Changes of peripheral blood α-L-fucosidase activity in patients with rheumatoid arthritis: a cross-sectional study.

Rheumatoid arthritis (RA), a systemic autoimmune disease with approximately 1% prevalent population worldwide, which the etiology is still unclear. RA cannot be completely cured at present, which seriously affects the quality of life of patients. This study is to compare the peripheral blood α-L-fucosidase (AFU) between RA and healthy persons. A cross-sectional study was performed using total of 96 patients with RA served as case group and another 94 age-matched healthy volunteers served as a control group. AFU assay is detected by continuous monitoring method using Toshiba TBA-120FR (Tokyo, Japan) fully automatic biochemical analyzer in Japan, and the reagent is purchased from Zhejiang Quark Biological Company (Zhejiang, China). Statistical analysis was performed using SPSS 24.0 (SPSS, Inc., Chicago, IL, USA). AFU activity in peripheral blood of RA patients were lower than healthy controls. The higher AFU activity, the shorter the course of disease (r=-0.2790, P=0.0065). The activity of lactate dehydrogenase in patients with RA is higher than that of healthy control, but the activity of acetylcholinesterase is lower than that of normal people. Finally, AFU activity was negatively correlated with the activity of lactate dehydrogenase (r=-0.2381, P=0.0208) and positively correlated with the activity of acetylcholinesterase (r=0.2985, P=0.0035). Changes of peripheral blood AFU activity might be associated with progression of disease in RA patients. The changes of AFU activity may lead to disturbances in glucose and lipid metabolism.

Secondary hemophagocytic lymphohistiocytosis in pediatric patients with visceral leishmaniasis and Epstein-Barr virus infection.

Visceral leishmaniasis-associated hemophagocytic lymphohistiocytosis (VL-HLH) is indistinguishable from those of HLH of other etiologies due to the overlap symptoms, posing a serious threat to life. In this study, we aimed to provide insights for early diagnosis and improve outcomes in pediatric patients with VL-HLH. We retrospectively analyzed the clinical and laboratory data of 10 pediatric patients with VL-HLH and 58 pediatric patients with Epstein-Barr virus-associated hemophagocytic lymphohistiocytosis (EBV-HLH). The median time from symptom onset to cytopenia in patients with VL-HLH and EBV-HLH was 11 days (interquartile range, 7-15 days) and five days (interquartile range, 3.75-9.25 days) (P = 0.005). Both groups showed liver injury and increased lactate dehydrogenase levels; however the levels of aspartate aminotransferase, alanine aminotransferase, direct bilirubin, and lactate dehydrogenase in patients with VL-HLH were significantly lower than those in patients with EBV-HLH (P < 0.05). The fibrinogen and triglyceride levels were almost normal in VL-HLH patients but were significantly altered in EBV-HLH cases ( P < 0.05). The positive rate of first bone marrow microscopy examination, anti-rK39IgG detection, and blood metagenomic next-generation sequencing was 50%, 100%, and 100%, respectively. After VL diagnosis, eight patients were treated with sodium stibogluconate and two were treated with liposomal amphotericin B. All the patients with VL-HLH recovered. Our study demonstrates that regular triglyceride and fibrinogen levels in pediatric patients with VL-HLH may help in differential diagnosis from EBV-HLH. VL-HLH is milder than EBV-HLH, with less severe liver injury and inflammatory responses, and timely treatment with antileishmanial agents is essential to improve the outcomes of pediatric patients with VL-HLH.

Prediction of acute lung injury assessed by chest computed tomography, oxygen saturation/fraction of inspired oxygen ratio, and serum lactate dehydrogenase in patients with COVID-19

IntroductionIn treating acute hypoxemic respiratory failure (AHRF) caused by coronavirus disease 2019 (COVID-19), clinicians choose respiratory therapies such as low-flow nasal cannula oxygenation, high-flow nasal cannula oxygenation, or mechanical ventilation after assessment of the patient's condition. Chest computed tomography (CT) imaging contributes significantly to diagnosing COVID-19 pneumonia. However, the costs and potential harm to patients from radiation exposure need to be considered. This study was performed to predict the quantitative extent of COVID-19 acute lung injury using clinical indicators such as an oxygenation index and blood test results. MethodsWe analyzed data from 192 patients with COVID-19 AHRF. Multiple logistic regression was used to determine correlations between the lung infiltration volume (LIV) and other pathophysiological or biochemical laboratory parameters. ResultsAmong 13 clinical parameters, we identified the oxygen saturation/fraction of inspired oxygen ratio (SF ratio) and serum lactate dehydrogenase (LD) concentration as factors associated with the LIV. In the binary classification of an LIV of ≥20 % or not and with the borderline LD = 2.2 × [SF ratio]−182.4, the accuracy, precision, diagnostic odds ratio, and area under the summary receiver operating characteristic curve were 0.828, 0.818, 23.400, and 0.870, respectively. ConclusionsThese data suggest that acute lung injury due to COVID-19 pneumonia can be estimated using the SF ratio and LD concentration without a CT scan. These findings may provide significant clinical benefit by allowing clinicians to predict acute lung injury levels using simple, minimally invasive assessment of oxygenation capacity and biochemical blood tests.

