The effects of the magnetic field on germination and seedling growth of chickpea (Cicer arietinum L.) and sunflower (Helianthus annuus L.)

Organisms interact with their environment and effects of environmental factors vary depending on ecology and tolerance levels. However magnetic field is an inevitable factor for all organisms. The aim of the study was to investigate the effects of different magnetic field (MF) applications on germination percentage, pigment content and antioxidant capacity of two important agricultural plant (Sunflower and Chickpea) species. Initially, seeds were exposed to 5 mT, 10 mT and 20 mT magnetic field generated by Helmholtz coil for detection of germination effects. Then seedling test was survived at the same conditions. MF was applied 20 minutes for every day at the same time period. According to germination results, MF application to sunflower and chickpea seeds was resulted with increase in germination percentage compared to control. 20 mT application caused decrease in shoot length of sunflower seedlings. On the contrary, 20 mT MF application resulted with increase in shoot length of chickpea seedlings. All magnetic field strengths increased carotenoid levels in chickpea seedlings. Also, MF application affected the phenolic and flavonoid contents of sunflower and chickpea seedlings. Depending on the increase in secondary metabolites, DPPH and FRAP activities varied. As a conclusion, MF application contributed to effect on plant metabolism and it has the potential to be used in agricultural applications.

Time of Application of Desiccant Herbicides Affects Photosynthetic Pigments, Physiological Indicators, and the Quality of Cowpea Seeds.

Chemical desiccation is widely used in agriculture to anticipate harvest and mitigate the effects of adverse environmental conditions. It is applied to both grains and seeds. Although this practice is widely used, there are still significant gaps in understanding the effects of different herbicide application times on seed quality and plant physiological responses. The objective of this study was to evaluate the effects of different herbicide application times on cowpea, focusing on seed quality, physiological responses, and biochemical composition, including chlorophylls, carotenoids, sugars, and proline, under nocturnal desiccation. In the first experiment, eight herbicides and two mixtures were applied at night: diquat, flumioxazin, diquat + flumioxazin, glufosinate ammonium, saflufenacil, carfentrazone, diquat + carfentrazone, atrazine, and glyphosate. All of the tested herbicides caused a reduction in normal seedling formation, with the diquat + carfentrazone combination resulting in 100% abnormal seedlings. A significant decrease in chlorophyll levels (chlorophyll a: 63.5%, chlorophyll b: 50.2%) was observed using diquat, which indicates damage to photosynthetic processes, while the carotenoid content increased. Total soluble sugars and proline were also negatively impacted, reflecting physiological stress and metabolic changes in seedlings. In the second experiment, three application times were tested with diquat, diquat + flumioxazin, and diquat + carfentrazone. Nocturnal application showed the most significant reduction in chlorophyll levels and increased carotenoid levels. Application at noon and late afternoon also significantly changed the soluble sugar and proline levels. These results indicate that the herbicide application time directly influences the seeds' physiological quality.

Determination of the effects of pre-harvest bagging treatment on kiwifruit appearance and quality via transcriptome and metabolome analyses

Bagging is an effective cultivation strategy to produce attractive and pollution-free kiwifruit. However, the effect and metabolic regulatory mechanism of bagging treatment on kiwifruit quality remain unclear. In this study, transcriptome and metabolome analyses were conducted to determine the regulatory network of the differential metabolites and genes after bagging. Using outer and inner yellow single-layer fruit bags, we found that bagging treatment improved the appearance of kiwifruit, increased the soluble solid content (SSC) and carotenoid and anthocyanin levels, and decreased the chlorophyll levels. We also identified 41 differentially expressed metabolites and 897 differentially expressed genes (DEGs) between the bagged and control ‘Hongyang’ fruit. Transcriptome and metabolome analyses revealed that the increase in SSC after bagging treatment was mainly due to the increase in D-glucosamine metabolite levels and eight DEGs involved in amino sugar and nucleotide sugar metabolic pathways. A decrease in glutamyl-tRNA reductase may be the main reason for the decrease in chlorophyll. Downregulation of lycopene epsilon cyclase and 9-cis-epoxycarotenoid dioxygenase increased carotenoid levels. Additionally, an increase in the levels of the taxifolin-3′-O-glucoside metabolite, flavonoid 3′-monooxygenase, and some transcription factors led to the increase in anthocyanin levels. This study provides novel insights into the effects of bagging on the appearance and internal quality of kiwifruit and enriches our theoretical knowledge on the regulation of color pigment synthesis in kiwifruit.

