Hepatotoxic Test Of Burdock Roots (Arctium Lappa L) On Histopathology Hepar Of Male White Rats (Rattus Norvegicus

Burdock (Arctium lappa L) is a homologous medical plant that is considered having with many advantages as a traditional medicine. Burdock root contains several substances such as inulin, essential oils, tannins, quercetin,iron, and arctigenin. This study was conducted to determine investigates effect of burdock root extract on the hepar histopathology of male white rats. This study used an experimental method with 20 rats and was divided into 4 groups. The positive control group was given gentamicin induction intramuscularly. The negative control group was given distilled water. The 150 mg/kgBW dose group and the 450 mg/kgBW dose group were given burdock root extract with treatment for 14 days and surgery was performed oin the 15th day to take hepatic organs to make histopathology preparations. The results of the this study were are interpreted by scoring and tabulated, the analysis was is done carried out descriptively with the results, in positive control group found mild damage, the negative group found no damage, in the extract dose group 150mg/kgBW found necrosis, bleeding and vacuolization, and in the extract dose group 450mg/kgBW found degeneration, necrosis, bleeding and vacuolization. The conclusion of this study is the hepatoprotective potential of burdock root extract (Arctium lappa L.) and its related effects on liver tissue in an experimental model. On the other hand, parasitic worms Taenia taeniaeformis were found in the liver which interfered with the interpretation of the results.

Human Mesenchymal Stem Cell Promotes Burn Wound Healing by Inducing FGF and VEGF in Diabetic Rat.

<b>Background and Objective:</b> The hBM-MSCs have a high level of differentiation and proliferation in the healing process of cuts and burns. This study aimed to determine the role of hBM-MSCs in the formation of granulation tissue in diabetic burnt rats. <b>Materials and Methods:</b> Thirty rats were divided into two groups; phosphate-buffered saline (PBS) as a control group and a treatment group (BM-MSCs 2×10<sup>6</sup> cells/mL). Both groups were treated as hyperglycaemia by injecting alloxan. Rats were anesthetized using xylazine and ketamine and given full-depth burns on the dorsal using a heated plate. Rat skin tissue was excised on the 3rd, 7th and 14th day and histopathological preparations were made using immunohistochemical staining to determine the expression of the growth factors of FGF and VEGF. The results were analyzed using Tukey's t-test advanced. <b>Results:</b> The FGF level increased statistically significantly on day 3, 7 and 14 in the treatment group. Otherwise, on day 14 there was a significant difference between the control group and the treatment group (p = 0.017). The VEGF expression also showed an increase on days 3 and 7 but decreased on day 14. The VEGF level was not statistically significant between the control and treatment groups. <b>Conclusion:</b> The hBM-MSCs increased the formation of granulation tissue by expressing a high level of FGF which plays a role at the beginning of new blood vessel formation and VEGF affects the end of the formation of new blood vessels.

Relationship of Stromal Tumor Infiltrating Lymphocytes (STILs) with Grading Histopathology in Penis Squamous Cell Carcinoma

ABSTRACT Background:Penile squamous cell carcinoma is the most common penile malignance found in the world. Assessment of stromal tumor infiltrating lymphocytes (STILs) associated with grading against penile squamous cell carcinoma can provide a potential prognostic picture and help determine the action. Method:Cross-sectional analytical study with 32 samples of histopathological preparations diagnosed as penile squamous cell carcinoma at H. Adam Malik Hospital Medan. The assessment of stromal TILs was carried out by assessing the number of lymphocyte inflammatory cells in the peritumoral stroma and then determined the assessment score, zero score no lymphocytes (minimum), score one found 0-10% lymphocytes (moderately), score two found 20-40% lymphocytes (massive) , score three found 50-90% lymphocytes. Grading assessments are categorized into grade I (well differentiated), grade II (moderately differentiated), and grade III (poorly differentiated). Result:The most grading found was grade II (43.8%) and the most STILs found were score three (50.0%). There is a significant relationship between grading and STILs in penile squamous cell carcinoma (p value=0.0001) i.e. the higher the STILs score , the lower the tumor grade. Conclusion:There is a relationship between STILs and histopathological grading in Penile Squamous Cell Carcinoma so that it can be used as an indikasi prognosis in patients with penile squamous cell carcinoma.

