Quantum Chemistry-Based Prediction of Electron Ionization Mass Spectra for Environmental Chemicals.

There is a lack of experimental electron ionization high-resolution mass spectra available to assist compound identification. The in silico generation of mass spectra by quantum chemistry can aid annotation workflows, in particular to support the identification of compounds that lack experimental reference spectra, such as environmental chemicals. We present an open-source, semiautomated workflow for the in silico prediction of electron ionization high-resolution mass spectra at 70 eV based on the QCxMS software. The workflow was applied to predict the spectra of 367 environmental chemicals, and the accuracy was evaluated by comparison to experimental reference spectra acquired. The molecular flexibility, number of rotatable bonds, and number of electronegative atoms of a compound were negatively correlated with prediction accuracy. Few analytes are predicted to sufficient accuracy for the direct application of predicted spectra in spectral matching workflows (overall average score 428). The m/z values of the top 5 most abundant ions of predicted spectra rarely match ions in experimental spectra, evidencing the disconnect between simulated fragmentation pathways and empirical reaction mechanisms.

High-Resolution Electron Ionization Mass Spectrometry of Stannane: Deconvolution of Superimposed Fragmentation Patterns.

In a pulsed laser plasma driven extreme ultraviolet (EUV) light, tin droplets undergo evaporation, eventually depositing on different surfaces. The removal of surface bound tin is commonly achieved with a hydrogen plasma, resulting in the formation of stannane (SnH4). The mechanisms leading to the formation and decomposition of stannane remain incompletely understood. To analyze these mechanisms mass spectrometrically, a reference is crucial, necessitating a high-resolution and thoroughly characterized mass spectrum of stannane. In this paper, a high-resolution 70 eV electron ionization (EI) mass spectrum of stannane is presented. The mass spectrum comprises all ten natural isotopes of the stannane fragments generated through EI. Utilizing the custom analysis program RASP, the relative distribution of fragments is calculated from the isotopically superimposed mass signals, offering crucial insights into the occurring processes. Furthermore, the dependence of fragment formation on ion source pressure and temperature was determined.

Antibacterial activity of five Indonesian medicinal plants and the isolation of compounds from Plectranthus scutellarioides

The purpose of this study is to investigate the antibacterial properties of five Indonesian medicinal plant extracts, namely Physalis angulata, Loranthus parasiticus, Plectranthus scutellarioides, Cyperus rotundus, and Terminalia catappa. The isolation of compounds from P. scutellarioides against pathogenic bacteria was also investigated. The assessment of the antibacterial activity of the extracts was conducted using the microdilution-3-(4,5-dimethylthiazol- 2-yl)-2,5-diphenyltetrazolium bromide method against Gram-positive bacteria (Bacillus subtilis and Staphylococcus aureus) and Gram-negative bacteria (Escherichia coli). The plant extract with the highest antibacterial activity was further purified using various chromatographic assays to isolate bioactive compounds. High Resolution-Electron Ionization Mass Spectroscopy/Electrospray Ionization Mass Spectroscopy (HR-ESIMS/EIMS) and Nuclear Magnetic Resonance (NMR) spectrometers were used to elucidate the structure of isolated compounds. Among all the plants tested, P. scutellarioides showed the strongest antibacterial activity against S. aureus [minimum inhibitory concentration (MIC 100 μg/ml)] and B. subtilis (MIC 200 μg/ml). Three compounds were isolated from P. scutellarioides including an abietane diterpene, 2,16-diacetoxy-6,11,12,14,17-pentahydroxy-abieta-5,8,11,13- tetraene-7-one (1), 5,6,7,3′,4′,5′-hexamethoxy flavone (2), and 5,6,7,8,3′,4′,5′-heptamethoxy flavone (3). Compounds 2 and 3 were recognized to be first isolated from this plant. Compound 1 exerted antibacterial activity against S. aureus with a MIC value of 60 μM, but was inactive against B. subtilis and E. coli (MIC > 200 μM). Compounds 2 and 3 were inactive against S. aureus, B. subtilis, and E. coli (MIC > 200 μM).