Prognostic value of lactate dehydrogenase in patients with uveal melanoma treated with immune checkpoint inhibition.

We performed the meta-analysis to explore the predictive value of lactate dehydrogenase (LDH) levels in uveal melanoma (UM) patients receiving immune checkpoint inhibitors (ICIs). Eligible articles were obtained through EMBASE, PubMed, Google Scholar, and the Cochrane Library, until March 23, 2023. The clinical outcomes evaluated in this study encompassed overall survival (OS) and progression-free survival (PFS). This meta-analysis comprised eight studies with a combined total of 383 patients. The results showed that patients with high LDH levels had noticeably worse OS (HR: 3.445, 95% CI: 2.504-4.740, p < 0.001) and PFS (HR: 1.720, 95% CI: 1.429-2.070, p < 0.001). Subgroup analysis confirmed that the upper limit of normal was the ideal cut-off value for LDH. In multivariate analysis, we also found that high LDH levels significantly predicted shorter OS (HR: 3.405, 95% CI: 1.827-6.348, p < 0.001) and PFS (HR: 2.519, 95% CI: 1.557-4.076, p < 0.001) in UM patients. The sensitivity analysis and publication bias test supported the reliability of our results. In UM patients treated with ICIs, the LDH levels were reliable indicators of prognosis.

The Relationship between anosmia and ageusia with biochemical markers, severity, and duration of the disease in patients with COVID-19

Introduction: Due to the increasing prevalence of COVID-19 and its effects on the sense of taste and smell, we analyzed Blood electrolyte levels and biomarkers in COVID-19 patients who have a sign of anosmia and ageusia in Zanjan, Iran, and its relationship with biochemical blood indicators.&#x0D; Method: The retrospective study included all hospitalized patients with confirmed COVID-19. We registered laboratory parameters. A questionnaire that validity and reliability have already been confirmed was used to assess anosmia and ageusia. Statistical analysis was evaluated using a bivariate Bayesian logistic regression in the binomial distribution.&#x0D; Results: A total of 450 COVID-19 patients completed the study (221 females). The mean age of the patients was 56.36±17.34 years. 31.8% and 24.9% of patients reported anosmia and ageusia. There was no significant relationship between anosmia and ageusia with age, gender, place of hospitalization, marriage status, duration of hospitalization, and CT scan (p&lt;0.05). The Male's platelet was 18.72 lower than the female’s (p=0.002). Male's C-reactive protein was 4.96 units higher than female(p=0.002). In hospitalized persons for less than four days and people under 39 years of age, CRP levels were lower (P=0.001, P=0.019 respectively). The Levels of lactate dehydrogenase in patients with anosmia were 51.72 units less than in patients without anosmia (p=0.010).&#x0D; Conclusion: These results suggest that anosmia and ageusia are prevalent symptoms in Iranian COVID-19 patients. More information on serum biomarkers would help us to establish a greater degree of accuracy on this matter.

Анализировали симптомы заболевания, результаты лабораторных исследований крови, ликвора, активность лактатдегидрогеназы (ЛДГ) ликвора. Тяжесть больных с серозными менингитами оценена по шкале тяжести AMSS. Была установлена статистически достоверно высшая активность лактатдегидрогеназы ликвора у пациентов с серозными менингитами. Активность ЛДГ зависела от уровня цитоза ликвора, концентрации в ликворе белка, общего количества лимфоцитов в периферической крови, что указывает на возможность использования исследования активности ЛДГ ликвора в качестве дополнительного показателя в диагностике серозных менингитов у детей.

