Impact of arbuscular mycorrhizal fungi on maize rhizosphere microbiome stability under moderate drought conditions.

With an alarming increase in global greenhouse gas emissions, unstable weather conditions are significantly impacting agricultural production. Drought stress is one of the frequent consequences of climate change that affects crop growth and yield. Addressing this issue is critical to ensure stable crop productivity under drought conditions. Arbuscular mycorrhizal fungi (AMF) establish symbiotic relationships with plants and enhance their resistance to adverse conditions. Effects of arbuscular mycorrhizal associations on the rhizosphere microbiome and root transcriptome under drought conditions have not been explored. Here, we investigated the effects of AMF and drought stress on rhizosphere microorganisms and root transcriptome of maize plants grown in chernozem soil. We used high-throughput sequencing data of bacterial 16S rRNA and fungal internal transcribed spacer regions (ITS) to identify rhizosphere microorganisms. Transcriptomic data were used to assess gene expression in maize plants under different treatments. Our results show that AMF maintains the composition of maize rhizosphere microorganisms under drought stress. In particular, the bacterial and fungal phyla maintained were Actinomycetes and Ascomycota, respectively. Transcriptomic data indicated that AMF influenced gene expression in maize plants under drought stress. Under drought stress, the expression of SWEET13, CHIT3, and RPL23A was significantly higher in the presence of AMF than it was without AMF inoculation, indicating better sugar transport, reduced malondialdehyde accumulation, and improved water use efficiency in AMF-inoculated maize plants. These findings suggest that AMF can enhance the resistance of maize to moderate drought stress by stabilising plant physical traits, which may help maintain the structure of the rhizosphere microbial community. This study provides valuable theoretical insights that should aid the utilization of AMF in sustainable agricultural practices.

The Effects of Warming and Nitrogen Application on the Stoichiometric Characteristics of Arbuscular Mycorrhizal Fungi in Forest Ecosystems

Arbuscular mycorrhizal (AM) fungi are one of the most widely distributed microorganisms in all terrestrial ecosystems, playing an important role in supplying nitrogen (N) and phosphorus (P) to plants and in nutrient cycling. The contribution discusses the responses of rises in temperature and atmospheric N deposition to stoichiometric features of plant–soil–litter–microorganism–soil hydrolases in forest ecosystems. It summarizes the role that AM fungi play in the context of global change in carbon (C), N, and P stoichiometric characteristics of forest plant–soil systems. In this study, under conditions of warming with N application, it said the AM fungi will strongly influence the stoichiometric characteristics of C, N, and P in forest ecosystems. In addition to that, the presence of AM fungi may weaken certain impacts of global change on nutrient limitations in plants, increasing their plant dependency on mycorrhizal symbionts. AM fungi also seem to control soil nutrient transformation but simultaneously enhance soil nutrient stability, accelerate litter decomposition, and shorten the cycling time of litter C, N, and P.

Arbuscular mycorrhizal fungi regulation of soil carbon fractions in coal mining restoration: Dominance of glomalin-related soil proteins revealed by stable carbon isotopes (δ13C)

Arbuscular mycorrhizal fungi (AMF) play a pivotal role in soil carbon cycling by orchestrating the recruitment of microbial communities and establishing hyphal networks. Understanding whether these attributes of AMF are instrumental in restoring soil carbon pools in areas disrupted by coal mining is crucial for forecasting soil organic carbon (SOC) dynamics in post-disturbance restoration sites. This study conducted a year-long, in-situ leaf decomposition experiment (comprising three shrubs: Amorpha fruticose, Caragana, and Sea buckthorn; two treatments: inoculated with AMF and without AMF inoculation) at the mycorrhizal restoration demonstration base in Inner Mongolia's mining area. Utilizing stable isotope technology, we investigated the mycorrhizal regulatory effects on aggregate carbon dynamics during the leaf decomposition process. Our findings showed that the presence of AMF hyphae accelerated carbon transformation within aggregates, resulting in augmented content of mineral-bound carbon (+6.1 %) and δ13C abundance (7.1 %) within microaggregates in sea buckthorn plots. Analysis employing random forest and partial least squares regression models corroborated that the regulation of mycorrhizal dynamics on aggregate carbon composition after fresh leaf input was indirectly influenced by GRSP rather than directly by AMF hyphae. In summary, our findings elucidate the evolving patterns of aggregate carbon dynamics following AMF inoculation in coal mining reclamation sites under field conditions. This underscored the efficacy of planting sea buckthorn and inoculating AMF during the vegetation restoration process of open-pit mine reclamation land in Inner Mongolia.

