Pattern of Antibiotic Consumption in Two Italian Production Chains Differing by the Endemic Status for Porcine Reproductive and Respiratory Syndrome.

The aim of this case study was to quantify antibiotic (AB) use in Italian weaning (W) and fattening (F) units differentiated for porcine reproductive and respiratory syndrome (PRRS) occurrence. Farms were classified as either PRRS negative (–) or PRRS positive (+) based on the circulation of the virus among the animals. In all the farms, the modified live PRRS virus (PRRSV) vaccine was provided to all the animals. In the PRRS– farms, the level of circulating antibodies was low, and the disease, in its clinical form, did not occur. In the PRRS+ farms, the level of circulating antibodies against the virus was high, and the disease was recurrent. Data regarding AB consumption were collected from 2017 to 2020, and the active compounds (ACs) were expressed as milligrams of AC/total kilogram of body weight (BW) produced. Each AC was classified into one of four categories according to the European Medicines Agency classification of ABs for prudent and responsible use in animals: Avoid, Restrict, Caution, and Prudence. Data regarding the ACs in each category were analyzed using a linear model that included production phase, PRRS status, and their interaction as factors. Performance parameters, average age of the pigs at the end of each phase, daily live weight gain, feed-to-gain ratio, total losses, cost index, and medication costs were significantly influenced by the PRRS chain. The use of class B ABs was not affected by production phase or PRRS status. Conversely, for class C ABs, interaction between the two factors (p = 0.02) was observed; W/PRRS+ and F/PRRS+ showed the greatest AB use for this class (p = 0.003). For class D ABs, the interaction was significant (p = 0.01); class C and D ABs were used more in the weaning (p = 0.07) than in the fattening phase (p = 0.003). For the weaning phase, the use of class C and D ABs was greater in the PRRS+ than in the PRRS– chain (p < 0.01). In conclusion, PRRS status affected the growth of pigs and economic performance. Moreover, PRRS status significantly influenced the use of ABs during all the growing periods with the greatest impact being on the weaning phase.

Comparison of performances between three commercial ELISA kits for detection of antibodies against Porcine Reproductive and Respiratory Syndrome Virus (PPRSV) in swine sera samples

Porcine Reproductive and Respiratory Syndrome (PRRS) is one of the most economically important diseases in pigs, worldwide. Just in the US, the total costs to the swine industry have been estimated at $664 million per year. Therefore, the continuous and reliable monitoring of the PRRS status of a pig herd is required in order to prevent and reduce costs due to this infection. Mostly used methods for diagnosis of PRRS infection nowadays are serological (ELISA) and molecular (PCR) ones. This study aimed to assess the sensitivity and specificity of three different commercially available ELISA kits for detection of antibodies against PRRSV (IDEXX PRRS X3 Ab Test (IDEXX, USA), INgezim PRRS Universal (Ingenasa, Spain), Pigtype PRRSV Ab (Qiagen, Germany)) using 91 blood serum samples collected from pigs in Serbia. Our study showed no significant differences in specificity and sensitivity between three commercially available ELISA kits. However, IDEXX ELISA proved to be more reliable kit for detecting antibodies against PRRSV with sensitivity of 97,4% and specificity of 98,1%, considering INgezim and Qiagen kits specificity of 92,5% and 83%, respectively, and sensitivity of 94,7 % for both kits. In order to achieve maximal reliability of obtained results, ELISA diagnostic protocol for diagnosis of PRRS infection should be complemented with additional tests such as PCR and virus neutralization test.

Assessment of reproductive and growth performance of pigs on commercial swine farms in southern Kyushu, Japan.

The objective of the present study was to assess the productivity of pigs to investigate the time-dependent change in productivity and compare productivity according to breeding company group and porcine reproductive and respiratory syndrome (PRRS) status on commercial swine farms in southern Kyushu, Japan. Data used in the present study were 245 annual productivity records from 2014 to 2018 obtained from 72 farms. Average sow inventory was 481.2 sows. Mean numbers of pigs weaned per sow per year and market pigs sold per sow per year were 23.7 and 21.6, respectively. Pigs born alive per litter increased from 11.1 to 11.9 pigs from 2014 to 2018 (p<.05). Farms using domestic breeding companies had similar numbers in 2016 to those using international breeding companies, but fewer pigs in 2014, 2015, 2017, and 2018 (p<.05). Farms with an unknown or unstable PRRS status had fewer pigs born alive per litter and pigs weaned per sow per year than those with stable or negative PRRS statuses (11.2±0.06 vs. 11.8±0.08 pigs and 22.6±0.38 vs. 25.0±0.22 pigs, respectively; p<.05). These results can be used to establish feasible targets and standards of performance to identify problem areas and improve production.

