Toxicological assessment of bromoxynil and 2-methyl-4-chlorophenoxyacetic acid herbicide in combination on Cirrhinus mrigala using multiple biomarker approach

The widespread application of herbicides raises concerns about their impact on non-target aquatic organisms. This study aimed to evaluate the toxicity of a commercially available herbicide formulation containing Bromoxynil+MCPA (2-Methyl-4-chlorophenoxyacetic acid) on Cirrhinus mrigala (economically significant fish). A total of 210 juvenile fish were subjected to a triplicate experimental setup, with 70 fish allocated to each replicate, exposed to seven different concentrations of herbicide: 0 mg/L, 0.133 mg/L, 0.266 mg/L, 0.4 mg/L, 0.5 mg/L, 0.66 mg/L, and 0.8 mg/L, respectively, for a duration of 96 h. The median lethal concentration (LC50) was determined to be 0.4 mg/L. Significant hematological alterations were observed, including decreases in RBC counts, hemoglobin, hematocrit, and lymphocyte counts, along with an increase in erythrocyte indices. Biochemical analysis revealed elevated levels of neutrophils, WBCs, bilirubin, urea, creatinine, ALT, AST, ALP, and glucose in treated groups. Morphological abnormalities in erythrocytes and histopathological changes in gills, liver, and kidneys were noted. Pathological alterations in gills, liver and kidneys including epithelial cell uplifting, lamellar fusion, hepatolysis, and renal tubule degeneration were observed. Oxidative stress biomarkers such as TBARS (Thiobarbituric Acid Reactive Substance), ROS (Reactive Oxygen Species), and POD (Peroxides) activity increased, while antioxidant enzymatic activities decreased as toxicant doses increased from low to high concentrations. The study reveals that Bromoxynil+MCPA significantly disrupts physiological and hematobiochemical parameters in Cirrhinus mrigala, which highlights the substantial aquatic risks. In conclusion, the herbicide formulation induced significant alterations in various fish biomarkers, emphasizing their pivotal role in assessing the environmental impact of toxicity. This multi-biomarker approach offers valuable insights regarding the toxicological effects, thereby contributing substantially to the comprehensive evaluation of environmental hazards.

Oxidative stress and histopathological effects by microplastic beads, in the crayfish Procambarus clarkii, and fiddler crab Leptuca pugilator

The present study was aimed at evaluating the in vivo effects of microplastics (MP), in terms of oxidative stress and histopathological effects, in two crustacean species: Procambarus clarkii and Leptuca pugilator. In addition, MP accumulation in the hepatopancreas (HP) of both species was also determined. Adults of both crayfish and crabs were exposed for one month to fluorescent polystyrene beads (size: 1 μm) at nominal concentrations of 1000 or 5000 particles/mL. During the exposure, animals were maintained under controlled feeding, aeration, temperature, and photoperiod conditions. At the end of the exposure, HP and hemolymph (HL) samples were harvested for analysis of oxidative damage and total antioxidant levels. Additionally, the presence of MPs in both tissues was confirmed. Significant differences with the control groups were observed in lipid peroxidation levels in HP in animals exposed to the lowest concentration in P. clarkii and to the highest concentration in L. pugilator. A marked increase in antioxidant levels was also observed in the HL at both concentrations in P. clarkii, and at the highest MPs concentration in L. pugilator. Moreover, several histopathological changes were detected in both gills and HP, including hypertrophied lamellae, lifting or collapse of gill epithelia, loss of normal shape of hepatopancreatic tubules, and epithelial atrophy in the HP tissue. We conclude that exposure to MP beads at selected concentrations results in oxidative damage, induces histopathological changes in gills and HP, and triggers an antioxidant response in two crustacean species.