Arbuscular Mycorrhizae Affect Soil Nitrogen Fertilizer Utilization, Denitrification Functional Genes, and N2O Emissions During Biochar Amendment

Arbuscular mycorrhizal fungi (AMF) can form symbionts with plant roots, acquire soil nitrogen, and affect nitrous oxide (N2O) production. Biochar, as a soil additive for the management of agricultural soil, affects soil nitrogen (N) utilization and plant growth. However, how AMF regulates soil N unitization, the denitrification process, and N2O emissions in plant–soil systems remains largely unknown, particularly under the biochar amendment. In this study, a microcosm experiment was conducted to investigate the impacts of different mycorrhizal treatments (CK: neither AMF hyphae nor plant roots; AMF: only AMF hyphae; AMF + R: AMF hyphae and plant roots) on plant growth, soil N fertilizer utilization, N2O production and consumption, functional gene abundance, and N2O emission at two biochar addition levels (B0: no biochar; B1: biochar addition rate of 10 g·kg−1 soil) in a maize planting soil system. The results revealed that AMF alone and AMF with plant root treatments enhanced the fresh weight of maize plants by 10.15% and 19.23% and decreased soil inorganic N contents by 33.28% and 75.56%, respectively. The combination of biochar, AMF, and plant roots showed the largest increase in maize plant biomass. The AMF and AMF with root treatments all significantly decreased the nirS + nirK/nosZ ratio and N2O emissions at two biochar levels. The presence of AMF and plant roots during biochar amendment showed the smallest nirS + nirK/nosZ ratio and N2O emissions. The AMF combined with biochar and AMF and plant roots combined with biochar treatments increased nirS + nirK/nosZ by 24.32% and 26.90% and decreased N2O accumulation emission by 21.12% and 38.13%, respectively. The results imply that biochar, AMF, and plant roots reduced N2O emissions directly by reducing soil N and increasing soil N unitization efficiency and indirectly by shifting the N2O production and consumption gene abundance in agroecosystems. These findings suggest that the addition of biochar and AMF and/or the presence of plant roots can interact to alleviate soil N2O emissions by manipulating plant inorganic N acquisition and the soil denitrification process.

Selection for agronomic traits in intermediate wheatgrass increases responsiveness to arbuscular mycorrhizal fungi

Societal Impact StatementAgricultural practices have had a negative impact on the physical, chemical, and biological components of soil. Perennial cropping systems that facilitate positive soil microbial interactions could not only rebuild soils but also sustain productivity through expected variations in environmental conditions. Here, we show the presence of arbuscular mycorrhizal (AM) fungi, soil symbionts that can improve host performance and soil health, increased the growth of intermediate wheatgrass, a novel perennial grain crop, in populations that have been increasingly bred for desirable agricultural characteristics. The right pairing of intermediate wheatgrass and a beneficial AM fungal community could lead to more sustainable agroecosystems.Summary Intermediate wheatgrass (IWG) is a novel perennial grain that can provide many soil health benefits in agroecosystems; however, little is known about how selection for agronomic traits has impacted interactions with soil biota. Here, we assess how the selection for agronomic traits in IWG has impacted its relationship with arbuscular mycorrhizal (AM) fungi. First, growth response to AM fungi was compared across five generations of IWG with varying degrees of selection. Second, variation in AM fungal responsiveness was compared among genets of IWG individuals within a more advanced generation. Finally, a meta‐analysis was performed on all published studies exploring AM fungal inocula effects on IWG performance to increase understanding of selection effects. AM fungal responsiveness increased with selection for agronomic traits, responsiveness varied among genets in the advanced generation, and a majority of genets performed better in the presence of AM fungi. The meta‐analysis supported the findings that AM fungal responsiveness has increased with selection in IWG. Further studies are needed to realize the combined potential soil health and sustainability benefits of IWG and AM fungi, including assessment of symbiotic benefits beyond biomass production, identification of IWG traits correlated with responsiveness, and characterization of AM fungal community response to IWG.