Development of a biosecurity assessment tool and the assessment of biosecurity levels by this tool on Japanese commercial swine farms

It is well known that infectious diseases such as porcine reproductive and respiratory syndrome (PRRS) and porcine epidemic diarrhea (PED) decrease herd productivity and lead to economic loss. It is believed that biosecurity practices are effective for the prevention and control of such infectious diseases. Therefore, the objective of the present study was to investigate whether or not an association between biosecurity level and herd productivity, as well as disease status exists on Japanese commercial swine farms. The present study was conducted on 141 farms. Biosecurity in each farm was assessed by a biosecurity assessment tool named BioAsseT. BioAsseT has a full score of 100 and consists of three sections (external biosecurity, internal biosecurity and diagnostic monitoring). Production data for number of pigs weaned per sow per year (PWSY) and post-weaning mortality per year (PWM) were collected for data analysis. Regarding PRRS status, the farms were categorized into two groups: unknown or unstable and stable or negative. In addition, these farms were categorized based on their PED status, either positive or negative. The total BioAsseT score was associated with herd productivity: as total score increased by 1, PWSY increased by 0.104 pigs and PWM decreased by 0.051 % (P < 0.05). Herd productivity was associated with the score of external and internal biosecurity (P < 0.05), but did not correlate with the score of diagnostic monitoring. Regarding PRRS status, farms with an unknown or unstable status had lower total score than those with stable or negative status (P < 0.05). Similarly, PED positive farms had a lower total score compared to PED negative farms (P < 0.05). In conclusion, the present study provides evidence for the association between high biosecurity levels and increased herd productivity as well as a decreased risk for novel introductions of infectious diseases such as PED.

Effects of high inclusion of soybean meal and a phytase superdose on growth performance of weaned pigs housed under the rigors of commercial conditions.

Two studies were conducted to determine whether soybean meal (SBM) use in nursery pig diets can be increased by superdosing with phytase. In Exp. 1, 2,550 pigs (BW of 5.54 ± 0.09 kg) were used to evaluate the optimal level of phytase in low- or high-SBM diets. Two SBM levels (low and high) and 4 phytase doses (0, 1,250, 2,500, and 3,750 phytase units [FTU]/kg) were combined to create 8 dietary treatments in a 2 × 4 factorial arrangement. Pigs were fed a 3-phase feeding program, with each period being 10, 10, and 22 d, respectively. Inclusion of low and high SBM was 15.0 and 25.0%, respectively, for Phase 1; 19.0 and 29.0%, respectively, for Phase 2; and 32.5% for the common Phase 3 diet. Pigs fed diets with high SBM had improved G:F for Phase 1 and 2 and overall ( < 0.01) compared with low-SBM diets. Phytase quadratically improved G:F during Phase 3 and overall ( < 0.05), with the optimum phytase dose being 2,500 FTU/kg. High-SBM diets tended ( = 0.09) to decrease stool firmness (determined daily from d 1 to 10) only on d 2. In Exp. 2, 2,112 pigs (BW of 5.99 ± 0.10 kg) were used to evaluate the impact of high levels of SBM and phytase on performance, stool firmness, mortality, and morbidity in weaned pigs originating from a porcine reproductive and respiratory syndrome (PRRS) virus-positive sow farm. Pigs were fed a 3-phase feeding program as in Exp. 1. Three levels of SBM (low, medium, or high) and 2 phytase levels (600 or 2,600 FTU) were combined to create 6 dietary treatments in a 3 × 2 factorial arrangement. Inclusion of SBM was 15.0, 22.5, and 30.0% for Phase 1 and 20.0, 27.5, and 35.0% for Phase 2 for low, medium, and high SBM, respectively, and 29.0% for the common Phase 3 diet. Inclusion of SBM did not affect growth performance. The percentage of pigs removed for medical treatment linearly declined with increasing SBM levels ( = 0.04). High-SBM diets tended ( < 0.10) to decrease stool firmness during d 4 and 5 and high phytase tended ( < 0.10) to improve stool firmness on d 2 and 4. Analyzed PRRS titers in saliva samples collected on d 20 and 42 confirmed the PRRS status of the pigs; however, viral load was not impacted by dietary treatments ( ≥ 0.11). Results indicate that SBM levels in early nursery diets can be increased without decreasing growth performance and may be favorable in pigs originating from PRRS-positive sow farms by reducing costs of medical treatments. Supplementation of phytase at superdose levels can improve growth performance independently from the level of SBM in the diet.

