Plakevulin A induces apoptosis and suppresses IL-6-induced STAT3 activation in HL60 cells

(+)-Plakevulin A (1), an oxylipin isolated from an Okinawan sponge Plakortis sp. inhibits enzymatic inhibition of DNA polymerases (pols) α and δ and exhibits cytotoxicity against murine leukemia (L1210) and human cervix carcinoma (KB) cell lines. However, the half-maximal inhibitory concentration (IC50) value for cytotoxicity significantly differed from those observed for the enzymatic inhibition of pols α and β, indicating the presence of target protein(s) other than pols. This study demonstrated cytotoxicity against human promyelocytic leukemia (HL60), human cervix epithelioid carcinoma (HeLa), mouse calvaria-derived pre-osteoblast (MC3T3-E1), and human normal lung fibroblast (MRC-5) cell lines. This compound had selectivity to cancer cells over normal ones. Among these cell lines, HL60 exhibited the highest sensitivity to (+)-plakevulin A. (+)-Plakevulin A induced DNA fragmentation and caspase-3 activation in HL60 cells, indicating its role in apoptosis induction. Additionally, hydroxysteroid 17-β dehydrogenase 4 (HSD17B4) was isolated from the HL60 lysate as one of its binding proteins through pull-down experiments using its biotinylated derivative and neutravidin-coated beads. Moreover, (+)-plakevulin A suppressed the activation of interleukin 6 (IL-6)-induced signal transducer and activator of transcription 3 (STAT3). Because the knockdown or inhibition of STAT3 induces apoptosis and HSD17B4 regulates STAT3 activation, (+)-plakevulin A may induce apoptosis in HL60 cell lines by suppressing STAT3 activation, potentially by binding to HSD17B4. The present findings provide valuable information for the mechanism of its action.

Anti-cancer drug-mediated increase in mitochondrial mass limits the application of metabolic viability-based MTT assay in cytotoxicity screening.

The high-throughput metabolic viability-based colorimetric MTT test is commonly employed to screen the cytotoxicity of different chemotherapeutic drugs. The assay assumes a cell density-dependent linear correlation with the MTT spectral absorbance. Therefore, the present study aimed to compare the cytotoxicity assessment between the MTT assay and gold standard cell number enumeration. The cytotoxicity was induced by Cisplatin, Etoposide, and Doxorubicin in human lung epithelial adenocarcinoma cells (A549) and cervix carcinoma (HeLa) cell lines. The mitochondrial mass was estimated, and immunoblotting of succinate dehydrogenase (SDH-A) was performed following drug treatment in both cell lines. Student's t-test paired analysis was employed to calculate the significance of the results, where the value p < 0.05 was considered statistically significant. The drug-induced cytotoxic response estimated by MTT absorbance did not show any significant difference with respect to control, and no correlation was observed with the enumerated cell number in both A549 and HeLa cells. Interestingly, per-cell metabolic viability was found to be increased by 1.18 to 3.26-fold (p < 0.05) following drug treatment. Further, mechanistic investigation revealed a drug concentration-dependent significant increase in mitochondrial mass (1.21 to 4.2-fold) and upregulation of SDH protein (50-70%) as well as enzymatic activity with respect to control in both A549 and Hela cells. The limitation of the MTT assay for drug-induced cytotoxicity assessment is due to increased mitochondrial mass and SDH upregulation in surviving cells, leading to enhanced formazan formation. This leads to a lack of correlation between cell number and MTT spectral absorbance, suggesting that the MTT assay may provide an erroneous conclusion for cytotoxicity assessment.

