Osmotically Induced Proline Accumulation in Lotus Corniculatus Leaves is Affected by Light and Nitrogen Source

Proline accumulation in osmotically stressed leaves of Lotus corniculatus was stimulated by increasing light intensity (photon fluence density, PFD). Treatment with propanil limited proline accumulation in response to light and osmotic stress, indicating a dependence of proline synthesis on photosynthetic NADPH. Drought stress induced proline accumulation in L. corniculatus both in nitrate-fed plant (NFP) and ammonium-fed plants (AFP), although higher proline concentration was observed in AFP than in NFP after 24 h of drought stress. Changes in proline accumulation induced by drought stress in plants grown under different nitrogen regimes could not be explained by changes of either total protein or amino acids, consistent with specifically altered regulation of proline synthesis. Under control conditions, alanine, aspartate and glutamate were the predominant amino acids in NFP; conversely, in AFP, arginine and ornithine were the predominant amino acids. Only the NFP regime showed changes in the concentrations of specific amino acids under drought stress a decrease in alanine, aspartate and glutamate and increased gama-aminobutyric acid. In AFP and especially NFP, proline accumulation under osmotic stress was associated with increased ornithine amino transferase activity. An increase of both activity and protein of ferredoxin-dependent glutamate synthase was observed in osmotic-stressed NFP; inversely both decreased in drought-stressed AFP. PFD and nitrogen source are therefore shown to be regulators of proline accumulation in L. corniculatus osmotically stressed plants.

Changes of glucose metabolism and skin-collagen neogenesis in vitamin B6deficiency

The mechanism of pellagrous changes in skin caused by a deficiency of vitamin B6 was studied in respect to neogenesis of proline in skin collagen and glucose metabolism. In vitamin B6 deficiency the insulin/glucagon coefficient in serum decreased significantly from 3.02 to 2.32, indicating a metabolic change towards gluconeogenesis. A deficiency of vitamin B6 caused a decrease in the levels of vitamin B6-dependent enzymes, such as ornithine aminotransferase, alanine aminotransferase, and aspartate aminotransferase, which also contribute to gluconeogenesis. Because the conversion of ornithine to proline via pyrroline-5-carboxylate was suppressed due to the decrease in ornithine aminotransferase activity, the amount of proline in the skin collagen fraction also decreased significantly in vitamin B6-deficient rats compared with the pair-fed control. These results suggest that the pellagrous lesions in vitamin B6-deficiency are caused by an impaired synthesis of proline from ornithine, which results in the suppression of collagen neogenesis in the skin.

Vitamin B6-Responsive Ornithine Aminotransferase Deficiency with a Novel Mutation G237D

Ornithine aminotransferase (OAT) deficiency (MIM: 258870) is a rare congenital metabolic disorder characterized by gyrate atrophy of the choroid and retina. Here, we report a 37-year-old male with gyrate atrophy of the choroid and retina who has been treated for 18 years. At the age of 7 years, the patient consulted an ophthalmologist due to progressive loss of vision. A large atrophied area was observed in his retina, and OAT deficiency was suspected. At the age of 19 years, amino acid analysis revealed high serum ornithine levels (1,140 nmol/ml), with the normal range being 40-100 nmol/ml. He was treated with vitamin B(6) 300 mg/day for 6 months, which successfully reduced his serum ornithine levels by 20-30%. For 18 years since, his serum ornithine levels have been maintained with vitamin B(6) medication. There was no further impairment of vision or increase in the atrophied area, as judged by ophthalmoscopic examination. OAT activity was undetected in white blood cells of the patient and was 105% and 45% of normal values in his wife and son, respectively. OAT gene analysis revealed a novel mutation of Gly237Asp in exon 7 (710G > A) in both alleles of the patient, while his son was a heterozygote for the mutation. Notably, this novel mutation is associated with a vitamin B6-responsive phenotype. Therefore, early diagnosis and treatment with vitamin B(6) may prevent loss of vision in some patients with OAT deficiency.

