Abstract 5348: Impaired self-renewal and colitis susceptibility of colonic mucosa in mice lacking AKR1B8

Abstract Background: Ulcerative colitis (UC) is a major clinically defined form of inflammatory bowel disease which affects individuals throughout a life and associates with an increased risk of developing colorectal cancer. Aldo-keto reductase 1B10 (AKR1B10), isolated by our laboratory, is a novel monomeric NADPH-dependent enzyme with reductive activity towards reactive carbonyls. AKR1B10 also acts as a novel regulator of de novo fatty acid synthesis and lipid metabolism via regulating acetyl-CoA carboxylase-α (ACCA) stability. AKR1B10 is primarily expressed in normal human intestines, but lost in colorectal cancer and predisposition conditions, such as UC and adenomatous polyps, suggesting its pathogenic role in the these diseases. Aldo-keto reductase 1B8 (AKR1B8) is the mouse ortholog of human AKR1B10, having the similar enzyme activity to α,β-unsaturated carbonyls and capability of regulating fatty acid synthesis and lipid metabolism in mice. Thereby, we generated an AKR1B8 knockout (-/-) mouse strain to understand the etiopathogenic role of AKR1B10/8 in the UC and associated colorectal cancer to Methods: AKR1B8 -/- mouse strain was produced by homozygous recombination. Dextran sulfate sodium (DSS) was utilized to induce colitis and associated colorectal tumors. In detail, AKR1B8 -/- mice were fed with 2% DSS in drink water for four cycles (e.g., 7 days DSS, 14 days water). Inflammatory activity and masses were recorded, and histopathology of chronic colitis, crypt architecture, and micro-neoplastic lesions was examined. Profiles of multiple cytokines and reactive oxidative species in colonic mucosa were estimated. Potential DNA mutations were identified using whole Exome sequencing. Results: Targeted disruption of AKR1B8 disturbed the self-renewal and injury recovery of colonic cryptic cells due to decrease in lipid synthesis from butyrate. AKR1B8 deficient mice are highly susceptible to colitis induced by DSS at 2% and to carbonyl and oxidative DNA damage, leading to colitis-associated tumorigenesis and malignant progression. Histopathological studies showed that AKR1B8 -/- mice suffered severe inflammatory lesions and high-grade dysplasia. Importantly, visible large masses were seen in AKR1B8 -/- mice, but not in wild type that underwent the same DSS treatment. Furthermore, 269 gene variants (in exons) were identified uniquely in AKR1B8 -/- colitis mucosa and tumors, not in WT colitis. Among them, 20 genes were mutated in colitis only and 28 genes in tumors alone. A total of 221 genes were mutated in both AKR1B8 -/- colitis mucosa and tumors. These gene mutants may represent the DNA damage that drives the malignant transition of colitis. Conclusions: AKR1B8 (AKR1B10 in humans) is a critical protein that regulates the dynamic homeostasis of self-renewal and barrier function of colonic epithelium. AKR1B8 deficiency in colonic epithelium is a critical etiopathogenic factor of UC and its tumorigenic transformation. Note: This abstract was not presented at the meeting. Effect of Ru Dopant Amount on the Electrocatalytic Capability of Ti/SnO2-Sb-Gd-Ru Electrodes in the Phenol Oxidation

