Sustainable Isolation of Bioactive Compounds and Proteins from Plant-Based Food (and Byproducts).

Plant-based food produces significantly less greenhouse gases, and due to its wealth of bioactive components and/or plant-based protein, it becomes an alternative in a sustainable food system. However, the processing and production of products from plant sources creates byproducts, which can be waste or a source of useful substances that can be reused. The waste produced during the production and processing of food is essentially nutrient- and energy-rich, and it is recognized as an excellent source of secondary raw materials that could be repurposed in the process of manufacturing and preparing food, or as feed for livestock. This review offers an overview of the sources and techniques of the sustainable isolation of bioactive substances and proteins from various sources that might represent waste in the preparation or production of food of plant origin. The aim is to uncover novel approaches to use waste and byproducts from the process of making food to provide this waste food an additional benefit, not forgetting the expectations of the end user, the consumer. For the successful isolation of bioactive ingredients and proteins from food of plant origin, it is crucial to develop more eco-friendly and efficient extraction techniques with a low CO2 footprint while considering the economic aspects.

Bioassay-guided isolation and in silico study of antihyperlipidemic compounds from Onosma hispidum Wall

Isolation of bioactive compounds from plants and their therapeutic evaluation is crucial in the pursuit of novel phytochemicals and contributes an indispensable role in drug discovery and design. The literature has documented the hypolipidemic effect of numerous Onosma species. Taking that into consideration, the current study was designed to isolate, purify and evaluate the antihyperlipidemic potential of leaves of Onosma hispidum Wall. For the first time, the bioassay-guided isolation led to the separation of 3 compounds that were identified by spectroscopic techniques as o-phthalic acid bis-(2-ethyl decyl)-ester (1), bis (2-ethyloctyl) phthalate (2), and 1,2 benzenedicarboxylic acid bis(2-methyl heptyl) ester (3). Lipase inhibition assay was utilized to scrutinize the antihyperlipidemic potential of methanolic extract fractions and subsequently isolated compounds. Further, the isolated compounds were employed for in silico studies via molecular docking, molecular mechanics with generalized born and surface area solvation (MM-GBSA), and MD simulations with Pancreatic Lipase Colipase (PDB ID: 1LPB). Molecular docking and MM-GBSA of isolated compounds were employed to explain the mode of binding between the protein–ligand complex and binding free energy calculation, respectively. Since compound (3) displayed the best docking score of −6.689 kcal/mol as compared to orlistat −5.529 kcal/mol with PDB: 1LPB. So, it was chosen for MD simulations to evaluate ligand stability and flexibility of the complex which was validated by the fluctuation of α-carbon chain, root mean square deviation (RMSD), root mean square fluctuation (RMSF), and type of interactions involved which authenticated the in vitro lipase inhibitory potential. Overall, in silico and in vitro results validated that compound (3) could be exploited as a promising pancreatic lipase inhibitor. Communicated by Ramaswamy H. Sarma

Gas Chromatography-Mass Spectrometry Analysis, Phytochemical Screening, Antioxidant and Antibacterial Activity of Methanol Root Extract of Rhaphiostylis beninensis (Hook.f.) Planch. [family ICACINACEAE] against Uropathogens

In Nigeria, urinary tract infections (UTIs) are amid the most prevalent bacterial contagions, with a higher incidence rate in women. Depending on the severity of the infection, different antibiotics are used to treat UTIs. Herbal remedies have, on the other hand, been seen as the paranormal means of treating UTIs while antibiotic resistance has emerged as a problem for medics. This study examined the methanol extract of Rhaphiostylis beninensis (MRB) for its phytochemical and bioactive components, antioxidant, and antibacterial activities. From the study, the methanol extract's antioxidant activities grew stronger as concentration increased. Alkaloids, saponin, tannins, cardiac glycosides, flavonoids, and reducing sugar—giving the extract its pharmacological potential—were among the phytochemicals identified from the phytochemical screening. The GC-MS Chromatogram showed that there are 42 bioactive compounds in the MRB with the most prominent compounds being (Z,Z)-9,12-Octadecadienoic acid 19.96%, Lupeol 18.96%, cis-13-Octadecenoic acid 9.96%, n-Hexadecanoic acid 9.87%, 3',5'-Dimethoxyacetophenone 6.67%, 4-((1E)-3-Hydroxy-1-propenyl)-2-methoxyphenol 3.21%, Octadecanoic acid 2.98%, Capsaicin 2.58%, α-Amyrin 2.53%, 9-Octadecenoic acid (Z)-, 2,3-dihydroxypropyl ester 2.49%, and Vitamin E 2.45%. The MRB showed significant minimum inhibitory concentration values against Pseudomonas putida PA25T, Escherichia coli EC3AT, Pseudomonas aeruginosa PA129AT and Serratia marcescens and a significant MBC value against Escherichia coli EC3AT. This study emphasizes the medicinal efficacy of R. beninensis root and indicated that the plant could make a promising candidate for the isolation of bioactive compounds that could be synthesized as medications for the treatment of bacteria in uropathogenic disorders.