The Effect of Neonicotinoids Exposure on Oreochromis niloticus Histopathological Alterations and Genotoxicity

This study aimed to examine the side effects of selected neonicotinoids (Acetamiprid, Aceta, and Imidacloprid, Imid) on Oreochromis niloticus juveniles. The acute toxicity, Probit method, revealed an LC50 of 195.81 and 150.76 ppm for Aceta/96 h and Imid/72 h respectively. The fish were divided into three groups that were exposed, for 21 days (n = 5/replicate), to 1/10 of the LC50 of either neonicotinoids, however, the third was an unexposed control group. Results of erythrocytic micronucleus (MN), and nuclear abnormalities (NA) showed that Aceta and Imid exposure caused a significant (p < 0.05) increase in MN by ~ 2.2 and ~ 10 folds, respectively relative to control. NAs occurred at the order of kidney-shaped > budding > binucleated in Aceta, however, budding > binucleated > kidney-shaped was noticed in the Imid group. Histopathological changes in gills, liver, and muscles were observed significantly in both exposed groups with more severity in the Imid group. Collectively, Aceta and Imid have potential genotoxicity and histopathological alterations in O. niloticus.

Oxidative Stress and Histopathological Changes in Gills and Kidneys of Cyprinus carpio following Exposure to Benzethonium Chloride, a Cationic Surfactant.

One cationic surfactant with a wide spectrum of microbiocidal activity is benzethonium chloride (BEC). Despite being widely used, the toxicity data on vertebrate organisms are limited. Therefore, we aimed to evaluate within this study the acute toxicity of BEC on the gills and kidneys of Cyprinus carpio (European carp). An alteration of the antioxidant enzymes activities (glutathione reductase, glutathione peroxidase and glutathione S-transferase) was noticed after 96 h of exposure, along with an elevation of lipid peroxidation and decreased concentration of reduced glutathione, which confirmed that BEC was able to induce toxicity to these tissues. These metabolic effects were correlated with unspecific structural changes observed in gills and kidneys, having moderate degree of severity (such as an increase of melanomacrophages aggregation incidence and cytoplasm vacuolation of goblet cells in collecting tubules) and generally being compatible with life for the exposure time studied. The most severe structural effects were observed in gills after 96 h, noticing a lamellar aneurysm, hemorrhages and lamellar epithelium disruption due to the blood vessels and pillar cells damages and increased blood flow inside the lamellae. By our research we can confirm the utility of biochemical and histological analyses in the fish organs as tools for monitoring the water quality and ecotoxicological potential of chemicals.

Determination of Median Lethal Concentration (LC50) for Endosulfan, Heptachlor and Dieldrin Pesticides to African Catfish, Clarias gariepinus and Their Impact on Its Behavioral Patterns and Histopathological Responses.

Pesticides such as endosulfan, heptachlor and dieldrin persist in aquatic environments as a result of their resistance to biodegradation. However, there is no adequate information about the toxicity of endosulfan, heptachlor and dieldrin to the aquatic organism, African catfish (Clarias gariepinus)—a high valued widely distributed commercially interesting species. The current experiment was performed with the aim to determine the median lethal concentration (LC50) of endosulfan, heptachlor and dieldrin to African catfish (Clarias gariepinus); their behavioral abnormalities and histopathological alterations in several vital organs. A total of 324 juvenile fish were exposed for 96 h to six concentrations of endosulfan and dieldrin at 0, 0.001, 0.002, 0.004, 0.008 and 0.016 ppm, and to heptachlor at concentrations of 0, 0.02, 0.04, 0.08, 0.16 and 0.32 ppm for dose-response tests. The study demonstrated that the species is highly susceptible to those contaminants showing a number of behavioral abnormalities and histopathological changes in gill, liver and muscle. The 96-h LC50 value of endosulfan, dieldrin and heptachlor for the African catfish was found as 0.004 (0.001−0.01) mg/L, 0.006 mg/L and 0.056 (0.006−0.144) mg/L, respectively. Abnormal behaviors such as erratic jerky swimming, frequent surfacing movement with gulping of air, secretion of mucus on the body and gills were observed in response to the increasing exposure concentrations. Histopathological alterations of liver, gill and muscle tissues were demonstrated as vacuolization in hepatocytes, congestion of red blood cells (RBCs) in hepatic portal vein; deformed secondary lamellae and disintegrated myotomes with disintegrated epidermis, respectively. These findings are important to monitor and responsibly manage pesticide use in and around C. gariepinus aquacultural areas.