Anthoxanthum odoratum (Poaceae) as a bioindicator of arbuscular mycorrhizal fungi abundance prior to native revegetation on agricultural soils

ABSTRACT Mycorrhizal fungi, which form symbiotic associations with plant roots, can be critical to the successful establishment of plants. Up to 90% of land plant species form associations with arbuscular mycorrhizal fungi (AMF), which develop fungal structures inside host roots. Common site preparation practices prior to ecological restoration, such as soil disturbance and herbicide application, may lessen the availability of AMF, potentially jeopardising plant establishment. However, the state of the soil microbiome can be difficult to assess directly. This study examines the potential of the widespread pasture grass Anthoxanthum odoratum (Sweet Vernal, Poaceae) as a bioassay of AMF abundance in soils. We present a rapid clearing and staining protocol for visualising AMF structures in the roots of A. odoratum. We then compare % colonisation on roots of naturally established A. odoratum plants collected from undisturbed pastures vs. sprayed and tilled pastures. Roots of A. odoratum collected from sprayed and tilled pastures showed a significantly lower average AMF colonisation at 43.8% compared with 57.6% on roots from undisturbed pastures. However, all plants sourced from the sprayed and tilled pastures still showed some AMF structures. Our results strongly suggest that A. odoratum can be a useful bioindicator of AMF presence and abundance in soils, offering a promising tool for seed-based restoration on retired agricultural land.

Globally Distributed Arbuscular Mycorrhizal Fungi Associated With Invasive Cinchona pubescens on Santa Cruz Island, Galápagos.

The presence of arbuscular mycorrhizal fungi (AMF) is essential for the survival and establishment of most plant species in nature. The reproductive success of invasive plant species in a particular habitat could also depend on these AM fungi. Cinchona pubescens, commonly known as quinine, is highly invasive on Santa Cruz Island, Galápagos, but at the same time severely endangered in its native range on mainland Ecuador due to overexploitation in the past. In this study, we aim at determining the AMF communities associated with C. pubescens at both locations to investigate whether the successful invasion of C. pubescens on Santa Cruz is related to its association with a particular community of AMF. For this, roots of C. pubescens trees were sampled at three sites, one site on Santa Cruz and two sites in the province of Loja, on mainland Ecuador. Communities of AM fungi were determined through the molecular cloning and sequencing of the 18S nrDNA gene and through the delimitation of Operational Taxonomic Units (OTUs), associated with the plant roots. We found 36 AMF OTUs associated with C. pubescens, most of them belonging to the genus Glomus. The highest richness of AMF OTUs was detected in samples from sites located on mainland Ecuador. The AMF communities between Santa Cruz and mainland Ecuador were significantly different, and only five OTUs were shared between both locations. Two dominant OTUs in C. pubescens from Santa Cruz were detected but no dominant OTUs from mainland Ecuador. Almost two thirds of the OTUs associated with C. pubescens had a wide global distribution. Our results suggest that the successful invasion of C. pubescens on Santa Cruz could have been facilitated by local generalist AMF and not by particular AMF. The observed generalist AMF from both locations could be important for conservation plans of restoring the endangered C. pubescens in the native forests on mainland Ecuador.

