Lipoxygenase Inhibitory Activities of Some Plant Extracts and Chemical Compounds

Lipoxygenases (LOXs, linoleate: oxygen reductase, E.C. 1.13.11.12) are a family of non-heme ironcontaining dioxygenases. LOXs are associated with several inflammation-related diseases such as arthritis, asthma, cardiovascular, kidney, skin and allergic diseases, neurodegenerative disorders, cancer and metabolic syndrome. In this study, the inhibitory effects of ethanolic extracts prepared from 12 different plants and 12 different chemical compounds were investigated on the activity of LOX which has an important value in the health area. All the plant extracts and chemical substances used in our study showed LOX inhibitory effect. The enzyme inhibitory activities of the extracts and chemical compounds were increasing in a dose-dependent manner. The results obtained in the present study indicate that plant extracts and chemical compounds examined can be a potential source of novel antiinflammatory therapeutics.

Relationship between total phenolic contents and biological properties of propolis from 20 different regions in South Korea

BackgroundPropolis (or bee glue), collected from botanical sources by honey bee, has been used as a popular natural remedies in folk medicine throughout the world. This study was conducted to assess growth inhibitory effects of ethanol extracts of propolis (EEPs) from 20 different regions in South Korea on human intestinal bacteria as well as their human β-amyloid precursor cleavage enzyme (BACE-1), acetylcholinesterase (AChE) inhibitory, antioxidant, antiproliferative, and anti-human rhinovirus activities.MethodsThe Bonferroni multiple-comparison method was used to test for significant differences in total polyphenol and flavonoid contents among EEP samples using SAS 9.13 program. Correlation coefficient (r) analysis of the biological activities of EEP samples was determined using their 50 % inhibition concentration or minimal inhibitory concentration values and their polyphenol or flavonoid contents in 20 native Korean EEP samples.ResultsThe amounts of total polyphenol and flavonoids in the Korean EEP samples ranged from 49 to 239 mg gallic acid equivalent (GAE)/g EEP (Brazilian, Chinese, and Australian samples, 127–142 mg GAE/g EEP) and from 21 to 50 mg quercetin equivalent (QE)/g EEP (Brazilian, Chinese, and Australian samples, 33–53 mg QE/g EEP), respectively. Correlation coefficient analysis showed that total polyphenol contents may be negatively correlated with 2,2-diphenyl-1-picrylhydrazyl free radical scavenging activity (r = −0.872) and total flavonoid content has no correlation with the activity (r = 0.071). No direct correlation between BACE-1 inhibition, AChE inhibition, or antiproliferative activity and total polyphenol or total flavonoid content in Korean EEP samples was found. Gram-positive and Gram-negative bacteria were observed to have different degrees of antimicrobial susceptibility to the EEP samples examined, although ciprofloxacin susceptibility among the bacterial groups did not differ greatly.ConclusionsFurther studies will warrant possible applications of propolis as potential therapeutic BACE-1 blocker, antioxidant, antiproliferative agent, and antimicrobial agent.

INHIBITORY EFFECT OF ETHANOLIC EXTRACT OF CURANGA FEL-TERRAE (PUGUN TANO) LEAVES ON ACETYLCHOLINE MUSCARINIC-3 RECEPTORS INDUCED ON ISOLATED GUINEA PIG TRACHEAL

ABSTRACTObjective: The study was aimed to investigate the inhibitory effect of ethanolic extract of Curanga fel-terrae leaves (EECFL) against acetylcholine(ACh)-induced contraction of the ACh-muscarinic-3 receptor.Materials and Methods: The study of the inhibitory effect of the ethanolic extract on the contraction by ACh concentration series (10 M) wasconducted in vitro using isolated guinea pig tracheal organ in the Krebs solution.Results: Early incubation of tracheal organ with EECFL (0.5, 1, 2 and 4 mg/ml) before contracted by the series of ACh concentration produces thedecrease of ACh contraction. The concentration series of ACh sigmoid curve rightward shift without decreasing the maximal contraction. The resultsof double-reciprocal plot of ACh shows the mean value of the 1/y-intercept of each extract was not different with the control.Conclusion: The EECFL showed competitive antagonist effect on ACh-muscarinic-3 receptors.Keywords: Curanga fel-terrae, Pugun tano, Acetylcholine-muscarinic-3 receptors, Competitive antagonist, Ethanolic extract.−8-10−3

Inhibitory Effect of Ethanolic Extract Isolated from Radix Pulasatillae Seeds against Proliferation of A549 Human Lung Cancer Cells

The present study was evaluated the anticancer activity of Radix Pulsatillae ethanol extract at first by using high throughput screening HTS for herb fractions screening in concentration of 100μM to identify the active fraction andthen we measured the cytotoxic effects by using morphology changes, and detected apoptosis by Hoechst 33258staining. Our results showed that Radix Pulsatillae ethanol extract has a strong toxicity against A549 lung cancer cells and low toxicity with mice normal splenocytes and requires more investigation to identify the small natural molecule with new anticancer activity and further mechanistic for cancer pathways.

