Vitamin B12 Protects Against Genotoxicity Induced by Cisplatin.

Cisplatin is an effective synthetic chemotherapeutic drug used for cancer treatment. Vitamin B12 has been shown to possess anti-genotoxic activity. This study aimed to investigate the effect of vitamin B12 on chromosomal damage induced by cisplatin. The level of sister chromatid exchanges (SCEs) and chromosomal aberrations (CAs) were measured in cultured human blood lymphocytes treated with cisplatin and/or vitamin B12. The results showed a significantly elevated frequency of CAs and SCEs of cisplatin-treated cultures compared to the control (P < 0.05). The CAs and SCEs induced by cisplatin were significantly lowered by pretreatment of cell cultures with vitamin B12. In addition, cisplatin caused a slight reduction in the mitotic index (MI), while vitamin B12 did not modulate the effect of cisplatin on MI. Vitamin B12 can protect human lymphocytes against genotoxicity associated with cisplatin.

Study of antimutagenic activity of medicinal plant infusions Crataegus sanguinea Pall. family Rosaceae in plant test systems

Medicinal plants, integral to traditional medicine systems, are rich sources of biologically active substances that benefit physiological and biochemical processes within living organisms. Amongst these activities, antimutagenic and genoprotective properties stand out, offering mitigation against genotoxic effects induced by adverse environmental factors on genetic material. This study delves into the mutagenic and antimutagenic capacities of aqueous and alcoholic infusions derived from Crataegus sanguinea Pall. (Rosaceae family), utilising Hordeum vulgare L. as a plant-based test subject. The assessment employed the metaphase chromosome analysis technique. Results indicate that these infusions exhibit no mutagenic activity, with the level of chromosomal aberrations in barley seeds treated with these infusions not exceeding the natural mutation rate in a statistically significant manner. When infusions were combined with Methyl methanesulfonate (positive control) exposure – irrespective of exposure sequence – a statistically significant attenuation in MMS-induced mutagenesis was observed (p&lt;0.01). A 56-60% reduction quantified the antimutagenic efficacy of C.sanguinea infusions. This metric underscores the infusions’ capability to inhibit MMS-induced mutagenesis by 50–60%, positioning these water and alcohol-based extracts of common hawthorn as viable candidates for safeguarding against chemically induced mutagenic factors.

Small extrachromosomal circular DNA harboring targeted tumor suppressor gene mutations supports intratumor heterogeneity in mouse liver cancer induced by multiplexed CRISPR/Cas9

BackgroundPrimary liver cancer has significant intratumor genetic heterogeneity (IGH), which drives cancer evolution and prevents effective cancer treatment. CRISPR/Cas9-induced mouse liver cancer models can be used to elucidate how IGH is developed. However, as CRISPR/Cas9 could induce chromothripsis and extrachromosomal DNA in cells in addition to targeted mutations, we wondered whether this effect contributes to the development of IGH in CRISPR/Cas9-induced mouse liver cancer.MethodsCRISPR/Cas9-based targeted somatic multiplex-mutagenesis was used to target 34 tumor suppressor genes (TSGs) for induction of primary liver tumors in mice. Target site mutations in tumor cells were analyzed and compared between single-cell clones and their subclones, between different time points of cell proliferation, and between parental clones and single-cell clones derived from mouse subcutaneous allografts. Genomic instability and generation of extrachromosomal circular DNA (eccDNA) was explored as a potential mechanism underlying the oscillation of target site mutations in these liver tumor cells.ResultsAfter efficiently inducing autochthonous liver tumors in mice within 30–60 days, analyses of CRISPR/Cas9-induced tumors and single-cell clones derived from tumor nodules revealed multiplexed and heterogeneous mutations at target sites. Many target sites frequently displayed more than two types of allelic variations with varying frequencies in single-cell clones, indicating increased copy number of these target sites. The types and frequencies of targeted TSG mutations continued to change at some target sites between single-cell clones and their subclones. Even the proliferation of a subclone in cell culture and in mouse subcutaneous graft altered the types and frequencies of targeted TSG mutations in the absence of continuing CRISPR/Cas9 genome editing, indicating a new source outside primary chromosomes for the development of IGH in these liver tumors. Karyotyping of tumor cells revealed genomic instability in these cells manifested by high levels of micronuclei and chromosomal aberrations including chromosomal fragments and chromosomal breaks. Sequencing analysis further demonstrated the generation of eccDNA harboring targeted TSG mutations in these tumor cells.ConclusionsSmall eccDNAs carrying TSG mutations may serve as an important source supporting intratumor heterogeneity and tumor evolution in mouse liver cancer induced by multiplexed CRISPR/Cas9.

