Buttressing Effect as a Key Design Principle towards Highly Efficient Palladium/N-Heterocyclic Carbene Buchwald-Hartwig Amination Catalysts.

The backbone substitution of the standard 1,3-bis(2,6-diisopropylphenyl)-2H-imidazol-2-ylidene (IPr) ligand by dimethylamino groups was previously shown to induce a dramatic improvement in the catalytic efficiency of the corresponding Pd-PEPPSI (pyridine-enhanced pre-catalyst preparation, stabilization, and initiation) pre-catalysts in N-arylation reactions. Herein, a thorough structure/activity study towards rationalizing this beneficial effect has been described. In addition to the previously reported IPrNMe2 and IPr(NMe2)2 ligands, the new IPrNiPr2 and IPr(NMe2,Cl) ligands, which bear one bulkier diisopropylamino group and a combination of dimethylamino and chloro substituents, respectively, have been designed and analyzed in the study. The influence of the backbone substitution was found to be steric in origin and is related to the well-known buttressing effect encountered in arene chemistry. The usefulness and versatility of this approach was demonstrated through the development of a highly efficient catalytic system for the challenging arylation of bulky α,α,α-trisubstituted primary amines. The optimized system based on the [PdCl(η3 -cinnamyl)(IPr(NMe2)2 )] or [PdCl(η3 -cinnamyl)(IPrNiPr2 )] pre-catalysts operates under unprecedented mild conditions (catalyst loadings: 0.5-2 mol %, reaction temperatures: 40-60 °C) with a wide substrate scope.

New compounds based on a benzimidazole nucleus: synthesis, characterization and cytotoxic activity against breast and colon cancer cell lines

Benzimidazolium salts and their Pyridine Enhanced Precatalyst Preparation Stabilization and Initiation (PEPPSI) palladium N-heterocyclic carbene (Pd-NHC) based complexes have been synthesized and their structures characterized with a number of different instrumental techniques including NMR (1H and 13C), IR, EI-MS (for 2), X-ray (for 1, 2 and 4) and elemental analysis. The cytotoxicity of all the compounds was tested using the human embryonic kidney (HEK-293T), human breast epithelial adenocarcinoma (MDA-MB-231), and human colon epithelial colorectal adenocarcinoma (DLD-1) cell lines. The benzimidazolium salts (2–5) had more cytotoxic activity against cancerous cells compared with the metal complexes (6–9), which curiously exhibited no activity against any of the cell lines. Based on the IC50 values, compound 5 displayed the highest in vitro anticancer activity among compounds 2–9.

Flexible Steric Bulky Bis(Imino)acenaphthene (BIAN)-Supported N-Heterocyclic Carbene Palladium Precatalysts: Catalytic Application in Buchwald-Hartwig Amination in Air.

To achieve efficient palladium-catalyzed cross-coupling reaction under mild reaction conditions with the flexible steric bulk strategy, a series of Pd-PEPPSI (PEPPSI: pyridine-enhanced precatalyst preparation, stabilization, and initiation) complexes C1-C6 were synthesized and characterized, in which unsymmetric flexible steric bulk was introduced on the N-aryl of ancenaphthyl skeleton. These well-defined palladium complexes were found to be excellent precatalysts for Buchwald-Hartwig amination of aryl chlorides with amines in air. The electronic effect of the Pd-PEPPSI complexes and the effect of ancillary pyridine ligands were evaluated, among which complex C3 exhibited the most efficiency. It was demonstrated that the cross-coupling products were obtained in excellent yields in the presence of 0.5-0.1 mol % palladium loading. A wide range of aryl- and heteroaryl chlorides as well as various amines were compatible. The oxidative addition of aryl chlorides is revealed to be the rate-determining step in the catalytic cycle. The catalytic activity can be enhanced by introducing electron-donating groups to the Pd-PEPPSI complexes. This type of Pd-PEPPSI precatalyst showed the most efficiency reported to date for the challenging C-N cross-coupling reactions requiring no anhydrous and inert atmosphere protections, suggesting flexible steric bulk as a promising catalyst design strategy.