Rational design of geranylgeranyl diphosphate synthase enhances carotenoid production and improves photosynthetic efficiency in Nicotiana tabacum

Restricted genetic diversity can supply only a limited number of elite genes for modern plant cultivation and transgenesis. In this study, we demonstrate that rational design enables the engineering of geranylgeranyl diphosphate synthase (NtGGPPS), an enzyme of the methylerythritol phosphate pathway (MEP) in the model plant Nicotiana tabacum. As the crucial bottleneck in carotenoid biosynthesis, NtGGPPS1 interacts with phytoene synthase (NtPSY1) to channel GGPP into the production of carotenoids. Loss of this enzyme in the ntggpps1 mutant leads to decreased carotenoid accumulation. With the aim of enhancing NtGGPPS1 activity, we undertook structure-guided rational redesign of its substrate binding pocket in combination with sequence alignment. The activity of the designed NtGGPPS1 (a pentuple mutant of five sites V154A/I161L/F218Y/I209S/V233E, d-NtGGPPS1) was measured by a high-throughput colorimetric assay. d-NtGGPPS1 exhibited significantly higher conversion of IPP and each co-substrate (DMAPP ~1995.5-fold, GPP ~25.9-fold, and FPP ~16.7-fold) for GGPP synthesis compared with wild-type NtGGPPS1. Importantly, the transient and stable expression of d-NtGGPPS1 in the ntggpps1 mutant increased carotenoid levels in leaves, improved photosynthetic efficiency, and increased biomass relative to NtGGPPS1. These findings provide a firm basis for the engineering of GGPPS and will facilitate the development of quality and yield traits. Our results open the door for the structure-guided rational design of elite genes in higher plants.

Impact of chitosan on nickel bioavailability in soil, the accumulation and tolerance of nickel in Calendula tripterocarpa

Excessive heavy metals in medicinal plants cause critical health issues to humans. Therefore, in the present study, the effect of soil amendment with chitosan (0, 0.125, 0.25, 0.5, and 1%) on bioavailability and tolerance of nickel in Calendula tripterocarpa grown in a soil spiked with Ni (100 and 150 mg/kg soil) was investigated. The results showed that Ni toxicity significantly reduced plant growth and content of chlorophyll a, b but increased carotenoid levels, lipid peroxidation, and catalase (CAT) and superoxide dismutase (SOD) activities in roots and shoots. The Ni bioaccumulation was significantly higher in shoots than roots. The soil amendment with chitosan reduced Ni bioavailability in soil, as well as lowered the biological accumulation of Ni in roots and shoots, and Ni transfer to leaves. The chitosan application also increased growth parameters and levels of chlorophyll a, b and carotenoids under both normal and Ni stress conditions. Furthermore, chitosan reduced the level of malondialdehyde and the activities of SOD and CAT in roots and shoots under Ni stress. In conclusion, results indicated that chitosan through lowering bioavailability of Ni in soils can remarkably relieve adverse effects of Ni toxicity in C. tripterocarpa.

Mutagenic effects of sodium azide on pineapple micropropagant growth and biochemical profile within temporary immersion bioreactors

Sodium azide (NaN3) is widely used to induce mutagenesis within in vitro plant systems. However, since this mutagenesis is undirected, its unintended effects demand characterization. This study investigated the mutagenic effects of sodium azide (0-0.45 mM) on selected growth (shoot multiplication rate and shoot cluster fresh weight) and biochemical (aldehydes, chlorophylls, carotenoids and phenolics) parameters in pineapple micropropagants within temporary immersion bioreactors (TIBs). The content of soluble phenolics in the culture medium was also evaluated. Irrespective of the concentration NaN3 decreased shoot multiplication rate (by 87% relative to the control at 0.45 mM) and fresh weight (by 66% relative to the control at 0.45 mM). Levels of chlorophyll a and b, and soluble phenolics in the culture medium were also negatively correlated with NaN3 concentration. Interestingly, NaN3 application increased shoot carotenoid and soluble phenolic levels but had no significant effect on a range of established plant stress biomarkers: cell wall-linked phenolic levels, malondialdehyde and other aldehydes. Given that 0.19 mM NaN3 decreased shoot multiplication rate by 50% and resulted in propagants that displayed no morphologically abnormalities, increased levels of photoprotective pigments (relative to the control) and no significant increase in lipid peroxidation products, the mutagen can be used at this concentration to induce pineapple mutagenesis in TIB based studies aimed at producing agriculturally-useful mutants.

