Utilizing transcriptomics and proteomics to unravel key genes and proteins of Oryza sativa seedlings mediated by selenium in response to cadmium stress

BackgroundCadmium (Cd) pollution has declined crop yields and quality. Selenium (Se) is a beneficial mineral element that protects plants from oxidative damage, thereby improving crop tolerance to heavy metals. The molecular mechanism of Se-induced Cd tolerance in rice (Oryza sativa) is not yet understood. This study aimed to elucidate the beneficial mechanism of Se (1 mg/kg) in alleviating Cd toxicity in rice seedlings.ResultsExogenous selenium addition significantly improved the toxic effect of cadmium stress on rice seedlings, increasing plant height and fresh weight by 20.53% and 34.48%, respectively, and increasing chlorophyll and carotenoid content by 16.68% and 15.26%, respectively. Moreover, the MDA, ·OH, and protein carbonyl levels induced by cadmium stress were reduced by 47.65%, 67.57%, and 56.43%, respectively. Cell wall metabolism, energy cycling, and enzymatic and non-enzymatic antioxidant systems in rice seedlings were significantly enhanced. Transcriptome analysis showed that the expressions of key functional genes psbQ, psbO, psaG, psaD, atpG, and PetH were significantly up-regulated under low-concentration Se treatment, which enhanced the energy metabolism process of photosystem I and photosystem II in rice seedlings. At the same time, the up-regulation of LHCA, LHCB family, and C4H1, PRX, and atp6 functional genes improved the ability of photon capture and heavy metal ion binding in plants. Combined with proteome analysis, the expression of functional proteins OsGSTF1, OsGSTU11, OsG6PDH4, OsDHAB1, CP29, and CabE was significantly up-regulated under Se, which enhanced photosynthesis and anti-oxidative stress mechanism in rice seedlings. At the same time, it regulates the plant hormone signal transduction pathway. It up-regulates the expression response process of IAA, ABA, and JAZ to activate the synergistic effect between each cell rapidly and jointly maintain the homeostasis balance.ConclusionOur results revealed the regulation process of Se-mediated critical metabolic pathways, functional genes, and proteins in rice under cadmium stress. They provided insights into the expression rules and dynamic response process of the Se-mediated plant resistance mechanism. This study provided the theoretical basis and technical support for crop safety in cropland ecosystems and cadmium-contaminated areas.

The Metabolism, Detrimental Effects, and Signal Transduction Mechanism of Reactive Oxygen Species in Plants under Abiotic Stress

Plants are a major source of bio based materials, and their productivity is significantly impacted by environmental adversities with the continuous deterioration of the environment. The disruption of reactive oxygen species (ROS) production and scavenging balance is a common consequence of various abiotic stresses in plants, resulting in the excessive accumulation of ROS. However, it is crucial to acknowledging the dual role of ROS, acting both as harmful substances and signaling molecules. ROS can interact with membrane proteins, initiating a chain reaction of membrane lipid peroxidation. This process leads to a decrease in membrane lipid unsaturation and fluidity, as well as an increase in membrane permeability. Inhibited enzyme activity even lead to loss of protein function, caused DNA damage and mutations, blocked DNA replication and transcription, eventually leading to cell death. This can result in significant reductions in crop yield and biomass in bio based agricultural production. ROS also played vital role in signal molecule transduction in the process of plant resistance to stress, and worked in concert with other signal molecules to induce the stress resistance genes’ expression and enhance plant resistance. Hereto, scavenging mechanism for ROS in plants is of the essence to regulate the concentration of ROS, thus determining which role ROS play. The thermal dissipation of excess light energy effectively avoids ROS production, and the enzymatic and non-enzymatic antioxidant defense systems in plants work cooperatively to clear excessive ROS. This review provides a comprehensive summary of the production pathway, detrimental effects, scavenging mechanism and signal transduction of ROS in plants under abiotic stress.

Oxidative Stress Response Mechanisms Sustain the Antibacterial and Antioxidant Activity of Quercus ilex.

