Effects of Using Mechanical Brushes on the Productive Performance of Dairy Cows

Intensive farming can reduce production costs and maximize animal production efficiency; however, it also causes many adverse effects on the welfare of dairy cows. A mechanical brush is an automated grooming device that promotes the grooming behavior of dairy cattle, thereby helping to alleviate stress. In the present study, we evaluated the effects of using mechanical brushes on the production performance of dairy cows by comprehensively analyzing their milk production, health status, and reproductive performance. The cows were assigned to 6 groups: 109 lactating dairy cows (brush treatment) and 105 controls (without brush treatment), 64 dry milk dairy cows (brush treatment) and 49 controls (without brush treatment), and 198 perinatal cows (brush treatment) and 65 controls (without brush treatment). We found an increasing trend in the daily utility time and usage frequency of mechanical brushes for each cow during the lactating period (7.73 ± 4.02 min/d and 2.90 ± 1.22 times/d, respectively), dry period (15.97 ± 14.16 min/d and 4.21 ± 2.91 times/d, respectively), and perinatal period (25.15 ± 19.05 min/d and 5.45 ± 3.83 times/d, respectively) (p < 0.01 and p < 0.05, respectively). The installation location of the mechanical brush significantly affected the frequency of its usage during the different periods. The head was the preferred body part for using the mechanical brush during the lactation and dry periods (59.32% and 44.54%, respectively), while the hip was the main preferred grooming part during the perinatal period (40.17%). Overall, the time, frequency, and preferred body part of dairy cows that used mechanical brushes varied across different physiological stages. Additionally, mechanical brush use in lactating and dry dairy cows significantly improved cleanliness of the body’s surface (p < 0.05) and enhanced milk production of lactating cows (p < 0.01), particularly for cows with four and five parities. Thus, the use of mechanical brushes could improve the production performance of dairy cows and enhance sustainability of large-scale farms.

Effects of supplementing rumen-protected arginine on performance of transition cows

An experiment was conducted to determine the effects of supplementing rumen-protected arginine (RPA) on productive performance in dairy cows. One-hundred and 2 cows were blocked by parity and then by energy-corrected milk (ECM) yield. Within block, cows were randomly assigned to control (CON) that received 200 g/d of a mixture of hydrogenated soybean oil and heat-treated soybean meal to supply 30 g of metabolizable protein (MP), or 200 g/d of a product containing 30 g of metabolizable arginine (RPA), which increased the dietary arginine from 5.7 to 7.5% of the MP from 250 d of gestation to 21 d postpartum. After 21 d postpartum, cows were fed the same diet and data were collected until 84 d postpartum. Cows fed RPA produced an additional 2.5 kg of colostrum (5.3 vs. 7.8 ± 1.0 kg) and 220 g more immunoglobulin G (526 vs. 746 ± 93 g) than CON cows. Supplementing RPA increased the yields of milk (32.8 vs. 34.9 ± 1.0 kg/d), ECM (37.8 vs. 40.9 ± 1.2 kg/d), and milk total solids (4.48 vs. 4.86 ± 0.14 kg/d) in the first 21 DIM. The benefits of RPA extended beyond the period of supplementation, with a 6.4% increase in yield of ECM per kg of dry matter consumed in all cows (1.88 vs. 2.00 ± 0.05 kg/kg) and an increase in ECM yield, but only in parous cows (44.2 vs. 48.5 ± 1.5 kg/d). Feeding RPA increased the concentrations of urea N in plasma pre- (12.5 vs. 13.9 ± 0.4 mg/dL) and postpartum (11.6 vs. 13.2 ± 0.4 mg/dL), and in milk during the first 21 d postpartum (11.0 vs. 12.0 ± 0.3 mg/dL). Treatment did not affect the concentrations of AA in plasma prepartum, but feeding RPA tended to increase citrulline (72.5 vs. 77.5 ± 2.7 μM), whereas RPA either tended to decrease isoleucine (129.5 vs. 120.9 ± 5.7 μM) or decreased the concentrations of leucine (181.3 vs. 170.2 ± 6.4 μM) and valine (293.2 vs. 276.7 ± 10.4 μM) postpartum. Feeding RPA increased the relative expression of transcripts involved in AA transport (SLC38A4), urea cycle (ARG1), and gluconeogenesis (PC, PEPCK, and G6PC) in hepatic tissue. Feeding diets to supply additional arginine as RPA during the transition period benefited productive performance in dairy cows that extended beyond the period of supplementation despite minor changes in plasma AA concentrations.

PSV-2 Rumen fluid metabolomics of dairy cows fed combinations of essential oil blend, onion peel extract and prebiotics using the RUSITEC system

