Metformin as an alternative therapeutic intervention in hemorrhagic shock

Hemorrhagic shock (HS) continues to significantly contribute to civilian and military mortality, underscoring the critical need for new resuscitation strategies. Recent discussions have emphasized the pivotal role of impaired oxygen delivery and cellular metabolism in exacerbating HS-related damage. Metformin, a well-established diabetes medication, exhibits promising potential to enhance cellular metabolism and activate pathways linked to improved oxygen uptake, thereby optimizing cellular energy management. Thus, this study aimed to assess metformin's effcacy in ameliorating outcomes in animals subjected to HS. To achieve this, Wistar rats were surgically prepared with femoral artery and vein catheters. HS was induced by controlled blood withdrawal to maintain a mean arterial pressure (MAP) within the range of 35-40 mmHg for 90 minutes. Throughout the protocol, blood pressure (BP) and heart rate (HR) were assessed. The animals were divided into two groups: Lactated Ringer's solution (LR) and LR treated with Metformin (Met). Metformin was administered in three bolus injections: at the start of resuscitation (10mg), 45 minutes into resuscitation (5mg), and 90 minutes into resuscitation (5mg). After 120 minutes, the animals were euthanized, and blood samples were collected for subsequent analysis, including metabolomic profiling of the tricarboxylic acid (TCA) cycle, amino acids, and lipids. Additionally, trypsin-like enzyme activity in the plasma and markers of brain injury were evaluated. Animals treated with Met showed significantly higher systolic blood pressure after resuscitation (LR: 74±5 mmHg vs. LR+Met: 94±3 mmHg) with no significant changes in HR observed. Brain injury markers such as GFAP (Glial fibrillary acidic protein), S100B (astrocytic protein specific to the central nervous system), Tau (microtubule-associated protein), and NFL (axonal protein neurofilament light) were assessed. Animals reperfused with LR and treated with Met displayed a 22% reduction in GFAP levels compared to those reperfused with LR alone, while other markers remained unchanged. Met treatment also led to a reduction in plasmatic metabolites, including Lactate (30%), Succinate (24%), and Fumarate (22%), along with an increase in Citrate (23%) and α-ketoglutarate (24%)—all constituents of the TCA cycle. Additionally, an increase in amino acid metabolites and a decrease in lipid metabolites were observed. Recent research has suggested that HS results in elevated levels of trypsin-like enzymes in the plasma and that inhibiting these enzymes might improve outcomes during resuscitation. Our study revealed that treatment with Met reduced approximately 60% of trypsin-like enzyme activity in the plasma compared to the LR-only group. In conclusion, our study demonstrates that Met, when administered in conjunction with LR as a resuscitation solution, significantly enhances post-resuscitation BP compared to LR alone. This improved outcome may be associated with alterations in the TCA cycle and the inhibition of trypsin-like enzyme activity. However, further investigations are necessary to elucidate the precise role of these changes in the observed enhancements within this experimental model. Pyrazoles have a multifaceted anti-inflammatory effect targeting prostaglandin E2, cyclooxygenases and leukocytes’ oxidative burst

Elevated levels of prostaglandin E2 have been implicated in the pathophysiology of various diseases. Anti-inflammatory drugs that act through the inhibition of cyclooxygenase enzymatic activity, thereby leading to the suppression of prostaglandin E2, are often associated with several side effects due to their non-specific inhibition of cyclooxygenase enzymes. Consequently, the targeted suppression of prostaglandin E2 production with innovative molecules and/or mechanisms emerges as a compelling therapeutic strategy for the treatment of inflammatory-related diseases. Therefore, in this study, a systematic analysis of 28 pyrazole derivatives was conducted to explore their potential mechanisms for reducing prostaglandin E2 levels. In this context, the evaluation of these derivatives extended to examining their capacity to reduce prostaglandin E2in vitro in human whole blood, inhibit cyclooxygenase-1 and cyclooxygenase-2 enzymes, modulate cyclooxygenase-2 expression, and suppress oxidative burst in human leukocytes. The results enabled the establishment of significant structure-activity relationships, elucidating key determinants for their activities. In particular, the 4-styryl group on the pyrazole moiety and the presence of chloro substitutions were identified as key determinants. Pyrazole 8 demonstrated the capacity to reduce prostaglandin E2 levels by downregulating cyclooxygenase-2 expression, and pyrazole-1,2,3-triazole 18 emerged as a dual-acting agent, inhibiting human leukocytes' oxidative burst and cyclooxygenase-2 activity. Furthermore, pyrazole 26 demonstrated effective reduction of prostaglandin E2 levels through selective cyclooxygenase-1 inhibition. These results underscore the multifaceted anti-inflammatory potential of pyrazoles, providing new insights into the substitutions and structural frameworks that are beneficial for the studied activity.

