Preparation of pure phase silver nanoparticles: Morphological, optical, binding interactions with bovine serum albumin and antioxidant potential studies

Pure silver nanoparticles(AgNPs) were synthesized using Senecio madagascariensis plant extract and oleic acid as an additional capping agent. The powder X-ray diffraction patterns of the silver nanoparticles were indexed to the face-centred cubic phase of Ag. TEM images of the nanoparticles revealed polydisperse spherical nanoparticles in a 9 to 22 nm range. The GC-MS revealed terpenes as some phytochemicals extracted, which the FTIR bands corroborated. The results confirmed that the increase in oleic acid concentration increased the size of the silver nanoparticles. The nanoparticles' optical band gaps, Eg, were 2.26 – 2.56 eV, which decreased with particle size. The AgNPs exhibited potential antioxidant activity, as demonstrated by the DPPH scavenging assay. The DPPH scavenging activity was highest, with AgNP1 at 80% and an IC50 of 1.668 μg/mL. Fluorescence and ultraviolet (UV) titrations were used to study the interaction of silver nanoparticles (AgNP1) with bovine serum albumin (BSA). The Kapp value of the AgNP1 was calculated to be 3.14 × 104 ± 0.02 Lmol−1, KSV calculated as 7.38 × 104 ± 0.12 M−1. In the study, AgNPs bind to BSA potentially through a static quenching mechanism with one binding site that leads to developing a ground state complex. It is proposed that AgNPs bond to BSA's surface spontaneously based on determining thermodynamic characteristics and binding constants.

Fabrication of Silver Nanoparticles Using Acalypha paniculata Extract, AI-Based Interaction Analysis and Its Activity Explication

This study showcases eco-friendly silver nanoparticle synthesis using Acalypha paniculata herb and its biological activity. Artificial Intelligence (AI) tools are used to analyze the interaction between phytoconstituent and silver ion to find the capping and nanoparticle fabricating phytoconstituents. Ethanolic extract was added to silver nitrate solution and irradiated under sunlight until the formation of silver nanoparticles. These particles were sedimented, characterised by spectral technique and biological activity was measured. Finally, a Computational simulation algorithm was used to visualize the interactions of phytoconstituent and silver nanoparticles. Ultra Violet (UV)-visible spectrum revealed a visible peak at 420nm in the brownish solution, Dynamic Light Scattering (DLS) size of 54.7nm, spherical crystalline morphology of the nanoparticle by Transmission Electron Microscope (TEM) and X-Ray Diffraction (XRD) analysis. The synthesized silver nanoparticle antagonised Escherichia coli, and Bacillus cereus and showed the Minimum Inhibitory Concentration (MIC) of 65 µg/mL, and 78 µg/mL respectively and Minimum Bacterial Concentration (MBC) of 52.5 µg/mL for B.cereus and 61.5µg/mL for E.coli. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radicals IC50 was found to be 97.414 µg/mL and it revealed the dose-dependent antioxidant activity of phyto-capped silver particles. The cytotoxicity of fabricated nanoparticles was studied on Vero cells and the results indicate that IC50 of 47.28 µg/mL. Furthermore, an AI-based study concludes that Alloaromadendrene adsorbed highly by the Silver (Ag) ion compared to the nitrate with an affinity of -30.6755 kcal/mol. This research introduces an innovative, cost-effective method for stable Acalypha paniculata Silver Nanoparticles (AP-AgNPs) fabrication, crucial for diverse biomedical uses.

Size- and Shape-Dependent Interactions of Lipid-Coated Silver Nanoparticles: An Improved Mechanistic Understanding through Model Cell Membranes and In Vivo Toxicity.

