Field Detection of Uranyl in Coastal Water of China Using a Portable Device via DNA Photocleavage.

The urgent need for field detection of uranium in seawater is 2-fold: to provide prompt guidance for uranium extraction and to prevent human exposure to nuclear radiation. However, current methods for this purpose are largely hindered by bulky instrumentation, high costs of developed materials, and severe matrix interferences, which limit their further application in the field. Herein, we demonstrated a portable and label-free strategy for the field detection of uranyl in seawater based on the efficient photocleavage of DNA. Further experiments confirmed the generation of ultraviolet (UV) light-induced reactive oxygen species (ROS), such as O2•- and •OH, which fragmented oligomeric DNA in the presence of uranyl and UV light. Detailed studies showed that DNA significantly enhances uranyl absorption in the UV-visible region, leading to the generation of more ROS. A fluorescence system for the selective detection of uranyl in seawater was established by immobilizing two complementary oligonucleotides with the fluorescent dye SYBR Green I. The strategy of UV-induced photocleavage offers high selectivity, excellent interference immunity, and high sensitivity for uranyl, with a detection limit of 6.8 nM. Additionally, the fluorescence can be visually detected using a 3D-printed miniaturized device integrated with a smartphone. This method has been successfully applied to the on-site detection of uranyl in seawater in 18 Chinese coastal cities and along the coast of Hainan Island within 3 min for a single sample. The sample testing and field analysis results indicate that this strategy has promising potential for real-time monitoring of trace uranyl in China's coastal waters. It is expected to be utilized for the rapid assessment of nuclear contamination and nuclear engineering construction.

Targeted Light-Induced Immunomodulatory Strategy for Implant-Associated Infections via Reversing Biofilm-Mediated Immunosuppression.

The clinical treatment efficacy for implant-associated infections (IAIs), particularly those caused by Methicillin-resistant Staphylococcus aureus (MRSA), remains unsatisfactory, primarily due to the formation of biofilm barriers and the resulting immunosuppressive microenvironment, leading to the chronicity and recurrence of IAIs. To address this challenge, we propose a light-induced immune enhancement strategy, synthesizing BSA@MnO2@Ce6@Van (BMCV). The BMCV exhibits precise targeting and adhesion to the S.aureus biofilm-infected region, coupled with its capacity to catalyze oxygen generation from H2O2 in the hypoxic and acidic biofilm microenvironment (BME), promoting oxygen-dependent photodynamic therapy efficacy while ensuring continuous release of manganese ions. Notably, targeted BMCV can penetrate biofilms, producing ROS that degrade extracellular DNA, disrupting the biofilm structure and impairing its barrier function, making it vulnerable to infiltration and elimination by the immune system. Furthermore, light-induced reactive oxygen species (ROS) around the biofilm can lyse S.aureus, triggering bacterium-like immunogenic cell death (ICD), releasing abundant immune costimulatory factors, facilitating the recognition and maturation of antigen-presenting cells (APCs), and activating adaptive immunity. Additionally, manganese ions in the BME act as immunoadjuvants, further amplifying macrophage-mediated innate and adaptive immune responses and reversing the immunologically cold BME to an immunologically hot BME. We prove that our synthesized BMCV elicits a robust adaptive immune response in vivo, effectively clearing primary IAIs and inducing long-term immune memory to prevent recurrence. Our study introduces a potent light-induced immunomodulatory nanoplatform capable of reversing the biofilm-induced immunosuppressive microenvironment and disrupting biofilm-mediated protective barriers, offering a promising immunotherapeutic strategy for addressing challenging S.aureus IAIs.

Efficient light-induced reactive oxygen species production from a far-red ER-targeting BODIPY dye.

Generation of reactive oxygen species (ROS) within the ER evokes stress leading to immunogenic cell death. A red light activated BODIPY dye capable of subcellular localization within the ER producing high quantum yields of ROS is reported. The ability of this dye to act as a photodynamic therapy (PDT) agent in breast cancer cells suggests promising organelle-targeted therapeutics.