Individual Risk Assessment for Population Living on the Territories Long-Term Polluted by Organochlorine Pesticides

The long-term storage of unutilized pesticides raised new problems of long-term environmental contamination. The study presents the results of surveying 151 individuals in 7 villages living close to pesticide-contaminated localities. All individuals have been surveyed concerning their consumption habits and lifestyle characteristics. An assessment of the general exposure risks of the local population was carried out using the analysis of pollutants in food products and the average levels of their consumption in the region. The cohort risk evaluation revealed that the greatest risk was associated with the regular consumption of cucumbers, pears, bell peppers, meat, and milk. The new model to estimate individual risks of long-term pesticide pollution was proposed as a calculation of the combined action of 9 risk factors, including individual genotypes, age, lifestyle, and personal pesticide consumption rates. The analysis of the predictive ability of this model showed that the final score for individual health risks corresponded to the development of chronic diseases. A high level of chromosomal aberrations was evidenced for individual genetic risk manifestations. The combined influence of all risk factors revealed contributions of 24.7% for health status and 14.2% for genetic status, while other impacts go to all unaccounted factors.

Biological (microfloral) factors of influence on cytogenetic stability during chemical mutagenesis

ABSTRACT The article presents the results of a study of chromosomal mutations in residents living in the Aral Sea disaster zone. The present study was designed to evaluate the impact of the combined effect of a chemical mutagen (nickel) and bacterial microflora on the level of chromosomal aberrations (CA) in peripheral blood lymphocytes. This study used classical methods of cell cultivation, methods for determining chromosomal aberrations, a cytomorphological method for assessing epithelial cells, and an atomic absorption method for determining trace elements in the blood. The article shows that with an increase of chemical agent in the blood, the number of cells with features of damage and cells with contamination by microflora increases. Both of these factors cause an increase in the frequency of chromosomal aberrations. The article demonstrates how being exposed to a chemical factor increases chromosomal mutations, and also damages membrane components, which leads to a decrease in the barrier and protective function of the cell, and as a result also affects the level of chromosomal aberrations.

Cytogenetic features in adolescent girls with abnormal uterine bleeding which is comorbid with multi­factor diseases

Objective — to study the state of chromosomal apparatus in adolescent girls with abnormal uterine bleeding which is comorbid with multi-­factor diseases.&#x0D; Materials and methods. Cytogenetic analysis has been carried out in 25 girls aged 14 to 17 with abnormal uterine bleeding (AUB) which is comorbid with multifactor diseases (endocrine and nervous). The comparison group included 25 age-matching healthy girls. All the girls have been examined in the Laboratory of Medical Genetics of the Institute. Statistical processing of the study results has been carried out following the Student’s t­test with the use of Excel software package.&#x0D; Results and discussion. According to the data of cytogenetic analysis, it has been established that the total level of chromosome aberrations in blood lymphocytes of sick girls constituted 5.44 % and exceeded this value by a factor of 2.2 in comparison with the frequency in healthy girls. A 1.8-­fold increase of the number of chromosome aberrations was observed; among them the paired acentric fragments have been recorded twice as often. Chromatid­type aberrations in blood lymphocytes were observed in 2.9 times oftener in girls with AUB, mainly due to an increase in the number of single acentric fragments. The frequency of genomic disorders was 0.60 % that is five times more than their frequency in blood lymphocytes of healthy girls.&#x0D; Conclusions. Cytogenetic analysis conducted in adolescent girls with abnormal uterine bleeding and comorbid pathology have revealed an increase of the overall level of chromosome aberrations in comparison with healthy girls. Paired acentric fragments prevailed among chromosome aberrations, single acentric fragments prevailed among chromatid-­type aberrations, polyploid cells prevailed among genomic disorders.

