Atrial natriuretic factor-like immunoreactivity in spinal cord and in primary sensory neurons of spinal and trigeminal ganglia of guinea-pig: correlation with tachykinin immunoreactivity*

Atrial natriuretic factor (ANF) is a cardiac hormone with various functions in body homeostasis. It is also processed in the brain and in the peripheral nervous system where it appears to play a role as a neuromodulator. Little is known about the presence of ANF throughout the spinal cord of the guinea-pig. We therefore examined the distribution of ANF and its possible interrelation with primary sensory afferents in this species. Using enzyme-and fluorescence-immunohistochemistry on deparaffinized sections. ANF-like immunoreactivity was found to be present in nerve fibers in laminae I/II of the spinal cord and in neurons of spinal and trigeminal ganglia. Tachykinins and ANF coexisted in very few fibers of the spinal cord but did not coexist in primary sensory spinal or trigeminal neurons. Our results indicate that spinal ANF-immunoreactive fibers are of dual origin, primary sensory and non-primary sensory. The possibly heterogeneous source of the non-primary sensory ANF, its possible coexistence with other co-transmitters and functional implications are discussed.

Distribution of neuropeptide Y-like immunoreactivity in the rat central nervous system—I. Radioimmunoassay and chromatographic characterisation

The distribution of neuropeptide Y-like immunoreactivity in the rat brain was investigated by means of immunochemical techniques. In the first part of the study (present paper) neuropeptide Y radioimmunoassays were characterised and the chromatographic properties and regional distribution of neuropeptide Y-like immunoreactivity was investigated. The second part of the study (accompanying paper) involved immunohistochemical techniques. Extracts from several regions of rat brain were found to contain immunoreactivity that behaved like synthetic porcine neuropeptide Y in three test systems: 1. (1) dilution in the radioimmunoassay (test of antigenic properties), 2. (2) gel chromatography (molecular weight), 3. (3) reverse phase high performance liquid chromatography (solubility properties). Experiments were conducted to optimise the extraction of neuropeptide Y. Boiling 0.1 M sodium phosphate buffer, pH 7.4, extracted at least two times as much immunoreactivity from whole brain pieces as other buffers. The nature of the extracted immunoreactivity was confirmed using chromatography. Experiments (using added iodinated or unlabelled neuropeptide Y standards) demonstrated that the differences between extraction media could not be explained by differential recovery of the peptide, although differences in recovery between media existed. Tissue sample weight was found to influence neuropeptide Y recovery. Evidence that rat neuropeptide Y-like immunoreactivity was not identical to the porcine peptide was obtained from experiments which demonstrated an early eluting peak of immunoreactivity in addition to the main peak on high performance liquid chromatograms. This material could be generated by oxidation of extracted rat neuropeptide Y, suggesting the presence in the rat peptide of a methionine residue. Some evidence of high molecular weight neuropeptide Y precursors was obtained from chromatography of hypothalamus extracts. Bovine pancreatic polypeptide-like material represented < 1% of the amounts of neuropeptide Y in the brain. The distribution of neuropeptide Y-like immunoreactivity was non-uniform in the rat brain with highest concentrations observed in the hypothalamus, amygdaloid complex and periaqueductal central gray matter. Other regions of forebrain contained moderate to high concentrations including olfactory tubercle, striatum, nucleus accumbens, neocortex and hippocampus. Negligible amounts were detected in the cerebellum. In spinal cord immunoreactivity was concentrated in the dorsal horn, although measurable amounts were found in the ventral horn. The neurointermediate but not anterior lobe of the pituitary contained neuropeptide Y.

Changes in TRH immunoreactivity in spinal cord after experimental spinal injury

Concentrations of immunoreactive TRH (TRH- ir ) in rat spinal cord were examined after traumatic injury. TRH- ir was significantly increased at the injury site (thoracic region) and rostral areas (cervical region), and significantly decreased below the injury site. Changes were delayed and time-dependent. Since TRH may serve as an excitatory neurotransmitter within the spinal cord, these changes may contribute to the functional alterations observed after spinal injury.

Increased dynorphin immunoreactivity in spinal cord after traumatic injury.

Opiate antagonists, at high doses, have been shown to improve physiological variables and outcome after experimental spinal injury. Dynorphin appears to be unique amongst opioids in producing hindlimb paralysis after intrathecal injection. Taken together, these findings suggest a possible pathophysiological role for endogenous opioids, particularly dynorphin, in spinal injury. In the present studies we examined the relationship between changes in dynorphin immunoreactivity (Dyn-ir) in rat spinal cord after traumatic injury and the subsequent motor dysfunction. Trauma was associated with significantly increased Dyn-ir at the injury site, but not distant from the lesion. Dyn-ir was found elevated as early as 2 h and as late as 2 weeks after trauma, and was significantly correlated with the degree of injury. These data are consistent with the hypothesis that dynorphin systems may be involved in the secondary injury that follows spinal trauma.

Effects of traumatic injury on dynorphin immunoreactivity in spinal cord

Traumatic spinal cord injury in rats resulted in a significant eleation of dynorphin A immunoreactivity in spinal cord tissue at the level of, and below, the site of injury. [Leu 5]enkephalin levels in the same tissue samples were not significantly altered following severe injury. Dynorphin A immunoreactivity was found in the fraction relatively enriched in synaptosomes after subcellular fractionation of spinal cord tissue. The dynorphin A content of this fraction was not significantly changed following injury, suggesting that dynorphin containing nerve terminals and axons are not severely damaged as a result of the injury.
