Escherichia coli-based synthesis of cadmium sulfide nanoparticles, characterization, antimicrobial and cytotoxicity studies.

The present research describes the synthesis of cadmium sulfide (CdS) nanoparticles from Escherichia coli under the influence of bacterial enzyme sulphate reductase and study on their cytotoxicity for applications in cancer therapy. Escherichia coli cells were used to synthesize CdS nanoparticles under different concentrations of cadmium chloride and sodium sulfide. The morphology of the nanoparticles was analysed using scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDX) was used for elemental analysis of nanoparticles. Fourier-transform infrared spectroscopy analysis (FTIR) was performed to assess the functional groups of the nanoparticles. Crystalline nature of nanoparticles was assessed using powder X-ray diffraction (XRD). Antibacterial studies of CdS nanoparticles were carried out on foodborne pathogens and cytotoxicity studies were carried out on Mus musculus skin melanoma (B16F10) and human epidermoid carcinoma (A431) cell lines. CdS nanoparticle showed more cytotoxic effect on cancer cells compared with standard 5-aminolevulinic acid (5-ALA). The Escherichia coli-synthesized CdS nanoparticles showed highest zone of inhibition in the ratio 4:1 of cadmium chloride and sodium sulfide on all tested bacterial strains. The nanoparticles were also tested for haemolytic activity on RBC cells, which exhibited lower cytotoxicity than sodium dodecyl sulphate which was used as positive control. The cytotoxicity of CdS nanoparticles assessed on A431 cells showed an inhibition of 81.53% at 100μM concentration while the cytotoxicity assessed on B16F10 cells showed an inhibition of 75.71% at 200μM concentration which was much efficient than 5-ALA which showed an inhibition of 31.95% at a concentration against B16F10 cells and 33.45% against A431 cells at a concentration of 1mM. Cadmium sulfide nanoparticles were thus found to be highly toxic on cancer cells compare with standard anticancerous drug 5-ALA.

Multifloroside Suppressing Proliferation and Colony Formation, Inducing S Cell Cycle Arrest, ROS Production, and Increasing MMP in Human Epidermoid Carcinoma Cell Lines A431.

Multifloroside (4), together with 10-hydroxyoleoside 11-methyl ester (1), 10-hydroxyoleoside dimethyl ester (2), and 10-hydroxyligustroside (3), are all secoiridoids, which are naturally occurring compounds that possess a wide range of biological and pharmacological activities. However, the anti-cancer activity of 1–4 has not been evaluated yet. The objective of this work was to study the anti-cancer activities of 1–4 in the human epidermoid carcinoma cell lines A431 and the human non-small cell lung cancer (NSCLC) cell lines A549. The results indicate that 1–4 differ in potency in their ability to inhibit the proliferation of human A431 and A549 cells, and multifloroside (4) display the highest inhibitory activity against A431 cells. The structure-activity relationships suggest that the o-hydroxy-p-hydroxy-phenylethyl group may contribute to the anti-cancer activity against A431 cells. Multifloroside treatment can also inhibit cell colony formation, arrest the cell cycle in the S-phase, increase the levels of reactive-oxygen-species (ROS), and mitochondrial membrane potential (MMP), but it did not significantly induce cell apoptosis at low concentrations. The findings indicated that multifloroside (4) has the tendency to show selective anti-cancer effects in A431 cells, along with suppressing the colony formation, inducing S cell cycle arrest, ROS production, and increasing MMP.

Silibinin induced apoptosis of human epidermal cancer A431 cells by promoting mitochondrial NOS

The antitumor effects of silibinin are of increasing interest, though its mechanism is not yet clear. The goal of this study was to clarify the mechanism of silibinin-induced cell death in the A431 human epidermoid carcinoma cell line. We used a cell viability assay, flow cytometry, nitric oxide (NO) assay, and western blotting to examine relationships between silibinin, NO generation and apoptosis in A431 cells. Silibinin inhibited A431 cell growth in a dose-dependent manner, inducing mitochondrial damage, and apoptosis at a high dose. At the same time, high dose silibinin increased NO levels in A431 cells and the endothelial nitric oxide synthase (eNOS) inhibitor NG-nitro-L-arginine methylester (L-NAME) attenuated silibinin-induced cell growth inhibition. By western blotting, silibinin caused increased eNOS phosphorylation in the mitochondria. The AMP-activated protein kinase inhibitor compound C significantly decreased p-eNOS expression, while blocking eNOS did not affect p-AMPK levels, suggested that AMPK acted upstream of eNOS. This study showed that silibinin increased NO levels in A431 cells by activating the AMPK–eNOS pathway, leading to mitochondrial dysfunction and apoptosis. In this mechanism of action, mitochondrial eNOS played an important role. The results provided new understanding of the functions of intracellular NO.