Polyamine Synthesis from Proline in the Developing Porcine Placenta1

Polyamines (putrescine, spermidine, and spermine) are essential for placental growth and angiogenesis. However, little is known about polyamine synthesis in the porcine placenta during conceptus development. The present study was conducted to test the hypothesis that arginine and proline are the major sources of ornithine for placental polyamine production in pigs. Placentae, amniotic fluid, and allantoic fluid were obtained from gilts on Days 20, 30, 35, 40, 45, 50, 60, 90, and 110 of the 114-day gestation (n = 6 per day). Placentae as well as amniotic and allantoic fluids were analyzed for arginase, proline oxidase, ornithine aminotransferase (OAT), ornithine decarboxylase (ODC), proline transport, concentrations of amino acids and polyamines, and polyamine synthesis using established radiochemical and chromatographic methods. Neither arginase activity nor conversion of arginine into polyamines was detected in the porcine placenta. In contrast, both proline and ornithine were converted into putrescine, spermidine, and spermine in placental tissue throughout pregnancy. The activities of proline oxidase, OAT, and ODC as well as proline transport, polyamine synthesis from proline, and polyamine concentrations increased markedly between Days 20 and 40 of gestation, declined between Days 40 and 90 of gestation, and remained at the reduced level through Day 110 of gestation. Proline oxidase and OAT, but not arginase, were present in allantoic and amniotic fluids for the production of ornithine (the immediate substrate for polyamine synthesis). The activities of these two enzymes as well as the concentrations of ornithine and total polyamines in fetal fluids were highest at Day 40 but lowest at Days 20, 90, and 110 of gestation. These results indicate that proline is the major amino acid for polyamine synthesis in the porcine placenta and that the activity of this synthetic pathway is maximal during early pregnancy, when placental growth is most rapid. Our novel findings provide a new base of information for future studies to define the role of proline in fetoplacental growth and development.

Effect of drought on polyamine metabolism, yield, protein content and in vitro protein digestibility in tepary (Phaseolus acutifolius) and common (Phaseolus vulgaris) bean seeds

AbstractThe objective of the present study was to investigate the effect of drought on the metabolism of proline, ornithine and polyamines, and the enzymes involved in their synthesis and degradation, in tepary and common bean seeds. Furthermore, the effect on seed yield, protein content and in vitro protein digestibility was also studied. Genotypes of tepary bean (Phaseolus acutifolius) used were: PI‐301‐801 from Nicaragua; G‐400‐35 from Oaxaca, Mexico; L‐242‐24 and L‐242‐36 from Sonora, Mexico; L‐172 from Arizona, USA; and the common bean (Phaseolus vulgaris) variety Flor de Mayo Bajío (FMB) from Mexico. The cultivars were grown in a greenhouse under controlled temperature, light and humidity. Water content in plants (WCP) decreased when irrigation was stopped (1 and 2 weeks after blooming). Common bean had the lowest WCP value (73.75%), while the best genotypes were PI‐301‐801 (91.62%), G‐400‐35 (88.70%) and L‐242‐24 (85.73%). Tepary bean plants had superior water absorption capacity, which helped to increase yield. De novo synthesis of drought proteins may explain the increased protein content present under drought conditions. The results suggest that water stress induced ornithine amino transferase activity and ornithine concentration. Proline also increased under drought conditions (p < 0.05). Genotypes L‐242‐24 and G‐400‐35, which had the highest yield, also had a major concentration of polyamines under water stress. The results also suggest that arginase activity participated in amino acid synthesis such as ornithine and, consequently, in the protein concentration under water stress. Genotypes G‐400‐35 and L‐242‐24 are good alternatives for cultivation in arid zones. Copyright © 2003 Society of Chemical Industry

Rat colon ornithine and arginine metabolism: coordinated effects after proliferative stimuli.