The development of new natural antibiotics is considered as the heart of several investigations in the nutraceutical field. In this work, leaves of Quercus ilex L. treated by tropospheric ozone (O3) and nitrogen (N) deposition, exhibited a clear antimicrobial efficacy against five multi-drug resistant (MDR) bacterial strains (two gram-positive and three gram-negative). Under controlled conditions, it was studied how simulated N deposition influences the response to O3 and the antibacterial and antioxidant activity, and antioxidant performance. The extraction was performed by ultra-pure acetone using two different steps. A higher antioxidant activity was measured in the presence of interaction between O3 and N treatments on Quercus leaves. At the same time, all organic extracts tested have shown bacteriostatic activity against all the tested strains with a MIC comprised between 9 and 4 micrograms/mL, and a higher antioxidant efficacy shown by spectrophotometric assay. Stronger antimicrobial activity was found in the samples treated with O3, whereas N-treated plants exhibited an intermediate antibacterial performance. This performance is related to the stimulation of the non-enzymatic antioxidant system induced by the oxidative stress, which results in an increase in the production of antimicrobial bioactive compounds.

Arbuscular mycorrhizal fungi enhanced resistance to low-temperature weak-light stress in snapdragon (Antirrhinum majus L.) through physiological and transcriptomic responses.

Low temperature (LT) and weak light (WL) seriously affects the yield and quality of snapdragon in winter greenhouse. Arbuscular mycorrhizal fungi (AMF) exert positive role in regulating growth and enhancing abiotic stress tolerance in plants. Nevertheless, the molecular mechanisms by AMF improve the LT combined with WL (LTWL) tolerance in snapdragon remain mostly unknown. We compared the differences in root configuration, osmoregulatory substances, enzymatic and non-enzymatic antioxidant enzyme defense systems and transcriptome between AMF-inoculated and control groups under LT, WL, low light, and LTWL conditions. Our analysis showed that inoculation with AMF effectively alleviated the inhibition caused by LTWL stress on snapdragon root development, and significantly enhanced the contents of soluble sugars, soluble proteins, proline, thereby maintaining the osmotic adjustment of snapdragon. In addition, AMF alleviated reactive oxygen species damage by elevating the contents of AsA, and GSH, and the activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR), monodehydroascorbate reductase (MDHAR), and glutathione reductase (GR). RNA-seq analysis revealed that AMF regulated the expression of genes related to photosynthesis (photosystem I related proteins, photosystem II related proteins, chlorophyll a/b binding protein), active oxygen metabolism (POD, Fe-SOD, and iron/ascorbate family oxidoreductase), plant hormone synthesis (ARF5 and ARF16) and stress-related transcription factors gene (bHLH112, WRKY72, MYB86, WRKY53, WRKY6, and WRKY26) under LTWL stress. We concluded that mycorrhizal snapdragon promotes root development and LTWL tolerance by accumulation of osmoregulatory substances, activation of enzymatic and non-enzymatic antioxidant defense systems, and induction expression of transcription factor genes and auxin synthesis related genes. This study provides a theoretical basis for AMF in promoting the production of greenhouse plants in winter.

Enhanced resistance to heat and fungal infection in transgenic Trichoderma via over-expressing the HSP70 gene

Heat stress is one of the major abiotic stresses affecting the growth, sporulation, colonization and survival of Trichoderma viride. This study aimed to gain a better insight into the underlying mechanism governing the heat stress response of T. viride Tv-1511. We analysed the transcriptomic changes of Tv-1511 under normal and heat stress conditions using RNA sequencing. We observed that Tv-1511 regulates the biosynthesis of secondary metabolites through a complex network of signalling pathways. Additionally, it significantly activates the anti-oxidant defence system, heat shock proteins and stress-response-related transcription factors in response to heat stress. TvHSP70 was identified as a key gene, and transgenic Tv-1511 overexpressing TvHSP70 (TvHSP70-OE) was generated. We conducted an integrated morphological, physiological and molecular analyses of the TvHSP70-OE and wild-type strains. We observed that TvHSP70 over-expression significantly triggered the growth, anti-oxidant capacity, anti-fungal activity and growth-promoting ability of Tv-1511. Regarding anti-oxidant capacity, TvHSP70 primarily up-regulated genes involved in enzymatic and non-enzymatic anti-oxidant systems. In terms of anti-fungal activity, TvHSP70 primarily activated genes involved in the synthesis of enediyne, anti-fungal and aminoglycoside antibiotics. This study provides a comparative analysis of the functional significance and molecular mechanisms of HSP70 in Trichoderma. These findings provide a valuable foundation for further analyses.