Safety evaluation of recombinant Newcastle disease virus expressing IBV multi-epitope chimeric live vaccine.

Newcastle Disease (ND) and Infectious Bronchitis (IB) are two significant diseases that pose threats to the poultry industry, caused by Newcastle disease virus (NDV) and Infectious bronchitis virus (IBV), respectively. Currently, the control and prevention of these diseases primarily rely on vaccination. However, commercial ND and IB vaccines face challenges such as poor cross-protection of inactivated IBV strains and interference from live vaccines when used together, leading to immunization failures. Previously, we reported the successful rescue of a recombinant NDV expressing multiple epitopes of IBV, named rNDV-IBV-T/B, which showed promising immunoprotective efficacy against both NDV and IBV. This study focuses on the biosafety of the genetically modified recombinant vaccine candidate rNDV-IBV-T/B. Immunization was performed on day-old chicks, ducklings, goslings, and ICR mice. Observations were recorded on clinical symptoms, body weight changes, and post-mortem examination of organs, as well as histopathological preparations of tissue samples. The results indicated that the rNDV-IBV-T/B vaccine candidate had no adverse effects on the growth of targeted animals (chickens) and non-target species (ducks, geese) as well as in mammals (mice). Additionally, histopathological slides confirmed that the vaccine is safe for all tested species. Further studies evaluated the potential of rNDV-IBV-T/B to spread horizontally and vertically post-immunization, and its environmental safety. The findings revealed that the vaccine candidate lacks the capability for both horizontal and vertical transmission and does not survive in the environment. In conclusion, the rNDV-IBV-T/B strain is safe and holds potential as a new chimeric live vaccine for ND and IB.

Differences of mandibular condylar cartilage thickness of Mus musculus after given different intensities of physical stress

Objective: This study aimed to evaluate the differences in the thickness of Mus musculus mandible cartilage after being given low, moderate, and high-intensity physical stress. Materials and Methods: This research is an experimental laboratory study. A total of 12 Mus musculus were divided into four groups, which are the control group(C), low (LS), moderate(MS), and high-intensity physical stress group(HS). The physical stress group was given treatment with swimming exercises using 3%, 6%, and 9% of body weight with a duration of 40%, 70%, and maximum swimming time calculation. After the experiment was carried out, the animal was decapitated and deparaffinized using xylol to dissolve paraffin in tissue to make histopathology preparations that were stained with hematoxylin-eosin. The thickness of mandibular condylar cartilage was calculated under a light microscope with a magnification of 400x and analyzed using the One Way ANOVA test to determine if the relationship between the sets of data in the treatment group is statistically significant (p<0.05). Results: The thickness of the mandibular cartilage was measured to be thinner in the MS group (56.30 ± 6.11) and the HS group (78.04 ± 19.31) compared to the C group 85,85 ± 7,20. Statistical analysis with One Way ANOVA test showed that all groups had a significant difference in the thickness of mandibular condylar cartilage (p=0.049). Conclusions: There was a difference in the thickness of the mandibular cartilage after giving low, moderate, and high-intensity physical stress.

HISTOPATHOLOGICAL FEATURES OF DOGS SKIN WITH ATOPIC DERMATITIS AFTER TREATMENT USING ECO ENZYME

Atopic dermatitis is one of the most common diseases in dogs that is multifactorial and usually attacks dogs aged between 6-36 months. This study was aimed to determine the histopathological appearance of the skin of dogs suffering from atopic dermatitis after treated with eco enzyme. Eco enzyme is a liquid product resulting from the fermentation of organic kitchen waste product such as fruit and vegetable dregs, molasses, and water. This study used five dogs suffering from atopic dermatitis which were divided into two groups. Group A: three dogs samples were bathed with 10% eco enzyme liquid every three days for 28 days. In Group B: two samples of dogs were bathed with 10% eco enzyme every three days until the 9th day, followed by a 2% eco enzyme bath once a week (13th, 20th, 27th day). Both treatment groups started from day 0 until day 28. Skin samples were taken using the biopsy method and histopathological changes were observed by making histopathological skin preparations stained with Hematoxylin and Eosin. Observations of the preparations were carried out using a light microscope. Data were analyzed using the Analysis of Variance test followed by the Duncan multiple range test and the Kruskall Wallis test followed by the Mann-Whitney test, then the data was explained descriptively. The results of the Anova test showed that the thickness of the dogs epidermis treated with 2% and 10% eco enzyme was significantly different (P<0.05). The Kruskall Wallis test for scoring inflammatory cell infiltration and degeneration showed that there was a significant difference (P<0.05) in the administration of 2% and 10% eco enzyme. Eco enzyme liquid is able to reduce atopic dermatitis lesions in dogs. Giving eco enzyme at a concentration of 10% and followed by a concentration of 2% with an extended application time can return the histological structure of the dog’s skin to normal, especially the thickness of the epidermis, reduce inflammatory cell infiltration, and reduce degeneration.