Metal–BODIPY complexes: versatile photosensitizers for oxidizing amyloid-β peptides and modulating their aggregation profiles

Metal–BODIPY complexes, rationally designed as photosensitizers, can effectively oxidize amyloidogenic peptides upon photoactivation at a relatively longer wavelength, resulting in altering their aggregation profiles.

Efficient Fresh Lamp Light-Harvesting Films with the Self-Activating Continuous and Recyclable Bactericidal Ability for Ultrapersistent Freshness of Perishable Muscle Food.

A large quantity of perishable muscle food is being wasted due to harmful bacteria infestation during the sales and circulation each year and facing challenges. In this study, a self-activated bactericidal active film (PLA/g-C3N4@PCN-224) responsive to fresh lamp light was prepared, which showed excellent hydrophobicity, water vapor resistance, and thermal stability. Due to the synergistic effect between light-induced reactive oxygen species and the high specific surface area of g-C3N4@PCN-224, this film still maintains 99.99% bactericidal efficacy against Escherichia coli and Staphylococcus aureus after 10 days of continuous bactericidal activity test. The results of cell and hemolysis experiments indicated that the film was safe and nontoxic and can effectively preserve fresh pork for 7 days. Moreover, the film also exhibited a recyclable and efficient killing activity. A strategy for achieving ultrapersistent freshness of perishable muscle food was provided.

Photodynamic-responsive gelatin-based coating with high utilization curcumin loaded bilayer nanoencapsulation: A promising environmental food packaging

The development and application of curcumin-mediated antimicrobial photodynamic techniques (PDT) in food preservation are highly anticipated to resist microbial contamination and prevent food spoilage. In this study, high-utilization curcumin-loaded bilayer nanoencapsulation was prepared to incorporated into a gelatin-based edible coating for beef preservation. Bilayer nanoencapsulation composed of shellac and poly-γ-glutamic acid (CS-NPs) improved the encapsulation efficiency of shellac to curcumin by >1.5 times. The incorporation of CS-NPs improved the compact of coating structure with hydrogen bonds. In food simulants, coatings possessed control release properties and the release mechanism was Fick diffusion (without the addition of γ-PGA) and non-Fick diffusion (with the addition of γ-PGA). These prepared coatings exhibited excellent barrier, antibacterial (antibacterial ratio > 95 %), and antioxidant properties (scavenging ratio > 90 %). Curcumin mediated antimicrobial photodynamic techniques (PDT) of the coatings were verified with the activity of blue light-induced reactive oxygen species (ROS). The shelf-life of beef was extended by the coating with blue light. In summary, the design of bilayer CS-NPs significantly improved the utilization of curcumin which provided a high-efficiency strategy for PDT-responsive food packaging with environmental practical.

Stimuli-Responsive Pressure-Strain Sensor-Based Conductive Hydrogel for Alleviated Non-alcoholic Fatty Liver Disease by Scavenging Reactive Oxygen Species in Adipose Tissue.

