Yttrium iron garnet for hyperthermia applications: Synthesis, characterization and in-vitro analysis.

Exclusive magnetocaloric properties of orthoferrites offer advantages for their application in the magnetic hyperthermia as well as imaging applications. In the present study, the effect of yttrium concentration on the magnetic characteristics of the iron oxide based nanomaterials was analyzed to assess their potential for the hyperthermia applications. The Sol-gel method was used to synthesize the Yttrium Iron Garnet (YIG) based nanoparticles, using different molar ratios of Fe and Y precursors, followed by the calcination at 900, 1000 and 1100°C. XRD analysis determined the formation of the pure phase of yttrium iron garnet Y3Fe5O12 (YIG) at 0.5 molar ratio of yttrium at all the calcination temperatures and pure phase of yttrium iron perovskite YFeO3 (YIP) for 1molar ratio of yttrium at 1000 and 1100°C. The mean particle size was observed in the range of 100 to 400nm. The magnetic characterization studies showed the highest saturation magnetization for the sample containing 0.5molar ratio of the yttrium calcinated at 1000°C. The magnetization values were linearly related to the contents of YIG phases in the synthesized samples. Induction heating of YIG resulted in the hyperthermia temperature (42 to 44°C) in 13min with the SAR values 114.65W/g at 1mg/ml. The prepared samples showed no in-vitro toxic effects on the MG63 cells (>90% cell viability). In addition, in-vitro treatment at hyperthermia temperature for 15min reduced cell viability of cancer cells (A549) to 55%, while no toxic effect was observed on MG 63 cells. The present study postulates Yttrium Iron Garnet as an effective therapeutic agent for hyperthermia cancer treatment.

3D in silico study of magnetic fluid hyperthermia of breast tumor using Fe3O4 magnetic nanoparticles.

Modeling and simulation of the temperature distribution, the mass concentration, and the heat transfer in the breast tissue are hot issues in magnetic fluid hyperthermia treatment of cancer. The breast tissue can be visualized as a porous matrix with saturated blood. In this paper, 3D in silico study of breast cancer hyperthermia using magnetic nanoparticles (MNPs) is conducted. The 3D FEM models are incorporated to investigate the infusion and backflow of nanofluid in the breast tumor, the diffusion of nanofluid, temperature distribution during the treatment, and prediction of the fraction of tumor necrosis while dealing with the thermal therapy. All the hyperthermia procedures are simulated and analyzed on COMSOL Multiphysics. The sensitivity of frequency and amplitude of the applied magnetic field (AMF) is investigated on the heating effect of the tumor. The mesh dependent solution of Penne's bioheat model is also analyzed. The simulated results demonstrate successful breast cancer treatment using MNPs with minimum side effects. Validation of current simulations results with experimental studies existing in literature advocates the success of our therapy. The increase in the amplitude and frequency of the AMF increases of the temperature in the tumor. The variation of mesh from coarser to finer increased the temperature through small fractions. We have also simulated the magnetic induction problem where the magnetic field is generated by current-carrying coil conductors induce heat in nearby breast tumors due to excitation of MNPs by magnetic flux. This research will aid treatment protocols and real-time clinical breast cancer treatments.

Impact of Different Set of Precipitants on the Formation of BaFe12O19 Phase Nanomaterials.

This work deals with the preparation of BaFe12O19 materials by soft chemical co-precipitation method using a different set of precipitants namely, (i) NaOH and Na₂CO₃, (ii) NH₂CONH₂ and NH₄OH, and (iii) NH₄HCO₃ and NH₄OH. The influence of these precipitants on the different properties is also investigated by various characterizations. The precipitants used in this work considerably affected the phase formation of BaFe12O19 materials. The thermal analyses reveal the thermal decomposition of the intermediate phase as well as the crystallization of BaFe12O19 phase. Due to the amorphous nature, all the co-precipitated products were annealed for 2 h at 1000 °C. Among the different annealed samples, the NaOH and Na₂CO₃ set of precipitant derived materials exhibit nearly single hexagonal phase of crystalline BaFe12O19 nanomaterials with an average crystalline size of ˜48 nm with a = 0.5888(4) nm and c = 2.320(2) nm. The particle size observed in micrograph was rather much higher than the average crystalline size obtained from XRD for the best sample. The BaFe12O19 nanomaterials studied in the present work have been found potential applications in magneto-optic recording media, biomolecular separations, magnetic carriers for drug targeting, permanent magnet, chip inductors, microwave absorbers, and hyperthermia cancer treatment.