Polycyclic aromatic hydrocarbon (PAH) activation of AhR inhibits glutathione S-transferase (GST) activity in primary human small airway epithelial cells (hSAECs)

Background: PAH represents the organic phase of particulate matter (PM) in air pollution emitted via the incomplete combustion of various compounds (i.e. diesel fuel, wood, and plastics). It is widely regarded that PAH activation of aryl hydrocarbon receptor (AhR) plays an important role in pro-inflammatory signaling, however the mechanisms remain unknown. In contrast- endogenous AhR agonists, such as 6-Formylindolo[3,2-b]carbazole (FICZ), exert pleiotropic protective effects against inflammation, fibrosis, and oxidative stress. The objective of our research program is to delineate the differences between activation of AhR by PAH (with adverse effects) compared to its beneficial homeostatic activation by endogenous FICZ. PM may induce oxidative stress in part through inhibition of antioxidant defense mechanisms. GSTs are a superfamily of enzymes that plays an important role in protecting cells from oxidative damage (by conjugating reduced glutathione to substrates under pro-oxidizing conditions). We hypothesize that PAH activation of AhR contributes to oxidative stress via inhibition of GST activity in primary hSAECs. Methods: Primary hSAECs were subjected to varying dosages of PAH [0, 250, 500, 1000, 5000 ng/mL] to model airborne PM exposure levels, and to increasing doses of FICZ [0, 0.50, 10, 100, 1000 nM] to model normal and excessively high endogenous ligand activation of AhR. We determined the relative potencies of PAH and FICZ by measuring CYP1A1 and CYP1B1 gene expression. Oxidative stress was measured using DHE-labeling and GST activity was measured with enzymatic kits (Sigma Aldrich). AhR activity was pharmacologically inhibited using 10μM CH223191. Statistical analysis was conducted using SigmaPlot 14.0 (Systat Software). Results: PAH and FICZ maximally stimulate hSAEC CYP1A1 and CYP1B1 expression at 1000ng/mL and 100nM, respectively, and are therefore defined as equipotent concentrations used in our studies. In a representative experiment in hSAECs, PAH (1000ng/mL) significantly increased hSAEC oxidative stress vs FICZ (100nM) n=6, p<0.01), while significantly decreasing GST specific activity vs equipotent FICZ activation of AhR in primary hSAECs from a 66-year-old female without lung disease (26.6±0.874 vs 28.3±.611 nmol/mL/min); n=6 from 2 independent assays; p=0.032. Similar differential effects of PAH vs FICZ activation of AhR on GST were seen in additional experiments with male hSAECs and Calu-3 cells (male). Importantly, CH223191 inhibition of GST enzymatic activity suggests that AhR plays an important role in protecting cells from oxidative stress. Conclusions: Our studies indicate that airborne PAH exposure can have detrimental health effects on the lung via aberrant AhR signaling. The Antityrosinase Activity of Mitragyna speciosa Korth Leaf Ethanolic Extract

The kratom plant (Mitragyna speciosa Korth) is a the plant that is traditionally used to soften the skin. Kratom leaves have very strong antioxidant activity so they are predicted also to have anti-tyrosinase activity. The potential inhibition of tyrosinase enzyme activity is thought to come from the content of flavonoid compounds, polyphenols, steroids, tannins, and quinones. This research aims to determine the tyrosinase inhibitory activity of ethanol extract of M. speciosa leaves. The inhibitory activity of the tyrosinase enzyme using L-DOPA as substrate and kojic acid as positive control. Antityrosinase testing was carried out by in vitro method using UV-vis spectrophotometer through determination of IC50 value. The results of the antityrosinase activity test of ethanol extract of M. speciosa leaves showed an IC50 value of 2117.708 ppm. The ethanol extract of M. speciosa leaves is scientifically proven to have antityrosinase activity with a weak category.