Perfluorobutanoic acid triggers metabolic and transcriptional reprogramming in wheat seedlings

The environmental risks of fluorinated alternatives are of great concern with the phasing out of perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate. Here, multi-omics (i.e., metabolomics and transcriptomics) coupled with physiological and biochemical analyses were employed to investigate the stress responses of wheat seedings (Triticum aestivum L.) to perfluorobutanoic acid (PFBA), one of the short-chain per- and polyfluoroalkyl substances (PFAS) and PFOA alternatives, at environmentally relevant concentrations (0.1–100 ng/g). After 28 days of soil exposure, PFBA boosted the generation of OH and O2− in wheat seedlings, resulting in lipid peroxidation, protein perturbation and impaired photosynthesis. Non-enzymatic antioxidant defense systems (e.g., glutathione, phenolics, and vitamin C) and enzymatic antioxidant copper/zinc superoxide dismutase were strikingly activated (p < 0.05). PFBA-triggered oxidative stress induced metabolic and transcriptional reprogramming, including carbon and nitrogen metabolisms, lipid metabolisms, immune responses, signal transduction processes, and antioxidant defense-related pathways. Down-regulation of genes related to plant-pathogen interaction suggested suppression of the immune-response, offering a novel understanding on the production of reactive oxygen species in plants under the exposure to PFAS. The identified MAPK signaling pathway illuminated a novel signal transduction mechanism in plant cells in response to PFAS. These findings provide comprehensive understandings on the phytotoxicity of PFBA to wheat seedlings and new insights into the impacts of PFAS on plants.

NO and H2S Contribute to Crop Resilience against Atmospheric Stressors.

Atmospheric stressors include a variety of pollutant gases such as CO2, nitrous oxide (NOx), and sulfurous compounds which could have a natural origin or be generated by uncontrolled human activity. Nevertheless, other atmospheric elements including high and low temperatures, ozone (O3), UV-B radiation, or acid rain among others can affect, at different levels, a large number of plant species, particularly those of agronomic interest. Paradoxically, both nitric oxide (NO) and hydrogen sulfide (H2S), until recently were considered toxic since they are part of the polluting gases; however, at present, these molecules are part of the mechanism of response to multiple stresses since they exert signaling functions which usually have an associated stimulation of the enzymatic and non-enzymatic antioxidant systems. At present, these gasotransmitters are considered essential components of the defense against a wide range of environmental stresses including atmospheric ones. This review aims to provide an updated vision of the endogenous metabolism of NO and H2S in plant cells and to deepen how the exogenous application of these compounds can contribute to crop resilience, particularly, against atmospheric stressors stimulating antioxidant systems.

Agroindustrial By-Products as a Source of Biostimulants Enhancing Responses to Abiotic Stress of Horticultural Crops.