Abstract Dairy products have a crucial role in the diets of over 6 billion people worldwide, providing essential nutrients such as energy, high-quality protein, and micronutrients. With dairy cows accounting for nearly 81% of global milk production, strategies to increase milk production sustainably are vital, especially with the U.S. being the second-largest producer. Essential oil blends (EOB), onion peel extract (OPE), and prebiotics such as mannan oligosaccharides (MOS) and galacto-oligosaccharides (GOS) offer promising avenues to enhance bovine milk production. These additives have the potential to modify rumen ecology and fermentation activity, while also mitigating greenhouse gas emissions in dairy cows. Therefore, this study investigated the effects of combining EOB, OPE, GOS, and/or MOS on specific rumen metabolites and metabolic pathways using the rumen simulation technique (RUSITEC) system. Rumen-cannulated non-lactating Holstein Friesian cows (n = 3) were used as rumen fluid donors. Total mixed ration (TMR) consisted of corn silage (60%), alfalfa hay (20%), and concentrate (20%). Using a completely randomized design, 16 fermentation vessels were divided into 4 groups with 4 replicates each over a 9-d period. The treatments included a control group (CON; TMR only), GEO (TMR+GOS+EOB+OPE), MEO (TMR+MOS+EOB+OPE), and OLEO (TMR+OLG+EOB+OPE). The EOB comprised of garlic, lemongrass, cumin, lavender, and nutmeg (4:2:2:1:1), while OLG contained GOS and MOS in equal proportions. EOB was added at 10 µL/g, and OPE, GOS, MOS, and OLG were included at 30 mg/g TMR. Rumen effluents were collected over 5 d for quantitative untargeted metabolome analysis. Analysis of rumen effluents using liquid chromatography-mass spectrometry and MetaboAnalyst 5.0 revealed 495 detected and identified metabolites. Volcano plot analysis showed no significant differences (P ≥ 0.05) in metabolite abundance between the CON and MEO, GEO, or OLEO treatments. Partial least squares-discriminant analysis (PLS-DA) also indicated no clear separation between the CON and treatment groups, suggesting no significant alteration in the metabolome. Compared with CON, the relative concentrations of dopamine 4-O-glucuronide, proline, and histamine were less while pyrocatechol was greater in MEO group. Isomer 1 of 2-hydroxyhepta-2,4-dienedioic acid, histamine, and dopamine 4-O-glucuronide were less while 2-aminomuconic acid and isomer 1 of hulupinic acid were greater in the GEO group. Histamine was less and hydroxypyridine was greater in the OLEO group. Phosphonate and phosphinate metabolism tended to be upregulated in OLEO. Meanwhile, metabolic pathways related to amino acids, vitamins, nitrogen metabolism, and primary bile acid biosynthesis remained unaltered by MEO, GEO and OLEO treatments. In summary, the study demonstrated that the inclusion of MEO, GEO, and OLEO had no adverse effects on rumen metabolites. Future research should focus on combinations of nutraceuticals that could positively impact nutrient metabolism and production performance of dairy cows.

The Effects of trans-10, cis-12 Conjugated Linoleic Acid on the Production Performance of Dairy Cows and the Expression and Transcription Regulation of Lipid Metabolism-Related Genes in Bovine Mammary Epithelial Cells.

Dietary fatty acids (FAs) determine the quality of dairy products. The trans-10, cis-12 conjugated linoleic acid (t10c12-CLA) is commonly considered an FA factor leading to milk fat depression syndrome (MFDs) in dairy cow. However, its effect on dairy cow performance and involvement in milk fat metabolism have been insufficiently explored. This study administered 136.17 g/day of rumen-protected CLA (RP-CLA) to dairy cows and found a diminution in milk fat percentage and a trend of increasing milk protein percentage on day 21 postpartum. Lactose content, milk yield, and net energy for lactation were unaffected. In the cell experiments, Oil Red O staining showed a notable increase in lipid droplets. Gene and protein expression analysis showed that 300 μM t10c12-CLA upregulated the expression of CD36, DGAT2, and ADRP, while downregulating the expression of ACACA, FASN, SREBP1, FABP3, FATP3, ACSL4, LPIN1, DGAT1, BTN1A1, XDH, SNAP23, and VAMP4. This provides a possible mechanistic pathway for the contradictory phenomenon of t10c12-CLA reducing milk fat while increasing lipid droplets. Overall, t10c12-CLA, as a long-chain fatty acid, can promote lipid droplet synthesis but may reduce milk fat by inhibiting lipid droplet fusion and secretion, FAs de novo synthesis, and triglyceride biosynthesis.

Microbial inoculum effects on the rumen epithelial transcriptome and rumen epimural metatranscriptome in calves

Manipulation of the rumen microbial ecosystem in early life may affect ruminal fermentation and enhance the productive performance of dairy cows. The objective of this experiment was to evaluate the effects of dosing three different types of microbial inoculum on the rumen epithelium tissue (RE) transcriptome and the rumen epimural metatranscriptome (REM) in dairy calves. For this objective, 15 Holstein bull calves were enrolled in the study at birth and assigned to three different intraruminal inoculum treatments dosed orally once weekly from three to six weeks of age. The inoculum treatments were prepared from rumen contents collected from rumen fistulated lactating cows and were either autoclaved (control; ARF), processed by differential centrifugation to create the bacterial-enriched inoculum (BE), or through gravimetric separation to create the protozoal-enriched inoculum (PE). Calves were fed 2.5 L/d pasteurized waste milk 3x/d from 0 to 7 weeks of age and texturized starter until euthanasia at 9 weeks of age, when the RE tissues were collected for transcriptome and microbial metatranscriptome analyses, from four randomly selected calves from each treatment. The different types of inoculum altered the RE transcriptome and REM. Compared to ARF, 9 genes were upregulated in the RE of BE and 92 in PE, whereas between BE and PE there were 13 genes upregulated in BE and 114 in PE. Gene ontology analysis identified enriched GO terms in biological process category between PE and ARF, with no enrichment between BE and ARF. The RE functional signature showed different KEGG pathways related to BE and ARF, and no specific KEGG pathway for PE. We observed a lower alpha diversity index for RE microbiome in ARF (observed genera and Chao1 (p < 0.05)). Five microbial genera showed a significant correlation with the changes in host gene expression: Roseburia (25 genes), Entamoeba (two genes); Anaerosinus, Lachnospira, and Succiniclasticum were each related to one gene. sPLS-DA analysis showed that RE microbial communities differ among the treatments, although the taxonomic and functional microbial profiles show different distributions. Co-expression Differential Network Analysis indicated that both BE and PE had an impact on the abundance of KEGG modules related to acyl-CoA synthesis, type VI secretion, and methanogenesis, while PE had a significant impact on KEGGs related to ectoine biosynthesis and D-xylose transport. Our study indicated that artificial dosing with different microbial inocula in early life alters not only the RE transcriptome, but also affects the REM and its functions.

