Spike synchrony as a measure of Gestalt structure

The function of spike synchrony is debatable: some researchers view it as a mechanism for binding perceptual features, others – as a byproduct of brain activity. We argue for an alternative computational role: synchrony can estimate the prior probability of incoming stimuli. In V1, this can be achieved by comparing input with previously acquired visual experience, which is encoded in plastic horizontal intracortical connections. V1 connectivity structure can encode the acquired visual experience in the form of its aggregate statistics. Since the aggregate statistics of natural images tend to follow the Gestalt principles, we can assume that V1 is more often exposed to Gestalt-like stimuli, and this is manifested in its connectivity structure. At the same time, the connectivity structure has an impact on spike synchrony in V1. We used a spiking model with V1-like connectivity to demonstrate that spike synchrony reflects the Gestalt structure of the stimulus. We conducted simulation experiments with three Gestalt laws: proximity, similarity, and continuity, and found substantial differences in firing synchrony for stimuli with varying degrees of Gestalt-likeness. This allows us to conclude that spike synchrony indeed reflects the Gestalt structure of the stimulus, which can be interpreted as a mechanism for prior probability estimation.

Brightness induction by contextual influences in V1: a neurodynamical account

Brightness induction is the modulation of the perceived intensity of an area by the luminance of surrounding areas and reveals fundamental properties of neural organization in the visual system. Several phenomenological models have been proposed that successfully account for psychophysical data (Pessoa et al. 1995, Blakeslee and McCourt 2004, Barkan et al. 2008, Otazu et al. 2008). Neurophysiological evidence suggests that brightness information is explicitly represented in V1 and neuronal response modulations have been observed following luminance changes outside their receptive fields (Rossi and Paradiso, 1999). In this work we investigate possible neural mechanisms that offer a plausible explanation for such effects. To this end, we consider the model by Z.Li (1999) which is based on biological data and focuses on the part of V1 responsible for contextual influences, namely, layer 2–3 pyramidal cells, interneurons, and horizontal intracortical connections. This model has proven to account for phenomena such as contour detection and preattentive segmentation, which share with brightness induction the relevant effect of contextual influences. In our model, the input to the network is derived from a complete multiscale and multiorientation wavelet decomposition which makes it possible to recover an image reflecting the perceived intensity. The proposed model successfully accounts for well known pyschophysical effects (among them: the White's and modified White's effects, the Todorović, Chevreul, achromatic ring patterns, and grating induction effects). Our work suggests that intra-cortical interactions in the primary visual cortex could partially explain perceptual brightness induction effects and reveals how a common general architecture may account for several different fundamental processes emerging early in the visual pathway. Diverse excitatory and inhibitory synaptic plasticity outcomes in complex horizontal circuits near a functional border of adult neocortex

The primary somatosensory cortex (SI) is topographically organized into a map of the body. This organization is dynamic, undergoing experience-dependent modifications throughout life. It has been hypothesized that excitatory and inhibitory synaptic plasticity of horizontal intracortical connections contributes to functional reorganization. However, very little is known about synaptic plasticity of these connections; particularly the characteristics of inhibitory synaptic plasticity, its relationship to excitatory synaptic plasticity, and their relationship to the functional organization of the cortex. To investigate this, we located the border between the forepaw and lower jaw representation of SI in vivo, and used whole cell-patch electrophysiology to record post-synaptic excitatory and inhibitory currents in complex horizontal connections in vitro. Connections that remained within the representation (continuous) and those that crossed from one representation to another (discontinuous) were stimulated differentially, allowing us to examine differences associated with the border. To induce synaptic plasticity, tetanic stimulation was applied to either continuous or discontinuous pathways. Tetanic stimulation induced diverse forms of excitatory and inhibitory synaptic plasticity, with LTP dominating for excitation and LTD dominating for inhibition. The border did not restrict plasticity in either case. In contrast, tetanization elicited LTP of monosynaptic inhibitory responses in continuous, but not discontinuous connections. These results demonstrate that continuous and discontinuous pathways are capable of diverse synaptic plasticity responses that are differentially inducible. Furthermore, continuous connections can undergo monosynaptic inhibitory LTP, independent of excitatory drive onto interneurons. Thus, coordinated excitatory and inhibitory synaptic plasticity of horizontal connections are capable of contributing to functional reorganization.

