Sulfate ionic liquids promoted FeCl3-catalyzed dehydration of propargyl alcohols

Conjugated enynes are an important class of compounds that could be widely used in medicinal chemistry and organic synthesis. Herein, various sulfate ionic liquids with FeCl3 were used to catalyze the dehydration of propargyl alcohols to prepare conjugated enynes. Sulfate ionic liquids with different substituent chain lengths or cations exhibited high catalytic activity, affording the highest conjugated enyne yield of 96 % with selectivity of 97 %. At the same time, the by-product naphthofuran derivative was isolated and verified by the single crystal X-ray diffraction for the first time. Kinetic studies found that the addition of sulfate ionic liquids increased the rate of formation of desired product conjugated enyne and inhibited the production of by-product naphthofuran derivative. Deuterated kinetic isotope studies were carried out by using the deuterated substrate PhCC(CD3)2OH and PhCC(CH3)2OD. A large value of H/D kinetic isotope effects (KIE1 = 5.5) suggested that the βC—H bond cleavage was involved in the rate-determining step and the formation of desired conjugated enyne followed the E2 elimination mechanism. An inverse kinetic isotope effects (KIE3 = 0.66) suggested that the formation of by-product naphthofuran derivative followed the SN2 mechanism. Density functional theory (DFT) calculations were conducted to reveal the activation of sulfate anion on the methyl group of propargyl alcohol. Based on the crystal structure of by-product, kinetic studies, deuterated kinetic isotope studies and DFT calculations, a cooperative mechanism of dehydration of propargyl alcohol over sulfate ionic liquids and FeCl3 was proposed.

Heteroatom modified polymer immobilized ionic liquid stabilized ruthenium nanoparticles: Efficient catalysts for the hydrolytic evolution of hydrogen from sodium borohydride

Ruthenium nanoparticles stabilised by polymer immobilized ionic liquids catalyse the hydrolytic release of hydrogen from sodium borohydride. The composition of the polymer influences performance and ruthenium nanoparticles stabilised by an amine-decorated imidazolium-based polymer immobilised ionic liquid (RuNP@NH2-PIILS) was the most efficient with a maximum initial turnover frequency (TOF) of 177 moleH2.molRu−1.min−1, obtained at 30°C with a catalyst loading of 0.08 mol%; markedly higher than that of 69 molH2.molRu−1.min−1 obtained with 5 wt% Ru/C and one of the highest to be reported for a RuNP catalyst. The apparent activation energy (Ea) of 38.9 kJ mol−1 for the hydrolysis of NaBH4 catalysed by RuNP@NH2-PIILS is lower than that for the other polymer immobilized ionic liquid stabilised RuNPs, which is consistent with its efficacy. Comparison of the initial rates of hydrolysis in H2O and D2O catalysed by RuNP@NH2-PIILS gave a primary kinetic isotope effect (kH/kD) of 2.3 which supports a mechanism involving rate limiting oxidative addition of one of the O-H bonds in a strongly hydrogen-bonded surface-coordinated [BH3H−]—-H2O ensemble. The involvement of a surface-coordinated borohydride is further supported by an inverse kinetic isotope effect of 0.65 obtained from a comparison of the initial rates for the hydrolysis of NaBH4 and NaBD4 under the conditions of catalysis i.e., at a high hydride/catalyst mole ratio. Interestingly though, when the comparison of the initial rates of hydrolysis of NaBH4 and NaBD4 was conducted in dilute solution with a hydride/catalyst mole ratio of 1 a kinetic isotope effect (kH/kD) of 2.72 was obtained; this would be more consistent with concerted activation of both an O-H and B-H bond in the rate limiting step, possibly via a concerted oxidative addition-hydride transfer in the surface-coordinated hydrogen-bonded ensemble. Catalyst stability and reuse studies showed that RuNP@NH2-PIILS retained 71% of its activity over five runs; the gradual drop in the initial TOF with run number appears to be due to passivation of the catalyst by the sodium borate by-product as well as an increase in viscosity of the reaction mixture rather than leaching of the catalyst.

Oxidation of Hypophosphorous Acid by a Ruthenium(VI) Nitrido Complex in Aqueous Acidic Solution. Evidence for a Proton-Coupled N-Atom Transfer Mechanism.