HISTOPATOLOGICAL OF WHITE RATS' KIDNEY CAUSED BYAPPLICATION OF MIMOSIN FROM SIMPLISIA OF THE LEUCAENA LEAF

Mimosine is a toxic substance contained in Leucaena leucocephala leaves. The toxic effects of mimosine can disrupt several tissues and organs, especially the kidneys. This study aims to determine the histopathology of white rat kidney exposed to mimosine with different doses. Male white rats with Wistar strain, 2 months old and 300-350 g body weight were being used in this study. The 20 rats used were divided into four treatment groups, namely P0 (negative control), P1 (positive control, given standard mimosine 15 mg/head/day), P2 given leucena leaf simplisia at a dose of 50 mg/head/day orally, P3 given leucaena leaf simplisia at a dose of 150 mg/head/day. On the 16th day of necropsy, the skin organs were taken and placed in 10% NBF. After the kidney organs were fixed, histopathology preparations were made using HE staining. Histopathological examination was performed including three lesion variables, namely congestion, bleeding, and inflammation. The severity of the lesions were scored as 0, 1, 2 and 3 respectively for normal, mild, moderate and severe lesions. Data were analyzed using non-parametric Kruskal-Wallis and Mann- Whitney tests. The results of the research showed that the mimosine substance in leucaena leaf simplicia caused some histopathological changes in the kidneys of white rats like congestion, bleeding and inflammation and there was a difference in dose effect for each giving of mimosine from leucaena leaf simplicia, between the dose of 50 mg/head/day and the 150 mg/head/day. Administration of mimosine from leucaena leaf simplicia can cause histopathological changes in the kidneys of white rats and there is an effect of the difference in the dose of mimosine from leucaena leaf simplicia between a dose of 50 mg/head/day and a dose of 150 mg/day. tail/day on changes in kidney pathology of white rats, especially in the form of bleeding and inflammation. With this research, it hoped that in the future further research can be carried out on leucaena leaf simplicia with a longer administration period so that the results obtained are more accurate.

Test Of The Effectiveness Of Oral Administration Of Red Dragon Fruit (Hylocereus Polyrhizus) In Preventing Dyslipidemia In Male Wistar Rats (Rattus Norvegicus) Given A High High-Cholesterol Diet

Hyperlipidemia can result from obesity, which causes excess fat to build in the tissue under the skin and spread to other organs and tissues. Indonesia has an alarming dyslipidemia rate. Diet and exercise are vital. Hypolipidemia is in red dragon fruit (Hylocereus spp.) The study examined whether oral red dragon fruit extract prevented dyslipidemia in male Wistar rats (Rattus norvegicus) fed a high-cholesterol diet. This research used 24 mice as test animals. The research procedures included acclimation of the test animals, dragon fruit extract, phytochemical tests, preparation of the test animals, treatment with four groups, dyslipidemia parameter checks, histopathological preparations, and observations. Normal tests for total cholesterol, LDL, LSD, and HDL yielded p-values> 0.05. Homogeneity tests with a 5% significance level yielded p values < 0.05. This study's one-way Anova test yields a significance value of 0.000, or <0.05, whereas the post-hoc LSD test yields <0.05. The best red dragon fruit extract dose for lowering total and LDL cholesterol and raising HDL is 120 mg/mL. The phytochemical experiments on red dragon fruit extract (Hylocereus polyrhizus) show promise for improving the lipid profile.