Mechanistic evaluation of contrasting modulation in conformation and catalytic performance of urease by surface interaction of graphene oxide and amine- functionalized graphene oxide nanosheets

The increasing use of graphene-based nanomaterials raises concerns about their potential environmental impact as ecotoxicants. This article examines the interaction and effects of graphene oxide (GO) and its amine-derived (GONH2) 2-D nanosheets on urease dynamics, a crucial enzyme in soil nutrient cycling, through spectroscopic, microscopic, and computational analyses. The findings reveal the distinct interaction mechanisms: GO formed ground - and excited-state complexes with urease; thus, fluorescence quenching was static and dynamic via hydrophobic interaction. In contrast, GONH2 nanosheets formed a ground-state complex by static quenching involving Van der Waals and hydrogen bonding. These interactions were energetically favored and altered tryptophan (Trp) residues’ microenvironment more than tyrosine (Tyr). The average exciton lifetime of urease in the presence of GO have decreased from 4.34 ns to 3.647 ns at the mere concentration of 7.11 µM, while for GONH2, no discernable change (4.16 ns) was observed even at the concentration of 16.75 µM. Primarily, the strong interaction of GO leads to the rearrangement of polypeptide structure, reduced α-helices to 12 %, and increased β-sheets by six times. In contrast, with GONH2, α-helices were reduced to 79 %, and β-sheets were increased by 1.8 times at the similar concentration. Molecular docking revealed that neither GO nor GONH2 interacted with the active site of urease, and binding was primarily via hydrophobic, Van der Waals, and hydrogen bonding, concurrent with in-vitro incubation results. Positive total energy (3.85 kJ mol−1) during molecular docking analysis of urease-GO interaction and inhibition in enzyme activity to 62 %, altered Vmax value (0.04606 ± 0.001 mM/s), without affecting the Km (4.78045 ± 0.12 mM) suggested that inhibition was noncompetitive and steric clashes due to disruptive binding might drove the inhibition. Meanwhile, negative total internal energy (-3.97 kJ mol−1) during urease + GONH2 binding and enhanced enzymatic activity by 32 % in-vitro without altering Vmax (0.063091 ± 0.003 mM/s) and Km (4.953943 ± 0.2 mM) values suggested that urease's binding site was well-complemented by GONH2 nanosheets, serving as a redox-mediator and radical-quencher.

Discovery of N-(4-((6-(3,5- Dimethoxyphenyl)-9H-purine derivatives as irreversible covalent FGFR inhibitors

Fibroblast growth factor receptor (FGFR) is an attractive target for cancer therapy, but existing FGFR inhibitors appear to hardly meet the demand for clinical application. Herein, a number of irreversible covalent FGFR inhibitors were designed and synthesized by selecting several five- and six-membered azaheterocycles as parent scaffold with different substituents to take over the hydrophobic region in the active pocket of FGFR proteins. Among the resulting target compounds, III-30 showed the most potent effect on enzyme activity inhibition and anti-proliferative activity against the tested cancer cell lines. Significantly, III-30 could inhibit the enzyme activity by achieving irreversible covalent binding with FGFR1 and FGFR4 proteins. It could also regulate FGFR-mediated signaling pathway and mitochondrial apoptotic pathway to promote cancer cell apoptosis and inhibit cancer cell invasion and metastasis. Moreover, III-30 had a good metabolic stability and showed relatively potent anti-tumor activity in the MDA-MB-231 xenograft tumor mice model.

Advances in Research on the Activity Evaluation, Mechanism and Structure-Activity Relationships of Natural Antioxidant Peptides.

