Comparative evaluation of the chiral recognition potential of single-isomer sulfated beta-cyclodextrin synthesis intermediates in non-aqueous capillary electrophoresis

The enantioselectivity of neutral single-isomer synthetic precursors of sulfated-β-cyclodextrins was studied. Four neutral single-isomer cyclodextrins substituted on the secondary side with acetyl and/or methyl functional groups, heptakis(2-O-methyl-3,6-dihydroxy)-β-cyclodextrin (HM-β-CD), heptakis(2,3-di-O-acetyl-6-hydroxy)-β-cyclodextrin (HDA-β-CD), heptakis(2,3-di-O-methyl-6-hydroxy)-β-cyclodextrin (HDM-β-CD), heptakis(2-O-methyl-3-O-acetyl-6-hydroxy)-β-cyclodextrin (HMA-β-CD), and their sulfated analogs the negatively charged heptakis(2,3-di-O-methyl-6-sulfato)-β-cyclodextrin (HDMS-β-CD) and heptakis(2,3-di-O-acetyl-6-sulfato)-β-cyclodextrin (HDAS-β-CD) were investigated by non-aqueous capillary electrophoresis in the view of enantiodiscrimination for various drugs and related pharmaceutical compounds. The focus of the present work was on the chiral selectivity studies of the neutral derivatives, which are the synthesis intermediates of the sulfated products. The chiral recognition experiments proved that among the neutral compounds the HMA-β-CD shows remarkable enantioselectivity towards chiral guests in non-aqueous capillary electrophoresis, while HM-β-CD, HDA-β-CD and HDM-β-CD failed to resolve any of the 25 studied racemates under the applied experimental conditions. In order to get deeper insight into the molecular interactions between the studied single-isomer cyclodextrin and chiral fluoroquinolones (ofloxacin, gatifloxacin and lomefloxacin) and β-blockers (propranolol), 1H and ROESY NMR experiments were performed. The 2-O-methylation in combination with the 3-O-acetylation of the host was evidenced to exclusively carry the essential spatial arrangement for chiral recognition.

Determination of tamoxifen and its metabolites using micelle to solvent stacking in nonaqueous capillary electrophoresis.

Micelle to solvent stacking was implemented for the recently established NACE-C(4) D method to determine tamoxifen and its metabolites in standard samples and human plasma of breast cancer patients. For stacking, the standard samples and extract after liquid-liquid extraction (LLE) were prepared in methanol and the resulting sample solution was pressure injected after a micellar plug of SDS. Factors that affected the stacking such as SDS concentration, micelle, and sample plug length were examined. The sensitivity enhancement factor (peak height from stacking/peak height from typical injection of sample in BGE) was 15-22. The method detection limits with LLE were in the range of 5-10 ng/mL, which was lower than the established method (where the LLE extract was also prepared in methanol) with reported method detection limits of 25-40 ng/mL. The intraday and interday repeatability were in the range of 1.0-3.4% and 3.8-6.5%, respectively.

Determination of Curcuminoids and Their Degradation Products in Turmeric (Curcuma longa) Rhizome Herbal Products by Non-aqueous Capillary Electrophoresis with Photodiode Array Detection

Non-aqueous capillary electrophoresis (NACE) method was optimised for simultaneous determination of major bioactive curcuminoids (CCD) viz., curcumin (CC), desmethoxycurcumin (DC) and bisdesmethoxycurcumin (BC), and some of the degradation products viz., vanillin (VN), vanillic acid (VA), ferulic acid (FA) and 4-hydroxybenzaldehyde (HB) in turmeric milk and herbal products. Separation was achieved using non-aqueous background electrolyte (NA-BGE), consisting of a mixture of sodium tetraborate, sodium hydroxide, methanol and 1-propanol in a single run by capillary electrophoresis (CE). In the present study, a novel ultrasonication-assisted phase separation method (US-PS) was used for extracting the analytes in turmeric milk and the extract was directly injected into the capillary without any pretreatment. High-performance thin layer chromatographic (HPTLC) method for the separation of the analytes was also evaluated on pre-coated silica gel plate. Two different high-performance liquid chromatographic (HPLC) conditions were reported in the literature for the separation of above-mentioned target compounds. The proposed method NACE is simple, fast, convenient and economical.