Characterization of squalene-induced PgCYP736B involved in salt tolerance by modulating key genes of abscisic acid biosynthesis

Cytochrome P450 genes as the one of the largest superfamily genes mediate a wide range of plant biochemical pathways. In this study, a full-length cytochrome P450 monooxygenase (CYP736B) cDNA was isolated and characterized from Panax ginseng. It was revealed that the deduced amino acid of PgCYP736B shares a high degree of sequence homology with CYP736A12 encoded by P. ginseng. Expression of PgCYP736B was differentially induced not only during a Pseudomonas syringae infection (7.7-fold) and wounding (47.3-fold) but also after exposure to salt (7.4-fold), cold (8.3-fold), and drought stress (3.24-fold). The gene transcription was highly affected by methyl jasmonate (476-fold) in the ginseng, suggesting that PgCYP736B was elicitor-responsive. Furthermore, we overexpressed the PgCYP736B gene in Arabidopsis and found that PgCYP736B is a transmembrane protein. Overexpression of PgCYP736B in Arabidopsis conferred enhanced resistance to salt stress via decreased H2O2 accumulation, increased carotenoid levels, and through abscisic acid biosynthesis gene expression. Our results suggest that the induction of ginsenoside biosynthetic pathway genes along with PgCYP736B by an exogenous supply of 10–100 μM of squalene most likely affects the metabolite profile of ginsenoside triterpenoid. Overall, our findings indicate that PgCYP736B protects ginseng from salt stress and may contribute to triterpenoid biosynthesis.

A Single Amino Acid Substitution in an ORANGE Protein Promotes Carotenoid Overaccumulation in Arabidopsis.

Carotenoids are crucial for plant growth and human health. The finding of ORANGE (OR) protein as a pivotal regulator of carotenogenesis offers a unique opportunity to comprehensively understand the regulatory mechanisms of carotenoid accumulation and develop crops with enhanced nutritional quality. Here, we demonstrated that alteration of a single amino acid in a wild-type OR greatly enhanced its ability to promote carotenoid accumulation. Whereas overexpression of OR from Arabidopsis (Arabidopsis thaliana; AtOR) or from the agronomically important crop sorghum (Sorghum bicolor; SbOR) increased carotenoid levels up to 2-fold, expression of AtOR(His) (R90H) or SbOR(His) (R104H) variants dramatically enhanced carotenoid accumulation by up to 7-fold in the Arabidopsis calli. Moreover, we found that AtOR(Ala) (R90A) functioned similarly to AtOR(His) to promote carotenoid overproduction. Neither AtOR nor AtOR(His) greatly affected carotenogenic gene expression. AtOR(His) exhibited similar interactions with phytoene synthase (PSY) as AtOR in posttranscriptionally regulating PSY protein abundance. AtOR(His) triggered biogenesis of membranous chromoplasts in the Arabidopsis calli, which shared structures similar to chromoplasts found in the curd of the orange cauliflower (Brassica oleracea) mutant. By contrast, AtOR did not cause plastid-type changes in comparison with the controls, but produced plastids containing larger and electron-dense plastoglobuli. The unique ability of AtOR(His) in mediating chromoplast biogenesis is responsible for its induced carotenoid overproduction. Our study demonstrates OR(His/Ala) as powerful tools for carotenoid enrichment in plants, and provides insights into the mechanisms underlying OR(His)-regulated carotenoid accumulation.