Prediction of Hospital Discharge Outcome from Changes in Cerebrospinal Fluid/serum Albumin Quotient and Cerebrospinal Fluid Lactate Dehydrogenase in Patients with Cryptococcal meningitis

Aim: This study aims to provide reliable prognostic factors for patients with cryptococcal meningitis (CM). Patients & methods: Clinical characteristics and laboratory findings of CM patients were retrospectively reviewed. Results: Sixty-three patients with CM were enrolled and 38/63 were confirmed to be HIV serology positive. Among clinical characteristics, headache, nausea and/or vomiting, and fever were the most common symptoms. Among cerebrospinal fluid (CSF) parameters, changes in leukocyte count, lactate dehydrogenase and chloride were significantly associated with theoutcome. An increased CSF/serum albumin quotient (QAlb) was indicative of an unfavorable outcome in HIV-negative patients. Conclusion: CSF lactate dehydrogenase and QAlb may improve the prediction of outcomes in patients with CM.

Association Between Body Mass Index and Admission Hemoglobin and Lactate Dehydrogenase in Patients Admitted for Sickle Cell Disease — A Retrospective Study

▪Introduction:Based on data in children, the presence of obesity may be associated with a milder variant of sickle cell disease. Data in children suggests that patients able to attain a normal or above normal body mass index (BMI) have better health outcomes than patients with low BMI. The association between BMI and severity of sickle disease in adults is less well studied. The present retrospective study aims to examine the association between increasing body mass index and markers of disease and flare severity in adults with sickle cell disease.Methods:A retrospective chart review was conducted at a tertiary care hospital from 2010 to 2016. Data on 156 patients was collected. Data collected included BMI, complete blood counts (on admission and baseline at prior visit), lactate dehydrogenase levels (LDH), serum chemistries and medications administered during hospitalization. BMI was examined as a continuous variable as well as categorical, with patients split into normal weight (BMI 20-25), underweight (BMI <20) and obese categories (BMI >25).Results:Of the 156 individuals included in the study, 29 (19%) were underweight, 81 (52%) were normal weight and 46 (29%) were obese. Increasing BMI was associated with increased admission hemoglobin (β=0.93;p=0.008) (Figure 1a), a higher value of lowest hemoglobin during hospitalization (β=0.10; p=0.023) and lower values of LDH (β=-19; p=0.025) (Figure 1b). As compared to underweight individuals, obese or normal weight individuals tended to have higher values of hemoglobin throughout hospitalization, as well as lower values of LDH, although this was not consistent across all study subgroups (Table 1).Discussion:The present study suggests that the presence of obesity, or indeed normal weight status is associated with improved laboratory parameters in adult patients with sickle cell disease. It is possible that obesity or normal weight status is a protective risk factor characterized by milder sickle crises. However, the presence of milder disease characteristics (as determined by higher hemoglobin and lower LDH levels) may represent a milder variant of sickle cell disease that “allows” patients to achieve normal or above normal body weights. A greater understanding of the mechanism underlying this phenomenon is required in order to determine optimal therapeutic strategies and clinical impact. The Predictive Value of Lactate Dehydrogenase in Patients with Chemoradiotherapy for Esophageal Squamous Cell Carcinoma

Increasing evidence has indicated that higher levels of lactate dehydrogenase (LDH) is associated with poor prognosis in various malignancies; however, the correlation between LDH levels and clinical outcomes in esophageal squamous cell carcinoma (ESCC) has been poorly studied. The objective of this study was to assess whether LDH has prognostic value in patients with ESCC undergoing chemoradiotherapy. A total of 614 ESCC patients were enrolled in this retrospective study from January 1, 2012 to December 31, 2016.The optimal cutoff points for LDH were determined by X-tile software. we analyzed the association between the levels of LDH and clinicopathological characteristics, as well as the relationship between LDH levels and responses. Kaplan-Meier method with log-rank test was used to explore the prognostic value for overall survival (OS) and progression-free survival (PFS). And then We examined the prognostic value for OS and PFS by univariate and multivariate Cox regression models. A 1:3 propensity score matching was used to compensate for differences in baseline characteristics. The optimal cutoff points of LDH was 134U/L. The median OS in the high LDH group was 31.3 months, and more than 50% of patients with low LDH were still survive to the last follow-up (P = 0.020).The median PFS of low LDH and high LDH were 40.1 and 18.0 months, respectively (P = 0.010).Gender,Cyfra21-1,tumor length, cN/M stage, treatment options and LDH were independent prognostic indicators for OS. Gender, Cyfra21-1, tumor length, cN/M stage, treatment options, chemotherapy cycles, and LDH were independent prognostic indicators for PFS. Unfortunately, there was no significant difference for OS and PFS by Kaplan–Meier method after matching, but the survival curve showed a tendency to separate between the two groups. The present study revealed the existence of a significant association between the pretreatment serum LDH level and OS in patients with ESCC receiving treatment with chemoradiotherapy as well as a significant association with PFS. It implied that the management of patients with high LDH levels should be changed. However, it is worth noting that this conclusion was not certified after matching. Although the consistent conclusions were not obtained in predicting the prognosis of ESCC patients with LDH before and after matching, a multicenter, large-sample prospective study is needed to further verify the conclusions of this study before this method can be applied to conventional clinical studies.