The widespread use of silver nanoparticles (AgNPs) in various applications and industries has brought to light the need for understanding the complex relationship between the physicochemical properties (shape, size, charge, and surface chemistry) of AgNPs that affect their ability to enter cells and cause toxicity. To evaluate their toxicological outcomes, this study systematically analyzed a series of homogeneous hybrid lipid-coated AgNPs spanning sizes from 5 to 100 nm with diverse shapes (spheres, triangles, and cubes). The hybrid lipid membrane comprises hydrogenated phosphatidylcholine (HPC), sodium oleate (SOA), and hexanethiol (HT), which shield the AgNP surface from surface oxidation and toxic Ag+ ion release to minimize its contribution to toxicity. To reduce any significant effects by surface chemistry, the HPC, SOA, and HT membrane composition ratio was kept constant, and the AgNPs were assessed using embryonic zebrafish (Danio rerio). While a direct comparison cannot be drawn due to the lack of complementary sizes below 40 nm for triangular plates and cubes due to synthetic challenges, significant mortality was observed for spherical AgNPs (AgNSs) of 5, 20, 40, and 60 nm at 120 h postfertilization at concentrations ≥6 mg Ag/L. In contrast, the 10, 80, and 100 nm AgNSs, 40, 70, and 100 nm triangular plate AgNPs (AgNPLs), and 55, 75, and 100 nm cubic AgNPs (AgNCs) showed no significant mortality at 5 days postfertilization following exposure to AgNPs at concentrations up to 12 mg Ag/L. With constant surface chemistry on the AgNPs, size is the dominant factor driving toxicological responses, with smaller nanoparticles (5 to 60 nm) being the most toxic. Larger AgNSs, AgNCs, and AgNPLs from 75 to 100 nm do not show any evidence of toxicity. However, when closely examining sizes between 40 and 60 nm for AgNSs, AgNCs, and AgNPLs, there is evidence that discriminates shape as a driver of toxicity since sublethal responses generally were observed to follow a pattern, suggesting toxicity is most significant for AgNSs followed by AgNPLs and then AgNCs, which is the least toxic. Sum frequency generation vibrational spectroscopy showed that irrespective of size or shape, all hybrid lipid-coated AgNPs interact with membrane surfaces and "snorkel" between phases into the lipid monolayer with minimal energetic cost. These findings decisively demonstrate that not only smaller AgNPs but also the shape of the AgNPs influences their biological compatibility.

Ionic and nanoparticulate silver alleviate the toxicity of inorganic mercury in marine microalga Chaetoceros muelleri.

Toxicological effects of silver nanoparticles (SNPs) in different organisms have been studied; however, interactions of SNPs with other environmental pollutants such as mercury are poorly understood. Herein, bioassay tests were performed according to ΟECD 201 guideline to assess the toxic effects induced by mercury ions (mercury chloride, MCl) on the marine microalga Chaetoceros muelleri in the presence of SNPs or silver ions (silver nitrate, SN). Acute toxicity tests displayed that the presence of SNPs or SN (0.01mg L-1) significantly reduced the toxicity of MCl (0.001, 0.01, 0.1, 1, 10, and 100mg L-1) and increased the IC50 of MCl from 0.072 ± 0.014 to 0.381 ± 0.029 and 0.676 ± 0.034mg L-1, respectively. In the presence of SN or SNPs, the mercury-reducing effect on algal population growth significantly decreased. Considering the increase of IC50, the mercury toxicity decreased approximately 5.44 and 9.66 times in the presence of SNPs or SN, respectively. The chlorophyll a and c contents decreased at all exposures; however, the decrease by MCl-SNPs and MCl-SN was significantly less than MCl except at 1mg L-1. The lowering effect of MCl-SN on chlorophyll contents was less than MCl and MCl-SNPs. MCl exposure induced significant raises in total protein content (TPC) at concentrations < 0.01mg L-1, with a maximum of ~ 70.83% attained at 100mg L-1. The effects of MCl-SNPs and MCl-SN on TPC were significantly less than MCl. Total lipid content (TLC) at all MCl concentrations was higher than the control, while at coexposure to MCl-SN, TLC did not change until 0.01mg L-1 compared with the control. The effects of MCl-SN and MCL-SNPs on TPC and TLC were in line with toxicity results, and were significantly less than those of MCl individually, confirming their antagonistic effects on MCl. The morphological changes of algal cells and mercury content of the cell wall at MCl-SN and MCl-SNPs were mitigated compared with MCl exposure. These findings highlight the mitigatory impacts of silver species on mercury toxicity, emphasizing the need for better realizing the mixture toxicity effects of pollutants in the water ecosystem.