Enhanced accumulation of carotenoids in sweetpotato plants overexpressing IbOr-Ins gene in purple-fleshed sweetpotato cultivar

Sweetpotato [Ipomoea batatas (L.) Lam] is an important root crop that produces low molecular weight antioxidants such as carotenoids and anthocyanin. The sweetpotato orange (IbOr) protein is involved in the accumulation of carotenoids. To increase the levels of carotenoids in the storage roots of sweetpotato, we generated transgenic sweetpotato plants overexpressing IbOr-Ins under the control of the cauliflower mosaic virus (CaMV) 35S promoter in an anthocyanin-rich purple-fleshed cultivar (referred to as IbOr plants). IbOr plants exhibited increased carotenoid levels (up to 7-fold) in their storage roots compared to wild type (WT) plants, as revealed by HPLC analysis. The carotenoid contents of IbOr plants were positively correlated with IbOr transcript levels. The levels of zeaxanthin were ∼12 times elevated in IbOr plants, whereas β-carotene increased ∼1.75 times higher than those of WT. Quantitative RT-PCR analysis revealed that most carotenoid biosynthetic pathway genes were up-regulated in the IbOr plants, including PDS, ZDS, LCY-β, CHY-β, ZEP and Pftf, whereas LCY-ɛ was down-regulated. Interestingly, CCD1, CCD4 and NCED, which are related to the degradation of carotenoids, were also up-regulated in the IbOr plants. Anthocyanin contents and transcription levels of associated biosynthetic genes seemed to be altered in the IbOr plants. The yields of storage roots and aerial parts of IbOr plants and WT plants were not significantly different under field cultivation. Taken together, these results indicate that overexpression of IbOr-Ins can increase the carotenoid contents of sweetpotato storage roots.

Testing the interactive effects of testosterone and parasites on carotenoid‐based ornamentation in a wild bird

Testosterone underlies the expression of most secondary sexual traits, playing a key role in sexual selection. However, high levels might be associated with physiological costs, such as immunosuppression. Immunostimulant carotenoids underpin the expression of many red-yellow ornaments, but are regulated by testosterone and constrained by parasites. We manipulated testosterone and nematode burdens in red grouse (Lagopus lagopus scoticus) in two populations to tease apart their effects on carotenoid levels, ornament size and colouration in three time-step periods. We found no evidence for interactive effects of testosterone and parasites on ornament size and colouration. We showed that ornament colouration was testosterone-driven. However, parasites decreased comb size with a time delay and testosterone increased carotenoid levels in one of the populations. This suggests that environmental context plays a key role in determining how individuals resolve the trade-off between allocating carotenoids for ornamental coloration or for self-maintenance needs. Our study advocates that adequately testing the mechanisms behind the production or maintenance of secondary sexual characters has to take into account the dynamics of sexual trait expression and their environmental context.

Phytoene synthase activity controls the biosynthesis of carotenoids and the supply of their metabolic precursors in dark‐grown Arabidopsis seedlings

Carotenoids are plastidial isoprenoids essential for plant life. In Arabidopsis thaliana carotenoid biosynthesis is strongly upregulated when seedlings that germinate in the dark (etiolated) emerge from the soil and light derepresses photomorphogenesis, causing etioplasts to become chloroplasts. We found that carotenoid biosynthesis is also induced when deetiolation is derepressed in the absence of actual light, eventually resulting in improved greening (chlorophyll accumulation) upon illumination. The increased production of carotenoids in the dark correlates with an upregulated activity of phytoene synthase (PSY; the first committed enzyme of carotenogenesis) and the induction of PSY gene expression in cotyledons (where carotenoids accumulate in dark-grown seedlings). The metabolic precursors for carotenoid synthesis under these conditions are mostly supplied by the plastidial methylerythritol 4-phosphate (MEP) pathway. Accumulation of flux-controlling MEP pathway enzymes, such as deoxyxylulose 5-phosphate synthase (DXS), is post-transcriptionally increased when deetiolation is derepressed in the dark. Unlike the situation observed in light-grown plants, however, the sole overexpression of DXS in dark-grown seedlings does not increase carotenoid accumulation. By contrast, induced expression of a PSY-encoding transgene results in increased carotenoid levels and a concomitant post-transcriptional accumulation of DXS. These data provide evidence for a feedback mechanism by which PSY controls metabolic flux to the carotenoid pathway in plants.

