Melatonin alleviates UV-B stress and enhances phenolic biosynthesis in rosemary (Rosmarinus officinalis) callus.

Although used in in vitro culture to boost secondary metabolite production, UV-B radiation can seriously affect plant growth if not properly dosed. Rosemary callus can be used as an important source of effective ingredients in the food and medicine industry. To balance the positive and negative effects of UV-B on rosmary callus, this study investigated the effects of melatonin on rosemary callus under UV-B radiation. The results showed that melatonin improved rosemary callus growth, with fresh weight and dry weight increased by 15.81% and 8.30%, respectively. The addition of 100 μM melatonin increased antioxidant enzyme activity and NO content in rosemary callus. At the same time, melatonin also significantly reduced membrane lipid damage and H2O2 accumulation in rosemary callus under UV-B stress, with malondialdehyde (MDA) and H2O2 contents reduced by 13.03% and 14.55%, respectively. In addition, melatonin increased the total phenol and rosmarinic acid contents in rosemary callus by 19% and 54%, respectively. Melatonin significantly improved the antioxidant activity of the extracts from rosemary callus. These results suggest that exogenous melatonin can alleviate the adverse effects of UV-B stress on rosemary callus by promoting NO accumulation while further enhancing phenolic accumulation and biological activity.

Short-Term Liquid Nitrogen Storage of Pyrostegia venusta Embryos: Effects on Germination, Phenotypic and Biochemical Characteristics, and In Vitro Secondary Metabolite Production

In this study, short-term liquid nitrogen (LN) storage was used as a strategy to conserve Pyrostegia venusta embryos, and its effects on in vitro germination, phenotypic and biochemical characteristics, and in vitro secondary metabolite production were assessed. Embryos stored in LN for 1 and 7 days presented a higher germination rate and germination speed index compared to those of the control (non-cryostored embryos). Short-term LN storage also favored the phenotypic characteristics of seedlings. LN storage significantly affected the proteins (PTN), soluble sugar (SS) and reducing sugar (RS) contents, oxidative metabolism, and phenylalanine ammonia-lyase (PAL) activity, as well as the total phenolic compound, flavonoid, phytosterol, and alkaloid levels in seedlings regenerated from embryos cryostored for 7 days. Benzoic acid derivatives and flavonoids were observed in regenerated non-acclimatized seedlings. LN storage did not affect the survival rate or phenotypic characteristics of seedlings during acclimatization. Acclimatization promoted significant changes in PTN, SS and RS contents, oxidative metabolism, and PAL activity in seedlings from embryos cryostored for 7 days. Roots from acclimatized seedlings exhibited the highest phenolic, phytosterol, and total alkaloid levels. Differences in the chromatographic profiles of the acclimatized seedlings compared with the non-acclimatized seedlings were observed. LN storage can be an effective means of ex situ conservation of P. venusta genetic resources.

1H-NMR-based metabolomics profiling of Beauveria bassiana on different growth media for the determination of its secondary metabolites

The entomopathogenic fungus (EPF) Beauveria bassiana is commercially used as a microbial insecticide against agricultural pests. Some strains of B. bassiana protect crops from pathogens. B. bassiana produces bioactive secondary metabolites (SMs) with potential applications. The present study aims to monitor the growth and behaviour of B. bassiana in different growth media and the effect of different growth media on the production of SMs. The fungal culture was recultivated and then inoculated in broth and agar growth media to perform this study. After 12 days of incubation of B. bassiana in potato dextrose broth (PDB) and a specific medium at temperatures between 28 - 30℃, SMs were extracted from broth cultures using liquid-liquid extraction. To harvest the SMs, the broth media of each culture was extracted using ethyl acetate. After the evaporation of the ethyl acetate using a rotary evaporator, the crude extracts were obtained. The crude extracts were then subjected to 1H-NMR and 2D HSQC analysis using a 600 MHz NMR spectrometer to monitor the presence of different SMs in the crude extracts. The analysis revealed that different types of SMs were produced by B. bassiana under different media and conditions. However, some SMs were detected in the 1H- NMR spectrum of the PDB extract that was completely absent in specific Fingal medium, further confirmed by the 2D HSQC experiment. In conclusion, this study revealed that modifying the media constituents and conditions, such as variable temperatures, could produce different SMs, as predicted by the NMR analysis.