Supplementation with Combined Additive Improved the Production of Dairy Cows and Their Offspring with Maintenance of Antioxidative Stability.

Oxidative stress damage in periparturient cows decreases both production and their health; supplementation with complex additives during the periparturient period has been used as an important strategy to enhance the antioxidant status and production of dairy cows. The periparturient cows not only risk a negative energy balance due to reduced dry matter intake but also represent a sensitive period for oxidative stress. Therefore, we have developed an immunomodulatory and nutritional regulation combined additive (INC) that hopefully can improve the immune status and production of cows during the periparturient period and their offspring health and growth by improving their antioxidant stress status. The INC comprised a diverse array of additives, including water-soluble and fat-soluble vitamins, Selenomethionine, and active dry Saccharomyces cerevisiae. Forty-five multiparous Holstein cows were randomly assigned to three treatments: CON (no INC supplementation, n = 15), INC30 (30 g/d INC supplementation, n = 15), and INC60 (60 g/d INC supplementation, n = 15) based on last lactation milk yield, body condition score, and parity. Newborn calves were administered 4 L of maternal colostrum originating from the corresponding treatment and categorized based on the treatment received by their respective dams. The INC not only served to maintain the antioxidative stress system of dairy cows during the periparturient period but also showed a tendency to improve the immune response (lower tumor necrosis factor and interleukin-6) during the perinatal period. A linear decrease in concentrations of alkaline phosphatase postpartum and β-hydroxybutyrate was observed with INC supplementation. Milk fat yield, milk protein yield, and energy-corrected milk yield were also increased linearly with increasing additive supplementation. Calves in the INC30 group exhibited greater wither height and chest girth but no significant effect on average daily gain or body weight. The diarrhea frequency was linearly decreased with the incremental level of INC. Results indicate that supplementation with INC in peripartum dairy cows could be a major strategy to improve immune response, decrease inflammation, maintain antioxidant stress status in transition dairy cows, and have merit in their calves. In conclusion, this study underlines the benefits of INC supplementation during the transition period, as it improved anti-inflammatory capacity, could positively impact antioxidative stress capacity, and eventually enhanced the production performance of dairy cows and the health and growth of calves.

Sirtuin 5 alleviates apoptosis and autophagy stimulated by ammonium chloride in bovine mammary epithelial cells.

Ammonia (NH3) is an irritating and harmful gas that affects cell apoptosis and autophagy. Sirtuin 5 (SIRT5) has multiple enzymatic activities and regulates NH3-induced autophagy in tumor cells. In order to determine whether SIRT5 regulates NH3-induced bovine mammary epithelial cell apoptosis and autophagy, cells with SIRT5 overexpression or knockdown were generated and in addition, bovine mammary epithelial cells were treated with SIRT5 inhibitors. The results showed that SIRT5 overexpression reduced the content of NH3 and glutamate in cells by inhibiting glutaminase activity in glutamine metabolism, and reduced the ratio of ADP/ATP. The results in the SIRT5 knockdown and inhibitor groups were comparable, including increased content of NH3 and glutamate in cells by activating glutaminase activity, and an elevated ratio of ADP/ATP. It was further confirmed that SIRT5 inhibited the apoptosis and autophagy of bovine mammary epithelial cells through reverse transcription-quantitative PCR, western blot, flow cytometry with Annexin V FITC/PI staining and transmission electron microscopy. In addition, it was also found that the addition of LY294002 or Rapamycin inhibited the PI3K/Akt or mTOR kinase signal, decreasing the apoptosis and autophagy activities of bovine mammary epithelial cells induced by SIRT5-inhibited NH3. In summary, the PI3K/Akt/mTOR signal involved in NH3-induced cell autophagy and apoptosis relies on the regulation of SIRT5. This study provides a new theory for the use of NH3 to regulate bovine mammary epithelial cell apoptosis and autophagy, and provides guidance for improving the health and production performance of dairy cows.