A visible light- and reactive oxygen species (ROS)-responsive pressure/strain sensor based on carbon dot (CD)-loaded conductive hydrogel was developed for detecting high-fat diet (HFD) and preventing the risk of non-alcoholic fatty liver disease. The designed nanoparticle consisted of a diselenide polymer dot (dsPD) loaded with a visible light-responsive CD to form dsPD@CD (DSCD). The influence of visible light irradiation and ROS on DSCD facilitated the electron transport, enhancing the conductivity of DSCD-embedded hydrogel (DSCD hydrogel) from 1.3 to 35.9 mS/m. Alternatively, the tensile modulus of the DSCD hydrogel enhanced to 223% after light-induced ROS treatment, which simultaneously impacted the capacitive response (120%). The hydrogel implantation into inguinal white adipose tissue of HFD mice showed 82% higher conductivity and 83% enhanced pressure sensing response to HFD-generated high ROS levels compared with the normal diet-fed mice. Additionally, the ROS scavenging activity of DSCD hydrogel was confirmed by the downregulation of ROS-responsive genes, such as Sod2, Nrf2, and catalase (Cat) in murine primary hepatocytes isolated from fatty liver-induced mice. In addition, in vivo animal studies also confirmed the suppression of hepatic lipogenesis, as shown by decreased Pparγ and Fasn expression and hypertrophy of adipocytes in HFD mice. The distinguishable real-time wireless resistance response observed with pressure sensing indicates the potential application of the device for monitoring the risk of non-alcoholic fatty liver disease. STATEMENT OF SIGNIFICANCE: : A visible-light-induced ROS-responsive carbon dot-loaded conductive hydrogel was developed for the detection of HFD-induced alterations in ROS levels by evaluating the conductivity and electrochemical responses with applied pressure/strain. The implanted hydrogel facilitates the recovery of the inflated adipocytes induced by NAFLD, which reduces fat accumulation in the liver, preventing the risk of NAFLD. Real-time detection based on the resistance response during local compression of the hydrogel is possibly performed utilizing a wireless sensing device, demonstrating the ease of NAFLD monitoring.

Visible light-responsive chitosan/sodium alginate/QDs@ZIF-8 nanocomposite films with antibacterial and ethylene scavenging performance for kiwifruit preservation

The deterioration of postharvest fruits and vegetables caused by ethylene and microorganisms leads to global resource waste. Therefore, the development of multifunctional packaging materials that scavenge ethylene and inhibit microorganisms is essential to extend the shelf life of fruits and vegetables and minimize waste. Herein, the excellent photocatalytic performance of QDs@ZIF-8 nanoparticles (NPs) was incorporated into chitosan (CS) and sodium alginate (SA) matrix by layer-by-layer (LBL) assembly method to obtain CS/SA/QDs@ZIF-8 nanocomposite films. These as-prepared films exhibited excellent antibacterial (antibacterial ratio >99%) and ethylene scavenging properties (scavenging ratio >40%) due to the synergistic effect between visible light-induced reactive oxygen species (ROS) and the higher specific surface area of ZIF-8. The extended shelf life of kiwifruit further confirmed the enhanced preservation performance of CS/SA/QDs@ZIF-8 nanocomposite films. In addition, with the addition of QDs@ZIF-8 NPs, the color, and transparency of the films decreased, but the mechanical strength, thermal stability, hydrophobicity, and barrier properties were improved. In summary, the successful construction of ethylene scavenging and antibacterial films provides new ideas for the innovation of environmentally adaptive and functionalization of films.

Neurite growth induced by red light-caused intracellular reactive oxygen species production through cytochrome c oxidase activation.

The applications of red-light photobiomodulation (PBM) to enhance neurite growth have been proposed for many years. However, the detailed mechanisms require further studies. In the present work we used a focused red-light spot to illuminate the junction of the longest neurite and the soma of a neuroblastoma cell (N2a), and demonstrated enhanced neurite growth at 620nm and 760nm with adequate illumination energy fluences. In contrast, 680nm light showed no effect on neurite growth. The neurite growth was accompanied with the increase of intracellular reactive oxygen species (ROS). Using Trolox to reduce the ROS level, this red light-induced neurite growth was hindered. Suppressing the activities of cytochrome c oxidase (CCO) by using either a small-molecule inhibitor or siRNA abrogated the red light-induced neurite growth. These results suggest that red light-induced ROS production through the activation of CCO could be beneficial for neurite growth.

Room-Temperature Phosphorescence and Cellular Phototoxicity Activated by Triplet Dynamics in Aggregates of Push-Pull Phenothiazine-Based Isomers.