Efek Terapi Pemberian Vitamin E Terhadap Kerusakan Histopatologi Ginjal Tikus Putih (Rattus norvegicus) Yang Diinduksi Dexamethasone

The purpose of this study was to determine whether or not there was a therapeutic effect of vitamin E administration on dexamethasone-induced kidney histopathological damage of white rats. This study used 20 white rats (Rattus norvegicus) aged 2-3 months with a body weight of 200 grams divided into 5 groups, namely negative control (K) without dexamethasone and vitamin E, positive control group (P0) given dexamethasone subcutaneously at a dose of 0.13 mg / Kg BB, treatment group 1 (P1) given dexamethasone subcutaneously at a dose of 0.13 mg / Kg body weight and vitamin E orally at a dose of 150 mg / Kg body weight, treatment group 2 (P2) given dexamethasone subcutaneously at a dose of 0.13 mg / Kg body weight and vitamin E orally at a dose of 200 mg / Kg body weight, and treatment group 3 (P3) given dexamethasone subcutaneously at a dose of 0.13 mg / Kg body weight and vitamin E orally at a dose of 250 mg / kg body weight. Adaptation is carried out for 7 days. Experimental animals are then terminated and kidney organs are taken to make histopathological preparations with HE staining then the preparations are observed under a microscope. The observed parameter is damage to the glomerulus and proximal tubules of the kidneys. The results showed that dexamethasone was able to damage the kidneys characterized by necrosis of the glomerulus and hydropic degeneration of the proximal tubules of the kidneys shown in the positive control group (P0). In the group given vitamin E, only the P3 group with a dose of vitamin E 250 mg / kg body weight was able to provide a therapeutic effect on damage to the glomerulus and renal tubules due to the toxic effects of dexamethasone.

Expression of Kidney White Rats Diabetes Mellitus after Therapy of Sinom Ethyl Acetate Fraction Mixed Lime and Honey

This study aims to prove that the administration of the ethyl acetate fraction sinom mixture of lime and honey (SCJM) can improve the histological structure of the kidney of Spraque Dawley rats (SD) diabetic were divided into six groups with four replications each. The K1 group as a positive control was not given a sample, the K2 group was given a sample dose of 50 mg/kg BW, the K3 group was given a sample dose of 100 mg/kg BW, and the K4 group was given a sample dose of 150 mg/kg BW, the K5 group was given an extract dose of 200 mg/kg BW, and positive control group K6 which was carried out for 21 days. The rats were then sacrificed and the kidney organs were taken for histopathological preparation with Hematoxylin Eosin (HE) staining. The results showed that there was fatty degeneration and necrosis of the kidneys. Kruskal Wallis test results from all treatments showed p = 0.001 for both fatty degeneration and necrosis (p<0.05).abstract is written along 100-200 words with a single space. Reliable Method for Estimating Nerve Fiber Density in Epidermis Using Routine Histopathologic Tissue Preparation: A Promising Diagnostic Tool for Small Fiber Neuropathy.

Practical yet reliable diagnostic tools for small-fiber neuropathy are needed. We aimed to establish a histopathologic protocol for estimating intraepidermal nerve fiber density (eIENFD) on formalin-fixed, paraffin-embedded tissue (FFPE), evaluate its reliability through intraobserver and interobserver analyses, and provide normative reference values for clinical use. Sixty-eight healthy participants underwent nerve conduction studies and quantitative sensory testing. Skin biopsies from the distal and proximal leg were taken and processed using routine immunohistochemistry (anti-PGP9.5 antibodies) on thin 5 µm sections. eIENFD was assessed with a modified counting protocol. Interobserver and intraobserver reliabilities were excellent (ICC=0.9). eIENFD was higher in females than males (fibers/mm, 14.3±4.4 vs. 11.6±5.8, P <0.05), decreased with age ( r s =-0.47, P <0.001), and was higher proximally than distally (15.0±5.5 vs. 13.0±5.3, P =0.002). Quantile regression equations for the fifth percentile of distal and proximal eIENFD were presented: 13.125-0.161×age (y)-0.932×sex (male=1; female=0) and 17.204-0.192×age (y)-3.313×sex (male=1; female=0), respectively. This study introduces a reliable and reproducible method for estimating epidermal nerve fiber density through immunostaining on 5-µm thin FFPE tissue samples. Normative data on eIENFD is provided. Regression equations help identify abnormal decreases in small nerve fiber density.

