Online Two-Dimensional Liquid Chromatography for Determination of Selected Flavonoids in Wine Samples

The development of two-dimensional chromatography systems is crucial in many scientific fields as it provides improved separation efficiency and selectivity of complex mixtures. By separating compounds based on two independent properties, these systems can offer a more detailed understanding of sample composition. This increased level of detail is particularly useful in industries such as pharmaceuticals, environmental analysis, and proteomics, where complex mixtures with similar properties need to be differentiated. A new chromatography system with two dimensions was developed to separate and quantify specific flavonoids in German wine samples. The first dimension used a homemade hydrophilic interaction liquid chromatographic column, while the second dimension used a reversed-phase liquid chromatographic column. This 2D ZIC-HILIC x RP technique was able to address the problem of peak overlapping that commonly occurs in one-dimensional liquid chromatography, especially for natural products. The study presents analytical and statistical data for both techniques, and suggests that the use of two-dimensional chromatography can improve the resolution of conventional one-dimensional liquid chromatography. In a conventional ZIC-HILIC-HPLC system, apigenin and naringenin were detected together but were successfully separated and measured using a 2DLC system. The limits of detection (LOD) for apigenin and naringenin were found to be 0.0705 µg/mL and 0.1300 µg/mL, respectively, while the limits of quantification (LOQ) were 0.2136 µg/mL and 0.3939 µg/mL, respectively. The recovery rate for both compounds was over 99%, with a relative standard deviation (RSD) of less than 1.5%. The linearity range for both compounds was between 0.005 µg/mL to 10 µg/mL, and the correlation coefficient (r2) was determined to be 0.9998. For hesperidin and rutin, the LOD and LOQ were found to be 0.0336 µg/mL and 0.1018 µg/mL, and 0.0120 µg/mL and 0.0363 µg/mL, respectively. The linearity range for both compounds was between 0.005 µg/mL to 12 µg/mL, and the correlation coefficient (r2) was determined to be 0.9999.

Two-dimensional liquid chromatography with reversed phase in both dimensions: A review

Two-dimensional liquid chromatography (2D-LC), and in particular comprehensive two-dimensional liquid chromatography (LC×LC), offers increased peak capacity, resolution and selectivity compared to one-dimensional liquid chromatography. It is commonly accepted that the technique produces the best results when the separation mechanisms in the two dimensions are completely orthogonal; however, the use of similar separation mechanisms in both dimensions has been gaining popularity as it helps avoid difficulties related to mobile phase incompatibility and poor column efficiency.The remarkable advantages of using reversed phase in both dimensions (RPLC×RPLC) over other separation mechanisms made it a promising technique in the separation of complex samples. This review discusses some physical and practical considerations in method development for 2D-LC involving the use of RP in both dimensions. In addition, an extensive overview is presented of different applications that relied on RPLC×RPLC and 2D-LC with reversed phase column combinations to separate components of complex samples in different fields including food analysis, natural product analysis, environmental analysis, proteomics, lipidomics and metabolomics.

Simultaneous analysis of highly polar and multi-residue-type pesticides by heart-cutting 2D-LC-MS

Liquid chromatography-mass spectrometry (LC-MS) is currently the gold-standard technique for the analysis of non-volatile small organic molecules. However, one-dimensional liquid chromatography (1D-LC) cannot efficiently deal with mixtures of analytes with different physicochemical properties and, thus, specific chromatographic behaviour. As an alternative, this work proposes a two-dimensional liquid chromatography/high-resolution mass spectrometry (2D-LC-HRMS) approach for the simultaneous analysis of compounds with different polarities. It is based on the combination of hydrophilic interaction liquid chromatography (HILIC) in the first dimension (1D) and reversed-phase chromatography (RPLC) in the second dimension (2D), employing the heart-cutting methodology. The coupling between 1D and 2D was performed by a multiple heart-cutting (MHC) interface equipped with an active solvent modulation (ASM) valve. The aim of the study was the development of a 2D-LC methodology able to (i) acquire the 1D and 2D content by MS in a single analytical run, avoiding the loss of information caused by the MHC algorithm for filling the sampling loops; (ii) overcome the breakthrough problem caused by solvent incompatibility, modifying the 2D gradient during the ASM phase for this purpose. To evaluate the 2D-LC approach, pesticide residue analysis was proposed, selecting 20 pesticides covering a wide range of polarities (log Kow from −3.2 to 4.3) and including some of the so-called single residue method pesticides because of the difficulty of including them in 1D-LC multi-residue methods with satisfactory chromatographic resolution. The proposed strategy was to transfer in a single cut the void volume from the HILIC separation (consisting of the nonpolar pesticides) to the 2D for analysis under RPLC conditions. The developed assembly was assessed in a vegetable matrix (tomato) employing a hybrid QuEChERS/QuPPe sample treatment based on acetonitrile and methanol extraction. The proposed setup may be extended for 2D-LC applications where it is essential to acquire the entire content of both dimensions in a single data file just by coupling a selection valve to the MHC interface.

