DMHGNN: Double multi-view heterogeneous graph neural network framework for drug-target interaction prediction

Accurate identification of drug-target interactions (DTIs) plays a crucial role in drug discovery. Compared with traditional experimental methods that are labor-intensive and time-consuming, computational methods for drug-target interactions prediction are more popular in recent years. Conventional computational methods almost simply view heterogeneous network constructed by the drug-related and protein-related dataset instead of comprehensively exploring drug-protein pair (DPP) information. To address this limitation, we proposed a Double Multi-view Heterogeneous Graph Neural Network framework for drug-target interaction prediction (DMHGNN). In DMHGNN, one multi-view heterogeneous graph neural network is based on meta-paths and denoising autoencoder for protein-, drug-related heterogeneous network learning, and another multi-view heterogeneous graph neural network is based on multi-channel graph convolutional network for drug-protein pair similarity network learning. First, a meta-path-based graph encoder with the attention mechanism is used for substructure learning of complex relationships from heterogeneous network constructed by proteins, drugs, side-effects and diseases, obtaining key information that is easy to be ignored in global learning of heterogeneous networks, and multi-source neighbouring features for drugs and proteins are learned from heterogeneous network via denoising auto-encoder model. Then, multi-view graphs of drug-protein pairs (DPPs) including the topology graph, semantics graph and collaborative graph with shared weights are constructed, and the multi-channel graph convolutional network (GCN) is utilized to learn the deep representation of DPPs. Finally, a multi-layer fully connection network is trained to predict drug-target interactions. Experiments have demonstrated its effectiveness and better performance than state-of-the-art methods.

MGACL: Prediction Drug-Protein Interaction Based on Meta-Graph Association-Aware Contrastive Learning.

The identification of drug-target interaction (DTI) is crucial for drug discovery. However, how to reduce the graph neural network's false positives due to its bias and negative transfer in the original bipartite graph remains to be clarified. Considering that the impact of heterogeneous auxiliary information on DTI varies depending on the drug and target, we established an adaptive enhanced personalized meta-knowledge transfer network named Meta Graph Association-Aware Contrastive Learning (MGACL), which can transfer personalized heterogeneous auxiliary information from different nodes and reduce data bias. Meanwhile, we propose a novel DTI association-aware contrastive learning strategy that aligns high-frequency drug representations with learned auxiliary graph representations to prevent negative transfer. Our study improves the DTI prediction performance by about 3%, evaluated by analyzing the area under the curve (AUC) and area under the precision-recall curve (AUPRC) compared with existing methods, which is more conducive to accurately identifying drug targets for the development of new drugs.

DTRE: A model for predicting drug-target interactions of endometrial cancer based on heterogeneous graph

Endometrial cancer is one of the most common gynecological malignancies affecting women worldwide, posing a serious threat to women’s health. Moreover, the identification of drug-target interactions (DTIs) is typically a time-consuming and costly critical step in drug discovery. In order to identify potential DTIs to enhance targeted therapy for endometrial cancer, we propose a deep learning model named DTRE (Drug-Target Relationship Enhanced) based on a heterogeneous graph to predict DTIs, which utilizes the relationships between drugs and targets to effectively capture their interactions. In the heterogeneous graph, nodes represent drugs and targets, and edges represent their interactions, then the representations of drugs and targets are learned through graph convolutional network, graph attention network and attention mechanism. Experimental results on the dataset proposed in this paper show that the AUC and AUPR of DTRE achieve 0.870 and 0.872 respectively, significantly outperforming comparative models and indicating that DTRE can effectively predict DTIs when applied to large-scale data. Additionally, DTRE also predicts the potential DTIs for endometrial cancer, providing new insights into targeted therapy for it.

Enhancing generalizability and performance in drug-target interaction identification by integrating pharmacophore and pre-trained models.