An update on biotechnological intervention mediated by plant tissue culture to boost secondary metabolite production in medicinal and aromatic plants

AbstractSince prehistoric times, medicinal and aromatic plants (MAPs) have been employed for various therapeutic purposes due to their varied array of pharmaceutically relevant bioactive compounds, i.e. secondary metabolites. However, when secondary metabolites are isolated directly from MAPs, there is occasionally very poor yield and limited synthesis of secondary metabolites from particular tissues and certain developmental stages. Moreover, many MAPs species are in danger of extinction, especially those used in pharmaceuticals, as their natural populations are under pressure from overharvesting due to the excess demand for plant‐based herbal remedies. The extensive use of these metabolites in a number of industrial and pharmaceutical industries has prompted a call for more research into increasing the output via optimization of large‐scale production using plant tissue culture techniques. The potential of plant cells as sources of secondary metabolites can be exploited through a combination of product recovery technology research, targeted metabolite production, and in vitro culture establishment. The plant tissue culture approach provides low‐cost, sustainable, continuous, and viable secondary metabolite production that is not affected by geographic or climatic factors. This study covers recent advancements in the induction of medicinally relevant metabolites, as well as the conservation and propagation of plants by advanced tissue culture technologies.

Enhanced metabolite yield with compensatory biomass reduction revealed by moisture stress induction in <i>Centella asiatica</i> (L.)

The exposure to any kind of stress tends to accelerate the secondary metabolism in medicinal plants increasing the production of secondary metabolites. The present investigation was undertaken to study the effect of moisture stress (100, 75, 50, 25 and 10% pot capacity) and control (as without plant) on growth, yield and metabolite content of Centella asiatica var. ‘Arka Prabhavi’, for two growing seasons under polyhouse conditions. Results revealed that moisture stress treatments had a significant effect on all observed growth and yield traits. Plants maintained at 100% PC exhibited luxurious vegetative growth with maximum leaf length (6.28 cm), leaf breadth (8.14 cm), petiole length (22.32 cm) and fresh biomass yield (164 g/pot). Cumulative water transpired and water use efficiency of the plants was also observed to be maximum at 100% PC. In contrary to biomass yield, increased asiaticoside (1.864%, 1.892%), madecassoside (2.856%, 3.382%) and total triterpenoid content (5.356%, 5.578%) at higher moisture stress levels of 75% and 50% PC, respectively, was observed. Hence, it is appropriate to grow Centella either at 100% or 75% PC to get optimum biomass and metabolite yield on a commercial scale.

An Overview on Nocardiopsis Species Originating From North African Biotopes as a Promising Source of Bioactive Compounds and In Silico Genome Mining Analysis of Three Sequenced Genomes.

Actinobacteria are renowned for their prolific production of diverse bioactive secondary metabolites. In recent years, there has been an increasing focus on exploring "rare" genera within this phylum for biodiscovery purposes, notably the Nocardiopsis genus, which will be the subject of the present study. Recognizing the absence of articles describing the research process of finding bioactive molecules from the genus Nocardiopsis in North African environments. We, therefore, present a historical overview of the discoveries of bioactive molecules of the genus Nocardiopsis originating from the region, highlighting their biological activities and associated reported molecules, providing a snapshot of the current state of the field, and offering insights into future opportunities and challenges for drug discovery. Additionally, we present a genome mining analysis of three genomes deposited in public databases that have been reported to be bioactive. A total of 36 biosynthetic gene clusters(BGCs) were identified, including those known to encode bioactive molecules. Notably, a substantial portion of the BGCsshowed little to no similarity to those previously described, suggesting the possibility that the analyzed strains could be potential producers of new compounds. Further research on these genomes is essential to fully uncovering their biotechnological potential. Moving forward, we discuss the experimental designs adopted in the reported studies, as well as new avenues to guide the exploration of the Nocardiopsis genus in North Africa.