Identification of 1,2,4-triazole-bearing Bis-Schiff base hybrid scaffolds: In vitro and in silico insights to develop promising anti-urease and anti-cancer agents

The present work describes the synthetic procedure of 1,2,4-triazole derivatives (1–14) synthesized efficiently in a single step reaction. Basic skeleton confirmation was achieved through different characterization approaches, such as nuclear magnetic resonance(1HNMR and13CNMR) and high-resolution electron ionization mass spectrometry(HREI-MS). In vitro biological assessment of these scaffolds as anti-urease and anti-cancer agents was analyzed in presence of their standard compounds thiourea (IC50 = 9.30 ± 0.20 µM)and Tetrandrine (IC50 = 11.70 ± 0.10 µM), respectively. Inhibition ranges were observed for all scaffolds with IC50 = 2.10 ± 0.20–––37.40 ± 1.10 µM and IC50 = 4.10 ± 0.10–––39.90 ± 0.20 µM, respectively. Among the screened, the potent behavior were shown by scaffolds 3 (IC50 = 2.10 ± 0.20 and 4.10 ± 0.10 µM), 4(IC50 = 3.20 ± 1.10 and 5.20 ± 0.20 µM), 5(IC50 = 4.20 ± 0.30 and 5.60 ± 0.10 µM), 13(IC50 = 5.80 ± 1.10 and 5.90 ± 0.10 µM)and 14(IC50 = 4.30 ± 1.20 and 6.10 ± 1.20 µM).These analogs show the excellent inhibition owing to the spellbinding reactivity of –CF3, −F, −Cl and –OH moieties, which decelerate enzyme potential via effective binding between drug molecule and enzyme. These compounds were further investigated by means of molecular docking studies which explore the binding interaction of ligand within the binding pockets of enzymes. In this regard, excellent results were found in both the activity profiles of these scaffolds. Additionally, ADMET investigations also strengthen the drug profile of the potent compounds.

Exploring Phytochemical Composition and In Vivo Anti-Inflammatory Potential of Grape Seed Oil from an Alternative Source after Traditional Fermentation Processes: Implications for Phytotherapy.

This study aimed to analyze the composition of grape seed oil (GSO) derived from an alternative source after traditional fermentation processes and its potential anti-inflammatory effects using an in vivo model of carrageenan-induced inflammation in mice. Gas chromatography high-resolution electron ionization mass spectrometry (GC-HR-EIMS) analysis identified eight main components in the GSO extract, including myristic acid methyl ester, palmitoleic acid methyl ester, methyl isoheptadecanoate, cis-linoleic acid, oleic acid methyl ester, linoleic acid stereoisomer, linoleic acid ethyl ester, and methyl (6E, 9E, 12E, 15E)-docose-6,9,12,15-tetraenoate. No significant differences were observed in the main fatty acids between commercially available grape seed oil and GSO extract obtained from fermented grape seeds. In the carrageenan-induced inflammation model, treatment with GSO resulted in a significant reduction in paw edema at 180 min, as in the reduction observed with diclofenac treatment. Combined treatment with GSO and diclofenac showed enhanced anti-inflammatory effects. Additionally, GSO exhibited antioxidative effects by decreasing the levels of glutathione (GSH) and malondialdehyde (MDA) in the serum. Chronic treatment with GSO for ten days did not provide a protective effect on inflammation. These findings suggest that GSO could be used as an alternative raw material and could possess anti-inflammatory and antioxidative properties. Further studies are needed to explore its potential therapeutic applications.

New pyrrolopyridine-based thiazolotriazolesas diabetics inhibitors: enzymatic kinetics andin silico study.

Aim: To synthesizepyrrolopyridine-based thiazolotriazoles as a novel class ofα-amylase and α-glucosidase inhibitors and to determine their enzymatic kinetics. Methodology: Pyrrolopyridine-based thiazolotriazole analogs (1-24) were synthesized and characterized through proton nuclear magnetic resonance, carbon-13nuclear magnetic resonance andhigh-resolution electron ionization mass spectrometry. Results: All synthesized analogs displayed good inhibitory potential of α-amylase and α-glucosidase ranging 17.65-70.7μM and 18.15-71.97μM, respectively, compared with the reference drug, acarbose (11.98μM and 12.79μM). Analog 3 was the most potent among thesynthesized analogs, having α-amylase and α-glucosidase inhibitory activity at17.65 and 18.15μM, respectively. Thestructure-activity relationship and binding modes of interactions between selected analogs were confirmed via docking and enzymatic kinetics studies. The compounds (1-24) were tested for cytotoxicity against the3T3 mouse fibroblast cell line and wereobserved to benontoxic.

