Identification of key genes of papillary thyroid carcinoma by integrated bioinformatics analysis.

Background: Papillary thyroid carcinoma (PTC) is one of the fastest-growing malignant tumor types of thyroid cancer. Therefore, identifying the interaction of genes in PTC is crucial for elucidating its pathogenesis and finding more specific molecular biomarkers.Methods: Four pairs of PTC tissues and adjacent tissues were sequenced using RNA-Seq, and 3745 differentially expressed genes were screened (P<0.05, |logFC|>1). The enrichment analysis indicated that the vast majority of differentially expressed genes (DEGs) may play a positive role in the development of cancer. Then, the significant modules were analyzed using Cytoscape software in the protein–protein interaction network. Survival analysis, TNM analysis, and immune infiltration analysis of key genes were analyzed. And the expression of ADORA1, APOE, and LPAR5 genes were verified by qPCR in PTC compared with matching adjacent tissues.Results: Twenty-five genes were identified as hub genes with nodes greater than 10. The expression of 25 genes were verified by the GEPIA database, and the overall survival and disease-free survival analyses were conducted with Kaplan–Meier plotter. We found only three genes were confirmed with our validation and were statistically significant in PTC, namely ADORA1, APOE, and LPAR5. Further analysis found that the mRNA levels and methylation degree of these three genes were significantly correlated with the TNM staging of PTC. And these three genes were related to PTC immune infiltration. Verification of the expression of these three genes by RT-qPCR and Western blot further confirmed the reliability of our results.Conclusion: Our study identified three genes that may play key regulatory roles in the development, metastasis, and immune infiltration of papillary thyroid carcinoma.

Silencing of Long Non-coding RNA ENST00000606790.1 Inhibits the Malignant Behaviors of Papillary Thyroid Carcinoma through the PI3K/AKT Pathway

ABSTRACT Purpose This study aimed to investigate the role and mechanism of lncRNA ENST00000606790.1 (ENST) in promoting the progression of papillary thyroid carcinoma (PTC). Methods The expression of ENST in human PTC and normal para-cancerous thyroid (NPTC) tissues or cell lines was determined by RT-qPCR. Cell growth was determined by CCK8 assay. Cell colony formation was determined by cell colony formation assay. Cell cycle analysis was performed by staining cells with PI (Propidium Iodide). Cell invasion was assessed by transwell assay. Protein expression was examined by western-blot. siRNA was constructed to inhibit the expression of ENST. 740-Y-P was used to activate PI3K. The correlation between ENST expression and clinical outcomes was analyzed. Results ENST was significantly up-regulated in PTC tissues or PTC cell lines (PTC and IHH4 cell lines), compared to NPTC tissues or normal cell lines, respectively. High expression of ENST was strongly correlated to lymph node metastasis and tumor size at diagnosis. Silencing of ENST significantly inhibited cell growth and colony formation, arrested the cell cycle at G2/M phase, upregulated the expression of CHK1, downregulated the expression of CDC25C, and inhibited cell invasion. Silencing of ENST significantly down-regulated the expression of PI3K, p-PI3K, AKT, and p-AKT in IHH4 cells. Furthermore, treatment with the PI3K activator 740-Y-P partially abolished the effect of silencing of ENST on PTC cells. Conclusions Overall, our results demonstrated that ENST can promote PTC progression by activating the PI3K/AKT signaling pathway, suggesting that ENST can serve as a potential biomarker and new therapeutic target for patients with PTC.

PD-L1, PDK-1 and p-Akt are correlated in patients with papillary thyroid carcinoma.