Polybutylene terephthalate-based stationary phase for ion-pair-free reversed-phase liquid chromatography of small interfering RNA. Part 2: Use for selective comprehensive two-dimensional liquid chromatography

With the increasing numbers of nucleic acid-based pharmaceuticals like antisense oligonucleotides (ASO), small interfering ribonucleic acid (siRNA) entering the market, research facilities, pharmaceutical industries and also regulatory authorities have been looking for efficient analytical methods for these synthetic oligonucleotides (ON). Besides of conventional one-dimensional (1D) reversed-phase liquid chromatography with or without ion-pairing (IP-RP-LC, RP-LC), hydrophilic liquid chromatography (HILIC) and mixed-mode chromatography (MMC), two-dimensional (2D) approaches combining two orthogonal chromatographic techniques also become more relevant due to the high structural complexity of oligonucleotides. Recently, we tested a polybutylene terephthalate(PBT)-based stationary phase under ion-pairing free RP mode for the liquid chromatography electrospray ionization mass spectrometry (LC-ESI-MS) analysis of siRNA (Patisiran). In this study, retention profile and chromatographic orthogonality, respectively, were compared to other LC-modes like HILIC, IP-RPLC, another ion-pair free cholesterol-bonded RPLC and MMC considering their normalized retention times. Finally, because of higher orthogonality, the ion-pairing free PBT-bonded RPLC as first dimension (1D) was hyphenated with HILIC in the second dimension (2D) in a selective comprehensive 2D-LC setup leading to an enhanced resolution for peak purity evaluation of the main ON entities.

Both heart-cutting and comprehensive applications are described and discussed in this review, highlighting the potential of 2D-LC-MS for the analysis of such complex samples.

Diluting modulation-based two dimensional-liquid chromatography coupled with mass spectrometry for simultaneously determining multiclass prohibited substances in cosmetics

Developing efficient and comprehensive screening methods for prohibited substances in cosmetics is critical for ensuring the quality and safety of cosmetics used in everyday life. This study proposed a heart-cutting two-dimensional liquid chromatography-mass spectrometry (2D-LC-MS) method based on online diluting modulation for detecting multiclass prohibited substances in cosmetics. The 2D-LC-MS method combines HILIC and RPLC techniques. Compounds near the dead time that the first dimensional HILIC could not separate were transferred to the second dimensional RPLC by valve switch, achieving good separation with a wide range of polarities. Moreover, the online diluting modulation solved the problem of mobile phase incompatibility, realizing an excellent column-head focusing effect and reducing the loss of sensitivity. Besides, the first dimensional analysis did not restrict the flow rate of the second dimensional analysis owing to the diluting modulation. We demonstrated the 2D-LC-MS system by determining 126 prohibited substances in cosmetic products, including hormones, local anesthetics, anti-infectives, adrenergic agents, antihistamines, pesticides, and other chemicals. All correlation coefficients of the compounds were above 0.9950. The LODs and the LOQs ranged from 0.000259 ng/mL to 16.6 ng/mL and 0.000864 ng/mL to 55.3 ng/mL, respectively. The RSDs% for intra-day and inter-day precision were within 6% and 14%, respectively. Compared with conventional one-dimensional liquid chromatography methods, the established method expanded the analytical coverage of cosmetics-prohibited substances with reduced matrix effects for most compounds and improved sensitivity for polar analytes. The results indicated that the 2D-LC-MS method was a powerful tool for screening multiclass prohibited substances in cosmetics.