Elicitors directed in vitro growth and production of stevioside and other secondary metabolites in Stevia rebaudiana (Bertoni) Bertoni

Stevia rebaudiana (stevia) is a plant in the Asteraceae that contains several biologically active compounds including the antidiabetic diterpene glycosides (e.g. stevioside, rebaudioside and dulcoside) that can serve as zero-calorie sugar alternatives. In this study, an elicitation strategy was applied using 5% polyethylene glycol (PEG), sodium chloride (NaCl; 50 and 100 mM) and gibberellic acid (2.0 and 4.0 mg/L GA3) to investigate their effect on shoot morphogenesis, and the production of phenolics, flavonoids, total soluble sugars, proline and stevioside, as well as antioxidant activity, in shoot cultures of S. rebaudiana. Herewith, the media supplemented with 2 mg/L and 4 mg/L GA3 exhibited the highest shooting response (87% and 80%). The augmentation of lower concentrations of GA3 (2 mg/L) in combination with 6-benzylaminopurine (BAP) resulted in the maximum mean shoot length (11.1 cm). The addition of 100 mM NaCl salts to the media led to the highest observed total phenolics content (TPC; 4.11 mg/g-DW compared to the control 0.52 mg/g-DW), total flavonoids content (TFC; 1.26 mg/g-DW) and polyphenolics concentration (5.39 mg/g-DW) in shoots cultured. However, the maximum antioxidant activity (81.8%) was observed in shoots raised in media treated with 50 mM NaCl. The application of 2 mg/L of GA3 resulted in the highest accumulation of proline (0.99 μg/mL) as compared to controls (0.37 μg/mL). Maximum stevioside content (71 µL/mL) was observed in cultures supplemented with 100 mM NaCl and 5% PEG, followed by the 4 mg/L GA3 treatment (70 µL/mL) as compared to control (60 µL/mL). Positive correlation was observed between GA3 and stevioside content. Notably, these two compounds are derived from a shared biochemical pathway. These results suggest that elicitation is an effective option to enhance the accumulation of steviosides and other metabolites and provides the groundwork for future industrial scale production using bioreactors.

Silver nanoparticles: Elicitor of anticancer metabolite biosynthesis and growth-metabolites parameters in Tanacetum parthenium L.

Nanoparticles in appropriate dose can used as an eco-friendly abiotic elicitor to induce specific and novel secondary metabolites with high performance. The present study was conducted to investigate the silver nanoparticles (Ag-NPs) at 0, 200, 400, and 600 ppm on morpho-physiological, yield, and phytochemical parameters of feverfew (Tanacetum parthenium L.). The essential oil was obtained by hydro-distillation and analyzed by using GC-FID and GC/MS. The HPLC was used to quantifying the parthenolide content. The results showed that the effect of Ag-NPs application on studied traits were in dose depended manner. The Ag-NPs treatment at 200 ppm increased the fresh weight, biological yield, dry weight, and flower yield up to 29.12 %, 31.61 %, 28.71 %, and 44.93 % respectively. However, the growth of treated plants with higher concentrations of Ag-NPs (400 and 600 ppm) was decreased compared with untreated plants. All Ag-NPs treatments caused a significant increment of the chlorophyll index, parthenolide content (as a valuable anticancer compound) and essential oil content. The highest content of chlorophyll index (49.90), parthenolide content (0.96 mg g−1 DW) and essential oil content (0.84 %) was measured in spayed plants with 600 ppm Ag-NPs. Also, the Ag-NPs application considerably affected the essential oil quality. With increasing the Ag-NPs concentration the oxygenated monoterpenes represented by camphor and trans-chrysanthenyl acetate were increased so that the highest content of camphor (45.13 %) and trans-chrysanthenyl acetate (32.04 %) was obtained at 600 ppm. Generally, it can be concluded that Ag-NPs could act as an elicitor to induce the more production of favorable secondary metabolites.