Metaplasticity of horizontal connections in the vicinity of focal laser lesions in rat visual cortex

Focal cortical injuries are accompanied by a reorganization of the adjacent neuronal networks. An increased synaptic plasticity has been suggested to mediate, at least in part, this functional reorganization. Previous studies showed an increased long-term potentiation (LTP) at synapses formed by ascending fibres projecting onto layers 2/3 pyramidal cells following lesions in rat visual cortex. This could be important to establish new functional connections within a vertical cortical column. Importantly, horizontal intracortical connections constitute an optimal substrate to mediate the functional reorganization across different cortical columns. However, so far little is known about their potential implication in the functional rewiring post-lesion. Here, we investigated possible alterations of synaptic plasticity of horizontal connections in layers 2/3 in an 'ex vivo-in vitro' model of focal laser lesion in rat visual cortex. LTP at these synapses was found to be enhanced post-lesion, whereas long-term depression (LTD) was impaired, revealing a metaplastic shift toward strengthening of these synapses. Furthermore, we disclosed a prolonged decay-time constant of NMDAR-dependent currents, which can contribute to the enhanced LTP. Taken together these data revealed that a laser lesion-induced focal damage of the visual cortex is accompanied by a facilitated potentiation of horizontal synaptic connections in the vicinity of the focal injury. This specific strengthening of synaptic plasticity at horizontal connections in layers 2/3 might be one important cellular mechanism to compensate focal injury-mediated dysfunction in the cerebral cortex.

Severe and extensive neonatal hearing loss in cats results in auditory cortex plasticity that differentiates into two regions

We examined the response characteristics of primary auditory cortex (A1) neurons in adult cats partially but extensively deafened by ototoxic drugs 2-8 days after birth. The damage evoked extensive A1 topographic map reorganization as also found by others, but a novel finding was that in the majority of cats with low-frequency edges to the cochlear lesion, the area of reorganization segregated into two areas expressing the same novel frequency inputs but differentiated by neuronal sensitivity and responsiveness. Immediately adjacent to normal A1 is an approximately 1.2-mm-wide area of reorganization in which sensitivity and responsiveness to sound are similar to that in normal A1 in the same animals and in unlesioned adult animals. Extending further into deprived A1 is a more extensive area of reorganization where neurons have poorer sensitivity and responsiveness to new inputs. These two areas did not differ in response-area bandwidth and response latency. We interpret these novel changes as the cortical consequences of severe receptor organ lesions extending to low-frequency cochlear regions. We speculate that the two areas of A1 reorganization may reflect differences in the transcortical spatial distribution of thalamo-cortical and horizontal intracortical connections. Qualitatively similar changes in response properties have been seen after retinal lesions producing large areas of visual cortical reorganization, suggesting they might be a general consequence of receptor lesions that deprive large regions of cortex of normal input. These effects may have perceptual implications for the use of cochlear implants in patients with residual low-frequency hearing.

Modular Organization of Finger Movements by the Human Central Nervous System

The motor system may generate automated movements, such as walking, by combining modular spinal motor synergies. However, it remains unknown whether a modular neuronal architecture is sufficient to generate the unique flexibility of human finger movements, which rely on cortical structures. Here we show that finger movements evoked by transcranial magnetic stimulation (TMS) of the primary motor cortex reproduced distinctive features of the spatial representation of voluntary movements as identified in previous neuroimaging studies, consistent with naturalistic activation of neuronal elements. Principal component analysis revealed that the dimensionality of TMS-evoked movements was low. Principal components extracted from TMS-induced finger movements resembled those derived from end-postures of voluntary movements performed to grasp imagined objects, and a small subset of them was sufficient to reconstruct these movements with remarkable fidelity. The motor system may coordinate even the most dexterous movements by using a modular architecture involving cortical components.