Ornithine decarboxylase (ODC) catalyzes the first step in the polyamine biosynthetic pathway, a highly regulated pathway in which activity increases during rapid growth. Other enzymes also metabolize ornithine, and in hepatomas, rate of growth correlates with decreased activity of these other enzymes, which thus channels more ornithine to polyamine biosynthesis. Ornithine is produced from arginase cleavage of arginine, which also serves as the precursor for nitric oxide production. To study whether short-term coordination of ornithine and arginine metabolism exists in rat colon, ODC, ornithine aminotransferase (OAT), arginase, ornithine, arginine, and polyamine levels were measured after two stimuli (refeeding and/or deoxycholate exposure) known to synergistically induce ODC activity. Increased ODC activity was accompanied by increased putrescine levels, whereas OAT and arginase activity were reduced by either treatment, accompanied by an increase in both arginine and ornithine levels. These results indicate a rapid reciprocal change in ODC, OAT, and arginase activity in response to refeeding or deoxycholate. The accompanying increases in ornithine and arginine concentration are likely to contribute to increased flux through the polyamine and nitric oxide biosynthetic pathways in vivo.

Transforming Growth Factor-β 1 Stimulates l -Arginine Transport and Metabolism in Vascular Smooth Muscle Cells

Transforming growth factor-beta(1) (TGF-beta(1)) contributes to arterial remodeling by stimulating vascular smooth muscle cell (VSMC) growth and collagen synthesis at sites of vascular injury. Because L-arginine is metabolized to growth-stimulatory polyamines and to the essential collagen precursor L-proline, we examined whether TGF-beta(1) regulates the transcellular transport and metabolism of L-arginine by VSMCs. TGF-beta(1) increased L-arginine uptake, and this was associated with a selective increase in cationic amino acid transporter-1 (CAT-1) mRNA. In addition, TGF-beta(1) stimulated L-arginine metabolism by inducing arginase I mRNA and arginase activity. TGF-beta(1) also stimulated L-ornithine catabolism by elevating ornithine decarboxylase (ODC) and ornithine aminotransferase (OAT) activity. TGF-beta(1) markedly increased the capacity of VSMCs to generate the polyamine putrescine and L-proline from extracellular L-arginine. The TGF-beta(1)-mediated increase in putrescine and L-proline production was reversed by methyl-L-arginine, a competitive inhibitor of cationic amino acid transport, or by hydroxy-L-arginine, an arginase inhibitor. Furthermore, the formation of putrescine was inhibited by the ODC inhibitor alpha-difluoromethylornithine, and L-proline generation was blocked by the OAT inhibitor L-canaline. L-Canaline also inhibited TGF-beta(1)-stimulated type I collagen synthesis. These results demonstrate that TGF-beta(1) stimulates polyamine and L-proline synthesis by inducing the genes that regulate the transport and metabolism of L-arginine. In addition, they show that TGF-beta(1)-stimulated collagen production is dependent on L-proline formation. The ability of TGF-beta(1) to upregulate L-arginine transport and direct its metabolism to polyamines and L-proline may contribute to arterial remodeling at sites of vascular damage.

A Cortisol Surge Mediates the Enhanced Expression of Pig Intestinal Pyrroline-5-Carboxylate Synthase during Weaning

Citrulline synthesis from glutamine is enhanced remarkably in enterocytes of weanling pigs, but the molecular mechanism(s) involved are not known. The objective of this study was to determine whether a cortisol surge mediates the enhanced expression of intestinal citrulline-synthetic enzymes during weaning. Jejunal enterocytes were prepared from 29-d-old weanling pigs treated with or without metyrapone (an inhibitor of cortisol synthesis), or from age-matched unweaned pigs. The mRNA levels and activities of phosphate-dependent glutaminase (PDG), pyrroline-5-carboxylate synthase (P5CS), ornithine aminotransferase (OAT), carbamoyl-phosphate synthase I (CPS-I) and ornithine carbamoyltransferase (OCT) were determined. The mRNA levels for PDG, P5CS, OAT and OCT were 139, 157, 102 and 55% higher, respectively, in weanling pigs compared with suckling pigs. The activities of PDG and P5CS were 38 and 692% higher, respectively, in weanling pigs compared with unweaned pigs, but the activities of OAT, CPS-I and OCT did not differ between these two groups of pigs. The effects of metyrapone administration to weanling pigs were as follows: 1) prevention of a cortisol surge, 2) abolition of the increases in both mRNA levels and activity of P5CS, 3) no alteration in the mRNA levels and activities of PDG and CPS-I, 4) increases in the mRNA levels for OAT (216%) and OCT (39%) and in OAT activity (30%), and 5) prevention of the increase in intestinal synthesis of citrulline from glutamine. These results suggest that increased P5CS activity reflects in large part the increased levels of P5CS mRNA and is responsible for the increased synthesis of citrulline from glutamine in enterocytes of weanling pigs; these increases may be mediated by a cortisol surge during weaning that can be blocked by metyrapone administration.