Identification of complementary McLafferty rearrangement ions at m/z 222 and 224 of methylstearate using high-resolution electron ionization time-of-flight mass spectrometer

All fragment ions observed in high-resolution electron ionization (EI) mass spectra of methyl stearate have been completely classified into three different series, such as methyl ester (CnHmO2), aliphatic hydrocarbon (CnHm) and acylium (CnHmO) ions. From the EI mass spectra classified into aliphatic hydrocarbon series, complementary McLafferty rearrangement (compMcLR) ion at m/z 224 and complementary McLafferty rearrangement+1 (compMcLR+1) ion at m/z 222 were identified from the standpoints of notable intensity and rearrangement reactions. The fragmentation of the keto- and enol-form molecular ions M.+ was discussed on the basis of the sites of unpaired electron (radical) and positive charge, which were determined by the spin density evaluated by density functional theory (DFT). The structure of M.+ determined by DFT rationally indicated that the compMcLR and compMcLR+1 ions at m/z 224 and 222 are produced by charge migration, while the McLR and McLR+1 ions at m/z 74 and 75 are produced by charge retention. The difference in the peak abundance of m/z 74 and 224 ions were evaluated from the difference in the ionization energy of the corresponding neutral fragments, based on Stevenson's rule.

Origin of Decomposition in a Family of Molybdenum Precursor Compounds.

The bis(tert-butylimido)-molybdenum(VI) framework has been used successfully in the design of vapor-phase precursors for molybdenum-containing thin films, so understanding its thermal behavior is important for such applications. Here, we report the thermal decomposition mechanism for a series of volatile bis(alkylimido)-dichloromolybdenum(VI) adducts with neutral N,N'-chelating ligands, to probe the stability and decomposition pathways for these molecules. The alkyl groups explored were tert-butyl, tert-pentyl, 1-adamantyl, and a cyclic imido (from 2,5-dimethylhexane-2,5-diamine). We also report the synthesis of the new tert-octyl imido adducts, (tOctN)2MoCl2·L (L = N,N,N',N'-tetramethylethylenediamine or 2,2'-bipyridine), which have been fully characterized by spectroscopic techniques as well as single-crystal X-ray diffraction and thermal analysis. We found that the decomposition of all compounds follows the same general pathway, proceeding first by the dissociation of the chelating ligand to give the coordinatively unsaturated species (RN)2MoCl2. Subsequent dimerization results in either an imido bridged adduct, [(RN)Mo(μ-NR)Cl2]2, or a chloride bridged adduct, [(RN)2Mo(μ-Cl)Cl]2, depending on the size of the R group. The dimeric species then likely undergoes an intramolecular γ-hydrogen transfer to yield a nitrido-amido adduct, (RHN)MoNCl2, and an alkene. Ultimately, the resulting molybdenum species appears to decompose into free tert-alkylamine and Mo2N or Mo2C. The thermolysis reactions have been monitored using 1H NMR spectroscopy, and the volatile decomposition products were analyzed using gas chromatography-mass spectrometry. A key intermediate has also been detected using electron ionization high-resolution mass spectrometry. Finally, a detailed computational investigation supports the mechanism outlined above and helps explain the relative stabilities of different N,N'-chelated bis(alkylimido)-dichloromolybdenum(VI) adducts.

Chemical constituents and their anti-neuroinflammatory activities from the bark of Taiwan incense cedar, Calocedrus formosana

One undescribed C40 terpenoid, calomacroquinoic acid; four undescribed diterpenes, 5α,6α-epoxy-7α-hydroxyferruginol, 15-ethoxysugiol, 7-methoxy-6,7-secoabieta-8,11,13-triene-6,12-diol, and ethyl 7,8-secoabieta-11,14-dioxo-7-ate; two compounds isolated from Nature for the first time, 6β,7α-dihydroxyferruginol and 12-O-methyltaxochinon; and six known compounds were successfully identified from the bark of Taiwan incense cedar Calocedrus formosana. Structures of all isolates were elucidated by physical data (appearance, ultraviolet, infrared, specific rotation, and X-ray) and spectroscopic data (1D and 2D nuclear magnetic resonance, and high-resolution electron ionization mass spectrometry). The biosynthetic pathway of calomacroquinoic acid is also described in the current study. Nitric oxide production in lipopolysaccharide (LPS)-stimulated BV-2 microglia cells was inhibited by 6,7-dehydroferruginol, 7α,11-dihydroxy-12-methoxy-8,11,13-abietriene, and trans-communic acid. Altogether, the bark of C. formosana possessed several potential natural therapeutics against inflammation-related neuronal diseases.