N-BUTYL SUBSTITUTED N-HETEROCYCLIC CARBENE-PD(II)-PYRIDINE (PEPPSI) COMPLEXES: SYNTHESIS, CHARACTERIZATION AND CATALYTIC ACTIVITY IN THE SUZUKI-MIYAURA REACTION

A series of N-butyl substituted imidazolium salts, ( 1a-c ) and their pyridine enhanced precatalyst preparation stabilization and initiation ( PEPPSI ) themed palladium N -heterocyclic carbene complexes ( 2a-c ) were synthesized and characterized. Pd-NHC complexes were fully determined by elemental analysis and spectroscopic. The synthesized complexes were tested in Suzuki-Miyaura cross-coupling reaction. These complexes were found to be efficient catalysts for the Suzuki-Miyaura reaction of phenylboronic acid with aryl bromides.

N-Methylphthalimide-substituted benzimidazolium salts and PEPPSI Pd-NHC complexes: synthesis, characterization and catalytic activity in carbon-carbon bond-forming reactions.

A series of novel benzimidazolium salts (1–4) and their pyridine enhanced precatalyst preparation stabilization and initiation (PEPPSI) themed palladium N-heterocyclic carbene complexes [PdCl2(NHC)(Py)] (5–8), where NHC = 1-(N-methylphthalimide)-3-alkylbenzimidazolin-2-ylidene and Py = 3-chloropyridine, were synthesized and characterized by means of 1H and 13C{1H} NMR, UV–vis (for 5–8), ESI-FTICR-MS (for 2, 4, 6–8) and FTIR spectroscopic methods and elemental analysis. The synthesized compounds were tested in Suzuki–Miyaura cross-coupling (for 1–8) and arylation (for 5–8) reactions. As catalysts, they demonstrated a highly efficient route for the formation of asymmetric biaryl compounds even though they were used in very low loading. For example, all compounds displayed good catalytic activity for the C–C bond formation of 4-tert-butylphenylboronic acid with 4-chlorotoluene.

An alternative approach to PEPPSI catalysts: from palladium isonitriles to highly active unsymmetrically substituted PEPPSI catalysts.

A series of new pyridine-enhanced precatalyst preparation, stabilization, and initiation (PEPPSI)-type complexes bearing different types of carbene ligands was prepared by the modular and convergent template synthesis strategy. Nitrogen acyclic carbenes, saturated and unsaturated five-membered NHC, saturated six-membered NHCs, and five-membered N-heterocyclic oxo-carbene (NHOC) ligands on palladium were prepared this way. These new organometallic compounds then were tested in Suzuki and Negishi cross-coupling reactions by using substrates with one or two substituents in ortho-position of the new CC bond being formed. Both aryl chlorides and bromides were tested as coupling partners. In some cases, the new ligands gave results similar to Organ's successful IPr-based and IPent-based PEPPSI derivatives, with aryl bromides 0.05 mol % catalyst load still gave satisfactory results, with aryl chlorides 0.5 mol % were needed.

PEPPSI‐Effect on Suzuki–Miyaura Reactions Using 4,5‐Dicyano‐1,3‐dimesitylimidazol‐2‐ylidene‐Palladium Complexes: A Comparison between trans‐Ligands