Micorrizas arbusculares y endófitos septados en tres especies de Thelypteridaceae de ambientes riparios de las Yungas (Tucumán, Argentina)

Background and aims: The status of the presence of arbuscular mycorrhizal fungi (AMF) in lycophytes and ferns has been analysed in 11% of them. This work aims to characterise and quantify the AMF colonisation and to characterise septate endophytes present on the roots of the Thelypteridaceae: Amauropelta jujuyensis, A. nubicola, and Pelazoneuron patens. M&M: Ten individuals per species were collected in August 2022. Conventional staining and techniques were used to estimate colonisation. Results: The root of Amauropelta jujuyensis, A. nubicola, and Pelazoneuron patens showed AMF with Arum and Paris morphological types simultaneously. The hyphal colonization showed no significant difference among species, arbuscular colonization was significantly higher in A. jujuyensis, while vesicular colonization was considerably higher in A. nubicola. In addition, septate endophytes were evident in the species of the genus Amauropelta. Conclusion: The association of AMF and other fungal endophytes with the native Argentine Thelypteridaceae Amauropelta jujuyensis, A. nubicola, and Pelazoneuron patens is evidenced for the first time in riparian environments of the Yungas. The coexistence of Arum and Paris morphological types in the three studied fern species is demonstrated.

Mycorrhizal fungi compromise production of endophytic alkaloids, increasing plant susceptibility to an aphid herbivore

Abstract Symbiosis plays a critical role in plant biology. Temperate grasses often associate with several symbiotic fungi simultaneously, including Epichloë endophytes and arbuscular mycorrhizal (AM) fungi, in shoots and roots, respectively. These symbionts often modulate plant–herbivore interactions by influencing nutritional traits (i.e. AM fungi‐mediated nutrient uptake) and/or the secondary chemistry (i.e. endophytic alkaloids) of their host plant. Moreover, such grasses also accumulate large amounts of silicon (Si) from the soil, which can be deposited in tissues to act as a physical anti‐herbivore defence. Recent evidence suggests that both endophytes and AM fungi independently facilitate Si uptake. However, the consequences of their interactions with piercing‐sucking insects (i.e. aphids), or whether Si supply, endophytes, and AM fungi interact in this regard, are currently unknown. While Si deposition may be less effective against aphids than other herbivores (i.e. chewing caterpillars), Si supply can also alter plant secondary metabolite defences, which could affect sucking insects. In a factorial greenhouse experiment, we evaluated whether these components, acting alone or in combination, altered (1) foliar primary chemistry, (2) Si and symbiont‐chemical (endophytic alkaloids) defences, as well as (3) performance of the bird cherry‐oat aphid (Rhopalosiphum padi) feeding on tall fescue (Festuca arundinacea). Endophytes decreased all aphid performance parameters, including population growth and reproduction by 40%, but their impact was reversed by the presence of AM fungi, leading to a 52% increase in aphid performance compared with plants solely hosting endophytes. This improvement in performance was associated with reduced loline alkaloid levels and higher shoot nitrogen in AM‐endophytic plants. Endophytes and AM fungi exhibited antagonism, with endophytes reducing AM colonization by 34% and AM presence decreasing endophyte loline alkaloids by 44%. While both fungi jointly increased Si accumulation by 39% under Si‐supplied conditions, Si had no noticeable effects on aphids. Moreover, although Si supply had no identifiable effects on AM colonization, it reduced endophyte peramine alkaloids by 24%. Synthesis. Our findings indicate that symbiotic fungal partnerships and silicon provision may benefit plants but could weaken anti‐herbivore defences when combined. Revealing the complex interactions among diverse fungal symbionts and showcasing their effects on different anti‐herbivore defences (chemical and physical) and herbivore performance for the first time.

Arbuscular mycorrhizal fungi equalize differences in plant fitness and facilitate plant species coexistence through niche differentiation.

Mycorrhizal fungi are essential to the establishment of the vast majority of plant species but are often conceptualized with contradictory roles in plant community assembly. On the one hand, host-specific mycorrhizal fungi may allow a plant to be competitively dominant by enhancing growth. On the other hand, host-specific mycorrhizal fungi with different functional capabilities may increase nutrient niche partitioning, allowing plant species to coexist. Here, to resolve the balance of these two contradictory forces, we used a controlled greenhouse study to manipulate the presence of two main types of mycorrhizal fungus, ectomycorrhizal fungi and arbuscular mycorrhizal fungi, and used a range of conspecific and heterospecific competitor densities to investigate the role of mycorrhizal fungi in plant competition and coexistence. We find that the presence of arbuscular mycorrhizal fungi equalizes fitness differences between plants and stabilizes competition to create conditions for host species coexistence. Our results show how below-ground mutualisms can shift outcomes of plant competition and that a holistic view of plant communities that incorporates their mycorrhizal partners is important in predicting plant community dynamics.