Abstract 2586: Therapeutic potential of the topical treatment of miR-634 ointment for cutaneous squamous cell carcinoma

Abstract Cutaneous squamous cell carcinoma (CSCC) is the second most common nonmelanoma skin cancers. Surgery represents the gold standard for CSCC treatment, however it is needed to develop the noninvasive and effective treatments as the nonsurgical options for CSCC patients whose conditions are not suitable for or who refuse operation. On the other hand, microRNAs (miRs) are endogenous small non-coding RNAs negatively regulate gene expression by interfering with the translation or stability of target transcripts. Some tumor-suppressive miRs can target multiple cancer-promoting genes concurrently and may be useful as a therapeutic agent for cancer therapy. We have previously found that overexpression of miR-634 activated the mitochondrial apoptotic pathway by directly targeting genes associated with cytoprotective processes, including mitochondrial homeostasis, anti-apoptosis, antioxidant ability, and autophagy, in various cancer cells. The development of drug delivery system (DDS) is critical for the implementation of miR-based therapeutics. While topical pharmacotherapy represents a nonsurgical option to consider in CSCC, we validated the therapeutic potential of topical treatment of miR-634 ointment in mice model of CSCC. In xenograft tumors of A431 cells, a human epidermoid carcinoma cell line, in mice the topical treatment of ointment incorporated double-strand miR-634 mimics significantly reduced tumor growth. Furthermore, the topical treatment of miR-634-ointment also inhibited tumor growth in carcinogen-induced mouse skin papilloma model. Taken together, these findings suggest that topical treatment of miR-634-ointment may be useful as a noninvasive and effective treatments for CSCC. Oxidative Response and Micronucleus Centromere Assay in HEp-2 Cells Exposed to Fungicide Iprodione.

The fungicide agents are a key component in the fruits and vegetables production. The Iprodione residues are one of the pesticide more frequently found in food products. The available data about the cytotoxicity of iprodione and its metabolites are scarce and do not allow characterization of its genotoxic potential and define the risk assessment.The human larynx epidermoid carcinoma cell line (HEp-2) has been shown to be sensitive to the toxic effects of xenobiotics of different origin and have been often used in citotoxicity and genotoxicity studies. The purpose of this paper is to evaluate the induction of genotoxicity and the role of oxidative stress in HEp-2cell line by exposure to the IP. The MTT test for viability resulted in CL50 85.86 (77.05-95.68) μg/mL of Iprodione. On the basis of this result, we proceeded to expose the cells to the sublethal concentrations (below the CL50) during 24 h to analyze the mitotic index and nuclear division index in order to determine the subcytotoxic concentrations of IP which the genotoxicity was evaluated. The subcytotoxic concentrations of 7, 17, and 25 μg/mL IP induced aneugenic effects as micronuclei centromere positive whereas 17 μg/mL was a threshold for centromere negative micronuclei induction in HEp-2 cells. The abnormal mitosis was induced for exposition of Hep-2 cells to the three concentrations. According to the result obtained, citotoxicity and genotoxicity oxidative stress studies were performed in 1.5, 7.0, and 25 μg/mL of IP. The results showed that the GSH intracellular content, the SOD activity and the levels of oxidative damage of the proteins were affected lead to redox imbalance. The decreased in the SOD activity and protein oxidation were in according to the result obtained to genotoxicity, suggesting that different biological targets could be affected.

Facile synthesis of silver nanoparticles using Averrhoa bilimbi L and Plum extracts and investigation on the synergistic bioactivity using in vitro models