In this study, we report a comprehensive time-resolved spectroscopic investigation of the excited-state deactivation mechanism in three push-pull isomers characterized by a phenothiazine electron donor, a benzothiazole electron acceptor, and a phenyl π-bridge where the connection is realized at the relative ortho, meta, and para positions. Spin-orbit charge-transfer-induced intersystem crossing takes place with high yield in these all-organic donor-acceptor compounds, leading also to efficient production of singlet oxygen. Our spectroscopic results give clear evidence of room-temperature phosphorescence not only in solid-state host-guest matrices but also in highly biocompatible aggregates of these isomers produced in water dispersions, as rarely reported in the literature. Moreover, aggregates of the isomers could be internalized by lung cancer and melanoma cells and display bright luminescence without any dark cytotoxic effect. On the other hand, the isomers showed significant cellular phototoxicity against the tumor cells due to light-induced reactive oxygen species generation. Our findings strongly suggest that nanoaggregates of the investigated isomers are promising candidates for imaging-guided photodynamic therapy.

Cherry fruit anthocyanins cyanidin-3-O-glucoside and cyanidin-3-O-rutinoside protect against blue light-induced cytotoxicity in HaCaT cells

Blue light derived from multiple sources, including sunlight, generates reactive oxygen species (ROS) and negatively affects the skin in a manner similar to that of ultraviolet light. Cyanidin-3-O-glucoside (C3OG) and cyanidin-3-O-rutinoside (C3OR) are anthocyanin antioxidants that have protective effects on various tissues and cell types. However, the effects of anthocyanins on blue light-mediated changes remain unconfirmed. In this study, we determined the protective effects of C3OG and C3OR isolated and purified from waste cherry fruits (Prunus serrulata L. var. tomentella Nakai) against the blue light-induced ROS formation and inflammatory responses in HaCaT cells. It is showed that the treatment of C3OG and C3OR significantly reduced the blue light-induced cytotoxicity and ROS production in a dose dependent manner. Furthermore, we found that focal adhesion kinase (FAK) is a major upstream of blue light-induced expression of inflammatory cytokines (TNF-α, IL-6 and IL-8), and these effects were attenuated by C3OG or C3OR treatment. In the initial reaction, blue lights increased the phosphorylation of inhibitory-κB Kinase α (IKKα), c-jun N-terminal kinase (JNK), and p38. The phosphorylation of these intracellular proteins was reduced via FAK inhibitor, NAC (ROS scavenger), and anthocyanin treatments. After 24 h of blue light irradiation, C3OG or C3OR treatment markedly inhibited caspase-3-mediated apoptosis and cleaved-FAK-mediated anoikis, which is cell detachment-induced apoptosis. Therefore, our results indicate that C3OG and C3OR effectively protected human keratinocytes from harmful blue light-induced cytotoxicity and inflammation.

Light-Mediated Modulation of Enzyme-Mimetic Activity of CuMnO2 Nanosheets.

As typical copper-based-manganese delafossites, CuMnO2 nanoflakes (CuMnO2 NFs) display excellent light absorption capacity, showing great promise for facile regulation of catalysis reactions by light stimulation. Here, we report that the as-prepared CuMnO2 NFs possess intrinsic peroxidase- and oxidase-like activities, which are capable of catalyzing the oxidation of the enzymatic substrate 3,3',5,5'-tetramethylbenzidine (TMB) in the presence of H2O2 or O2, respectively. Additionally, CuMnO2 NFs exhibit significant light-enhanced dual enzyme-like performance (peroxidase and oxidase) for accelerating the oxidation of TMB, owing to a much greater light-induced reactive oxygen species (ROS) yield. The light-induced hydroxyl radicals (·OH) and superoxide radicals (O2•-) hold the key to rapidly catalyzing the oxidation of TMB into the two-electron charge transfer complex diamine, subsequently improving the oxidation capacity and enzyme-like activity of CuMnO2 NFs as peroxidase and oxidase mimics. These findings confirm an efficient way to improve the enzyme-like performance of nanozymes and regulate the reactive oxygen species (ROS) level by simply employing irradiated light. This work also provides a general method for constructing external field-modulated smart nanozymes and an insightful approach to the corresponding enzyme-mimetic reaction mechanism.