A new strain of Saccharomyces cerevisiae in diets of lactating Holstein cows improved feed efficiency and lactation performance

Abstract This study compared the effects of feeding a new strain of Saccharomyces cerevisiae HSA2020 with a commercial strain on in vitro rumen fermentation and production performance of dairy cows. Permeate was used as a substrate for the laboratory production of the new strain of S. cerevisiae after the hydrolysis by β–galactosidase (5000 µ/mL at 37°C). Two experiments were conducted: in Experiment 1, the effects of three levels (1, 2 and 3 g/kg dry matter) of S. cerevisiae on in vitro ruminal fermentation kinetics were evaluated. In Experiment 2, for 60 days, sixty multiparous Holstein cows (639±24.8 kg BW, 3±1 parity, 7±1 days in milk, with a previous milk production of 23±2.0 kg/d) during the previous lactation, were randomly assigned to 3 treatments in a completely randomized design. Cows were fed without any additives (control treatment) or supplemented with 2 g/kg feed daily of laboratory produced (PY)or commercial (CY) S. cerevisiae. In Experiment 1, inclusion of PY and CY increased (P&lt;0.05) gas production, propionate, and nutrient disappearance, while decreased (P&lt;0.05) methane production and protozoal count. Moreover, in Experiment 2, PY followed by CY increased (P&lt;0.01) nutrient digestibility, and serum concentrations of total protein, albumin, and glucose (P&lt;0.05). Higher daily milk yield, and milk energy output were observed with PY and CY without affecting concentrations of milk components or milk fatty acid profile. Compared to control, increased feed efficiency was observed with PY and CY. Compared to PY, CY increased serum concentrations of urea-N and decreased triglycerides, while PY decreased serum aspartate transaminase and increased concentration of conjugated linoleic acids in milk. In early lactating cow diets, both strains of S. cerevisiae improved production performance at 2 g/kg improved production performance, and minimal differences between strains were found.

Effect of carbohydrate source on energy-nitrogen balance and methane emissions in lactating dairy cows

The chemical characteristics of Cenchrus clandestinus grass, nutritional base in dairy production systems, demand the energy supplementation. Corn grain is the most used food for this purpose, but considering its demand and prices, the objective of this work was to evaluate its partial replacement by alternative non-fibrous carbohydrates sources on methane emissions, energy and nitrogen balance and productive performance of dairy cows. Sixteen multiparous Holstein cows were fed Cenchrus clandestinus plus one of four concentrate supplements formulated with corn (Zea mays) or a partial replacement of corn with sorghum (Sorghum vulgare), cassava (Manihot esculenta) or citrus pulp (Citrus sp.). Inclusion of sorghum, cassava or citrus pulp did not affect dry matter intake, dry matter digestibility, enteric methane emissions, energy and nitrogen balance, milk yield or composition. The results allow to conclude that these ingredients can partially replace corn in the diet of lactating dairy cows.

Effect of Calving Season on Productive Performance of Dairy Cows

The aim of the present research was to study the influence of the calving season in conditions of the upcoming climate changes on the productive traits of dairy cows in Bulgaria. The study was conducted on a cattle farm with a capacity of 500 dairy cows, which were loose-housed in open free-stall barns (shed-type). In the research, 286 lactations of 199 Holstein cattle from the studied farm were included. The cows with the highest average milk yield for lactation—8522.2 kg—calved in the spring, while the cows with the lowest milk yield—8082.7 kg—calved in the summer. Cows that calved in the spring had the highest maximum daily milk yield (lactation peak)—38 kg—whereas cows that calved in the summer had the lowest—35.7 kg. Regarding the composition indicators of milk, fat, and protein content, no significant effect of the calving season was found, but there was a tendency for the lowest values for the percentage of fat in milk to be reported for cows that calved in the summer—3.68%—and the highest for those calved in the spring—3.71%. Regarding the percentage of protein in the milk, the lowest values were observed for cows that calved in autumn—3.19%—and the highest for cows that calved in summer—3.27%.

Effects of feeding rumen-protected lysine during the postpartum period on performance and amino acid profile in dairy cows: A meta-analysis

Lysine is one of the limiting AA in the diets of dairy cows and is typically fed as rumen-protected Lys (RPL). We hypothesized that supplementation of RPL during the postpartum period would improve the productive performance in dairy cows. Objectives were to use meta-analytic methods to explore the effects of feeding RPL on performance and blood AA profile in lactating dairy cows. An additional objective was to identify an optimal concentration (%) of Lys in MP (LYSMP) and determine if responses to LYSMP were associated with the concentration (%) of Met in MP (METMP). The literature was systematically reviewed, and 13 experiments, comprising 40 treatment means and 594 lactating cows, were included in the meta-analysis. All experiments had a nonsupplemental control (CON; n = 17 treatment means), or a group supplemented with RPL (n = 23 treatment means). Cows supplemented with RPL were supplied additionally with a mean (±standard deviation) 19.3 ± 10.3 g/d metabolizable Lys (5.1-40.6 g/d). Meta-analytical statistics were used to estimate the weighted mean difference in STATA. Mixed models were fitted to the data to investigate the linear and quadratic effects of LYSMP, METMP, and interactions between LYSMP and METMP. All models included the random effect of experiment and weighting by the inverse of the SE of the means squared. Cows that began receiving RPL in early lactation (≤90 DIM) or for an extended duration (≥70 DIM) produced 1.51 kg/d more milk compared with CON cows. Increasing digestible LYSMP from 6.5% to 8.5% linearly increased yields of milk, FCM, ECM, and milk fat by 1.8, 2.5, 2.4, and 0.10 kg/d, respectively, and tended to increase milk protein yield and body weight gain by 0.07 and 0.09 kg/d, respectively, without a concurrent increase in DMI. Interactions between the linear effects of LYSMP and METMP were observed for FCM/DMI or ECM/DMI. In a diet with low METMP (e.g., 1.82% of MP), a digestible supply of 7.40% LYSMP would result in 1.46 and 1.47 kg/kg FCM/DMI or ECM/DMI, respectively; however, with high digestible METMP (e.g., 2.91% of MP), supplying 7.40% of digestible LYSMP would result in 1.68 and 1.62 kg/kg FCM/DMI or ECM/DMI, respectively. Increasing digestible LYSMP from 6.5% to 8.5% linearly increased blood concentrations of Lys by 16.6 µM, whereas blood concentrations of Met and Ala decreased by 4.6 and 6.0 µM, respectively. Nevertheless, an interaction was also observed between LYSMP and METMP for blood concentrations of total EAA because as METMP increased, the positive response to LYSMP on total EAA was also increased, suggesting a competitive mobilization of AA and their utilization in various body tissues. Only 4 out of the 13 experiments in this meta-analysis involved primiparous cows; thus, insufficient data were available to understand the role of supplemental RPL in primiparous cows. Collectively, feeding RPL improved productive performance, and the increments were maximized up to 9.25% of LYSMP in multiparous dairy cows.