AbstractThe bacterial communities (Gram-negative and Gram-positive) form the biofilms which oppose the mode of action of antibiotics and affecting the immune system of the human. These chronic infections related to biofilm are always hard to be cured because of their inherent resistance to both antimicrobial agents and host defense. The present study is devoted to the synthesis of silver nanoparticles (AgNPs) using aqueous extracts ofAverrhoa bilimbileaf and Plum fruit (Prunus bokharensis) and its inhibitory effect onStaphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, andSalmonella typhi. Here, AgNPs) were successfully prepared by green synthesis method obtained by the reduction of silver nitrate into silver ions. The synthesized AgNPs were characterized by UV-Vis Spectroscopy, XRD, FTIR, FESEM-EDAX and DLS analysis. The antimicrobial effects of the two extract were evaluated by the disk diffusion method. The green synthesized AgNPs exhibited good antibacterial activity against both Gram-negative and Gram-positive. They were also analyzed for their significant antibacterial activities against all the test cultures exposed whereasEscherichia coliandSalmonella typhitopped among other test cultures. The anti-proliferative activity of phytochemical mediated synthesis of Ag NPs was investigated for their cytotoxicity in Vero and Human epidermoid larynx carcinoma cell lines (HEp-2) based on their viability using MTT assay. The present study also represents the synergistic bioactivity of silver nanoparticles using in vitro models.

Polymers with distinctive anticancer mechanism that kills MDR cancer cells and inhibits tumor metastasis

Although mortality continues to decline over the past two decades, cancer is still a pervasive healthcare problem worldwide due to the increase in the number of cases, multidrug resistance (MDR) and metastasis. As a consequence of multidrug resistance, cancer treatment must rely on a host of chemotherapeutic agents and chemosensitizers to achieve remission. To overcome these problems, a series of biodegradable triblock copolymers of PEG, guanidinium-functionalized polycarbonate and polylactide (PEG-PGCx-PDLAy) is designed as chemotherapeutic agents. These copolymers self-assemble into micellar nanoparticles, and are highly effective against various cancer cell lines including human breast cancer (BCap37), liver cancer (HepG2), lung cancer (A549) and epidermoid carcinoma (A431) cell lines as well as MDR Bats-72 and Bads-200 cancer cells that were developed from BCap37. Multiple treatments with the polymers at sub-lethal doses do not induce resistance. The polymers kill cancer cells by a non-apoptotic mechanism with significant vacuolization and subsequent membrane disruption. In vivo antitumor efficacy is evaluated in a metastatic 4T1 subcutaneous tumor model. Treatment with stereocomplexes of PEG-PGC43-PLLA19 and PEG-PGC43-PDLA20 at a dose of 20 mg/kg of mouse body weight suppresses tumor growth and inhibits tumor metastasis in vivo. These polymers show promise in the treatment of cancer without the onset of resistance.

MAPK/ERK regulation of P53 in human epidermoid carcinoma cell line A431

Objective: To observe the impact of activation and inhibition of mitogen activated protein kinases (MAPK)/extracellular signalregulated protein kinase (ERK) signaling pathway on the proliferation and apoptosis of cutaneous squamous cell carcinoma (SCC). cells and investigate the interaction mechanism between MAPK/ERK signaling pathway and tumor suppressor gene P53 in SCC. Methods: Human A431 cells were cultured and divided into MAPK/ERK inhibition groups with low-, medium- and highconcentration of inhibitors (PD98059 + DMSO), MAPK/ERK activation groups with low-, medium- and high-concentration of stimuli (IGF + PBS) and blank control group (DMSO). The cell proliferation in vitro was detected by MTT assay, with the cell apoptosis detected by flow cytometry (FCM) and the protein expression of P-ERK and P53 detected by western blot in each group. Results: The A431 cell proliferation was inhibited by different concentrations of PD98059 with a clear concentration - effect and time - effect relationship ( p < .05); and the cell proliferation was promoted by the different concentrations of IGF with a clear concentration - effect and time - effect relationship ( p < .05). The FCM results showed a significant increase in the apoptosis rate of A431 cells which were treated with PD98059, with a clear concentration - effect relationship ( p < .05); while the apoptosis rate was decreased significantly after A431 cells were treated with IGF, also with a concentration - effect relationship ( p < .05). The western blot results showed that the expression of P-ERK protein was decreased but the expression of P53 was increased after A431 cells were treated with PD98059. With the concentration of PD98059 going up, the decrease in P-ERK and the increase in P53 were more significant ( p < .05); while the expression of P-ERK protein was increased but the expression of P53 was decreased after A431 cells were treated with IGF. With the concentration of IGF going up, the increase in P-ERK and the decrease in P53 were more significant ( p < .05). According to Pearson correlation analysis, the expression of P53 was negatively correlated to that of P-ERK ( p < .05). Conclusions: After MAPK/ERK signaling pathway was activated by IGF in A431 cells, the expression of pro-apoptotic factor P53 was decreased with the ability of cell proliferation enhanced and the ability of apoptosis reduced. However, after the inhibition of MAPK/ERK signaling pathway, the expression of pro-apoptotic factor P53 was increased with the ability of cell proliferation reduced and the ability of apoptosis increased.