Together with other abiotic stresses such as drought and high temperatures, salt stress is one of the most deleterious environmental factors affecting plant development and productivity, causing significant crop yield reductions. The progressive secondary salinisation of irrigated farmland is a problem as old as agriculture but is aggravated and accelerated in the current climate change scenario. Plant biostimulants, developed commercially during the last decade, are now recognised as innovative, sustainable agronomic tools for improving crop growth, yield, plant health and tolerance to abiotic stress factors such as water and soil salinity. Biostimulants are a disparate collection of biological extracts, natural and synthetic organic compounds or mixtures of compounds, inorganic molecules and microorganisms, defined by the positive effects of their application to crops. The growing interest in biostimulants is reflected in the increasing number of scientific reports published on this topic in recent years. However, the processes triggered by the biostimulants and, therefore, their mechanisms of action remain elusive and represent an exciting research field. In this review, we will mainly focus on one specific group of biostimulants, protein hydrolysates, generally produced from agricultural wastes and agroindustrial by-products-contributing, therefore, to more sustainable use of resources and circular economy-and primarily on the consequences of their application on the abiotic stress resistance of horticultural crops. We will summarise data in the scientific literature describing the biostimulants' effects on basic, conserved mechanisms activated in response to elevated salinity and other abiotic stress conditions, such as the control of ion transport and ion homeostasis, the accumulation of osmolytes for osmotic adjustment, or the activation of enzymatic and non-enzymatic antioxidant systems to counteract the induced secondary oxidative stress. The collected information confirms the positive effects of biostimulants on crop tolerance to abiotic stress by enhancing morphological, physiological and biochemical responses, but also highlights that more work is needed to further establish the molecular mechanisms underlying biostimulants' effects.

Biodegradation of chloroxylenol by an aerobic enrichment consortium and a newly identified Rhodococcus strain.

Chloroxylenol is a commonly used antimicrobial agent in antibacterial and disinfection products, which has been detected in various environments, such as wastewater treatment plants, rivers, seawater, and even drinking water, with concentrations ranging from ng/L to mg/L. However, the biodegradation of chloroxylenol received limited attention with only sporadic reports available so far. In this study, an efficient chloroxylenol-degrading consortium, which could degrade 20 mg/L chloroxylenol within two days, was obtained after five months of enrichment. Amplicon sequencing analysis revealed a decrease in the α-diversity (e.g., Shannon index and Inv_Simpson index) of the community during the domestication process. Microbial community dynamics were uncovered, with sequences affiliated to Achromobacter, Pseudomonas, and Rhodococcus identified as the most abundant taxonomic groups. From the consortium, five pure isolates were obtained; however, it was found that only one strain of Rhodococcus could degradechloroxylenol. Strain Rhodococcus sp. DMU2021 could degrade chloroxylenol efficiently under the conditions of temperature 30-40 °C, and neutral/alkaline conditions. Chloroxylenol was toxic to strain DMU2021 and triggered both enzymatic and non-enzymatic antioxidant systems in response. This study provides novel insights into the biodegradation process of chloroxylenol, as well as valuable bioresources for bioremediation.

Possible Markers for Distinguishing benign and Malignant Thyroid Tumors and Predicting Malignancy in Patients with Genetic Predisposition to Cancer.

We hypothesized that mutations in several genes disrupt oxidative metabolism, increasing the risk of developing tumors and their malignancy in patients with a family predisposition to cancer. The purpose of our study was to assess the characteristics of oxidative metabolism in patients with malignant and benign tumor with and without a family history of cancer and identify the marker predicting the likelihood of malignancy. We conducted a study on patients with thyroid pathology (thyrotoxicosis, benign tumor pathology of the thyroid gland, and thyroid cancer) who underwent treatment at LLC "Oncology Scientific Research Center" in Tbilisi, Georgia between 2020-2021. In patients' blood the thyroid hormones content, the oxidative metabolism parameters (activity of nonenzymatic antioxidant system (TAA), malondialdehyde (MDA) content), geometrical and rheological (deformability index (EDI), membrane proteins content) characteristics of erythrocytes were determined. in the patient's blood serum with benign tumor (47 patients) MDA exceeded (p<0.005) and TAA decreased (p<0.005) in comparison to the control level; in patients with thyroid cancer (35 patients), MDA also exceeded (p<0.005), while TAA increased (p<0.005) up to the control level. In patients with benign and malignant tumors, the size of erythrocytes increased compared to the control indicators (p<0.005); in patients with thyroid cancer and benign tumors with a family history of cancer (29 patients) EDI increased (p<0.005), content of GLUT1 in erythrocyte membranes decreased (p<0.005) compared to the control level. Alterations in redox metabolism play a crucial role in tumor formation; an imbalance between anti-/pro-oxidant systems may contribute to tumor formation and support its progression into a more malignant state. Thyroid cancer is characterized by a reduction in erythrocyte deformability, related to TSH levels. These alterations are less detectable in patients with benign thyroid tumors with a family history of cancer.