Identification and quantification of polymeric impurity in block copolymer by one-dimensional and two-dimensional liquid chromatography coupled to high-resolution mass spectrometry and evaporative light scattering detector

Identifying and quantifying polymeric impurities in a polymeric material is critical for understanding material quality and performance, but it remains a challenge requiring developing new characterization methods. In this work, a comprehensive two-dimensional liquid chromatography method with simultaneous evaporative light scattering and high-resolution mass spectrometry detection was developed to separate and identify a polymeric impurity in alkyl alcohol-initiated polyethylene oxide/polybutylene oxide diblock copolymer. Size exclusion chromatography was implemented in the first dimension, and gradient reversed-phase liquid chromatography using a large-pore C4 column was applied in the second dimension using an active solvent modulation valve as the interface to minimize the polymer breakthrough. The two-dimensional separation significantly reduced the complexity of the mass spectra data compared to the one-dimensional separation, and the combination of retention time and mass spectral interpretation led to the successful identification of the water-initiated triblock copolymer impurity. This identification was confirmed by comparison with the synthesized triblock copolymer reference material. A one-dimensional LC method with evaporative light scattering detection was employed to quantify the triblock impurity. The impurity level in three samples made with the different processes was determined to be in the range of 9–18 wt% using the triblock reference material as the standard.

Trapping array-based preparative two-dimensional liquid chromatography for the purification of four components in tobacco leaves

Two-dimensional liquid chromatography (2D-LC) has gained increased attention because of its high peak capacity for separating complex samples. However, preparative 2D-LC aimed at isolating compounds is significantly different compared with one-dimensional liquid chromatography (1D-LC) in terms of method development and system configuration; thus, it is less developed than its analytical counterpart. The use of 2D-LC in large-scale product preparation has rarely been reported. Hence, a preparative 2D-LC system was developed in this study. The system was composed of one set of preparative LC modules as a separation system, with a dilution pump, switch valves, and trap column array as the interface, to enable the simultaneous isolation of several compounds. Tobacco was used as a sample, and the developed system was applied to isolate nicotine, chlorogenic acid, rutin, and solanesol. The chromatographic conditions were developed by investigating the trapping efficiency of different types of trap column packings, and chromatographic behaviors under different overload conditions. The four compounds were isolated in one 2D-LC run with high purity. The developed system features low cost because it employs medium-pressure isolation, excellent automation owing to its use of an online column switch, high stability, and capability for large-scale production. The isolation of chemicals from tobacco leaves as pharmaceutical raw materials could aid in the development of the tobacco industry and promote the local agricultural economy.

Theoretical and practical comparison of RPLC and RPLC × RPLC: how to consider dilution effects and sensitivity in addition to separation power?

The objective of this work was to provide an unbiased comparison of one-dimensional reversed-phase liquid chromatography (1D-RPLC) and comprehensive two-dimensional RPLC (RPLC × RPLC), through calculations and experimental verifications. For this purpose, various quality descriptors were evaluated, including peak capacity, analysis time, dilution factor, number of runs in the second dimension, and injection volume. The same strategy was applied to small pharmaceuticals and peptides. Whatever the analysis time between 30 and 200min, short columns of only 30 × 2.1mm packed with sub-2-µm particles should be selected in both dimensions of the 2D-LC setup to obtain the best compromise in terms of peak capacity and sensitivity. The peak capacity in RPLC × RPLC vs. RPLC was significantly improved for analysis times beyond 5min. However, extra-column volume located after the second-dimension column was found to be particularly critical for peptides, and up to 50% lower peak capacity was observed with MS vs. UV detection. Contrary to common belief, higher dilution is not always observed in RPLC × RPLC. With adequate analytical conditions, better sensitivity (in theory fivefold and in practice three- to fivefold) could be achieved in RPLC × RPLC compared to 1D-RPLC, regardless of the analysis time.