Papillary thyroid carcinoma (PTC) is the most common type of thyroid carcinoma. To investigate the clinical significance of programmed death ligand 1 (PD-L1) and phosphoinositide-dependent protein kinase 1 (PDK1) in PTC. A total of 194 PTC patients were recruited. Contralateral normal thyroid tissues were obtained and used as controls (n = 80). The expression levels of PD-L1, PDK1 and p-Akt were determined using immunohistochemistry. The PD-L1, PDK1 and p-Akt were upregulated in cancer tissues compared to the normal tissues. The mean optical density (MOD) values of PD-L1, PDK1 and p-Akt were significantly higher in the PTC tissues. The expression of PD-L1 positively correlated with the levels of PDK1 and p-Akt. In addition, the expression of PD-L1, PDK1 and p-Akt in PTC patients without chronic lymphocytic thyroiditis (CLT) was significantly higher than the expression of those proteins in the CLT patients. The patients with higher expression levels of PD-L1, PDK1 or p-Akt had remarkably larger tumors and higher rates of TNM III-IV, capsular infiltration, lymph node metastasis, and of recurrence. The Kaplan-Meier curve showed that patients with lower expression of PD-L1, PDK1 or p-Akt had significantly longer recurrence-free time. The logistic regression analysis revealed that only CLT, PD-L and capsular infiltration were risk factors for patients' five-year recurrence. The PD-L1, PDK1 and p-Akt were found to be positively correlated with a poor prognosis in PTC.

FOXN3 suppresses the growth and invasion of papillary thyroid cancer through the inactivation of Wnt/β-catenin pathway

Forkhead box N3 (FOXN3) is a subtype of FOX family that has been demonstrated to be implicated in several cancers. However, the role of FOXN3 in papillary thyroid carcinoma (PTC) and its mechanisms have not yet been investigated. Our results showed that FOXN3 was markedly down regulated in PTC tissues and cell lines. Overexpression of FOXN3 suppressed the proliferation, colony formation, migration, and invasion in PTC cells. Overexpression of FOXN3 also prevented EMT process in PTC cells, as shown by the increased E-cadherin expression level and decreased expression levels of N-cadherin and vimentin. In addition, overexpression of FOXN3 inhibited tumor growth of PTC in vivo. Furthermore, overexpression of FOXN3 caused significant decreases in expression levels of β-catenin, c-Myc, and cyclin D1. Additionally, activation of Wnt/β-catenin pathway reversed the effects of FOXN3 on PTC cells. In conclusion, these findings indicated that FOXN3 exerted a tumor suppressive activity in PTC, which was mediated by Wnt/β-catenin pathway.

Potential five-mRNA signature model for the prediction of prognosis in patients with papillary thyroid carcinoma.

Although the mortality rate of papillary thyroid carcinoma (PTC) is relatively low, the recurrence rates of PTC remain high. The high recurrence rates are related to the difficulties in treatment. Gene expression profiles has provided novel insights into potential therapeutic targets and molecular biomarkers of PTC. The aim of the present study was to identify mRNA signatures which may categorize PTCs into high-and low-risk subgroups and aid with the predictions for prognoses. The mRNA expression profiles of PTC and normal thyroid tissue samples were obtained from The Cancer Genome Atlas (TCGA) database. Differentially expressed mRNAs were identified using the ‘EdgeR’ software package. Gene signatures associated with the overall survival of PTC were selected, and enrichment analysis was performed to explore the biological pathways and functions of the prognostic mRNAs using the Database for Visualization, Annotation and Integration Discovery. A signature model was established to investigate a specific and robust risk stratification for PTC. A total of 1,085 differentially expressed mRNAs were identified between the PTC and normal thyroid tissue samples. Among them, 361 mRNAs were associated with overall survival (P<0.05). A 5-mRNA prognostic signature for PTC (ADRA1B, RIPPLY3, PCOLCE, TEKT1 and SALL3) was identified to classify the patients into high-and low-risk subgroups. These prognostic mRNAs were enriched in Gene Ontology terms such as ‘calcium ion binding’, ‘enzyme inhibitor activity’, ‘carbohydrate binding’, ‘transcriptional activator activity’, ‘RNA polymerase II core promoter proximal region sequence-specific binding’ and ‘glutathione transferase activity’, and Kyoto Encyclopedia of Genes and Genomes signaling pathways such as ‘pertussis’, ‘ascorbate and aldarate metabolism’, ‘systemic lupus erythematosus’, ‘drug metabolism-cytochrome P450 and ‘complement and coagulation cascades’. The 5-mRNA signature model may be useful during consultations with patients with PTC to improve the prediction of their prognosis. In addition, the prognostic signature identified in the present study may reveal novel therapeutic targets for patients with PTC.