Polycystic ovary syndrome and its management: In view of oxidative stress.

In the past two decades, oxidative stress (OS) has drawn a lot of interest due to the revelation that individuals with many persistent disorders including diabetes, polycystic ovarian syndrome (PCOS), cardiovascular, and other disorders often have aberrant oxidation statuses. OS has a close interplay with PCOS features such as insulin resistance, hyperandrogenism, and chronic inflammation; there is a belief that OS might contribute to the development of PCOS. PCOS is currently recognized as not only one of the most prevalent endocrine disorders but also a significant contributor to female infertility, affecting a considerable proportion of women globally. Therefore, the understanding of the relationship between OS and PCOS is crucial to the development of therapeutic and preventive strategies for PCOS. Moreover, the mechanistic study of intracellular reactive oxygen species/ reactive nitrogen species formation and its possible interaction with women's reproductive health is required, which includes complex enzymatic and non-enzymatic antioxidant systems. Apart from that, our current review includes possible regulation of the pathogenesis of OS. A change in lifestyle, including physical activity, various supplements that boost antioxidant levels, particularly vitamins, and the usage of medicinal herbs, is thought to be the best way to combat this occurrence of OS and improve the pathophysiologic conditions associated with PCOS.

Eugenol improves salt tolerance via enhancing antioxidant capacity and regulating ionic balance in tobacco seedlings.

Salt stress inhibits plant growth by disturbing plant intrinsic physiology. The application of exogenous plant growth regulators to improve the plant tolerance against salt stress has become one of the promising approaches to promote plant growth in saline environment. Eugenol (4-allyl-2- methoxyphenol) is the main ingredient in clove oil and it is known for its strong antioxidant and anti-microbial activities. Eugenol also has the ability of inhibiting several plant pathogens, implying the potential use of eugenol as an environmental friendly agrichemical. However, little is known about the possible role of eugenol in the regulation of plant tolerance against abiotic stress. Therefore, here we investigated the effectiveness of phytochemical eugenol in promoting salt tolerance in tobacco seedlings through physiological, histochemical, and biochemical method. The seedling roots were exposed to NaCl solution in the presence or absence of eugenol. Salt stress inhibited seedling growth, but eugenol supplementation effectively attenuated its effects in a dose-dependent manner, with an optimal effect at 20 µM. ROS (reactive oxygen species) accumulation was found in seedlings upon salt stress which was further resulted in the amelioration of lipid peroxidation, loss of membrane integrity, and cell death in salt-treated seedlings. Addition of eugenol highly suppressed ROS accumulation and reduced lipid peroxidation generation. Both enzymatic and non-enzymatic antioxidative systems were activated by eugenol treatment. AsA/DHA and GSH/GSSG were also enhanced upon eugenol treatment, which helped maintain redox homeostasis upon salinity. Eugenol treatment resulted in an increase in the content of osmoprotectants (e.g. proline, soluble sugar and starch) in salt-treated seedlings. Na+ levels decreased significantly in seedlings upon eugenol exposure. This may result from the upregulation of the expression of two ionic transporter genes, SOS1 (salt-hypersensitive 1) and NHX1 (Na+/H+ anti-transporter 1). Hierarchical cluster combined correlation analysis uncovered that eugenol induced salt tolerance was mediated by redox homeostasis and maintaining ionic balance in tobacco seedlings. This work reveals that eugenol plays a crucial role in regulating plant resistant physiology. This may extend its biological function as a novel biostimulant and opens up new possibilities for improving crop productivity in the saline agricultural environment.