Quantification strategies for two-dimensional liquid chromatography datasets using regions of interest and multivariate curve resolution approaches

In this work, three chemometrics-based approaches are compared for quantification purposes when using two-dimensional liquid chromatography (LC×LC-MS), taking as a study case the quantification of amino acids in commercial drug mixtures. Although the approaches have been already used for one-dimensional gas or liquid chromatography, the main novelty of this work is the demonstration of their applicability to LC×LC-MS datasets. Besides, steps such as peak alignment and modelling, commonly applied in this type of data analysis, are not required with the approaches proposed here. In a first step, regions of interest (ROI) strategy is used for the spectral compression of the LC×LC-MS datasets. Then the first strategy consists of building a calibration curve from the areas obtained in this ROI compression step. Alternatively, the ROI intensity matrices can be used as input for a second analysis step employing the multivariate curve resolution alternating least squares (MCR-ALS) method. The main benefit of MCR-ALS is the resolution of elution and spectral profiles for each of the analytes in the mixture, even in the case of strong coelutions and high signal overlapping. Classical MCR-ALS based calibration curve from the peak areas resolved only applying non-negativity constraints (second strategy) is compared to the results obtained when an area correlation constraint is imposed during the ALS optimization (third strategy). All in all, similar quantification results were achieved by the three approaches but, especially in prediction studies, the more accurate quantification is obtained when the calibration curve is built from the peak areas obtained with MCR-ALS when the area correlation constraint is imposed.

Green approaches to comprehensive two-dimensional liquid chromatography (LC × LC)

Comprehensive two-dimensional liquid chromatography (LC×LC) has been adopted in the analysis of complex mixtures because it can achieve much higher peak capacity and resolution than one-dimensional liquid chromatography. The ever-growing demand for protecting the environment necessitates reducing or eliminating hazardous solvents to improve the environmental friendliness of analytical procedures. Acetonitrile is one of class II solvents which are hazardous to the environment and should be phased out and replaced by eco-friendlier alternatives. Herein, we introduce the use of propylene carbonate as an eco-friendly mobile phase component in comprehensive two-dimensional liquid chromatography to separate a mixture of 39 pharmaceutical compounds. Two sets of experiments were developed. In the first set of experiments, two emulated on-line LC × LC systems were developed using Solvent X (propylene carbonate: ethanol, 60:40) as a green organic modifier in the first dimension (1D) and ethanol in the second dimension (2D). Upon comparing the separation performance of these two systems to two systems described in the literature that used acetonitrile and methanol instead, no significant differences in terms of selectivity, peak capacity and orthogonality were found. In the second set of experiments, three comprehensive LC × LC systems were developed using Solvent X or acetonitrile in the first dimension, while ethanol or methanol were used in the second dimension. The same stationary phases were used in the three systems, but the organic modifiers were changed in one or both dimensions in order to confirm that acetonitrile can be replaced by a green alternative without losing separation efficiency. There was no significant difference in terms of selectivity, peak capacity and orthogonality between the three systems.

Comprehensive profiling of Sanguisorba officinalis using offline two-dimensional mixed-mode liquid chromatography × reversed-phase liquid chromatography, tandem high-resolution mass spectrometry, and molecular network.

The profiling of natural products is important in modern biological sciences and new drug development. However, the separation and characterization of complex herbal extracts are significantly challenging for researchers in the biochemical field. Herein, an offline two-dimensional mixed-mode liquid chromatography × reversed-phase liquid chromatography system is developed. Our system exhibits high orthogonality and is composed of a newly prepared stationary phase in the first dimension and a traditional C18 phase in the second dimension, and is operated in combination with a high-resolution mass spectrometry and molecular network. Sanguisorba officinalis L. is studied using the proposed method owing to its bioactivity. With the aid of orthogonal separation, the ionization of the individual components is improved. The number of detected compounds and separated peaks are significantly increased when one-dimensional liquid chromatography is upgraded to two-dimensional liquid chromatography. In addition, 270 compounds (127 of which are tentatively characterized as new compounds, and further confirmation is needed) are successfully characterized based on their fragmentation patterns under the guidance of molecular network, while only 95 compounds are characterized using one-dimensional liquid chromatography and high-resolution mass spectrometry. The results indicate that the developed offline two-dimensional mixed-mode liquid chromatography × reversed-phase liquid chromatography, tandem high-resolution mass spectrometry, and molecular network method are effective for profiling complex samples.

Strategies to circumvent the solvent strength mismatch problem in online comprehensive two-dimensional liquid chromatography.