Histological Morphology of Gill, Skin and Liver of Nile Tilapia Oreochromis Niloticus Fingerling Exposed to Jatropha Curcas (Linnaeus I779) Seed Powder

The effect of Jatropha curcas seed powder on the histopathology of selected organs of Oreochromis niloticus fingerlings was investigated. O. niloticus fingerlings of 6.5cm-10cm in length and 14.7g-18g in weight were exposed to different concentrations of J.curcas seed powder at 133.3mg/l, 200mg/l, 266.7mg/l, 333.3mg/land 400mg/l. The acute toxicity of the toxicant to O. niloticusfingerlings was carried out using bioassay method under laboratory condition and the 96h LC 50 was 439.23mg/l with maximum admissible concentration of the toxicant to be 4.3923mg/l – 43.923mg/l which is derived by multiplying the 96h LC 50 with an application factor of 0.01 – 0.1 . After 96hr of exposure, Histopathological changes of gill, liver and skin were observed. In the gill there was Degeneration of the gill arch,Lamellae are severely stunted,Shortened and High degeneration of gill arch, filament &amp; lamellae and Severe submucosal congestion, secondary lamellae are absent at the highest concentration. In the skin there wasNecrosis of the skinandHypertrophy, Necrosis and erosion of dermal cell. In the liver Disorientation of the liver parenchyma structures, Hyperplasia and disorientation of hepatic cells and marked sinusoidal congestion was observed. The pattern of response of O. niloticus fingerlings at the various concentration of J. curcas seed powder varied widely at the exposure duration. The observable effect of the toxicant on the examined organs could be could serve as a baseline information needed to develop models of J. curcas seed effects on ecological systems.

Perfluorinated compounds disrupted osmoregulation in Oryzias melastigma during acclimation to hypoosmotic environment

Perfluorinated compounds (PFCs) are a type of ubiquitous contaminants spreading in the estuarine and coastal areas. Anadromous fish should deal with hypoosmotic challenge with PFCs stress during their migration from seawater to estuaries. However, few studies have been carried out to investigate the adverse impact of PFCs on fish osmoregulation and the underlying mechanism. In this study, Oryzias melastigma, an euryhaline fish model, were exposed to four representative PFC congeners including perfluorobutane sulfonate (PFBS), perfluorooctane sulfonates (PFOS), perfluorooctanoic acid (PFOA), and perfluorododecanoic acid (PFDoA) separately under both seawater and freshwater conditions. Histopathological changes in gills, ion homeostasis, Na+/K+-ATPase (NKA) activity, as well as the expression of related genes was detected upon exposure. Results showed that PFCs induced morphological changes in gills, disturbed the levels of major ions (Na+, Ca2+, Mg2+), and inhibited the NKA activity. Transcriptome analysis in fish gills during the acclimation to freshwater revealed that PFCs influenced the osmoregulation mainly by interfering with the endocrine system, signal transduction, as well as cellular community and motility. Validation with qRT-PCR confirmed that the mRNA expressions of osmoregulatory genes encoding hormones and receptors, as well as ion transmembrane transporters were disturbed by PFCs. Longer chain homolog (PFOS) showed a greater impact on osmoregulation than the shorter chain homolog (PFBS). Within the same carbon chain, sulfonic congener (PFOS) induced more serious injury to gills than carboxylic congener (PFOA). The interaction between PFCs and salinity varied in different adverse outcome. These results help to further understand the mechanism of how PFCs influence osmoregulation and elicit the need to assess the ecological risk of PFCs and other pollutants on anadromous migration.

Heavy Metal Bioaccumulation and Related Histopathological Changes in Gills, Liver and Muscle of Solea aegyptiaca Fish Inhabiting the Lake Qarun, Egypt.