SnO2 electrodes doped with Sb have been used in the process of anodic oxidation of contaminants organic due to its electrochemical properties, high performance and its low cost [1]. Recent studies have found that by introducing elements of rare earths, especially Gd in Ti/SnO2-Sb electrodes, favors a be tter performance in the catalytic activity for the electrooxidation of phenol [2], however, the main problem presented by this type of electrode is its short life time which limits its applications to industrial wastewater treatment. Therefore, increase the active life time of these anodes has a significant importance to develop treatment technologies of organic contaminants in wastewater. Different investigations have been directed to extend the useful life of SnO2-Sb anodes incorporating dopants metallic, as platinum, ruthenium [3] and Iridium or by the introduction of a interlayer of IrO2, RuO2 [4] between the surface of Ti and the SnO2-Sb film. The results reported indicate that the life time is increased, although decreases the electrocatalytic activity for pollutant elimination due to that reduces the potential of the oxygen evolution reaction (OER). This paper reports the study of influence of the Ru content on the electrocatalytic capability of Ti/SnO2-Sb(5%)-Gd(1%) electrodes in the electrooxidation of phenol.The electrodes were synthesized using the Pechini method [5] and calcined at 600° C. The relationship molar acid ethylene glycol/citric/metals was 3/14/1, respectively. The molar relations of Sn/Sb(5%) and Sn/Gd(1%) were constants and Ru amount was variable: Sn/Ru(0, 0.25, 0.5, 0.75%). The obtained electrodes were characterized by X-ray, microscopy electron scanning (SEM), photoelectron spectroscopy (XPS). The electrochemical characterization of electrodes was carried out by cyclic voltammetry, determination of Tafel slope with respect to the oxygen evolution reaction, the roughness factor and accelerated life tests. The electrocatalytic activity of electrodes was evaluated with respect to the electrooxidation of a phenol solution (100 ppm) to constant current (60 mA). The phenol solution was analyzed during the electrolysis by UV-Vis spectroscopy, liquid chromatography high efficiency (HPLC) and total organic carbon (TOC).The results indicate that the incorporation of Ru in the system Ti/SnO2-Sb-Gd, induces changes in crystallinity and morphology of the material, also modifies the antimony content and the oxygen content in the SnO2 network on the surface of the film. The percentages of Sb+5 were much higher on the surface for Sn/Ru(0.25%) in comparison to others electrode materials. The presence of Ru slightly modifies the potential onset of the oxygen evolution reaction (OER). Therefore, with regard to the system Ti/SnO2-Sb(5%)-Gd(1%), the addition of small amounts of Ru does not produce a substantial effect on variation in phenol degradation and TOC removal as shown in Fig. 1, in addition, the presence of Ru increases the life time of the electrodes. Synthesis and Characterization of Flexible and Free-Standing Carbon Nanotube, Graphene Films and Their Composites for Li-Ion Batteries

The objective of this communication is to present our results on the synthesis and the characterization of flexible and free standing films made from Carbon Nanotube (CNT), reduced graphene oxide (rGO) [1] and their composites (CNT/rGO). The films were obtained by filtration method from aqueous suspension of CNT, rGO, or CNT/rGO. The obtained materials are used as negative electrode in Li-ion systems. CNT films with a thickness below 50 µm are integrated on an electrodeposited Cu current collector (15 µm thick) according to the method describe by Luais et al [2], whereas CNT or CNT/rGO flexible free standing films with a thickness of 50 µm can be directly used without the need of a metallic current collector as anode material for battery cycling. All results will be discussed on the basis of SEM observations (as synthetized films and after cycling for post-mortem characterizations), Raman spectroscopy analysis, galvanostatic cycling, and electrochemical impedance spectroscopy. CNT free standing electrodes exhibit specific discharge capacities up to 300 mA.h.g-1 at C/20 and 200 mA.h.g-1at 5C. The effect of rGO/CNT ratio in composite electrodes will be presented as an improvement of the electrochemical performances compared to CNT free standing electrodes. Abstract LB-79: Combined treatment with HER3 antibody MM-121/SAR 256212 and EGFR antibody cetuximab in pre-clinical models of head and neck cancer.