A Review of Quantitative Tools Used to Assess the Epidemiology of Porcine Reproductive and Respiratory Syndrome in U.S. Swine Farms Using Dr. Morrison’s Swine Health Monitoring Program Data

Porcine reproductive and respiratory syndrome (PRRS) causes far-reaching financial losses to infected countries and regions, including the U.S. The Dr. Morrison’s Swine Health Monitoring Program (MSHMP) is a voluntary initiative in which producers and veterinarians share sow farm PRRS status weekly to contribute to the understanding, in quantitative terms, of PRRS epidemiological dynamics and, ultimately, to support its control in the U.S. Here, we offer a review of a variety of analytic tools that were applied to MSHMP data to assess disease dynamics in quantitative terms to support the decision-making process for veterinarians and producers. Use of those methods has helped the U.S. swine industry to quantify the cyclical patterns of PRRS, to describe the impact that emerging pathogens has had on that pattern, to identify the nature and extent at which environmental factors (e.g., precipitation or land cover) influence PRRS risk, to identify PRRS virus emerging strains, and to assess the influence that voluntary reporting has on disease control. Results from the numerous studies reviewed here provide important insights into PRRS epidemiology that help to create the foundations for a near real-time prediction of disease risk, and, ultimately, will contribute to support the prevention and control of, arguably, one of the most devastating diseases affecting the North American swine industry. The review also demonstrates how different approaches to analyze and visualize the data may help to add value to the routine collection of surveillance data and support infectious animal disease control.

Land altitude, slope, and coverage as risk factors for Porcine Reproductive and Respiratory Syndrome (PRRS) outbreaks in the United States.

Porcine reproductive and respiratory syndrome (PRRS) is, arguably, the most impactful disease on the North American swine industry. The Swine Health Monitoring Project (SHMP) is a national volunteer initiative aimed at monitoring incidence and, ultimately, supporting swine disease control, including PRRS. Data collected through the SHMP currently represents approximately 42% of the sow population of the United States. The objective of the study here was to investigate the association between geographical factors (including land elevation, and land coverage) and PRRS incidence as recorded in the SHMP. Weekly PRRS status data from sites participating in the SHMP from 2009 to 2016 (n = 706) was assessed. Number of PRRS outbreaks, years of participation in the SHMP, and site location were collected from the SHMP database. Environmental features hypothesized to influence PRRS risk included land coverage (cultivated areas, shrubs and trees), land altitude (in meters above sea level) and land slope (in degrees compared to surrounding areas). Other risk factors considered included region, production system to which the site belonged, herd size, and swine density in the area in which the site was located. Land-related variables and pig density were captured in raster format from a number of sources and extracted to points (farm locations). A mixed-effects Poisson regression model was built; and dependence among sites that belonged to a given production system was accounted for using a random effect at the system level. The annual mean and median number of outbreaks per farm was 1.38 (SD: 1.6), and 1 (IQR: 2.0), respectively. The maximum annual number of outbreaks per farm was 9, and approximately 40% of the farms did not report any outbreak. Results from the final multivariable model suggested that increments of swine density and herd size increased the risk for PRRS outbreaks (P < 0.01). Even though altitude (meters above sea level) was not significant in the final model, farms located in terrains with a slope of 9% or higher had lower rates of PRRS outbreaks compared to farms located in terrains with slopes lower than 2% (P < 0.01). Finally, being located in an area of shrubs/ herbaceous cover and trees lowered the incidence rate of PRRS outbreaks compared to being located in cultivated/ managed areas (P < 0.05). In conclusion, highly inclined terrains were associated with fewer PRRS outbreaks in US sow farms, as was the presence of shrubs and trees when compared to cultivated/ managed areas. Influence of terrain characteristics on spread of airborne diseases, such as PRRS, may help to predicting disease risk, and effective planning of measures intended to mitigate and prevent risk of infection.