Antioxidant peptides are a class of biologically active peptides with low molecular weights and stable antioxidant properties that are isolated from proteins. In this review, the progress in research on the activity evaluation, action mechanism, and structure-activity relationships of natural antioxidant peptides are summarized. The methods used to evaluate antioxidant activity are mainly classified into three categories: in vitro chemical, in vitro cellular, and in vivo animal methods. Also, the biological effects produced by these three methods are listed: the scavenging of free radicals, chelation of metal ions, inhibition of lipid peroxidation, inhibition of oxidative enzyme activities, and activation of antioxidant enzymes and non-enzymatic systems. The antioxidant effects of natural peptides primarily consist of the regulation of redox signaling pathways, which includes activation of the Nrf2 pathway and the inhibition of the NF-κB pathway. The structure-activity relationships of the antioxidant peptides are investigated, including the effects of peptide molecular weight, amino acid composition and sequence, and secondary structure on antioxidant activity. In addition, four computer-assisted methods (molecular docking, molecular dynamics simulation, quantum chemical calculations, and the determination of quantitative structure-activity relationships) for analyzing the structure-activity effects of natural peptides are summarized. Thus, this review lays a theoretical foundation for the development of new antioxidants, nutraceuticals, and cosmetics.

Application of the thin layer chromatography method for the determination of the degree of hydrolysis of soy lecithin

The aim of this study was the determination of the degree of enzymatic hydrolysis of soy lecithin using thin layer chromatography in combination with a modern imaging data processing program. Hydrolysis of phospholipids was carried by the action of the enzyme phospholipase A2 (PLA2) in the presence of calcium ions for 2 h at a temperature of 50°C and pH 5.7. Chromatographic separation of the components of the reaction medium was carried out 10, 20, 30, 60, 90, and 120 min after adding the PLA2 in the chloroform: methanol: ammonium hydroxide system in a ratio of 6.5 : 2.5 : 0.4 (v/v/v). The following phospholipids and their lysoforms were identified: Rf = 0.41±0.03 for phosphatidylethanolamine (PE), Rf = 0.28±0.02 for phosphatidylcholine (PC), Rf = 0.09±0.01 for phosphatidylinositol (PI), Rf = 0.06±0.01 for phosphatidic acid (PA), Rf = 0.15±0.02 for lysophosphatidylethanolamine (LPE), Rf = 0.08±0.01 for lysophosphatidylcholine (LPC), Rf = 0.03±0.01 for lysophosphatidylinositol (LPI) and Rf = 0.02±0.01 for lysophosphatidic acid (LPA). It was shown that soy lecithin contained PC, PE, PI, and PA before the enzymatic reaction, the content of these compounds after the addition of the PLA2 decreases due to the formation of LPC, LPE, LPI, and LPA, respectively. The dependence of the degree of hydrolysis (α, %) of PC on the duration of the enzymatic reaction was obtained. It was established that the intensive conversion of PC to LPC under the action of the PLA2 occurred during 60 min (α = 82%). Further conversion of PC occurred insignificantly: 84% after 90 min and 87% after 120 min, which was due to the inhibition of enzyme activity by the final reaction product.

Synthesis, bioactivity evaluation and theoretical study of nicotinamide derivatives containing diphenyl ether fragments as potential succinate dehydrogenase inhibitors

To obtain nicotinamide fungicides with higher inhibitory activity against Rhizoctonia solani, 24 nicotinamide derivatives were designed and synthesized by introducing the diphenyl ether fragments into the structure of boscalid. Biological assays revealed that the obtained compounds exhibited varying degrees of antifungal activity. Among them, compound 2-chloro-N-(2-(2,4-dichlorophenoxy)phenyl)nicotinamide (7g) exhibited prominent biological activity against R. solani (EC50 = 0.034 mg/L), surpassing the inhibitory effect of boscalid. Experimentally, compound 7g was a promising fungicide with low toxicity that targeted succinate dehydrogenase, as confirmed by the tests of cytotoxicity, inhibition of enzyme activity, mitochondrial membrane potential, and mycelial respiration. This study highlighted the potential of nicotinamide derivatives containing diphenyl ether fragments designed based on theoretical calculations as succinate dehydrogenase inhibitor (SDHI) fungicides.