Genotype-phenotype correlation of a pyridoxine-responsive form of gyrate atrophy

Two clinical subtypes of gyrate atrophy (GA) have been defined based on in vivo or in vitro evidence of response to vitamin B6 (pyridoxine), which is the cofactor of the enzyme ornithine aminotransferase (OAT) shown to be defective in GA. We identified the E318K mutation in the OAT gene, heterozygously in three patients and homozygously in one patient, all of whom were vitamin B6-responsive by previous in vivo and in vitro studies. Dose-dependent effects of the E318K mutation were observed in the homo- and heterozygotes in the OAT activity, increase of OAT activity in the presence of pyridoxal phosphate, and apparent Km for pyridoxal phosphate. The highest residual level of OAT activity and mildness of clinical disease correlated directly with the dose of the mutant E318K allele present in the patient.

A variant of ornithine aminotransferase from mouse small intestine.

The ornithine aminotransferase (OAT) activity of mouse was found to be highest in the small intestine. The mitochondrial OAT from mouse small intestine was purified to homogeneity by the procedures including heart treatment, ammonium sulfate fractionation, octyl-Sepharose chromatography, and Sephadex G-150 gel filtration. Comparing to the amino acid sequence of mouse hepatic OAT, six N-terminal amino acid residues have been deleted in intestinal OAT. However, the subsequent sequence was identical with that of hepatic OAT. The molecular weights of both intestinal and hepatic OAT were estimated as 46 kDa by SDS-gel electrophoresis and as 92 kDa by gel filtration, indicating that both native OATs are dimeric. Biochemical properties of intestinal OAT, such as molecular weight, pH optimum and K(m) values for L-ornithine and alpha-ketoglutarate, were similar to those of hepatic OAT. However, intestinal OAT was more labile than hepatic OAT to tryptic digestion.

Crystal structure of human recombinant ornithine aminotransferase

Ornithine aminotransferase (OAT), a pyridoxal-5′-phosphate dependent enzyme, catalyses the transfer of the δ-amino group of l-ornithine to 2-oxoglutarate, producing l-glutamate-γ-semialdehyde, which spontaneously cyclizes to pyrroline-5-carboxylate, and l-glutamate. The crystal structure determination of human recombinant OAT is described in this paper. As a first step, the structure was determined at low resolution (6Å) by molecular replacement using the refined structure of dialkylglycine decarboxylase as a search model. Crystallographic phases were then refined and extended in a step-wise fashion to 2.5 Å by cyclic averaging of the electron density corresponding to the three monomers within the asymmetric unit. Interpretation of the resulting map was straightforward and refinement of the model resulted in an R-factor of 17.1% ( R free=24.3%). The success of the procedure demonstrates the power of real-space molecular averaging even with only threefold redundancy. The α 6-hexameric molecule is a trimer of intimate dimers with a monomer-monomer interface of 5500 Å 2per subunit. The three dimers are related by an approximate 3-fold screw axis with a translational component of 18Å. The monomer fold is that of a typical representative of subgroup 2 aminotransferases and very similar to those described for dialkylglycine decarboxylase from Pseudomonas cepacia and glutamate-1-semialdehyde aminomutase from Synechococcus. It consists of a large domain that contributes most to the subunit interface, a C-terminal small domain most distant to the 2-fold axis and an N-terminal region that contains a helix, a loop and a three stranded β-meander embracing a protrusion in the large domain of the second subunit of the dimer. The large domain contains the characteristic central seven-stranded β-sheet (agfedbc) covered by eight helices in a typical α/β fold. The cofactor pyridoxal-5′-phosphate is bound through a Schiff base to Lys292, located in the loop between strands f and g. The C-terminal domain includes a four-stranded antiparallel β-sheet in contact with the large domain and three further helices at the far end of the subunit. The active sites of the dimer lie, about 25Å apart, at the subunit and domain interfaces. The conical entrances are on opposite sides of the dimer. In the active site, R180, E235 and R413 are probable substrate binding residues. Structure-based sequence comparisons with related transaminases in this work support that view. In patients suffering from gyrate atrophy, a recessive hereditary genetic disorder that can cause blindness in humans, ornithine aminotransferase activity is lacking. A large number of frameshift and point mutations in the ornithine aminotransferase gene have been identified in such patients. Possible effects of the various point mutations on the structural stability or the catalytic competence of the enzyme are discussed in light of the three-dimensional structure.