The oxidation of hypophosphorous acid (H3PO2) by a ruthenium(VI) nitrido complex, [(L)RuVI(N)(OH2)]+ (RuVIN; L = N,N'-bis(salicylidene)-o-cyclohexyldiamine dianion), has been studied in aqueous acidic solutions at pH 0-2.50. The reaction has the following stoichiometry: 2[(L)RuVI(N)(OH2)]+ + 3H3PO2 + H2O → 2[(L)RuIII(NH2P(OH)2)(OH2)]+ + H3PO3. The pseudo-first-order rate constant, kobs, depends linearly on [H3PO2], and the second-order rate constant k2 depends on [H+] according to the relationship k2 = k[H+]/([H+] + Ka), where k is the rate constant for the oxidation of H3PO2 molecule and Ka is the dissociation constant of H3PO2. At 298.0 K and I = 1.0 M, k = (2.04 ± 0.19) × 10-2 M-1 s-1 and Ka = (6.38 ± 0.63) × 10-2 M. A kinetic isotope effect (KIE) of 2.9 ± 0.1 was obtained when kinetic studies were carried out with D3PO2 at pH 1.16, suggesting P-H bond cleavage in the rate-determining step. On the other hand, when the kinetics were determined in D2O, an inverse KIE of 0.21 ± 0.03 (H3PO2 in H2O vs H3PO2 in D2O) was found. On the basis of experimental results and DFT calculations, the proposed mechanism involves an acid-catalyzed tautomerization of H2P(O)(OH) to HP(OH)2; the latter molecule is the reacting species which reacts with RuVIN via a proton-coupled N-atom transfer pathway.

Enhancement in the metal efficiency of Ru/TiO2 catalyst for guaiacol hydrogenation via hydrogen spillover in the liquid phase

Decrease in the metal dispersion of noble-metal-loaded catalysts with high metal loading is a major factor reducing noble-metal efficiency. Herein, we investigated the enhancement in metal efficiency of Ru/TiO2 catalyst by utilizing hydrogen spillover in the liquid-phase. Ru was highly dispersed at low loadings (0.1 and 0.5 wt%), while larger nanoparticles were formed at higher loadings (1–5 wt%). The hydrogen spillover in liquid phase was activated at reaction temperature (100 °C) as the Ru dispersion decreased, which was confirmed through physical dilution experiments, hydrogen temperature-programmed-reduction, and kinetic analysis. Isotope experiment was conducted using D2O, observing inverse kinetic isotope effect (IKIE) for the high-Ru-loading catalysts. Based on the understanding of the hydrogen spillover in the liquid phase, the low metal efficiency of high-Ru-loading catalysts resulting from low dispersion could be compensated simply by physically mixing pristine TiO2, which played a role as new active sites when liquid-phase hydrogen spillover was activated.

Density functional theory calculations of nitrogen and oxygen equilibrium isotope fractionations in NO3−–NO2−–H2O aqueous system reveal inverse kinetic isotope effects during nitrite oxidation

Nitrate and nitrite play key roles in the nitrogen cycle on Earth's surface. The isotope fractionation during nitrate reduction on enzymatic level involves multiple steps, including transfer of free NO3− to the activate site on nitrate reductase and NO2− equilibration with ambient water. However, the isotope fractionation factors of 15N and 18O among NO3−, NO2−, and H2O molecules in aqueous phases are poorly constrained. It strongly impedes the understanding of the involved processes and using stable isotopes to quantitatively examine the biogeochemical nitrogen cycle. In this contribution, we employ the density functional theory method with the Urey-Bigeleisen-Goeppert-Mayer model to predict the nitrogen and oxygen equilibrium isotope fractionation factors of NO3−, NO2−, and H2O molecules in gaseous and aqueous phases. Our calculation results show that the solvent effect has a large influence on equilibrium isotope fractionation for oxygen in water (+10.1‰ between liquid and vapor water at 25 °C), which is different to the little solvent effects on both oxygen and nitrogen in nitrate and nitrite. The calculated temperature-dependent equilibrium isotope fractionations between nitrogen of NO3− and NO2−, and oxygen of H2O and NO2− are consistent with previous laboratory experiments. Our results confirm that the oxygen equilibrium isotope fractionations between NO3− and NO2− should be +9.4‰ at 25 °C. Integrating the new results and previously reported kinetic isotope effects of nitrate reduction, we demonstrate inverse kinetic isotope effects for both nitrogen (+15.4‰) and oxygen (+5.2‰) during nitrite oxidation, which falls in the range of previous experiments. The new results enable us to use both nitrogen and oxygen isotopes as a bonded isotope tool to quantitatively assess the nitrogen cycle in low-temperature environments.