Analyzing Efficacy and Safety of Anti-Fungal Blue Light Therapy via Kernel-Based Modeling the Reactive Oxygen Species Induced by Light.

The goal of this study is to investigate the efficacy, safety, and mechanism of ABL for inactivating Candida albicans (C. albicans), and to determine the best wavelength for treating candida infected disease, by experimental measurements and dynamic modeling. The changes in reactive oxygen species (ROS) in C. albicans and human host cells under the irradiation of 385, 405, and 415 nm wavelengths light with irradiance of 50 mW/cm2 were measured. Moreover, a kernel-based nonlinear dynamic model, i.e., nonlinear autoregressive with exogenous inputs (NARX), was developed and applied to predict the concentration of light-induced ROS, whose kernels were selected by a newly developed algorithm based on particle swarm optimization (PSO). The ROS concentration was increased respectively about 10-12 times in C. albicans and about 3-6 times in human epithelial cells by the ABL treatment with the same fluence of 90 J/cm2. The NARX models were respectively fitted to the data from the experiments on both types of cells. Besides, four different kernel functions, including Gaussian, Laplace, linear and polynomial kernels, were compared in their fitting accuracies. The errors with the Laplace kernel turned out to be only 0.2704 and 0.0593, as respectively fitted to the experimental data of the C. albicans and human host cells. The results demonstrated the effectiveness of the NARX modeling approach, and revealed that the 415 nm light was more effective as an anti-fungal treatment with less damage to the host cells than the 405 or 385 nm light. The kernel-based NARX model identification algorithm offers opportunities for determining the effective and safe light dosages in treating various fungal infection diseases.

An advanced multifunctional prodrug combining photodynamic therapy with chemotherapy for highly efficient and precise tumor ablation

It is urgently required to develop the molecular agents with high therapeutic efficacy and low side effects for precise cancer therapy. Herein, an advanced multifunctional prodrug B-BDP-CL-CPT, consisting of a glutathione (GSH)-responsive boron dipyrromethene (BDP)-based photosensitizer, a chemo drug camptothecin (CPT), a reactive oxygen species (ROS)-sensitive chain linking BDP with CPT, and a biotin fragment with specific affinity to biotin-receptor, was designed and synthesized. Owing to the introduction of 2,4-dinitrobenzenesulfonate (DNBS) as the quencher into BDP core and chemical modification of CPT at the active site, B-BDP-CL-CPT is non-cytotoxic. Having biotin as the targeting ligand, the prodrug can be efficiently internalized by biotin-acceptor positive cancer cells, and then activated to generate substantial light-induced ROS through removal of the stopper DNBS by intracellular GSH which is abundant in cancer cells. The resulting ROS can not only serve for photodynamic therapy (PDT) but also further cleave thioketal linkage causing the release of CPT, to achieve dual PDT and chemotherapy. And the combination of preferential accumulation with specific activation in tumor tissues for this prodrug is conducive to improving the dual therapeutic outcome meanwhile greatly reducing the photo-damage and the release of CPT in adjacent healthy tissues. In addition, the activated B-BDP-CL-CPT can give bright fluorescence signals which are favor for imaging-guided PDT and real-time tracking the release of therapeutic agent. The in vitro and in vivo investigations have been performed to demonstrate the targeted and combined anticancer activities of this prodrug against HepG2 and HeLa cancer cells, and the mice with H22 tumors. This study proposes a new tactics for highly efficient and precise cancer treatment.