Spastin locally amplifies microtubule dynamics to pattern the axon for presynaptic cargo delivery

Neurons rely on the long-range trafficking of synaptic components to form and maintain the complex neural networks that encode the human experience. With a single neuron capable of forming thousands of distinct en passant synapses along its axon, spatially precise delivery of the necessary synaptic components is paramount. How these synapses are patterned, as well as how the efficient delivery of synaptic components is regulated, remains largely unknown. Here, we reveal a novel role for the microtubule (MT)-severing enzyme spastin in locally enhancing MT polymerization to influence presynaptic cargo pausing and retention along the axon. In human neurons derived from induced pluripotent stem cells (iPSCs), we identify sites stably enriched for presynaptic components along the axon prior to the robust assembly of mature presynapses apposed by postsynaptic contacts. These sites are capable of cycling synaptic vesicles, are enriched with spastin, and are hotspots for new MT growth and synaptic vesicle precursor (SVP) pausing/retention. The disruption of neuronal spastin level or activity, by CRISPRi-mediated depletion, transient overexpression, or pharmacologic inhibition of enzymatic activity, interrupts the localized enrichment of dynamic MT plus ends and diminishes SVP accumulation. Using an innovative human heterologous synapse model, where microfluidically isolated human axons recognize and form presynaptic connections with neuroligin-expressing non-neuronal cells, we reveal that neurons deficient for spastin do not achieve the same level of presynaptic component accumulation as control neurons. We propose a model where spastin acts locally as an amplifier of MT polymerization to pattern specific regions of the axon for synaptogenesis and guide synaptic cargo delivery.

Impacts of nano-acetamiprid pesticide on faba bean root metabolic response and soil health

The associated benefits and potential environmental risks of nanopesticides on plant and soil health, particularly in comparison with traditional pesticides, have not been systematically elucidated. Herein, we investigated the impacts of the as-synthesized nano-acetamiprid (Nano-Ace, 20 nm) at low (10 mg/L), medium (50 mg/L), high (100 mg/L) doses and the corresponding high commercial acetamiprid (Ace, 100 mg/L) on the physiological and metabolic response of faba bean (Vicia faba L.) plants, as well as on rhizosphere bacterial communities and functions over short-, medium- and long-term exposures. Overall, Nano-Ace exposure contributed to basic metabolic pathways (e.g., flavonoids, amino acids, TCA cycle intermediate, etc.) in faba bean roots across the whole exposure period. Moreover, Nano-Ace exposure enriched rhizosphere beneficial bacteria (e.g., Streptomyces (420.7%), Pseudomonas (33.8%), Flavobacterium (23.3%)) and suppressed pathogenic bacteria (e.g., Acidovorax (44.5%)). Additionally, Nano-Ace exposure showed a trend of low promotion and high inhibition of soil enzyme activities (e.g., invertase, urease, arylsulfatase, alkaline phosphatase) involved in soil C, N, S, and P cycling, while the inhibition was generally weaker than that of conventional Ace. Altogether, this study indicated that the redox-responsive nano-acetamiprid pesticide possessed high safety for host plants and soil health.

Inhibitory protein-protein interactions of the SIRT1 deacetylase are choreographed by post-translational modification.

Regulation of SIRT1 activity is vital to energy homeostasis and plays important roles in many diseases. We previously showed that insulin triggers the epigenetic regulator DBC1 to prime SIRT1 for repression by the multifunctional trafficking protein PACS-2. Here, we show that liver DBC1/PACS-2 regulates the diurnal inhibition of SIRT1, which is critically important for insulin-dependent switch in fuel metabolism from fat to glucose oxidation. We present the x-ray structure of the DBC1 S1-like domain that binds SIRT1 and an NMR characterization of how the SIRT1 N-terminal region engages DBC1. This interaction is inhibited by acetylation of K112 of DBC1 and stimulated by the insulin-dependent phosphorylation of human SIRT1 at S162 and S172, catalyzed sequentially by CK2 and GSK3, resulting in the PACS-2-dependent inhibition of nuclear SIRT1 enzymatic activity and translocation of the deacetylase in the cytoplasm. Finally, we discuss how defects in the DBC1/PACS-2-controlled SIRT1 inhibitory pathway are associated with disease, including obesity and non-alcoholic fatty liver disease.