Arginine boxes and the argR gene in Streptomyces clavuligerus: evidence for a clear regulation of the arginine pathway.

The argR gene of Streptomyces clavuligerus has been located in the upstream region of argG. It encodes a protein of 160 amino acids with a deduced M(r) of 17117 for the monomer. Transformants containing the amplified argR gene showed lower activity (50%) of the biosynthetic ornithine carbamoyltransferase (OTC) activity and higher levels (380%) of the catabolic ornithine aminotransferase (OAT) activity than control strains. Amplification of an arginine (ARG) box-containing sequence results in a 2- to 2.5-fold derepression of ornithine acetyltransferase and OTC, suggesting that the repressor is titrated out. Footprinting experiments using the pure homologous arginine repressor (AhrC) of B. subtilis showed a protected 38 nt region (ARG box) in the coding strand upstream of argC. The protected region contained two tandemly repeated imperfect palindromic 18-nt ARG boxes. The repressor-operator interaction was confirmed by bandshift experiments of the DNA fragment containing the protected region. By computer analysis of the Streptomyces sequences available in the databases, a consensus ARG box has been deduced for the genus Streptomyces. This is the first example of a clear regulation of an amino acid biosynthetic pathway in Streptomyces species, challenging the belief that actinomycetes do not have a well-developed regulatory system of these pathways.

Exogenous spermine induces maturation of the liver in suckling rats

In the present study, we investigated the effects of spermine on postnatal liver maturation in suckling rats. The animals were given spermine either per os (8 micromol) or by intraperitoneal injection (1 micromol), once daily for three or five days. The percentage of liver cells in different cell cycle phases and of diploid cells in the parenchyma was estimated. The protein content, ornithine aminotransferase (OAT) activity, and content of DNA polyamines and receptors for polymeric immunoglobulins (RPI) were also measured in liver extracts. The ingestion of spermine had the following effects: the percentage of the cells in S and G2M phases of the cell cycle diminished the percentage of diploid cells increased the content of polymeric immunoglobulin receptors increased; the OAT activity increased; the contents of putrescine and spermidine decreased and almost reached adult values; and the spermidine/spermine ratio became similar to that observed in the liver of adult rats. These phenomena were detected 40 hours after the beginning of oral spermine treatment. The intraperitoneal injection of spermine had no effect on the OAT activity, but it decreased the spermidine content and enhanced the spermine content. Our data demonstrated for the first time that dietary polyamines play a role in the initiation of liver postnatal maturation in suckling rats. (Hepatology 1996 Nov;24(5):1206-10)

Cell growth in arginine- and ornithine-deprived medium and conversion of glutamate to ornithine and arginine in rat hepatoma cells

A subline growing in medium without arginine and ornithine was established from a rat Reuber hepatoma cell line (R-Y121B·cho). The subline designated R-Y117B·cho was able to grow in glutamine, arginine and ornithine-free, glutamate-supplemented medium. Arginine synthesis from glutamate requires four urea cycle enzymes and another two enzymes, glutamate semialdehyde dehydrogenase and ornithine aminotransferase. Since R-Y121B·cho cells have all the urea cycle enzymes, two other enzyme activities were determined. The activities of ornithine aminotransferase and glutamate semialdehyde dehydrogenase were similar in R-Y117B·cho and its parental R-Y121B·cho cells, but R-Y117B·cho cells had higher conversion of glutamate to arginine than parental cells.