In drug discovery, it is crucial to assess the drug-target binding affinity (DTA). Although molecular docking is widely used, computational efficiency limits its application in large-scale virtual screening. Deep learning-based methods learn virtual scoring functions from labeled datasets and can quickly predict affinity. However, there are three limitations. First, existing methods only consider the atom-bond graph or one-dimensional sequence representations of compounds, ignoring the information about functional groups (pharmacophores) with specific biological activities. Second, relying on limited labeled datasets fails to learn comprehensive embedding representations of compounds and proteins, resulting in poor generalization performance in complex scenarios. Third, existing feature fusion methods cannot adequately capture contextual interaction information. Therefore, we propose a novel DTA prediction method named HeteroDTA. Specifically, a multi-view compound feature extraction module is constructed to model the atom-bond graph and pharmacophore graph. The residue concat graph and protein sequence are also utilized to model protein structure and function. Moreover, to enhance the generalization capability and reduce the dependence on task-specific labeled data, pre-trained models are utilized to initialize the atomic features of the compounds and the embedding representations of the protein sequence. A context-aware nonlinear feature fusion method is also proposed to learn interaction patterns between compounds and proteins. Experimental results on public benchmark datasets show that HeteroDTA significantly outperforms existing methods. In addition, HeteroDTA shows excellent generalization performance in cold-start experiments and superiority in the representation learning ability of drug-target pairs. BiTGNN: Prediction of drug–target interactions based on bidirectional transformer and graph neural network on heterogeneous graph

Drug–target interaction (DTI) is a widely explored topic in the field of bioinformatics and plays a pivotal role in drug discovery. However, the traditional bio-experimental process of drug–target interaction identification requires a large investment of time and labor. To address this challenge, graph neural network (GNN) approaches in deep learning are becoming a prominent trend in the field of DTI research, which is characterized by multimodal processing of data, feature learning and interpretability in DTI. Nevertheless, some methods are still limited by homogeneous graphs and single features. To address the problems, we mechanistically analyze graph convolutional neural networks (GCNs) and graph attentional neural networks (GATs) to propose a new model for the prediction of drug–target interactions using graph neural networks named BiTGNN [Bidirectional Transformer (Bi-Transformer)–graph neural network]. The method first establishes drug–target pairs through the pseudo-position specificity scoring matrix (PsePSSM) and drug fingerprint data, and constructs a heterogeneous network by utilizing the relationship between the drug and the target. Then, the computational extraction of drug and target attributes is performed using GCNs and GATs for the purpose of model information flow extension and graph information enhancement. We collect interaction data using the proposed Bi-Transformer architecture, in which we design a bidirectional cross-attention mechanism for calculating the effects of drug–target interactions for realistic biological interaction simulations. Finally, a feed-forward neural network is used to obtain the feature matrices of the drug and the target, and DTI prediction is performed by fusing the two feature matrices. The Enzyme, Ion Channel (IC), G Protein-coupled Receptor (GPCR) and Nuclear Receptor (NR) datasets are used in the experiments, and compared with several existing mainstream models, our model outperforms in Area Under the ROC Curve (AUC), Specificity, Accuracy and the metric Area Under the Precision–Recall Curve (AUPR).

Drug-Online: an online platform for drug-target interaction, affinity, and binding sites identification using deep learning

BackgroundAccurately identifying drug-target interaction (DTI), affinity (DTA), and binding sites (DTS) is crucial for drug screening, repositioning, and design, as well as for understanding the functions of target. Although there are a few online platforms based on deep learning for drug-target interaction, affinity, and binding sites identification, there is currently no integrated online platforms for all three aspects.ResultsOur solution, the novel integrated online platform Drug-Online, has been developed to facilitate drug screening, target identification, and understanding the functions of target in a progressive manner of “interaction-affinity-binding sites”. Drug-Online platform consists of three parts: the first part uses the drug-target interaction identification method MGraphDTA, based on graph neural networks (GNN) and convolutional neural networks (CNN), to identify whether there is a drug-target interaction. If an interaction is identified, the second part employs the drug-target affinity identification method MMDTA, also based on GNN and CNN, to calculate the strength of drug-target interaction, i.e., affinity. Finally, the third part identifies drug-target binding sites, i.e., pockets. The method pt-lm-gnn used in this part is also based on GNN.ConclusionsDrug-Online is a reliable online platform that integrates drug-target interaction, affinity, and binding sites identification. Drug-target Affinity Prediction by Molecule Secondary Structure Representation Network.