Cold atmospheric pressure plasma as eco-friendly technology prolonged the vase life and improved the quality of cut rose flowers

Cold atmospheric pressure plasma is a relatively new technology characterized by safety and high efficiency, that is increasingly attracting attention in agriculture and food industries especially in post-harvest technology. This experiment addresses the potential use of plasma-activated water (PAW) for increment of cut rose flower longevity and quality. A complete randomized design was used in this study, and each treatment had three replicates with six flowers in each replicate. Based on the findings of this study, it seems that PAW application as a preservation solution improves plant water status during postharvest storage and reducing the electrolyte leakage and maintaining the relative water content (RWC). Data from the experiment showed that the plasma activated water decreased the malondialdehyde content and maintains the structure and integrity of the cell membrane through the activation of enzymatic and non-enzymatic antioxidant systems and phenolic compounds synthesis ultimately leading to postharvest quality and prolonged vase life of cut rose flowers. As PAW is more cost-effective than many other preservatives and is eco- friendly with fewer side effects, so it could be used in preservative solutions as an alternative.

Defense responses of the green microalgae Chlorella vulgaris to the vanadium pentoxide nanoparticles

Although vanadium-based nanomaterials have found extensive use in industry, their influence on ecosystems and living organisms is not yet well investigated. In this study, hydrothermal methods were utilized for the synthesis of vanadium pentoxide nanoparticles (V2 O5 NPs). The gained NPs were characterized using XRD, FT-IR, EDS, DLS, SEM and TEM techniques. Subsequently, the toxic effects of V2 O5 NPs on the model green microalgae Chlorella vulgaris were evaluated. According to the obtained results, V2 O5 NPs caused a significant reduction in cell number and biomass production of algae in a dose and time dependent manner. Moreover, flow cytometric analysis confirmed a reduction in the quantity of living cells. Scanning electron microscopy showed plasmolysis and deformation of the cells after exposure to nanoparticles. The photosynthetic pigments and phenolics content exhibited a decrease in comparison with the control sample. Although, non-enzymatic antioxidant system in C. vulgaris displayed an average action, antioxidant enzymes, including superoxide dismutase (SOD), catalase (CAT), and ascorbate peroxidase (APX) showed a dose dependent increasing trend. These intercellular reactions designated the activation of the antioxidant defense system in response to the induced oxidative stress by V2 O5 NPs.

Essential oil profiles, improvement of enzymatic and non-enzymatic antioxidant systems in different populations of Salvia subg. Perovskia by irrigation management

Salvia subg. Perovskia is an aromatic medicinal plant from the Lamiaceae family. Essential oil (EO) content and composition, along with enzymatic and non-enzymatic antioxidants, were evaluated in 18 plant populations under three levels of irrigation for two consecutive years. Based on the GC-MS analysis, the main components of EO were borneol (1.18–36.53%), followed by camphor (0.54–32.17%), 1,8-cineole (12.44–29.26%), δ-3-carene (0.39–21.20%), myrcene (0.59–16.28%), and α- pinene (0.79–12.87%) in the studied treatment. Except for malondialdehyde and hydrogen peroxide in leaves and roots, all the examined parameters showed lower values in the first harvest year compared to those of the second year. The activities of antioxidant enzymes, total phenolics, and flavonoids of the extracts were substantially enhanced as stress intensified to reach maximum values in leaves under severe stress and in roots under moderate stress conditions. The maximum root tanshinones content was observed under moderate water deficit conditions, while the highest EO content was obtained in plants exposed to severe stress conditions. Taking into account the evidence provided by this study, it can be inferred that the use of water deficit stress can serve as an effective method to stimulate and improve antioxidant properties, as well as the quantity and quality of secondary metabolites in Salvia subg. Perovskia.