Abstract Background: The EGFR monoclonal antibody cetuximab is approved for treating head and neck squamous cell cancer (SCCHN) in combination with radiation therapy for locally advanced disease and chemotherapy for recurrent and metastatic disease. Intrinsic or compensatory HER3 signaling and sustained PI3K/Akt activation may play a role in resistance to EGFR-targeted therapy. However, the role of HER3 in resistance to EGFR-targeted therapy in SCCHN has not been elucidated. Therefore, we first tested whether the combination of MM-121/SAR 256212 (M), a HER3 antibody being co-developed by Merrimack Pharmaceuticals and Sanofi, with cetuximab (C) may be more effective than M or C alone in treatment of SCCHN. Methods: We screened for the expression of EGFR, HER3 and their activated forms in 12 SCCHN cell lines. We then evaluated the activity of C, M, and their combination (CM) both in vitro by colony formation assay and in vivo by using SCCHN xenograft models (Tu212 and SCC47). In the in vivo experiment, mice were treated by intraperitoneal injection (i.p.) of C, M or CM twice per week for 4 weeks. The experimental groups include a PBS control, C (6.25ug/dose), M at low dose (300ug/dose, LD), M at high dose (600ug/dose, HD), the combination with LD (CMLD), and the combination with HD (CMHD). In addition, we examined the effect of M and C on HER3 and its downstream pathways in SCCHN cell lines by western blot assay. Results: pHER3 was detected in 9/12 while pEGFR, EGFR and HER3 were expressed in all of 12 SCCHN cell lines. The colony formation assay showed that Tu212 cells were significantly inhibited by CM compared to the control (p <0.001), M (p<0.001) and C (p=0.009) alone. Similar results were also observed using the SCC47 cell line. Western blot analysis confirmed the down-regulation of activated HER3 and its downstream phospho-Akt and phospho-S6 ribosomal protein by CM. Our in vivo study showed significant tumor growth inhibition in both MM-121 LD (p<0.001) and MM-121HD (p<0.001) compared to the control. Moreover, the treatment with both CMLD and CMHD significantly suppressed Tu212 xenograft tumor growth compared to the PBS control (both p<0.0001), C alone (both p<0.0001), and MLD (p=0.0046 and 0.0008, respectively), but only CMHD showed significant inhibition compared to MHD (p = 0.02). There was no significant difference between CMLD and CMHD (p=0.5994). Studies using another SCCHN xenograft model (SCC47) showed a similar inhibitory effect from the treatments by C, M and CM. Conclusion: This study shows that the combination of cetuximab and MM-121 is significantly more active than MM-121 or cetuximab alone in models of head and neck cancer in which both EGFR and HER3 are activated. 585: Distinct vaginal microbiome profiles are associated with host mitochondrial DNA (mtDNA) haplogroup and SNP variants

diac function in a mouse model of IUGR. STUDY DESIGN: Laparotomy was performed on pregnant C57BL/6J mice at gestational day 18 and pups were divided into 3 groups (n 12). Control: Sham operated; IUGR surgically induced by ligation of a branch of the uterine artery; IGF1Treated by intra placental injection of Ad hIGF1 after ligation. Pups were delivered on day 20, cross-fostered to CD1 mice, sorted by gender and at week 8 a western high fat diet introduced. Cross-sectional, 2D and Doppler transthoracic echocardiography was performed at 12 weeks evaluating ejection fraction (EF), fractional shortening (FS) and left ventricular (LV) dimensions. Data were analyzed using ANOVA. RESULTS: IUGR mice demonstrated significantly reduced EF (43.5 8.3 vs 60 5.1vs 59 5.7,p 0.001) and FS (22 4.6 vs 32.2 3.2 vs 31.3 3.7,p 0.001) compared to SHAM that was normalized in the treatment group (FigA). IUGR significantly increased LVID;S (3.6 0.4vs3.1 0.2vs2.9 0.2,p 0.001) as compared to SHAM, restored to normal in IGF1 Treated (FigB). These findings were the same when data was stratified by sex. CONCLUSION: Intra-placental gene transfer of Ad hIGF1 rescues IUGR induced cardiac dysfunction in mice exposed to high fat diet. The synonymous point mutation T6776C demonstrated strong association with microbial encoded fatty acid metabolism pathways (right panels). CONCLUSION: Several Caucasion haplogroups and mtDNA variants show significant association with specific microbial taxa and metabolic pathways from the posterior fornix of the vagina. We speculate that these findings may partially explain infectious, microbiome and host associations in perinatal disorders.

Abstract LB-156: The role of AKR1B10 in early tumorigenesis depends on AMP-activated protein Kinase-α checkpoint

Abstract The role of AKR1B10 in early tumorigenesis depends on AMP-activated protein Kinase-α checkpoint Jun Ma and Deliang Cao 1Department of Medical Microbiology, Immunology, & Cell Biology, Simmons Cancer Institute, Southern Illinois University School of Medicine. 913 N. Rutledge Street, Springfield, IL 62794. Aldo-keto reductase 1B10 (AKR1B10) is upregulated in 71.4% ductal carcinoma in situ (DCIS) and 83.6% invasive breast cancer. Furthermore, AKR1B10 expression is correlated significantly with tumor size, node status and disease-related survival. In malignant cells, AKR1B10 promotes cell growth and survival via provoking cellular lipid synthesis and eliminating cytotoxic α, β-unsaturated carbonyl compounds. Therefore, AKR1B10 may play an oncogenic role in tumor growth and progression. This study is aimed to explore the role of AKR1B10 in early tumorigenesis. Interestingly, our data show that the expression of AKR1B10 in MCF10A cell, a normal breast epithelium cell line, led to elevation of p53 levels and premature senescence. However, in RAO-1 cells, an hTERT-immortalized breast epithelium cell line, AKR1B10 expression promoted glucose uptake, glycolysis and fatty acid synthesis, and subsequent cell growth instead. Further studies revealed that AMP-activated protein kinase-alpha (AMPKα), which is an important metabolic barrier during cell transformation, was activated upon AKR1B10 expression in both cell lines. On the other hand, down-regulation of AMPKα by shRNA rescued AKR1B10-induced senescence in MCF10A while partially abrogated pro-metabolism effect of AKR1B10 in RAO-1 cells and caused cell growth inhibition. In summary, the role of AKR1B10 in early tumorigenesis depended on AMPKα checkpoint. When this checkpoint is intact, AKR1B10 activates AMPKα and triggers p53 activation and consequential cellular senescence. On the contrary, when this checkpoint is dysfunctional, such as in cells with constitutive expression of telomerase, AKR1B10 activates AMPKα, enhances glucose and lipid metabolism and promotes cell proliferation. Pollution Data Uneven Across China