In vitro inhibitory effects of ethanol extract of Danshen (Salvia miltiorrhiza) and its components on the catalytic activity of soluble epoxide hydrolase

Background: Soluble epoxide hydrolase (sEH) has been demonstrated to be a key enzyme involved in the pathologic development of several cardiovascular diseases and inflammation, and inhibition of sEH is therefore very helpful or crucial for the treatment of ischemia-reperfusion injury, cardiac hypertrophy, hypertension and inflammation. Danshen, the dried root of Salvia miltiorrhiza (Fam. Labiatae), has been used for the treatment of cardiovascular and cerebrovascular diseases in China and other countries for hundreds of years. Recent studies indicated that Danshen and its preparations also have potential for the management of inflammation. However, little information is available about the possibility of Danshen and its components on sEH inhibition.Purpose and methods: Danshen extracts and its constituents were tested for sEH inhibition using its physiological substrate, 8,9-EET, based on a LC–MS/MS assay in this study.Results: Among the tested 15 compounds, tanshinone IIA and cryptotanshinone were found to be the potent (Ki = 0.87 μM) and medium (Ki = 6.7 μM) mixed-type inhibitors of sEH, respectively. Salvianolic acid C (Ki = 8.6 μM) was proved to be a moderate noncompetitive sEH inhibitor. In consistent with the inhibition results of the pure compounds, the 75% ethanol extract of Danshen (EE, IC50 = 86.5 μg/ml) which contained more tanshinone IIA and cryptotanshinone exhibited more potent inhibition on sEH than the water extract (WE, IC50 > 200 μg/ml) or 1 M NaHCO3 (BE, IC50 > 200 μg/ml) extract.Conclusion: These data indicated that using the ethanol fraction of Danshen and increasing the amounts of tanshinone IIA, cryptotanshinone and salvianolic acid C, especially the contents of tanshinone IIA in Danshen extract or preparations to enhance the inhibitory effects on sEH might be efficient ways to improve its cardiovascular protective and anti-inflammatory effects, and that herbal medicines could be an untapped reservoir for sEH-inhibition agents and developing sEH inhibitors from the cardiovascular protective and anti-inflammatory herbs is a promising approach.

Dalbergia odorifera Extract Ameliorates UVB-Induced Wrinkle Formation by Modulating Expression of Extracellular Matrix Proteins.

Preclinical Research Emerging evidence suggests that Dalbergia odorifera T. Chen (Leguminosae), an indigenous medicinal herb, has therapeutic potential. This study examined the antiwrinkle effects of ethanol extracts of D. odorifera in UVB-irradiated human skin cells. Ethanol extracts of D. odorifera and thier constituents, dalbergin and sativanone, induced expression of collagen type I and transforming growth factor (TGF)-β1 in human dermal fibroblasts. In HR-1 hairless mice exposed to UVB, the ethanol extract reduced wrinkle formation and skin thickness. This inhibitory effect of ethanol extract was associated with the restoration of collagen type I, TGF-β1, and elastin to levels approaching those in skin tissues not exposed to UVB, which was accompanied by the reduction of matrix metalloproteinase-2 and upregulation of tissue inhibitors of metalloproteinase (TIMP)-2 and TIMP-3 in skin tissue exposed to UVB. These results suggest that the ethanol extracts prevent some effects of photoaging and maintain skin integrity by regulating the degradation of the extracellular matrix proteins. © 2015 Wiley Periodicals, Inc.

Inhibitory activities of Perilla frutescens britton leaf extract against the growth, migration, and adhesion of human cancer cells.