Identification of drug-target interactions (DTI) is a crucial step in drug development with high specificity and low toxicity. To accelerate the process, computer-aided DTI prediction algorithms have been used to screen compounds or targets rapidly. Furthermore, DTI prediction can be used to identify potential targets for existing drugs, thus uncovering new indications and repositioning them. Therefore, it is of great importance to develop efficient and accurate DTI prediction algorithms. Current algorithms usually represent drugs as extracted features, which are learned by convolutional neural networks (CNNs) from its linear representation, or utilize graph neural networks (GNNs) to learn its graph representation. However, these methods either lose information or fail to capture the structural information of the drug. To address this issue, a novel molecule secondary structure representation network (MSSRN) is proposed to learn drug characterization more accurately. Firstly, the network performs relational graph convolutional networks (R-GCNs) on the drug's molecular graph and integrates drug sequence convolutions to learn the sequential information. Secondly, inspired by the attention mechanism, spatial importance weights of the drug sequence are calculated to guide R-GCNs to learn the topological information of the drug. A drug-target affinity model, called MSSRN-DTA, was then constructed by using MSSRN to learn drug structure and CNN to learn protein sequence. The effectiveness of the proposed method is verified by comparing it with other alternative methods and baseline models on two benchmark datasets.

BINDTI: A bi-directional Intention network for drug-target interaction identification based on attention mechanisms.

The identification of drug-target interactions (DTIs) is an essential step in drug discovery. In vitro experimental methods are expensive, laborious, and time-consuming. Deep learning has witnessed promising progress in DTI prediction. However, how to precisely represent drug and protein features is a major challenge for DTI prediction. Here, we developed an end-to-end DTI identification framework called BINDTI based on bi-directional Intention network. First, drug features are encoded with graph convolutional networks based on its 2D molecular graph obtained by its SMILES string. Next, protein features are encoded based on its amino acid sequence through a mixed model called ACmix, which integrates self-attention mechanism and convolution. Third, drug and target features are fused through bi-directional Intention network, which combines Intention and multi-head attention. Finally, unknown drug-target (DT) pairs are classified through multilayer perceptron based on the fused DT features. The results demonstrate that BINDTI greatly outperformed four baseline methods (i.e., CPI-GNN, TransfomerCPI, MolTrans, and IIFDTI) on the BindingDB, BioSNAP, DrugBank, and Human datasets. More importantly, it was more appropriate to predict new DTIs than the four baseline methods on imbalanced datasets. Ablation experimental results elucidated that both bi-directional Intention and ACmix could greatly advance DTI prediction. The fused feature visualization and case studies manifested that the predicted results by BINDTI were basically consistent with the true ones. We anticipate that the proposed BINDTI framework can find new low-cost drug candidates, improve drugs' virtual screening, and further facilitate drug repositioning as well as drug discovery. Prediction of Drug-Target Interactions with High-Quality Negative Samples and A Network-Based Deep Learning Framework.

Identification of drug-target interactions (DTIs) plays a crucial role in drug discovery. Compared to traditional experimental methods, computer-based methods for predicting DTIs can significantly reduce the time and financial burdens of drug development. In recent years, numerous machine learning-based methods have been proposed for predicting potential DTIs. However, a common limitation among these methods is the absence of high-quality negative samples. Moreover, the effective extraction of multisource information of drugs and proteins for DTI prediction remains a significant challenge. In this paper, we investigated two aspects: the selection of high-quality negative samples and the construction of a high-performance DTI prediction framework. Specifically, we found two types of hidden biases when randomly selecting negative samples from unlabeled drug-protein pairs and proposed a negative sample selection approach based on complex network theory. Furthermore, we proposed a novel DTI prediction method named HNetPa-DTI, which integrates topological information from the drug-protein-disease heterogeneous network and gene ontology (GO) and pathway annotation information of proteins. Specifically, we extracted topological information of the drug-protein-disease heterogeneous network using heterogeneous graph neural networks, and obtained GO and pathway annotation information of proteins from the GO term semantic similarity networks, GO term-protein bipartite networks, and pathway-protein bipartite network using graph neural networks. Experimental results show that HNetPa-DTI outperforms the baseline methods on four types of prediction tasks, demonstrating the superiority of our method. TAG-DTA: Binding-region-guided strategy to predict drug-target affinity using transformers