Addition of polyester microplastic fibers to soil alters the diversity and abundance of arbuscular mycorrhizal fungi and affects plant growth and nutrition

Microplastics (MPs) represent an emerging factor in global environmental change and are increasingly found in soils. However, the extent to which they affect plants and their interactions with the soil microbiome is poorly understood. Here, we test the hypothesis that increasing levels of polyester MP fibers in soil alter plant growth and nutrient acquisition responses to arbuscular mycorrhizal (AM) fungi via changes in AM fungal colonization and community composition. We used Sorghum drummondii as a model species in a fully factorial greenhouse experiment. Plants were exposed to soil treatments with 0, 0.2, 1, and 3 % MP polyester fibers either in the presence or absence of an assembled AM fungal community comprising 13 species across three families with contrasting life-history strategies. We found that the 1 % MP treatment promoted plant biomass irrespective of the presence of AM fungi. While no changes in macronutrient concentrations in plant tissues were seen, there was a significant increase in B and Mn when relatively low amounts of MPs were added, and this effect was modulated by AM fungi. Furthermore, there were shifts in AM fungal community composition in response to MP, favoring taxa such as Gigaspora sp. while negatively affecting ruderal taxa like Glomus sp. Overall, our data indicate that MP polyester fibers present in soil can in some cases be beneficial to plants and AM fungal interactions. However, the implications of these findings over the long-term and in the context of ecological repercussions of MP pollution in the environment remain to be seen.

Enhancing olive tree (Olea europaea) rhizosphere dynamics: Co-inoculation effects of arbuscular mycorrhizal fungi and plant growth- promoting rhizobacteria in field experiments

We investigated the outcome of the interaction between the arbuscular mycorrhizal fungus (AMF) Rhizophagus (R) irregularis DAOM 197198 and the Plant-Growth-Promoting Rhizobacteria (PGPR) (mix of Bacillus megaterium, Burkholdria cedrus and Streptomyces beta-vulgaris) by conducting an olive field experiment. Our data provide evidence that the co-inoculation of R. irregularis and PGPR has important effects on the rhizosphere microbial community. The largest proportional increase was found for the PLFA biomarkers indicative of Gram-negative bacteria (16:1ω9, 18:1ω7 and 18:1ω9), fungi (18:2ω6) and actinobacteria (10Me16:0 and 10Me18:0). Microbial inoculants application of all tested treatments caused a significant decrease in the level of trehalose in the olive rhizosphere. The most pronounced decrease was observed in the plant inoculated with R. intraradices only, suggesting that the presence of AMF may have relaxed the bacterial stress. Co-inoculation of PGPR and AMF significantly improved the nutritional status of olive roots. Specifically, the interaction of PGPR and R. intraradices led to an increase in N (26 %), P (60 %), Fe (25 %), Mn (18 %), Zn (26 %), B (22 %) and Cu (14 %) compared with the control. We also found that the co-inoculation of AMF with PGPR causes a shift in the accumulation of secondary metabolites in olive roots. In particular, the most important effect induced by AMF was an improvement of oleuropein concentration, while co-inoculation of R. irregularis and PGPR positively modulated verbascoside concentration. The novelty of the present work lies in the use of microbial inoculants in the field of olive trees. This approach provided direct information regarding the advantages of using AMF and PGPR inoculants, allowing the reduction of chemical inputs and positively influencing the olive tree performance.