BACKGROUND/OBJECTIVESPerilla frutescens Britton leaves are a commonly consumed vegetable in different Asian countries including Korea. Cancer is a major cause of human death worldwide. The aim of the current study was to investigate the inhibitory effects of ethanol extract of perilla leaf (PLE) against important characteristics of cancer cells, including unrestricted growth, resisted apoptosis, and activated metastasis, using human cancer cells.MATERIALS/METHODSTwo human cancer cell lines were used in this study, HCT116 colorectal carcinoma cells and H1299 non-small cell lung carcinoma cells. Assays using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide were performed for measurement of cell growth. Soft agar and wound healing assays were performed to determine colony formation and cell migration, respectively. Nuclear staining and cell cycle analysis were performed for assessment of apoptosis. Fibronectin-coated plates were used to determine cell adhesion.RESULTSTreatment of HCT116 and H1299 cells with PLE resulted in dose-dependent inhibition of growth by 52-92% (at the concentrations of 87.5, 175, and 350 µg/ml) and completely abolished the colony formation in soft agar (at the concentration of 350 µg/ml). Treatment with PLE at the 350 µg/ml concentration resulted in change of the nucleus morphology and significantly increased sub-G1 cell population in both cells, indicating its apoptosis-inducing activity. PLE at the concentration range of 87.5 to 350 µg/ml was also effective in inhibiting the migration of H1299 cells (by 52-58%) and adhesion of both HCT116 and H1299 cells (by 25-46%).CONCLUSIONSThese results indicate that PLE exerts anti-cancer activities against colon and lung cancers in vitro. Further studies are needed in order to determine whether similar effects are reproduced in vivo.

Synergistic interaction of extracts of garlic (Allium sativum) and propolis against methicillin-resistant Staphylococcus aureus

Methicillin-resistant Staphylococcus aureus (MRSA) is a public health problem, being a cause of severe diseases in hospitals and communities in general. To confront this contingency at present, the effectiveness, in combination with diverse natural products is being studied in order to inhibit this microorganism. The objective of the present work was to evaluate the combined inhibitory effects of ethanolic extracts of garlic (Allium sativum) and propolis (Propolis –ppl-) against MRSA strains. We tested two types of extracts: at 20 and 30% for each. Microbial resistance assays were evaluated by the macrodilution method and the combinations were assessed by isobolographic studies. The study strains were divided into two groups based on their resistance to garlic as sensitive (36.8 ± 7.4 mg/mL) and resistant (67.2 ± 8.9 mg/mL). The results show that for strains catalogued as sensitive, the combinations in both concentrations suggest a synergistic effect; on the other hand, for strains catalogued as resistant, the combinations no longer presented a synergic effect. Inhibitory effect of ethanol extract of Ocimum sanctum on osteopontin mediated metastasis of NCI-H460 non-small cell lung cancer cells.

BackgroundOsteopontin (OPN) is one of important molecular targets in cancer progression, metastasis as a calcium-binding, extracellular-matrix-associated protein of the small integrin-binding ligand and, N-linked glycoprotein. In the present study, anti-metastatic mechanism of ethanol extracts of Ocimum sanctum (EEOS) was elucidated on OPN enhanced metastasis in NCI-H460 non- small cell lung cancer cells.MethodsCell viability was measured by MTT assay. Adhesion and invasion assays were carried out to see that EEOS inhibited cell adhesion and invasion in OPN treated and non-treated NCI-H 460 cells. RT-PCR was used to determine the mRNA levels of uPA, uPAR, and EGFR.ResultsEEOS significantly inhibited cell adhesion and invasion in OPN treated and non treated NCI-H460 cells, though EEOS did not show any toxicity up to 200 μg/ml. EEOS effectively attenuated the expression of OPN and CD44 and also OPN activated the expression of CD44 in NCI-H460 cells. In addition, EEOS effectively suppressed the expression of phosphatidylinositide 3-kinases (PI3K) and cyclooxygenase 2 (COX-2) and the phosphorylation of Akt at protein level in OPN treated NCI-H460 cells. Also, EEOS significantly attenuated the expression of urokinase plasminogen activator (uPA), its receptor (uPAR) and epidermal growth factor receptor (EGFR) at mRNA level and reduced vascular endothelial growth factor (VEGF) production and MMP-9 activity in OPN treated NCI-H460 cells. Furthermore, PI3K/Akt inhibitor LY294002 enhanced anti-metastatic potential of EEOS to attenuate the expression of uPA and MMP-9 in OPN treated NCI-H 460 cells.ConclusionOverall, our findings suggest that anti-metastatic mechanism of EEOS is mediated by inhibition of PI3K/Akt in OPN treated NCI-H460 non-small cell lung cancer cells.