Metabolomics combined with chemometric analysis to identify α-glucosidase inhibitors in Phaleria macrocarpa fruit extracts and its molecular docking simulation

Phaleria macrocarpa is a medicinal plant widely available in Malaysia. The plant parts have been traditionally used as an antidiabetic remedy. This study aimed to identify the putative inhibitors of the α-glucosidase enzyme from P. macrocarpa using a gas chromatography-mass spectrometry (GC–MS)-based metabolomics approach and further subjected them to in silico molecular docking analysis to elucidate the possible mechanism of action. This study assessed the inhibitory potential of P. macrocarpa fruit extracts (aqueous, 20 %, 40 %, 60 %, 80 %, and 100 % ethanol) against the α-glucosidase enzyme using GC–MS and chemometric analysis. Orthogonal partial least squares (OPLS) combined with GC–MS analysis were applied to correlate the inhibition of enzyme activity to the metabolites profile of P. macrocarpa. All the potential inhibitors identified were further docked to the yeast (Saccharomyces cerevisiae) protein crystal structure (PDB3A4A). The generated score scatter plot of OPLS showed a recognizable separation of all the extracts into six different clusters. GC–MS, incorporated with multivariate data analysis techniques, was used to identify significant chemical markers. Methyl α-d-glucopyranoside, squalene, palmitic acid, myo-inositol and isoquinoline metabolites were identified as putative inhibitors against α-glucosidase activity from P. macrocarpa. In conclusion, the GC–MS-based metabolomics approach identified potential chemical markers of P. macrocarpa that could be utilized in the development of herbal based medicine.

Abstract 3366: FoRx-06-428 is a novel PARG inhibitor with potent anti-tumor efficacy in preclinical cancer models

Abstract The formation of poly(ADP-ribose) (PAR) chains from NAD+ precursors by PARP enzymes is a characteristic posttranslational modification during DNA damage repair. PAR chains function as docking platforms for DNA repair proteins that are required to resolve DNA lesions. Equally important is the subsequent removal of PAR chains from protein substrates to conclude the repair process and restore genomic integrity. Poly(ADP-ribose) glycohydrolase (PARG) is the key enzyme in this process and has recently garnered significant attention as a novel cancer target. FoRx-06-428 is a novel, orally bioavailable and highly potent, low molecular weight PARG inhibitor. It binds to the catalytic domain of human PARG with high affinity, as evidenced by surface plasmon resonance (SPR), and displays potent inhibition of enzymatic activity. FoRx-06-428 shows selective reduction of viability in a panel of cancer cell lines highlighting therapeutic potential in multiple cancer indications. Pharmacological inhibition of PARG leads to accumulation of PAR chains and is indicative of target engagement, accompanied by modulation of DNA damage response markers, such as phosphorylation of replication protein A (pRPA) and phosphorylation of H2AX (γH2AX). Compelling anti-tumor efficacy as single agent was observed in several xenograft models (breast and gastric cancer) with excellent tolerability. Dose-dependent accumulation of PAR chains was demonstrated in vivo and constitutes a tractable target engagement biomarker for clinical exploration. Sensitivity to PARG inhibition is partly driven by distinct genomic instabilities, such as defects in homologous recombination (HR) repair, however the activity of PARG inhibitors is also influenced by aberrations in other cancer-relevant pathways. This report represents the first disclosure of the pharmacological characterization of FoRx-06-428 and highlights the value of PARG inhibition as a novel targeted approach to treat DNA repair-deficient cancers. Effects of different fermentation methods on chemical composition, antioxidant activity, and enzymatic inhibition of fermented fig juice

ABSTRACT In order to develop nutritious and healthy fermented fig juice, foure samples fermented were made using natural fermentation and fermentation by lactobacillus. Then the total acidity, total sugar, reducing sugar, total phenols, total flavones and characteristic organic acids, antioxidant activity, and inhibition of enzyme activity in fermented fig juice were compared. The results showed that natural fermentation could produce more reducing sugar, lactic acid, flavonoids, and exhibit stronger DPPH radical scavenging capacity and total reducing power. Inhibition of tyrosinase and pancreatic lipase was detected in fermented fig juice, of which the inhibition rate of tyrosinase by natural fermentation was 81.80%, and the inhibition rate of pancreatic lipase by mixed cultures was 38.72%, which was higher than that of other groups. The results indicated the functional activity of fermented fig juice could be improved by fermentation, and the natural fermentation method was found superior.

Chemical characterization, multivariate analysis and comparison of biological activities of different parts of Fraxinus mandshurica.