Integrated High-Throughput Screening and Large-Scale Isobolographic Analysis to Accelerate the Discovery of Radiosensitizers With Greater Selectivity for Cancer Cells

High-throughput screening (HTS) platforms have been widely used to identify candidate anticancer drugs and drug-drug combinations; however, HTS-based identification of new drug-ionizing radiation (IR) combinations has rarely been reported. Herein, we developed an integrated approach including cell-based HTS and computational large-scale isobolographic analysis to accelerate the identification of radiosensitizing compounds acting strongly and more specifically on cancer cells. In a 384-well plate format, 160 compounds likely to interfere with the cell response to radiation were screened on human glioblastoma (U251-MG) and cervix carcinoma (ME-180) cell lines, as well as on normal fibroblasts (CCD-19Lu). After drug exposure, cells were irradiated or not and short-term cell survival was assessed by high-throughput cell microscopy. Computational large-scale dose-response and isobolographic approach were used to identify promising synergistic drugs radiosensitizing cancer cells rather than normal cells. Synergy of a promising compound was confirmed on ME-180 cells by an independent 96-well assay protocol, and finally, by the gold-standard colony forming assay. We retained 4 compounds synergistic at 2 isoeffects in U251-MG and ME-180 cell lines and 11 compounds synergistically effective in only one cancer cell line. Among these 15 promising radiosensitizers, 5 compounds showed limited toxicity combined or not with IR on normal fibroblasts. Overall, this study demonstrated that HTS chemoradiation screening together with large-scale computational analysis is an efficient tool to identify synergistic drug-IR combinations, with concomitant assessment of unwanted toxicity on normal fibroblasts. It sparks expectations to accelerate the discovery of highly desired agents improving the therapeutic index of radiation therapy.

Furanocoumarins from the twigs of Ficus chlamydocarpa (Moraceae)

Two furanocoumarin derivatives, 3-methoxypsoralen (1) and 3,5-dimethoxypsoralen (2), along with nine known compounds, friedelinol (3), 3-oxo-11β-hydroxyoleanan-12-ene (4), lupeol (5), taraxer-3-one (6), a mixture of β-sitosterone (7a) and stigmast-4,22-dien-3-one (7b), ergosterol (8), 9,19-cyclolanost-3-one-24,25-diol (9), oleanan-12-ene-3,11-dione (10), and β-sitosterol 3-O-β-D-glucopyranoside (11) were isolated from the twigs of Ficus chlamydocarpa. Their structures were established by NMR spectroscopic analyses and HRESIMS. The structure of 1 was further confirmed from its single crystal X-ray diffraction. The crude extract, fractions and some isolated compounds were assessed for their preliminary antibacterial activity and cytotoxicity. One of the fractions (FB-B3) exhibited inhibition against the bacterial strain Pseudomonas agarici and induced a remarkable cytotoxic activity toward the human cervix carcinoma cell line KB-3-1 (IC50 0.166 mg/mL), and compounds 1, 6, and 7 showed moderate antibacterial activity against Bacillus subtilis and Micrococcus luteus.

Three new iridoids from the aerial parts of Nepeta teucriifolia Willd. and antiproliferative activities of extracts

The aerial parts of Nepeta teucriifolia Willd. were extracted with the solvents of different polarities. The antiproliferative activities of the extracts were evaluated against rat brain tumor (C6) and human cervix carcinoma (HeLa) cell lines. The phytochemical screening of the extracts was performed with TOF-LC/MS. The CH2Cl2 and EtOAc extracts showed considerable antiproliferative activities against HeLa cells at higher concentration (250 µg mL−1). The CH2Cl2 extract was found more active than the others on both cells. The phytochemical studies of the active extract led to the isolation of three new iridoids, teucriifolian A-C (1–3). The structure elucidations of the new compounds were performed using HPLC-TOF/MS, 1D and 2D NMR techniques. The compounds 1–3 were evaluated in terms of their antiproliferative activities against HeLa and C6 cells, respectively. The results indicated that only 2 had moderate antiproliferative activity against HeLa cells at 250 µg mL−1.

Self-Assembly and Multifaceted Bioactivity of a Silver(I) Quinolinate Coordination Polymer.