Solid polymer electrolyte and antimicrobial performance of Polyvinyl alcohol/Silver nanoparticles composite film

A nanocomposite based on polyvinyl alcohol (PVA) is developed using silver nanoparticles (Ag NPs) prepared by the chemical reduction method. The nanocomposite is analyzed by X-ray diffraction method (XRD) and Fourier transform infrared (FTIR) analyses. XRD analysis confirms the purity of the synthesized silver nanoparticles with the average crystallite size of 15 nm. It also shows the presence of nanoparticles in the nanocomposite films. The interaction of silver nanoparticles with the polymer matrix is demonstrated by the FTIR spectrum. Scanning electron microscope (SEM), Energy Dispersive X-ray Spectroscopy (EDAX) and UV–Visible spectroscopy confirm the formation of silver nanoparticles with spherical morphology and surface Plasmon resonance at 424 nm. Further, nanocomposite as solid polymer electrolyte and antimicrobial properties of the nanocomposite are illustrated. The addition of nanoparticles increases the electrical conductivity (1.7866 × 10-6 Scm−1) of the PVA silver nanocomposite film that is measured from electrochemical impedance spectroscopy through Cole-Cole plots. The PVA/Silver nanoparticles composite films show a high zone of inhibition for both Gram-negative and Gram-positive bacteria than pure PVA films. The electrochemical performance shows the ability to use as a solid polymer electrolyte in electrochemical devices. Polyvinyl alcohol (PVA)/Silver nanoparticles composite films with high anti-bacterial activity can be used for wound dressings and active food packaging.

Interaction of silver nanoparticles with marine/lake snow in early formation stage.

Marine and lake snows play an important ecological role in aquatic systems, and recent researches have also revealed their interactions with various pollutants. In this paper, the interaction of silver nanoparticles (Ag-NPs), a typical nano-pollutant, with marine/lake snow in the early formation stage was investigated by roller table experiments. Results indicated Ag-NPs promoted the accumulation of larger marine snow flocs while inhibited the development of lake snow. The promotion effect of AgNPs might be attributed to their oxidative dissolution into low-toxic silver chloride complexes in seawater, and the subsequent incorporation into marine snow, which would enhance the rigidity and strength of larger flocs and favor the development of biomass. Conversely, Ag-NPs mainly existed in the form of colloidal nanoparticles in lake water and their strong antimicrobial effect suppressed the growths of biomass and lake snow. In addition, Ag-NPs could also affect the microbial community of marine/lake snow, including impact on microbial diversity, and elevation on abundances of extracellular polymeric substances (EPS) synthesis genes and silver resistance genes. This work has deepened our understanding of the fate and ecological effect of Ag-NPs via the interaction with marine/lake snow in aquatic environments.

Cytochrome c - silver nanoparticle interactions: Spectroscopy, thermodynamic and enzymatic activity studies

Cytochrome c, an iron containing metalloprotein in the mitochondria of the cells with an oxide/redox property, plays key role in the cell apoptotic pathway. In this study, the interaction of silver nanoparticles (AgNPs) with cytochrome c (Cyt c) was investigated by using a combination of spectroscopic, imaging and thermodynamic techniques, including dynamic light scattering (DLS), ultraviolet–visible (UV–vis) spectroscopy, transmission electron microscopy (TEM), fluorescence spectroscopy, near and far circular dichroism (CD) spectroscopy, and isothermal titration calorimetry (ITC). DLS and UV–vis analysis evidenced the formation of surface complexes of Cyt c on AgNPs. The saturation of surface coverage of AgNPs was observed at 4.36 Cyt c molecules per nm2 of AgNPs. The surface complexation resulted in a promotion of the Ag dissolution overtime. The negative sign of enthalpic (ΔH) contribution suggested that electrostatic forces are indicative forces in the interaction between protein and AgNPs. Moreover, the fluorescence spectra revealed that the conformation of protein was altered around tryptophan (Trp) and tyrosine (Tyr) residues indicating the alteration of the tertiary structure of Cyt c. CD analysis evidenced that the secondary structure of Cyt c was modified under AgNPs-Cyt c interactions and the binding of Cyt c onto AgNPs resulted in remarkable structural perturbation around the active site heme, which in turn alter the protein enzymatic activity. The results of the present study contributed to a deeper insight on the mechanisms of interaction between NPs and biomacromolecules and could help establish the in vivo fate of AgNPs on cellular redox homeostasis.