Differential protein expressions in papillary thyroid carcinoma patients with or without Hashimoto's thyroiditis

Objective: To explore the differential protein expressions in papillary thyroid carcinoma (PTC) with or without Hashimoto's thyroiditis (HT). Methods: Tissue microarray was prepared and the protein expression levels of v-Raf murine sarcoma viral oncogene homolog B1 (BRAF), vascular endothelial growth factor (VEGF), cyclinD1, mesothelial cell (MC) , CD56 and Galectin3 in the PTC tissues with or without HT were detected by immunohistochemical staining. Results: The positive expression rates of BRAF protein in the PTC tissues with or without HT groups were 55.4% (36/65) and 63.6% (42/66), respectively, without significant difference (P=0.336). The positive expression rates of VEGF protein in the PTC tissues with or without HT groups were 25.7% (19/74) and 25.8%(17/66), respectively, without significant difference (P=0.991). The positive expression rates of cyclin D1 protein in the PTC tissues with or without HT groups were 93.4% (71/76) and 97.6% (80/82), without significant difference (P=0.206). The positive expression rates of MC protein in the PTC tissues with or without HT groups were 86.1% (62/72) and 83.5%(71/85), without significant difference (P=0.654). The positive expression rates of Galectin3 protein in the PTC tissues with or without HT groups were 98.7% (76/77) and 97.5% (78/80), without significant difference (P=0.583). The positive expression rates of CD56 in the PTC tissues and adjacent thyroid follicular epithelial cells were 27.4% (32/117) and 65.0% (76/117), respectively, and the difference was statistically significant (P=0.001). The positive expression rates of CD56 in PTC tissues with or without HT were 35.5% (24/68) and 16.5% (13/79), respectively, and the difference was statistically significant (P=0.009). Conclusions: There are no significant differences in the expressions of BRAF, VEGF, CyclinD1, MC and Galectin3 between the PTC tissues with or without HT. However, the significantly differential expression of CD56 between the two group suggests that CD56 may be related to the pathogenesis of PTC with HT. CD56 may be used as a potential molecular marker in PTC diagnosis.

Circ_LDLR promoted the development of papillary thyroid carcinoma via regulating miR-195-5p/LIPH axis

BackgroundEmerging studies have demonstrated that circular RNAs (circRNAs) are key regulators for tumorigenesis in cancers, including papillary thyroid carcinoma (PTC). In this study, we aimed to explore the effects of circ_LDLR on PTC.MethodsQuantitative real-time polymerase chain reaction (qRT-PCR) was performed to determine the levels of circ_LDLR, miR-195-5p and lipase H (LIPH). RNase R digestion assay and Actinomycin D assay were utilized to analyze the characteristics of circ_LDLR. Colony formation assay and 3-(4,5-dimethyl-2-thiazolyl)-2, 5-diphenyl-2-H-tetrazolium bromide (MTT) assay were conducted to evaluate cell proliferation. Western blot assay was used for the determination of protein levels. Flow cytometry analysis was applied to determine cell apoptosis. Transwell assay was performed to determine cell migration and invasion. Dual-luciferase reporter assay was used to verify the associations among circ_LDLR, miR-195-5p and LIPH. The murine xenograft model was constructed to explore the roles of circ_LDLR in vivo.ResultsCompared to normal tissues and cells, circ_LDLR was upregulated in PTC tissues and cells. Silencing of circ_LDLR suppressed PTC cell colony formation, proliferation, migration and invasion and promoted apoptosis in vitro and hampered tumor growth in vivo. For mechanism investigation, circ_LDLR could regulate LIPH expression via sponging miR-195-5p. Moreover, miR-195-5p inhibition restored the effects of circ_LDLR knockdown on the malignant behaviors of PTC cells. MiR-195-5p overexpression inhibited PTC cell colony formation, proliferation, migration and invasion and facilitated apoptosis by targeting LIPH.ConclusionCirc_LDLR knockdown decelerated PTC progression by regulating miR-195-5p/LIPH axis, which might provide a novel therapeutic target for PTC.

Correlation analysis between the pre-operative contrast-enhanced ultrasound parameters and biological characteristics of papillary thyroid carcinoma and associated risk factors for prognosis after radiofrequency ablation.