Cytogenetic analysis of the population living in close proximity to the destroyed warehouses of banned pesticides

Assessment of genotoxicity of pesticides and biomonitoring of their impact on the population living in the area of ​​disposal of endangered stocks of pesticides are of exceptional importance for regulating the ecological state of the environment and protecting the population. The purpose of this study was to assess the level of cytogenetic damage (chromosomal aberrations) in residents of the villages of Karakastek and Umbetala, Zhambul district, Almaty region, located in areas contaminated with pesticides banned for occurrence more than 10 years ago. The soils of the settlements are contaminated with organochlorine pesticides such as aldrin, deldrin, eldrin, chlorobenzilate and DDT. In food, their content does not exceed the MPC. However, there was an excess of MPC for heavy metals - Ni and Co. The residents of v. Karakastek and Umbetali showed a significant excess of the total frequency of chromosomal aberrations compared to the control (p≤0.01). The occurrence of cases of shift of mutations from the chromatid adrenal glands in the foci of inflammation, the destruction of the nature of chemical mutagenesis. An analysis of the state and dynamics of the level of chromosomal aberrations in the population living in territories contaminated with pesticides can help in developing a rationale for predicting long-term biomedical events and normal control over the health of the population. Clinical and genetic aspect of resistant depression in adolescents

The work considers and distinguishes the psychopathological symptom complexes of depression in adolescents that are resistant to therapy: anxious, obsessive, behavioral. The level of spontaneous and induced mutagenesis in the blood lymphocytes of adolescents with depression was determined in vitro, which was twice the level of chromosomal aberrations after introducing a mutagen into the culture mixture, which indicates a pronounced individual and group hidden chromosomal instability of patients with depression. The constants of the lack of effectiveness of therapeutic intervention in adolescents with depression were determined (infantilism, personal anxiety, cogni- tive component of depression, noncompliance, addictions, comorbidity of depression with anorexia nervosa, suicidal behavior, recurrent depression, insufficient titration of antidepressant medication, unpleasant side effects of antidepressants).

MEDICAL AND RADIOBIOLOGICAL ASPECTS OF RADIATION COMDPLICATIONS IN PATIENTS WITH ONCOGINECOLOGICAL PROFILE

Summary. Aim: to analyze the literature regarding the causes of the occurrence and features of distant complications of radiation therapy of cancer patients; investigation of the frequency and spectrum of spontaneous aberrations of chromosomes in the lymphocytes of peripheral blood of patients with oncogynecological profile (body cancer and cervix) before the onset of radiation therapy. Object and methods: peripheral blood lymphocyte test system with metaphase analysis of chromosome aberrations of 32 primary of cancer patients (follow-up group) and 30 conditionally healthy donors (comparison group). The examination of patients was performed before the onset of radiation therapy. Results: based on the analysis of literature data, the path to a personalized approach to the planning of radiation therapy for patients with oncogyne5 cological profile, the treatment of which is complicated by radiation lesions from the organs and tissues of the pelvis. The clinical and radiobiological aspects of the formation of radiation complications are considered in detail, the search for genetic indicators for the detection of patients with a high risk of developing radiation complications is justified. The results of cytogenetic examinations of patients with endometrial cancer and cervical cancer are close and indicate a 6-fold increase in the frequency of spontaneous chromosome aberrations compared to the population rate. In the spectrum of chromosomal restructures, complex restructures are recorded, which is uncharacteristic of the spontaneous level of aberration in healthy donors, as well as increased levels of chromatide type aberrations. Conclusions: the increased level of spontaneous chromosomal aberrations in T-lymphocytes of primary oncogynecological patients and the predominance of chromatid-type aberrations in the spectrum of registered chromosomal rearrangements indicate that genetic instability is formed in healthy cells before the start of radiation therapy, which predicts the risk of distant radiation complications, including the occurrence of secondary tumors radiation genesis. The examination of patients with the use of cytogenetic test will provide the most reasonable conclusion about the individual radio sensitivity of the patient to the onset of radiation therapy and will contribute to increasing its effectiveness, as well as improving the quality of life.