Antimicrobial Effects of Folk Medicinal Plants From the North of Iran Against Mycobacterium tuberculosis

Background: Medicinal plants have been used traditionally in Golestan province (north of Iran), against Mycobacterium tuberculosis or the clinical signs of tuberculosis (TB). Objectives: This study aimed to define the inhibitory effects of ethanolic extracts of six of these medicinal plants against Mycobacterium tuberculosis. Materials and Methods: Peganum harmala (seed extract), Punica granatum (peel extract), Digitalis sp. (leaf extract), fruit extract of Citrus lemon, Rosa canina and Berberis vulgaris were extracted in ethanol and their activity against M. tuberculosis isolates were determined by the agar diffusion method. The zone of inhibition (at 200 to 1.6 mg/mL) was measured and the results were compared with isoniazid and rifampin as standard positive controls. Also the concentration of vitamin C of each the extracts was evaluated. Results: The ethanolic extract of Peganum harmala seed and Punica granatum peel exhibited potential activity against all M. tuberculosis isolates with mean inhibitory zone of 18.7 and 18.8 mm, at 200 mg/mL concentration. The mean inhibitory zone around isoniazid and rifampinwere 19.2 and 18.8 mm. Ethanolic extract of Citrus lemon showed moderate inhibitory activity only against sensitive (non MDR; non multi drug resistant) strains of M. tuberculosis, and Digitalis sp. showed inhibitory effects on five isolates. Ascorbic acid content was 43.3 mg/dL in Punica granatum and Digitalis sp. and only 9.1 mg/dL in ethanolic extract of Peganum harmala. Conclusions: The highest content of vitamin C was observed in the extract of Punica granatum, which was observed to be highly active against Mycobacterium tuberculosis, while the P. harmala must have contained other phytochemical constituents that contributed to the anti-tuberculosis effects of this plant. Our findings showed that ethanolic extracts of P. granatum and P. harmala had anti-TB effects comparable to isoniazid and rifampin and can be good candidates for novel and safe natural products against tuberculosis.

Saussurea lappa extract suppresses TPA-induced cell invasion via inhibition of NF-κB-dependent MMP-9 expression in MCF-7 breast cancer cells.

BackgroundSaussurea lappa (SL) has been used as a traditional herbal medicine to treat abdominal pain and tenesmus, and has been suggested to possess various biological activities, including anti-tumor, anti-ulcer, anti-inflammatory, anti-viral, and cardiotonic activities. The effect of SL on breast cancer metastasis, however, is unknown. Cell migration and invasion are crucial in neoplastic metastasis. Matrix metalloproteinase-9 (MMP-9), which degrades the extracellular matrix, is a major component in cancer cell invasion.MethodsCell viability was examined by MTT assay, whereas cell motility was measured by invasion assay. Western blot, Real-time PCR, and Zymography assays were used to investigate the inhibitory effects of ESL on matrix metalloproteinase-9 (MMP-9) expression level in MCF-7 cells. EMSA confirmed the inhibitory effects of ESL on DNA binding of NF- κB in MCF-7 cells.ResultsCells threated with various concentrations of Saussurea lappa (ESL) for 24 h. Concentrations of 2 or 4 μM did not lead to a significant change in cell viability or morphology. Therefore, subsequent experiments utilized the optimal non-toxic concentration (2 or 4 μM) of ESL. In this study, we investigated the inhibitory effect of ethanol extract of ESL on MMP-9 expression and cell invasion in 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced MCF-7 cells. ESL inhibited the TPA-induced transcriptional activation of nuclear factor-kappa B (NF-κB). However, this result obtained that ESL did not block the TPA-induced phosphorylation of the kinases: p38, ERK, and JNK. Therefore, ELS-mediated inhibition of TPA-induced MMP-9 expression and cell invasion involves the suppression of NF-kB pathway in MCF-7 cells.ConclusionsThese results indicate that ELS-mediated inhibition of TPA-induced MMP-9 expression and cell invasion involves the suppression of NF-kB pathway in MCF-7 cells. Thus, ESL has potential for controlling breast cancer invasiveness in vitro.