The proper assessment of target-specific compound selectivity is paramount in the drug discovery context, promoting the identification of drug-target interactions (DTIs) and the discovery of potential leads. On that account, the accurate prediction of an unbiased drug-target binding affinity (DTA) metric is pivotal to understanding the binding process. Most in silico computational approaches, however, neglect the inter-dependency of the proteomics, chemical, and pharmacological spaces and the explainability during the model construction. Furthermore, these methods have yet to actively include information associated with binding pockets during the learning process, which is essential to DTA prediction performance and model explainability. In this study, we propose an end-to-end binding-region-guided Transformer-based architecture that simultaneously predicts the 1D binding pocket and the binding affinity of DTI pairs, where the prediction of the 1D binding pocket guides and conditions the prediction of DTA. This architecture uses 1D raw sequential and structural data to represent the proteins and compounds, respectively, and combines multiple Transformer-Encoder blocks to capture and learn the proteomics, chemical, and pharmacological contexts. The predicted 1D binding pocket conditions the attention mechanism of the Transformer-Encoder used to learn the pharmacological space in order to model the inter-dependency amongst binding-related positions. The results show that the proposed architecture, TAG-DTA, achieved the best performance in DTA prediction compared to state-of-the-art benchmarks, including in unknown subsets of the proteomics and chemical representation spaces. Moreover, the 1D binding pocket prediction increases the discriminative power and robustness of the aggregate representation of the pharmacological space and improves the DTA prediction performance. Overall, this research study validates the applicability of an end-to-end Transformer-based architecture in the context of drug discovery, and that combining computationally different yet contextually related tasks is critical to new findings in the DTI domain. Additionally, it shows that TAG-DTA is capable of providing increasing DTI and prediction understanding due to the nature of the attention blocks and prediction of the 1D binding pocket. VGAEDTI: drug-target interaction prediction based on variational inference and graph autoencoder

MotivationAccurate identification of Drug-Target Interactions (DTIs) plays a crucial role in many stages of drug development and drug repurposing. (i) Traditional methods do not consider the use of multi-source data and do not consider the complex relationship between data sources. (ii) How to better mine the hidden features of drug and target space from high-dimensional data, and better solve the accuracy and robustness of the model.ResultsTo solve the above problems, a novel prediction model named VGAEDTI is proposed in this paper. We constructed a heterogeneous network with multiple sources of information using multiple types of drug and target dataIn order to obtain deeper features of drugs and targets, we use two different autoencoders. One is variational graph autoencoder (VGAE) which is used to infer feature representations from drug and target spaces. The second is graph autoencoder (GAE) propagating labels between known DTIs. Experimental results on two public datasets show that the prediction accuracy of VGAEDTI is better than that of six DTIs prediction methods. These results indicate that model can predict new DTIs and provide an effective tool for accelerating drug development and repurposing.

Novel perspective for protein-drug interaction analysis: atomic force microscope.

Proteins are major drug targets, and drug-target interaction identification and analysis are important factors for drug discovery. Atomic force microscopy (AFM) is a powerful tool making it possible to image proteins with nanometric resolution and probe intermolecular forces under physiological conditions. We review recent studies conducted in the field of target protein drug discovery using AFM-based analysis technology, including drug-driven changes in nanomechanical properties of protein morphology and interactions. Underlying mechanisms (including thermodynamic and kinetic parameters) of the drug-target interaction and drug-modulating protein-protein interaction (PPI) on the surfaces of models or living cells are discussed. Furthermore, challenges and the outlook for the field are likewise discussed. Overall, this insight into the mechanical properties of protein-drug interactions provides an unprecedented information framework for rational drug discovery in the pharmaceutical field.

CoaDTI: multi-modal co-attention based framework for drug-target interaction annotation.

The identification of drug-target interactions (DTIs) plays a vital role for in silico drug discovery, in which the drug is the chemical molecule, and the target is the protein residues in the binding pocket. Manual DTI annotation approaches remain reliable; however, it is notoriously laborious and time-consuming to test each drug-target pair exhaustively. Recently, the rapid growth of labelled DTI data has catalysed interests in high-throughput DTI prediction. Unfortunately, those methods highly rely on the manual features denoted by human, leading to errors. Here, we developed an end-to-end deep learning framework called CoaDTI to significantly improve the efficiency and interpretability of drug target annotation. CoaDTI incorporates the Co-attention mechanism to model the interaction information from the drug modality and protein modality. In particular, CoaDTI incorporates transformer to learn the protein representations from raw amino acid sequences, and GraphSage to extract the molecule graph features from SMILES. Furthermore, we proposed to employ the transfer learning strategy to encode protein features by pre-trained transformer to address the issue of scarce labelled data. The experimental results demonstrate that CoaDTI achieves competitive performance on three public datasets compared with state-of-the-art models. In addition, the transfer learning strategy further boosts the performance to an unprecedented level. The extended study reveals that CoaDTI can identify novel DTIs such as reactions between candidate drugs and severe acute respiratory syndrome coronavirus 2-associated proteins. EFMSDTI: Drug-target interaction prediction based on an efficient fusion of multi-source data.