Effects of prepartum concentrate feeding on reticular pH, plasma energy metabolites, acute phase proteins, and milk performance in grass silage–fed dairy cows

We investigated how concentrate feeding during the last 21 d of pregnancy affects reticular pH, inflammatory response, dry matter (DM) intake, and production performance of dairy cows. We hypothesized that adding concentrates to dairy cows' diet before calving reduces the decrease in reticular pH postpartum and thus alleviates inflammatory response. We also hypothesized that prepartum concentrate feeding increases DM intake postpartum and consequently improves milk performance. Two feeding experiments were conducted using a randomized complete block design. In each experiment, 16 multiparous Finnish Ayrshire cows were paired based on parity, expected calving date, body weight, and milk yield of the previous lactation. Within the pairs, cows were randomly allocated on one of the 2 dietary treatments 21 d before expected calving. In experiment 1 (Exp1), diets were ad libitum feeding of grass silage as a sole feed or supplemented with increasing amounts of concentrate offered separately (increased to 4 kg/d by d -7). In experiment 2 (Exp2), diets were ad libitum feeding of a total mixed ration containing either grass silage, barley straw, and rapeseed meal (64%, 28%, and 8% on DM basis, respectively) or grass silage, barley straw, and cereal-based concentrate mixture (49%, 29%, and 30% on DM basis, respectively). Following calving, all the cows were fed similarly and observed until d 56 postpartum. Feed intake and milk yield were recorded daily, and reticular pH was monitored continuously by reticular pH bolus. Blood samples were collected at the beginning of the experiments, 7 d before the expected calving date, 1 d (in Exp1) or 5 d (in Exp2), 10 d, and 21 d postpartum. In Exp1, concentrate feeding increased metabolizable energy intake and tended to increase DM and crude protein intake prepartum. Moreover, prepartum concentrate feeding increased the concentrations of plasma β-hydroxybutyrate and insulin, but differences in nonesterified fatty acids, glucose, or acute phase proteins were not observed. After calving, prepartum diet did not affect DM or nutrient intake, plasma energy metabolites, or milk production in Exp1. Although prepartum concentrate feeding increased reticular pH on the first day of lactation, it elevated plasma concentrations of serum amyloid-A and haptoglobin postpartum in the grass silage-based diet. In Exp2, adding concentrates to the diet based on a mixture of grass silage and straw did not affect prepartum DM intake or plasma concentrations of nonesterified fatty acids, glucose, or insulin. Adding concentrates to prepartum diet increased plasma concentration of β-hydroxybutyrate before calving as in Exp1. After calving, prepartum concentrate feeding increased DM and nutrient intake during the second week of lactation in Exp2, but no effects were observed thereafter. In contrast to our hypothesis, prepartum concentrate feeding decreased reticular pH after calving in Exp2, but no differences in inflammatory markers were observed. Based on this study, close-up concentrate feeding in diets based on grass silage with or without straw does not alleviate the decrease in reticular pH or mitigate inflammatory response postpartum.

Dietary Neutral Detergent Fiber Levels Impacting Dairy Cows' Feeding Behavior, Rumen Fermentation, and Production Performance during the Period of Peak-Lactation.

This study investigated the impact of dietary neutral detergent fiber (NDF) levels (25.49%, 28.65%, 31.66%, and 34.65%, respectively) on the feeding behavior, rumen fermentation, cellulolytic bacteria, and production performance of dairy cows during peak lactation. A feeding experiment was conducted using four fistulated Holstein dairy cows (600 ± 25 kg) with days in milk (50 ± 15 days), employing a 4 × 4 Latin square design to assign the cows to four groups. The results demonstrated that increasing NDF levels in the diet had the following effects: (1) A linear decrease in dry matter intake (DMI), NDF intake, and physically effective NDF8.0 (peNDF8.0) intake; a linear increase in the average time spent eating and ruminating, as well as the time spent eating and ruminating per kilogram of dry matter (DM); a quadratic response in the time spent ruminating per kilogram of NDF and peNDF8.0. (2) A linear increase in average pH value, acetate concentration, and the proportions of Fibrobacter succinogenes and Ruminococcus flavefaciens among total bacteria; a linear decrease in ammonia nitrogen (NH3-N) concentration, microbial crude protein (MCP), total volatile fatty acid (TVFA), propionate, butyrate, and lactate. (3) A linear decrease in milk yield, milk protein percentage, and nitrogen efficiency of dairy cows; a linear increase in milk fat percentage and milk urea nitrogen (MUN) concentration. Based on the combined results, it was found that diets with 25% and 34% NDF had detrimental effects on the feeding behavior, rumen fermentation, and production performance of dairy cows. However, the diet with 28% NDF showed superior outcomes in production performance compared to the one with 31% NDF. Therefore, it is strongly recommended to include a diet containing 28% NDF during the critical peak lactation period for dairy cows.

