Objective To detect the expression profiling of microRNA (miRNA) and the expression level of miRNA-429 in human gastric carcinoma.Methods The miRNAs expression of 7 normal gastric tissues,52 malignant tissues,gastric cancer cell line SGC7901 and gastric epithelial cell line 1 ( GES-1 ) was detected by using miRNA gene chips.Real-time reverse transcriptase-polymerase chain reaction (qRT-PCR) was used to quantify the expression level of miR-429 in 52 gastric cancer tissues and their paracancerous tissues.Results The results of gene chip showed that 23 miRNA were significantly up-regulated,such as miR-21,miR-30b,etc.,and 31 miRNA down-regulated,such as miR-429,miR-200,etc.The average expression of miR-429 was significantly reduced by 63.4% in gastric cancer samples as compared with that in normal paracancerous tissues (P <0.01 ).miR-429 expression was reduced by 76.2%in SGC-7901 cells as compared with non-tumor gastric cells (P <0.01 ).Conclusion miR-429 might play a role in the pathogenesis of gastric carcinoma.