SERS of Human Red Blood Cells in Non-Resonant Conditions: Benefits, Limitations, and Complementary Tools (CytoViva and GFAAS)

Herein, Raman and surface-enhanced Raman spectroscopies (SERS) were successfully employed to establish the chemical interactions of citrate-capped silver nanoparticles (AgNPs, 10–15 nm) with human red blood cells (RBCs). The Raman/SERS spectra offered spectral evidence for the cellular uptake of AgNPs and the subsequent change in the conformation of the most abundant component, hemoglobin (Hb), from oxyhemoglobin to deoxyhemoglobin. The spectral characterization of AgNPs’ interactions with other RBC biomarkers (membrane proteins and lipids) was impeded by the dominant Hb bands, even for non-resonant Hb conditions. CytoViva hyperspectral imaging and graphite furnace atomic absorption spectroscopy (GFAAS) served as complementary tools to effectively address the challenges related to a single excitation line (632.8 nm) and the resolution of the confocal Raman microscope (0.5–1.0 µm). CytoViva confirmed the RBC-AgNP interactions through hyperspectral signatures and facilitated the label-free localization of AgNPs extracellularly and intracellularly. Irreversible agglutination of RBCs was noted after 24 h of exposure, raising concerns about the toxicity of AgNPs of biocompatible citrate coatings. GFAAS validated the Raman/SERS results by quantifying the proportion of AgNPs absorbed by RBCs, which was significant (~48% AgNPs by mass), mostly at the membrane (60% RBCs), and size dependent (no large AgNPs or AgNP-aggregates in RBCs, after 12–24 h).

Comparative Analysis of the Interaction of Silver Nanoparticles with Hexokinase from Trypanosoma brucei and Humans.

Regardless of the efforts to ease cases of human African trypanosomiasis (HAT), an increased number of cases get reported annually. This is because of drug resistant Trypanosoma brucei (Tb), the causative agent of the illness. This has renewed the need for creative methods to find new anti-trypanosomal drugs. The blood stream form (BSF) of the parasite depends exclusively on the glycolytic pathway for energy production while it is in the human host. Interruptions in this pathway efficiently kills the parasite. Trypanosoma brucei hexokinase (TbHK) is the first enzyme in glycolysis, and any effectors or inhibitors of TbHK would have potential as anti-trypanosomal agents. TbHK and human glucokinase (hGCK) were over-expressed with a 6 histidine-tag in E. coli BL21(DE3) cells having the pRARE2 plasmid. TbHK had thermal and pH stability between 30°C and 55°C and 7.5 and 8.5, respectively, while hGCK exhibited thermal and pH stability between 30°C and 40°C and 7.0 and 8.0, respectively. Kinetically, TbHK had a Km of 39.3 µM, Vmax of 0.066 µmol.min-1.mL-1, kcat of 2.05 min-1 and kcat/Km of 0.0526 min-1.µmol-1. hGCK exhibited a Km of 4.5 µM, Vmax of 0.032 µnmol.min-1.mL-1, kcat of 11.25 min-1, and kcat/Km of 2.5 min-1.µmol-1. Interaction kinetic studies of silver nanoparticles (AgNPs) (0.1 µM) of average size of 6 nm with TbHK and hGCK were conducted. AgNPs selectively inhibited TbHK over hGCK. TbHK showed a non-competitive inhibition with a 50% and 28% decrease in Vmax, and kcat/km, respectively. HsGCK showed a 33% increase in affinity, 9% decrease in Vmax, and a 50% increase in enzyme efficiency. The observed pattern of hGCK and AgNPs falls under the uncompetitive inhibition. The observed highly selective inhibitory effects of AgNPs between TbHK and hGCK may be used in development of new anti-trypanosomal drugs.