Network, cluster and risk factor analyses for porcine reproductive and respiratory syndrome using data from swine sites participating in a disease control program

The objectives of this study were to describe networks of Ontario swine sites and their service providers (including trucking, feed, semen, gilt and boar companies); to categorize swine sites into clusters based on site-level centrality measures, and to investigate risk factors for porcine reproductive and respiratory syndrome (PRRS) using information gathered from the above-mentioned analyses. All 816 sites included in the current study were enrolled in the PRRS area regional control and elimination projects in Ontario. Demographics, biosecurity and network data were collected using a standardized questionnaire and PRRS status was determined on the basis of available diagnostic tests and assessment by site veterinarians. Two-mode networks were transformed into one-mode dichotomized networks. Cluster and risk factor analyses were conducted separately for breeding and growing pig sites. In addition to the clusters obtained from cluster analyses, other explanatory variables of interest included: production type, type of animal flow, use of a shower facility, and number of neighboring swine sites within 3km. Unadjusted univariable analyses were followed by two types of adjusted models (adjusted for production systems): a generalizing estimation equation model (GEE) and a generalized linear mixed model (GLMM). Results showed that the gilt network was the most fragmented network, followed by the boar and truck networks. Considering all networks simultaneously, approximately 94% of all swine sites were indirectly connected. Unadjusted risk factor analyses showed significant associations between almost all predictors of interest and PRRS positivity, but these disappeared once production system was taken into consideration. Finally, the vast majority of the variation on PRRS status was explained by production system according to GLMM, which shows the highly correlated nature of the data, and raises the point that interventions at this level could potentially have high impact in PRRS status change and/or maintenance.

Measuring Progress on the Control of Porcine Reproductive and Respiratory Syndrome (PRRS) at a Regional Level: The Minnesota N212 Regional Control Project (Rcp) as a Working Example.

Due to the highly transmissible nature of porcine reproductive and respiratory syndrome (PRRS), implementation of regional programs to control the disease may be critical. Because PRRS is not reported in the US, numerous voluntary regional control projects (RCPs) have been established. However, the effect of RCPs on PRRS control has not been assessed yet. This study aims to quantify the extent to which RCPs contribute to PRRS control by proposing a methodological framework to evaluate the progress of RCPs. Information collected between July 2012 and June 2015 from the Minnesota Voluntary Regional PRRS Elimination Project (RCP-N212) was used. Demography of premises (e.g. composition of farms with sows = SS and without sows = NSS) was assessed by a repeated analysis of variance. By using general linear mixed-effects models, active participation of farms enrolled in the RCP-N212, defined as the decision to share (or not to share) PRRS status, was evaluated and used as a predictor, along with other variables, to assess the PRRS trend over time. Additionally, spatial and temporal patterns of farmers’ participation and the disease dynamics were investigated. The number of farms enrolled in RCP-N212 and its geographical coverage increased, but the proportion of SS and NSS did not vary significantly over time. A significant increasing (p<0.001) trend in farmers’ decision to share PRRS status was observed, but with NSS producers less willing to report and a large variability between counties. The incidence of PRRS significantly (p<0.001) decreased, showing a negative correlation between degree of participation and occurrence of PRRS (p<0.001) and a positive correlation with farm density at the county level (p = 0.02). Despite a noted decrease in PRRS, significant spatio-temporal patterns of incidence of the disease over 3-weeks and 3-kms during the entire study period were identified. This study established a systematic approach to quantify the effect of RCPs on PRRS control. Despite an increase in number of farms enrolled in the RCP-N212, active participation is not ensured. By evaluating the effect of participation on the occurrence of PRRS, the value of sharing information among producers may be demonstrated, in turn justifying the existence of RCPs.