Regulations on environmental disclosure adopted by China three years ago have improved the behavior of polluting companies, claims a Beijing-based environmental group. But the new rules are unevenly followed across China, with municipal governments in the eastern part of the country more willing to disclose environmental data than are authorities in the west. In May 2008, the central government in Beijing adopted two regulations that improve public access to official environmental data. One law was implemented by the State Council, China’s top decision-making body. The other was adopted by the country’s Ministry of Environmental Protection. Among other obligations, cities now have to let citizens see what complaints were filed against companies and what follow-up actions, if any, authorities have taken. Use of dietary supplements among US adults with asthma

Methods: Data from the 2005–2006 National Health and Nutrition Examination Survey were analyzed. Asthma was defined by self-report of ever receiving a diagnosis and still having asthma currently. Dietary supplements were collected by direct inspection of products where possible (88% of the time), and were transcribed by trained nutritionists. Multivariate logistic procedure was performed to determine independent associations of supplement use with participant characteristics. Results: The prevalence of using a dietary supplement in the past month was similar between adults with asthma (50.2%, 8.6 million) and those without asthma (54%, 104.3 million). Among asthmatic adults, multivitamins/multiminerals (40.1%) were the most commonly used supplements, followed by vitamin B 12 (23.3%), vitamin C (19.9%), calcium (15.2%), vitamin E (14.7), folic acid (12.7%), vitamin B 6 (11.2%), thiamin (10.8%), niacin (10.8%), B complex vitamins (10.4%), and riboflavin (10.3%). Asthma adults who were supplement users tended to be older (50.2 ± 19.2 versus 45.2 ± 18.0 years for supplement users versus nonusers, respectively, P = 0.009), white/Hispanic, women, and had excellent/very good self-reported health. Forty-three percent of supplement users took only one supplement. Middle-aged and older asthma adults were more likely to use multiple supplements concurrently. Conclusion: Use of dietary supplements is popular in US adults with asthma, similar to the general adult population. It is important to take supplement use into account in clinical care for asthma patients and in the design and analysis in epidemiological and intervention studies of nutrition and asthma.

It was found that the degradation of phenolic EDCs is promoted to various extents by the addition of humic acid. Results show that the removal rate of estrone by permanganate within 3 min of reaction time is increased from approximately 10% in control experiments to 68% when 0.08 mg l−1 of humic acid is present in the reaction system. The oxidation experiment of nine types of phenolic EDC by permanganate confirms that the promotion induced by the addition of humic acid varies in the cases of different EDCS. Results also indicate that the promotion induced by the presence of humic acid takes effect under acidic and neutral pH conditions, while the effect of pH plays a dominant role for the degradation of phenolic EDCs under alkaline conditions. Differring from previous studies, which mainly focused on optimizing water treatment processes to enhance algae cells removal, this study paid more attention to the effect of algae population characteristics on their removal efficiency during conventional water treatment processes. The results showed that the size, shape, surface structures and the activity of algae cells affected the algal removal efficiency. The remaining algae species in the effluent of coagulation–filtration processes were mainly micro-algae. The smaller the size of algae cells was, the lower was the removal efficiency. The special shapes of algae cells, such as ellipsoidal, bacilliform and piece, and special surface structures, such as thorns and flagellae, might have a negative impact on algae removal. Moreover, the diversity of algae species in water also affected the efficiencies of micro-algae cell removal. The more the diversity of algae species were, the higher were the algae removal efficiencies.