Establishment of a callus induction system of Saxifraga stolonifera Meerb. and its response to different elicitors

Elicitation is a well-known strategy for increasing the accumulation of secondary metabolites in callus cultures. In this study, a callus induction system for Saxifraga stolonifera Meerb. was developed, and elicitors were used to enhance secondary metabolite production. Tender leaves of S. stolonifera were used as explants for in vitro culture. We examined callus induction characteristics of S. stolonifera and its response to various elicitors by assessing the sterilization conditions of the explants, varying hormone ratios, and determining the optimal concentrations of salicylic acid (SA), Sodium nitroprusside (SNP) and methyl jasmonate (MeJA). The results revealed that sterilizing the explants with 70% ethanol for 40 s, followed by 0.1% mercury chloride for 10 min, resulted in a survival rate of 82.22% during tissue culture. Following these sterilization methods, callus formation was successfully induced on Murashige and Skoog medium supplemented with 2 mg/L 6-benzylaminopurine and 0.15 mg/L naphthaleneacetic acid, achieving an induction rate of 88.88%. Additionally, the study identified SA as the most effective elicitor for increasing the levels of gallic acid, protocatechuic acid, and quercitrin in the callus, with optimal concentrations of salicylic acid being 60, 40, and 60 μM, respectively. SNP at 40 μM, 80 μM, and 20 μM concentrations was identified as the best elicitor for promoting the accumulation of bergenin, chlorogenic acid, and quercetin, respectively. This study established appropriate conditions for cultivating S. stolonifera callus and identified the optimal elicitor concentrations for maximizing secondary metabolite production.

Filamentous morphology: a new frontier for genetic modification of filamentous fungal cell factories

Filamentous fungi are a group of eukaryotic microorganisms widely found in nature. Some filamentous fungi have been developed as "cell factories" and extensively used for the production of recombinant proteins, organic acids, and secondary metabolites due to their strong protein secretion capabilities or effective synthesis of many natural products. The growth morphology of filamentous fungi significantly influences the quality and quantity of fermented products. Previous research conducted by the authors' group revealed that an increase in hyphal branches leads to enhanced protein secretion during liquid fermentation. With the development of morphological engineering of filamentous fungi, an increasing number of studies have focused on modifying fungal mycelium morphology to improve the yield of target metabolites during fermentation. While there have been a few reviews on the relationship between fungal fermentation morphology and productivity, research in this area is rapidly developing and requires updates. The paper presents a comprehensive review of domestic and international research reports, along with the authors' own research findings, to systematically review the morphological patterns of filamentous fungi, the impact of fungal morphology on industrial fermentation, as well as methods and strategies for regulating mycelial morphology. The aim of this review is to enhance the understanding of relevant domestic scholars regarding the morphological development of filamentous fungi and provide ideas for the rational engineering of fungal strains suitable for industrial fermentation.

Enhancement of anti-cancer compounds in fungal elicited-Oldenlandia umbellata culture.

Our study focused on enhancing the production of anthraquinone derivatives in Oldenlandia umbellata using fungal elicitors. Aspergillus niger, Mucor prayagensis, and Trichoderma viride were used to elicit the anthraquinone derivatives in root cultures. The elicitation process led to an increase in the production of phytochemicals and secondary metabolites, with the highest total protein content observed in A. niger-elicited plants. We performed qualitative and quantitative phytochemical screening of the 80% methanol extract of the plants. Using reverse phase-ultra-fast liquid chromatography, we identified and quantified five anthraquinone compounds: aloe-emodin, rhein, emodin, chrysophanol, and alizarin. The in vitro root samples elicited with A. niger and M. prayagensis exhibited four and three anthraquinone derivatives, respectively, whereas those elicited with T. viride showed only two derivatives. Interestingly, chrysophanol content was the highest in A. niger-elicited root samples. We constructed a system pharmacology framework consisting of 40 nodes and 45 edges with 34 interacting genes. We also identified human proteins that interact with these derivatives, and inferred their roles in cancer-associated pathways. These anthraquinone derivatives interact with various proteins in multiple pathways, including apoptosis, human cytomegalovirus infection, proteoglycans in cancer, MAPK signaling, and hepatitis C, highlighting their potential therapeutic applications in cancer treatment.