Accurate identification of Drug Target Interactions (DTIs) is of great significance for understanding the mechanism of drug treatment and discovering new drugs for disease treatment. Currently, computational methods of DTIs prediction that combine drug and target multi-source data can effectively reduce the cost and time of drug development. However, in multi-source data processing, the contribution of different source data to DTIs is often not considered. Therefore, how to make full use of the contribution of different source data to predict DTIs for efficient fusion is the key to improving the prediction accuracy of DTIs. In this paper, considering the contribution of different source data to DTIs prediction, a DTIs prediction approach based on an effective fusion of drug and target multi-source data is proposed, named EFMSDTI. EFMSDTI first builds 15 similarity networks based on multi-source information networks classified as topological and semantic graphs of drugs and targets according to their biological characteristics. Then, the multi-networks are fused by selective and entropy weighting based on similarity network fusion (SNF) according to their contribution to DTIs prediction. The deep neural networks model learns the embedding of low-dimensional vectors of drugs and targets. Finally, the LightGBM algorithm based on Gradient Boosting Decision Tree (GBDT) is used to complete DTIs prediction. Experimental results show that EFMSDTI has better performance (AUROC and AUPR are 0.982) than several state-of-the-art algorithms. DeepFusionDTA: Drug-Target Binding Affinity Prediction With Information Fusion and Hybrid Deep-Learning Ensemble Model

Identification of drug-target interaction (DTI) is the most important issue in the broad field of drug discovery. Using purely biological experiments to verify drug-target binding profiles takes lots of time and effort, so computational technologies for this task obviously have great benefits in reducing the drug search space. Most of computational methods to predict DTI are proposed to solve a binary classification problem, which ignore the influence of binding strength. Therefore, drug-target binding affinity prediction is still a challenging issue. Currently, lots of studies only extract sequence information that lacks feature-rich representation, but we consider more spatial features in order to merge various data in drug and target spaces. In this study, we propose a two-stage deep neural network ensemble model for detecting drug-target binding affinity, called DeepFusionDTA, via various information analysis modules. First stage is to utilize sequence and structure information to generate fusion feature map of candidate protein and drug pair through various analysis modules based deep learning. Second stage is to apply bagging-based ensemble learning strategy for regression prediction, and we obtain outstanding results by combining the advantages of various algorithms in efficient feature abstraction and regression calculation. Importantly, we evaluate our novel method, DeepFusionDTA, which delivers 1.5 percent CI increase on KIBA dataset and 1.0 percent increase on Davis dataset, by comparing with existing prediction tools, DeepDTA. Furthermore, the ideas we have offered can be applied to in-silico screening of the interaction space, to provide novel DTIs which can be experimentally pursued. DTI-HETA: prediction of drug-target interactions based on GCN and GAT on heterogeneous graph.

Drug-target interaction (DTI) prediction plays an important role in drug repositioning, drug discovery and drug design. However, due to the large size of the chemical and genomic spaces and the complex interactions between drugs and targets, experimental identification of DTIs is costly and time-consuming. In recent years, the emerging graph neural network (GNN) has been applied to DTI prediction because DTIs can be represented effectively using graphs. However, some of these methods are only based on homogeneous graphs, and some consist of two decoupled steps that cannot be trained jointly. To further explore GNN-based DTI prediction by integrating heterogeneous graph information, this study regards DTI prediction as a link prediction problem and proposes an end-to-end model based on HETerogeneous graph with Attention mechanism (DTI-HETA). In this model, a heterogeneous graph is first constructed based on the drug-drug and target-target similarity matrices and the DTI matrix. Then, the graph convolutional neural network is utilized to obtain the embedded representation of the drugs and targets. To highlight the contribution of different neighborhood nodes to the central node in aggregating the graph convolution information, a graph attention mechanism is introduced into the node embedding process. Afterward, an inner product decoder is applied to predict DTIs. To evaluate the performance of DTI-HETA, experiments are conducted on two datasets. The experimental results show that our model is superior to the state-of-the-art methods. Also, the identification of novel DTIs indicates that DTI-HETA can serve as a powerful tool for integrating heterogeneous graph information to predict DTIs.