Isolation, identification, and characterization of α- asarone, from hydromethanolic leaf extract of Acorus calamus L. and its apoptosis-inducing mechanism in A549 cells

Due to the presence of several active secondary metabolites, the traditional Indian and Chinese medicinal herb Acorus calamus L. has been utilized for both medical and culinary purposes since ancient times. A recent report has underscored the promising cytotoxic effect of A. calamus leaves extract against non-small cell lung cancer A549 cells. Thus, we want to separate the bioactive substance from the hydromethanolic extract of A. calamus leaves in the current investigation. Thin-layer chromatography was used to separate the compounds and different spectroscopic methods (UV, FTIR, NMR, and LCMS/MS) were used for the structure prediction. α-asarone was found to be the main bioactive compound present and it was isolated from A. calamus leaves extract. It exerted a good cytotoxic effect with an IC50 value of 21.43 ± 1.27 μM against A549 cells and IC50 value of 324.12 ± 1.32 μM against WI-38 cells. The induction of apoptosis in A549 cells by α-asarone was reaffirmed by the diverse differential staining methods including DAPI, Acridine Orange/Ethidium Bromide, and Giemsa staining. Additionally, α-asarone induced mitochondrial membrane potential (ΔΨm) dissipation with a concomitant increase in the production of ROS. Furthermore, it also increased expressions of caspase-3, caspase-9, caspase-8, DR4, and DR5 genes in A549 cells. In conclusion, α-asarone-induced apoptotic cell death in non-small lung cancer cells (A549) as a result of loss of mitochondrial function, increased ROS production, subsequent activation of an internal and extrinsic caspase pathway, and altered expression of genes controlling apoptosis. As a whole, α-asarone is a plausible therapeutic agent for managing lung cancer. Active Compound Phenyl Ether from Potential Endophytic Fungi of Syzygium zeylanicum

Endophytic fungi can be found in various types of plants, especially medicinal plants, such as Syzygium zeylanicum, one of the many medicinal plants found in Indonesia. This study aimed to obtain isolates of endophytic fungi from the bark of Syzygium zeylanicum, L., with antibacterial and antioxidant activities and identify their bioactive compounds. Characterization of endophytic fungi was carried out morphologically and fungi that showed high bioactivity were characterized molecularly. Antibacterial activity was carried out by disc diffusion method and antioxidant activity by DPPH method. Isolation of bioactive compounds was carried out by chromatographic techniques and determination of the structure of pure chemical compounds by spectroscopic analysis. In this study, 3 isolates of endophytic fungi were obtained. Based on their morphological characterization, the endophytic fungi belong to the genera Trichoderma (Code SZT2 and SZT6), and Sclerotium (Code SZT8). Trichoderma mushroom extract (SZT2) gave high antibacterial activity (74.0 % against E. coli; 78.2 % against S. aureus; 80.5 % against S. tyhpi and 74.0 % against B. subtilis). The antioxidant activity was very strong SZT2 (IC50 = 5.83 µg/mL) SZT2 potential endophytic fungus based on molecular analysis is Trichoderma koningi. SZT2 mushroom extract has an active compound in the form of compound 3-hydroxy-4(hydroxy(4-hydroxyphenyl)methyl)-ɣ-butyrolactone based on the results of compound isolation using NMR and HMR. These endophytic fungal compounds can be developed into new sources of antibacterial and antioxidant bioactive compounds through a series of further studies. Alternatives to classic solvents for the isolation of bioactive compounds from Chrysochromulina rotalis