The role of horizontal intracortical connections in "long-range" spatial interactions

Previous studies have used a conservative definition of CRF size, the minimum response field (MRF). But MRF measurement misses parts of the CRF that are too insensitive to generate spikes when stimulated alone. We have measured the size of the CRF in macaque V1 neurons using a grating summation technique. On average the MRF underestimates the area of the CRF by a factor of 4 at high contrast. At low contrast, the suppressive surround is weakened and the area of summation increases by an additional factor of 6. Using published visuotopic maps, we projected our measured CRFs onto the cortical surface, and found that the majority have radii that correspond to horizontal cortical distances of 2-6 mm.

Nature and interaction of signals from the receptive field center and surround in macaque V1 neurons.

Information is integrated across the visual field to transform local features into a global percept. We now know that V1 neurons provide more spatial integration than originally thought due to the existence of their nonclassical inhibitory surrounds. To understand spatial integration in the visual cortex, we have studied the nature and extent of center and surround influences on neuronal response. We used drifting sinusoidal gratings in circular and annular apertures to estimate the sizes of the receptive field's excitatory center and suppressive surround. We used combinations of stimuli inside and outside the receptive field to explore the nature of the surround influence on the receptive field center as a function of the relative and absolute contrast of stimuli in the two regions. We conclude that the interaction is best explained as a divisive modulation of response gain by signals from the surround. We then develop a receptive field model based on the ratio of signals from Gaussian-shaped center and surround mechanisms. We show that this model can account well for the variations in receptive field size with contrast that we and others have observed and for variations in size with the state of contrast adaptation. The model achieves this success by simple variations in the relative gain of the two component mechanisms of the receptive field. This model thus offers a parsimonious explanation of a variety of phenomena involving changes in apparent receptive field size and accounts for these phenomena purely in terms of two receptive field mechanisms that do not themselves change in size. We used the extent of the center mechanism in our model as an indicator of the spatial extent of the central excitatory portion of the receptive field. We compared the extent of the center to measurements of horizontal connections within V1 and determined that horizontal intracortical connections are well matched in extent to the receptive field center mechanism. Input to the suppressive surround may come in part from feedback signals from higher areas.

Strengthening of horizontal cortical connections following skill learning.

Learning a new motor skill requires an alteration in the spatiotemporal pattern of muscle activation. Motor areas of cerebral neocortex are thought to be involved in this type of learning, possibly by functional reorganization of cortical connections. Here we show that skill learning is accompanied by changes in the strength of connections within adult rat primary motor cortex (M1). Rats were trained for three or five days in a skilled reaching task with one forelimb, after which slices of motor cortex were examined to determine the effect of training on the strength of horizontal intracortical connections in layer II/III. The amplitude of field potentials in the forelimb region contralateral to the trained limb was significantly increased relative to the opposite 'untrained' hemisphere. No differences were seen in the hindlimb region. Moreover, the amount of long-term potentiation (LTP) that could be induced in trained M1 was less than in controls, suggesting that the effect of training was at least partly due to LTP-like mechanisms. These data represent the first direct evidence that plasticity of intracortical connections is associated with learning a new motor skill.

A neural model of contour integration in the primary visual cortex.

Experimental observations suggest that contour integration may take place in V1. However, there has yet to be a model of contour integration that uses only known V1 elements, operations, and connection patterns. This article introduces such a model, using orientation selective cells, local cortical circuits, and horizontal intracortical connections. The model is composed of recurrently connected excitatory neurons and inhibitory interneurons, receiving visual input via oriented receptive fields resembling those found in primary visual cortex. Intracortical interactions modify initial activity patterns from input, selectively amplifying the activities of edges that form smooth contours in the image. The neural activities produced by such interactions are oscillatory and edge segments within a contour oscillate in synchrony. It is shown analytically and empirically that the extent of contour enhancement and neural synchrony increases with the smoothness, length, and closure of contours, as observed in experiments on some of these phenomena. In addition, the model incorporates a feedback mechanism that allows higher visual centers selectively to enhance or suppress sensitivities to given contours, effectively segmenting one from another. The model makes the testable prediction that the horizontal cortical connections are more likely to target excitatory (or inhibitory) cells when the two linked cells have their preferred orientation aligned with (or orthogonal to) their relative receptive field center displacements.