Highly sensitive wavelength-dependent nonaqueous capillary electrophoresis for simultaneous screening of various synthetic organic dyes

A novel multi-wavelength nonaqueous capillary electrophoresis (MW-NACE) technique based on wavelength-dependent laser-induced fluorescence (LIF) detection was investigated for the simultaneous screening of various synthetic organic dyes. Multi-wavelength excitation light sources were utilized to excite different organic dyes [e.g., 543nm for crystal violet (CV), methyl violet B (MVB), methyl violet B base (MBB), rhodamine 6G (R6G), and rhodamine B base (RBB); 635nm for nile blue A (NBA) and methylene blue (MB)] simultaneously. Using a nonaqueous buffer system composed of 15mM sodium borate and 835mM acetic acid in 100% ethanol (pH=5.4), all dyes were analyzed within 15min with excellent resolution (R≥4.0) under an electric field of 500V/cm. Calibration curves showed excellent linearity with square of correlation coefficients (r2) greater than 0.9908 over wide dynamic ranges of 0.4–50μM for CV, 0.8–50μM for MVB, 1.5–50μM for MBB, 0.08–5nM for R6G, 0.06–10μM for MB, 0.02–10μM for NBA, and 0.13–10 pM for RBB. The detection limits (S/N=3) of 40fM to 0.5μM were 10–200,000 times lower than those of previous detection methods. While adjacent peaks were not well distinguished with baseline separation in a single capillary, the devised technique was faster and more sensitive than conventional aqueous and nonaqueous CE approaches, thereby enabling the quantitative analysis of various dyes based on wavelength-dependent fluorescence detection with different excitation wavelengths.

Large-Volume Sample Staking of Rice Polyphenols Prior to Their Determination by Non-aqueous Capillary Electrophoresis

A rapid non-aqueous capillary electrophoretic (NACE) method for the separation of (−)-epicatechin (EPI), (+)-catechin (−CAT), kaempferol (KAE), quercetin (QUR), naringanin (NAR), ferulic acid (FA), and p-coumaric acid (p-CA) has been developed and applied to the determination of these compounds in different rice varieties. All seven compounds were separated on capillary of 50 μm × 68 cm (60-cm effective length) using 20 mmol L−1 borate buffer of pH 9.0 and 5 % acetonitrile in methanol. Large-volume sample stacking (LVSS) technique was optimized and used to preconcentrate non-detectable polyphenols of white polished rice. Rice extracts were concentrated on-line by LVSS prior to separation by non-aqueous capillary electrophoresis. An improvement of 10–55 times in detectability was achieved with injection at 50 mbar for 30 s followed by voltage inversion (−20 kV) for 5 s. Linear calibration range of 1–300 μg L−1 and 0.01–60 μg L−1 was observed for NACE and NACE-LVSS method respectively, with the detection limit of 0.33–2.0 and 0.006–0.19 μg L−1. Good reproducibility with standard deviations of less than 5 % was achieved. Polyphenol contents of different rice varieties were determined using developed method.

Nonaqueous Capillary Electrophoresis Mass Spectrometry.

The term nonaqueous capillary electrophoresis (NACE) commonly refers to capillary electrophoresis with purely nonaqueous background electrolytes (BGE). Main advantages of NACE are the possibility to analyze substances with very low solubility in aqueous media as well as separation selectivity that can be quite different in organic solvents (compared to water)-a property that can be employed for manipulation of separation selectivities. Mass spectrometry (MS) has become more and more popular as a detector in CE a fact that applies also for NACE. In the present chapter, the development of NACE-MS since 2004 is reviewed. Relevant parameters like composition of BGE and its influence on separation and detection in NACE as well as sheath liquid for NACE-MS are discussed. Finally, an overview of the papers published in the field of NACE-MS between 2004 and 2014 is given. Applications are grouped according to the field (analysis of natural products, biomedical analysis, food analysis, analysis of industrial products, and fundamental investigations).

Chiral separation by nonaqueous capillary electrophoresis using l-sorbose–boric acid complexes as chiral ion-pair selectors

A chiral nonaqueous capillary electrophoresis (NACE) method using l-sorbose–boric acid complexes as the chiral ion-pair selectors was developed for enantioseparation of nineteen chiral analytes.