Evaluation of porcine reproductive and respiratory syndrome virus (PRRSV) in Binh Duong province by Elisa and RT-PCR

Porcine reproductive and respiratory syndrome (PRRS), which cause heavy losses to the breeding economy in many countries, including Vietnam, has been a top concern of the farm. Testing PRRSV antibodies and the presence of PRRSV have an important role in the evaluation and control of PRRS. The method used to diagnose PRRS are ELISA and RT-PCR, however, these methods are often used individually, not combined, lead to errors in judgment PRRS status of the herd. In this study, ELISA, RT-PCR and PCR steps 2 are combined, to assess the status of the herd PRRS circulation both antigens and antibodies. As a result, ELISA (+) - RT-PCR (-) accounted for the highest proportion of 48% (36/75). Combination ELISA (-) - RT-PCR (+) accounted for 5.333% (4/75) and this consortium focused in farm 3. Combination ELISA (+) - RT-PCR (+) accounted for 4% (3/75). In a total of 75 pigs, there were 42.67% negative for both reactions. The pig was the conclusion is negative for PRRSV and retained.

Spread of porcine circovirus associated disease (PCVAD) in Ontario (Canada) swine herds: Part I. Exploratory spatial analysis

BackgroundThe systemic form of porcine circovirus associated disease (PCVAD), also known as postweaning multisystemic wasting syndrome (PMWS) was initially detected in the early 1990s. Starting in 2004, the Canadian swine industry experienced considerable losses due to PCVAD, concurrent with a shift in genotype of porcine circovirus type 2 (PCV2). Objectives of the current study were to explore spatial characteristics of self-reported PCVAD distribution in Ontario between 2004 and 2008, and to investigate the existence and nature of local spread.ResultsThe study included 278 swine herds from a large disease-monitoring project that included porcine reproductive and respiratory syndrome (PRRS) virus-positive herds identified by the diagnostic laboratory, and PRRS virus-negative herds directly from the target population. Herds were included if they had growing pigs present on-site and available geographical coordinates for the sampling site. Furthermore, herds were defined as PCVAD-positive if a producer reported an outbreak of circovirus associated disease, or as PCVAD-negative if no outbreak was noted. Spatial trend was investigated using generalized additive models and time to PCVAD outbreak in a herd using Cox's proportional hazard model; spatial and spatio-temporal clustering was explored using K-functions; and location of most likely spatial and spatio-temporal clusters was investigated using scan statistics. Over the study period, the risk of reporting a PCVAD-positive herd tended to be higher in the eastern part of the province after adjustment for herd PRRS status (P = 0.05). This was partly confirmed for spread (Partial P < 0.01). Local spread also appeared to exist, as suggested by the tentative (P = 0.06) existence of spatio-temporal clustering of PCVAD and detection of a spatio-temporal cluster (P = 0.04).ConclusionsIn Ontario, PCVAD has shown a general trend, spreading from east-to-west. We interpret the existence of spatio-temporal clustering as evidence of spatio-temporal aggregation of PCVAD-positive cases above expectations and, together with the existence of spatio-temporal and spatial clusters, as suggestive of apparent local spread of PCVAD. Clustering was detected at small spatial and temporal scales. Other patterns of spread could not be detected; however, survival rates in discrete Ontario zones, as well as a lack of a clear spatial pattern in the most likely spatio-temporal clusters, suggest other between-herd transmission mechanisms.

Heat and social rank impact behavior and physiology of PRRS-virus-infected pigs

Changes in thermal environment can invoke a stress response in pigs, which in turn can potentially impact their immune system and disease susceptibility. We investigated effects of heat stress and social rank on behavior, immune responsiveness, and performance of pigs challenged with porcine reproductive and respiratory syndrome (PRRS) virus. Sixty-four 7-week-old PRRS-naïve pigs were assigned to each of four experimental treatments consisting of a 2 × 2 factorial design: PRRS (PRRS− or PRRS+) and environmental temperature (24 °C or 32 °C). Blood samples were taken prior to and at days 7 and 14 post-inoculation, and alveolar macrophages were collected via bronchoalveolar lavage at day 14. Total white blood cell counts, natural killer cytotoxicity, macrophage numbers, macrophage subpopulations, and performance measures were all significantly affected by social rank, heat stress, and/or infection status of the pig. Heat stress and PRRS status also significantly influenced the amount of time pigs spent lying with or without contacting another animal. Cortisol and various immune measures were also affected by PRRS status. These results show not only that intranasal inoculation with PRRS virus affects physiological, behavioral, and performance measures in growing pigs, but that social rank influences pigs' immune responsiveness to PRRS as well. Moreover, heat stress does not have additive negative impact on physiological or performance traits in pigs challenged with PRRS virus.