DESEMPEÑO PRODUCTIVO DE VACAS LECHERAS ALIMENTADAS CON PRADERAS DE KIKUYO SOBRESEMBRADAS CON CENTENO EN SISTEMAS DE PRODUCCIÓN DE LECHE EN PEQUEÑA ESCALA EN LOS VALLES ALTOS DE MÉXICO

&lt;p&gt;&lt;strong&gt;Background: &lt;/strong&gt;Small-scale dairy systems contribute to mitigate rural poverty as they provide a stable source of incomes to farming families, and also contribute with over 30% of national milk production. However, the dependency on external inputs affects their profitability and sustainability, which added to difficult agroclimatic conditions and limiting management factors as high stocking rate in pastures. It is therefore necessary to evaluate feeding strategies better adapted to conditions in these systems to improve cattle diets based on quality forages reducing external dependency and thus, costs. A means to increase herbage yields in pastures is overseeding with annual fast-growing cereals. &lt;strong&gt;Objective: &lt;/strong&gt;The objective was to evaluate the productive performance of dairy cows on two feeding strategies based on grazing, and the analysis of costs in small-scale dairy systems in the highlands of central Mexico. &lt;strong&gt;Methodology: &lt;/strong&gt;Eight Holstein cows were used on continuous day grazing (8 h/day) under on-farm research with a double cross-over design. Treatments ere: continuous intensive grazing of Kikuyu grass pastures (KY) or Kikuyu grass overseeded with rye (KYCEN) from 6:00 to 16:00 h, plus 4.5 kg DM/day of commercial concentrate. Two groups of four cows balanced for days in milk and daily milk yield were formed, and each group randomly assigned to one of the treatment sequences: KY-KYCEN-KY y KYCEN-KY-KYCEN, with three experimental periods of 14-day each, with 10 days for adaptation to diets and four days for sampling. Pastures were assessed with a split-plot design. Records were for milk yield and composition, live weight and body condition. A partial budget analysis was performed to compare income and feeding costs. &lt;strong&gt;Results: &lt;/strong&gt;There were no statistical differences (P&amp;gt;0.05) in productive performance, with mean 3.5% fat corrected milk yield of 17 kg/cow/day, with 36.6 g/kg, 28.5 g/kg and 43.4 g/kg for milk fat, protein and lactose respectively. There were no significant differences (P&amp;gt;0.05) between treatments for sward height or herbage mass, but there were significant differences (P&amp;lt;0.05) for DM and OM content between treatments, and for CP, NDF, ADF, IVDMD, and EM (P&amp;lt;0.05) between periods. &lt;strong&gt;Implications:&lt;/strong&gt; In the face of complex agroecological scenarios for small-scale dairy systems, it is necessary to assess forages better adapted for these conditions as the inclusion of a fast growing cereal as rye to increase pasture production and increase the forage base in the diets with farm resources as Kikuyo grass, in order to increase the profitability of farms. &lt;strong&gt;Conclusions: &lt;/strong&gt;Since there were no statistical differences in the productive performance of dairy cows, or in economic performance, the conclusion is that treatment KY is the recommended option for this season. Treatment KYCEN requires expenditures in seed and labour, so that it is not a viable option for small-scale dairy systems in this season.&lt;/p&gt;

Associations of neonatal acute phase response with first lactation performance in dairy cows

During the first 3 wk of life, the immune system of newborn ruminants starts to work, as indicated by fluctuations in the concentrations of proinflammatory cytokines and acute phase proteins (APP). They have been shown to be markers for short and long-term weight gain in ruminants. This observational study investigated these proteins as possible indicators of first lactation performance of dairy cows. A total of 117 dairy calves from a single farm were enrolled in the study. Serum and fecal samples were taken once a week for the first 3 wk of life. Cryptosporidium spp. infection and its treatment were monitored and accounted for in statistical analysis. The concentrations of the APP serum amyloid A (SAA) and haptoglobin (Hp), and the proinflammatory cytokines interleukin-6 (IL-6) and tumor necrosis factor α (TNF-α) were analyzed in serum. After the first lactation, health and performance data of the cows were retrieved, and associations between APP and cytokine concentrations with first lactation outcomes were investigated through linear and logistic regression. This study found a negative association between the concentration of Hp in the second week of life and average daily weight gain at one year. The SAA concentration measured during the second and third weeks of life was positively associated with age at first calving. IL-6, SAA, and Hp measured in the second week of life were positively associated with calving to conception interval. The concentrations of IL-6 and TNF-α during the first week of life were associated with higher odds of reproductive issues during the first lactation period. These markers can be used to help understand underlying processes that influence animal health and production. In conclusion, inflammatory responses during the first week of life are related to reproductive success, whereas the second and third weeks of life seem to influence the future productive performance in dairy cows.