Pollution of Lake Qarun with heavy metals has become a more serious concern during recent years as it affects the physicochemical characteristics of the water, sediments and biological components and may damage aquatic organisms as well. Lake Qarun receives the agricultural and sewage wastewater loaded with heavy metals through two main drains; El-Batts and El-Wadi. Therefore, an annual increase in the accumulation of pollutants and heavy metals has been detected in all its components (e.g. water, sediment and fish). The present study aimed to evaluate the seasonal variation of heavy metals’ contents (Fe, Cu, Zn, Cd, Pb and Ni) in water and the bioaccumulation factor of these heavy metals in Solea aegyptiaca fish samples, collected from Lake Qarun. Thus, information was provided regarding histological changes in gills, liver and muscle organs. This study was carried out along Lake Qarun at four sites (Eastern, Southern, Northern and Western) during the winter and summer seasons, 2019. The results revealed that summer was higher in heavy metal concentration in water than winter. Additionally, the eastern and southern sectors showed increased water concentration of heavy metals. Concerning fish samples, the present study exhibited that, the maximum values of bioaccumulation factor were recorded in the gills and liver, whereas the minimum values were observed in the muscles. The gills of Solea aegyptiaca accumulate higher levels of Zn, pb and Ni, while, Fe, Cu and Cd accumulated mainly in the liver. Results showed obvious histopathological alternations and clear damage in gills, liver and muscles of Solea aegyptiaca collected from the eastern and southern sectors of Lake Qarun.

Histopathological changes in gills and liver of Clarias gariepinus fingerlings exposed to acute concentrations of dry cell battery

Waste dry cell batteries are frequently improperly disposed and subsequently washed into water bodies-- causing deleterious effects on fish particularly Clarias gariepinus which inhabits diverse freshwater habitats. Acute toxicity of water-soluble fractions of waste dry cell batteries was investigated on C. gariepinus fingerlings under laboratory conditions in 96 hours. Ten (10) C. gariepinus fingerlings were exposed to acute concentrations (0.31, 0.63, 1.25, 2.50, and 5.00 g/L) of waste dry cell batteries and a control (0.00 g/L), each duplicate replicated. Histopathological alterations evident in the gills were lamellar fusion, hyperplasia, inter-lamella space occlusion, hypertrophy and erosion of secondary lamellae. The liver showed nuclear and hepatocytes degeneration, vacuolation and portal congestion. Acute concentrations of water-soluble fractions of waste dry cell batteries caused significant (P&lt;0.05) changes in the histomorphology of the gills and liver of C. gariepinus fingerlings, therefore indiscriminate disposal of waste dry cell batteries around riparian ecosystem should be safeguarded to reduce the declining diversity and abundance of freshwater fish species.&#x0D; Keywords: African catfish, 96 hr.LC50, Fingerlings, Histopathology Zinc-carbon battery

Estudo histopatológico em brânquias de juvenis de Rhamdia quelen submetidos a ensaio toxicológico crônico com ibuprofeno

The present study aimed at evaluating histopathological changes in gills of Rhamdia quelenjuveniles submitted to different concentrations of ibuprofen (0.0; 0.5; 5.0 and 50.0 mg/L); concentrations were determined from LC50acute assays of 5.0mg/L(120h). For each concentration four replicates were made, with four copies each. The experiment had a total duration of 120 hours in a chronic toxicological trial. After completion, the animals were euthanized, laparotomized, and the gills were localized, isolated and fixed in 10% formaldehyde, preserved in 70% alcohol and subsequently submitted to histological routine technique for paraffin inclusion. Fivecuts (6μM) per specimen were made and stained with Hematoxylin and Eosin (H&amp;E). The Average Alteration Score(AAS) and Histopathological Alteration Indices (HAI) were determined according to the degree of the lesion and then classified in stages I, II and III which produced respectively Lamellar derangement, Lamella’s epithelium rupture and aneurysm with more expressive alterations. For all concentrations the drug led to histological alterations for Rhamdia quelen, putting in evidence damages that go from moderate to severe in the gills of the analysed fishes. Ibuprofen caused branchial lesions in R.quelenwith a progressive effect in high concentrations, suggesting that the presence of this drug could cause abnormalities and favour mortality in this species.

