential benefits and richness of M.vulgare L. harvested from the two study areas and suggested it as a potential source of natural antioxidants that could be used in the food and pharmaceutical fields.

In vitro anti-inflammatory, antioxidant and antibacterial activities of the Algerian species Micropus bombycinus

The present study reports the in vitro pharmacological properties of the methanolic extract obtained from the species Micropus bombycinus Lag. The quantification of total phenolic content was performed using the Folin-Ciocalteu method. The antioxidant activity was evaluated by four different assays. Moreover, the in vitro anti-inflammatory activity was assessed using the albumin denaturation method and the antibacterial activity was estimated by agar disk diffusion assay against ten bacterial strains. The Micropus bombycinus methanolic extract contains several classes of secondary metabolites known for their biological properties. A high content of polyphenols was estimated (115.32 ± 0.47 μg GAE/ mg of plant extract). The MeOH extract possessed strong antioxidant activities in various assays that subsequently correlated with its phenolic content. It had also the ability to inhibit thermally-induced protein denaturation with a percentage of 27.51% at the concentration of 400 μg/mL. Furthermore, this extract showed antibacterial activity against all the tested strains with zones of inhibition ranging from 9 to 28 mm. It can be concluded that Micropus bombycinus could be an interesting source for potential bioactive compounds as inhibitors of pathogenic bacteria, and it helps to cure chronic inflammatory (Rheumatoid arthritis) and oxidative stress-related diseases.

Physicochemical characterization, fatty acid profile, antioxidant activityand antibacterial potential of cacay oil, coconut oil and cacay butter.

The Amazon region is rich in genetic resources such as oilseeds which have potentially important local commercial exploitation. Despite its high concentration of bioactive compounds, cacay (Caryodendron orinocense Karst.) oil is poorly investigated and explored. Thus, this study focuses on the physicochemical characterization (moisture, density, and saponification, iodine, and acidity values), fatty acid composition as determined by gas chromatograph mass spectrometry (GC/MS), total phenolic content (TPC), and antioxidant activity (DPPH and ABTS radical scavenging assay) of cacay oil, coconut oil and a coconut/cacay oil blend, also known as cacay butter. The antibacterial activity of cacay oil was additionally evaluated. Our study demonstrated that cacay oil presents a high amount of polyunsaturated fatty acid (PUFA) (58.3%) with an emphasis on linoleic acid and a lower acidity value (2.67 ± 0.01 cg I2/g) than butter and coconut oil, indicating a low concentration of free fatty acids. In contrast, cacay butter and coconut oil presented higher saturated fatty acid percentages (69.1% and 78.4%, respectively) and higher saponification values (242.78 and 252.22 mg KOH/g, respectively). The samples showed low moisture and relative density between 912 and 916 kg/m3. The hydrophilic fraction of cacay oil was highlighted in the quantification of TPC (326.27 ± 6.79 mg GAE/kg) and antioxidant capacity in vitro by DPPH radical scavenging assay (156.57 ± 2.25 μmol TE/g). Cacay oil inhibited the growth of Bacillus cereus (44.99 ± 7.68%), Enterococcus faecalis (27.76 ± 0.00%), and Staphylococcus aureus (11.81 ± 3.75%). At long last, this is the first study reporting the physicochemical characterization and bioactive properties of cacay butter. Coconut oil and cacay butter showed great oxidative stability potential due to higher contents of saturated fatty acids. Moreover, cacay oil presents as an alternative source of raw materials for cosmetic and biotechnology industries due to its high concentration of PUFA and for being a rich source of phenolic compounds.

Drying Effects on Phenolics and Free Radical-Scavenging Capacity of Rhus pachyrrhachis and Rhus virens Used in Traditional Medicine.

Rhus pachyrrhachis and Rhus virens are medicinal plant species with important uses in northeastern Mexico. They belong to a complex of Rhus species called “lantriscos”, which are used for medicinal applications. The medicinal effects of these species are based on traditional use, however, they require phytochemical research to validate their medicinal properties, as well as structural characterization for their correct identification during the collecting practice and uses. The phytochemical potential of aqueous extracts from R. pachyrrhachis and R. virens was analyzed by the quantification of total phenolic content (TPC), free radical-scavenging potential, and total flavonoids, with a comparison of four drying methods, and some phenolic compounds were identified. Furthermore, the stems and leaves of both species were anatomically characterized to establish a differentiation. R. pachyrrhachis and R. virens showed similar values of phytochemical contents, although the TPC content (0.17 mg of gallic acid equivalent per gram of dry weight, GAE/g DW) was higher in R. virens. The drying method used affected the metabolite contents, and this behavior was related to the species. Regarding the phenolic compounds, shikimic acid, galloylquinic acid, and gallic acid were identified in both species, however, quinic acid was only found in Rhus pachyrrhachis, while vanillic acid O-hexoside was identified only in Rhus virens. At the anatomical level, the pubescence associated with trichomes on the leaves of Rhus pachyrrhachis was highlighted as the main differential characteristic.