Predicting Drug-Target Affinity Based on Recurrent Neural Networks and Graph Convolutional Neural Networks.

Drug development requires a lot of money and time, and the outcome of the challenge is unknown. So, there is an urgent need for researchers to find a new approach that can reduce costs. Therefore, the identification of drug-target interactions (DTIs) has been a critical step in the early stages of drug discovery. These computational methods aim to narrow the search space for novel DTIs and to elucidate the functional background of drugs. Most of the methods developed so far use binary classification to predict the presence or absence of interactions between the drug and the target. However, it is more informative but also more challenging to predict the strength of the binding between a drug and its target. If the strength is not strong enough, such a DTI may not be useful. Hence, the development of methods to predict drug-target affinity (DTA) is of significant importance Method: We have improved the GraphDTA model from a dual-channel model to a triple-channel model. We interpreted the target/protein sequences as time series and extracted their features using the LSTM network. For the drug, we considered both the molecular structure and the local chemical background, retaining the four variant networks used in GraphDTA to extract the topological features of the drug and capturing the local chemical background of the atoms in the drug by using BiGRU. Thus, we obtained the latent features of the target and two latent features of the drug. The connection of these three feature vectors is then inputted into a 2 layer FC network, and a valuable binding affinity is the output. We used the Davis and Kiba datasets, using 80% of the data for training and 20% of the data for validation. Our model showed better performance when compared with the experimental results of GraphDTA Conclusion: In this paper, we altered the GraphDTA model to predict drug-target affinity. It represents the drug as a graph and extracts the two-dimensional drug information using a graph convolutional neural network. Simultaneously, the drug and protein targets are represented as a word vector, and the convolutional neural network is used to extract the time-series information of the drug and the target. We demonstrate that our improved method has better performance than the original method. In particular, our model has better performance in the evaluation of benchmark databases.

Supervised graph co-contrastive learning for drug-target interaction prediction.

Identification of Drug-Target Interactions (DTIs) is an essential step in drug discovery and repositioning. DTI prediction based on biological experiments is time-consuming and expensive. In recent years, graph learning-based methods have aroused widespread interest and shown certain advantages on this task, where the DTI prediction is often modeled as a binary classification problem of the nodes composed of drug and protein pairs (DPPs). Nevertheless, in many real applications, labeled data are very limited and expensive to obtain. With only a few thousand labeled data, models could hardly recognize comprehensive patterns of DPP node representations, and are unable to capture enough commonsense knowledge, which is required in DTI prediction. Supervised contrastive learning gives an aligned representation of DPP node representations with the same class label. In embedding space, DPP node representations with the same label are pulled together, and those with different labels are pushed apart. We propose an end-to-end supervised graph co-contrastive learning model for DTI prediction directly from heterogeneous networks. By contrasting the topology structures and semantic features of the drug-protein-pair network, as well as the new selection strategy of positive and negative samples, SGCL-DTI generates a contrastive loss to guide the model optimization in a supervised manner. Comprehensive experiments on three public datasets demonstrate that our model outperforms the SOTA methods significantly on the task of DTI prediction, especially in the case of cold start. Furthermore, SGCL-DTI provides a new research perspective of contrastive learning for DTI prediction. The research shows that this method has certain applicability in the discovery of drugs, the identification of drug-target pairs and so on.

HIDTI: integration of heterogeneous information to predict drug-target interactions

Identification of drug-target interactions (DTIs) plays a crucial role in drug development. Traditional laboratory-based DTI discovery is generally costly and time-consuming. Therefore, computational approaches have been developed to predict interactions between drug candidates and disease-causing proteins. We designed a novel method, termed heterogeneous information integration for DTI prediction (HIDTI), based on the concept of predicting vectors for all of unknown/unavailable heterogeneous drug- and protein-related information. We applied a residual network in HIDTI to extract features of such heterogeneous information for predicting DTIs, and tested the model using drug-based ten-fold cross-validation to examine the prediction performance for unseen drugs. As a result, HIDTI outperformed existing models using heterogeneous information, and was demonstrating that our method predicted heterogeneous information on unseen data better than other models. In conclusion, our study suggests that HIDTI has the potential to advance the field of drug development by accurately predicting the targets of new drugs.