Ionic Liquids in Capillary Electrophoresis.

Recently, a great interest was drawn toward ionic liquids (ILs) in analytical separation techniques. ILs possess many properties making them excellent additives in capillary electrophoresis (CE) background electrolytes (BGE). The most important property is the charge of the dissolved ions in BGE enabling the cations to interact with deprotonated silanol groups on the capillary surface and thereby modifying the electroosmotic flow (EOF). Ionic and/or proton donor-acceptor interactions between analyte and IL are possible interactions facilitating new kinds of separation mechanisms in CE. Further advantages of ILs are the high conductivity, the environmentally friendliness, and the good solubility for organic and inorganic compounds. The most commonly used ILs in capillary electrophoresis are dialkylimidazolium-based ILs, whereas for enantioseparation a lot of innovative chiral cations and anions were investigated.ILs are reported to be additives to a normal CE background electrolyte or the sole electrolyte in CE, nonaqueous CE (NACE), micellar electrokinetic chromatography (MEKC), and in enantioseparation. An overview of applications and separation mechanisms reported in the literature is given here, in addition to the enantioseparation of pseudoephedrine using tetrabutylammonium chloride (TBAC) as IL additive to an ammonium formate buffer containing β-cyclodextrin (β-CD).

Determination of tamoxifen and its metabolites in human plasma by nonaqueous capillary electrophoresis with contactless conductivity detection.

A new approach for the quantification of tamoxifen and its metabolites 4-hydroxytamoxifen, N-desmethyltamoxifen, and 4-hydroxy-N-desmethyltamoxifen (endoxifen) in human plasma samples using NACE coupled with contactless conductivity detection (C4 D) is presented. The buffer system employed consisted of 7.5 mM deoxycholic acid sodium salt, 15 mM acetic acid, and 1 mM 18-crown-6 in 100% methanol. The complete separation of all targeted compounds (including endoxifen racemate) could be achieved within 6 min under optimized conditions. The proposed method was validated and showed good linearity in the range from 100 to 5000 ng/mL with correlation coefficients between 0.9922 and 0.9973, LODs in the range of 25-40 ng/mL, and acceptable reproducibility of the peak area (intraday RSD 2.2-3.1%, n = 4; interday (3 days) RSD 6.0-8.8%, n = 4). The developed method was successfully demonstrated for the quantification of tamoxifen and its metabolites in human plasma samples collected from breast cancer patients undertaking tamoxifen treatment.

Quantitation of sunitinib, an oral multitarget tyrosine kinase inhibitor, and its metabolite in urine samples by nonaqueous capillary electrophoresis time of flight mass spectrometry

A rapid, sensitive, and specific method was developed and validated using a nonaqueous-capillary electrophoresis method with TOF-MS for determination of sunitinib and N-desethyl sunitinib in human urine. In order to avoid ionic suppression a urine samples dilution with methanol 1:10 previous step was used. This was the only treatment step to urine samples before the injection. Despite this dilution of the urine, the detection limit was as low as 0.07 mg/L for sunitinib and 0.15 mg/L for N-desethyl sunitinib. Separation of compounds was achieved with a mixture of 5 mM ammonium formate in methanol. The calibration curves were linear over the range of 0.5-50.0 mg/L for the two analyzed compounds. The within-run and between-run precisions were within 5%, while the accuracy ranged from 96.0 to 100.4%. This method can be used in routine clinical practice to monitor sunitinib and N-desethyl sunitinib drugs in the urine of cancer patients treated with once daily administration.

Separation of linear synthetic polymers in non-aqueous capillary zone electrophoresis using cationic surfactant

A method for separating water-insoluble and neutral synthetic polymers using non-aqueous capillary zone electrophoresis (NACZE) was developed. The non-aqueous solvent system comprising a mixture of tetrahydrofuran, acetonitrile, and ethanol containing cetyltrimethylammonium chloride was used for solubilizing and conferring positive charges to the polymers. A mixture of polystyrene (PS, Mn=6500) and polybutadiene (PBD, Mn=5900) was successfully separated by the NACZE method using cationic surfactants. Evaluation of the effect of the molecular weight of the polymers on the electrophoretic behavior demonstrated that PSs with different molecular weights (Mn=6500, 10,200, 19,600, 200,000) were co-eluted as a single peak. That is, the apparent electrophoretic mobility of the PSs was independent of the molecular weight. In contrast, evaluation of PBD and polycarbonate (PC) demonstrated that the solubility of polymers in the medium affected the apparent electrophoretic mobility of the polymers, where low solubility resulted in reduced apparent electrophoretic mobility. Using the proposed method, poly(styrene-co-methylmethacrylate)s with different compositions were successfully separated.