The present study aimed to assess the correlation between the contrast-enhanced ultrasound (CEUS) parameters and biological characteristics of papillary thyroid carcinoma (PTC), and identify risk factors of recurrence for the prognosis after radiofrequency ablation. A total of 72 patients with PTC were included. Disease recurrence and metastasis were evaluated based on the imaging performance and laboratory tests. Perfusion parameters were recorded and compared. After biopsy, the tumor tissues were subjected to hematoxylin and eosin staining, and the expression levels of P53 and Ki67 were detected using immunohistochemistry. The association between the CEUS parameters and the expression of P53 and Ki67 was analyzed. Risk factors for disease recurrence were analyzed using logistic regression. Compared with normal tissues, PTC tissue was characterized by a slow perfusion pattern with no obvious enhancement. The peak time and intensity were lower, while the initial increasing time and mean transit time (MTT) were longer in the PTC tissue. The expression levels of P53 and Ki67 in PTC tissues were associated with age, local infiltration, lymph node metastasis and the number of lesions, but not with gender, tumor size, histological type or degree of differentiation. The initial increasing time, peak time and MTT were negatively correlated, while the peak intensity was positively correlated with the expression of P53 and Ki67 in PTC tissues. Multivariate logistic regression analysis suggested that lymph node metastasis, age, local infiltration, peak intensity and MTT were risk factors for disease recurrence. In conclusion, quantitative CEUS parameters are closely linked to the biological characteristics of PTC and may serve as indicators for the prognosis of patients.

Diagnostic significance of DNA methylation of PTEN and DAPK in thyroid tumors.

DNA Methylation of the tumour suppressor gene leading to gene silencing plays an important role in thyroid tumour development. The purpose was to determine the DNA methylation status of phosphatase and tensin homolog (PTEN) and death-associated protein kinase (DAPK) in patients with thyroid nodules and to explore whether they can be used as molecular diagnostic tools to differentiate benign and malignant thyroid nodules. Thyroid tissue and blood samples were obtained from normal healthy individuals (controls) and patients suffering from clinically diagnosed thyroid nodular disease [papillary thyroid carcinoma (PTC), adenoma and nodular goitre]. DNA methylation level, mRNA expression and protein expression of PTEN and DAPK in the thyroid tissues and peripheral blood were detected using methylation-specific PCR, semi-quantitative reverse transcription PCR and Western blot, respectively. Diagnostic sensitivity, specific and accuracy of detection were evaluated between blood and thyroid tissue. There was a significant increase in the level of DNA methylation of PTEN and DAPK in PTC (P<.05) compared with controls and other types of thyroid nodules. Levels of the mRNA of both PTEN and DAPK were lower in PTC in both peripheral blood and tissue samples compared with controls, while there was concomitant decrease of both PTEN and DAPK protein expression in PTC tissues (P<.05). There was no significant difference in diagnostic specificity, sensitivity and accuracy between blood sample and thyroid tissues. Hypermethylated status of both PTEN and DAPK in peripheral blood and tissue samples can be useful biomarkers for clinical diagnosis and, distinguishing of benign and malignant thyroid nodules.

MiR-7-5pand miR-451 as diagnostic biomarkers for papillary thyroid carcinoma in formalin-fixed paraffin-embedded tissues.

MiR-7-5p and miR-451 are important members of the small RNA family, which have been shown to be significantly downregulated in various human tumors and play a key role in the occurrence and development of tumors. However, little is known about their role in endocrine malignancies. This study aimed to investigate the diagnostic value of miR-7-5p and miR-451 levels in formalin-fixed paraffin-embedded tissues of papillary thyroid carcinoma (PTC) patients, as well as the relationship between clinicopathological characteristics and the two miRNAs. Quantitative real-time PCR (qRT-PCR) was performed to detect the expression levels of miR-7-5p and miR-451 in 101 PTC tissues and in 40 nodular goiter tissues (controls). MiR-7-5p and miR-451 levels were significantly downregulated in PTC patients compared with controls (P < 0.001). MiR-7-5p expression was further downregulated in tumors with larger diameter and advanced tumor stages (all P < 0.05). Moreover, the two miRNAs showed great capability of discriminating PTC patients from controls with 89.5% (miR-7-5p) and 76.8% (miR-451) diagnostic accuracy. Furthermore, according to univariable/multivariate logistic regression, miR-7-5p was significantly associated with PTC (P < 0.001). In conclusion, MiR-7-5p and miR-451 may be used as potential diagnostic biomarkers for identification and validation of PTC patients. Moreover, miR-7-5p appears to be associated with the aggressiveness of PTC.