Antimutagenicity and Anticancer Effects of Biebersteinia multifida DC

Aims: The purpose of this research is to examine antimutagenicity and anticancer effect of Biebersteinia multifida. Study Design: Currently cancer is considered as one of the main factors of mortality globally. Many chemicals in our environment can cause genetic mutations and are potentially responsible for millions of cancer-related deaths. Nowadays the scientists are looking for food materials which can potentially prevent the cancer occurrence. Place and Duration of Study: Biebersteinia multifida DC. dried roots were purchased from local market in Tehran bazar, the center of Tehran province, Iran, between June 2012 and January 2013. Methodology: In this study human leukemia pre B-cells (Nalm-6) were cultured in RPMI 1640 [Sigma], supplemented with 10% fetal bovine serum (FBS), penicillin -streptomycin and L-glutamine. The cultures were incubated at 37oC, 5% CO2 and then inhibitory effect of ethanolic extract on their proliferation was measured by MTT assay. The ethanolic extract was evaluated in terms of antimutagenicity properties by a standard reverse mutation assay (Ames Test). This was performed with histidine auxotroph strain of Salmonella Original Research Article Annual Research &Review in Biology, 4(6): 906-913, 2014 907 typhimurium (TA100).Thus, it requires histidine from a foreign supply to ensure its growth. The aforementioned strain gives rise to reverted colonies when expose to carcinogen substance (Sodium Azide) . Results: The ethanolic extract prevented the reverted mutations and the hindrance percent 51.2% in antimutagenicity test. During MTT, human leukemia pre B-cells revealed to have a meaningful cell death when compared with controls (P = .01). Conclusion: This study demonstrates the antimutagenicity effect of Biebersteinia multifida DC, and suggests that it may be potentially usefull as anticancer agent.

Protective Effect of Chlorophyllin and Lycopene from Water Spinach Extract on Cytotoxicity and Oxidative Stress Induced by Heavy Metals in Human Hepatoma Cells

The purpose of this study was to examine the inhibitory effects of ethanol extract of water spinach (EEWS) containing chlorophyll and lycopene on cytotoxicity and oxidative stress in liver induced by heavy metals. The (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) MTT assay and dichlorofluorescein (DCF) assay were conducted to measure cytotoxicity and inhibition of reactive oxygen species (ROS), respectively. Cytotoxicity was prevented at a concentration of 11.7 mg/L of EEWS. Both sodium copper chlorophyllin (SCC) and lycopene in EEWS were identified by ultraperformance liquid chromatography–photodiode array–electrospray ionization–mass spectroscopy (UPLC-PDA-ESI-MS/MSn) as major components at m/z 722.64 and 535.45, respectively. The concentrations of SCC and lycopene were 0.12 and 0.04 mg from 100 g of dried powder, respectively. Approximately 99% cytotoxicity induced by Cd was inhibited by EEWS. However, the inhibitory effect attributed to generation of ROS was similar with SCC, lycopene, and EEWS. Our results indicated that EEWS was effective in reducing cytotoxicity and oxidative stress produced by heavy metals in a HepG2 cell. Data suggest that the possible mechanism underlying the preventive action of SCC might be associated with diminished absorption of metal ions by chelating and blocking metal-mediated generation of ROS, while lycopene effects may be attributed to its high number of conjugated dienes that act as most potent singlet oxygen quenchers.