Determination of gambogic acid in Gamboge by non-aqueous capillary electrophoresis

Gambogic acid (GA), a kind of caged xanthones, has low solubility in water. A non-aqueous capillary electrophoresis (NACE) was established for the determination of GA in Gamboge based on the optimized conditions. The effect of 20% - 60% methanol or acetonitrile spiked in running solution was investigated. The effects of compositions, concentration, pH, additives like β-cyclodextrin in running buffer were thoroughly studied. Applied voltage and applied temperature were also observed. Optimal electrophoretic conditions were as follows: 20 mmol/L sodium borohydride solution (pH 9. 86) containing 40% (v/v) acetonitrile, 10 mmol/L β-cyclodextrin as running buffer, applied voltage of 10 kV, capillary temperature of 30 °C and detection wavelength of 280 nm. The calibration curve had good linearity in the range of 2-2 000 mg/L with the correlation coefficient of 0. 999 6. The limit of quantification (S/N= 3) of the method was 2 mg/L. The quantifications of GA in Gamboge from different producing places including Vietnam, Thailand, Burma, India were 1. 67-472.40 mg/g with the RSD (n= 3) of 1.12% -2.60%. The content of Gamboge from Vietnam is obviously low while the others are high. The recoveries of GA spiked in real samples ranged from 95. 2% to 105. 6%. The method of NACE is simple, efficient and of good reproducibility, can be served as a novel reference to identify and control the quality of Gamboge.

Determination of enantiomers by FESI-sweeping with an acid-labile sweeper in nonaqueous capillary electrophoresis.

In this work, a facile and highly efficient on-line concentration strategy based on a coupling of field enhanced sample injection (FESI) and sweeping was developed for the determination of trace enantiomers (propranolol, PL) by nonaqueous capillary electrophoresis (NACE). In this FESI-sweeping method, the use of a sample of high acidity and low conductivity (pH* = 2.5, 4.0 μS cm(-1)) allowed for a large amount of analyte injection. Then, the concentration of the analytes was carried out by sweeping based on the interaction of an acid-labile anionic selector, di-n-butyl L-tartrate-boric acid complex acid, and cationic analytes. Simultaneously, the concentrated analytes were released and focused at the boundary of the acid sample solution and separation buffer due to the decomposition of the selector in the acid sample solution. Under the optimum conditions, a 21,000-fold sensitivity enhancement upon normal capillary zone electrophoresis (CZE) was achieved for PL enantiomers. The detection limits of R-propranolol and S-propranolol were 0.26 ng mL(-1) and 0.31 ng mL(-1), respectively. Eventually, the FESI-sweeping method was applied to detect PL enantiomers in plasma, saliva, and urine.

Separation and analysis of triazine herbcide residues by capillary electrophoresis

Triazines are widely used in agriculture around the world as selective pre- and post-emergence herbicides for the control of broad leaf and grassy weeds. With high toxicity and persistence, triazines can contaminate the environment and crops, so the development of rapid and sensitive methods for the determination of different triazines is necessary. Capillary electrophoresis comprises a group of techniques used to separate chemical mixtures. Analytical separation is based on different electrophoretic mobilities. This review focuses on the analysis of triazine herbicides with different modes of capillary electrophoresis, including capillary zone electrophoresis, micellar electrokinetic capillary electrophoresis, capillary electrochromatography and nonaqueous capillary electrophoresis. Determinations of triazines in various matrices such as surface water, groundwater, vegetables, soil and grains are emphasized.