Antioxidant Potential and Angiotensin-Converting Enzyme (ACE) Inhibitory Activity of Orotic Acid-Loaded Gum Arabic Nanoparticles.

Orotic acid (OA) nanoparticles were prepared using the freeze-drying method. The antihypertensive activity and antioxidant capacity of OA and orotic acid-loaded gum arabic nanoparticles (OAGANPs) were examined using the angiotensin-converting enzyme (ACE), 1,1-diphenyl-2-picrylhydrazyl (DPPH), nitric oxide (NO), and β-carotene assays, as well as the quantification of total phenolic content (TPC). The DPPH and NO scavenging activities of OAGANPs were significantly higher than those of the OA solution. The β-carotene bleaching assay of OAGANPs showed a dose-dependent trend, while 500μg/ml was significantly more effective than the other concentrations, which exerted 63.4% of the antioxidant activity. The in vitro antihypertensive assay revealed that the OAGANPs exhibited the most potent ACE inhibition activity, when compared to the OA solution. Hence, results revealed the potential of preparing the OA as a nanoparticle formulation in enhancing the antioxidant and antihypertensive properties compared to the OA solution.

Quantification of Total Phenolic and Flavonoid content in leaves of Cucumis melo var agrestis using UV- spectrophotometer

Plants and plant-derived products contain a wide range of phytochemicals, such as alkaloids, flavanoids, glycosides which are thought to have the medicinal properties. Cucumis melo var agrestis is belongs to the family of cucurbitaceae and well distributed in rural and coastal areas. This plant is consumed by rural people in Tamil Nadu, India. Phenolic and flavanoid contents plays the important role in antioxidant mechanism thereby cure many of the diseases. Therefore current research is aimed to quantification of total phenolic and flavanoid content present in leaves of CMA. Hydroalcoholic leaf extract of CMA (HALEC) was evaluated its phytochemicals using chemical tests and quantification of total phenolic content and total flavonoid contents by standard curve of Gallic acid and Quercetin using UV- Spectrophotometer. Our results revealed that HALEC has the phytochemicals like alkaloids, flavonoids, tannins, carbohydrates, saponins, glycosides, proteins and amino acids and quantitative tests confirms the presence of total phenolic content was 77.82 mg/g of extract calculated as Gallic acid equivalent and total flavonoid content was 30.06 mg/g of extract calculated as Gallic acid equivalent in HALEC. This research work is useful to animal study on leaf extract of CMA, Isolation of active principles and detection of exact mechanism.

Potentilla erecta (L.) rhizomes as a source of phenolic acids

The aim of this study was to evaluate the content of major phenolic acids from Potentilla erecta rhiozomes. Water and ethanol-water mixture was used for extraction of these compounds. The extracts were also evaluated for the quantification of total phenolic content and the antioxidant capacity. The contents of phenolic acids and resulting antioxidant activities are dependent on the nature of extracting solvent due to the presence of different antioxidant compounds. Results showed that P. erecta rhiozomes contained high amount of gallic and p-HBA acids. The contents of chlorogenic and protocatechuic acids in the extracts of Potentilla species have not been reported yet. The results suggested that the extracts could be used as the active cosmetics ingredients and nutraceuticals.

Determination of Total Phenolic Compounds and Antioxidant Activity of Ethanolic Extracts of Propolis Using ATR–FT-IR Spectroscopy and Chemometrics

Fourier transform mid-infrared spectroscopy equipped with attenuated total reflectance (FT-IR–ATR) combined to partial least squares (PLS) regression was used for the quantification of total phenolic contents (TPCs) and antioxidant activities in 98 samples of ethanolic extract of propolis (EEP) from the southwest region of Parana, Brazil. The Pearson’s correlation coefficients were applied, and results ranged from 0.96 to 0.88 and showed higher correlation coefficients among TPC and ferric-reducing antioxidant power (FRAP) followed by 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) ABTS and 2,2-diphenyl-1-picrylhydrazyl hydrate (DPPH). Calibration was performed using a Savitzky-Golay filter (15 pt) and first derivative as well as standard normal variate (SNV) and mean center correction pretreatments. The determination coefficient in the calibration models ranged from 0.95 to 0.87. The range error ratio (RER) indicates the quality of estimation of the models and the results obtained were 10.0, 8.11, 16.8, and 8.99 for TPC, DPPH, ABTS, and FRAP, respectively. Thus, the results obtained for calibration and prediction parameters indicated that the models for DPPH, FRAP, and TPC have a low predictive capacity which complicates the data modeling. However, the ABTS model is validated and can be used for quantification of antioxidant activity of new extracts of propolis, being useful as an alternative to rapid analysis, reducing waste generation and cost, and indicating that the mid-infrared spectroscopy associated with PLS regression can be used to predict ABTS radical scavenger.