Visible-Light-Driven, Iridium-Catalyzed Hydrogen Atom Transfer: Mechanistic Studies, Identification of Intermediates, and Catalyst Improvements.

The harvesting of visible light is a powerful strategy for the synthesis of weak chemical bonds involving hydrogen that are below the thermodynamic threshold for spontaneous H2 evolution. Piano-stool iridium hydride complexes are effective for the blue-light-driven hydrogenation of organic substrates and contra-thermodynamic dearomative isomerization. In this work, a combination of spectroscopic measurements, isotopic labeling, structure–reactivity relationships, and computational studies has been used to explore the mechanism of these stoichiometric and catalytic reactions. Photophysical measurements on the iridium hydride catalysts demonstrated the generation of long-lived excited states with principally metal-to-ligand charge transfer (MLCT) character. Transient absorption spectroscopic studies with a representative substrate, anthracene revealed a diffusion-controlled dynamic quenching of the MLCT state. The triplet state of anthracene was detected immediately after the quenching events, suggesting that triplet–triplet energy transfer initiated the photocatalytic process. The key role of triplet anthracene on the post-energy transfer step was further demonstrated by employing photocatalytic hydrogenation with a triplet photosensitizer and a HAT agent, hydroquinone. DFT calculations support a concerted hydrogen atom transfer mechanism in lieu of stepwise electron/proton or proton/electron transfer pathways. Kinetic monitoring of the deactivation channel established an inverse kinetic isotope effect, supporting reversible C(sp2)–H reductive coupling followed by rate-limiting ligand dissociation. Mechanistic insights enabled design of a piano-stool iridium hydride catalyst with a rationally modified supporting ligand that exhibited improved photostability under blue light irradiation. The complex also provided improved catalytic performance toward photoinduced hydrogenation with H2 and contra-thermodynamic isomerization.

Analysis of nitrite oxidation process and nitrification performance by nitrogen and oxygen isotope fractionation effect

The N and O isotope fractionation effects in NO2−-N oxidation and nitrification performance of an activated sludge system treating municipal wastewater are unknown. The nitrifying sludge was cultured under different temperature (33 ± 1 °C, 25 ± 1 °C,and 18 ± 1 °C) and dissolved oxygen (DO: 0.5–1 mg/L, 3–4 mg/L, and 7–8 mg/L). The inverse kinetic isotope effects of N and O (15εNO2 and 18εNO2) were −0.62‰ to −7.08‰ and −0.87‰ to −1.68‰ in the process of NO2−-N oxidation, respectively. 15εNO3 gradually increased with increasing of temperature (15εNO3–33°C (14.49‰) > 15εNO3–25°C (10.43‰) > 15εNO3–18°C (7.3‰)), while the 15εNO3:18εNO3 was maintained at 1.02–5.32. The increase of temperature improved the nitrification activity, which promoted the fractionation effect, but the change of DO did not highlight this difference. The exchange of NO2−-N and H2O (XNOB) was 32.5 ± 1.5%, and the kinetic isotope effect of H2O participating in the reaction (18εk, H2O, 2) was 22.57 ± 1.79‰, indicating that H2O was involved in the NO2−-N oxidation rather than DO. In summary, the elevated temperature enhanced the fractionation effect of NO2−-N oxidation. This study provides a new perspective to reveal the mechanism of NO2−-N oxidation, optimize the process of nitrogen removal from wastewater and further control water eutrophication.

The complete mechanism of an aldol condensation in water.