AbstractThe PEPPSI (Pyridine Enhanced Precatalyst Preparation, Stabilization and Initiation) complexes 12–15 with the structure [PdCl2{(CN)2IMes}(3‐R‐py)] (12: R = H; 13: R = Cl; 14: R = Br; 15: R = CN) bearing the maleonitrile‐based N‐heterocyclic carbene (NHC) (CN)2IMes ({(CN)2IMes}: 4,5‐dicyano‐1,3‐dimesitylimidazol‐2‐ylidene) were prepared. Solid state structures of 14 and 15 were obtained. Complexes 14 and 15 adopt a slightly distorted square‐planar coordination geometry in the solid state with the substituted pyridine ligand trans to the NHC. Catalytic activities of precatalysts 12–15 were studied and subsequently compared to complexes [PdCl2{(CN)2IMes}(PPh3)] (4) and [PdCl(dmba){(CN)2IMes}] (5) recently reported by our group in the Suzuki–Miyaura reaction of various aryl halides and phenylboronic acid. Reactions using previously reported [PdCl2(IMes)(py)] (IMes: 1,3‐dimesitylimidazol‐2‐ylidene) (1) were also carried out and their results contrasted to those involving 12–15, 4 and 5. Differences in initiation rates and the catalytically active species related to the seven complexes in regards to the “throw away ligand” were investigated. Poisoning experiments with mercury show that palladium nanoparticles are responsible for the catalytic activity.

Efficient PEPPSI-Themed Palladium N-Heterocyclic Carbene Precatalysts for the Mizoroki–Heck Reaction

Two sets of six PEPPSI (pyridine enhanced precatalyst preparation, stabilization, and initiation) themed palladium complexes of amide-functionalized N-heterocyclic carbenes [PdBr2(NHC)(Py)], where NHC = 1-acetamido-3-R-imidazolin-2-ylidene, 1-acetamido-3-R-benzimidazolin-2-ylidene and Py = pyridine, were prepared. Solid-state structures of all these complexes were determined by single-crystal X-ray diffraction methods. All complexes under scrutiny were found to adopt a trans arrangement with square-planar geometry. These complexes showed excellent catalytic activity toward the Mizoroki–Heck cross-coupling reaction of aryl chlorides and styrene. Benzimidazole-derived complexes exhibited better catalytic activity than imidazole-based complexes. Palladium complexes of the N-fused heterocycle derived abnormal N-heterocyclic carbenes for the much-preferred Cu-free and the amine-free Sonogashira coupling in air

Abnormal N-heterocyclic carbenes of the N-fused heterocycles were employed in stabilizing a series of PEPPSI (Pyridine Enhanced Precatalyst Preparation Stabilization and Initiation) themed complexes of the type (abnormal-NHC)PdI2(pyridine). Specifically, imidazo[1,2-a]pyridine based abnormal complexes, trans-{(1-ethyl-2-(4-R-phenyl)-imidazol-3-ylidene[1,2-a]pyridine)}PdI2(pyridine) [R=F (1b), Cl (2b), Br (3b) and Me (4b)] were synthesized from the reaction of corresponding imidazo[1,2-a]pyridinium iodide salts (1–4)a with PdCl2 in pyridine in the presence of K2CO3 as a base. The (1–4)b complexes carried out the Sonogashira couplings of the aryl bromide and iodide substrates with terminal alkynes in a mixed aqueous medium in air under the Cu-free and amine-free conditions. The density functional theory studies revealed that in the (1–4)b complexes, the Ccarbene–Pd σ-bonding interaction was significantly stronger than the trans Pd–Npyridine σ-bonding interaction.

Understanding the lability of a trans bound pyridine ligand in a saturated six-membered N-heterocyclic carbene based (NHC)PdCl2(pyridine) type complex: A case study

The lability of a trans “throwaway” pyridine ligand in a PEPPSI (Pyridine Enhanced Precatalyst Preparation, Stabilization and Initiation) themed saturated six-membered N-heterocyclic carbene complex namely, (1,3-difurfuryl tetrahydropyrimidin-2-ylidene)PdCl2(pyridine) (2), was investigated with the aid of a combined experimental and computational studies. In this regard, a new saturated six-membered N-heterocyclic carbene complex 2 was synthesized from the reaction of 1,3-difurfuryl tetrahydropyrimidin-1-ium chloride (1) with PdCl2 in the presence of KN(SiMe3)2 as a base in pyridine. Significantly enough, the complex 2 represents the first structurally characterized example of a PEPPSI themed trans-(NHC)PdX2(pyridine) (X=halide) type of a complex supported over a saturated six-membered N-heterocyclic carbene ligand. The lability of a trans “throwaway” pyridine ligand in 2 was probed by a displacement reaction with PPh3 to yield the corresponding phosphine analog, (1,3-difurfuryl tetrahydropyrimidin-2-ylidene)PdCl2(PPh3) (3), at room temperature under ambient condition. Detailed density functional theory (DFT) studies aimed at understanding the lability of the trans “throwaway” pyridine ligand in 2, against a PPh3 ligand yielding 3, points towards a kinetic factor rather than a thermodynamic one as a reason for the lability of the pyridine in 2.