Coordination polymers have emerged as a new class of potent biologically active agents due to a variety of important characteristics such as the presence of bioactive metal centers and linkers, low toxicity, stability, tailorable structures, and bioavailability. The research on intermediate metabolites has also been explored with implications toward the development of selective anticancer, antimicrobial, and antiviral therapeutic strategies. In particular, quinolinic acid (H2quin) is a recognized metabolite in kynurenine pathway and potent neurotoxic molecule, which has been selected in this study as a bioactive building block for assembling a new silver(I) coordination polymer, [Ag(Hquin)(μ-PTA)]n·H2O (1). This product has been prepared from silver oxide, H2quin, and 1,3,5-triaza-7-phosphaadamantane (PTA), and fully characterized by standard methods including single-crystal X-ray diffraction. Compound 1 has revealed distinctive bioactive features, namely (i) a remarkable antiviral activity against herpes simplex virus type 1 (HSV-1) and adenovirus 36 (Ad-36), (ii) a significant antibacterial activity against clinically important bacteria (Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa), and (iii) a selective cytotoxicity against HeLa (human cervix carcinoma) cell line. The present work widens a growing family of bioactive coordination polymers with potent antiviral, antibacterial, and antiproliferative activity.

Ficusanolide A and ficusanolide B, two new cinnamic acid derivative stereoisomers and other constituents of the stem barks of Ficus exasperata Vahl. (Moraceae)

Phytochemical investigation of the stem barks of Ficus exasperata Vahl. (Moraceae) led to the isolation of two new cinnamic acid derivatives stereoisomers, named ficusanolide A (1) and ficusanolide B (2) along with twelve known compounds: ficusanol (3), umbelliferone-6-carboxylic acid (4), oxypeucedanin hydrate (5), marmesin (6), decursinol (7), β-amyrin acetate (8), lupeol (9), betulinic acid (10), ursolic acid (11), a mixture of stigmasterol (12) and β-sitosterol (13), sitosteryl-3-O-β-D-glucopyranoside (14); and one hemisynthetic derivative : per acetylated betulinic acid (15). Their structures were established by the means of their physical data (melting point, rotatory power), their spectroscopic data, particularly IR, NMR (1H, 13C, DEPT, COSY, HSQC and HMBC) data, and HR-ESIMS data. Crude extract, compounds 1, 2, 3, 5, 6, 7, 9, 10, 11, as well as the semisynthetic derivative 15 were evaluated for their cytotoxic activity on the human cervix carcinoma cell line KB-3−1 and the human colon cancer cell line HT-29. Ursolic acid 11 showed a moderate activity on both cancer cells tested with IC50s of 50.9 μM and 34.4 μM respectively.

A new phenylpropanoid glucoside from Psorospermum tenuifolium Kotschy (Hypericaceae)

A new phenylpropanoid glycoside named psorospermoside (1) possessing an allose unit was isolated from the Psorospermum tenuifolium Hook f. bark extract together with eleven known compounds. Their structures were elucidated using spectroscopic and spectrometric methods including 1D, 2D-NMR, and ESI-MS, as well as by comparison of their data with those reported in the literature. All the isolated compounds were assessed for their cytotoxicity effect on the human cervix carcinoma cell line KB3-1. Emodin (2) and its congener 2-geranylemodin (3) displayed significant cytotoxicity with IC50 values of 11.4 µM and 19.0µM, respectively. Furthermore, the chemotaxonomic significance of the isolated compounds was also discussed.