On-line comprehensive two-dimensional liquid chromatography is a powerful technique for the separation of highly complex samples. Due to the addition of the second dimension of separation, impressive peak capacities can be obtained within a reasonable analysis time compared to one-dimensional liquid chromatography. In online comprehensive two-dimensional liquid chromatography, the separation power is maximized by selecting two separation dimensions as orthogonal as possible, which most often requires the combination of different mobile phases and stationary phases. The online transfer of a given solvent from the first dimension to the second dimension may cause severe injection effects in the second dimension, mostly due to solvent strength mismatch. Those injection effects may include peak broadening, peak distortion, peak splitting or breakthrough phenomenon. They are often found to reduce significantly the peak capacity and the peak intensity. To overcome such effects, arising specifically in online comprehensive two-dimensional liquid chromatography, different methods have been developed over the years. In this review, we focused on the most recently reported ones. A critical discussion, supported by a theoretical approach, gives an overview of their advantages and drawbacks.

Multiple Heart-Cutting Two-Dimensional Liquid Chromatography: Recent Developments and Applications

Background: Over the last few decades, there has been a growing interest in Two- Dimensional Liquid Chromatography (2D-LC). This advanced technology has become a relevant solution for solving all the analytical challenges encountered in the pharmaceutical industry. The primary purpose of this review is to give a brief account of the developing techniques notable in 2D-LC. Methods: Review of recent literature on advanced separation techniques for complex mixtures revealed that 2D-LC is the most widely used technique in the pharmaceutical laboratory for the separation of complex samples. Among the various techniques used in 2D-LC, comprehensive 2D-LC and heart-cutting 2D-LC are highly used techniques. To enhance the resolution, further development has been made to heart-cutting 2D-LC called multiple heart-cutting 2D-LC, which has recently become a very popular technique in the pharmaceutical industry. Therefore research articles mentioning the use of multiple heart-cutting 2D-LC for separation of complex mixtures were reviewed. Results: The most crucial benefit of multiple heart-cutting 2D-LC is its reduced effort in method development and also the storage of multiple sample sections for further analysis. The multiple heartcutting 2D-LC is suited, preferably for method development and impurity analysis of pharmaceutical substances, fine chemicals and offers better peak separation, which is otherwise not possible by comprehensive 2D-LC. Conclusion: The advantage of two-dimensional liquid chromatography over one-dimensional liquid chromatography is its high resolving power. Further improvement in resolution was achieved by using the advanced multiple heart-cutting 2D-LC which appears to have an imperative role in pharmaceutical and food analysis.

Liquid Chromatographic Separation Using a 2 μm i.d. Open Tubular Column at Elevated Temperature.

We have experimentally demonstrated the extraordinarily high resolving power of liquid chromatography (LC) using a narrow open tubular (OT) column. In this work, we show that we can further increase its efficiency, peak capacity, and separation speed by elevating the operation (or column) temperature; all of these three numbers can be improved without mutual compromises. We use a mixture of five amino acids as a sample and show that we can increase the efficiency by 34%-260% and the separation speeds by 7%-10% by raising the operation temperature from 30 to 70 °C. When we use a 2 μm i.d. × 80 cm in length OT column coated with OTMS at a temperature of 70 °C, we can frequently obtain peak capacities of 700-800 within 20-30 min for separating cytochrome C digests. By increasing the column length to 160 cm, we can obtain a peak capacity of 2720 within 143 min for separating a complex peptide sample. This peak capacity is the highest peak capacity to date for one-dimensional LC separations. Importantly, heating the column is easy to implement and does not cost much, and many commercial LC systems already have compartments to control column temperatures. Running LC using a narrow OT column at an elevated temperature should broaden the applications of OT-LC in chemical and biochemical analyses.

Multi-dimensional LC-MS: the next generation characterization of antibody-based therapeutics by unified online bottom-up, middle-up and intact approaches.