The effect of a herbal anthelmintic (garlic and neem) on the production performance of Karan Fries cows

The present study was conducted to evaluate the effect of a herbal anthelmintic (Garlic and Neem combination) on the production parameters of Karan Fries cows. For this study, a total of 28 freshly calved cows (1-3 parity), were divided into four groups with 7 cows in each group (T1, T2, T3, T4). The cows in T1 were kept as the control. In T2, a commercial dewormer was used, whereas in T3, a single dose of garlic and neem in the ratio of 1:5 @ 4g/kg BW was used 7 days post calving, and in T4, in addition to the first dose, a second dose was also supplemented on day 60 after calving. The milk samples were collected at 0, 1, 2, 3, 4, 5, 6, 7, 8 fortnights after the deworming treatment and analyzed for milk composition and SCC. From the 5th fortnight onwards, a significantly higher (P≤0.05) daily milk yield (kg) was observed in T1, T2, T3 groups compared to T1. The overall average Somatic Cell Count (SCC) was found to be significantly lower in T4 followed by T3, compared to T2, which was further significantly lower than T1. From the 3rd to the 8th fortnight, protein (%) was observed to be significantly (p≤0.05) higher in the anthelmintic treated groups (significantly higher in T4 followed by T2, T3) compared to T1. From the 4th to the 8th fortnight, fat (%) was significantly (P≤0.05) higher in T4 compared to T1. The overall SNF (%) in T2, T3 and T4 differed significantly (P≤0.05) from the control group (T1). The overall value of lactose (%) was found to be significantly higher in the T4 group compared to the other groups. During the 7th and 8th fortnight significantly higher (P≤0.05) lactose (%) was observed in the T4 group compared to the other groups. This study concluded that besides providing better anthelmintic effects in comparison to chemical anthelmintics, the garlic and neem combination might improve the production performance of dairy cows.

Lactational performance of cows fed extruded flaxseed in commercial dairy herds

The objective of the current on-farm trial was to assess the impact of feeding extruded flaxseed on milk yield and composition. Thirty commercial dairy herds located in the province of Québec, Canada were recruited. The experiment began with a baseline period of 2 months during which each cow received their regular diets. Data collected during this period were used as covariate. Farms were then randomly allocated into a control group (n = 15; 767 cows) which continued to receive their regular diets, or a treatment group (n = 15; 863 cows) which received diets supplemented with extruded flaxseed (0.7 kg/d per cow) during an experimental period of 7 months. Significance was declared at P ≤ 0.05 and tendencies at 0.05 < P ≤ 0.10. Feeding extruded flaxseed did not affect feed intake but increased milk yield by 1.1 kg/d per cow, and feed efficiency by 6.5%. Dietary addition of extruded flaxseed increased milk fat (tendency) and lactose yield, whereas milk protein yield was similar between treatments. Estimated CH4 intensity were reduced by 1.3 g/L of milk (−9.2%) in herds receiving extruded flaxseed. Feeding extruded flaxseed increased milk fat concentration of cis-9, cis-12, cis-15 18:3 and total n-3 fatty acids. Results of the current on-farm trial confirm observations made under experimental conditions that feeding moderate levels of extruded flaxseed improves production performance in dairy cows.

Effects of a mastitis J5 bacterin vaccination on the productive performance of dairy cows: An observational study using propensity score matching techniques

Inferring causal effects between variables when utilizing observational data is challenging due to confounding factors not controlled through a randomized experiment. Propensity score matching can decrease confounding in observational studies and offers insights about potential causal effects of prophylactic management interventions such as vaccinations. The objective of this study was to determine potential causality and impact of vaccination with an Escherichia coli J5 bacterin on the productive performance of dairy cows applying propensity score matching techniques to farm-recorded (e.g., observational) data. Traits of interest included 305-d milk yield (MY305), 305-d fat yield (FY305), 305-d protein yield (PY305), and somatic cell score (SCS). Records from 6,418 lactations generated by 5,121 animals were available for the analysis. Vaccination status of each animal was obtained from producer-recorded information. Confounding variables considered were herd-year-season groups (56 levels), parity (5 levels: 1, 2, 3, 4, and ≥5), and genetic quartile groups (4 levels: top 25% through bottom 25%) derived from genetic predictions for MY305, FY305, PY305, and SCS, as well as for the genetic susceptibility to mastitis. A logistic regression model was applied to estimate the propensity score (PS) for each cow. Subsequently, PS values were used to form pairs of animals (1 vaccinated with 1 unvaccinated control), depending on their PS similarities (difference in PS values of cows within a match required to be <20% of 1 standard deviation of the logit of PS). After the matching process, 2,091 pairs of animals (4,182 records) remained available to infer the causal effects of vaccinating dairy cows with the E. coli J5 bacterin. Causal effects estimation was performed using 2 approaches: simple matching and a bias-corrected matching. According to the PS methodology, causal effects of vaccinating dairy cows with a J5 bacterin on their productive performance were identified for MY305. The simple matched estimator suggested that vaccinated cows produced 163.89 kg more milk over an entire lactation when compared with nonvaccinated counterparts, whereas the bias-corrected estimator suggested that such increment in milk production was of 150.48 kg. Conversely, no causal effects of immunizing dairy cows with a J5 bacterin were identified for FY305, PY305, or SCS. In conclusion, the utilization of PS matching techniques applied to farm-recorded data was feasible and allowed us to identify that vaccination with an E. coli J5 bacterin relates to an overall milk production increment without compromising milk quality.