Silver nanoparticle interactions with glycated and non-glycated human serum albumin mediate toxicity.

Introduction: Biomolecules bind to and transform nanoparticles, mediating their fate in biological systems. Despite over a decade of research into the protein corona, the role of protein modifications in mediating their interaction with nanomaterials remains poorly understood. In this study, we evaluated how glycation of the most abundant blood protein, human serum albumin (HSA), influences the formation of the protein corona on 40nm silver nanoparticles (AgNPs) and the toxicity of AgNPs to the HepG2 human liver cell line. Methods: The effects of glycation on AgNP-HSA interactions were quantified using circular dichroism spectroscopy to monitor protein structural changes, dynamic light scattering to assess AgNP colloidal stability, zeta potential measurements to measure AgNP surface charge, and UV-vis spectroscopy and capillary electrophoresis (CE) to evaluate protein binding affinity and kinetics. The effect of the protein corona and HSA glycation on the toxicity of AgNPs to HepG2 cells was measured using the WST cell viability assay and AgNP dissolution was measured using linear sweep stripping voltammetry. Results and Discussion: Results from UV-vis and CE analyses suggest that glycation of HSA had little impact on the formation of the AgNP protein corona with protein-AgNP association constants of ≈2x107 M-1 for both HSA and glycated HSA (gHSA). The formation of the protein corona itself (regardless of whether it was formed from HSA or glycated HSA) caused an approximate 2-fold decrease in cell viability compared to the no protein AgNP control. While the toxicity of AgNPs to cells is often attributed to dissolved Ag(I), dissolution studies showed that the protein coated AgNPs underwent less dissolution than the no protein control, suggesting that the protein corona facilitated a nanoparticle-specific mechanism of toxicity. Overall, this study highlights the importance of protein coronas in mediating AgNP interactions with HepG2 cells and the need for future work to discern how protein coronas and protein modifications (like glycation) may alter AgNP reactivity to cellular organisms.

Exploring the In‐Vitro Antibacterial Activity and Protein (Human Serum Albumin, Human Hemoglobin and Lysozyme) Interaction of Hexagonal Silver Nanoparticle Obtained from Wood Extract of Wild Cherry Shrub

AbstractHere, the silver nanoparticles were synthesized using aqueous wood extract of cerasus macrocarpa (wild Cherry) shrub. This green synthesis method was both eco‐friendly and straightforward. The nanoparticles were characterized using FT‐IR, zeta potential, ultraviolet‐visible, TEM, and SEM‐EDX analysis. Green synthesized AgNPs demonstrated effective antibacterial activity against gram‐positive and gram‐negative bacteria (S. aureus and E. coli). Furthermore, fluorometric, circular dichroism and UV‐vis were used to study the interaction of green synthesized AgNPs with Human serum albumin (HSA (, Human hemoglobin (HHb), and Lysozyme (Lys) in simulated physiological conditions. The studies of interaction with HSA, HHb and Lys show good binding affinity for green synthesized silver nanoparticles; the binding affinity was found in the order Lys&gt;HSA&gt;HHb at 298.15 K. The results show that the native conformation of HSA, HHb, and Lys was preserved at the secondary structure level, which is crucial in providing insights into the side effects of newly synthesized drugs on their carriers. The fluorescence data represented that green synthesized AgNPs quench the intrinsic fluorescence of proteins through a static quenching procedure.