Impact of climate change on the yield of medicinal plants in recent years

Climate change significantly impacts the yield and quality of medicinal plants due to alterations in temperature, precipitation patterns, and increased frequency of extreme weather events. These changes affect the growth, secondary metabolite production, and geographical distribution of medicinal plants, leading to reduced yields and compromised medicinal properties. Adaptive strategies such as developing climate-resilient plant varieties, sustainable agricultural practices, and enhanced conservation efforts are essential to mitigate these effects. Increased research and collaborative efforts are necessary to safeguard these vital resources for future generations.

Optimization of polysaccharide conditions and analysis of antioxidant capacity in the co-culture of Sanghuangporus vaninii and Pleurotus sapidus

Fungal polysaccharides are commonly utilized in the food industry and biomedical fields as a natural and safe immune modulator. Co-culturing is a valuable method for enhancing the production of secondary metabolites. This study used intracellular polysaccharide (IPS) content as a screening index, co-culturing seven different fungi with Sanghuangporus vaninii. The seed pre-culture liquid culture time was selected through screening, and conditions were assessed using single factor experimentation, a Plackett-Burman (PB) design, and response surface methodology (RSM) optimization. RSM optimization was conducted, leading to the measurement of antioxidant capacity. Results indicated that the co-culture of S. vaninii and Pleurotus sapidus exhibited the most effective outcome. Specifically, pre-culturing S. vaninii and P. sapidus seed cultures for 2 days and 0 days, respectively, followed by co-culturing, significantly increased IPS content compared to single-strain culturing. Further optimization of co-culture conditions revealed that yeast extract concentration, liquid volume, and S. vaninii inoculum ratio notably influenced IPS content in the order of yeast extract concentration > liquid volume > S. vaninii inoculum ratio. Under the optimal conditions, IPS content reached 69.9626 mg/g, a 17.04% increase from pre-optimization co-culture conditions. Antioxidant capacity testing demonstrated that co-cultured IPS exhibited greater scavenging abilities for DPPH and ABTS free radicals compared to single strain cultures. These findings highlight the potential of co-culturing S. vaninii and P. sapidus to enhance IPS content and improve antioxidant capacity, presenting an effective strategy for increasing fungal polysaccharide production.

Not just candy: A herbivore‐induced defence‐related plant protein in honeydew enhances natural enemy fitness