COVID-19 and the World with Co-Morbidities of Heart Disease, Hypertension and Diabetes

Newly emerging severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) causing coronavirus disease 2019 (COVID-19) pandemic has now spread across the globe in past few months while affecting 26 million people and leading to more than 0.85 million deaths as on 2nd September, 2020. Severity of SARS-CoV-2 infection increases in COVID-19 patients due to pre-existing health co-morbidities. This mini-review has focused on the three significant co-morbidities viz., heart disease, hypertension, and diabetes, which are posing high health concerns and increased mortality during this ongoing pandemic. The observed co-morbidities have been found to be associated with the increasing risk factors for SARS-CoV-2 infection and COVID-19 critical illness as well as to be associated positively with the worsening of the health condition of COVID-19 suffering individuals resulting in the high risk for mortality. SARS-CoV-2 enters host cell via angiotensin-converting enzyme 2 receptors. Regulation of crucial cardiovascular functions and metabolisms like blood pressure and sugar levels are being carried out by ACE2. This might be one of the reasons that contribute to the higher mortality in COVID-19 patients having co-morbidities. Clinical investigations have identified higher levels of creatinine, cardiac troponin I, alanine aminotransferase, NT-proBNP, creatine kinase, D-dimer, aspartate aminotransferase and lactate dehydrogenase in patients who have succumbed to death from COVID-19 as compared to recovered individuals. More investigations are required to identify the modes behind increased mortality in COVID-19 patients having co-morbidities of heart disease, hypertension, and diabetes. This will enable us to design and develop suitable therapeutic strategies for reducing the mortality. More attention and critical care need to be paid to such high risk patients suffering from co-morbidities during COVID-19 pandemic.

The diagnostic value of serum C-reactive protein, procalcitonin, interleukin-6 and lactate dehydrogenase in patients with severe acute pancreatitis

ObjectiveAcute pancreatitis (AP) is an inflammatory disease with rapid progression. In severe cases, it can cause systemic inflammatory response syndrome (SIRS), multiple organ failure (POF) and even death. The study aimed to investigate the diagnostic value of C-reactive protein (CRP), procalcitonin (PCT), interleukin-6 (IL-6) and lactate dehydrogenase (LDH) in patients with severe AP. MethodsAP patients (n = 153) divided into mild AP patients (n = 81) and severe AP patients (n = 72) were selected from June 2014 to June 2016. The demographic information (age, sex) and the hematological parameters (WBC, PLT, CRP, PCT, IL-6, LDH and so on) were analyzed. ResultsSignificant differences were found out of CRP, PCT, IL-6 and LDH values between AP patients and controls (P < 0.05), even those results had significant difference between MAP group and SAP group (P < 0.05). In SAP group, the cut-off values of CRP, PCT, IL-6 and LDH were 16.62, 2.29, 16.66, 273.04; sensitivity 55.6%, 77.8%, 80.2%, 82.7%; specificity 73%, 94%, 85%, 96% and AUC 0.637, 0.929, 0.886, 0.919, respectively. The AUC of combined detection of CRP, PCT, IL-6 and LDH was 0.989 (95%CI). ConclusionThe combined detection of CRP, PCT, IL-6 and LDH has a high diagnostic value for judging the severity of AP.