UV Light Stimulated Activation of Camkii in Human Epidermoid Carcinoma A431 Cells

Summary The Ca2+/calmodulin dependent kinase II (CaMKII) represents a family of serine/threonine kinases with diverse functions, implicated in various signalling pathways in the cell, especially in those controlling cellular proliferation and cellular death. The human epidermoid carcinoma cell line A431 is an established model for the study of the cell cycle and cancer-associated signalling, in which the role of CaMKII is not so well elucidated. Therefore, the present study aimed to investigate the role of CaMKII in the induction of apoptosis in the cell line A431 following ultraviolet (UV) irradiation and to determine the downstream events with a focus on the caspase cascade. For the latter purpose, we employed a method initially developed for the simultaneous determination of the activity of 10 different caspase enzymes. Following exposure to UV light, we observed activation of the activator caspase-8 followed by activation of the effector caspase-3, triggering apoptosis in the epidermoid carcinoma cell line A431. The results supported a pivotal role of caspases and demonstrated that a protein kinase cascade involving caspases-8 and -3 is the main pathway driven in UV-induced apoptosis in the human carcinoma cell line A431, and apoptotic stimuli triggering this pathway passed through activation of the kinase CaMKII.

The internalization kinetics and intracellular spatial distribution of functionalized diatomite nanoparticles in human lung epidermoid carcinoma cell line have been investigated by confocal fluorescence and Raman microscopy. In this context, Raman imaging due to its non‐destructive, chemically selective and label‐free working principle provides evidence that the nanovectors are internalized and co‐localize with lipid environments, suggesting an endocytic internalisation route. Macroalgae Extracts From Antarctica Have Antimicrobial and Anticancer Potential.

Background: Macroalgae are sources of bioactive compounds due to the large number of secondary metabolites they synthesize. The Antarctica region is characterized by extreme weather conditions and abundant aggregations of macroalgae. However, current knowledge on their biodiversity and their potential for bio-prospecting is still fledging. This study evaluates the antimicrobial and cytotoxic activity of different extracts of four macroalgae (Cystosphaera jacquinotii, Iridaea cordata, Himantothallus grandifolius, and Pyropia endiviifolia) from the Antarctic region against cancer and non-cancer cell lines.Methods: The antimicrobial activity of macroalgae was evaluated by the broth microdilution method. Extracts were assessed against Staphylococcus aureus ATCC 19095, Enterococcus faecalis ATCC 4083, Escherichia coli ATCC29214, Pseudomonas aeruginosa ATCC 9027, Candida albicans ATCC 62342, and the clinical isolates from the human oral cavity, namely, C. albicans (3), C. parapsilosis, C. glabrata, C. lipolytica, and C. famata. Cytotoxicity against human epidermoid carcinoma (A-431) and mouse embryonic fibroblast (NIH/3T3) cell lines was evaluated with MTT colorimetric assay.Results: An ethyl acetate extract of H. grandifolius showed noticeable antifungal activity against all fungal strains tested, including fluconazole-resistant samples. Cytotoxicity investigation with a cancer cell line revealed that the ethyl acetate extract of I. cordata was highly cytotoxic against A-431 cancer cell line, increasing the inhibitory ratio to 91.1 and 95.6% after 24 and 48 h exposure, respectively, for a concentration of 500 μg mL−1. Alpha-Tocopheryl Succinate Selectively Affects the Morphology and Motility of Normal and Tumor Epithelial Cells

Alpha-tocopheryl succinate (α-TS) is a vitamin-E derivative with potential antitumor properties. In this study, we have investigated the effect of α-TS on the morphology and motility of the A431 human epidermoid carcinoma cell line and HaCaT immortalized nontumorigenic human keratinocytes. Phase contrast and fluorescent microscopy, in vitro wound healing assay (scratch test), and scanning electron microscopy have been used. In both cell lines, alteration of cell shape, actin-cytoskeleton reorganization, cell-surface smoothing, disappearance of microvillae, and cell-motility inhibition have been shown. These effects are similar in both cell lines, but in the HaCaT cell line they occur after treatment with higher α-TS doses than for A431cells. Thus, we have demonstrated the selectivity of the effect of α-TS on tumor cells derived from human-skin stratified epithelia. The results of this work may be useful for further investigations focused on development of antitumor agents with selective action toward tumor cells.