This paper demonstrates a sequential partitioning method for isolating bioactive compounds from Chrysochromulina rotalis using a polarity gradient, replacing classic and hazardous solvents with greener alternatives. Seventeen solvents were evaluated based on their Hansen solubility parameters and for having a similar polarity to the solvents they would replace, four of which were selected as substitutes in the classic fractionation process. Considering the fatty acid and carotenoid recovery yields obtained for each of the solvents, it has been proposed to replace hexane (HEX), toluene (TOL), dichloromethane (DCM) and n-butanol (BUT) with cyclohexane, chlorobenzene, isobutyl acetate and isoamyl alcohol, respectively. In addition, cytotoxic activity was observed when the TOL and DCM solvent extracts were tested against tumour cell lines, demonstrating the antiproliferative potential of compounds containing, for example, fucoxanthin, fatty acids, peptides, isoflavonoids or terpenes, among others.

Comparative Studies of DPPH Radical Scavenging Activity and Content of Bioactive Compounds in Maca (Lepidium meyenii) Root Extracts Obtained by Various Techniques

The effect of the extraction conditions on the DPPH radical scavenging activity and isolation of bioactive compounds from the maca (Lepidium meyenii) root was investigated. Different extraction techniques (maceration, maceration with shaking, ultrasound-assisted extraction, and reflux extraction) were compared. Moreover, the effect of the extraction time and two various solvents (water and ethanol) was studied. The antioxidant activity of extracts was determined by the spectrophotometric method with the DPPH radical, while total phenolic content (TPC) was analyzed by the Folin–Ciocalteu method. Using gas chromatography with a mass selective detector (GC-MS), some characteristics of maca bioactive compounds were identified in the extracts: benzylalkamides (macamides), sterols, nitriles, fatty acids, and their derivatives. The influence of various factors on the extraction process of health-promoting antioxidant compounds from maca root was discussed. It was found that water was a more effective solvent than ethanol for obtaining extracts characterized by high radical scavenging activity and phenolics content. Nevertheless, some ethanol-extractable valuable compounds specific for maca, e.g., macamides or fatty acids derivatives, were not present in water extracts. In developing nutritional and therapeutic formulations based on maca extracts, it is important to take into account that the bioactivity of maca extracts varies depending on the solvent used.

Vinca (Catharanthus roseus) containing phytochemicals and pharmacological profile

Catharanthus roseus (Vinca plant) is an important plant having many medicinal uses. It is popularly known as Madagascar periwinkle. It is small in size perennial herbaceous evergreen plant that was native to the Madagascar island .It has attracted increasing attention due to it being shown to possess a range of phytochemicals with various biological activities such as antioxidant, antibacterial, antifungal, antidiabetic and anticancer properties. It delivers about 130 alkaloids predominantly ajmalcine, vinceine, resperine, vincristine, vinblastine and raubasin. Remarkably, vinblastine and vincristine isolated from this plant were the first plant-derived anticancer agents deployed for clinical use. This paper provides an overview of the traditional use and phytochemical profiles of C. roseus, and summarises updated techniques of the preparation of dried material, extraction and isolation of bioactive compounds from this plant. In addition, purported health benefits of the extracts and bioactive compounds derived from this plant were also addressed to support their potential as therapeutic agents.

Optimized Isolation Procedure for the Extraction of Bioactive Compounds from Spent Coffee Grounds

Due to the worldwide consumption of coffee, many tons of spent coffee grounds (SCGs) are discarded each year, as a by-product of coffee preparation. Not only their disposal is costly, but also it may cause the release of compounds that can endanger the environment. However, there are valuable chemical compounds that can be extracted from SCGs and used in the food industry. The aim of this study was to investigate the main parameters affecting the extraction of caffeine and polyphenols (i.e., chlorogenic acid, neochlorogenic acid, and caffeic acid) and to evaluate the antioxidant properties of the extracts. To this end, extraction solvent, temperature, time, and liquid-to-solid ratio were studied. A response surface methodology was used to optimize the extraction process. According to the results, the caffeine content of the optimum extract was found to be 6.14 mg/g in dry SCGs, the total polyphenol content was 19.85 mg gallic acid equivalents/g, while the ferric reducing antioxidant power and DPPH scavenging values were 136.69 μmoL ascorbic acid equivalents/g and 230.41 μmoL DPPH/g, respectively. The experimental values were in close agreement with the predicted ones, highlighting the potential of SCGs to be used for the isolation of bioactive compounds with the proposed extraction procedure.