The costs and benefits of symbiotic interactions: variable effects of rhizobia and arbuscular mycorrhizae on Vigna radiata accessions

BackgroundThe symbiosis among plants, rhizobia, and arbuscular mycorrhizal fungi (AMF) is one of the most well-known symbiotic relationships in nature. However, it is still unclear how bilateral/tripartite symbiosis works under resource-limited conditions and the diverse genetic backgrounds of the host.ResultsUsing a full factorial design, we manipulated mungbean accessions/subspecies, rhizobia, and AMF to test their effects on each other. Rhizobia functions as a typical facilitator by increasing plant nitrogen content, plant weight, chlorophyll content, and AMF colonization. In contrast, AMF resulted in a tradeoff in plants (reducing biomass for phosphorus acquisition) and behaved as a competitor in reducing rhizobia fitness (nodule weight). Plant genotype did not have a significant effect on AMF fitness, but different mungbean accessions had distinct rhizobia affinities. In contrast to previous studies, the positive relationship between plant and rhizobia fitness was attenuated in the presence of AMF, with wild mungbean being more responsive to the beneficial effect of rhizobia and attenuation by AMF.ConclusionsWe showed that this complex tripartite relationship does not unconditionally benefit all parties. Moreover, rhizobia species and host genetic background affect the symbiotic relationship significantly. This study provides a new opportunity to re-evaluate the relationships between legume plants and their symbiotic partners.

Testing the joint effects of arbuscular mycorrhizal fungi and ants on insect herbivory on potato plants

Main conclusionAnts, but not mycorrhizae, significantly affected insect leaf-chewing herbivory on potato plants. However, there was no evidence of mutualistic interactive effects on herbivory.Plants associate with both aboveground and belowground mutualists, two prominent examples being ants and arbuscular mycorrhizal fungi (AMF), respectively. While both of these mutualisms have been extensively studied, joint manipulations testing their independent and interactive (non-additive) effects on plants are rare. To address this gap, we conducted a joint test of ant and AMF effects on herbivory by leaf-chewing insects attacking potato (Solanum tuberosum) plants, and further measured plant traits likely mediating mutualist effects on herbivory. In a field experiment, we factorially manipulated the presence of AMF (two levels: control and mycorrhization) and ants (two levels: exclusion and presence) and quantified the concentration of leaf phenolic compounds acting as direct defenses, as well as plant volatile organic compound (VOC) emissions potentially mediating direct (e.g., herbivore repellents) or indirect (e.g., ant attractants) defense. Moreover, we measured ant abundance and performed a dual-choice greenhouse experiment testing for effects of VOC blends (mimicking those emitted by control vs. AMF-inoculated plants) on ant attraction as a mechanism for indirect defense. Ant presence significantly reduced herbivory whereas mycorrhization had no detectable influence on herbivory and mutualist effects operated independently. Plant trait measurements indicated that mycorrhization had no effect on leaf phenolics but significantly increased VOC emissions. However, mycorrhization did not affect ant abundance and there was no evidence of AMF effects on herbivory operating via ant-mediated defense. Consistently, the dual-choice assay showed no effect of AMF-induced volatile blends on ant attraction. Together, these results suggest that herbivory on potato plants responds mainly to top-down (ant-mediated) rather than bottom-up (AMF-mediated) control, an asymmetry in effects which could have precluded mutualist non-additive effects on herbivory. Further research on this, as well as other plant systems, is needed to examine the ecological contexts under which mutualist interactive effects are more or less likely to emerge and their impacts on plant fitness and associated communities.

Arbuscular Mycorrhizal Fungi Mediate the Acclimation of Rice to Submergence.

Flooding is a critical factor that limits the establishment of a symbiosis between rice and arbuscular mycorrhizal fungi (AMF) in wetland ecosystems. The distribution of carbon resources in roots and the acclimation strategies of rice to flooding stress in the presence of AMF are poorly understood. We conducted a root box experiment, employing nylon sheets or nylon meshes to create separate fungal chambers that either prevented or allowed the roots and any molecules to pass through. We found that the mycorrhizal colonization rate and the expression of genes OsD14L and OsCERK1, which are involved in fungal perception during symbiosis, both increased in mycorrhizal rice roots following intermittent flooding compared to continuous flooding. Furthermore, AMF inoculation affected root morphological traits, facilitating both shallower and deeper soil exploration. Increased submergence intensity led to carbohydrate deprivation in roots, while high mycorrhizal colonization increased soil oxygen consumption and decreased the neutral lipid concentration in roots. However, mycorrhizal inoculation increased the rice photosynthesis rate and facilitated acclimation to submergence by mediating the expression of the genes OsCIPK15 and OsSUB1A to enhance rice shoot elongation and the sugar concentration in roots as a result of reduced competition for carbon between rice and AMF under different flooding conditions.