Tuning riboflavin derivatives for photodynamic inactivation of pathogens

The development of effective pathogen reduction strategies is required due to the rise in antibiotic-resistant bacteria and zoonotic viral pandemics. Photodynamic inactivation (PDI) of bacteria and viruses is a potent reduction strategy that bypasses typical resistance mechanisms. Naturally occurring riboflavin has been widely used in PDI applications due to efficient light-induced reactive oxygen species (ROS) release. By rational design of its core structure to alter (photo)physical properties, we obtained derivatives capable of outperforming riboflavin’s visible light-induced PDI against E. coli and a SARS-CoV-2 surrogate, revealing functional group dependency for each pathogen. Bacterial PDI was influenced mainly by guanidino substitution, whereas viral PDI increased through bromination of the flavin. These observations were related to enhanced uptake and ROS-specific nucleic acid cleavage mechanisms. Trends in the derivatives’ toxicity towards human fibroblast cells were also investigated to assess viable therapeutic derivatives and help guide further design of PDI agents to combat pathogenic organisms.

Tunable Antibacterial Activity of a Polypropylene Fabric Coated with Bristling Ti3C2Tx MXene Flakes Coupling the Nanoblade Effect with ROS Generation

Polypropylene (PP) is a thermoplastic polymer widely used as a medical textile in healthcare applications due to its low cost and superior performance. However, it does not show antibacterial properties leading to the possibility of pathogen transmission. Herein, we have developed an antibacterial medical fabric by facile self-assembly of delaminated two-dimensional (2D) Ti3C2Tx MXene flakes bristling on the surface of PP fibers. The increasing amount of MXene in the coating solution from 1 up to 32 mg/mL allowed for edge-on assembly of MXene flakes on the PP surface and tracking the evolution of the band gap for a restacked structure. Characterization of the PP/Ti3C2Tx nanocomposite has proven that it exhibited highly effective antibacterial, robust coating, and chemically/thermally stable properties. The in vitro microbiological studies against Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus have shown that PP/Ti3C2Tx reduced the bacterial viability up to 100%, as driven by synergistic membrane stress mediated by physical contact and light-induced reactive oxygen species (ROS) generation. Moreover, the use of l-ascorbic acid for MXene stabilization allowed for achieving excellent thermal stability of the PP/Ti3C2Tx nanocomposite upon accelerated thermal aging. Collectively, this work provides a facile surface engineering strategy for designing medical fabrics with outstanding functional performances. By demonstrating the exceptional performance of the stabilized MXene in a self-assembly nanocomposite structure, we are opening the door for MXenes to be applied in other biomedical fields.

LED Light-Induced ROS Differentially Regulates Focal Adhesion Kinase Activity in HaCaT Cell Viability.

In this study, changes in cell signaling mechanisms in skin cells induced by various wavelengths and intensities of light-emitting diodes (LED) were investigated, focusing on the activity of focal adhesion kinase (FAK) in particular. We examined the effect of LED irradiation on cell survival, the generation of intracellular reactive oxygen species (ROS), and the activity of various cell-signaling proteins. Red LED light increased cell viability at all intensities, whereas strong green and blue LED light reduced cell viability, and this effect was reversed by NAC or DPI treatment. Red LED light caused an increase in ROS formation according to the increase in the intensity of the LED light, and green and blue LED lights led to sharp increases in ROS formation. In the initial reaction to LEDs, red LED light only increased the phosphorylation of FAK and extracellular-signal regulated protein kinase (ERK), whereas green and blue LED lights increased the phosphorylation of inhibitory-κB Kinase α (IKKα), c-jun N-terminal kinase (JNK), and p38. The phosphorylation of these intracellular proteins was reduced via FAK inhibitor, NAC, and DPI treatments. Even after 24 h of LED irradiation, the activity of FAK and ERK appeared in cells treated with red LED light but did not appear in cells treated with green and blue LED lights. Furthermore, the activity of caspase-3 was confirmed along with cell detachment. Therefore, our results suggest that red LED light induced mitogenic effects via low levels of ROS–FAK–ERK, while green and blue LED lights induced cytotoxic effects via cellular stress and apoptosis signaling resulting from high levels of ROS.

Free-Radical Scavenger NSP-116 Protects the Corneal Epithelium against UV-A and Blue LED Light Exposure.