Two new triterpenoid fatty acid esters from Schefflera barteri Harms (Araliaceae)§

Two new fatty acid esters of triterpenoids (1–2) together with eleven known compounds (3–13) were obtained after investigation of the CH2Cl2-MeOH (1:1) crude extract from the leaves of Schefflera barteri Harms. All these compounds (1–13) were isolated for the first time from this plant among which compounds 3, 4, 6 and 9–13 were also isolated from the genus Schefflera for the first time. The structures of the isolated compounds were elucidated by analyses of their spectroscopic data (1D and 2D NMR, and MS). The antibacterial and cytotoxic activities of crude extracts, fractions and compounds (1, 2, 5, 6, 8 and 9) were investigated against both Gram-negative and Gram-positive bacteria strains as well as on human cervix carcinoma and colon adenocarcinoma cancer cell lines, respectively. They showed weak to significant activity towards the strains and malignant cells used.

Further Approaches in the Design of Antitumor Agents with Response to Cell Resistance: Looking toward Aza Crown Ether-dtc Complexes.

The dianionic aza crown ether-dtc N,N'-bis(dithiocarbamate)-1,10-diaza-18-crown-6 (L2-) is a versatile ligand capable of yielding binuclear complexes with group 10 elements, also known as Ni-triade, [μ-(κ2-S,-S'-L)M2(PPh3)4]Cl2 (M = Pd (1), Pt (2)), [μ-(κ2-S,-S'-L)M2(PPh3)4](BPh4)2 (M = Pd (3), Pt (4)), and μ-(κ-S,-S'-L)Ni2(PPh3)2Cl2 (5), and has proven to be an excellent option to the design of metal-based drugs able to provide multiple response to cell resistance. Palladium and platinum complexes, 1 and 2, were tested for cytotoxicity in the human cervix carcinoma cell line HeLa-229, the human ovarian carcinoma cell line A2780, and the cisplatin-resistant mutant A2780cis, finding significant activity toward all three cancer cell lines, with low micromolar IC50 values, comparable to cisplatin. Markedly, against the cisplatin resistant cell line A2780cis, compound 2 exhibits better cytotoxic activity than the clinical drug (IC50 = 2.3 ± 0.2 μM for 2 versus 3.6 ± 0.5 μM for cisplatin). Moreover, an enhancement of the antitumor response is achieved when adding an equimolar amount of alkali metal chloride (NaCl or KCl) to the medium, for instance, testing compound 1 against the cisplatin-resistant A2780cis cells, the IC50 decreases from 9.3 ± 0.4 to 7.4 ± 0.3 and 5.4 ± 0.1 μM, respectively, after addition of the salt solution. For the platinum derivative 2, the IC50 improves by ca. 40% reaching 1.3 ± 0.1 μM when potassium chloride is added. Likewise, the resistant factor found for 2 (RF = 1) confirms that this complex circumvents cisplatin-resistance in A2780cis and is improved with the addition of potassium chloride (RF = 0.65). The presence of the aza crown ether moiety as linker in the systems studied herein is a key point since, in addition to allowing and facilitating interaction with alkali metal ions, this unit is flexible enough to adapt to a variety of environments, as confirmed by the X-ray crystal structures described, where different conformations and ways to fold in are found. In order to gain insight into the electronic and structural facts involved in the interaction of complex 2 with the alkali metal ions, a DFT study was performed, and the description of the molecular electrostatic potentials (MEPs) is also presented.

Meleagrin from marine fungus Emericella dentata Nq45: crystal structure and diverse biological activity studies

The crystal structure and unambiguous absolute configuration of meleagrin (1) isolated from fungus Emericella dentata Nq45 is reported herein to first time on the bases of single crystal X-ray diffraction. Together with 1, haenamindole (2), isorugulosuvine (3), secalonic acid D (4), ergosterol (5) and cerebroside A (6) were obtained and their structures were determined by ESI MS and NMR data analysis. Diverse biological activity of meleagrin (1) was investigated. Compound 1 pronounced potent cytotoxicity against the human cervix carcinoma cell line KB-3-1 and its multidrug resistant sub-clone KB-V1 of IC50 3.07 and 6.07 µM, respectively, in comparison with the reference (+) – griseofulvin (IC50 19, 19.5 µM). Based on the antibiofilm activity, compound 1 displayed as well potent activity against Staphylococcus aureus with an MIC of 0.25 mg/mL. Isolation of the producing fungus and taxonomical characterization is stated as well.