MiR-224-5p targets EGR2 to promote the development of papillary thyroid carcinoma.

Various microRNAs (miRNAs) have been reported to be involved in the pathogenesis and development of human cancers, including papillary thyroid carcinoma (PTC). However, the role of miR-224-5p in PTC progression remains unclear. Therefore, the purpose of this study is to illuminate the function of miR-224-5p in PTC. Expression of miR-224-5p and EGR2 was examined in PTC by quantitative Real Time-Polymerase Chain Reaction (qRT-PCR). Transwell assay was used to detect cell migration and invasion. Western blot analysis was used to detect epithelial-mesenchymal transition (EMT). The relationship between miR-224-5p and EGR2 was confirmed by Dual-Luciferase assay. Upregulation of miR-224-5p and downregulation of EGR2 expression were detected in PTC tissues and cells. Upregulation of miR-224-5p was found to be associated with TNM stage and lymph node metastasis. Meanwhile, it also predicted poor prognosis in PTC patients. Functionally, upregulation of miR-224-5p promoted cell metastasis and EMT in PTC. In addition, miR-224-5p was detected to directly target EGR2. EGR2 expression was negatively correlated with EGR2 expression in PTC. Of note, overexpression of EGR2 attenuated the carcinogenic effects of miR-224-5p in PTC. MiR-224-5p promotes cell migration, invasion, and EMT in PTC by targeting EGR2.

Aberrant Expression of Androgen Receptor Associated with High Cancer Risk and Extrathyroidal Extension in Papillary Thyroid Carcinoma

Male gender is a risk factor for mortality in patients with papillary thyroid carcinoma (PTC). This study investigated the impact of androgen receptor (AR) gene expression on the clinical features and progression of PTC. The levels of AR mRNA and protein in frozen, formalin-fixed, paraffin-embedded tissue samples from PTC and adjacent normal thyroid tissue were assessed by quantitative real-time polymerase chain reaction and immunohistochemical staining, respectively, and the relationships between AR expression and clinical features were analyzed. The thyroid cancer cell lines, BCPAP and TPC-1, were used to evaluate the effects of AR on the regulation of cell migration, and key epithelial–mesenchymal transition (EMT) markers. AR mRNA expression was significantly higher in normal thyroid tissue from men than women. The sex difference in AR mRNA expression diminished during PTC tumorigenesis, as AR mRNA expression levels were lower in PTC than normal thyroid tissues from both men and women. AR mRNA expression was significantly decreased in PTC patients with higher risk and in those with extrathyroidal extension. Overexpression of AR in BCPAP cells decreased cell migration and repressed the EMT process by down-regulating mRNA expression of N-cadherin, Snail1, Snail2, Vimentin, and TWIST1 and up-regulating E-cadherin gene expression. These results suggest that suppression of the androgen–AR axis may lead to aggressive tumor behavior in patients with PTC.

MiR-744–5p mediates lncRNA HOTTIP to regulate the proliferation and apoptosis of papillary thyroid carcinoma cells

Papillary thyroid carcinoma (PTC) is the most common malignancy in thyroid. miR-744–5p plays an efficient role in various cancers, but its role in PTC remains unknown. In this work, we aimed to explore the function of miR-744–5p and the mechanism by which miR-744–5p acted in PTC. We observed that miR-744–5p expression was significantly declined in PTC tissues and cell lines. The high level of miR-744–5p is significantly associated with a better clinical picture of PTC patients. Overexpression of miR-744–5p inhibited the proliferation, arrested the cell cycle, and promoted the apoptosis in PTC cells. Oppositely, down-regulation of miR-744–5p reversed the above tendencies. We also found that miR-744–5p down-regulated its downstream genes c-myc and attenuated cell proliferation induced by c-myc. Long non-coding RNA (lncRNA) HOTTIP was found to be up-regulated and to act as an oncogene in PTC. In this study, miR-744–5p bound to HOTTIP and was negatively regulated by HOTTIP. In conclusion, miR-744-5p acts as a tumor suppressor to inhibit proliferation and promotes the apoptosis of PTC cells via targeting c-myc. Moreover, miR-744-5p expression interferes with lncRNA HOTTIP ability to promote proliferation and downregulate apoptosis in papillary thyroid carcinoma.