The Development of Bulky Palladium NHC Complexes for the Most‐Challenging Cross‐Coupling Reactions

Palladium-catalyzed cross-coupling reactions enable organic chemists to form C-C bonds in targeted positions and under mild conditions. Although phosphine ligands have been intensively researched, in the search for even better cross-coupling catalysts attention has recently turned to the use of N-heterocyclic carbene (NHC) ligands, which form a strong bond to the palladium center. PEPPSI (pyridine-enhanced precatalyst preparation, stabilization, and initiation) palladium precatalysts with bulky NHC ligands have established themselves as successful alternatives to palladium phosphine complexes. This Review shows the success of these species in Suzuki-Miyaura, Negishi, and Stille-Migita cross-couplings as well as in amination and sulfination reactions.

An Efficient Low‐Temperature Stille–Migita Cross‐Coupling Reaction for Heteroaromatic Compounds by Pd–PEPPSI–IPent

The reactivity of Pd-PEPPSI (Pyridine, Enhanced, Precatalyst, Preparation, Stabilization, and Initiation) precatalysts in the Stille-Migita cross-coupling reaction between heteroaryl stannanes and aryl or heteroaryl halides was evaluated. In general, Pd-PEPPSI-IPent (IPent=diisopentylphenylimidazolium derivative) demonstrated high efficiency over a variety of challenging aryl or heteroaryl halides with thiophene-, furan-, pyrrole-, and thiazole-based organostannanes when compared with Pd-PEPPSI-IPr (IPr=diisopropylphenylimidazolium derivative). The transformations proceeded at appreciably lower temperatures (30-80 degrees C) than triarylphosphine-based Pd catalysts, improving the scope of this useful carbon-carbon bond-forming process.

Palladium complexes of abnormal N-heterocyclic carbenes as precatalysts for the much preferred Cu-free and amine-free Sonogashira coupling in air in a mixed-aqueous medium

A series of new PEPPSI (Pyridine Enhanced Precatalyst Preparation Stabilization and Initiation) themed precatalysts of abnormal N-heterocyclic carbenes for the highly desirable Cu-free and amine-free Sonogashira coupling in air in a mixed-aqueous medium is reported. Specifically, the PEPPSI themed (NHC)PdI2(pyridine) type precatalysts, 1b-4b, efficiently carried out the highly convenient Cu-free and amine-free Sonogashira coupling of aryl bromides and iodides with terminal acetylenes in air in a mixed aqueous medium. Complexes, 1b-4b, were synthesized by the direct reaction of the corresponding imidazo[1,2-a]pyridinium iodide salts, 1a-4a, with PdCl2 in pyridine in the presence of K2CO3 as a base while the imidazo[1,2-a]pyridinium iodide salts, 1a-4a, were in turn synthesized by the alkylation reactions of the respective imidazo[1,2-a]pyridine derivatives with alkyl iodides. The density functional theory (DFT) studies revealed that these imidazol-3-ylidene[1,2-a]pyridine derived abnormal carbenes are strongly sigma-donating and consequently significantly weaken the catalytically important labile trans pyridine ligand in 1b-4b.