Water stress‐induced alterations in the proline metabolism of drought‐susceptible and ‐tolerant cassava (Manihot esculenta) cultivars

The free proline levels and activities of ornithine aminotransferase (EC 2.6.1.13) and proline oxidase (EC 1.5.2.2), two of the enzymes involved in proline metabolism were studied during the induction of water stress in a drought susceptible (M‐4) and a drought tolerant (S‐1315) cultivar of cassava (Manihot esculenta Crantz). Water stress induced by polyethylene glycol (MW 6000, osmotic potential — 1.65 MPa) caused a ca 25‐fold increase in proline in young excised leaves of the susceptible cultivar (M‐4) while the increase was about 9‐fold in the tolerant cultivar (S‐1315). The activity of ornithine aminotransferase (OAT), a key enzyme involved in the biosynthesis of proline, was found to increase 3‐fold in water stressed leaves of M‐4 and about 2‐fold in those of S‐1315. The activity of proline oxidase, which is involved in the degradation of proline to pyrroline‐5‐carboxylate, was reduced by 50% in M‐4 and nearly 25% in S‐1315 on water stress. Comparison of the kinetic properties of OAT showed that the enzyme from water‐stressed leaves is more stable to heat inactivation compared to that of control. These results indicate that during water stress there are alterations in the metabolism of proline in cassava, and the extent of alteration varies between drought‐susceptible and ‐tolerant cultivars.

Proline and Proline Metabolising Enzymes in in-vitro Selected NaCl-tolerant Brassica juncea L. under Salt Stress

The effect of salt stress was studied on proline accumulation and the activities of proline metabolic pathway enzymes in seedling and leaf tissue of two genetically stable lines (SR2P1-2 and SR3P6-2) of in vitro selected NaCl-tolerant plants and parent cultivar Prakash of Brassica juncea L. Salt stress caused differential enhancement in proline level in both seedlings and leaf tissue of plants at different developmental stages. The magnitude of increase in proline content was higher in SR3P6-2 line in seedlings (34 fold at 140 meq-1 NaCl) as well as leaves (16 fold at 40 d after sowing at 100 meq-1 NaCl) compared to the parent cv. Prakash (29 fold in seedlings and five fold in leaves) and SR2P1-2 (21 fold in seedlings and five fold in leaves) at similar stress levels. Salt stress also resulted in changes in the activities of enzymes of proline metabolism. The activities of proline biosynthetic enzymes, pyrroline-5-carboxylate reductase and ornithine aminotransferase, increased under salt stress both in the seedlings and leaves. The range of increase in the activities of the two enzymes was relatively higher in SR3P6-2 (3·3-3·9 fold) compared to the SR2P1-2 (1·8-2·8 fold) and parent cv. Prakash (1·5-2·8 fold). The activity of proline degrading enzyme, proline oxidase, decreased under salt stress in both the tissues of all the lines; the reduction in activity was relatively greater in SR3P6-2 compared to SR2P1-2 or cv. Prakash. The trend of changes in the enzyme activities was in tune with the increase in proline level, the magnitude of change did not match the extent of increase in proline level.

Glutamine metabolism in chick enterocytes: absence of pyrroline-5-carboxylase synthase and citrulline synthesis.

This study was designed to determine whether pyrroline-5-carboxylate (P-5-C) synthase is deficient in chick enterocytes therefore resulting in the lack of synthesis of ornithine and citrulline from glutamine. Post-weaning pig enterocytes, which are known to contain P-5-C synthase and to synthesize both ornithine and citrulline from glutamine, were used as positive controls. Enterocytes were incubated at 37 degrees C for 0-30 min in the presence of 2 mM [U-14C]glutamine or 2 mM ornithine plus 2 mM NH4Cl. In chick enterocytes, glutamine was metabolized to NH3, CO2, glutamate, alanine and aspartate, but not to ornithine, citrulline, arginine or proline. Likewise, there was no formation of citrulline, arginine, alanine or aspartate from ornithine in chick enterocytes. Furthermore, the rate of conversion of ornithine into proline in chick enterocytes was only about 4% of that in cells from pigs. To elucidate the reason for the inability of chick enterocytes to synthesize ornithine and citrulline from glutamine, the activities of the enzymes involved were measured. No activity of P-5-C synthase or ornithine carbamoyltransferase was found in chick enterocytes, in contrast with cells from post-weaning pigs. It was also demonstrated that the activity of ornithine aminotransferase in chick enterocytes was only 3% of that in cells from pigs. Thus the present findings elucidate the biochemical reason for the lack of endogenous synthesis of ornithine and citrulline in chicks. Our results also explain previous observations that ornithine cannot replace arginine or proline in the diet of chicks. We suggest that the absence of P-5-C synthase and ornithine carbamoyltransferase in enterocytes is the metabolic basis for the nutritional requirement of arginine in the chick.