MiR-451a restrains the growth and metastatic phenotypes of papillary thyroid carcinoma cells via inhibiting ZEB1

MicroRNAs (miRNAs) are known to be critical regulators in cancer progression. MiR-451a is reported to be involved in the progression of many different forms of cancers, including osteosarcoma, colorectal cancer, and breast carcinoma. In this study, we illuminated the possible roles of miR-451a in the development of papillary thyroid carcinoma (PTC) cells in vitro and in vivo. MiR-451a was markedly down-expressed in PTC sample compared with paratumor tissue. Upregulation of miR-451a repressed PTC cells proliferation, migration ability and inhibited the invasiveness of PTC cells in vitro. Additional, miR-451a suppressed PTC cells growth and the lung metastasis of PTC cells in vivo, whereas downregulation of miR-451a caused opposite outcomes. Importantly, miR-451a inversely modulated the expression of Zinc Finger E-Box Binding Homeobox 1 (ZEB1) by directly binding to the 3′ untranslated region (UTR) of ZEB1 in PTC cells. The level of ZEB1 was negatively associated with miR-451a level in PTC tissues, and ZEB1 silencing mimicked the suppressive impacts of miR-451a on the proliferation, mobility, and invasive phenotypes of PTC cells. ZEB1 overexpression abrogated the inhibitory impacts of miR-451a on PTC cells. Together, this study revealed that miR-451a restrained the growth and metastatic phenotypes of PTC cells through targeting ZEB1.

Lysine-Specific Demethylase 1 Affects the Progression of Papillary Thyroid Carcinoma via HIF1α and microRNA-146a.

Lysine-specific demethylase 1 (LSD1) stabilizes hypoxia-inducible factor 1α (HIF1α) to advance tumor progression, while HIF1α functions as a transcription factor to increase the expression of microRNA-146a (miR-146a). We aim to investigate whether LSD1 affects the development of papillary thyroid carcinoma (PTC) via HIF1α and miR-146a. In vitro assays were performed with Nthy-ori 3-1, BHP5-16, BCPAP, K1, and BHP2-7 cell lines. In vivo assays were conducted with established xenograft tumors in nude mice. This study was conducted at our lab. PTC tissues and corresponding adjacent normal tissues were obtained from 45 patients hospitalized in Sun Yat-Sen Memorial Hospital. Assays were conducted using Nthy-ori 3-1, BHP5-16, BCPAP, K1, and BHP2-7 cell lines, as well as 50 male BALB/c nude mice. Cells were transfected with sh-LSD1, sh-GABPA, oe-LSD1, oe-HIF1α, miR-146a mimic, and miR-146a inhibitor. In addition, K1 cells expressing lv-oe-LSD1, lv-miR-146a inhibitor, lv-oe-LSD1 or miR-146a inhibitor were injected into the right side of the mice. LSD1 gene and protein expression patterns were analyzed in 45 clinical PTC tissue samples. Expression of LSD1, HIF1α, miR-146a, and GA-binding protein transcription factor alpha (GABPA), as well as their effects on PTC. LSD1 was highly expressed in clinical PTC tissues. LSD1 stabilized HIF1α and inhibited the degradation of its ubiquitin proteasome. HIF1α was enriched in the promoter region of miR-146a, an upregulated miRNA in PTC. HIF1α increased miR-146a expression to promote PTC progression in vitro, which was achieved by inhibiting GABPA, a target gene of miR-146a. LSD1 upregulated miR-146a to enhance the development and metastasis of PTC in nude mice. Our results show that LSD1 functions as an oncogene in PTC by upregulating HIF1α and miR-146a, elucidating an understanding of undefined mechanisms associated with tumor progression in PTC.