Fraxinus mandshurica (Oleaceae) is used as a traditional medicinal plant for the treatment of red eyes, menstrual disorders, excessive leucorrhea, chronic bronchitis and psoriasis. To perform chemical characterization of the secondary metabolites of F. mandshurica roots, bark, stems and leaves, 32 samples were collected from eight provinces in this study. A total of 64 chemical components were detected from four different parts of F. mandshurica by ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry. Meanwhile, a total of nine secoiridoids were obtained by natural product chemical extraction, isolation and identification methods. Quantitative analysis by high-performance liquid chromatography-diode array detection-mass spectrometry showed the highest total content of secoiridoids in the bark, which is also consistent with the traditional medicinal parts. The results of methodological validation showed that the correlation coefficient (R2) values were all >0.9993, indicating a good linear range of the standard curve, while the relative standard deviations of precision, reproducibility and stability were <3%, and the spiked recoveries ranged from 98.22 to 102.27%, indicating that the experimental method was reliable and stable. In addition, fingerprinting and a heatmap were established to demonstrate the content trends of F. mandshurica more visually from different origins. Multivariate analysis, including principal component analysis and partial least squares discriminant analysis, was performed to determine the chemical characteristics of different parts of F. mandshurica, and six characteristic secoiridoids that could be used to distinguish different origins were screened. Finally, the inhibition of tyrosinase, α-glucosidase, acetylcholinesterase and pancreatic lipase activities by the nine characteristic compounds and extracts from different parts were investigated, and the results showed that they all exhibited different degrees of enzyme activity inhibition and thus have potential applications in whitening and blemish removal, hypoglycemia, anti-Alzheimer's disease and anti-obesity as a new source of natural enzyme activity inhibitors. This study establishes an identification and evaluation method applicable to phytochemistry of different origins, which is a guideline for quality control, origin evaluation and clinical application of traditional medicinal plants. This is also an unprecedented study on the identification of the chemical composition of different parts of F. mandshurica, characteristic compounds and the inhibition of enzyme activity of extracts from different parts.

Cisplatin Disrupts Proteasome Bounce-Back Effect through Suppressing ZEB1/Nfe2l1 in Cholangiocarcinoma.

Bortezomib (BTZ) is a powerful proteasome inhibitor that has been approved for the treatment of haematologic malignancies. Its effectiveness has been assessed against different types of solid tumours. BTZ is ineffective in most solid tumours because of drug resistance, including cholangiocarcinoma, which is associated with a proteasome bounce-back effect. However, the mechanism through which proteasome inhibitors induce the proteasome bounce-back effect remains largely unknown. Cholangiocarcinoma cells were treated with BTZ, cisplatin, or a combination of both. The mRNA levels of Nfe2l1 and proteasome subunit genes (PSMA1, PSMB7, PSMD1, PSMD11, PSMD14, and PSME4) were determined using quantitative real time polymerase chain reaction (qPCR). The protein levels of nuclear factor-erythroid 2-related factor 1 (Nfe2l1) and proteasome enzyme activity were evaluated using western blotting and proteasome activity assays, respectively. Transcriptome sequencing was performed to screen for potential transcription factors that regulate Nfe2l1 expression. The effect of zinc finger E-box-binding homeobox 1 (ZEB1) on the expression of Nfe2l1 and proteasome subunit genes, as well as proteasome enzyme activity, was evaluated after the knockdown of ZEB1 expression with siRNA before treatment with BTZ. The transcriptional activity of ZEB1 on the Nfe2l1 promoter was detected using dual-luciferase reporter gene and chromatin immunoprecipitation assays. Cell viability was measured using the cell counting kit-8 (CCK-8) assay and cell apoptosis was assessed using western blotting and flow cytometry. Cisplatin treatment of BTZ-treated human cholangiocarcinoma cell line (RBE) suppressed proteasome subunit gene expression (proteasome bounce-back) and proteasomal enzyme activity. This effect was achieved by reducing the levels of Nfe2l1 mRNA and protein. Our study utilised transcriptome sequencing to identify ZEB1 as an upstream transcription factor of Nfe2l1, which was confirmed using dual-luciferase reporter gene and chromatin immunoprecipitation assays. Notably, ZEB1 knockdown using siRNA (si-ZEB1) hindered the expression of proteasome subunit genes under both basal and BTZ-induced conditions, leading to the inhibition of proteasomal enzyme activity. Furthermore, the combination treatment with BTZ, cisplatin, and si-ZEB1 significantly reduced the viability of RBE cells. Our study uncovered a novel mechanism through which cisplatin disrupts the BTZ-induced proteasome bounce-back effect by suppressing the ZEB1/Nfe2l1 axis in cholangiocarcinoma. This finding provides a theoretical basis for developing proteasome inhibitor-based strategies for the clinical treatment of cholangiocarcinoma and other tumours.