Direct treatment versus indirect: Thermo-ablative and mild hyperthermia effects.

Hyperthermia is a rapidly growing field in cancer therapy and many advances have been made in understanding and applying the mechanisms of hyperthermia. Secondary effects of hyperthermia have been increasingly recognized as important in therapeutic effects and multiple studies have started to elucidate their implications for treatment. Immune effects have especially been recognized as important in the efficacy of hyperthermia treatment of cancer. Both thermo-ablative and mild hyperthermia activate the immune system, but mild hyperthermia seems to be more effective at doing so. This may suggest that mild hyperthermia has some advantages over thermo-ablative hyperthermia and research into immune effects of mild hyperthermia should continue. Enhancing Targeted Cancer Treatment by Combining Hyperthermia and Radiotherapy Using Mn-Zn Ferrite Magnetic Nanoparticles.

Radiotherapy (RT) is a major treatment method for non-small-cell lung cancer (NSCLC), and development of new treatment modality is now critical to amplify the negative effects of RT on tumors. In this study, we demonstrated a nanoparticle-loaded block copolymer micellar system for cancer hyperthermia treatment (HT) that can be used for synergistic therapy under alternating magnetic field (AMF) and radiation field. Block copolymer micelles (polyethylene glycol-block-polycaprolactone, or PEG-PCL) containing hyaluronic acid (HA) and Mn-Zn ferrite magnetic nanoparticles (MZF) were fabricated via a two-step preparation. HA-modified Mn-Zn ferrite magnetic nanoparticles (MZF-HA) can be enriched in CD44 highly expressing tumor cells, such as A549 (human lung adenocarcinoma cell line), through an active targeting mechanism via receptor-ligand binding of HA and CD44 (HA receptor). MZF can generate thermal energy under an AMF, leading to a local temperature increase to approximately 43 °C at tumor sites for mild HT, and the increased tumor oxygenation can enhance the therapeutic effect of RT. In vitro experiments show that MZF-HA is able to achieve excellent specific targeting performance toward A549 cells with excellent biocompatibility as well as enhanced therapy performance under HT and RT in vitro by apoptosis flow cytometry. In the A549 subcutaneous tumor xenografts model, MRI confirms the enrichment of MZF-HA in tumor, and hypoxia immunohistochemistry analysis (IHC) proved the increased tumor oxygenation after HT. Furthermore, the tumor volume decreases to 49.6% through the combination of HT and RT in comparison with the 58.8% increase of the untreated group. These results suggest that the application of MZF-HA is able to increase the therapeutic effect of RT on A549 and can be used for further clinical NSCLC treatment evaluation.

Magnetic Nanoparticle Systems for Nanomedicine—A Materials Science Perspective

Iron oxide nanoparticles are the basic components of the most promising magneto-responsive systems for nanomedicine, ranging from drug delivery and imaging to hyperthermia cancer treatment, as well as to rapid point-of-care diagnostic systems with magnetic nanoparticles. Advanced synthesis procedures of single- and multi-core iron-oxide nanoparticles with high magnetic moment and well-defined size and shape, being designed to simultaneously fulfill multiple biomedical functionalities, have been thoroughly evaluated. The review summarizes recent results in manufacturing novel magnetic nanoparticle systems, as well as the use of proper characterization methods that are relevant to the magneto-responsive nature, size range, surface chemistry, structuring behavior, and exploitation conditions of magnetic nanosystems. These refer to particle size, size distribution and aggregation characteristics, zeta potential/surface charge, surface coating, functionalization and catalytic activity, morphology (shape, surface area, surface topology, crystallinity), solubility and stability (e.g., solubility in biological fluids, stability on storage), as well as to DC and AC magnetic properties, particle agglomerates formation, and flow behavior under applied magnetic field (magnetorheology).

Production and Characterization of Magnetic Fe3O4Nanoparticles Coated with PCL for Biomedical Applications