Cytotoxicity of hyaluronic acid coated chitosan nanoparticles containing nitric oxide donor against cancer cell lines

The incidence and mortality rates of all types of cancer have a global burden. The lack of specificity of chemotherapy is one of the major problems in cancer treatment and it is related with moderate to severe adverse effects. CD44 is a protein overexpressed in certain types of cancer cells and can be used as an anchor of attachment for nanoparticles decorated with hyaluronic acid (HA). HA is a natural polymer with high affinity for CD44 receptors. In this work, chitosan nanoparticles (CS NPs) were coated with HA to delivery nitric oxide (NO) donor to cancer cells. NO is a small molecule involved in several physiological processes and it has cytotoxicity against cancer cells. HA-coated and uncoated NO-releasing CS NPs were synthesized and characterized by different techniques. Uncoated and HA-coated NO-releasing CS NPs have hydrodynamic diameters of 142.80 ± 2.22 nm and 170.80 ± 0.14 nm, respectively, polydispersity index of 0.282 ± 0.010 and 0.37 ± 0.04, respectively, and a zeta potential values of + 25.20 ± 0.85 mV and + 15.60 ± 0.15 mV, respectively. As expected, the presence of HA layer on the surface of NO-releasing CS NPs increased hydrodynamic size ca. 20%. The encapsulation efficiency of the NO donor into CS NPs was found to be higher than 90%. The kinetic of NO release from uncoated and HA-coated CS NPs has two distinct phases, an initial burst followed by the establishment of a plateau, with a maximum of NO released at 40 mmol·L−1. At this concentration, NO is expected to have cytotoxic effects. The cytotoxicity of uncoated and HA-coated NO releasing CS NPs showed a concentration-dependent toxicity against human prostatic carcinoma (PC-3) and human uterine cervix carcinoma (HeLa) cell lines. Taken all together, uncoated and HA-coated NO-releasing CS NPs might find important biomedical applications, including cancer in treatment.

Synthesis, in vitro cytotoxicity activity against the human cervix carcinoma cell line and in silico computational predictions of new 4-arylamino-3-nitrocoumarin analogues

A new series of 4-arylamino-3-nitrocoumarin analogues (4–18) have been synthesized and characterized by sophisticated spectroscopic techniques (1H NMR, 13C NMR) and mass spectrometry. All the new synthesized compounds were evaluated for their in vitro cytotoxic activity against the human cervix carcinoma cell line (KB-3-1) using resazurin assay with (+)-griseofulvin as the positive control (IC50 = 19 μM). Among them, thiazolidinylidene derivative 17a that bearing malononitrile unit displayed the best cytotoxic potency with IC50 value of 21 μM. Also, in silico docking simulation studies were conducted on human DNA topoisomerase 1 (Top1) (PDB: 1T8I) to explore and interpret the interaction pattern between the selected compounds and target enzyme as well confirm the acquired cytotoxicity results. In addition to the above, in silico predictions of physicochemical properties, ADME (absorption, distribution, metabolism and excretion) parameters, oral toxicity and indication of toxicity targets were implemented for some title compounds.

Overcoming radioresistance in WiDr cells with heavy ion irradiation and radiosensitization by 2-deoxyglucose with photon irradiation