Effect of Curcumin Extract (Curcuma Longa) on Cancer Cell Lines in Vitro

Objectives: This study aims to detect the active compounds found in ethanolic crude extract of Curcuma longa rhizome and evaluate the cytotoxic effect of ethanolic extract on two malignant cell lines; Human laryngeal Carcinoma (HEP-2) cell line, murine mammary adenocarcinoma (AMN-3) cell line and one transformed cell line of Rat Embryogenic Fibroblast (REF-3). Methodology: Administrative Arrangement: This study was conducted in Iraqi Centre for Cancer and Medical Genetic Research (ICCMGR), Baghdad, Iraq into the period (1 / 1 / 2011 to 1 / 1 / 2012), periods of exposure of cell lines Different concentrations were prepared from ethanolic extract starting in (1 to 10000) µg/ml to were measured at) 24, 48, and 72 (hrs. in a microtitration plate under complete sterile conditions. Results: Chemical detection of extract appeared that the ethanolic extract contain flavonoid, saponin, phenols, resins, volatile oils, tannins, terpins and steroids while glycosids, alkaloids and cumarines gave negative results appeared the ethanolic extract of curcuma longa showed inhibition effect at 48 hr. period for all concentration on cancer cell lines. And amount the inhibition percentage of (AMN-3) cell line (88.3%) for treatment in concentration (6 µg/ml) after (24hr). of exposure time, and inhibition percentage begin in lower concentration on (Hep-2) cancer cell line and gave inhibition percentage (77.1%) after treatment in (2000 µg/ml) after (48hr). of exposure time and appeared the ethanolic extract on REF transformed cell line and gave higher inhibition percentage (93%) after treatment in (100 µg/ml) of exposure time. Conclusion: The ethanolic crude extract of Curcuma longa Which contain flavonoid, saponin, phenols, resins, Volatile Oils, tannins, terpins and steroids while alkaloids, glycosids, and cumarines gave negative results, In the in vitro study, The ethanolic crude extract of C. longa has a potent cytotoxic activity against REF transformed cell line, AMN-3 and Hep-2 cell lines in a concentration and time dependent manners (in vitro), The cell lines REF and AMN-3 were the most sensitive to the inhibitory effects of ethanolic extract of C. longa rhizomes, The cell line Hep-2 showed less response at low concentration while the extract had cytotoxic effect at highest concentration after exposure 48 hr. Recommendations: Using further different cancer cell lines as well as normal cell lines to study the cytotoxicity of Curcuma longa extract such as oral, stomach and colon cell lines. Using the comet test technique to detect the effects of plants extracts on DNA, use different solvent to extract of Curcuma longa for example methanol.

발아기간에 따른 벼(Oryza sativa L.)의 부위별 효소저해활성

This study investigated the enzyme inhibitory effects of ethanol extracts from the different parts of rough rice (Oryza sativar L.) from its germination period. Rough rice was germinated at for 6 days, then separated into hull+sprout and brown rice. -Glucosidase inhibitory activity had the highest value (39.38%) in hull+sprout extracts after 5 days of germination. -Amylase and DPP-4 inhibitory activity had the highest values (75.32% and 47.77%, respectively) in hull+sprout extracts after germination for 5 days. ACE inhibitory activity of hull+ sprout extracts increased from 43.16% at the beginning to 58.60% at 5 days, while brown rice extracts increased this activity from 0.88% at the beginning to 14.50% at 4 days. The xanthine oxidase inhibitory activity of hull+ sprout extracts increased from 62.02% at the beginning to 64.49% at one day, and then decreased. Lipase inhibitory activity had its highest value (55.17%) in hull+sprout extracts after germination for 5 days. These results indicate that the optimal germination period for increasing enzyme inhibitory activities may be 5 days, and that hull+sprout extracts have a higher enzyme inhibition activity than brown rice.

Study of the inhibitory effect of Ethanolic extract of( Quercus robur ,Cinnamomum zeylanicum and Thymus vulgaris ) on the growth of Staphylococcus aureus isolated from clinical mastitis in cow

The present study was designed to evaluate the inhibitory effects of three local plant ethanolic extracts (Quercus robur , Cinnamomum zeylanicum and Thymus vulgaris) against the growth of Staphylococcus aureus isolated from milk of cow infected with clinical mastits in culture media by using of agar well diffusion method. For this purpose graduate concentrates for each extract (50 , 100 , 200 , 400 )mg/ml prepared and tested .The result showed that the extract of Quercus robur was more effective followed by Thymus vulgaris and Cinnamomum zeylanicum.The statistical analysis by using ANOVA with LSD at level (p<0.05) showed that there was no significant differences between the effect of the studied concentrations of Quercus robur , Cinnamomum zeylanicum while for Thymus vulgaris we find that the concentration 100 mg\ml had a significant difference with the other studied concentration in inhibition of the growth of the tested bacteria. Tropical Atlantic marine macroalgae with bioactivity against virulent and antibiotic resistant Vibrio

The antibacterial activity of ethanol, methanol, hexane and acetone-based extracts of the macroalgae Padina gymnospora (PG), Hypnea musciformes (HM), Ulva fasciata (UF) and Caulerpa prolifera (CP) was investigated. The disk diffusion method was used to evaluate the algae antimicrobial effect against standard strains of Vibrio parahaemolyticus, Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa and Salmonella enterica and five virulent antibiotic-resistant strains of V. brasiliensis, V. xuii and V. navarrensis (isolated from the hemolymph of Litopenaeus vannamei). Ethanol extracts of PG and HM inhibited all Vibrio strains. E. coli and P. aeruginosa were only susceptible to ethanol extracts of PG. Among the methanol extracts, only UF was bioactive, inhibiting V. navarrensis. The observed inhibitory effect of ethanol extracts of PG, HM and UF against virulent antibiotic-resistant bacteria suggests these macroalgal species constitute a potential source of bioactive compounds.