Accelerated development of new therapeutics in an increasingly competitive landscape requires the use of high throughput analytical platforms. In addition, the complexity of novel biotherapeutic formats (e.g. fusion proteins, protein-polymer conjugates, co-formulations, etc.) reinforces the need to improve the selectivity and resolution of conventional one-dimensional (1D) liquid chromatography (LC). Liquid chromatography-mass spectrometry (LC-MS)-based technologies such as native LC-MS for intact mass analysis or peptide mapping (also called bottom-up approach)-based multi-attribute methods (MAM) have already demonstrated their potential to complement the conventional analytical toolbox for monoclonal antibody (mAb) characterization. Two-dimensional liquid-chromatography (2D-LC-MS) methods have emerged in the last ten years as promising approaches to address the increasing analytical challenges faced with novel antibody formats. However, off-line sample preparation procedures are still required for conventional 1D and 2D-LC-MS methods for the in-depth variant characterization at the peptide level. Multi-dimensional LC-MS (mD-LC-MS) combine sample preparation and multi-level (i.e. intact, reduced, middle-up and peptide) analysis within the same chromatographic set-up. This review presents an overview of the benefits and limitations of mD-LC-MS approaches in comparison to conventional chromatographic methods (i.e. 1D-LC-UV methods at intact protein level and 1D-LC-MS methods at peptide level). The current analytical trends in antibody characterization by mD-LC-MS approaches, beyond the 2D-LC-MS workhorse, are also reviewed, and our vision on a more integrated multi-level mD-LC-MS characterization platform is shared.

Multidimensional liquid chromatography approaches for analysis of food contaminants.

In the past years, multidimensional liquid chromatography became very widespread for the complete separation of non-volatile analytes in complex matrices. The main advantage of these techniques, especially when coupled with mass spectrometry, is the enhancement of separation power or peak capacity, due to an increase in selectivity and sensitivity of the two systems. With respect to conventional one-dimensional liquid chromatography, multidimensional liquid chromatography allows us to resolve potential co-elutions and also minimize the matrix effect thus providing a more accurate quantitative analysis. This review provides an overview on the presence of contaminants in food, the main sources of contamination, and finally, the techniques used to reveal their presence. All different modes, investigated so far on this topic for heart-cutting and comprehensive techniques were described. Advantages and disadvantages of each method are reported in addition to the main food applications.

Parallel gradients in comprehensive multidimensional liquid chromatography enhance utilization of the separation space and the degree of orthogonality when the separation mechanisms are correlated.

Herein, we argue that when the separation mechanisms are correlated in the two dimensions, the best results can be obtained with the use of parallel gradients in the second dimension, which makes the technique nearly as user-friendly as comprehensive two-dimensional gas chromatography. This has been illustrated through the separation of a mixture of 39 pharmaceutical compounds using reversed phase in both dimensions. Different selectivity in the second dimension was obtained through the use of different stationary phase chemistries and/or mobile phase organic modifiers. The best coverage of the separation space was obtained when parallel gradients were applied in both dimensions, and the same was true for practical peak capacity.

Evaluation of LC-MS and LC\xd7LC-MS in analysis of zebrafish embryo samples for comprehensive lipid profiling

In this study, both conventional one-dimensional liquid chromatography (1DLC) and comprehensive two-dimensional liquid chromatography (2DLC) coupled to a high-resolution time-of-flight mass spectrometer (HR-TOF MS) were used for full-scale lipid characterization of lipid extracts from zebrafish embryos. We investigated the influence on annotated lipids and different separation mechanisms (HILIC, C18, and PFP), and their different orders arranged in the first and the second dimensions. As a result, the number of lipid species annotated by conventional one-dimensional LC-MS was between 212 and 448. In contrast, the number of individual lipids species annotated by C18×HILIC, HILIC×C18, and HILIC×PFP were 1784, 1059, and 1123, respectively. Therefore, it was evident that the performance of comprehensive 2DLC, especially the C18×HILIC method, considerably exceeded 1DLC. Interestingly, a comparison of the HILIC×C18 and C18×HILIC approaches showed, under the optimized conditions, similar orthogonality, but the effective separation power of the C18×HILIC was much higher. A comparison of the HILIC×C18 and the HILIC×PFP methods demonstrated that the HILIC×PFP separation had superior orthogonality with a small increase on its effective peak capacity, indicating that the HILIC×PFP combination maybe a promising platform for untargeted lipidomics in complex samples. Finally, from the comprehensive lipid profiling respective, the C18×HILIC was selected for further studies.