ON THE ECOLOGICAL BASIS OF INTERSPECIFIC HOMOPLASY IN CAROTENOID-BEARING SIGNALS

Evidence that similar color patterns occur in unrelated animals with different habits undermines the traditional view that homoplasy evolves through shared ecological selection pressures. Carotenoid pigments responsible for many yellow to red signals exhibit two related properties that could link ecology with appearance by nontraditional means. Ecologic homoplasy could arise through ecophenotypy because all animals must obtain carotenoids through their diet. Such homoplasy also could be hidden from view because increased carotenoid levels are more strongly encoded by decreased reflectance over ultraviolet (UV) wavelengths invisible to humans. To explore these possibilities, I examined apparent matches or mismatches between color and ecology among insectivorous (low carotenoid diet) and frugivorous (high carotenoid diet) bird species in relation to the typical yellow and black plumage pattern of insectivorous, UV-sensitive titmice (Paridae). Diagnostic features of reflectance spectra indicated that all yellow plumages resulted from carotenoids, black plumages from melanins, and olive green plumages from codeposition of both pigments. However, reflectance by carotenoid-bearing plumages correlated with diet independent of plumage pattern; compared to the insectivores, frugivores had reduced amounts of UV reflectance, and to a lesser extent, "red shifts" in longer-wavelength reflectance. Furthermore, an asymptotic decrease in amount of UV with increased redness implied that plumage reflectance of insectivorous species differed more over UV wavelengths, whereas that of frugivorous species differed more over longer wavelengths. I verified that dietary links to plumage reflectance resulted from greater amounts of plumage carotenoids in frugivores, presumably due to their carotenoid-rich diets. All of these ecological associations transcended post-mortem or post-breeding color change, and phylogeny. Thus, predictable associations between avian-visible plumage reflectance, pigmentation, and diet across evolutionary scales may arise directly (diet per se) or indirectly (honest signaling of diet) by ecophenotypy, although various genetic factors also may play a role.

Carotenoid biotechnology in plants for nutritionally improved foods

Carotenoids participate in light harvesting and are essential for photoprotection in photosynthetic plant tissues. They also furnish non‐photosynthetic flowers and fruits with yellow to red colors to attract animals for pollination and dispersal of seeds. Although animals can not synthesize carotenoids de novo, carotenoid‐derived products such as retinoids (including vitamin A) are required as visual pigments and signaling molecules. Dietary carotenoids also provide health benefits based on their antioxidant properties. The main pathway for carotenoid biosynthesis in plants and microorganisms has been virtually elucidated in recent years, and some of the identified biosynthetic genes have been successfully used in metabolic engineering approaches to overproduce carotenoids of interest in plants. Alternative approaches that enhance the metabolic flux to carotenoids by upregulating the production of their isoprenoid precursors or interfere with light‐mediated regulation of carotenogenesis have been recently shown to result in increased carotenoid levels. Despite spectacular achievements in the metabolic engineering of plant carotenogenesis, much work is still ahead to better understand the regulation of carotenoid biosynthesis and accumulation in plant cells. New genetic and genomic approaches are now in progress to identify regulatory factors that might significantly contribute to improve the nutritional value of plant‐derived foods by increasing their carotenoid levels.

The Oxidative Stress-Sensitive yap1 Null Strain of Saccharomyces cerevisiae Becomes Resistant Due to Increased Carotenoid Levels upon the Introduction of the Chlamydomonas reinhardtii cDNA, Coding for the 60S Ribosomal Protein L10a

The Saccharomyces cerevisiae yap1 null strain was transformed with a Chlamydomonas reinhardtii cDNA expression library. A 688-bp cDNA fragment, coding for the 60S ribosomal protein L10a (RPL10a), restored the capacity of the S. cerevisiae yap1 null strain to resist oxidative stress. The rpl10a gene is a single-copy gene in C. reinhardtii and encodes a constitutively produced 1.35-kb mRNA. The deduced 214-residue amino acid sequence was highly related with RPL10a proteins from eukarya (between 46.1 and 63.7% identity) and archaea (between 24.5 and 29.2% identity). Resistant transformants were pink, due to increased carotenoid levels, with the same chemical structure as torularhodin, the main carotenoid of the pink yeast Rhodotorula mucilaginosa. The pink transformants showed high resistance levels against H2O2, paraquat, menadione, and UV light. Partial inhibition of the carotenoid synthesis by diphenylamine reduced the resistance levels, demonstrating the role of excess carotenoid synthesis in the resistance mechanism.