The base-catalyzed aldol condensation between benzaldehyde and p-acetylbenzoic acid in water shows an inverse solvent kinetic isotope effect, k3,D2O/k3,H2O, of 1.33 ± 0.03. The reaction is definitely faster in D2O. This is interpreted to mean that the rate-limiting step in a five-step mechanism is Step 5, the final elimination of hydroxide from the enolate intermediate, not the formation of that intermediate. This is the same result and the same conclusion as from earlier studies in aqueous acetonitrile and refutes a suggestion, based on computations, that the rate-limiting step would change in water. Those computations are criticized as implying impossibly large isotope effects.

Deuterium kinetic isotope effects as redox mechanistic criterions

AbstractThis account article focuses on deuterium kinetic isotope effects (KIEs) used as criterions to elucidate redox mechanisms including proton‐, hydrogen‐ and hydride‐transfer reactions. Hydrogen atom transfer (HAT) is composed of two elementary steps: electron transfer (ET) and proton transfer (PT), while hydride transfer is composed of three elementary steps: ET, PT, and ET. Large tunneling effects are often observed for proton‐coupled electron‐transfer (PCET) reactions of metal–oxygen complexes in which ET occurs to the metal center and PT occurs simultaneously to the ligand, exhibiting large KIEs. Whether HAT proceeds via sequential ET/PT, PT/ET, or concerted PCET (cPCET) depending on the redox properties of hydrogen donors and acceptors to exhibit different KIEs. Whether hydride transfer also proceeds via sequential ET/PT/ET, PT/ET/ET, or cPCET/ET depending on the redox properties of hydride donors and acceptors to exhibit different KIEs. Temperature dependence of KIEs for aldehyde deformylation reactions has enabled to distinguish two reaction pathways: one is a HAT and the other is a nucleophilic addition. The change of the mechanism from cPCET to sequential ET/PT is made possible by binding acids to the hydrogen and hydride acceptors when no KIE is observed. Inverse KIEs are also discussed for acid (or deuteron)‐promoted ET reactions.

Insertion products in the reaction of carbonyl oxide Criegee intermediates with acids: Chloro(hydroperoxy)methane formation from reaction of CH2OO with HCl and DCl

The reactions of carbonyl oxide Criegee intermediates with acids proceed predominantly by an insertion mechanism. We characterise the products from one of the simplest reactions of carbonyl oxides with inorganic acids, CH2OO + hydrogen chloride, which occurs via a 1,2-insertion in the H–Cl bond. Reactions of both HCl and DCl isotopologues yield product signal at the mass of the insertion product chloro(hydroperoxy)methane and a dissociative ionisation peak at the mass of the protonated (or deuteronated) Criegee intermediate. The isotopic composition of the insertion product has been measured for reaction mixtures where both HCl isotopologues are present, and the H/D ratio of the product is consistently higher (by a factor of 1.6 ± 0.3) than that of the reactants. This isotope selectivity in the products has smaller uncertainty than the ratio of measured rate coefficients and suggests a normal (k H > k D) kinetic isotope effect in the reaction. Theoretical kinetics calculations predict a small normal kinetic isotope effect for the overall reaction (k H / k D = 1.35 at 20 Torr N2 and k H / k D = 1.2 at 1 atm N2) but predict a substantial inverse kinetic isotope effect (k D > k H) for the stabilisation fraction, in disagreement with the experimental observation.

Kinetic and Dynamic Studies of the F(2P) + ND3 → DF + ND2 Reaction.

The fast F reaction with NH3 poses a big challenge to experimental studies because of secondary chemical and collisional reactions. The quasi-classical trajectory method is utilized to investigate the mode specificity, product energy disposal, and temperature dependence of the thermal rate coefficient of F + ND3 → DF + ND2 on a recently developed potential energy surface. The effect of isotopic substitution is explored by comparing the F + ND3 reaction with the F + NH3 reaction. The computed results permit a better understanding of the F + ammonia reaction. The DF vibrational state has a Λ-type distribution, in accordance with the experimental measurement by the fast flow reactor technique. The product ND2 is dominantly populated in the ground state, and a considerable amount of ND2 is produced in the fundamental states of the bending mode. The similar vibrational state distributions of HF and NH2 in the F + NH3 reaction indicate a weak isotopic substitution effect on the product energy disposal. Exciting the umbrella mode of ND3 suppresses the reaction at low energies below 5 kcal mol-1, in sharp contrast to the observation in the F + NH3 reaction. These dynamical behaviors can be partially explained by the sudden vector projection model. In addition, the thermal rate coefficient of F + ND3 shows no temperature dependence in the range between 150 and 2000 K. There exists an inverse kinetic isotope effect at temperatures from 150 to 1500 K.