Antioxidative-, Antimicrobial-, Enzyme Inhibition-, and Cytotoxicity-Based Fractionation and Isolation of Active Components from Monotheca buxifolia (Falc.) A. DC. Stem Extracts.

The current study elaborates the pharmacological potential of the methanolic extract and its fractions of the stems of Monotheca buxifolia based on thin-layer chromatography and column chromatography analyses, exploiting biological and phytochemical assays. The results suggest that bioassay-guided isolation and fractionation led to the accumulation of biologically active components in the most active fractions that resulted in the isolation of different compounds. Structural elucidation of the purified compounds was accomplished using spectroscopic one-dimensional (1H, 13C) and two-dimensional NMR (heteronuclear multiple quantum coherence, heteronuclear multiple bond coherence, and correlation spectroscopy) and spectrometric (electron ionization mass spectrometry and high-resolution electron ionization mass spectrometry) techniques. The n-hexane, CHCl3, and EtAOc fractions led to the isolation of lupeol from different fractions. 1-Triacontanol was also isolated from the n-hexane fraction, while benzoic acid, methyl benzoate, ursolic acid, and 3-hydroxybenzoic acid were obtained from the EtOAc fraction. The compounds depicted good-to-moderate total antioxidative potential and total reducing power activity and significant free-radical scavenging activity. All the compounds showed significant urease and lipase inhibitory activity with poor-to-moderate amylase inhibition. Significant zone of inhibition was observed against different bacterial strains by the isolated compounds. This work therefore states that bioassay-guided isolation plays a vital role in the isolation of biologically active constituents that can be exploited for drug development.

Anti-Hyperglycemic Activities, Molecular Docking and Structure-Activity Relationships (SARs) Studies of Endiandric Acids and Kingianins from Endiandra kingiana

Diabetes has become a severe chronic disease worldwide with patients significantly increasing daily. Due to the side effects of insulin and oral hypoglycaemic agents employed in diabetes treatment, scientists are working hard to develop alternative approaches from natural plants that inhibit α-amylase and α-glucosidase. Consequently, by performing a phytochemical analysis on the bark of Endiandra kingiana, the present study isolated 11 cyclic polyketides. Analyses with one-dimensional and two-dimensional nuclear magnetic resonance (1D- and 2D-NMR), highresolution electron ionization mass spectrometry (HRESIMS), and comparison with previous literature confirmed the compounds characteristics. Subsequently, the compounds were screened for in vitro α-amylase and α-glucosidase inhibiting activities. Compounds 9 and 2 exhibited potent inhibition towards α-amylase at 0.0008903 ± 0.5 and 0.02 ± 0.3 mg mL−1 of half-maximal inhibitory concentration (IC50) values, respectively. In the α-glucosidase inhibition assay, compounds 10 and 5 demonstrated good inhibition with IC50 values of 0.11 ± 0.08 and 0.14 ± 0.05 mg mL−1, respectively. The molecular docking examination demonstrated that the compounds adhered to the active sites on the C-terminal of the human pancreatic α-amylase (Protein Data Bank Identification (PDB ID): 2QV4, resolution: 1.97 Å) and maltase-glucoamylase (MGAM) (PDB ID: 3TOP, resolution: 2.88 Å), agreeing with α-amylase and α-glucosidase enzymes inhibitory reactions.

Development and application of an in-house library and workflow for gas chromatography–electron ionization–accurate-mass/high-resolution mass spectrometry screening of environmental samples

This work presents an optimized gas chromatography–electron ionization–high-resolution mass spectrometry (GC-EI-HRMS) screening method. Different method parameters affecting data processing with the Agilent Unknowns Analysis SureMass deconvolution software were optimized in order to achieve the best compromise between false positives and false negatives. To this end, an accurate-mass library of 26 model compounds was created. Then, five replicates of mussel extracts were spiked with a mixture of these 26 compounds at two concentration levels (10 and 100 ng/g dry weight in mussel, 50 and 500 ng/mL in extract) and injected in the GC-EI-HRMS system. The results of these experiments showed that accurate mass tolerance and pure weight factor (combination of reverse-forward library search) are the most critical factors. The validation of the developed method afforded screening detection limits in the 2.5–5 ng range for passive sampler extracts and 1–2 ng/g for mussel sample extracts, and limits of quantification in the 0.6–3.2 ng and 0.1–1.8 ng/g range, for the same type of samples, respectively, for 17 model analytes. Once the method was optimized, an accurate-mass HRMS library, containing retention indexes, with ca. 355 spectra of derivatized and non-derivatized compounds was generated. This library (freely available at https://doi.org/10.5281/zenodo.5647960), together with a modified Agilent Pesticides Library of over 800 compounds, was applied to the screening of passive samplers, both of polydimethylsiloxane and polar chemical integrative samplers (POCIS), and mussel samples collected in Galicia (NW Spain), where a total of 75 chemicals could be identified.