Extraction of perceptually salient contours by striate cortical networks.

We present a cortical-based model for computing the perceptual salience of contours embedded in noisy images. It has been suggested that horizontal intra-cortical connections in primary visual cortex may modulate contrast detection thresholds and pre-attentive "pop-out". In our model, horizontal connections mediate context-dependent facilitatory and inhibitory interactions among oriented cells. Strongly facilitated cells undergo temporal synchronization; and perceptual salience is determined by the level of synchronized activity. The model accounts for a range of reported psychophysical and physiological effects of contour salience. In particular, the model proposes that intrinsic properties of synchronization account for the increased salience of smooth, closed contours. Application of the model to real images is demonstrated.

Optically imaged maps of orientation preference in primary visual cortex of cats and ferrets

Feature maps in the cerebral cortex constitute orderly representations of response features created within the cortex; an example is the mapping of orientation-selective neurons in visual cortex. We have compared the properties of orientation maps in area 17 of cats and ferrets, obtained by optical imaging of intrinsic signals. Orientation maps in both species contain a quasi-periodic distribution of iso-orientation domains that are organized into a lattice of pinwheels. However, the spatial density of orientation domains and of pinwheels in ferret area 17 is nearly twice that in cat area 17. The ferret map also contains more discontinuities, or fractures, where orientation changes abruptly. The size of orientation domains scales with interdomain spacing, so that the ratio of the two is approximately the same in both species. Consistent with this finding, the orientation tuning width of individual pixels is similar in the two. The magnitude of orientation preference, however, is much lower in ferret compared to cat. The greater incidence of fractures in ferret appears to be due to proportionately greater overlap between domains of different orientations, particularly along fracture lines that link pinwheel centers. We hypothesize that a key determinant of orientation maps, the relationship between orientation domain size and spacing, expresses an anatomical link between sizes of thalamocortical arbors and horizontal intracortical connections in area 17.

Hand/Face Border as a Limiting Boundary in the Body Representation in Monkey Somatosensory Cortex

Horizontal intracortical connections may form one substrate for representational plasticity in somatosensory cortex. Electrophysiological mapping demonstrated the finer details of the representations of the hand, lower jaw, neck, and face in area 3b of normal macaque monkeys. Injections of two fluorescent tracers then defined the extent to which horizontal connections crossed from the face into the hand representations and vice versa in area 3b. Connections are widely distributed within cortical representations of skin areas innervated by cervical nerves or by the trigeminal nerve but do not cross a border defined by the anterior limit of the representation of skin innervated by the second cervical nerve. This border separates the representation of the muzzle, innervated only by the mandibular nerve, and the representation of the lower jaw and neck region, innervated by the second and third cervical nerves but overlapped by the mandibular nerve. Thus, the muzzle representation lacks connections with the hand and with the lower jaw and neck representations, but the representations of the hand and of the lower jaw and neck are strongly interconnected. Overlap of the hand and of the lower jaw and neck representations and of their horizontal intracortical connections may form one basis for expansions of the lower jaw representation into that of the hand when peripheral input from the hand is lost. Lack of connections with the rest of the face representation may limit this spread.

Control of spatiotemporal coherence of a thalamic oscillation by corticothalamic feedback.

The mammalian thalamus is the gateway to the cortex for most sensory modalities. Nearly all thalamic nuclei also receive massive feedback projections from the cortical region to which they project. In this study, the spatiotemporal properties of synchronized thalamic spindle oscillations (7 to 14 hertz) were investigated in barbiturate-anesthetized cats, before and after removal of the cortex. After complete ipsilateral decortication, the long-range synchronization of thalamic spindles in the intact cortex hemisphere changed into disorganized patterns with low spatiotemporal coherence. Local thalamic synchrony was still present, as demonstrated by dual intracellular recordings from nearby neurons. In the cortex, synchrony was insensitive to the disruption of horizontal intracortical connections. These results indicate that the global coherence of thalamic oscillations is determined by corticothalamic projections.