Immunological, hematological, and histological responses in blood, spleen and gill of Salmo caspius juveniles exposed in different water temperatures

Caspian brown trout, Salmo caspius, were subject to five temperatures (5°C, 8°C, 20°C, 25°C, and 28°C) for two durations 12 and 24 hours. The fish were maintained for 48 hours at a water temperature of 9-10°C to acclimatize.The water temperature was changed gradually at a rate of 2°C per hour to the respective temperatures. Exposure to temperatures of 25 and 28°C resulted in loss of all fish. Red blood cell, hemoglobin concentration, and hematocrit were decreased at 5°C compared to control (8°C) and 20°C in duration of 12h. Temperature tension did not affect the mean cell volume and mean cell hemoglobin value during the 12 and 24h duration (p >0.05). Neutrophils percentage was significantly higher at 5°C and 20°C, but the most percentage of lymphocytes was noted in the control group. Cortisol, glucose, and lactate values elevated in both periods when water temperature decreased to 5°C compare to the control and 20°C groups (p <0.05). The levels of albumin and total protein in the duration of 12h at 20°C were significantly higher than control and 5°C, but this trend was not seen in the exposure of 24h (p <0.05). The lysozyme activity and immunoglobin M were affected by varying temperatures (p <0.05), and highest levels were observed in control groups. Most histopathological changes in gill, such as epithelial hypertrophy, curling of lamella, and necrosis of the epithelial cell were observed at 20°C; whereasthese were less affected at 5°C and 8°C. Lowest hemorrhages in the spleen and lowest size of sinusoids were observed in 8°C and 5°C treatments respectively. Overall, temperature variation had a superior impact on gills than spleen. Taken together, the results of the present study showed that sudden variation beyond the optimum temperature leads to physiological and pathological changes.

Zinc Induced Histopathological Alternations in Gill, Liver, and Kidney of Freshwater Fish Gonoproktopterus kolus (Sykes)

The present study aims to investigate the histopathological changes in gill, liver, and kidney of fresh water fish Gonoproktopterus kolus (Sykes). The fingerlings of fish were exposed to Zinc (Zinc sulphate) in acute toxicity experiment (96 hrs.). The LC₀ and LC₅₀ concentrations were 2.430 ppm and 4.860 ppm respectively. After acute exposure, fish were sacrificed and tissues were obtained separately from control and LC₀ and LC₅₀ groups. These tissues were fixed in Bouin's fluid and standard protocol was followed to study histopathological alterations in gill, liver and kidney of experimental fish. In gills, fusion and degenerative changes were observed in secondary gill lamellae. In liver, hepatocytes showed hypertrophy due to degenerative changes in cellular structure. Empty hepatocytes showed shifting of nucleus. Kidney showed degenerative nuclei in the epithelial lining of the tubule. Haemopoetic tissue showed hypertrophy and disarray. Distended glomerulus was observed. Renal tubules showed disarrangement as compared to control. Thus, in present investigation, it was observed that heavy metal zinc is affecting the fishes at tissue level, which may interfere in physiological processes.

Effects of heavy metal pollution on hepatosomatic ındex and vital organ histology in Alburnus mossulensis from Karasu River

In this study, histological effects of water pollution to freshwater cyprinid, Alburnus mossulensis, was investigated. Water, sediment, and fish samples were taken from four stations in Karasu River (Erzurum, Turkey) between June and September in 2015? 2016. In water and sediment samples, the concentrations of Ti, V, Cr, Mn, Fe, Co, Ni, Cu, Zn, As, Se, Br, Sr, and Pb were detected. Histopathological changes in gill, liver, and kidney tissues were determined by degree of tissue change (DTC), and hepatosomatic index (HSI) was calculated. The mean concentrations of heavy metals in water and sediment samples were lower at station 4. The trend of some metal contents found in the fish was in increasing order of Zn < Cu < Cr < Pb. The observations on histopathology included cellular degeneration, congestion, dilatation, vacuolization, edema, vasodilatation, hyperplasia, hypertrophy, infiltration, melanomacrophage aggregates, glomerulopathy, fibrosis, and necrosis. It was observed that the DTC and HSI values in fish varied significantly from station to station. The results showed that the higher severity of histopathological alterations in the fish tissues was strongly correlated with agricultural and domestic activities in the area.