Internalization kinetics and cytoplasmic localization of functionalized diatomite nanoparticles in cancer cells by Raman imaging.

Porous biosilica nanoparticles obtained from diatomites (DNPs) have been recently demonstrated to be non-toxic nanovectors of therapeutic agents in cancer cells. In this work, the internalization kinetics and intracellular spatial distribution of functionalized DNPs incubated with human lung epidermoid carcinoma cell line (H1355) up to 72 hours are investigated by Raman imaging. The label-free Raman results are compared with confocal fluorescence microscopy and photoluminescence (PL) data. Raman bands specifically assigned to DNPs and cellular components provide evidence that the nanovectors are internalized and co-localize with lipid environments. A considerable DNPs uptake in cells is observed within 6 hours, with equilibrium being achieved after 18 hours. The obtained data show the presence of DNPs up to 72 hours, without damage to cell viability or morphology. The PL measurements performed on DNPs not penetrating the cells at different incubation times are strongly correlated with the results obtained by Raman imaging and confocal microscopy analyses.

Synthesis and cytotoxicity against KB and NCI-H187 cell lines of sporogen AO-1 analogues

20 Analogues of sporogen AO-1 were synthesized by chemical modification at α,β-unsaturated carbonyl, 3-hydroxyl and vinylic methyl groups of sporogen AO-1 precursor, and were evaluated for their cytotoxic activities against human oral epidermoid carcinoma (KB) and human small cell lung (NCI-H187) cancer cell lines. Structure-activity relationship study indicated the importance of α,β-unsaturated carbonyl moiety for both cancer cell lines. Vinylic methyl and R-configuration of 3-hydroxyl group were crucial for cytotoxicity toward KB cells. In contrast, conversion of vinylic methyl and 3-hydroxyl groups to ketone moieties afforded triketone 19 which displayed comparable cytotoxicity against NCI-H187 cells lines to sporogen AO-1, and was more potent than ellipticine, a standard drug. Interestingly, compound 19 was weakly cytotoxic toward Vero cells, whereas sporogen AO-1 showed strong cytotoxicity.

Establishment and characterization of arsenic trioxide resistant KB/ATO cells

Arsenic trioxide (ATO) is used as a chemotherapeutic agent for the treatment of acute promyelocytic leukemia. However, increasing drug resistance is reducing its efficacy. Therefore, a better understanding of ATO resistance mechanism is required. In this study, we established an ATO-resistant human epidermoid carcinoma cell line, KB/ATO, from its parental KB-3-1 cells. In addition to ATO, KB/ATO cells also exhibited cross-resistance to other anticancer drugs such as cisplatin, antimony potassium tartrate, and 6-mercaptopurine. The arsenic accumulation in KB/ATO cells was significantly lower than that in KB-3-1 cells. Further analysis indicated that neither application of P-glycoprotein inhibitor, breast cancer resistant protein (BCRP) inhibitor, or multidrug resistance protein 1 (MRP1) inhibitor could eliminate ATO resistance. We found that the expression level of ABCB6 was increased in KB/ATO cells. In conclusion, ABCB6 could be an important factor for ATO resistance in KB/ATO cells. The ABCB6 level may serve as a predictive biomarker for the effectiveness of ATO therapy.

Comparative Evaluation of Phenolic Profile, Antioxidative and Cytotoxic Activities of Different Geranium Species.

Cranesbill (Geranium species) are used traditionally as tonic, antidiabetic, antidiarrheal, antihemorrhoidal, diuretic and for the treatment of cough, fever, tonsillitis, urticaria, kidney pain and gastrointestinal ailments. The leaves of some Geranium species are consumed in traditional Turkish cousin as salad or added to pastry. In the present study, different extracts from G. psilostemon and G. stepporum of the plants were prepared and their SO, NO and ABTS radical scavenging activites were tested. Also, cytotoxic activities of extracts against KB human epidermoid carcinoma cell line tested by MTT method. While EtOAc extracts showed strong radical scavenging activity against SO, NO and ABTS radicals (IC50 of EtOAc extracts of G. psilostemon 29.4, 98.4, µg/ml for SO and NO respectively and TEAC value 0.371 ± 0.29 µM TE) cytotoxic activity against KB cell line was observed for the water extracts of tested species. In order to evaluate phytochemical content of the species; total phenolic, total flavonoid and total flavonol contents of the extracts were estimated and major compounds of the extracts were determined as gallic acid, methyl gallate and pusilagin using HPLC-DAD system. Taken together, the results showed that gallic acid derivatives are important for the bioactivities of Geranium species, and it may be promising possible sources for future novel antioxidants in food and pharmaceutical formulations.