NEWCASTLE DISEASE AND AVIAN INFLUENZA IN BROILER THAT VACCINATED ON ONE DAY OLD

Newcastle disease and Avian Influenza are highly contagious viral diseases that can affect poultry with high mortality rates, and are zoonotic diseases that endanger humans. In this case study, broiler chickens aged 12 days were vaccinated at hatching and came from Yangapi Village, Tembuku, Bangli showed clinical symptoms of drooping neck or torticollis, no appetite, and greenish white diluted feces. Examinations are continued at each laboratory to obtain a definite diagnosis. The methods used are anatomical pathology examination, histopathology preparations, HA-HI examination, bacterial identification, and qualitative examination for paracites. The results of anatomical pathology examination found pathognomonic lesions, like ptechie in the proventriculus and ventriculus, enlargement and congestion in the lungs, hemorrhagic trachea, and hyperemic intestines. The results of histopathological examination showed marked changes with single-core inflammatory cells or lymphoid suggesting an infectious agent in the form of a virus. The results of the virology laboratory examination with the HA and HI test showed that the case chicken was positively infected by Newcastle Disease and Avian Influenza viruses. Bacteriology laboratory by culture of lung and intestinal organ samples was carried out on Nutrient Agar, followed by selective media Eosin Methylene Blue Agar, primary tests, gram staining, biochemical tests and sugar tests until the results showed that Escherichia coli bacteria grew from the chicken intestines and were classified as normal flora. The results of the examination related to parasites in the feces of case chickens by qualitative methods, natif, sedimentation concentration, and floating were confirmed not to be found worm eggs or coccidia. From this case study, it can be concluded that the case chicken was infected with Newcastle Disease and Avian Influenza viruses.

Analysis of the Expression of LSF Transcription Factor in the Regulation of Transcription and TSG101 during the Neoplastic Transformation of Endometrial Cells.

Previous research indicates that carcinogenesis involves disrupting the functions of numerous genes, including factors involved in the regulation of transcription and cell proliferation. For these reasons, in endometrial carcinogenesis, we decided to investigate the expression of TSG101 (a suppressor of tumor transformation) and LSF (a transcription factor involved in numerous cellular processes, such as cell cycle regulation, cell growth, development, and apoptosis). LSF may be involved in the regulation of TSG101 expression. The research material consisted of endometrial cancer samples from 60 patients. The control group consisted of normal endometrium samples donated by 60 women undergoing surgery for benign diseases of the female reproductive organs. The samples were subjected to immunohistochemical staining with antibodies specific to TSG101 and LSF. Specific antibodies were used to identify TSG101 and LSF in the examined histopathological preparations. An approximately 14-fold lower risk of endometrial cancer development was observed in patients with TSG expression in more than 75% of the assessed cells (4% vs. 36%; OR = 0.07; p = 0.0182). There was a four-fold lower risk of endometrial cancer development in patients with LSF expression in more than 50% of the assessed cells (32% vs. 64%; OR = 0.26; p = 0.0262). A more than three-fold lower risk of endometrial cancer development was observed in patients with LSF expression in more than 75% of the assessed cells (24% vs. 52%; OR = 0.29; p = 0.0454). Endometrial cancer was diagnosed in those with a lower level of TSG101 expression than in those with a cancer-free endometrium. Decreased expression of TSG101 may be a marker of endometrial cancer, and increased expression of LSF when diagnosed with endometrial cancer may indicate greater advancement of the disease. These markers might be used as diagnostic and prognostic markers-however, there is a lack of a correlation between them.