Currently, magnetic nanoparticles are widely studied with regard to their application in cancer treatment. This study aims to show a straightforward strategy for the production of Fe3O4 nanoparticles (NPs) with biocompatible surface modifications with polycaprolactone (PCL) for biomedical purposes. The effects of the polymer coating on the properties of magnetite were evaluated. Crystallinity, morphology, composition, hydrodynamic size and magnetic properties of the produced nanoparticles were analysed via X-ray diffractometry (XRD), Transmission Electron Microscopy (TEM), Fourier-Transform Infrared Spectroscopy (FTIR), Dynamic Light Scattering (DLS) and Vibrating Sample Magnetometry (VSM), respectively. The proposed method produced nanoparticles of magnetite with an average size between 9 and 11 nm, with spherical morphology and superparamagnetic properties. Magnetization values were not compromised even when the highest amount of polymer was used in the surface modification. On the other hand, the coating resulted in the decrease of the hydrodynamic size of the composites, indicating greater colloidal stability when the polymer was present. The obtained nanoparticles showed maintenance of significant superparamagneticbehavior, even in the presence of PCL on their surface. This phenomenon would allow for their application as a further optimized vector in hyperthermia cancer treatment, controlled drug delivery and resonance imaging. Influence of Moving Heat Source on Skin Tissue in the Context of Two-Temperature Caputo–Fabrizio Heat Transport Law

Modeling and understanding heat transport and temperature variations within biological tissues and body organs are key issues in medical thermal therapeutic applications, such as hyperthermia cancer treatment. In this analysis, the bio-heat equation has been studied in the context of a new formulation using Caputo–Fabrizio (CF) heat transport law. The heat equation for this problem involving the Three-Phase (3P)-lag model under two-temperature theory. The Laplace transform technique is implemented to solve the governing equations. The influences of the CF order, the two-temperature parameter and moving heat source velocity on the temperature of skin tissues have been precisely investigated. The numerical inversion of the Laplace transform is carried out using the Zakian method. The numerical outcomes of conductive temperature and thermodynamic temperatures have been represented graphically. Excellent predictive capability is demonstrated for identification of appropriate procedure to select different CF order to reach effective heating in hyperthermia treatment. Significant effect of thermal therapy is reported due to the effect of CF order, the two-temperature parameter and the velocity of moving heat source as well.

Magnetic nanoparticles for hyperthermia in cancer treatment: an emerging tool.

Cancer remains as the major cause of death worldwide. The main reason why available therapies fail is that a vicious cycle in established which initiates multiple pathways and recurrence after metastasis. Hyperthermic treatment, which involves heating tumor tissues to a moderate temperature of 40-43°C, has emerged as an effective strategy for treating tumors. This method is highly efficient at destroying tumor cells and does not induce the side effects of conventional cancer treatments. On the other hand, hyperthermic treatment method can be co-administered with conventional treatments. Nanotechnology had created huge opportunities in almost all areas of research, including the field of hyperthermic treatment. The utilization of magnetic nanoparticles (MNPs) offers functionalities not possible using conventional magnetic materials. In this review, we detail recent developments and applications of MNPs for hyperthermic treatment and discuss future possibilities.

Numerical Analysis of Human Cancer Therapy Using Microwave Ablation

Microwave ablation is one type of hyperthermia treatment of cancer that involves heating tumor cells. This technique uses electromagnetic wave effects to kill cancer cells. A micro-coaxial antenna is introduced into the biological tissue. The radiation emitted by the antenna is absorbed by the tissue and leads to the heating of cancer cells. The diffuse increase in temperature should reach a certain value to achieve the treatment of cancer cells but it should be less than a certain other value to avoid damaging normal cells. This is why hyperthermia treatment should be carefully monitored. A numerical simulation is useful and may provide valuable information. The bio-heat equation and Maxwell’s equations are solved using the finite element method. Electro-thermal effects, temperature distribution profile, specific absorption rate (SAR), and fraction of necrotic tissue within cancer cells are analyzed. The results show that SAR and temperature distribution are strongly affected by input microwave power. High microwave power causes a high SAR value and raises the temperature above 50 °C, which may destroy healthy cells. It is revealed that with a power of 10 W, the tumor cells will be killed without damaging the surrounding tissue.

Thermosensitive Nanosystems Associated with Hyperthermia for Cancer Treatment.

Conventional chemotherapy regimens have limitations due to serious adverse effects. Targeted drug delivery systems to reduce systemic toxicity are a powerful drug development platform. Encapsulation of antitumor drug(s) in thermosensitive nanocarriers is an emerging approach with a promise to improve uptake and increase therapeutic efficacy, as they can be activated by hyperthermia selectively at the tumor site. In this review, we focus on thermosensitive nanosystems associated with hyperthermia for the treatment of cancer, in preclinical and clinical use.

Excellent predictive capability is demonstrated for identification of an appropriate procedure to select different kernel functions to reach effective heating in hyperthermia treatment. Significant effect of thermal therapy is reported due to the effect of delay time and the velocity of moving heat source as well.