Native Arbuscular Mycorrhizal Fungi Promote Plukenetia volubilis Growth and Decrease the Infection Levels of Meloidogyne incognita.

The use of arbuscular mycorrhizal fungi (AMF) offers promising benefits to agriculture in the Amazon regions, where soils are characteristically acidic and nutrient-poor. The purpose of this research was to investigate the potential effects of two recently described species of AMF (Nanoglomus plukenetiae and Rhizoglomus variabile) native to the Peruvian Amazon for improving the plant growth of Plukenetia volubilis (inka nut or sacha inchi) and protecting the roots against soil pathogens. Two assays were simultaneously conducted under greenhouse conditions in Peru. The first focused on evaluating the biofertilizer effect of AMF inoculation, while the second examined the bioprotective effect against the root knot nematode, Meloidogyne incognita. Overall, the results showed that AMF inoculation of P. volubilis seedlings positively improved their development, particularly their biomass, height, and the leaf nutrient contents. When seedlings were exposed to M. incognita, plant growth was also noticeably higher for AMF-inoculated plants than those without AMF inoculation. Nematode reproduction was significantly suppressed by the presence of AMF, in particular R. variabile, and especially when inoculated prior to nematode exposure. The dual AMF inoculation did not necessarily lead to improved crop growth but notably improved P and K leaf contents. The findings provide strong justification for the development of products based on AMF as agro-inputs to catalyze nutrient use and uptake and protect crops against pests and diseases, especially those that are locally adapted to local crops and cropping conditions.

Crab bioturbation reduces carbon storage in salt marshes under more robust mechanisms than plant invasiveness

IntroductionThe macrobenthos are crucial for the stability of estuarine ecosystems due to their burrowing behavior in the sediment and their uptake of nutrients from plants. These activities lead to significant alterations in both the morphological and biogeochemical processes within the region.MethodsHowever, there is limited investigation into carbon cycling processes specifically related to crab bioturbation. Additionally, few studies have examined enzymatic activities and carbon fractions in sediments from crab burrow walls at different crab growth stages. This study aims to explore the impact of both plant invasiveness and crab bioturbation on carbon storage.ResultsOur findings suggest that plant invasion leads to higher organic accumulation due to the reduction of recalcitrant organic carbon (ROC) (decreased by 11.6% in invasive and 62.6% in native site from April to December), faster photosynthetic rates (25.8 μmol/m2 s in invasive and 10.7 μmol/m2 s in native site), and an increased presence of arbuscular mycorrhizal fungi (AMF) in the soil over time. However, the increase of easy oxidized carbon (EOC) may lead to less carbon storage in soil (increase by 67.7% in invasive and 48.8% in native site from April to December). In addition to invasiveness, the bioturbation activities of macrobenthos also affect carbon storage. Sediments from crab burrows exhibit higher EOC content (33.6% more than the bulk soil) and higher levels of carbon cycling-related enzymes, including S-ACT, S-β-GC, and S-ACP activities (24.2%, 8.99%, and 135.6% higher than the bulk soil, respectively).DiscussionThese changes contribute to reduced carbon accumulation in the soil. Therefore, crab bioturbation is a more significant factor affecting carbon sink capacity than plant invasion.