Histopathology and immunohistochemistry of gills of Van fish (Alburnus tarichi Güldenstädt, 1814) infected with myxosporean parasites

ABSTRACT The Van fish are a cyprinid species endemic to Turkey’s largest soda lake, Lake Van, and have great economic value because they are a food source. Once a year, the fish take part in reproductive migration to the fresh waters flowing into the lake. The fish migrate from an extreme environment with high salinity (2.2%) and high pH (9.8). These fish are unable to reproduce in this alkaline environment and must migrate to fresh water during their breeding season. The aim of the present study is to report the presence of the myxosporean parasites on the gills and the pathological changes. Changes in gill histopathology, mucocytes, mitochondria-rich cells, expression of Heat Shock Protein 70 (Hsp70), and ATPase (NKA) were observed in the gill tissue. As a result of the histopathological changes in gills, infected fish had abundant plasmodia with different sizes. Plasmodia were found on gill filaments inside white ovoid-shaped structures. It was observed that plasmodia were contained on the primary filament which changed the histological structure of the gill tissue to a large extent. It was determined that the density and size of mucocytes in the infected areas of the gill tissue increased, whereas the number of mitochondria-rich cells decreased. Hsp70, an indicator of stress, was not different between normal and infected fish.

Effect of Biofilm of Aeromonas Hydrophila Oral Vaccine on Growth Performance and Histopathological Changes in Various Tissues of Striped Catfish, Pangasianodon Hypophthalmus (Sauvage 1878)

Biofilm of Aeromonas hydrophila oral vaccine was evaluated on growth and histopathological features of striped catfish, Pangasianodon hypophthalmus. Fish were fed with feed incorporating biofilm (BF) or free cell (FC) of A. hydrophila vaccine at 1010 cells/fish/day for 20 days and basal feed for 60 days. From 30th day onwards, till end of the trial, the mean weight gain (g/fish) was significantly higher (P less than 0.05) in BF treated group than both FC and control group. At the end of the study, histopathological changes in gills, liver and kidney were evaluated using haematoxylin eosin technique. The histopathological findings, in BF fed P. hypophthalmus, showed normal architechture of the gills, liver and kidney. However, fishes fed on FC and control group, resulted several histopathological abnormalities in all the organs.

Biochemical and haematological responses to thermal stress in Labeo rohita (Hamilton, 1822) fingerlings

An experiment was conducted to understand the effect of thermal shock on stress and recovery in Labeo rohita (Hamilton, 1822) fingerlings. L. rohita fingerlings were exposed to a temperature of 380C for 2 h and permitted to recuperate afterwards for 48 h. Primary and secondary stress parameters were analysed after 2, 4, 8, 12, 24 and 48 h of recovery. The activity of glucose-6-phosphate dehydrogenase and acetylcholine esterase reduced significantly (p&lt;0.05). Erythrocytes, haemoglobin, respiratory burst activity and lysozyme levels increased significantly (p&lt;0.05) by 7.31, 23.91, 109.09 and 70.13% after thermal shock, whereas leucocytes, serum protein, albumin and globulin showed a reverse trend. During recovery, biochemical parameters normalised in 48 h. However, immunological variables such as serum total protein, albumin and globulin, and histopathological changes in gill and kidney tissues did not return to normal condition.

Chronic exposure to dichloroacetamide induces biochemical and histopathological changes in the gills of zebrafish.

To evaluate the impact of DCAcAm on zebrafish gill, we measure the responses of antioxidant enzyme (superoxide dismutase, SOD), lipid peroxidation (malondialdehyde, MDA), ATPase (Na+ /K+ -ATPase and Ca2+ /Mg2+ -ATP) and histopathological changes of gill in adult zebrafish, after exposed to different concentrations of DCAcAm (0, 1, 10, 100, and 1000 μg L-1 ) for 30 days. Results indicated that DCAcAm first increased and then decreased SOD activity, and DCAcAm also lowered the activities of Na+ /K+ -ATPase and Ca2+ /Mg2+ -ATPase. These results indicated that high affinity of DCAcAm probably be a main factor, which can damage the structures of enzymes, thereby inhibiting the SOD and ATPase activities. Besides, histopathological investigation results also manifested that chronic exposure to DCAcAm can damage the gill tissues, disrupting the normal function of gills. We conclude that chronic exposure to DCAcAm was harmful to organisms, not only influence gill function, but also further cause damage on the gill tissues.