Antibacterial, Antioxidant Activities, GC-Mass Characterization, and Cyto/Genotoxicity Effect of Green Synthesis of Silver Nanoparticles Using Latex of Cynanchum acutum L

Green synthesis of nanoparticles is receiving more attention these days since it is simple to use and prepare, uses fewer harsh chemicals and chemical reactions, and is environmentally benign. A novel strategy aims to recycle poisonous plant chemicals and use them as natural stabilizing capping agents for nanoparticles. In this investigation, silver nanoparticles loaded with latex from Cynanchum acutum L. (Cy-AgNPs) were examined using a transmission electron microscope, FT-IR spectroscopy, and UV-visible spectroscopy. Additionally, using Vicia faba as a model test plant, the genotoxicity and cytotoxicity effects of crude latex and various concentrations of Cy-AgNPs were studied. The majority of the particles were spherical in shape. The highest antioxidant activity using DPPH was illustrated for CAgNPs (25 mg/L) (70.26 ± 1.32%) and decreased with increased concentrations of Cy-AGNPs. Antibacterial activity for all treatments was determined showing that the highest antibacterial activity was for Cy-AgNPs (50 mg/L) with inhibition zone 24 ± 0.014 mm against Bacillus subtilis, 19 ± 0.12 mm against Escherichia coli, and 23 ± 0.015 against Staphylococcus aureus. For phytochemical analysis, the highest levels of secondary metabolites from phenolic content, flavonoids, tannins, and alkaloids, were found in Cy-AgNPs (25 mg/L). Vicia faba treated with Cy-AgNPs- (25 mg/L) displayed the highest mitotic index (MI%) value of 9.08% compared to other Cy-AgNP concentrations (50-100 mg/L) and C. acutum crude latex concentrations (3%). To detect cytotoxicity, a variety of chromosomal abnormalities were used, including micronuclei at interphase, disturbed at metaphase and anaphase, chromosomal stickiness, bridges, and laggards. The concentration of Cy-AgNPs (25 mg/L) had the lowest level of chromosomal aberrations, with a value of 23.41% versus 20.81% for the control. Proteins from seeds treated with V. faba produced sixteen bands on SDS-PAGE, comprising ten monomorphic bands and six polymorphic bands, for a total percentage of polymorphism of 37.5%. Eight ISSR primers were employed to generate a total of 79 bands, 56 of which were polymorphic and 23 of which were common. Primer ISSR 14 has the highest level of polymorphism (92.86%), according to the data. Using biochemical SDS-PAGE and ISSR molecular markers, Cy-AgNPs (25 mg/L) showed the highest percentage of genomic template stability (GTS%), with values of 80% and 51.28%, respectively. The findings of this work suggest employing CyAgNPs (25 mg/L) in pharmaceutical purposes due to its highest content of bioactive compounds and lowest concentration of chromosomal abnormalities.

Background Level of Unstable Chromosome Aberrations in the Kazakhstan Population: A Human Biomonitoring Study.