Anticancer activity of Indonesian Melia azedarach L.: Phyto-chemistry, in vitro and in silico studies

This study describes the isolation of bioactive compounds and their in vitro anticancer activities from Melia azedarach L leaves from Karanglor, Karangan, Karanganom Klaten, Central Java-Indonesia. Isolated and characterised were the phenolics, kaempferol 7-O-rutinoside 1 and 4-methoxyresorcinol 3, The anticancer activity of the crude methanol extract of M. azedarach against a number cell lines (LNCaP, MDA-MB-231, and MCF-7) was found to be moderate; kaempferol derivatives were notable for their antiproliferative activities. Computational analysis, including molecular docking simulations and molecular dynamic studies, were used to investigate the feasibility of radiolabelled versions of 1 and 3 as theranostic agents targeting tyrosine kinase-type cell surface receptor HER2 in cancer - the iodinated derivatives 2 and 4, respectively, were used in this investigation as surrogates. The corresponding radioiodinated 2 and 4 were found to have high binding affinities of 10.1 and 5.7 kcal/mol, respectively, and have a good stability on the receptor.

Qualitative phytochemical screening and ftir spectroscopic analysis of mitracarpus hirtus leaf extracts

The present study aim is to analyze the phytochemicals present in Mitracarpus hirtus leaf extracts by using Qualitative phytochemical analysis, Fourier-transform infrared spectroscopy (FTIR). The leaf extracts were prepared using three different solvents. The phytochemical analysis Fourier transform infrared spectroscopy (FTIR) analysis were performed using standard methods. The FTIR spectroscopic studies revealed different characteristic peak values with various functional groups present in the compounds of respective extracts. The FT-IR analysis shows the presence of different functional groups such as alcohols, phenols, alkanes, carboxylic acids, aldehydes, ketones, alkenes, primary amines, aromatics, esters, ethers, alkyl halides and aliphatic amine compounds, which showed major compounds present in the leaf extracts. The present study generated the FTIR spectrum profile for the medicinally important plant Mitracarpus hirtus. The present study provides evidences that different extracts of Mitracarpus hirtus leaf is useful to cure many serious diseases which remained still problematic and for further isolation of bioactive compounds from the plant which could be of interest for the development of the new drug.

Phytochemical, Antimicrobial, Antioxidant, and In Vitro Cytotoxicity Evaluation of Echinops erinaceus Kit Tan

Wild plants are used by many cultures for the treatment of diverse ailments. However, they are formed from mixtures of many wanted and unwanted phytochemicals. Thus, there is a necessity to separate the bioactive compounds responsible for their biological activity. In this study, the chemical composition as well as antimicrobial and cytotoxic activities of Echinops erinaceus Kit Tan (Asteraceae) were investigated. This led to the isolation and identification of seven compounds, two of which are new (erinaceosin C3 and erinaceol C5), in addition to methyl oleate (C1) and ethyl oleate (C2), loliolide (C4), (E)-p-coumaric acid (C6), and 5,7,3`,5`-tetrahydroxy flavanone (C7). The structures of the isolated compounds were elucidated by 1D, 2D NMR, and HR-ESI-MS. The methanol extract showed the highest antimicrobial activity among the tested extracts and fractions. The n-hexane and EtOAc extracts showed remarkable antimicrobial activity against B. subtilus, P. aeruginosa, E. coli, and C. albicans. A cytotoxicity-guided fractionation of the most bioactive chloroform extract resulted in the isolation of bioactive compounds C1/C2, which showed significant cytotoxicity against HCT-116 and CACO2 cell lines (IC50 24.95 and 19.74 µg/mL, respectively), followed by compounds C3 (IC50 82.82 and 76.70 µg/mL) and C5 (IC50 99.09 and 87.27 µg/mL), respectively. The antioxidant activity of the bioactive chloroform fractions was screened. Molecular docking was used to explain the results of the antimicrobial and anticancer activities against five protein targets, including DNA gyrase topoisomerase II, enoyl-acyl carrier protein reductase of S. aureus (FabI), dihydrofolate reductase (DHFR), β-catenin, and human P-glycoprotein (P-gp).