Background and purposeRadiosensitizers and heavy ion irradiation could improve therapy for female patients with malignant tumors located in the pelvic region through dose reduction. Aim of the study was to investigate the radiosensitizing potential of 2-deoxy-d-glucose (2-DG) in combination with carbon ion-irradiation (12C) in representative cell lines of cancer in the female pelvic region. Materials and methodsThe human cervix carcinoma cell line CaSki and the colorectal carcinoma cell line WiDr were used. 2-DG was employed in two different settings, pretreatment and treatment simultaneous to irradiation. Clonogenic survival, α and β values for application of the linear quadratic model and relative biological effectiveness (RBE) were determined. ANOVA tests were used for statistical group comparison. Isobolograms were generated for curve comparisons. ResultsThe comparison of monotherapy with 12C versus photons yielded RBE values of 2.4 for CaSki and 3.5 for WiDr along with a significant increase of α values in the 12C setting. 2-DG monotherapy reduced the colony formation of both cell lines. Radiosensitization was found in WiDr for the combination of photon irradiation with synchronous application of 2-DG. The same setup for 12C showed no radiosensitization, but rather an additive effect. In all settings with CaSki, the combination of irradiation and 2-DG exhibited additive properties. ConclusionThe combination of 2-DG and photon therapy, as well as irradiation with carbon ions can overcome radioresistance of tumor cells such as WiDr.

Coumamarin: a first coumarinyl calcium complex isolated from nature.

The first calcium complex from nature, Coumamarin (1), 7-hydroxy-3-methoxy-2-oxo-2H-chromene-6-carboxylate Ca(II) complex, was isolated from Aspergillus sydowii ASTI, together with diorcinol (2), violaceol I (3), hydroxysydonic acid (4), cyclo (Trp-Phe), kojic acid, ergosterol, and uracil. The producing strain was isolated from marine water sample collected from Tiran Island, Red Sea, Egypt. Structure 1 was assigned by intensive 1D, 2D NMR, HR-ESIMS, and X-ray crystallography as well. Coumamarin is potentially active against certain tested bacteria and yeasts, while showing no cytotoxic activity against human cervix carcinoma cell line (KB-3-1). Taxonomically, the fungus was identified by phylogenetic analysis of its 18S rRNA gene sequence.

New naturally occurring phenolic derivatives from marine Nocardiopsis sp. AS23C: Structural elucidation and in silico computational studies

AbstractIn our persistent research for new bioactive secondary metabolites from marine actinomycetes, Nocardiopsis sp. AS23C strain was isolated from the marine Alga Sargassum arnaudianum collected in the Red Sea at Hurghada coast, Egypt. A large‐scale fermentation of the strain on M2 medium containing 50% sea water, followed by fractionation and purification of the obtained extract afforded a new phenolic acid derivative, 4‐amino‐6‐methylsalicylic acid (1a), together with 5‐methylresorcinol (2) and linoleic acid (3). Their molecular structures were proved through the sophisticated spectroscopic analyses including exhaustive 1D and 2D NMR (1 and 2‐Dimensional Nuclear Magnetic Resonance) as well mass spectroscopy techniques. The extract exhibited antibacterial activity against the Gram‐positive Staphylococcus aureus, Bacillus subtilis ATCC6051, and Streptomyces viridochromogenes Tü 57. An in vitro cytotoxicity assaying of 1a and 2 against the human cervix carcinoma cell line (KB‐3‐1), pronounced no obvious activity. Additionally, in silico predictions of physicochemical properties, ADME (absorption, distribution, metabolism and excretion) parameters, acute oral toxicity and indication of toxicity targets were performed for 1a and 2 in comparison with the antioxidant agent orsellinic acid.

Isoshamixanthone: a new pyrano xanthone from endophytic Aspergillus sp. ASCLA and absolute configuration of epiisoshamixanthone

Isoshamixanthone (1), a new stereoisomeric pyrano xanthone together with the previously known fungal metabolites, epiisoshamixanthone (2), sterigmatocystin (3), arugosin C (4), norlichexanthone (5), diorcinol (6), ergosterol and methyllinoleate, were obtained from the endophytic fungal strain Aspergillus sp. ASCLA isolated from leaf tissues of the medicinal plant Callistemon subulatus. The chemical structure of the new xanthone (1) was elucidated by extensive 1D, 2D NMR, and ESI HR mass measurements, and by comparison with literature data. The constitutions and absolute configurations of 1 and epiisoshamixanthone (2) were additionally confirmed by X-ray crystallography. Compounds 1,2 were evaluated for their potential anticancer activity using the human cervix carcinoma cell line (KB-3-1). The antimicrobial activities of the fungal extract and compounds 1,2 were studied using a panel of pathogenic microorganisms as well.