Unimolecular, Bimolecular, and Intramolecular Hydrolysis Mechanisms of 4-Nitrophenyl β-d-Glucopyranoside.

1,2-trans-Glycosides hydrolyze through different mechanisms at different pH values, but systematic studies are lacking. Here, we report the pH-rate constant profile for the hydrolysis of 4-nitrophenyl β-D-glucoside. An inverse kinetic isotope effect of k(H3O+)/k(D3O+) = 0.65 in the acidic region indicates that the mechanism requires the formation of the conjugate acid of the substrate for the reaction to proceed, with the heterolytic cleavage of the glycosidic C-O bond. Reactions in the pH-independent region exhibit general catalysis with a single proton in flight, a normal solvent isotope effect of kH/kD = 1.5, and when extrapolated to zero buffer concentration show a small solvent isotope effect of k(H2O)/k(D2O) = 1.1, consistent with water attack through a dissociative mechanism. In the basic region, solvolysis in 18O-labeled water and H2O/MeOH mixtures allowed the detection of bimolecular hydrolysis and neighboring group participation, with a minor contribution of nucleophilic aromatic substitution. Under mildly basic conditions, a bimolecular concerted mechanism is implicated through an inverse solvent isotope effect of k(HO-)/k(DO-) = 0.5 and a strongly negative entropy of activation (ΔS‡ = -13.6 cal mol-1 K-1). Finally, at high pH, an inverse solvent isotope effect of k(HO-)/k(DO-) = 0.5 indicates that the formation of 1,2-anhydrosugar is the rate-determining step.

Competition of quantum effects in H 2/D 2 sieving in narrow single-wall carbon nanotubes

Nanoporous materials have the potential to be used as molecular sieves to separate chemical substances in a mixture via selective adsorption and kinetic sieving. The separation of isotopologues is also possible via the so-called quantum sieving effect: the different effective size of isotopologues due to their different Zero Point Energy (ZPE). Here we compare the diffusion coefficients of Hydrogen and Deuterium in (8,0) Single Walled Carbon Nanotubes obtained with quantum dynamics simulations. The diffusion channels obtained present important contributions from resonances connecting the potential wells. These resonances, which are more important for H than for D , increase the low-temperature diffusivity of both isotopologues, but prevent the inverse kinetic isotope effect reported for similar nanostructured systems.

Temperature driven interchange of the effective size of proton with deuterium

We employ ab initio molecular dynamics to study the H/D isotope effect in order to introduce the usually neglected effect of temperature in answering the controversial question: Could deuterium be bigger than protium as suggested from neutron diffraction studies on benzene crystals? Especially that recent neutron powder diffraction study by Fortes and Capelli contradicts previous observations made by Dunitz and Ibberson. Our simulations of the dynamics of isolated benzene and perdeuterated isotopologues have shown surprisingly that, if an environment is not present, the deuterium atom is always larger than the hydrogen atom for temperatures from 100 K up to 1200 K with the rate of change increasing with temperature. Only the inclusion of quantum effects of the nuclei reverses the situation, with hydrogen atoms in isolated benzene becoming larger in volume at low temperatures. In benzene-crystal simulations, we have demonstrated that at some temperatures, above 400 K, deuterium atoms appear bigger than protium atoms. Further studies on the isotope effect and inverse kinetic isotope effect are required from different perspectives; in particular, our results support one of the competing mechanisms of olfaction, in which molecular shape (size) is important in the process. In consequence, the spectral mechanism, which assumes that shape/size of the molecule remains unchanged after deuteration, appears less favorable.

Regiospecific and Enantioselective Arylvinylcarbene Insertion of a C-H Bond of Aniline Derivatives Enabled by a Rh(I)-Diene Catalyst.