Antioxidant and hepatoprotective potentials of novel endophytic fungus Achaetomium sp., from Euphorbia hirta

To isolate, identify and evaluate the antioxidant, antimicrobial, hepatoprotective potentials, total phenolic content, flavonoid content, tannin content of ethyl acetate extract of endophytic fungus Achaetomium sp., isolated from Euphorbia hirta. Hepatoprotectivity of ethyl acetate extract of Achaetomium sp., was evaluated by CCl4 induced toxicity in HepG2 cells and subsequently analysed for cell viability using MTT assay. It also demonstrates antioxidant and antimicrobial potentials by DPPH radical scavenging assay and well diffusion assay respectively. Quantification of total phenolic content, tannin content and flavonoid content were assessed by spectroscopic methods. Phenols, flavonoids and tannins were the phytochemicals present in ethyl acetate extract of Achaetomium sp., with rich phenolic content exhibited potent hepatoprotective, antimicrobial and antioxidant activities. The hepatoprotective activity was recorded as of 72.13%±2.948% of cell viability at a concentration of 150μg/mL, whereas the standard silymarin showed 93.260%±0.784%. It was observed to be dose dependent, when CCl4 exposed HepG2 cells were treated with different concentrations of ethyl acetate extract. Antibacterial activity showed significant inhibition against Staphylococcus aureus, Pseudomonas aeroginosa, and Klebsiella pneumoniae. The antioxidant activity ranged from 66.890%±1.385% to 87.340%±0.289% with (44.02±1.57) μg of total phenolics, (54.54±1.82) μg of flavonoid content and (18.790±1.018) μg of tannin content. Ascorbic acid, BHT (butylated hydroxyl toluene) Gallic acid and Pyrogallol were used as standards which showed 98.370%±0.763%; 97.080%±0.636%; 94.890%±1.103% and 96.980%±0.098% reducing potential respectively. The results reveal that the metabolites produced by endophytic fungi isolated from Euphorbia hirta could be novel natural products that could lead to new drug discovery.

Antioxidant activity and total phenolic and flavonoids content variations of leaves extracts of white Horehound (Marrubium vulgare Linné) from three geographical origins.

To elucidate the effect of the sampling location of Marrubium vulgare L.leaves on phenolic contents and antioxidant proprieties of flavonoids extracts. M.vulgare L.leaves were collected from three different geographical locations belonging to northwest Algeria: Tessala (mountain region), M'sila forest (coastal region), and Ain Skhouna (steppe region). The flavonoid extraction was achieved using organic solvents with different polarities (methanol, chloroform, ethyl acetate, and hexane). Folin-Ciocalteu colorimetric method was used for quantification of total phenolic contents, and aluminum chloride assay for quantification of total flavonoid contents. The antioxidant properties of flavonoids extract were studied by free l,l-diphenyl-2-picrylhydrazyl radical-scavenging technique. Total phenolic and flavonoids concentrations varied respectively between 40.7 and 160mg gallic acid equivalents/g and 27.4 and 66.3mg catechin equivalents/g. The DPPH free radical-scavenging activity shows that the antioxidant activity of the flavonoid extracts varied significantly (P<0.001) depending on the type of the organic solvent used, and the sampling location. The methanol, chloroform and ethyl acetate extracts exhibited the highest percentages of inhibition unlike to the aqueous and hexane extracts. These percentages are ranged from 54.8 to 98.8% at 1000μg/mL. In general, M'sila forest flavonoids extracts showed the highestfree radical inhibition capacity; followed by those of Ain Skhouna and Tessala Mountain. The inhibitory concentration 50 (IC50) ranged from 33.7 to 774μg/mL and often exceeded those recorded by phenolic standards (ascorbic acid, gallic acid, caffeic acid, tannic acid and catechin). The phytochemical screening revealed the presence of some flavonoid classes, such as flavans and flavanols. The results suggested a potent antioxidant activity of M.vulgare flavonoids extracts, which may find its application in feature research for the food and the pharmacological industries.