Association of MicroRNA Expression and BRAFV600E Mutation with Recurrence of Thyroid Cancer.

Many miRNAs and cancer-related mutations have been proposed as promising molecular markers of papillary thyroid carcinoma (PTC). However, there are limited data on the correlation between miRNA expression, BRAFV600E mutation, and PTC recurrence. Therefore, to evaluate the potential of BRAFV600E mutation and five selected miRNAs (-146b, -222, -21, -221, -181b) in predicting PTC recurrence, these molecular markers were analyzed in 400 formalin-fixed, paraffin-embedded PTC tissue specimens. The expression levels of miRNAs were measured using qRT-PCR. It was demonstrated that expression levels of all analyzed miRNAs are significantly higher in recurrent PTC than in non-recurrent PTC (p < 0.05). Moreover, higher expression levels of miR-146b, miR-222, miR-21, and miR-221 were associated with other clinicopathologic features of PTC, such as tumor size and lymph node metastases at initial surgery (p < 0.05). No significant differences in the frequency of BRAFV600E mutation in recurrent PTC and non-recurrent PTC were determined. Our results suggest that miRNA expression profile differs in PTC that is prone to recurrence when compared to PTC that does not reoccur after the initial surgery while BRAFV600E mutation frequency does not reflect the PTC recurrence status. However, the prognostic value of the analyzed miRNAs is rather limited in individual cases as the pattern of miRNA expression is highly overlapping between recurrent and non-recurrent PTC.

Knockout of ASAP1 induces autophagy in papillary thyroid carcinoma by inhibiting the mTOR signaling pathway

Due to lymph node metastasis and infiltration, surgery for PTC (papillary thyroid carcinoma) is a high-risk treatment strategy. Our work reports for the first time that ASAP1 (ArfGAP with SH3 Domain, Ankyrin Repeat and PH Domain 1) is highly expressed in PTC and that its high expression is related to autophagy. Autophagy and ASAP1 expression in 40 PTC tissues and normal tissues were detected by immunofluorescence. We used the lentiviral CRISPR/Cas9 nickase to generate stable cell lines. The difference in autophagy levels between the ASAP1 KO group and the control group was determined by Western blot and immunofluorescence analyses. We added chloroquine (CQ) to verify that ASAP1 increased the formation of autophagosomes rather than reducing their degradation. The expression of mTOR activity-related proteins (P-P70S6K, P-MTOR) was studied by Western blotting. ASAP1 was upregulated while autophagy was downregulated in PTC tissues compared to normal tissues. Knockout of ASAP1 induced autophagy in both MDA-T32 and MDA-T85 cells. Knockout of ASAP1 attenuated the activation of the mTOR signaling pathway. Our studies demonstrated that ASAP1 is upregulated while autophagy is reduced in PTC tissues. In addition, knockout of ASAP1 induces autophagy in PTC by inhibiting the mTOR signaling pathway.

Induction of mTOR signaling pathway in the progression of papillary thyroid cancer

Papillary thyroid carcinoma (PTC) is the most frequent (more than 85%) malignancy among the four pathological types of thyroid cancer. Since enhancements of survival and recurrence rates of the disease as well as early diagnosis are essential, the genome-wide investigation for identifying robust biomarkers for PTC patients is necessary.In the current research, two independent gene expression omnibus (GEO) datasets resulted from systematically searching the NCBI-GEO database and applying the inclusion and exclusion criteria for PTC patients. Then, the identification of significant biomarkers through a meta-analysis approach used the associated genes of the mTOR signaling pathway. The survival analysis of the determined genes included overall survival (OS) and recurrence-free survival (RFS) rates based on The Cancer Genome Atlas dataset. Furthermore, the analysis and virtual screening process were carried out for finding potential drugs and drug-like ligands.The analysis outcomes reveal three statistically significant downregulated biomarkers IRS1, DEPTOR, and PRKCA between PTC and healthy tissues with experimental literature confirmation. Moreover, the survival analysis shows that DEPTOR and PRKCA have vital roles in RFS (p-value = .012) and OS (p-value = .045) rates, respectively. However, IRS1 does not present significant p-value in terms of RFS and OS rates. The GDIdbi webserver found twenty, three, and four potential drugs for PRKCA, DEPTOR, and IRS1. And two molecules had structural interactions with IRS1 with four hydrogen bond acceptors through the virtual screening procedure.Consequently, the critical role of genome-wide association studies (GWAS) should not be of ignorance in diagnosing PTC patients. Furthermore, the outcomes of such GWAS investigations need to be carefully involved in the future development of new therapeutic agents.