Regulation of Na+/H+ antiport system and nitrogen metabolism by melatonin and endogenous hydrogen sulfide confers resilience to drought and salt stress

The present study was conducted with the purpose of examining the role of endogenous hydrogen sulfide (H2S) in melatonin (Mel)-induced regulation of tolerance mechanisms to drought stress and salinity. The drought stress was induced by maintaining soil water level at 40% and salinity was induced by applying 100 mM sodium chloride (NaCl). The results demonstrate an augmentation in the degree of activation of the antioxidant defense mechanisms in response to drought stress and salinity. However, a significant decline in the K+/Na+ ratio, accompanied by an elevation in reactive oxygen species, lipid peroxidation, and electrolyte leakage, was witnessed as well. A remarkable decrease in the level of hydration and content of chlorophyll was also observed under the imposed stress. The reported findings indicate that the administration of 30 µM Mel effectively regulated the activity of the Na+-H+ antiporter and H+-ATPase. This regulation ultimately led to an increase in K+ influx and a decline in Na+ efflux, resulting in an enhanced K+/Na+ ratio. Application of Mel also demonstrated an improvement in nitrogen metabolism, as well as the enzymatic and non-enzymatic antioxidant defense system. Additionally, Mel application caused a remarkable increase in the accumulation of osmolytes in drought stress and salinity -suffered plants. The synergistic effect of these characteristics resulted in the resilience of the Mel-supplemented plants to drought stress and salinity. However, application of hypotaurine (a scavenger of H2S) counteracted the beneficial impact of Mel which once again resulted in the same condition that was initially created by the imposed stress. These findings imply that Mel, in coordination with H2S was remarkable in inducing plant adaptive responses to drought stress and salinity.

Oxidative Stress in Beta-thalassemia Patients: Role of Enzymatic and Non-enzymatic Modulators.

Oxidative stress is a pathophysiological state that arises due to an imbalance created between ROS generation and the antioxidant potential of the host cell. Transfusion- dependent beta-thalassemia major patients are at high risk of cellular and molecular damages induced by ROS mainly due to iron overload caused by repetitive blood transfusion. To assess polymorphism in VDR (rs2228570) and NOS2 (rs944725) in β-thalassemia patients. To analyze serum vitamin D levels of β-TM patients compared to healthy individuals. The present case-control study aimed to identify Vitamin D levels in the serum of β-thalassemia patients and compared it with healthy subjects. The study further analyzed VDR FOKI (rs2228570) polymorphism through ARMS-PCR. Expression profiling of VDR, anti-oxidant enzyme (SOD2 and GPx), and their respective regulator (HuR and NrF2) transcripts was done by the 2-ΔΔCt method. The study reports that there is no a significant difference between the Vitamin D levels among healthy and patients. VDR polymorphism analysis (rs2228570) demonstrates that although the C allele is prevalent in the study cohort, the frequency of the T allele is comparatively higher in β-thalassemia patients as compared to healthy subjects. Furthermore, patients express lower levels of anti-oxidant enzymes despite having increased oxidative stress. The study reports that β-thalassemia patients are at higher risk of cellular and molecular damages induced by oxidative stress and their associated pathologies inefficient enzymatic and non-enzymatic anti-oxidant defense systems.

Postharvest Storage Differentially Modulates the Enzymatic and Non-Enzymatic Antioxidant System of the Exocarp and Mesocarp of Hass Avocado: Implications for Disorders

The present study evaluated the performance of some enzymatic and non-enzymatic antioxidant systems against oxidative stress for 10 to 30 d of refrigeration (R) and 15 to 50 d in controlled atmosphere (CA) conditions in both exocarp and mesocarp of Hass avocados from early and late harvests and at shelf life (SL) or consumption maturity. The possible relationship of the antioxidant systems with the occurrence of physiological disorders is also evaluated. The results indicate that the enzymatic system—superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), phenylalanine ammonium lyase (PAL) and polyphenoloxidase (PPO)—as well as the non-enzymatic system—such as phenolic compounds (PC)—showed different responses to the stress generated during storage and shelf life. In general, SOD, CAT, PAL and PPO did not significantly vary in storage (R or CA). At consumption maturity, SOD, POD and PAL activities decreased in the mesocarp (RSL and CASL), while CAT increased in the exocarp for CASL15-50d. PC instead decreased in the exocarp as the harvest period progressed while it increased in the mesocarp. Physiological disorders (dark spots) showed only in refrigeration on the exocarp at R30d and in mesocarp at RSL30d coincident with low SOD and low SOD and POD activity values, as well as low PC contents (p-coumaric and its derivatives and caffeic acid derivatives), respectively. The results support the use of CA as a postharvest technology to prevent the development of physiological disorders through the joint action of antioxidative defenses during avocado transport to distant markets until consumption maturity is reached.