Extraction, Isolation of Bioactive Compounds and Therapeutic Potential of Rapeseed (Brassica napus L.).

Rapeseed (Brassica napus L.) is a herbaceous annual plant of the Cruciferous family, the Cabbage genus. This oilseed crop is widely used in many areas of industry and agriculture. High-quality oil obtained from rapeseed can be found in many industrial food products. To date, extracts with a high content of biologically active substances are obtained from rapeseed using modern extraction methods. Brassica napus L. seeds contain polyunsaturated and monounsaturated fatty acids, carotenoids, phytosterols, flavonoids, vitamins, glucosinolates and microelements. The data in this review show that rapeseed biocompounds have therapeutic effects in the treatment of various types of diseases. Some studies indicate that rapeseed can be used as an anti-inflammatory, antioxidant, antiviral, hypoglycemic and anticancer agent. In the pharmaceutical industry, using rapeseed as an active ingredient may help to develop new forms drugs with wide range of therapeutic effects. This review focuses on aspects of the extraction of biocompounds from rapeseed and the study of its pharmacological properties.

Genomic Investigation of Desert Streptomyces huasconensis D23 Reveals Its Environmental Adaptability and Antimicrobial Activity.

The harsh climatic conditions of deserts may lead to unique adaptations of microbes, which could serve as potential sources of new metabolites to cope with environmental stresses. However, the mechanisms governing the environmental adaptability and antimicrobial activity of desert Streptomyces remain inadequate, especially in extreme temperature differences, drought conditions, and strong radiation. Here, we isolated a Streptomyces strain from rocks in the Kumtagh Desert in Northwest China and tested its antibacterial activity, resistance to UV-C irradiation, and tolerance to hydrogen peroxide (H2O2). The whole-genome sequencing was carried out to study the mechanisms underlying physiological characteristics and ecological adaptation from a genomic perspective. This strain has a growth inhibitory effect against a variety of indicator bacteria, and the highest antibacterial activity recorded was against Bacillus cereus. Moreover, strain D23 can withstand UV-C irradiation up to 100 J/m2 (D10 = 80 J/m2) and tolerate stress up to 70 mM H2O2. The genome prediction of strain D23 revealed the mechanisms associated with its adaptation to extreme environmental and stressful conditions. In total, 33 biosynthetic gene clusters (BGCs) were predicted based on anti-SMASH. Gene annotation found that S. huasconensis D23 contains several genes and proteins associated with the biosynthesis of factors required to cope with environmental stress of temperature, UV radiation, and osmotic pressure. The results of this study provide information about the genome and BGCs of the strain S. huasconensis D23. The experimental results combined with the genome sequencing data show that antimicrobial activity and stress resistance of S. huasconensis D23 was due to the rich and diverse secondary metabolite production capacity and the induction of stress-responsive genes. The environmental adaptability and antimicrobial activity information presented here will be valuable for subsequent work regarding the isolation of bioactive compounds and provide insight into the ecological adaptation mechanism of microbes to extreme desert environments.

Isolation, Characterization and Anticancer Activity of Two Bioactive Compounds from Arisaema flavum (Forssk.) Schott.