Potential of Ciplukan Leaf (Physalis Angulate L.) Infusion as A Bronchodilator in Pulmonary Histopathology of Sprague Dawley White Rats

Ciplukan leaves contain alkaloid compounds and exhibit pharmacological activity as bronchodilators. Bronchodilators are utilized in chronic inflammatory respiratory conditions such as asthma, involving various cell types such as mast cells, eosinophils, and epithelial cells. The objective of this study was to evaluate the effectiveness of administering Ciplukan leaf infusion as a bronchodilator. A total of 24 Sprague Dawley rats were divided into 6 groups: normal control (K1), negative control (K2), positive control (K3), 200 mg/kg BW dose (K4), 400 mg/kg BW dose (K5), and 800 mg/kg BW dose (K6). On days 1 and 8, rats were sensitized with 0.35 mg OVA + 1 mg AlOH3 in 0.9% NaCl (i.p). On days 15, 17, and 19, rats were re-sensitized with inhaled OVA. From day 20 to 28, K1 and K2 received no treatment, K3 received 2 mL/day of salbutamol, and the dose groups were administered 2 mL/day of Ciplukan leaf infusion; all treatments were carried out simultaneously. On day 29, rats were dissected, and their lung organs were collected for histopathological preparation. Research findings revealed a reduction in bronchiolar epithelial thickness across groups in the following order: 495.05; 584.95; 334.69; 421.70; 482.81; 484.95 µm. The sequential lung eosinophil counts were 19; 44; 37; 23; 17; and 27, and mast cell counts were 9; 22; 13; 15; 13; and 7. Statistical analysis demonstrated significant differences at the 200 mg/kg BW dose for the parameters of bronchiolar epithelial thickness and eosinophil count, as well as significant differences in mast cell counts at the 800 mg/kg BW dose and negative control group.

Effect of Moringa Flower Extract (Moringa Oleifera) on Collagenization and Histopathology of Skin Tissue in the Healing of Dermapen Scars in Obese Male Wistar White Rats

Injury can be accidental, intentional, or disease-related. Moringa leaves may help heal wounds. The research aims to test and assess the effects of Moringa oleifera flower extract on dermapen wound healing collagenization in male Wistar strain white rats, an obese model, and their histological findings—quantitative True Experimental Study with Post-Test Only Control Group Design. Twenty-four male Wistar white rats (Rattus norvegicus) were sampled. Accumulation of Test Animals, Moringa Flower Extract Preparation, Phytochemical Screening / Secondary Metabolite Test, Treatment Process (200, 400, and mg/kgBW of Moringa flower extract), Wound Observation, Histopathological Skin Preparations, and Histopathological Examination of Preparations were the research procedures. Window data was analyzed using SPSS 25.0 and One Way ANOVA with a 95% confidence level (p &lt; 0.05). The Post Hoc Test with LSD was used for further analysis. The research results found Moringa flower extract at 200mg/Kg BW, 400mg/KgBW, and 600mg/KgBW could expedite wound healing and improve collagen density in obese mice's dermapen wounds. The 600mg dose was most beneficial. Research conclusions: Moringa flower extract aids dermapen wound healing, collagenization, and obesity weight loss, according to a study.

POTENCY OF OCIMUM SANCTUM L EXTRACT OINTMENT ON THE MANAGEMENT PARTIAL-THICKNESS PALPEBRAL LACERATION, IS IT A NOVELTY TO BE CONSIDERED AS AN ALTERNATIVE THERAPY?: REVIEW ON HISTOPATHOLOGICAL CHANGES IN EXPERIMENTAL MODEL

Introduction and Objective: Ocimum sanctum extract (OSE) has been shown to have antioxidant, anti-inflammatory, and antibacterial properties which are responsible for wound healing. This study aims to analyze the effect of 5%, 10%, and 15% OSE ointment on changes in collagen density and fibroblast count in Wistar rats with partial-thickness palpebral laceration. Methods: Laboratory experimental study with randomized post-test only control group design. The subjects were 30 rats divided into 5 groups: The placebo; Gentamicin 0.3%; OSE 5%; OSE 10% and OSE 15% group. The intervention was administered 2 times/day for 4 days. On the fifth day, the palpebral tissue was obtained to make histopathological preparations and to observe the collagen density and fibroblast count. Data were analyzed using Mann-Whitney, One-Way ANOVA, Kruskal Wallis, and Tamhane test. A p-value &lt;0.05 was assigned to the statistically significant. Results: The placebo group revealed loose collagen tissue and OSE 15% and gentamicin group revealed dense collagen tissue. The low fibroblast count was in the gentamicin and OSE 15% group and the highest fibroblast count was in the placebo group with a mean of 19.77±3.29; 22,67±0,73 and 60.9±10.66 cells, respectively. There was a significant difference (p&lt;0.001) in the collagen density and the fibroblast count based on the treatment groups. Posthoc tests revealed differences in the collagen density level and the fibroblast count between the placebo and the whole group. Conclusion: There was a significant difference between placebo and all groups of collagen density changes and fibroblast count. POTENCY OF OCIMUM SANCTUM EXTRACT L OINTMENT ON THE MANAGEMENT PARTIAL-THICKNESS PALPEBRAL LACERATION, IS IT A NOVELTY TO BE CONSIDERED AS AN ALTERNATIVE THERAPY?: REVIEW ON HISTOPATHOLOGICAL CHANGES IN EXPERIMENTAL MODEL