FDM-based 2D Numerical Study of Hyperthermia Cancer Treatment by Micro/Nano-Phase-Change Materials

Hyperthermia is one of the methods for cancer therapies by creating artificial fever. Protection of healthy tissues around the tumor tissue, which has a high temperature, is one of the major issues in this treatment. In present study, the two-dimensional transient model of biological tissue with a tumor is taken into account. The area of the tumor is embedded with micro/nano superparamagnetic materials, and the surrounding healthy tissues are embedded with microcapsules of micro/nano-phase-change materials (PCMs). The effect of concentrations, the radius of the microcapsule, the melting temperature and latent heat of phase-change nanoparticles and also superparamagnetic nanoparticles on thermal protection were examined numerically by using the finite difference method. The results showed that the use of PCMs causes sensible decreases in temperatures of the healthy tissue around the cancerous cells (in some places up to 3 ℃) that leads to treatment complications reduction. If the protection of healthy tissues is well done, it is possible to raise the temperature of the cancerous tissue to achieve more favorable treatment.

Biocompatibility of magnetic iron oxide nanoparticles for biomedical applications

Abstract Magnetic nanoparticles are highly promising for the usage in various biomedical applications including magnetic particle imaging (MPI), cancer hyperthermia treatment or as drug carriers. The present study aims at assessing in vitro biocompatibility of two commercially available magnetic iron oxide nanoparticle formulations: dextran-based magnetic nanoparticle synomag-D and bionized nanoferrite BNF-starch. Biological performance of both nanoparticle formulations were studied in human endothelial cells by analyzing cell viability and nanoparticle internalization in order to judge their suitability as theranostics.

The effects of intraparticle structure and interparticle interactions on the magnetic hysteresis loop of magnetic nanoparticles

Technologically relevant magnetic nanoparticles for biomedicine are rarely noninteracting single-domain nanoparticles; instead, they are often interacting, with complex physical and magnetic structures. In this paper, we present both experimental and simulated magnetic hysteresis loops of a system of magnetic nanoparticles with significant interparticle interactions and a well-defined intraparticle structure which are used for magnetic nanoparticle hyperthermia cancer treatment. Experimental measurements were made at 11 K on suspensions of magnetic nanoparticles dispersed in H2O which have been frozen in a range of applied magnetic fields to tune the interparticle interactions. Micromagnetic simulations of hysteresis loops investigated the roles of particle orientation with respect to the field and of particle chaining in the shape of the hysteresis loops. In addition, we present an analysis of the magnetic anisotropy arising from the combination of magnetocrystalline and shape anisotropy, given the well-defined internal structure of the nanoparticles. We find that the shape of the experimental hysteresis loops can be explained by the internal magnetic structure, modified by the effects of interparticle interactions from chaining.

Typical experiment vs. in-cell like conditions in magnetic hyperthermia: Effects of media viscosity and agglomeration

Abstract Magnetic nanoparticles (MNPs) can be used to transform electromagnetic energy into heat in hyperthermic treatment of cancer and other thermally activated therapies. The MNPs heating efficiency depends strongly on the combination of the MNPs’ structural properties and environmental conditions. MNPs hyperthermic yield is usually studied in diluted suspensions, although, in the actual therapy, the particles end mostly aggregated and fixed into cellular structures. In this work, the heating efficiency of low size dispersion Fe 3 O 4 MNPs, defined as the Specific Absorption Rate (SAR), was studied in two conditions: liquid suspension (ferrofluid FF, typical characterization state) and gel matrix (ferrogel FG, mimicking biological application environment). The samples were characterized by TEM, ZFC-FC and SAXS. Their magnetic response to radio-frequency fields was measured by induction in order to obtain SAR values from the magnetization cycles area. 3D maps of SAR versus field amplitude and frequency were elaborated in order to compare the response of fixed and suspended MNPs. Structural characterization shows FG’s MNPs agglomerated in a crystal-like mesostructure with a well defined interparticle distance. SAR results show a clear difference of behaviour between liquid and gel matrices, with larger SAR values for the FG sample indicating a lower resonance frequency, inside the studied region, for fixed MNP. Additionally, the local maximum suggested in FG’s SAR map indicates a behaviour outside linear response regimen as expected for the applied field amplitudes.

Biomedical Applications of Hyaluronic Acid-Based Nanomaterials in Hyperthermic Cancer Therapy.