LncRNA FALEC promotes proliferation, migration, and invasion of PTC cells through regulating Wnt/β-catenin signaling pathway.

The aim of this study was to explore the expression of long non-coding ribonucleic acid (lncRNA) FALEC (hereinafter referred to as FALEC) in papillary thyroid carcinoma (PTC) and its effects on the proliferation, invasion, and metastasis of PTC cells. Quantitative Reverse Transcription-Polymerase Chain Reaction (qRT-PCR) was performed to measure the expression level of FALEC in 48 cases of PTC tissues and cells. The small interfering (si)-FALEC was synthesized and transfected into PTC cells. Interference efficiency was confirmed via qRT-PCR assay. Subsequently, the effect of FALEC on the proliferation of PTC cells was determined by cell counting kit-8 (CCK-8) assay. Wound healing and transwell assays were conducted to detect the effects of FALEC on the invasion, migration, and metastasis of PTC cells. Additionally, changes in the protein expression of Wnt/β-catenin signaling pathway molecular markers was detected via Western blotting. The expression level of FALEC was significantly higher in PTC tissues than that of adjacent normal tissues. FALEC expression was significantly up-regulated in PTC cell lines, as well. CCK-8 assay revealed that the proliferation ability of PTC cells was remarkably weakened after down-regulation of FALEC in vitro. Wound healing and transwell assays demonstrated that, compared with si-normal control (NC) group, the migration and invasion capabilities declined significantly in si-FALEC group. Furthermore, the Western blotting analysis indicated that the expression of Wnt/β-catenin signaling pathway molecular markers was changed after the interference in FALEC expression. FALEC expression was up-regulated in PTC tissues and cell lines. Highly expressed FALEC facilitated the proliferation, migration, and invasion of PTC by regulating the Wnt/β-catenin signaling pathway.

In-depth characterization of miRNome in papillary thyroid cancer with BRAF V600E mutation

MicroRNAs (miRNAs) are small non-coding RNAs, which play a critical regulatory role in papillary thyroid carcinoma (PTC). BRAF V600E is a hotspot mutation occurring in a majority of PTC cases and is proposed to be associatedwith poor clinical outcomes. The relationship between BRAF V600E status and miRNA expression in PTC has not been comprehensively studied. In this study, we aimed to identify the differentially expressed miRNAs in PTCs with and without BRAF V600E in an unbiased manner. Five fresh frozen thyroid cancer tissues paired with their respective adjacent normal tissues from PTC patients were subjected to BRAF V600E genotyping using Sanger sequencing and small RNA deep sequencing (miRNAseq). MiRNAs differentially expressed between BRAF V600E-positive and BRAF V600E-negative PTC tissues were validated in silico using The Cancer Genome Atlas (TCGA) THCA datasets containing 420 samples. MiRNA target prediction and pathway enrichment analysis were performed to identify biological pathways altered in this cancer. We identified 174 differentially expressed miRNAs; 80 were significantly over-expressed, while 94 were underexpressed (adj. p-value &lt; 0.1; log2 fold change ? -1 or ? 1). Fifteen miRNAs were significantly differentially expressed only in BRAF V600E-positive PTC, and eight of these were validated in TCGA THCA dataset (hsa-miR-212, -132, -135b-3p/5p, -200b, -200a-3p/5p, -27a-3p/5p, -29a and -1296). Subsequent analysis revealed significant enrichment of cancer-relatedpathways including proteoglycans in cancer, ECM-receptor interaction and MAPK pathways in BRAF V600E-positive PTC. Using the miRNAseq and in silico validation using TCGA THCA study, we identified eight miRNAs that were differentially expressed in PTC tissues with BRAF V600E. This study also complemented the existing knowledge about deregulated miRNAs in PTC development.