Abstract&#x0D; Introduction &amp; Objectives : Ocimum sanctum extract (OSE) has been shown to have antioxidant, anti-inflammatory, and antibacterial properties which are responsible for wound healing. This study aims to analyze the effect of 5%, 10%, and 15% OSE ointment on changes in collagen density and fibroblast count in Wistar rats with partial-thickness palpebral laceration.&#x0D; Methods : Laboratory experimental study with randomized post-test only control group design. The subjects were 30 rats divided into 5 groups: The placebo; Gentamicin 0.3%; OSE 5%; OSE 10% and OSE 15% group. The intervention was administered 2 times/day for 4 days. On the fifth day, the eyelid tissue was obtained to make histopathological preparations and to observe the collagen density and fibroblast count. Data were analyzed using Mann-Whitney, One-Way ANOVA, Kruskal Wallis, and Tamhane test. A p-value &lt;0.05 was assigned to the statistically significant.&#x0D; Results : The placebo group revealed loose collagen tissue and OSE 15% and gentamicin group revealed dense collagen tissue. The low fibroblast count was in the gentamicin and OSE 15% group and the highest fibroblast count was in the placebo group with a mean of 19.77±3.29; 22,67±0,73 and 60.9±10.66 cells, respectively. There was a significant difference (p&lt;0.001) in the collagen density and the fibroblast count based on the treatment groups. Posthoc tests revealed differences in the collagen density level and the fibroblast count between the placebo and the whole group.&#x0D; Conclusion : There was a significant difference between placebo and all groups of collagen density changes and fibroblast count. There was no difference between OSE 15% and gentamicin both microscopically and statistically.

Gastric Histopathological Image of Winstar Rats Induced by Ethanol After Red Ginger Extract Administration

Background: Alcohol can cause damage to the gastric mucosal barrier and reverse diffusion of HCL which results in damage to gastric tissue. The flavonoids in red ginger act as exogenous antioxidants which can ward off free radicals which reduce damage to the gastric mucosa caused by alcohol.Objective: To determine the differences in the histopathology of the rats gastric induced by 40% ethanol after administration of red ginger (Zingiber officinale var. rubrum) extract at graded doses.Methods: A post-test only group design method. 25 rats samples were divided into 5 groups: K(-) standard feed; K(+) ethanol 40% 1.8ml/200g/day; P1 red ginger extract 250mg/KgBW; P2 red ginger extract 500mg/KgBW; and P3 red ginger extract 750mg/KgBW. After 60 minutes, 40% ethanol was given at a dose of 1.8ml/200g/day. Treatment was carried out for 30 days, then on day 31 termination was carried out and gastric histopathology preparations were made with hematoxylin-eosin staining. Calculation of hull damage scores using the Barthel-Manja criteria. Analysis of differences in gastric mucosal damage between groups using the Kruskal Wallis Test and Mann Withney Test.Results: The difference between groups obtained significant results (p=0.003). There were significant differences between the K- and K+ groups (p=0.016), the K- and P1 groups (p=0.032), between the K- and P2 groups (p=0.032), between the K+ and P3 groups (p =0.032). Conclusion: Administration of red ginger extract at a dose of 750mg/KgBW was proven to be effective in preventing damage to the gastric mucosa induced by 40% ethanol.