Preparation of Ag/C Nanocomposites Based on Silver Maleate and Their Use for the Analysis of Iodine Ions

In recent decades, metal-containing nanocomposites have attracted considerable attention from researchers. In the present study, a detailed analysis of the preparation of Ag/C nanocomposites through the thermolysis of silver maleate was carried out. Thermolysis products are nanocomposites containing silver nanoparticles (NPs) uniformly distributed in a stabilizing carbon matrix. The composition, structure, and properties of the obtained nanocomposites were studied using IR-spectroscopy, X-ray diffraction (XRD), atomic force microscopy (AFM), scanning electron microscopy (SEM), transmission electron microscopy (TEM), and energy dispersive X-ray spectroscopy (EDS). This article reports on the possibility of using Ag/C nanocomposites to create new indicator papers that are sensitive to iodide ions in the concentration range of 0.03–1.6 mg/L (0.24–12.6 μM). The developed papers are used in a technique based on the oxidation of iodides with the formation of molecular iodine, which is extracted in an air stream and transferred to a sensitive paper layer containing silver NPs. The interaction of silver NPs with iodine leads to optical changes that can be tracked using a conventional scanner.

Quantification of silver nanoparticle interactions with yeast Saccharomyces cerevisiae studied using single-cell ICP-MS.

Silver nanoparticles (AgNPs) have been used in many fields due to their anticancer, antimicrobial, and antiviral potential. Single-cell ICP-MS (SC-ICP-MS) is an emerging technology that allows for the rapid characterization and quantification of a metal analyte across a cell population in a single analysis. In this study, a new rapid and sensitive SC-ICP-MS method was developed to quantitatively study the interactions of AgNPs with yeast Saccharomyces cerevisiae. The method can quantify the cell concentration, silver concentration per cell, and profile the nanoparticle distribution in a yeast cell population. AgNP dosing time, concentration, and AgNP size were quantitatively evaluated for their effects on AgNP-yeast cell interactions. The results showed that the initial uptake of AgNPs was rapid and primarily driven by the mass of Ag per cell. The optimal dosing particle concentrations for highest uptake were approximately 1820, 1000, and 300 AgNPs/cell for 10, 20, and 40 nm AgNPs, respectively. Furthermore, this study also validated a washing method for the application to a microorganism for the first time and was used to quantitatively determine the amount of cell surface–adsorbed AgNPs and intracellular AgNPs. These results indicated that the mass (Ag in ag/cell) ratios of intracelluar vs cell surface-adsorbed AgNPs were similar for different AgNP sizes. This high throughput and ultrasensitive SC-ICP-MS method is expected to have many potential applications, such as optimization of methods for green synthesis of AgNPs, nanotoxicity studies, and drug delivery. In vitro study on the immunomodulatory effects of differently functionalized silver nanoparticles on human peripheral blood mononuclear cells.

The interaction of silver nanoparticles (AgNPs) with the immune system has not yet been sufficiently elucidated even though they belong to the most investigated and exploited group of nanomaterials. This study aimed to evaluate immunomodulatory effect of four different AgNPs on human peripheral blood mononuclear cells (hPBMCs). Fresh hPBMCs were exposed to the small sized (~ 10 nm) AgNPs immediately after isolation from the whole blood of healthy volunteers. The study considered coating-, time- and dose-dependent response of hPBMSc and stimulation of both early and intermediate activation of lymphocytes and monocytes using flow cytometry. The AgNPs differed in surface charge and were stabilised with polyvinyl pyrrolidone (PVP), poly-L-lysine (PLL), bis(2-ethylhexyl) sulfosuccinate sodium (AOT) or blood serum albumin (BSA). Response of hPBMCs to coating agents and ionic Ag form was evaluated to distinguish their effect from the AgNPs action as they may be released from the nanosurface. There was no significant effect of any tested AgNPs on relative count of hPBMCs subpopulations. The T-cells and monocytes were not activated after treatment with AgNPs, but the highest concentration of PLL- and BSA-AgNPs decreased density of CD4 and CD8 markers on T-helper and T-cytotoxic cells, respectively. The same AgNPs activated B- and NK-cells. Ionic Ag activated T-, B- and NK-cells, but at very higher concentration, whereas only PLL exhibited immunomodulatory activity. Green synthesis of silver nanoparticles by Cassytha filiformis L. extract and its characterization