Unravelling the cardiovascular symphony: Decoding the intricate influence of green tea polyphenols from leaf to heart

The extensive exploration of green tea's therapeutic potential in relation to cardiovascular ailments has sparked considerable global interest. The green tea polyphenols notably EGCG, ECG, EGC, EC, and various catechins, exhibit considerable promise as agents with cardioprotective attributes. The green tea polyphenolic compounds effectively counteract the detrimental impact of reactive oxygen species while concurrently modulating intricate cellular signalling pathways, thereby ameliorating the adverse effects of oxidative stress—a pivotal contributor to cardiovascular diseases. Furthermore, these compounds have demonstrated a capacity to function as antihypertensive agents, primarily through their facilitation of vasodilation, inhibition of angiotensin-converting enzyme activity, and deterrence of excessive vasoconstriction. Moreover, green tea polyphenols exert their influence on lipid profiles and cholesterol metabolism, presenting an additional avenue for cardiovascular health maintenance. By inhibiting key enzymes, namely ACAT and HMG-CoA reductases, involved in cholesterol esterification and synthesis, respectively, green tea polyphenols actively contribute to the preservation of a favourable lipid profile, effectively mitigating the risk of atherosclerosis development. In evaluating the bioavailability of green tea polyphenols, it is observed that they exhibit considerable stability in gastric conditions. However, substantial metabolic transformations occur within the intestinal and hepatic milieu, consequently influencing their availability and physiological activity within the human body. This comprehensive review proffers an array of interdisciplinary methodologies, encompassing in silico modelling, omics technologies, and innovative research approaches. These multifaceted avenues possess immense potential in unravelling novel insights and enhancing therapeutic strategies. Future investigations should seek to explore captivating prospects, including the identification of specific polyphenolic constituents or synergistic combinations that exhibit pronounced cardioprotective effects, thus guiding the development of targeted therapeutic interventions. By elucidating the molecular mechanisms underpinning the cardiovascular effects of green tea polyphenols, a greater understanding of the therapeutic targets involved in the prevention.

Differences in the activities of six soil enzymes in response to cadmium contamination of paddy soils in high geological background areas

East Yunnan province in southwest China is a region with elevated natural abundance (high geological background levels) of Cd due to high metal (loid) contents in the soils. Enzyme activities are useful indicators of metal (loid) toxicity in contaminated soils and whether Cd inhibits enzyme activities in paddy soils in high geological background areas is of considerable public concern. A pot experiment combined with field investigation was conducted to assess the effects of Cd on six soil enzymes that are essential to the cycling of C, N, and P in soils. Inhibitory effects of Cd fractions on enzyme activities were assessed using ecological dose-response models. The impact of soil properties on the inhibition of sensitive soil enzymes by Cd were assessed using linear and structural equation models. Cadmium was enriched in the paddy soils with 72.2 % of soil samples from high geological background areas exceeding the Chinese threshold values (GB 15618–2018) of Cd. Enzyme responses to Cd contamination varied markedly with a negative response by catalase but a positive response by invertase. Urease, β-glucosidase, and alkaline phosphatase activities were stimulated at low Cd concentrations and inhibited at high concentrations. The average inhibition ratios of β-glucosidase, urease, and catalase in high Cd levels were 19.9, 38.9, and 51.9%, respectively. Ecological dose-response models indicate that catalase and urease were the most Cd-sensitive of the enzymes studied and were suitable indicators of soil quality in high geological background areas. Structural equation modeling (SEM) indicates that soil properties influenced sensitive enzymes through various pathways, indicating that soil properties were factors determining Cd inhibition of enzyme activities. This suggests that Cd concentrations and soil physicochemical properties under a range of environmental conditions should be considered in addressing soil Cd pollution.