Medicinal plants play important role in the public health sector worldwide. Natural products from medicinal plants are sources of unlimited opportunities for new drug leads because of their unique chemical diversity. Researchers have focused on exploring herbal products as potential sources for the treatment of cancer, cardiac and infectious diseases. Arisaema flavum (Forssk.) is an important medicinal plant found in the northwest Himalayan regions of Pakistan. It is a poisonous plant and is used as a remedy against snake bites and scorpion stings. In this study, two bioactive compounds were isolated from Arisaema flavum (Forssk.) and their anticancer activity was evaluated against human breast cancer cell line MCF-7 using an MTT assay. The crude extract of Arisaema flavum (Forssk.) was subjected to fractionation using different organic solvents in increasing order of polarity. The fraction indicating maximum activity was then taken for isolation of bioactive compounds using various chromatographic and spectroscopic techniques such as column chromatography, thin-layer chromatography (TLC), gas chromatography-mass spectrometry (GC-MS), Fourier transform infrared spectroscopy (FTIR) and nuclear magnetic resonance spectroscopy (NMR). Crude extract of Arisaema flavum (Forssk.), as well as various fractions extracted in different solvents such as n-hexane, chloroform and ethyl acetate, were tested against human breast cancer cell line MCF-7 using an MTT assay. The crude extract exhibited significant dose-dependent anticancer activity with a maximum activity of 78.6% at 500 µg/mL concentration. Two compounds, hexadecanoic acid ethyl ester with molecular formula C18H36O7 and molar mass 284 and 5-Oxo-19 propyl-docosanoic acid methyl ester with molecular formula C26H50O3 and molecular mass 410, were isolated from chloroform fraction. These compounds were tested against the MCF-7cell line for cytotoxic activity and exhibited a significant (p < 0.00l) decrease in cell numbers for MCF-7 cells with IC50 of 25 µM after 48 h of treatment. Results indicated that Arisaema flavum (Forssk.) possesses compounds with cytotoxic activity that can further be exploited to develop anticancer formulations.

Optimization of the Extraction Parameters for the Isolation of Bioactive Compounds from Orange Peel Waste

More and more research is being focused on the production of value-adding products from waste materials. Food waste is not only a major global issue, but also an excellent source of bioactive compounds. In this study, the parameters that affect the extraction of the bioactive compounds (polyphenols, ascorbic acid, hesperidin, carotenoids) from orange peels, and their antioxidant properties, were optimized, using a response surface methodology (RSM) (examining the extraction temperature, time, and composition of the extraction solvent). In addition, the effect of two more techniques was examined [ultrasound (US) and pulsed electric field (PEF)], either separately or combined, so as to determine whether they can enhance the extraction of the compounds. From our results, it was apparent that orange peels are an excellent source of many bioactive compounds since the extracts contained hesperidin (16.26 mg/g dw), total polyphenols (34.71 mg GAE/g dw), ascorbic acid (1228.93 mg/100 g dw) and total carotenoids (52.98 μg CtE/g dw).

GC-MS Profiling, in vitro Antioxidant and Antimicrobial Activities of Kaempferia parviflora Wall. ex. Baker Rhizome Extract

Bakground: Kaempferia parviflora (zingiberaceae family) rhizomes are used for various pharmacological applications. The present study is aimed for putative identification of phyto-constituents in the chloroform extract of the rhizome of Kaempferia parviflora and to correlate with bioactivities like antioxidant and anti-microbial/fungal activities. Methods: Rhizome extract was prepared using Soxhlet extractor and undergone the phytoconstituents analysis of it by gas chromatography–mass spectrometry (GC-MS). Results: Identified twenty-five compounds, out of which 4H-1-Benzopyran- 4-0ne,5,7-dimethoxy-2-phenyl and Hydroxyurea showed the maximum and minimum relative abundance of 76.4% and 0.002% respectively. By agar well diffusion method, the extract (25 μg/ml) exhibited highest antibacterial potential against Micrococcus luteus (MTCC 106) (gram-positive bacteria) and anti-fungal potential against plant pathogen Rhizoctonia solani (MTCC 4633). The extract also showed the potent scavenging activities of 2,2-diphenyl-1-picryl-hydrazine (DPPH) and 2,2′-Azino-bis (3-Ethylbenzothiazoline-6-Sulfonic Acid) (ABTS) radicals with IC50 values of 3.0 ± 0.1 mg/ml and 19.4 ± 1.2 μg/ml respectively. Subsequently, lipid peroxidation assay was also used for estimation of antioxidant activity of this extract. The extract (20 μg/ml) inhibited lipid peroxidation by 80.0 ± 4.5%. Conclusion: The outcomes of present study provide scope for isolation of bioactive compounds from Kaempferia parviflora for medicinal application.