Nanotechnology is a new area of research with new possibilities in various fields. Nanotechnology is a having the potential to take forward the agriculture with new tools which promises to increase food production and also protect the agriculture crops from pests, diseases etc. The present study focuses on the green synthesis of silver nanoparticles using plant extracts of Cassytha filiformis. Aqueous silver ions when exposed to plant leaf extract were reduced and resulted in a color change to dark brown indicating the formation of silver nanoparticles by using UV–Vis Spectroscopy which showed peak at in range from 400 nm to 480 nm which are characteristic peaks of silver nanoparticles. Synthesized silver nanoparticles were also characterized by other techniques such as Field Emission Scanning Electron Microscopy (FESEM) showed even shaped spherical nanoparticles with particle size of 42 nm. Energy Dispersive X-Ray (EDX) showed absorption peak approximately at 3 keV. Fourier Transform Infra Red (FTIR) showed chemical interactions of silver nanoparticles and Atomic force microscope (AFM) depicted the three dimensional structure of silver nanoparticle.

Implementation of a Dynamic Co-Culture Model Abated Silver Nanoparticle Interactions and Nanotoxicological Outcomes In Vitro.

The incorporation of engineered nanoparticles (NPs) into everyday consumer goods, products, and applications has given rise to the field of nanotoxicology, which evaluates the safety of NPs within biological environments. The unique physicochemical properties of NPs have made this an insurmountable challenge, as their reactivity and variable behavior have given rise to discrepancies between standard cell-based in vitro and animal in vivo models. In this study, enhanced in vitro models were generated that retained the advantages of traditional cell cultures, but incorporated the modifications of (1) inclusion of an activated immune element and (2) the presence of physiologically-relevant dynamic flow. Following verification that the human alveolar epithelial and macrophage (A549/U937) co-culture could be successfully sustained under both static and dynamic conditions, these cultures, in addition to a standard A549 static model, were challenged with 10 nm citrate coated silver NPs (AgNPs). This work identified a reshaping of the AgNP-cellular interface and differential biological responses following exposure. The presence of dynamic flow modified cellular morphology and reduced AgNP deposition by approximately 20% over the static exposure environments. Cellular toxicity and stress endpoints, including reactive oxygen species, heat shock protein 70, and secretion of pro-inflammatory cytokines, were found to vary as a function of both cellular composition and flow conditions; with activated macrophages and fluid flow both mitigating the severity of AgNP-dependent bioeffects. This work highlights the possibility of enhanced in vitro systems to assess the safety of engineered NPs and demonstrates their effectiveness in elucidating novel NP-cellular interactions and toxicological profiles.

The Impact of Graphite Oxide Nanocomposites on the Antibacterial Activity of Serum.

Nanoparticles can interact with the complement system and modulate the inflammatory response. The effect of these interactions on the complement activity strongly depends on physicochemical properties of nanoparticles. The interactions of silver nanoparticles with serum proteins (particularly with the complement system components) have the potential to significantly affect the antibacterial activity of serum, with serious implications for human health. The aim of the study was to assess the influence of graphite oxide (GO) nanocomposites (GO, GO-PcZr(Lys)2-Ag, GO-Ag, GO-PcZr(Lys)2) on the antibacterial activity of normal human serum (NHS), serum activity against bacteria isolated from alveoli treated with nanocomposites, and nanocomposite sensitivity of bacteria exposed to serum in vitro (using normal human serum). Additionally, the in vivo cytotoxic effect of the GO compounds was determined with application of a Galleria mellonella larvae model. GO-PcZr(Lys)2, without IR irradiation enhance the antimicrobial efficacy of the human serum. IR irradiation enhances bactericidal activity of serum in the case of the GO-PcZr(Lys)2-Ag sample. Bacteria exposed to nanocomposites become more sensitive to the action of serum. Bacteria exposed to serum become more sensitive to the GO-Ag sample. None of the tested GO nanocomposites displayed a cytotoxicity towards larvae.