In-vitro α amylase and glycosidase inhibitory effect of ethanolic extract of antiasthmatic drug - Shirishadi

Asthma and diabetes have strong relationship; both are cause and effect of each other. Oxidative stress due to bronchial asthma may cause insulin resistance whereas lack of proper insulin can cause defective smooth muscle relaxant. There is no single medicine available that can manage both diseases, rather the mainstay treatment of bronchial asthma causes hyperglycemia. Keeping this problem in focus, in this study the hypoglycemic effect of an indigenous antiasthmatic Ayurvedic drug Shirishadi was evaluated. Pancreatic alpha amylase and glucosidase inhibitors offer an effective strategy to lower the level of post prandial hyperglycemia via control of starch breakdown. For evaluation of hypoglycemic activity of drug, in-vitro alpha amylase and alpha glucosidase enzyme inhibition was calculated. Ethanolic extract of compound showed 76.40% + 0.88% reduction in alpha amylase activity and 63.85% + 0.36% in alpha glucosidase activity with IC50 0.68 mg/ml and 2.89 mg/ml, respectively. This study suggests that the ethanolic extract of Shirishadi polyherbal compound effectively acts as alpha amylase and glucosidase inhibitor leading to a reduction in starch hydrolysis and hence acts as antiasthmatic as well as hypoglycemic drug.

α-Glucosidase and α-Amylase Inhibitory Activity of Senna surattensis

In this study, we investigated the inhibitory effects of ethanolic extract of the leaves of Senna surattensis (EESS) on α-glucosidase and α-amylase. We also studied the in vitro antidiabetic activity of S. surattensis using the glucose uptake by isolated rat hemidiaphragm model. In vitro studies using mammalian α-glucosidase extracted from the small intestine homogenate of mouse showed that the extract was found to be more effective in inhibiting the activities of maltase [half maximal inhibitory concentration (IC50): 209.15 μg/mL] and sucrase (IC50: 366.44 μg/mL) when compared with the control group (acarbose). The extract of S. surattensis were further quantified with respect to porcine pancreatic α-amylase inhibition using the chromogenic 3,5-dinitrosalicylic acid method. Interestingly, S. surattensis was also found to exhibit α-amylase (IC50: 123.95 μg/mL) inhibitory activity. The glucose uptake in the rat hemidiaphragm was significantly (p < 0.01) increased by EESS (220.95 ± 5.4 mg/g/30 minute) when compared with the control group. The total polyphenolic content of EESS was found to be 98 μg pyrocatechol/mg of the extract. These results suggest that EESS inhibited carbohydrate digestive enzymes and increased the peripheral uptake of glucose. This study endorses the use of this plant for further studies to determine their potential for managing type II diabetes.

Antioxidant properties and inhibitory effect of ethanolic extract of Struchium sparganophora (Ewuro Odo) leaf on α--amylase and α--glucosidase activities.

Recent studies have shown that vegetables consumption could lower the risk of diabetes mellitus. Therefore, this study sought to investigate the inhibitory effect of Struchium sparganophora (Ewuro Odo) leaf on key enzyme linked to type-2 diabetes (α-amylase and α-glucosidase) as well as assessing the effect of blanching (a commonly practiced food processing technique) of this leafy vegetable on these key enzymes. Fresh leaves of Struchium sparganophora were blanched in hot water for 10 minutes, and the 70% ethanolic extracts of the fresh and blanched vegetables were prepared and used for subsequent analysis. The antioxidant properties and interaction of the extracts on α - amylase and α - glucosidase activities was determined in vitro. The result revealed that Struchium sparganophora leaf scavenged DPPH free radical and also inhibited α -amylase and α - glucosidase activities in a dose dependent manner (0.05 -0.2 mg/ml). However, blanching of this leafy vegetables caused a significant (P<0.05) increase in the antioxidant properties as typified by the DPPH radical scavenging ability and reducing property but decrease their ability to inhibit α - amylase and α - glucosidase activities. This antioxidant properties and enzyme inhibition could be part of the mechanism by which green leafy vegetables exert their anti-diabetic properties. However, blanching of the vegetable could reduce their ability to inhibit both α - amylase and α - glucosidase activity, but enhance their antioxidant properties in vitro.