Changes of the antioxidant system in pear (Pyrus communis L.) fruits by foliar application of copper, selenium, iron, and zinc nanoparticles

Innovations in agricultural nanotechnology are a viable option for improving the antioxidant system in the pear crop helping to mitigate the different types of abiotic and biotic stresses whose effects have increased due to the advance of climate change. Therefore, foliar applications were made with nanoparticles (NPs) of copper (Cu), iron (Fe), and zinc (Zn) at 250 ppm, as well as applications with selenium (Se) at 50 ppm, and control with water in a commercial pear orchard (Bartlett variety), to evaluate its effect on the behavior of the enzymatic antioxidant system, the concentration of bioactive compounds, and the evaluation of damage by lipid peroxidation, hydrogen peroxide concentration, and activity of polyphenol oxidases (PPO) in the fruits. The results indicate that the Cu and Se NPs stimulated in the fruits a greater synthesis of total phenols, flavonoids, and ascorbic acid, achieving higher concentrations than the control fruits, and also exhibited greater antioxidant activity in vitro. An opposite effect was observed in the fruits of the treatment with Zn NPs, since the concentration of phenols and flavonoids was reduced by 33.14 and 34.24 %, respectively, as well as a decrease in antioxidant activity. On the other hand, the Cu and Fe NPs improved the activity of the enzymes superoxide dismutase, catalase, ascorbate peroxidase, and guaiacol peroxidase, additionally, the integrity of the membranes was maintained, thus avoiding enzymatic browning due to the activity of the PPOs in the fruits. The highest concentration of malondialdehyde and PPOs activity was observed in the fruits of the treatment with Se and Zn NPs. The results indicate that the application of Cu NPs at 250 ppm can improve the enzymatic and non-enzymatic antioxidant system in Bartlett cultivar pear fruits.

Effect of Nitric Oxide on Browning of Stem Tip Explants of Malus sieversii

Browning is a major problem in the tissue culture of woody plants. Previous studies have shown that nitric oxide (NO) plays a role in regulating plant responses to stress, but its effect on browning in the tissue culture of Malus remains unclear. This study aimed to investigate the impact of exogenous NO donor sodium nitroprusside (SNP) on the browning of Malus sieversii stem tip explants. The results demonstrated that the addition of 50 μM SNP significantly reduced explant browning. Further analysis revealed that exogenous NO decreased the browning index (BD) and levels of malondialdehyde (MDA), hydrogen peroxide (H2O2), and superoxide anion radical O2−. Additionally, NO treatment increased the activity of antioxidant enzymes such as superoxide dismutase (SOD), catalase (CAT), and ascorbate peroxidase (APX). NO treatment also enhanced the activity of phenylalanine ammonia lyase (PAL), which contributed to the accumulation of flavonoids and inhibited the activity of polyphenol oxidase (PPO) and peroxidase (POD), which are key enzymes involved in the browning process. Furthermore, 50 μM SNP significantly promoted the accumulation of non-enzymatic antioxidants such as proline (Pro), soluble sugar (SS), and soluble protein (SP). Therefore, the results suggest that NO is able to counteract excessive reactive oxygen species (ROS) damage by enhancing both the enzymatic and non-enzymatic antioxidant systems, resulting in a reduction in browning in stem tip explants. Consequently, an improvement in the in vitro propagation efficiency of Malus sieversii shoot tip explants can be achieved.