Abstract Herbivore feeding often increases secondary metabolite production in plants. These herbivore‐induced plant proteins might end up in honeydew excreted by phloem‐feeding insects. This is important because honeydew is one of the most abundant and accessible carbohydrate sources for natural enemies in many agroecosystems and these proteins can thus mediate many tri‐trophic interactions. Here, we hypothesized that defensive metabolites induced in the phloem by herbivory accumulate in the honeydew excreted by phloem‐feeding insects and, consequently, affect the fitness of the herbivores' natural enemies that feed on it. We used a tri‐trophic system consisting of citrus plants, the mealybug Planococcus citri and its primary parasitoid Anagyrus vladimiri. First, we assessed A. vladimiri fitness when fed on P. citri honeydew. We then collected honeydew of seven phloem‐feeding insects, including P. citri, and analysed their protein content. Finally, we analysed the effect of superoxide dismutase (SOD), an antioxidant enzyme associated with plant defences that was commonly found in the analysed honeydews, on A. vladimiri fitness. The fitness of A. vladimiri increased when fed on honeydew compared to a sucrose‐based diet, demonstrating that honeydew can contain compounds that benefit natural enemies. Proteomic analyses showed that defence‐related plant proteins were present in honeydew of seven phloem‐feeding insects analysed. Among these, the enzyme SOD was present in honeydew of all of them. Moreover, the levels of SOD were 10‐fold higher in the phloem of plants infested by P. citri than in that of uninfested plants. SOD was also actively excreted in P. citri honeydew, and we proved that it increases the fecundity of the parasitoid A. vladimiri. We conclude that enzymatic proteins induced by herbivory in the phloem and involved in plant defence, accumulate in the honeydew excreted by phloem‐feeding insects and, contrary to the current paradigm, at least some of these can have positive effects on the third trophic level. Unveiling Nature’s Secrets: Activating Silent Biosynthetic Genes Clusters in Fungi and Bacteria

Microorganisms such as fungi and bacteria are prosperous producers of secondary metabolites; moreover, they are home to a large number of silent biosynthetic gene clusters, but they are all poorly expressed. Thus, these resultant compounds remain cryptic or unknown. However, since these cluster products have many bioactivities, stimulating their production and gaining access to their related structures are considered a top priority. Here, we summarize the three strategies that have been the topic of current trends. Regulating of genetic investigation, heterogeneous and refactoring cluster expression, as well as elicitor and coculture screening are the strategies utilized to unlock the expression of biosynthetic gene clusters (BGCs). These all help to triggers the manufacturing of innovative metabolites having various functions.

Melatonin: The Multifaceted Molecule in Plant Growth and Defense.

Melatonin (MEL), a hormone primarily known for its role in regulating sleep and circadian rhythms in animals, has emerged as a multifaceted molecule in plants. Recent research has shed light on its diverse functions in plant growth and defense mechanisms. This review explores the intricate roles of MEL in plant growth and defense responses. MEL is involved in plant growth owing to its influence on hormone regulation. MEL promotes root elongation and lateral root formation and enhances photosynthesis, thereby promoting overall plant growth and productivity. Additionally, MEL is implicated in regulating the circadian rhythm of plants, affecting key physiological processes that influence plant growth patterns. MEL also exhibits antioxidant properties and scavenges reactive oxygen species, thereby mitigating oxidative stress. Furthermore, it activates defense pathways against various biotic stressors. MEL also enhances the production of secondary metabolites that contribute to plant resistance against environmental changes. MEL's ability to modulate plant response to abiotic stresses has also been extensively studied. It regulates stomatal closure, conserves water, and enhances stress tolerance by activating stress-responsive genes and modulating signaling pathways. Moreover, MEL and nitric oxide cooperate in stress responses, antioxidant defense, and plant growth. Understanding the mechanisms underlying MEL's actions in plants will provide new insights into the development of innovative strategies for enhancing crop productivity, improving stress tolerance, and combating plant diseases. Further research in this area will deepen our knowledge of MEL's intricate functions and its potential applications in sustainable agriculture.

A Genomics-Based Discovery of Secondary Metabolite Biosynthetic Gene Clusters in the Potential Novel Strain Streptomyces sp. 21So2-11 Isolated from Antarctic Soil.