Kazakhstan is known as a country with a complex radioecological situation resulting from different sources such as a natural radiation background, extensive activities of the industrial system of the former Soviet Union and a well-known testing of nuclear power weapons occurred in the Semipalatinsk Test Site (STS) area. The present study focuses on the assessment of the background of dicentric chromosomes in Kazakhstan’s population, which is the starting point in the dose assessment of irradiated people, since the baseline level of spontaneous dicentrics can vary significantly in different populations. In this context, aiming to determine the background frequency of chromosome aberrations in the population of Kazakhstan, considering the heterogeneity of natural radiation background levels of its large territory, a selection of 40 control subjects living in four cities of North, South, West and East Kazakhstan was performed. The cytogenetic study on the selected groups showed fairly low background frequency values of chromosome aberrations (0.84 ± 0.83 per 1000 cells), comparable with other data in the literature on general populations, reporting background frequency values between 0.54 and 2.99 per 1000 cells. The obtained results should be taken into account when constructing the dose–effect calibration curve used in cytogenetic biodosimetry, as a “zero” dose point, which will reduce the uncertainty in quantifying the individual absorbed dose in emergency radiological situations.

Effect of Gamma Radiation on Cytogenetic and Growth Endpoints of Allium cepa Seedlings in Long-Term Experiments

The effect of γ-radiation on the growth and cytogenetic endpoints of Allium cepa L. seedlings in a long period after irradiation in absorbed doses from 1.0 to 10.7 Gy were examined. The chromosome aberration rate was most sensitive to the radiation: it increased immediately after exposure at all doses. In the recovery period (up to 216 h) after irradiation, the chromosome aberration frequency naturally decreased but at the end of experiment in maximum doses remained above the control values. The impact of the initial level of chromosome aberrations on the inhibition of onion roots growth in the long terms after irradiation is discussed.

Alleviation of the Genotoxic Effect of Experimental Circulatory Brain Hypoxia in Rat Cornea under the Effect of Infrared Low-Level Laser Radiation.

Experimental modeling of oxidative stress was conducted by creating circulatory brain hypoxia, accompanied by development of free radical processes leading to genome instability through an example of anterior epithelial layer of rat cornea. We studied the influence of infrared low-level laser radiation (ILLLR) in the therapeutic dose and mode (pulse power 8 W, λ=0.89 μm). Exposure of animals with circulatory brain hypoxia (ischemia) to ILLLR reduced the level of chromosome aberrations in corneal epitheliocytes and led to change in the mitotic index. Correction of the damaging effects of ischemia in the experiment in the absence of mutagenic effect of ILLLR in therapeutic dose and mode of exposure indicates the possibility of applying this physical method for correcting after-effects of oxidative stress during progression of degenerative eye diseases in ophthalmic practice.

CYTOGENETIC STUDY OF MANIFESTATIONS OF THE UNIVERSAL PHENOMENON OF THE BYSTANDER RESPONSE.

to establish the level of chromosomal instability in peripheral blood lymphocytes (PBL) from healthyindividuals and cells from patients with B-cell chronic lymphocytic leukemia (CLL) under co-cultivation conditions and to determine the possibility of inducing manifestations of the universal phenomenon of bystander response in them. Cytogenetic analysis of uniformly stained chromosomes from human PBL, which differed by cytogenetic markers of sex; the presence of oncological transformation and in vitro irradiation of 137Cs in a dose of 0.50 Gy under the conditions of their joint cultivation was performed. The frequency of chromosome aberrations in PBL from healthy individuals when co-cultured with the blood from CLL patients was 3.35 per 100 cells, exceeded the control (1.48 per 100 cells, р < 0.01), did not significantly differ from the rate in non-irradiated blood T-lymphocytes from patients with B-cell CLL (3.18 per 100 cells, р > 0.05) and was lower than when co-cultivated with irradiated blood from CLL patients (5.00 per 100 cells, р < 0.01). In irradiated in vitro blood lymphocytes from CLL patients, the mean group level of chromosome aberrations under separate cultivation was 12.36 per 100 cells and exceeded the indicator during their co-cultivation with lymphocytes from healthy individuals (8.35 per 100 cells, р < 0.05). A tumor-induced bystander effect (TIBE) develops in PBL from healthy individuals when co-cultured with the blood from CLL patients and in T lymphocytes of B-cell CLL patients, the cytogenetic manifestation of which is an increase in the frequency of single chromatid fragments. The interaction of irradiated blood cells from CLL patients with lymphocytes from healthy individuals causes an increase in chromosomal instability in the latter due to the development of a radiation-induced bystander effect (RIBE) and leads to a decrease in the level of chromosomal instability in irradiated lymphocytes from CLL patients (rescue effect). An increase in the level of chromatid-type aberrations in T-lymphocytes of CLL patients during in vitro irradiation is a consequence of the development of RIBE against the background of TIBE.