Asymmetric insertion of an arylvinylcarbenoid into the C-H bond for direct enantioselective C(sp2)-H functionalization of aniline derivatives catalyzed by a rhodium(I)-diene complex was developed for the first time. The reaction occurred exclusively at the uncommon vinyl terminus site with excellent E selectivity and enantioselectivities, providing various chiral γ,γ-gem-diarylsubstituted α,β-unsaturated esters with broad functional group compatibility under simple and mild conditions. It provides a rare example of the asymmetric C-H insertion of arenes with selective vinylogous reactivity. Synthesis applications of this protocol were featured by several versatile product transformations. Systematic DFT calculations were also performed to elucidate the reaction mechanism and origin of the uncommon enantio- and regioselectivity of the Rh(I)-catalyzed C(sp2)-H functionalization reaction. The measured and computed inverse deuterium kinetic isotope effect supports the C-C bond-formation step as the rate-determining step. Attractive interactions between the chiral ligand and substrates were also proposed to control the enantioselectivity.

Inverse kinetic isotope effects in the charge transfer reactions of ammonia with rare gas ions.

In the absence of experimental data, models of complex chemical environments rely on predicted reaction properties. Astrochemistry models, for example, typically adopt variants of capture theory to estimate the reactivity of ionic species present in interstellar environments. In this work, we examine astrochemically-relevant charge transfer reactions between two isotopologues of ammonia, NH3 and ND3, and two rare gas ions, Kr+ and Ar+. An inverse kinetic isotope effect is observed; ND3 reacts faster than NH3. Combining these results with findings from an earlier study on Xe+ (Petralia et al., Nat. Commun., 2020, 11, 1), we note that the magnitude of the kinetic isotope effect shows a dependence on the identity of the rare gas ion. Capture theory models consistently overestimate the reaction rate coefficients and cannot account for the observed inverse kinetic isotope effects. In all three cases, the reactant and product potential energy surfaces, constructed from high-level ab initio calculations, do not exhibit any energetically-accessible crossing points. Aided by a one-dimensional quantum-mechanical model, we propose a possible explanation for the presence of inverse kinetic isotope effects in these charge transfer reaction systems.

An isotopic study of abiotic nitrite oxidation by ligand-bound manganese (III)

Redox transformations of nitrogen (N) play a critical role in determining its speciation and biological availability, thus defining the magnitude and extent of productivity in many ecosystems. A range of important nitrogen transformations often co-occur in regions hosting other redox-active elements, including sulfur, iron, and manganese (Mn), especially along sharp redox gradients within aquatic sediments. This proximity in “redox real estate” produces conditions under which multi-element interactions and coupled cycling are thermodynamically favored. While previous work has reported anoxic nitrification linked to the presence of manganese (Mn) oxides in sediments, a clear connection between the cycling of Mn and N has remained elusive. Soluble Mn(III), which is stabilized via ligand-complexation, has recently been shown to represent the dominant dissolved Mn species in many environments. Here, we examined the reactivity of ligand-stabilized Mn(III) with nitrite, using natural abundance stable nitrogen and oxygen isotopes to explore reaction dynamics under a range of conditions. Oxidation of nitrite to nitrate by Mn(III)-pyrophosphate proceeded abiotically under both oxygen replete and nitrogen-purged conditions. Kinetics and isotope systematics of this reaction were measured over a range of pH (5–8), with reaction rates decreasing with increasing pH. Under all treatments, an inverse kinetic isotope effect of −19.9 ± 0.7‰ was observed for N, remarkably similar to previously documented fractionation by nitrite-oxidizing organisms. Experiments using 18O-labeled water confirmed that the source of the additional oxygen atom was from water. These findings suggest that nitrite oxidation in environments hosting abundant ligand-bound Mn(III), including porewaters, estuaries, and coastal waters, may be facilitated in part by abiotic reactions with Mn, even under functionally anoxic conditions.

Kinetic and Spectroscopic Characterization of the Catalytic Ternary Complex of Tryptophan 2,3-Dioxygenase.