Synaptic integration fields and associative plasticity of visual cortical cells in vivo

Two major constraints in connectivity decide the spatial extent of visual cortical receptive fields, both during development and adult functioning: 1) feedforward input, extrinsic to visual cortex, is organized in an orderly projection to form a point-to-point mapping of the retina onto the cortical mantle and constitutes the core of the minimal discharge field (MDF) after amplification by local intracortical circuits; and 2) a second type of connectivity consists of long distance horizontal intracortical connections which are thought to favor the binding of local visual operations occurring simultaneously in different parts of the visual field. We review here possible factors, intrinsic to the considered cortical cell, that may control the effectiveness of post-synaptic integration. Their expression during sensory recognition could depend on the spatio-temporal distribution of active inputs onto the target cell dendrite and on their interplay with non-linearities of the membrane properties. On the basis of intracellular recordings in cat area 17, we have observed that peripheral responses (excitatory and inhibitory) could be boosted by coincident postsynaptic depolarization. This effect is lost if the response and the postsynaptic depolarization are mismatched by 1000 ms, suggesting that temporal correlation of central and peripheral responses could regulate in a non-linear manner the gain of center-surround interactions. This temporal selectivity is compatible with up-regulation of composite potentials by the transient voltage-dependent activation of slowly inactivating conductances in visual cortical neurons. A direct consequence of these different considerations is that the receptive field (RF) of neurons in visual pathways should not be considered as a static hardwired window probing the outer environment, but as an active filter which may continuously adapt and be updated as a function of global context and past experience.

The patterns and synaptic properties of horizontal intracortical connections in the rat motor cortex

1. The laminar distribution of synaptic activity in the primary motor cortex, elicited by stimulation of intracortical, horizontal afferents, was studied in young (12-17 days old) and adult rats using the in vitro brain slice preparation. Connectivity patterns were deduced from current-source density (CSD) analyses of field potential depth profiles and were confirmed by anatomic data of retrograde cell labeling after focal injections of a fluorescent tracer. 2. According to the CSD distributions, horizontal axons in layer II/III provide strong monosynaptic input to dendrites of layer II and III pyramidal cells in a distant column, and weaker monosynaptic input to layer V and VI cells by synapsing on dendritic fields at the border of layer III and V and in deep layer V. When these pathways are activated, layer II/III cells may relay excitatory activity to upper and deep layer V, as well as to other cells in layer II/III of the same column. Axons arising from layer V provide monosynaptic input to pyramidal cells in all layers of neighboring columns, by synapsing in two dendritic fields: one in the superficial layers and the other in middle layer V. Activation of these pathways may generate a disynaptic intracolumnar input from layer II/III cells to middle layer V, as well as to other cells in layer II/III. Similar patterns of synaptic activity were elicited by stimulation from 0.45 to 2 mm distal to the recorded column. There were no apparent differences between young and adult rats in the connectivity patterns revealed by the CSD analyses. 3. Tracer injections in layer III resulted in retrograde labeling of cells in layers II/III and V, at distances > 2 mm from the injection site, whereas injections in layer V resulted in retrograde labeling of cells at long distances in layer V and to a lesser extent in layer II/III. These findings indicate that neurons in layer V project, via horizontal axon collaterals, for long distances within layers III and V, whereas the horizontal axon collaterals of layer III cells are restricted, for the most part, to the superficial layers. 4. Suppression of inhibitory activity by bath application of the gamma-aminobutyric acid-A (GABAA) receptor antagonist bicuculline methiodide (BMI) did not alter the pattern of the CSD distributions. All synaptic currents present in the control medium were enhanced by application of BMI, although the effect was more pronounced on the polysynaptic components.(ABSTRACT TRUNCATED AT 400 WORDS)