HISTOPATOLOGICAL CHANGES OF WHITE RATS' SKIN CAUSED BY APPLICATION OF MIMOSIN FROM SIMPLISIA OF THE LAMTORO LEAF

Mimosine is one of the substances contained in lamtoro (Leucaena leucocephala) leaves. The toxic effect of mimosine on livestock is highly dependent on the concentration of mimosine in feed ingredients and the length of time livestock consume feed high in mimosine. This study aims to determine the histopathology of white rat skin exposed to mimosine compounds with different doses. This study used male white rats with Wistar strain, 2 months old and 300-350 g body weight. The 20 rats used were divided into four treatment groups, namely P0 (negative control, P1 (positive control, given standard mimosine 5 mg/head/day), P2 (given lamtoro leaf simplisia at a dose of 50 mg/head/day orally), P3 (given lamtoro leaf simplisia at a dose of 150 mg/head/day). On the 15th day of necropsy, the skin organs were taken and fixed using 10% NBF. After the skin organs were fixed, histopathology preparations were made using HE staining. Histopathologic examination was performed including three lesion variables: hair follicle necrosis, congestion, and inflammation. The severity of the lesions was scored as 0, 1, 2, and 3 for normal, mild, moderate and severe lesions, respectively. Data were then analyzed using Kruskal-Wallis and Mann-Whitney non-parametric tests. The results showed that exposure to mimosine from lamtoro leaf simplisia did not cause hair loss in experimental animals as it does in cattle. However, exposure to mimosine in P1 (5 mg/head/day) and P3 (150 mg/head/day) caused histopathological skin lesions in the form of congestion, necrosis, and mild inflammation. The results showed that exposure to mimosine from lamtoro leaf simplisia did not cause hair loss in experimental animals as it does in cattle. The conclusion of the study is that mimosine compounds from lamtoro leaf simplisia cause histopathological changes in white rat skin tissue, especially congestion lesions. However, there was no difference in the effect of mimosine administration from lamtoro leaf simplisia between a dose of 50 mg/head/day and a dose of 150 mg/head/day on the histopathology of rat skin tissue. Further research needs to be done on the effect of exposure to mimosine from lamtoro leaf simplisia with higher concentrations.

&lt;b&gt;Effect of curcumin nanoparticles on the number of preantral and antral follicles of white rats (&lt;i&gt;Rattus norvegicus&lt;/i&gt;) exposed to carbon black&lt;/b&gt;

Background: Continuous exposure to carbon black could affect the reproductive health of female animals, especially in the process of folliculogenesis. Curcumin nanoparticles are expected to maintain reproductive organ fertility through exposure to pollution containing carbon. Aim: The purpose of this research is to investigate the effect of curcumin nanoparticles on the number of preantral and antral ovarian follicles in white rats (Rattus norvegicus) exposed to carbon black. Methods: This study used 30 female rats divided into five groups. The negative (K-) and positive (K+) control groups were given aquades + Na-CMC 0.5% orally, while the treatment group received different doses of curcumin nanoparticles, namely in the P1 (50 mg/kgBB), P2 (100 mg/kgBB), and P3 (150 mg/kgBB) groups, respectively, via oral administration. After that, K+, P1, P2, and P3 were exposed to carbon black with a concentration of 1064 mg/m3 for 6 hours a day for 30 days. The ovarian organ would then be made histopathological preparations to examine the number of preantral and antral follicles with HE staining. Results: The results showed a significant difference (p&lt;0.05). The average number of preantral follicles in the K- group (27.17 ± 6.37), K+ (10.33 ± 6.22), P1 (26.17 ± 5.98), P2 (19.17 ± 3.71), and P3 (23.50 ± 10.36) (p &lt; 0.05) along with the average number of antral follicles in the K- group (18.50 ± 8.89), K+ (9.17 ± 2.14), P1 (17.67 ± 7.45), P2 (16.00 ± 5.30), and P3 (9.50 ± 5.09). Conclusion: The conclusion of this study shows that curcumin nanoparticles with different doses could affect and maintain preantral and antral ovarian follicles in white rats exposed to carbon black. It was also found that the dose of the P1 group (50 mg/kgBB) had the highest effectiveness in maintaining the number of ovarian follicles. The results of this study demonstrate therapeutic potential of curcumin nanoparticles as an affordable drug.