Streptomyces species are attractive sources of secondary metabolites that serve as major sources of antibiotics and other drugs. In this study, genome mining was used to determine the biosynthetic potential of Streptomyces sp. 21So2-11 isolated from Antarctic soil. 16S rRNA gene sequencing revealed that this strain is most closely related to Streptomyces drozdowiczii NBRC 101007T, with a similarity of 98.02%. Genome comparisons based on average nucleotide identity (ANI) and digital DNA-DNA hybridization (dDDH) showed that strain 21So2-11 represents a novel species of the genus Streptomyces. In addition to a large number of genes related to environmental adaptation and ecological function, a total of 28 putative biosynthetic gene clusters (BGCs) responsible for the biosynthesis of known and/or novel secondary metabolites, including terpenes, lantipeptides, polyketides, nonribosomal peptides, RiPPs and siderophores, were detected in the genome of strain 21So2-11. In addition, a total of 1456 BGCs were predicted to contribute to the biosynthesis of more than 300 secondary metabolites based on the genomes of 47 Streptomyces strains originating from polar regions. The results indicate the potential of Streptomyces sp. 21So2-11 for bioactive secondary metabolite production and are helpful for understanding bacterial adaptability and ecological function in cold terrestrial environments.

Volatile communication in Actinobacteria: a language for secondary metabolism regulation

BackgroundVolatile compounds are key elements in the interaction and communication between organisms at both interspecific and intraspecific levels. In complex bacterial communities, the emission of these fast-acting chemical messengers allows an exchange of information even at a certain distance that can cause different types of responses in the receiving organisms. The changes in secondary metabolism as a consequence of this interaction arouse great interest in the field of searching for bioactive compounds since they can be used as a tool to activate silenced metabolic pathways. Regarding the great metabolic potential that the Actinobacteria group presents in the production of compounds with attractive properties, we evaluated the reply the emitted volatile compounds can generate in other individuals of the same group.ResultsWe recently reported that volatile compounds released by different streptomycete species trigger the modulation of biosynthetic gene clusters in Streptomyces spp. which finally leads to the activation/repression of the production of secondary metabolites in the recipient strains. Here we present the application of this rationale in a broader bacterial community to evaluate volatiles as signaling effectors that drive the activation of biosynthesis of bioactive compounds in other members of the Actinobacteria group. Using cocultures of different actinobacteria (where only the volatile compounds reach the recipient strain) we were able to modify the bacterial secondary metabolism that drives overproduction (e.g., granaticins, actiphenol, chromomycins) and/or de novo production (e.g., collismycins, skyllamycins, cosmomycins) of compounds belonging to different chemical species that present important biological activities.ConclusionsThis work shows how the secondary metabolism of different Actinobacteria species can vary significantly when exposed in co-culture to the volatile compounds of other phylum-shared bacteria, these effects being variable depending on strains and culture media. This approach can be applied to the field of new drug discovery to increase the battery of bioactive compounds produced by bacteria that can potentially be used in treatments for humans and animals.

The Near-Gapless Penicillium fuscoglaucum Genome Enables the Discovery of Lifestyle Features as an Emerging Post-Harvest Phytopathogen.

Penicillium spp. occupy many diverse biological niches that include plant pathogens, opportunistic human pathogens, saprophytes, indoor air contaminants, and those selected specifically for industrial applications to produce secondary metabolites and lifesaving antibiotics. Recent phylogenetic studies have established Penicillium fuscoglaucum as a synonym for Penicillium commune, which is an indoor air contaminant and toxin producer and can infect apple fruit during storage. During routine culturing on selective media in the lab, we obtained an isolate of P. fuscoglaucum Pf_T2 and sequenced its genome. The Pf_T2 genome is far superior to available genomic resources for the species. Our assembly exhibits a length of 35.1 Mb, a BUSCO score of 97.9% complete, and consists of five scaffolds/contigs representing the four expected chromosomes. It was determined that the Pf_T2 genome was colinear with a type specimen P. fuscoglaucum and contained a lineage-specific, intact cyclopiazonic acid (CPA) gene cluster. For comparison, a highly virulent postharvest apple pathogen, P. expansum strain TDL 12.1, was included and showed a similar growth pattern in culture to our Pf_T2 isolate but was far more aggressive in apple fruit than P. fuscoglaucum. The genome of Pf_T2 serves as a major improvement over existing resources, has superior annotation, and can inform forthcoming omics-based work and functional genetic studies to probe secondary metabolite production and disparities in aggressiveness during apple fruit decay.