Prognostic significance of serum lactate dehydrogenase in patients undergoing radical cystectomy for bladder cancer

BackgroundTo investigate the prognostic significance of preoperative serum lactate dehydrogenase (LDH) in patients undergoing radical cystectomy for bladder cancer (BCa). Patients and methodsA cohort of 263 patients undergoing open or laparoscopic radical cystectomy between 2011 and 2016 was studied. Baseline characteristics, hematological variables, follow-up data were collected. Kaplan-Meier curves and Cox proportional hazard regression model were applied to assess the relationship between LDH and overall survival (OS), cancer-specific survival (CSS), and disease-free survival (DFS). ResultsAfter a median 34.2 (22.9–45.8) months follow-up, all-cause death, cancer-specific death, and disease recurrence occurred in 66 patients, 50 patients, and 91 patients. The elevation of serum LDH was associated with several unfavorable parameters, including advanced age, continent cutaneous urinary diversion, increased neutrophil-to-lymphocyte ratio, decreased lymphocyte-to-monocyte ratio. Patients with a higher serum LDH (> 220 U/L) had a worse OS (P < 0.001), CSS (P < 0.001) and DFS (P < 0.001). Multivariate Cox analysis suggested that elevated LDH was an independent predictor for OS (hazard ratio [HR]: 3.113, 95% confidence interval [CI]: 1.524–6.358; P = 0.002), CSS (HR: 4.564, 95% CI: 2.008–10.373; P < 0.001), DFS (HR: 2.051, 95% CI: 1.125–3.739; P = 0.019). Medical history of diabetes, high pT stage, and positive lymph node also were adverse predictors for oncological outcomes of BCa patients in multivariate analysis. ConclusionsPreoperative serum LDH is an independent prognostic biomarker for OS, CSS, and DFS in patients undergoing radical cystectomy for BCa, which can be incorporated into prognostic models.

Long non-coding RNA T-cell factor 7 in multiple myeloma: A potential biomarker for deteriorated clinical features and poor prognosis.

BackgroundThis study aimed to investigate the correlation of long non‐coding RNA T‐cell factor 7 (lnc‐TCF7) with clinical features and prognosis in patients with multiple myeloma (MM).MethodsTotally, 216 newly diagnosed symptomatic MM patients and 60 healthy controls (HCs) were enrolled. Bone marrow samples were collected from patients before treatment and from HCs on donation to detect lnc‐TCF7 expression in plasma cells by reverse transcription quantitative polymerase chain reaction. Besides, clinical response, progression‐free survival (PFS), and overall survival (OS) of patients were assessed.ResultsLnc‐TCF7 expression was increased in patients with MM compared with HCs. Lnc‐TCF7 expression was highest in international staging system (ISS) stage III patients, followed by ISS stage II patients, and then ISS stage I patients, while lnc‐TCF7 expression was similar in patients with different immunoglobulin subtypes and Durie‐Salmon stages. Regarding chromosomal abnormalities, lnc‐TCF7 expression positively correlated with t(4; 14) and Del(17p), whereas no correlation of lnc‐TCF7 expression with t(14; 16), 1q21 amplification, Del(13q), or hyperdiploid was observed in patients with MM. Furthermore, lnc‐TCF7 expression positively correlated with serum creatinine, beta‐2‐microglobulin, and lactate dehydrogenase in patients. Besides, lnc‐TCF7 was negatively associated with complete response but not overall response rate in patients. Additionally, patients with lnc‐TCF7 high expression exhibited shorter PFS and OS compared to patients with lnc‐TCF7 low expression.ConclusionLnc‐TCF7 might have clinical value in aiding disease management and prognosis prediction of MM.

Prognostic role of pretreatment lactate dehydrogenase in patients with metastatic renal cell carcinoma: A systematic review and meta-analysis.

To date, there remain uncertainties over the prognostic role of serum lactate dehydrogenase (LDH) in patients with metastatic renal cell carcinoma (mRCC). A systematic review and meta-analysis was performed. Eligible studies were retrieved from PubMed, Embase, Cochrane Library and Web of Science databases up to October 2019. The endpoints included overall survival (OS) and progression-free survival (PFS). Multivariable adjusted hazard ratios (HRs) and 95% confidence intervals (CIs) were used to evaluate each endpoint. Thirty observational studies of low to moderate risk of bias embracing 6754 patients with mRCC were included. The results showed that patients with a high pretreatment serum LDH had an inferior OS (HR: 2.15, 95% CI: 1.85-2.51; P<0.001) and PFS (HR: 1.76, 95% CI: 1.49-2.10; P<0.001). Subgroup analyses according to year of publication, study design, patient population, geographic region, sample size and NOS score did not alter the direction of results. There was significant publication bias for OS, but not for PFS. Sensitivity analyses further confirmed the robustness of the results. Our findings indicated that a high level of pretreatment serum LDH was associated with an inferior OS and DFS in patients with mRCC. Methodological limitations should be considered while interpreting these results.