The form more than the fatty acid profile of fat supplements influences digestibility but not necessarily the production performance of dairy cows

The form of a lipid supplement, its degree of saturation, and its fatty acid (FA) profile greatly influence digestibility and cow productive response. The objective in this study was to examine the effect of fat supplements that differ in their form or FA profile on nutrient digestibility and cow performance. Forty-two mid-lactation cows (128 ± 53 d) were assigned to 3 treatment groups according to milk yield, days in milk, and body weight. For 13 wk, the cows were fed rations that contained (on a dry matter basis) (1) 2.4% of calcium salts of fatty acids (CSFA) consisting of 45% palmitic acid (PA) and 35% oleic acid (OA; CS45:35); (2) 2.4% of CSFA consisting of 80% PA and 10% OA (CS80:10); or (3) 2.0% of free FA consisting of 80% PA and 10% OA (FF80:10). Rumen samples were taken to measure the ammonia and volatile FA concentrations, and fecal samples were taken to measure the digestibility. Preplanned comparisons were CS45:35 versus CS80:10 to assess 2 CSFA supplements with different FA profiles, and CS80:10 versus FF80:10 to assess similar FA profiles in different forms. Compared with CS45:35, CS80:10 decreased the milk yields, increased the fat percentage, and tended to increase the energy-corrected milk (ECM) yields. The fat percentage of milk was highest in the FF80:10 cows (4.02%), intermediate in the CS80:10 cows (3.89%), and lowest in the CS45:35 cows (3.75%). Compared with CS80:10, FF80:10 increased milk yields (50.1 vs. 49.4 kg/d, respectively), tended to increase fat percentage, and increased 4% fat-corrected milk (4% FCM; 49.1 vs. 47.7 kg/d, respectively) and ECM yields (49.5 vs. 48.2 kg/d, respectively). Treatment had no effect on dry matter intake (DMI), and compared with CS80:10 cows, the calculated energy balance was lower in the FF80:10 cows. The 4% FCM/DMI and ECM/DMI ratios were higher in the FF80:10 group compared with the CS80:10 group. Compared with the CS80:10 cows, the FF80:10 cows had a lower rumen pH, higher propionate, lower acetate/propionate ratio, and higher total VFA. Compared with CS45:35 cows, the apparent total-tract digestibilities of neutral detergent fiber and acid detergent fiber were higher in CS80:10 cows; whereas, the apparent total-tract digestibilities of dry matter, organic matter, protein, neutral detergent fiber, and acid detergent fiber were higher in the CS80:10 cows compared with the FF80:10 cows. Compared with the CS80:10 group, the apparent digestibility of total FA was 13.0 percentage points lower in the FF80:10 cows (79.1 vs. 66.1%, respectively), and similarly, the digestibilities of 16-carbon and 18-carbon FA were lower in the FF80:10 cows than in the CS80:10 cows. In conclusion, the form, more than the FA profile of fat supplements, influenced digestibility. Further, the CSFA supplements were more digestible than the free fatty acids, regardless of the FA profile. However, energy partitioning toward production appeared to be higher in the FF80:10 cows, although the digestibility of nutrients was lower than in the CSFA product with a similar FA profile.

Evidence that elevation of maternal somatic cell count could lead to production of offspring with inferior reproductive and productive performance in dairy cows during the first lactation period

Although the effect of mastitis on reproduction and production of lactating dairy cows has been vastly studied, little information is available about the association of maternal udder health status with offspring reproduction and production. Therefore, the present research was conducted to study the association between maternal average monthly somatic cell count (SCC) with reproduction, anti-Müllerian hormone (AMH) concentration, udder health status and milk production in the offspring. Based on maternal average monthly SCC (MSCC), offspring were classified into five categories including MSCC1 (SCC <200,000; n = 3005), MSCC2 (200,000 ≤ SCC <400,000; n = 252), MSCC3 (400,000 ≤ SCC <600,000; n = 103), MSCC4 (600,000 ≤ SCC <800,000; n = 40) and MSCC5 (SCC ≥800,000; n = 61). Data associated with reproduction, production and udder health status of offspring were retrieved from the herd database. In addition, blood samples were collected from a subset of offspring (n = 136) for measurement of serum AMH, as a reliable marker of ovarian reserves. Offspring in MSCC5 category had more services per conception and longer calving to conception interval than offspring in MSCC1 and MSCC2 categories (P < 0.05). The average number of SCC and risk of clinical mastitis in the offspring were not associated with MSCC (P > 0.05). But offspring in MSCC5 category produced less milk, fat and protein than offspring in MSCC1 category (P < 0.05). In addition, AMH concentration was lower in MSCC5 than MSCC1 category (P < 0.05). In conclusion, the present study showed that elevated maternal average monthly SCC could culminate in birth of offspring with inferior reproductive performance, smaller size of ovarian reserves and lower level of milk production during the first lactation period.