Understanding the Antifungal Mechanism of Ag@ZnO Core-shell Nanocomposites against Candida krusei.

In the present paper, facile synthesis of Ag@ZnO core-shell nanocomposites is reported where zinc oxide is coated on biogenic silver nanoparticles synthesized using Andrographis paniculata and Aloe vera leaf extract. Structural features of as synthesized nanocomposites are characterized by UV-visible spectroscopy, XRD, and FTIR. Morphology of the above core-shell nanocomposites is investigated by electron microscopy. As synthesized nanocomposite material has shown antimicrobial activity against Candida krusei, which is an opportunistic pathogen known to cause candidemia. The possible mode of activity of the above material has been studied by in-vitro molecular techniques. Our investigations have shown that surface coating of biogenic silver nanoparticles by zinc oxide has increased its antimicrobial efficiency against Candida krusei, while decreasing its toxicity towards A431 human epidermoid carcinoma cell lines.

Evaluation of Mannosidase and Trypsin Enzymes Effects on Biofilm Production of Pseudomonas aeruginosa Isolated from Burn Wound Infections.

Biofilm is an important virulence factor in Pseudomonas aeruginosa and has a substantial role in antibiotic resistance and chronic burn wound infections. New therapeutic agents against P. aeruginosa, degrading biofilms in burn wounds and improving the efficacy of current antimicrobial agents, are required. In this study, the effects of α-mannosidase, β-mannosidase and trypsin enzymes on the degradation of P. aeruginosa biofilms and on the reduction of ceftazidime minimum biofilm eliminating concentrations (MBEC) were evaluated. All tested enzymes, destroyed the biofilms and reduced the ceftazidime MBECs. However, only trypsin had no cytotoxic effect on A-431 human epidermoid carcinoma cell lines. In conclusion, since trypsin had better features than mannosidase enzymes, it can be a promising agent in combatting P. aeruginosa burn wound infections.

Synthesis biological screening and molecular docking studies of some tin (IV) Schiff base adducts

The search for an alternative to platinum anticancer agents is a major motivation for continuing investigations concerning the antitumor properties of other transition metal-based compounds. Keeping this in view, synthesis, antitumor and antimicrobial activity of diorganotin (IV) complexes was studied. A novel series of diorganotin (IV) complexes of the Schiff base ligand derived from 7-methoxy-2-hydroxy-1-naphthaldehyde, 1,2-phenylenediamine, Salicylaldehyde were synthesized. Physical and spectral examination was done through various techniques using elemental analyses, IR, 1H, 13C, 119Sn NMR, and 119mSn Mössbauer techniques respectively. The results obtained are in good agreement with 1:1:1 stoichiometry of Schiff base and 2:1 stoichiometry of the complexes. Octahedral geometry was assigned to all the synthesized complexes within six (6) coordination number around the tin. Antitumor activity was screened against human oral epidermoid carcinoma (KB) cell line. The diethyltin (IV) complex 2 showed the most promising cytotoxic results (IC50=0.35μM) against the cell line which is comparable with cisplatin (IC50=0.37μM). Docking studies revealed that these complexes can bind favorably within cisplatin binding site and the binding energy of complex 2 is more than that of cisplatin. Furthermore, binding of these complexes on human topoisomerase IIα enzyme and revealed that these complexes intercalating within the inter-strand of DNA showing interactions with DNA as well as protein that may results in DNA damage and cell death.

Characterization and in vitro evaluation of nimotuzumab conjugated with cisplatin-loaded liposomes

In this paper, we report the conjugation of the humanized monoclonal antibody nimotuzumab with cisplatin-loaded liposomes and the in vitro evaluation of its affinity for tumor cells. The conjugation procedure was performed through derivatization of nimotuzumab with N-succinimidyl S-acetylthioacetate (SATA) followed by a covalent attachment with maleimide groups at the end of PEG-DSPE chains located at the membrane of pre-formed liposomes. Confocal microscopy was performed to evaluate the immunoliposome affinity for EGFR antigens from human epidermoid carcinoma (A-431) and normal lung (MRC-5) cell lines. Results showed that the procedures implemented in this work do not affect the capability of the nimotuzumab-immunoliposomes to recognize the tumor cells, which overexpress the EGFR antigens.