Diverse polyketides and alkaloids from Penicillium sp. KHMM: structural elucidation, biological and molecular docking studies.

As a continuation of our earlier research concerning the investigation of microbial bioactive secondary metabolites from the terrestrial Penicillium sp.KH Link 1809 isolate KHMM, the fungus was re-cultivated on a large scale to explore its bioactive compounds intensively. Fifteen compounds, including seven alkaloids (1-7), one sesquiterpene (8), an acetylenic system (9), two sterols, and sphengolipid, were identified. Their structures were established on the bases of extensive one- and two-dimensional nuclear magnetic resonance and mass measurements, and by comparison with literature data. The antimicrobial activity of the fungal extract and the corresponding compounds were studied using a panel of pathogenic microorganisms, and their in vitro cytotoxicity against the human cervix carcinoma cell line (KB-3-1) was reported as well. The molecular docking of the isolated compounds showed promising affinities for the alkaloidal compounds 4-6 towards α, β tubulins.

Anti-proliferative activity of ethyl acetate extracts of Tamarix gallica L. grown at different climatic conditions in Algeria

Abstract The aerial parts of T. gallica collected from three different locations (arid, humid and semi-arid) were extracted using ethyl acetate. The crude extracts were subjected to phenolic appraisal and antiproliferative activity using ELISA and xCELLigence assays. The total phenolic and flavonoids were evaluated using appropriate techniques to give a yield of total phenolics ranging between 238.46 and 348.56 mg GAE (Gallic acid equivalent)/g dry weight extract. The flavonoids yield was found to vary from 36.6 to 103.14 mg QE (quercetin equivalent)/g dry weight extract. Moreover, the extracts were tested against rat brain tumor (C6) and human cervix carcinoma (HeLa) cell lines and displayed important differences in activity. These disparities highlighted the effect of climatic factors as quality determinants of secondary metabolites and therefore as a key control of the biological therapeutic effect.

Evaluation of the cytotoxic and genotoxic effects of mycotoxin fusaric acid

Fusaric acid (FA) is produced by several Fusarium species and is commonly found in grains. This investigation was performed to evaluate the cytotoxic and genotoxic effects of FA either in human cervix carcinoma (HeLa) cell line using 3-(4,5-dimethylthiazolyl-2)-2,5 diphenyltetrazolium bromide (MTT) assay and in human lymphocytes using chromosome aberrations (CAs), sister chromatid exchanges (SCEs), micronuclei (MN) as well as comet assay in vitro. The cells were treated with 0.78, 1.56, 3.125, 6.25, 12.50, 25, 50, 100, 200, and 400 µg/mL concentrations of FA. It has potent cytotoxic effect on HeLa cell line measured by MTT assay especially at higher concentrations (200, 400 µg/mL). The half of inhibitory concentration (IC50) evidenced by FA in the HeLa cells was 200 μg/mL at 24 h and between 200 and 400 μg/mL at 48 h. It was also observed that FA produced a significant decrease in mitotic index (MI) at 12.50 µg/mL compared to solvent control. Furthermore, it indicated a cytotoxic effect at the concentrations ranging from 25 to 400 μg/mL in human lymphocytes. The results of this research point out that being exposed to FA at high concentrations show cytotoxicity. Besides FA induced comet tail intensity at 3.125, 6.25, and 12.50 µg/mL concentrations in isolated human lymphocytes. On the other hand, no genotoxic effects were seen in human lymphocytes in vitro using CA, SCE and MN assays.