Highly Convenient Amine‐Free Sonogashira Coupling in Air in a Polar Mixed Aqueous Medium by trans‐ and cis‐[(NHC)2PdX2] (X=Cl, Br) Complexes of N/O‐Functionalized N‐Heterocyclic Carbenes

Two new trans- and cis-[(NHC)(2)PdX(2)] (X=Cl, Br) complexes of N/O-functionalized N-heterocyclic carbenes employed in a highly convenient amine-free Sonogashira cross-coupling reaction in air in a polar mixed aqueous medium are reported. Specifically, the trans-[{1-benzyl-3-(3,3-dimethyl-2-oxobutyl)imidazol-2-ylidene}(2)PdBr(2)] (3) and cis-[{1-benzyl-3-(N-tert-butylacetamido)imidazol-2-ylidene}(2)PdCl(2)] (4) complexes effectively catalyzed the Sonogashira cross-coupling reaction of aryl iodides with substituted acetylenes in air in a mixed solvent (DMF/H(2)O, 3:1 v/v) under amine-free conditions. Interestingly, these trans- and cis-[(NHC)(2)PdX(2)] (X=Cl, Br) complexes, with two N-heterocyclic carbene ligands, exhibited superior activity compared with the now popular PEPPSI (pyridine enhanced precatalyst preparation, stabilization and initiation)-themed analogues, trans-[(NHC)Pd(pyridine)X(2)] (X=Cl, Br), 3 a and 4 a, with one N-heterocyclic carbene ligand and a "throw away" pyridine ligand in a trans disposition to each other. The higher activities of 3 and 4 compared with PEPPSI analogues 3 a and 4 a are attributed to more-electron-rich metal centers, as revealed by DFT studies, in the former complexes and is in concurrence with a more electron-rich metal center being effective in facilitating the oxidative addition of aryl halide, often a rate-determining step in palladium-mediated cross-coupling reactions. Complexes 3 and 4 were prepared from the corresponding silver analogues by transmetalation with [(cod)PdCl(2)], whereas the corresponding PEPPSI analogues 3 a and 4 a were obtained directly from the imidazolium halide salts by reaction with PdCl(2) in pyridine in the presence of K(2)CO(3) as base.

Air-stable, convenient to handle Pd based PEPPSI (pyridine enhanced precatalyst preparation, stabilization and initiation) themed precatalysts of N/O-functionalized N-heterocyclic carbenes and its utility in Suzuki–Miyaura cross-coupling reaction

Several new air-stable, convenient to handle and easily synthesized Pd based PEPPSI (Pyridine Enhanced Precatalyst Preparation, Stabilization and Initiation) type precatalysts supported over N/O-functionalized N-heterocyclic carbenes (NHC) namely, trans-[1-(benzyl)-3-(N-t-butylacetamido)imidazol-2-ylidene]Pd(pyridine)Cl2 (), trans-[1-(2-hydroxy-cyclohexyl)-3-(benzyl)imidazol-2-ylidene]Pd(pyridine)Cl2 () and trans-[1-(o-methoxybenzyl)-3-(t-butyl)imidazol-2-ylidene]Pd(pyridine)Br2 (), have been designed. Specifically, the Pd-NHC complexes, , and , were conveniently synthesized from their respective imidazolium halide salts by the reaction with PdCl2 in pyridine in presence of K2CO3 as a base. A new imidazolium chloride salt, 1-(benzyl)-3-(N-t-butylacetamido)imidazolium chloride () was synthesized by the alkylation reaction of benzyl imidazole with N-t-butyl-2-chloroacetamide. The molecular structures of the imidazolium chloride salt, , and the Pd-NHC complexes, , and , have been determined by X-ray diffraction studies. The density functional theory studies of the , and complexes were carried out to in order to gain insight about their structure, bonding and the electronic properties. The nature of the NHC-metal bond in these complexes was examined using Charge Decomposition Analysis (CDA), which revealed that the N-heterocyclic carbene ligands are effective sigma-donors. In addition, the catalysis studies revealed that the Pd-NHC complexes, , and , are effective catalysts for the Suzuki-Miyaura type C-C cross-coupling reactions.