Soil compaction reversed the effect of arbuscular mycorrhizal fungi on soil hydraulic properties

Arbuscular mycorrhizal fungi (AMF) typically provide a wide range of nutritional benefits to their host plants, and their role in plant water uptake, although still controversial, is often cited as one of the hallmarks of this symbiosis. Less attention has been dedicated to other effects relating to water dynamics that the presence of AMF in soils may have. Evidence that AMF can affect soil hydraulic properties is only beginning to emerge. In one of our recent experiments with dwarf tomato plants, we serendipitously found that the arbuscular mycorrhizal fungus (Rhizophagus irregularis ‘PH5’) can slightly but significantly reduce water holding capacity (WHC) of the substrate (a sand–zeolite–soil mixture). This was further investigated in a subsequent experiment, but there we found exactly the opposite effect as mycorrhizal substrate retained more water than did the non-mycorrhizal substrate. Because the same substrate was used and other conditions were mostly comparable in the two experiments, we explain the contrasting results by different substrate compaction, most likely caused by different pot shapes. It seems that in compacted substrates, AMF may have no effect upon or even decrease the substrates’ WHC. On the other hand, the AMF hyphae interweaving the pores of less compacted substrates may increase the capillary movement of water throughout such substrates and cause slightly more water to remain in the pores after the free water has drained. We believe that this phenomenon is worthy of mycorrhizologists’ attention and merits further investigation as to the role of AMF in soil hydraulic properties.

Computer vision models enable mixed linear modeling to predict arbuscular mycorrhizal fungal colonization using fungal morphology

The presence of Arbuscular Mycorrhizal Fungi (AMF) in vascular land plant roots is one of the most ancient of symbioses supporting nitrogen and phosphorus exchange for photosynthetically derived carbon. Here we provide a multi-scale modeling approach to predict AMF colonization of a worldwide crop from a Recombinant Inbred Line (RIL) population derived from Sorghum bicolor and S. propinquum. The high-throughput phenotyping methods of fungal structures here rely on a Mask Region-based Convolutional Neural Network (Mask R-CNN) in computer vision for pixel-wise fungal structure segmentations and mixed linear models to explore the relations of AMF colonization, root niche, and fungal structure allocation. Models proposed capture over 95% of the variation in AMF colonization as a function of root niche and relative abundance of fungal structures in each plant. Arbuscule allocation is a significant predictor of AMF colonization among sibling plants. Arbuscules and extraradical hyphae implicated in nutrient exchange predict highest AMF colonization in the top root section. Our work demonstrates that deep learning can be used by the community for the high-throughput phenotyping of AMF in plant roots. Mixed linear modeling provides a framework for testing hypotheses about AMF colonization phenotypes as a function of root niche and fungal structure allocations.

Arbuscular mycorrhizal fungi reduce soil N2O emissions by altering root traits and soil denitrifier community composition

Arbuscular mycorrhizal fungi (AMF) increase the ability of plants to obtain nitrogen (N) from the soil, and thus can affect emissions of nitrous oxide (N2O), a long-lived potent greenhouse gas. However, the mechanisms underlying the effects of AMF on N2O emissions are still poorly understood, particularly in agroecosystems with different forms of N fertilizer inputs. Utilizing a mesocosm experiment in field, we examined the effects of AMF on N2O emissions via their influence on maize root traits and denitrifying microorganisms under ammonia and nitrate fertilizer input using 15N isotope tracer. Here we show that the presence of AMF alone or both maize roots and AMF increased maize biomass and their 15N uptake, root length, root surface area, and root volume, but led to a reduction in N2O emissions under both N input forms. Random forest model showed that root length and surface area were the most important predictors of N2O emissions. Additionally, the presence of AMF reduced the (nirK + nirS)/nosZ ratio by increasing the relative abundance of nirS-Bradyrhizobium and Rubrivivax with ammonia input, but reducing nosZ-Azospirillum, Cupriavidus and Rhodopseudomonas under both fertilizer input. Further, N2O emissions were significantly and positively correlated with the nosZ-type Azospirillum, Cupriavidus and Rhodopseudomonas, but negatively correlated with the nirS-type Bradyrhizobium and Rubrivivax. These results indicate that AMF reduce N2O emissions by increasing root length to explore N nutrients and altering the community composition of denitrifiers, suggesting that effective management of N fertilizer forms interacting with the rhizosphere microbiome may help mitigate N2O emissions under future N input scenarios.