Comparison of Salivary and Serum Alkaline Phosphates Level and Lactate Dehydrogenase Levels in Patients with Tobacco Related Oral Lesions with Healthy Subjects - A Step Towards Early Diagnosis

Aim:To evaluate and compare salivary and serum levels of Alkaline Phosphates and Lactate Dehydrogenase in patients without the habit of tobacco, in patients with the habit of tobacco, in patients with benign oral lesions and in patients with oral premalignant lesions and oral malignant lesions. Material and Methodology:This study was comprised of 500 subjects, Group I: 100 healthy individuals without the habit of tobacco usage formed the control group. Group II: 100 patients with the habit of tobacco/ smoking consumption without any oral lesion. Group III: 100 patients with benign oral lesions. Group IV: 100 patients having the history of tobacco consumption and having apparent precancerous lesions like leukoplakia, erythroplakia. Group V:100 patients having frank oral cancer. The grade of dysplasia in these patients was statically correlated with the levels of serum and salivary ALP and LDH. Results:This study revealed that there was high expression of both serum and salivary ALP and LDH in group IV and Group V as compared with the other groups and mean difference showed a statistically significant p value of less than 0.01. This study revealed that the in group V, the highest level of serum and salivary ALP was found in those patients who were reported with poorly differentiated oral cancer. Conclusion:Both Alkaline phosphates and Lactate dehydrogenase could be considered a sensitive markers for the detection of dysplasia with already existing precancancerous and cancerous lesions.

Prognostic Significance of Lactate Dehydrogenase in Patients Undergoing Surgical Resection for Laryngeal Squamous Cell Carcinoma

The aim is to estimate the prognostic value of lactate dehydrogenase (LDH) in patients undergoing surgical resection for laryngeal squamous cell carcinoma (LSCC). A total of 640 resected LSCC patients were included. Preoperative lactate dehydrogenase (LDH) was assessed. Kaplan-Meier survival analysis and Cox regression analysis were conducted for overall survival (OS) and recurrence-free survival (RFS). Kaplan-Meier analysis, univariate analysis and multivariate analysis demonstrated significant prognostic value for preoperative LDH. Although LDH was predictor of OS, it failed to be a predictor of RFS. The univariate HR and 95% CI of LDH were 0.484 and 0.357-0.658 (P < 0.0001). The multivariate analysis showed that LDH (HR = 0.518, 95% CI: 0.380-0.705, p < 0.0001) was related to OS. Elevated preoperative LDH >132 IU/L was significantly associated with better survival. Preoperative LDH might be an independent prognostic marker of OS in LSCC patients undergoing surgical resection.

Association of PFS with levels of pretreatment lactate dehydrogenase in patients with EML4-ALK rearrangement non-small cell lung cancer treated with ALK tyrosine kinase inhibitor.

e20513 Background: We performed a retrospective analysis to investigate the association between the lactate dehydrogenase (LDH) levels and progression-free survival (PFS) in patients with echinoderm microtubule-associated protein-like 4-anaplasticlymphoma kinase (EML4-ALK) rearrangement non-small cell lung cancer (NSCLC) receiving treatment with crizotinib. Methods: Advanced NSCLC patients with EML4-ALK rearrangement receiving treatment with crizotinib were enrolled at Peking Union Medical College and Cancer Hospital Chinese Academy of Medical Sciences between January 2007 and January 2016. Pre-treatment or post-treatment serum LDH levels were analyzed with progression-free survival (PFS) and patients’ clinical parameters. Results: Overall, 212 patients were studied. Kaplan-Meier univariate analysis showed that elevated pre-treatment LDH level (7.9 vs. 14.1 months, P = 0.004) were associated with PFS, while the mean value of post-treatment LDH level (14.3 vs. 13.3 months, P = 0.970) were not associated with PFS. Coxproportional hazards model also identified that pre-treatment LDH level (hazard ratio [HR] = 1.841, 95% confidence interval [CI] 1.062-3.190, P = 0.030) was associated with the PFS. Logistic regression analysis showed that post-treatment LDH level was associated with creatine kinase(OR = 6.712, 95% CI 3.395-13.273, P &lt; 0.01), CKMB (OR = 6.297, 95% CI 2.953-13.427, P &lt; 0.01), and hemoglobin(OR = 4.163, 1.741-9.956, P = 0.001). Conclusions: An elevated pre-treatment serum LDH level ( &gt; 250U/L) is significantly associated with shorter PFS in patients with EML4-ALK rearrangement NSCLC. Post-treatment elevated serum LDH level is associated with multiple factors including muscle damage and anemia, rather than PFS.