The first step of the kynurenine pathway for l-tryptophan (l-Trp) degradation is catalyzed by heme-dependent dioxygenases, tryptophan 2,3-dioxygenase (TDO) and indoleamine 2,3-dioxygenase. In this work, we employed stopped-flow optical absorption spectroscopy to study the kinetic behavior of the Michaelis complex of Cupriavidus metallidurans TDO (cmTDO) to improve our understanding of oxygen activation and initial oxidation of l-Trp. On the basis of the stopped-flow results, rapid freeze-quench (RFQ) experiments were performed to capture and characterize this intermediate by Mössbauer spectroscopy. By incorporating the chlorite dismutase-chlorite system to produce high concentrations of solubilized O2, we were able to capture the Michaelis complex of cmTDO in a nearly quantitative yield. The RFQ-Mössbauer results confirmed the identity of the Michaelis complex as an O2-bound ferrous species. They revealed remarkable similarities between the electronic properties of the Michaelis complex and those of the O2 adduct of myoglobin. We also found that the decay of this reactive intermediate is the rate-limiting step of the catalytic reaction. An inverse α-secondary substrate kinetic isotope effect was observed with a kH/kD of 0.87 ± 0.03 when (indole-d5)-l-Trp was employed as the substrate. This work provides an important piece of spectroscopic evidence of the chemical identity of the Michaelis complex of bacterial TDO.

Energy-dependent normal and unusually large inverse chlorine kinetic isotope effects of simple chlorohydrocarbons in collision-induced dissociation by gas chromatography-electron ionization-tandem mass spectrometry.

Kinetic isotope effects (KIEs) occurring in mass spectrometry (MS) can provide in-depth insights into the fragmentation behaviors of compounds of interest in MS. Yet, the fundamentals of KIEs in collision-induced dissociation (CID) in tandem mass spectrometry (MS/MS) are unclear, and information about chlorine KIEs (Cl-KIEs) of organochlorines in MS is particularly scarce. This study investigated the Cl-KIEs of dichloromethane, trichloroethylene, and tetrachloroethylene during CID using gas chromatography-electron ionization triple-quadrupole MS/MS. Cl-KIEs were evaluated with MS signal intensities. All the organochlorines presented large inverse Cl-KIEs (<1, the departures of Cl-KIEs from 1 denote the magnitudes of Cl-KIEs), showing the largest magnitudes of 0.797, 0.910, and 0.892 at the highest collision energy (60 eV) for dichloromethane, trichloroethylene, and tetrachloroethylene, respectively. For dichloromethane, both intra-ion and inter-ion Cl-KIEs were studied, within the ranges of 0.820-1.020 and 0.797-1.016, respectively, showing both normal and inverse Cl-KIEs depending on collision energies. The observed Cl-KIEs generally declined from large normal to extremely large inverse values with increasing collision energies from 0 to 60 eV but were inferred to be independent of MS signal intensities. The Cl-KIEs are dominated by critical energies at low internal energies of precursor ions, resulting in normal Cl-KIEs; while at high internal energies, the Cl-KIEs are controlled by rotational barriers (or looseness/tightness of transition states), which lead to isotope-competitive reactions in dechlorination and thereby inverse Cl-KIEs. It is concluded that the Cl-KIEs may depend on critical energies, bond strengths, available internal energies, and transition state looseness/tightness. The findings of this study yield new insights into the fundamentals of Cl-KIEs of organochlorines during CID and may be conducive to elucidating the underlying mechanisms of KIEs in collision-induced and photo-induced reactions in the actual world.

Inverse Solvent Isotope Effects in Enzyme-Catalyzed Reactions.

Solvent isotope effects have long been used as a mechanistic tool for determining enzyme mechanisms. Most commonly, macroscopic rate constants such as kcat and kcat/Km are found to decrease when the reaction is performed in D2O for a variety of reasons including the transfer of protons. Under certain circumstances, these constants are found to increase, in what is termed an inverse solvent kinetic isotope effect (SKIE), which can be a diagnostic mechanistic feature. Generally, these phenomena can be attributed to an inverse solvent equilibrium isotope effect on a rapid equilibrium preceding the rate-limiting step(s). This review surveys inverse SKIEs in enzyme-catalyzed reactions by assessing their underlying origins in common mechanistic themes. Case studies for each category are presented, and the mechanistic implications are put into context. It is hoped that readers may find the illustrative examples valuable in planning and interpreting solvent isotope effect experiments.