A simple method for patterning poly(dimethylsiloxane) barriers in paper using contact-printing with low-cost rubber stamps

This paper presents a simple and low-cost method for patterning poly(dimethylsiloxane) (PDMS) barriers in porous support such as paper for the construction of flexible microfluidic paper-based analytical devices (μPADs). The fabrication method consisted of contact-printing a solution of PDMS and hexane (10:1.5 w/w) onto chromatographic paper using custom-designed rubber stamps containing the patterns of μPADs. After penetrating the paper (∼30s), the PDMS is cured to form hydrophobic barriers. Under optimized conditions, hydrophobic barriers and hydrophilic channels with dimensions down to 949±88μm and 771±90μm (n=5), respectively, were obtained. This resolution is well-suitable for most applications in analytical chemistry. Chemical compatibility studies revealed that the PDMS barriers were able to contain some organic solvents, including acetonitrile and methanol, and aqueous solutions of some surfactants. This find is particularly interesting given that acetonitrile and methanol are the most used solvents in chromatographic separations, non-aqueous capillary electrophoresis and electroanalysis, as well as aqueous solutions of surfactants are suitable mediums for cell lyses assays. The utility of the technique was evaluated in the fabrication of paper-based electrochemical devices (PEDs) with pencil-drawn electrodes for experiments in static cyclic voltammetry and flow injection analysis (FIA) with amperometric detection, in both aqueous and organic mediums.

Analysis of aromatic acids by nonaqueous capillary electrophoresis with ionic-liquid electrolytes.

The separation of six kinds of aromatic acids by CZE with 1-ethyl-3-methylimidazolium chloride (EMIMCl) and 1-ethyl-3-methylimidazolium hydrogen sulfate (EMIMHSO4 ), two kinds of ionic liquids (ILs) as background electrolytes, and acetonitrile as solvent were investigated. The six kinds of aromatic acids can be separated under positive voltage with low IL concentration with either of the two ILs and separation with EMIMHSO4 is better in consideration of peak shapes and separation efficiency. But the migration order is different when the IL is different. Under negative voltage with high IL concentration, the six analytes can be separated with EMIMCl as background electrolytes and the migration order of the analytes is opposite to those with low concentration of EMIMCl as background electrolyte. The separations are based on the combination effects of heteroconjugation between the anions and cations in the ILs and the analytes, of which the heteroconjugation between the anions in the ILs and the analytes plays a dominant role. The heteroconjugation between the anions of the ILs and analytes is proton sensitive and only a very small amount of proticsolvents added into the electrolyte solution can harm the separation. When EMIMCl concentration is high, the heteroconjugation between the IL anions and the proton in the analytes make the effective mobility of the analytes much higher than the EOF and their migration direction reversed. Finally, the six aromatic acids in water samples were analyzed by nonaqueous CE with low concentration of EMIMHSO4 as background electrolytes with satisfactory results.

20 years of fatty acid analysis by capillary electrophoresis.

A review taking into account the literature reports covering 20 years of fatty acid analysis by capillary electrophoresis is presented. This paper describes the evolution of fatty acid analysis using different CE modes such as capillary zone electrophoresis, non-aqueous capillary electrophoresis, micellar electrokinetic capillary chromatography and microemulsion electrokinetic chromatography employing different detection systems, such as ultraviolet-visible, capacitively coupled contactless conductivity, laser-induced fluorescence and mass spectrometry. In summary, the present review signals that CE seems to be an interesting analytical separation technique that is very useful for screening analysis or quantification of the usual fatty acids present in different matrices, offering short analysis times and a simple sample preparation step as inherent advantages in comparison with the classical methodology, making it a separation technique that is very attractive for quality control in industry and government agencies.

Nonaqueous capillary electrophoresis of dextromethorphan and its metabolites.

This study deals with the nonaqueous capillary electrophoretic separation of dextromethorphan and its metabolites using a methanolic background electrolyte. The optimization of separation conditions was performed in terms of the resolution of dextromethorphan and dextrorphan and the effect of separation temperature, voltage, and the characteristics of the background electrolyte were studied. Complete separation of all analytes was achieved in 40 mM ammonium acetate dissolved in methanol. Hydrodynamic injection was performed at 3 kPa for 4 s. The separation voltage was 20 kV accompanied by a low electric current. The ultraviolet detection was performed at 214 nm, the temperature of the capillary was 25°C. These conditions enabled the separation of four analytes plus the internal standard within 9 min. Further, the developed method was validated in terms of linearity, sensitivity, and repeatability. Rat liver perfusate samples were subjected to the nonaqueous capillary electrophoretic method to illustrate its applicability.