The corneal epithelium is continuously exposed to oxygen, light, and environmental substances. Excessive exposure to those stresses is thought to be a risk factor for eye diseases. Photokeratitis is damage to the corneal epithelium resulting in a painful eye condition caused by unprotected exposure to UV rays, usually from sunlight, and is often found in people who spend a long time outdoors. In modern life, human eyes are exposed to artificial light from light-emitting diode (LED) displays of computers and smartphones, and it has been shown that short-wavelength (blue) LED light can damage eyes, especially photoreceptors. However, the effect of blue LED light on the cornea is less understood. In addition, it is important to develop new treatments for preserving human eyesight and eye health from light stress. Here, we used human corneal epithelial cells-transformed (HCE-T) cells as an in-vitro model to investigate the protective effect of NSP-116, an imidazolyl aniline derivative, against the oxidative stress induced by light in the corneal epithelium. Treatment with 10 µM NSP-116 significantly increased the cell viability and reduced the death ratio following UV or blue LED light exposure. Furthermore, NSP-116 treatment decreased light-induced reactive oxygen species production and preserved the mitochondrial membrane potential. Immunoblotting data showed that NSP-116 suppressed the stress response pathway. Finally, NSP-116 treatment prevented corneal epithelial apoptosis induced by blue LED light in an in-vivo mouse model. In conclusion, NSP-116 has a protective effect against oxidative stress and corneal cell death from both UV and blue LED light exposure.

Light-Induced Reactive Oxygen Species (ROS) Generator for Tumor Therapy through an ROS Burst in Mitochondria and AKT-Inactivation-Induced Apoptosis.

Mitochondria are identified as a valuable target for cancer therapy owing to their primary function in energy supply and cellular signal regulation. Mitochondria in tumor cells are depicted by excess reactive oxygen species (ROS), which lead to numerous detrimental results. Hence, mitochondria-targeting ROS-associated therapy is an optional therapeutic strategy for cancer. In this contribution, a light-induced ROS generator (TBTP) is developed for evaluation of the efficacy of mitochondria-targeting ROS-associated therapy and investigation of the mechanism underlying mitochondrial-injure-mediated therapy of tumors. TBTP serves as an efficient ROS generator with low cytotoxicity, favorable biocompatibility, excellent photostability, mitochondria-targeted properties, and NIR emission. In vivo and in vitro experiments reveal that TBTP exhibits effective anticancer potential. ROS generated from TBTP could destroy the integrity of mitochondria, downregulate ATP, decrease the mitochondrial membrane potential, secrete Cyt-c into cytoplasm, activate Caspase-3/9, and induce cell apoptosis. Moreover, RNA-seq analysis highlights that an ROS burst in mitochondria can kill tumor cells via inhibition of the AKT pathway. All these results prove that mitochondrial-targeted ROS-associated therapy hold great potential in cancer therapy.

Construction of precisely controllable and stable interface bonding Au-TiO2/PVDF composited membrane for biofouling-resistant properties

A novel approach combining atomic layer deposition (ALD) and photodeposition was proposed to prepare stable and antibacterial organic–inorganic composited membrane. These composited membranes were fabricated by atomic-scale coating of titanium on the surface of a commercial polyvinylidene fluoride (PVDF) membrane under vacuum, followed by Au photosensitization under ultraviolet (UV) irradiation. Thus, as-prepared Au-TiO2/PVDF composite membrane could exhibit similar mechanical stability and separation performance as pristine PVDF membrane because of chemisorbing between PVDF and TiO2. We have studied the antibacterial mechanism of increasing surface hydrophilicity to prevent bacterial adhesion and cytotoxic reactive oxygen sterilization. The results showed that light-induced reactive oxygen species dominate the antibacterial process. Additionally, Au-TiO2/PVDF composite membranes have potential degradation ability of dye, such as trace methyl orange in aqueous solution. Our present approach demonstrates great potential for the controllable preparation of inorganic modification on inert organic surface with different compositions and shapes for various applications.