Study of Genotoxic Properties of Thiosonide

The objective of the study: evaluation of mutagenic properties of thiosonide in tests of chromosomal aberrations in mammalian bone marrow cells and mutation accounting on Salmonella typhimurium strains.Subjects and methods. Studies were carried out using F1 hybrids of CBA×C57BL/6 mice, they received thiozonide in the equivalent of a therapeutic dose and 160 times above it, once and in the course of 5 injections. The genetic toxicity of thiosonide was evaluated on Salmonella typhimurium strains in the tested doses of 1,000, 100, 10, 1 and 0.1 μg per plate.Results. No mutagenic activity of thiosonide in the chromosomal aberration counting test in bone marrow cells of mice was demonstrated. The level of chromosome aberrations in the experiment did not exceed the corresponding control values. Thus, thiosonide at the therapeutic dose (25 mg/kg) and 160-fold increase above it (4,000 mg/kg) did not induce chromosomal aberrations in the bone marrow cells of mice either with single dosing or several intragastric administrations.According to the Ames test results, the number of revertant colonies in the solvent control in the CM- and CM+ variants was within the range of spontaneous fluctuations for these strains; thiosonide in all tested doses did not show mutagenic effect on the TA 100, TA 98, and TA 97 strains both with the system metabolic activation and without it.

Polymorphism of nucleolar organizer regions in different Ukrainian cattle breeds

Aim. To study the activity of nucleolar organizer regions (NORs) in different Ukrainian cattle breeds in terms of their apparent activity status in silver stain and possible relation with milk productivity. Methods. Chromosome prepara- tions using lymphocytes from the peripheral blood of 90 cows of different breeds were used in the study. NOR activity was determined by visual evaluation of concentrations of silver precipitation on NORs in individual chromosomes. A 50 % silver nitrate solution was used to stain chromosome preparations. NORs were detected as dark spots on telomeres of the corresponding chromosomes. Results. The cytological analysis of chromosome preparations from lymphocytes of first lactation cows detected NOR polymorphism in Ukrainian Red-and-Motley dairy cattle (URM), Ukrainian Black-and-White dairy breed (UBW), and hybrid cows, obtained by crossing Ukrainian Red-and-Motley dairy breed and Montbeliarde bulls (URM × M). First lactation cows of URM and UBM had higher or the same inci- dence of cells with four (29.8 and 30 %) and five (17.1 and 19.5 %) NORs, while in URM × M cows the incidence of cells with the same number of NORs was almost twice lower; cells with 7 and 8 NORs exceeded a similar index for other investigated breeds almost twice (2.5 against 4.5 % and 2.0 against 4.2 %). The highest level of chromosomal aberrations (CA) was observed in the group of animals with medium number (2 to 3 NORs per cell), and the lowest – in the group with a high number of NORs (from 6 to 7) with a reliable intergroup difference (p &lt; 0.01). NOR activity was the highest in the group of animals of local origin (URM × M) with a milk yield over 7,000 kg in 305 days of the first lactation and the lowest in the UBW cows with a milk yield of 4–5,000 kg during the first lactation. Conclusions. We determined the differences in the activity of nucleolar organizers between the investigated groups of cows of dairy breed. URM × M hybrids reliably (р ≤ 0.05) exceeded dairy UBW cows by this index. No statistically significant different was found between other investigated groups of animals by this trait. Higher dairy productivity was found in the animals with higher frequency of NORs in the chromosomes of metaphase cells. In our opinion, the number of active NORs demonstrates relative variability between their number and the rate of protein synthesis, required to implement the productivity traits of the investigated animals.