Towards understanding of electrolyte degradation in lithium-mediated non-aqueous electrochemical ammonia synthesis with gas chromatography-mass spectrometry.

Lithium-mediated electrochemical ammonia synthesis (LiMEAS) in non-aqueous media is a promising technique for efficient and green ammonia synthesis. Compared to the widely used Haber–Bosch process, the method reduces CO2 emissions to zero due to the application of green hydrogen. However, the non-aqueous medium encounters the alkali metal lithium and organic components at high negative potentials of electrolysis, which leads to formation of byproducts. To assess the environmental risk of this synthesis method, standardized analytical methods towards understanding of the degradation level and consequences are needed. Here we report on the implementation of an approach to analyze the liquid electrolytes after electrochemical ammonia synthesis via high-resolution gas chromatography-mass spectrometry (GCMS). To characterize the molecular species formed after electrolysis, electron ionization high-resolution mass spectrometry (EI-MS) was applied. The fragmentation patterns enabled the elucidation of the mechanisms of byproduct formation. Several organic electrolytes were analyzed and compared both qualitatively and quantitatively to ascertain molecular composition and degradation products. It was found that the organic solvent in contact with metallic electrodeposited lithium induces solvent degradation, and the extent of this decomposition to different organic molecules depends on the organic solvent used. Our results show GCMS as a suitable technique for monitoring non-aqueous electrochemical ammonia synthesis in different organic electrolytes.

Investigation of Fragmentation Pathways of Fentanyl Analogues and Novel Synthetic Opioids by Electron Ionization High-Resolution Mass Spectrometry and Electrospray Ionization High-Resolution Tandem Mass Spectrometry.

The global drug market is characterized by the fast development of new psychoactive substances such as fentanyl analogues and novel synthetic opioids, the detection of which is complicated by the lack of appropriate quality control procedures and references. Herein, we analyze the fragmentation pathways and characteristic ions of 25 novel fentanyl analogues and 5 novel synthetic opioids by electron ionization (EI) and electrospray ionization (ESI) high-resolution mass spectrometry to provide a reference for the identification of these species. In the ESI mode, fentanyl analogues mainly undergo piperidine ring degradation, phenethyl and piperidine ring dissociation, and piperidine ring and amide moiety cleavage, while piperidine ring degradation and phenethyl and piperidine ring dissociation are the major pathways in the EI mode. The five novel synthetic opioids largely undergo amide group dissociation and N-cyclohexyl bond cleavage in the ESI mode. Thus, this work facilitates the detection and quantitation of fentanyl analogues and novel synthetic opioids or other substances with similar structures in forensic laboratories.

A new indole alkaloid and other constituents from Monodora minor and Uvaria tanzaniae: their antitrypanosomal and antiplasmodial evaluation

Phytochemical investigation of the methanolic extract of Monodora minor Engl. & Diels (Annonaceae) stem bark yielded a new indole (E)-4-(1H-indol-5-yl)-but-3-en-2-one (1), a known indole 5-formyl-1H-indole (2) and an ubiquitous steroid sitosterol (3). The investigations of the methanolic extract of Uvaria tanzaniae Verdc. (Annonaceae) root bark yielded two previously reported C-benzylated dihydrochalcones namely uvaretin (4) and diuvaretin (5). Structures of the isolated compounds were elucidated based on NMR spectroscopy and high resolution electron ionization mass spectrometry (HR-EI-MS) data. All compounds were tested against Trypanosoma brucei brucei and Plasmodium falciparum. At a single concentration (20 μM) in the antitrypanosomal and antiplasmodial assays, compound 4 exhibited remarkable activities against T. brucei brucei and P. falciparum with percentage inhibition of 97.3% and 83.0% respectively, whereas compounds 1, 2, 3 and 5 were inactive. In a dose response antiplasmodial assay compound 4 exhibited moderate activity against P. falciparum with an IC50 value of 7.20 μM.

A novel five-step synthetic route to 1,3,4-oxadiazole derivatives with potent α-glucosidase inhibitory potential and their in silico studies.