Hyaluronic acid (HA) is a non-sulfated polysaccharide polymer with the properties of biodegradability, biocompatibility, and non-toxicity. Additionally, HA specifically binds to certain receptors that are over-expressed in cancer cells. To maximize the effect of drug delivery and cancer treatment, diverse types of nanomaterials have been developed. HA-based nanomaterials, including micelles, polymersomes, hydrogels, and nanoparticles, play a critical role in efficient drug delivery and cancer treatment. Hyperthermic cancer treatment using HA-based nanomaterials has attracted attention as an efficient cancer treatment approach. In this paper, the biomedical applications of HA-based nanomaterials in hyperthermic cancer treatment and combined therapies are summarized. HA-based nanomaterials may become a representative platform in hyperthermic cancer treatment.

An overview of available metamaterial-based antenna for non-invasive hyperthermia cancer treatment

This paper presents the outcome of a literature review that an overview of various types of antenna and metamaterial applicator performance towards cancerous tissue or cell for non-invasive hyperthermia cancer treatment (NIHCT) procedure. From the review, it shows that when LHM lens integrated with an antenna, focusing capabilities of the antenna towards the cancerous area can be improved. However, current applicators have a poor focusing effect when directed towards the actual tumor area. In conjunction with that, this paper proposes a new design of modified applicator that is microstrip antenna integrated with left-handed metamaterial (LHM) lens. The antenna termed microstrip-LHM (M-LHM) lens antenna is proposed for use in NIHCT. It is expected to improve the focusing capabilities of an antenna which is used to kill the cancerous area and thus improve the hyperthermia cancer treatment procedure success rate. In addition, this paper provides an overview of heating techniques used in hyperthermia to enhance focusing capabilities and a few metamaterial advantages that can improve the focusing effect and reduced the hot-spots. Specific Absorption Rate (SAR) will be investigated to evaluate the focusing abilities of the proposed applicator using the SEMCAD X Solver.

Iron-dextran as a thermosensitizer in radiofrequency hyperthermia for cancer treatment

Radiofrequency hyperthermia is a recently rediscovered oncotherapy rising in popularity. However, lack of a proper thermosensitizer limits current radiofrequency hyperthermia to be only slightly effective, mostly being used as a subsidiary to a standard oncotherapy. Here, we report that iron-dextran delivers iron ion to cancer cells for cancer-selective accumulation of the iron ion, which functions as a thermosensitizer for radiofrequency hyperthermia. Intravenous injection of iron-dextran to tumor-xenografted mice resulted in selective accumulation of iron ion in the targeted cancer cells. The accumulated iron ion in cancer cells dramatically reacted to radiofrequency wave to result in tumor-selective dielectric temperature increment without harming the surrounding normal tissue. The oncotherapeutic effect of was evaluated using tumor-xenografted mice. The overall anticancer efficacy of radiofrequency hyperthermia after injection of iron-dextran as a thermosensitizer in breast cancer-bearing mice was much better than the efficacy of paclitaxel, a standard chemotherapy drug for cancer. Moreover, hyperthermia using iron-dextran as a thermosensitizer completely eradicated cancer in the tumor xenografted mice. This work suggests that iron-dextran is an ideal thermosensitizer for radiofrequency hyperthermia. We believe that the application of iron-dextran as a thermosensitizer would be a major progress in hyperthermia cancer treatments.

Gold Nanoparticles for Photothermal Cancer Therapy.

Gold is a multifunctional material that has been utilized in medicinal applications for centuries because it has been recognized for its bacteriostatic, anticorrosive, and antioxidative properties. Modern medicine makes routine, conventional use of gold and has even developed more advanced applications by taking advantage of its ability to be manufactured at the nanoscale and functionalized because of the presence of thiol and amine groups, allowing for the conjugation of various functional groups such as targeted antibodies or drug products. It has been shown that colloidal gold exhibits localized plasmon surface resonance (LPSR), meaning that gold nanoparticles can absorb light at specific wavelengths, resulting in photoacoustic and photothermal properties, making them potentially useful for hyperthermic cancer treatments and medical imaging applications. Modifying gold nanoparticle shape and size can change their LPSR photochemical activities, thereby also altering their photothermal and photoacoustic properties, allowing for the utilization of different wavelengths of light, such as light in the near-infrared spectrum. By manufacturing gold in a nanoscale format, it is possible to passively distribute the material through the body, where it can localize in tumors (which are characterized by leaky blood vessels) and be safely excreted through the urinary system. In this paper, we give a quick review of the structure, applications, recent advancements, and potential future directions for the utilization of gold nanoparticles in cancer therapeutics.