Association of the DNA repair genes polymorphism with the frequency of chromosomal mutations and health status of the population of the Almaty region

The study utilized molecular-genetic, cytogenetic, and statistical analyzes methods. A cytogenetic and molecular genetic analysis of the inhabitants of the villages of Amangeldy, Belbulak, in whose territory warehouses of unused pesticides are located, was carried out. Residents of the Basshi village, on the territory of which there are no products of disposal of pesticides, were taken as control. Cytogenetic analysis showed a high level of chromosomal aberrations for the inhabitants of the Belbulak village, an increased level for the inhabitants of the Amangeldy village and a low level in the Basshi cohort. Molecular genetic analysis did not reveal a significant correlation between the polymorphisms of the XRCC1 Arg194Trp, XRCC3 Thr241Met and XPD Lys751Gln genes and the frequency of chromosomal aberrations. However, study determined the associative relationship of the polymorphism of the DNA repair gene XRCC1 Arg399Gln with the development of cardiovascular diseases in a population living near foci of pesticidal contamination.

Diacetyl is an aromatic popular synthetic fragrance that gives food a buttery taste used in ice cream, snacks and potting with butter, strawberry, caramel, or cheese flavor. The chromosomal aberrations and micronuclei are commonly used biomarkers of chromosomal damage, genome stability, and cancer risk assessment. In vivo trials are still important to assess the genetic toxicology of chemical products such as industrial chemicals, pharmaceuticals, and food additives. This study aimed at assessing the potential genotoxic effect of diacetyl and butter flavors on swiss albino mice using alterations in liver function enzymes, micronucleus (MN), and chromosomal aberrations (CA) assays. The results showed that exposure of swiss albino mice males to diacetyl and butter flavors induced (CA) and (MN) in a statistically highly significant manner compared to the control. Meanwhile, the biochemical analysis revealed that these substances caused an exceptional rise in liver function enzymes (AST, ALT, and ALP) activity in serum of treated experimental animals. In conclusion, both tested compounds have increased the chromosomal aberration, micronucleus test, and serum levels of liver function enzymes indicating their high potential of being cytotoxic and genotoxic materials.

Biological features of blood lymphocytes of the primary patients with endometrial cancer

The aim: to examine the radiosensitivity of chromosomes of T-lymphocytes in the blood of primary patients with endometrial cancer depending on the radiation dose. It was expected that the investigations would reveal a cytogenetic parameter as a predictor of radiosensitivity in non-malignant cells of patients exposed to curative irradiation.&#x0D; Materials and methods. Blood samples from 20 primary patients and 30 conditionally healthy donors were examined. Peripheral blood T-lymphocytes culture test system with metaphase chromosome aberration analysis was used. X-ray test-irradiation was performed at G0-stage of the cell cycle in the dose range of 0.5–3.0 Gy.&#x0D; Results. It was shown that the spontaneous level of chromosome aberrations in lymphocytes of primary patients before anti-tumour therapy is 7,82±0,33 aberrations/100 metaphases. This is more than 2-fold higher than the upper limit of average population index and approximately 6-fold higher than the data of own control. In our study during X-ray irradiation of cells cultures of patients, it was found for the first time that the total frequency of radiation-induced chromosome aberrations obeys the classical linear quadratic dose dependence with a predominance of linear component values; the frequency of radiation markers – also linear quadratic dose dependence, but with a predominance of quadratic component.&#x0D; Conclusions. High specificity of T-lymphocyte chromosomes to exposure to ionizing radiation as well as strict dependence of chromosome aberration yield on exposure dose justify their use as predictors of radiosensitivity of healthy cells from the tumour environment. The revealed dependences of induction of chromosomal damage in T-lymphocytes of patients with endometrial cancer prove the need for a personalized approach to plan the course of radiation therapy
