Prostaglandin-like substances in Propionibacterium acnes. IV. Effect on isolated human vessels.

The present study demonstrates a powerful vasoconstrictor activity of prostaglandin-like substances (PLS), extracted from P. acnes, on human blood vessels. PLS is about equipotent to PGE2 in its effect on human umbilical vessels, but the contractile response pattern is different. PLS therefore seems to have specific and different physiological characteristics.

Inhibition of platelet aggregation by placental extracts.

Abstract Extracts of human placenta and of human umbilical cord vessels were examined for anti-platelet aggregating activity. Placental extracts progressively inhibited aggregation induced by ADP but had little effect on that produced by adrenaline, collagen or ristocetin. In contrast, umbilical vessel extracts and PGI2 had an almost immediate inhibitory effect on aggregation induced by all four reagents. The effect of placental extracts was shown to be mediated through the degradation of ADP and was mimicked by purified human placental alkaline phosphatase. Since human placental alkaline phosphatase activity is known to be low in pregnancies associated with toxaemia, an alteration in this ADPase activity of the placenta may contribute to the pathogenesis of disseminated intravascular coagulation in eclampsia of pregnancy.

Differential release of prostaglandin E-like and F-like substances by endothelial cells cultured from human umbilical arteries and veins

Endothelial cells, isolated from human umbilical vessels and grown in culture, displayed morphological characteristics similar to those described for endothelial cells in vivo. After 7–10 days in culture, endothelial cells from human umbilical arteries and veins were found to release prostaglandin F-like substances in excess of prostaglandin E-like substances into the media bathing the cells. Quantitative differences in concentration of prostaglandin E-like and F-like substances as well as alterations in the rate of prostaglandin release were determined by the presence of some unidentified component(s) of fetal calf serum. Implications of prostaglandin release by cellular components of human umbilical vessels are discussed.

Effects of prostaglandins E1 and E2 and prostaglandin endoperoxides on the isolated human umbilical artery

Prostaglandin (PG) activity has been found in homogenates of human umbilical vessels (1). Indomethaoin inhibits the PG production and reduces the resting tone of human umbilical artery (HUA) strips (2). In the present study the motor responses of spiral HUA strips in Krebs solution to PGE1 and PGE2 and to two endoperoxide intermediates in the prostaglandin biosynthesis, i.e. prostaglandin G2 and prostaglandin H2 (3), were studied. PGE1 produced relaxation at 0, 1-3 μg/ml but contraction at 10-40 μg/ml. PGE2 (0, 04-20 μg/ml), PGG2 (0,001-0,4 μg/ml) and PGH2(0,001-0,4 μg/ml) produced contractions only within the concentration range used. The time lapse to reach the peak effect was consistently shorter for the endoperoxides than for PGE1 and PGE2. The results support the theory that local synthesis of prostaglandins might be an important mechanism in tone regulation of the human umbilical artery. 1. Karim, S.M.M. Electrical stimulation and drug responses of isolated human umbilical blood vessels

Histochemical studies and electrical stimulation of isolated human umbilical arteries and veins confirm the absence of noradrenergic and cholinergic innervations. The order of potency in producing contractions of the two isolated tissues was 5-hydroxytryptamine (5-HT) > noradrenaline (NA) > acetylcholine (ACH). The maximal contractions to NA and ACH were less than those to 5-HT. These findings were related to the closure of the umbilical blood vessels at birth.

Bradykinin production associated with oxygenation of the fetal lamb.

Previous studies showed that synthetic bradykinin produced in-vitro constriction of the ductus arteriosus of the fetal lamb and guinea pig, constriction of isolated human and lamb umbilical vessels, and pulmonary vasodilatation in the fetal lamb. The present study showed that bradykinin precursor, kininogen, is present in arterial blood of the fetal lamb by 61 days of gestation and that its concentration increases toward term. We studied the bradykinin-generating system in 6 exteriorized fetal lambs. Kininogen concentrations in left atrial blood decreased within 1 to 2 minutes after beginning ventilation with O 2 , and free bradykinin was detected in left atrial blood. Pulmonary arterial blood kininogen concentration was not significantly altered. Kininogen concentration in left atrial blood did not fall in 4 other lambs ventilated with N 2 but did fall after subsequent ventilation with O 2 . To study the effects of increased oxygenation in utero without lung expansion, 6 pregnant ewes were exposed to hyperbaric O 2 at 3.63 atmospheres absolute. Associated with a mean increase in brachial arterial blood Po 2 to 44 mm Hg, a value equivalent to that in the ventilated exteriorized fetus, kininogen concentration fell, and free bradykinin was detected in the brachial arterial blood of 3 of the 6 fetuses. In 4 other exteriorized lambs we measured kininogen concentration in brachial arterial and right ventricular blood at frequent intervals up to 30 minutes after ventilation with O 2 . The concentration in brachial arterial blood fell before that in right ventricular blood. The hypothesis is presented diat bradykinin is produced from kininogen in the lungs of the fetal lamb when oxygenated, and that the maximal rate of production occurs during the first few minutes after expansion of the lungs or exposure of the ewe and fetus to hyperbaric O 2 .

With a modified methylene blue immersion technique, it has been possible to demonstrate neural fibers in cleared whole mounts of human umbilical cords. The nerve fibers run in Wharton's jelly and give off branches to form a plexus in the walls of the umbilical vessels. It is suggested that vasomotor responses in the umbilical vessels are mediated via a neural mechanism.
