Single Molecule Dynamics on Hydrophobic Self-Assembled Monolayers

The interactions between adsorbate molecules and hydrophobic surfaces are of significant interest due to their importance in a variety of biological and separation processes. However, it is challenging to extrapolate macroscopic ensemble-averaged force measurements to molecular-level phenomena. Using total internal reflection fluorescence microscopy to image individual molecules at hydrophobic solid-aqueous interfaces, we directly observed dynamic behavior associated with the interactions between fluorescently labeled dodecanoic acid (our probe molecules) and self-assembled monolayers (SAM) comprising n-alkyltriethoxysilanes with systematically increasing chain length (from n = 4-18). In all cases, we observed at least two characteristic surface residence times and two diffusive modes, suggesting the presence of multiple distinct adsorbed populations. In general, the mean surface residence time increased and the mobility decreased with increasing SAM chain length, consistent with stronger probe-surface interactions. However, these trends were not primarily due to changes in characteristic residence times or diffusion coefficients associated with the individual populations but rather to a dramatic increase in the fraction associated with the long-lived slow-moving population(s) on long-chain SAMs. In particular, on longer (16-18 carbon) alkylsilane monolayers, the probe molecule exhibited far fewer desorption-mediated "flights" than on short (4-6 carbon) monolayers. Additionally, probes on the longer chain surfaces were much more likely to exhibit extended surface residence times as opposed to short transient surface visits.

Characterizing the Molecular Order of Phosphonic Acid Self-Assembled Monolayers on Indium Tin Oxide Surfaces

Self-assembled monolayers (SAMs) of alkanephosphonic acids with chain lengths between 8 and 18 carbon units were formed on thin films of indium tin oxide (ITO) sputter-deposited on silicon substrates with 400 nm thermally grown SiO(2). The silicon substrates, while not intended for use in near-IR or visible optics applications, do provide smooth surfaces that permit systematic engineering of grain size and surface roughness as a function of the sputter pressure. Argon sputter pressures from 4 to 20 mTorr show systematic changes in surface morphology ranging from smooth, micrometer-sized grain structures to <50 nm grains with 3× higher surface roughness. Near-edge X-ray absorption fine structure (NEXAFS) spectroscopy experiments are conducted for alkanephosphonic acids deposited on these wide range of ITO surfaces to evaluate the effects of these morphological features on monolayer ordering. Results indicate that long-chain SAMs are more highly ordered, and have a smaller tilt angle, than short-chain SAMs. Surprisingly, the 1-octadecyl phosphonic acids maintain their order as the lateral grain dimensions of the ITO surface shrink to ∼50 nm. It is only when the ITO surface roughness becomes greater than the SAM chain length (∼15 Å) that SAMs are observed to become relatively disordered.

Packing density and structure effects on energy-transfer dynamics in argon collisions with organic monolayers

A combined experimental and molecular-dynamics simulation study has been used to investigate energy-transfer dynamics of argon atoms when they collide with n-alkanethiols adsorbed to gold and silver substrates. These surfaces provide the opportunity to explore how surface structure and packing density of alkane chains affect energy transfer in gas-surface collisions while maintaining the chemical nature of the surface. The chains pack standing up with 12 degrees and 30 degrees tilt angles relative to the surface normal and number densities of 18.9 and 21.5 A(2)molecule on the silver and gold substrates, respectively. For 7-kJmol argon scattering, the two surfaces behave equivalently, fully thermalizing all impinging argon atoms. In contrast, these self-assembled monolayers (SAMs) are not equally efficient at absorbing the excess translational energy from high-energy, 35 and 80 kJmol, argon collisions. When high-energy argon atoms are scattered from a SAM on silver, the fraction of atoms that reach thermal equilibrium with the surface and the average energy transferred to the surface are lower than for analogous SAMs on gold. In the case of argon atoms with 80 kJmol of translational energy scattering from long-chain SAMs, 60% and 45% of the atoms detected have reached thermal equilibrium with the monolayers on gold and silver surfaces, respectively. The differences in the scattering characteristics are attributed to excitation efficiencies of different types of surface modes. The high packing density of alkyl chains on silver restricts certain low-energy degrees of freedom from absorbing energy as efficiently as the lower-density monolayers. In addition, molecular-dynamics simulations reveal that the extent to which argon penetrates into the monolayer is related to packing density. For argon atoms with 80-kJmol incident energy, we find 16% and 7% of the atoms penetrate below the terminal methyl groups of C(10) SAMs on gold and silver, respectively.

Grafting of single, stimuli-responsive poly(ferrocenylsilane) polymer chains to gold surfaces.

Redox-responsive poly(ferrocenylsilane) (PFS) polymer molecules were attached individually to gold surfaces for force spectroscopy experiments on the single molecule level. By grafting ethylenesulfide-functionalized PFS into the defects of preformed self-assembled monolayers (SAMs) of different omega-mercaptoalkanols on Au(111), the surface coverage of PFS macromolecules could be conveniently controlled. Atomic force microscopy (AFM), contact angle, as well as cyclic and differential pulse voltammetry measurements were carried out to characterize the morphology, wettability, and surface coverage of the grafted layers. The values of the PFS surface coverage were found to depend on the chain length of the omega-mercaptoalkanol molecules and on the concentration of the PFS solution but not on the insertion time or on the molar mass of PFS. The equilibrium surface coverages were successfully described by Langmuir adsorption isotherms. For low-surface coverage values (< 6.2 x 10(-4) chain/nm2), achieved by PFS insertion from very dilute solutions (8 x 10(-6) M) into long-chain SAMs, AFM and differential pulse voltammetry showed that surfaces exposing isolated individual polymer chains were obtained. The isolated PFS macromolecules were subjected to in situ AFM-based single molecule force spectroscopy (SMFS) measurements. The single chain elasticity of PFS in isopropanol (and ethanol) was fitted with the modified freely jointed chain (m-FJC) model. This procedure yielded a Kuhn segment length of 0.33 +/- 0.05 nm and a segment elasticity of 32 +/- 5 nN/nm.

Friction Force Microscopy of Self-Assembled Monolayers: Influence of Adsorbate Alkyl Chain Length, Terminal Group Chemistry, and Scan Velocity

Friction force measurements have been made using friction force microscopy for a series of self-assembled monolayers (SAMs) of alkanethiols of varying chain lengths and terminal groups adsorbed on gold. The chemistry of the tip was controlled by the deposition of a SAM. When carboxylic acid terminated tips were employed, the friction coefficients of hydroxyl and carboxylic acid terminated SAMs were found to be greater than those of methyl terminated SAMs. However, although the friction coefficients of short-chain methyl terminated SAMs were significantly greater than those of long-chain methyl terminated SAMs, there was not a significant difference between the values determined for short- and long-chain SAMs with polar terminal groups. When methyl terminated tips were employed, there was no difference between the behavior observed for hydroxyl and carboxylic acid terminated SAMs of equal chain length, but methyl terminated SAMs exhibited increased friction coefficients which were larger for short-chain adsorbates. Friction−velocity plots exhibited markedly different behavior for polar and nonpolar SAMs when carboxylic acid terminated tips were employed. These observations are explained in terms of the stabilization of the adsorbates by intermolecular hydrogen bonding in SAMs with polar terminal groups.