Arginine and ornithine metabolizing enzymes in testosterone-induced hypertrophic mouse kidney

Administration of testosterone to female mice causes hypertrophy of their kidneys with spectacular induction of ornithine decarboxylase and significant increase in the level of putrescine. We tried to find out whether testosterone treatment affects also the renal activities of enzymes participating in the formation and utilization of ornithine, specifically arginase and ornithine aminotransferase, and whether they are dependent on putrescine level. Swiss, CFW, DBA2 or F1 (CFW × DBA2) female and male mice were injected with testosterone (125 mg/kg) or CB 3717 (100 mg/kg). DFMO was applied in the drinking water. The activities of the enzymes were determined 24 hr or 5 days after administration of CB 3717 or testosterone, respectively. Renal activities of ornithine decarboxylase (ODC), arginase and ornithine aminotransferase (OAT) were found to be sex-differentiated. The highest activity of ODC was characteristic for the kidneys of males, whereas those of arginase and OAT for the kidneys of females. In the kidneys of testosterone-treated female mice a decrease (50%) of OAT, and a significant increase of arginase activities (up to 200%), were observed. In the males these changes were less pronounced. DFMO, which completely inhibited the activity of renal ODC, did not influence significantly the testosterone-induced arginase and the testosterone-decreased OAT. Arginase and OAT, in contrast to ODC, were not changed in CB 3717-induced hyperplastic kidney. The study showed testosterone-induced differential changes in the activity of two enzymes involved in ornithine biosynthesis and catabolism which accompanied ODC induction in female mouse kidney. However ODC induction was not a prerequisite for these enzymatic changes as they did not follow ODC activation by CB 3717, and were not affected by DFMO, a specific inhibitor of ODC.

Functional and metabolic changes in intestinal mucosa of rats after enteral administration of ornithine alpha-ketoglutarate salt.

Ornithine alpha-ketoglutarate salt efficiently improves the nutritional status of protein-depleted patients. Our aim was to explore the effects of ornithine alpha-ketoglutarate supplementation on intestinal physiology in healthy animals. Rats were given a nutritive mixture supplemented with ornithine alpha-ketoglutarate (1 g.kg-1 per day) by enteral route for 7 days. Controls received the diet supplemented with casein acid hydrolysate under isoenergetic and isonitrogenous conditions. An adaptive hyperplasia of the villi and an increase in the brush-border hydrolase activities were observed in rats receiving ornithine alpha-ketoglutarate. Because of the high ornithine aminotransferase activity, ornithine alpha-ketoglutarate-derived ornithine was extensively transaminated with a concomitant enhancement of ornithine decarboxylation. Surprisingly, with glutamate and putrescine, the products of ornithine transamination and decarboxylation, gamma-aminobutyric acid accumulated (10-fold to 16-fold) dramatically in the intestinal mucosa of rats treated with ornithine alpha-ketoglutarate. Because gamma-aminobutyric acid formation was completely prevented by the diamine oxidase inhibitor aminoguanidine but was not modified after inactivation of ornithine aminotransferase by 5-fluoromethylornithine, it is evident that gamma-aminobutyric acid is formed in the mucosa from ornithine via putrescine as an intermediate. It is assumed that enhanced gamma-aminobutyric acid formation in the intestinal mucosa by ornithine alpha-ketoglutarate treatment might be of physiologic importance in the regulatory processes of cell growth and differentiation.