A series of new N-aryl/aralkyl derivatives of 2-methyl-2-{5-(4-chlorophenyl)-1,3,4-oxadiazole-2ylthiol}acetamide were synthesized by successive conversions of 4-chlorobenzoic acid (a) into ethyl 4-chlorobenzoate (1), 4-chlorobenzoylhydrazide (2) and 5-(4-chlorophenyl)-1,3,4-oxadiazole-2-thiol (3), respectively. The required array of compounds (6a-n) was obtained by the reaction of 1,3,4-oxadiazole (3) with various electrophiles (5a-n) in the presence of DMF (N,N-dimethylformamide) and sodium hydroxide at room temperature. The structural determination of these compounds was done by infrared, 1 H-NMR (nuclear magnetic resonance), 13 C-NMR, electron ionization mass spectrometry, and high-resolution electron ionization mass spectrometry analyses. All compounds were evaluated for their α-glucosidase inhibitory potential. Compounds 6a, 6c-e, 6g, and 6i were found to be promising inhibitors of α-glucosidase with IC50 values of 81.72 ± 1.18, 52.73 ± 1.16, 62.62 ± 1.15, 56.34 ± 1.17, 86.35 ± 1.17, 52.63 ± 1.16 µM, respectively. Molecular modeling and ADME (absorption, distribution, metabolism, excretion) predictions supported the findings. The current synthesized library of compounds was achieved by utilizing very common raw materials in such a way that the synthesized compounds may prove to be promising drug leads.

Synthesis, molecular modeling, in vivo study, and anticancer activity of 1,2,4-triazole containing hydrazide-hydrazones derived from (S)-naproxen.

A new series of 1,2,4-triazole containing hydrazide-hydrazones derived from (S)-naproxen ( 7a-m) was synthesized in this study. The structures of these compounds were characterized by spectral (Fourier-transform infrared spectroscopy, 1 H-nuclear magnetic resonance (NMR), 13 C-NMR, and high-resolution electron ionization mass spectrometry) methods. Furthermore, molecular modeling of these compounds was studied on human methionine aminopeptidase-2. All synthesized compounds were screened for anticancer activity against three prostate cancer cell lines (PC3, DU-145, and LNCaP) using the 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium colorimetric method. Compound 7a showed the best activity against the PC3, DU-145 and LNCaP cancer cell lines with IC50 values of 26.0, 34.5, and 48.8 μM, respectively. Compounds 7b, 7k, and 7m showed anticancer activity against cancer cell lines PC3 and DU-145 with IC50 values of 43.0, 36.5, 29.3 μM and 49.8, 49.1, 31.6 μM, respectively. Compounds 7f and 7g showed anticancer activity against PC3 cells with IC50 values of 43.4 and 34.5 μM, respectively. To assess the biodistribution in mice of IRDye800, dye-labeled compound 7a or 100 μM of free dye was injected intravenously into the mice's tail. In vivo images were taken with in vivo imaging system spectrum device at 60, 120, 180, 240, 300, and 360 min after injection. At the end of 360 min, ex vivo studies were carried out to determine in which organs the dye was accumulated in the urogenital system. Ex vivo studies showed that the accumulation of compound 7a in the prostate is greater than that of the free dye, and it is concluded that compound 7a may be promising for the treatment of prostate cancer.

Isolation of a spirolactone norditerpenoid as a yeast Ca2+ signal transduction inhibitor from Kuji amber and evaluation of its effects on PPM1A activity

A different type of biologically active compound from Kuji amber (Late Cretaceous, Japan) before the K-Pg boundary [65 million years ago (Ma)] was isolated based on the growth-restoring activity of a mutant yeast involving Ca2+ signal transduction. It was identified as a spirolactone norditerpenoid, (4R*, 5S*, 8R*, 9R*, 10S*)-14,15,16,19-tetranor-labdan-13,9-olide (1) from spectral analyses with high-resolution electron ionization mass spectrometry (HREIMS), 1D and 2D nuclear magnetic resonance (NMR). Although the planar structure of 1 is known as an artificial derivative from marrubiin, it was isolated as a natural product from Kuji amber and its structure was elucidated for the first time. It had a growth-restoring activity against the mutant yeast through the direct or indirect inhibition of calcineurin activity [protein phosphatase, Mg2+/Mn2+-dependent 1A (PPM1A) activation]. Furthermore, the compound had potent inhibitory effect against the degranulation of rat basophilic leukemia 2H3 (RBL-2H3) cells.

Mass Spectrometry Study of New Polyamine Derivatives of Caffeine

The mass spectrometric fragmentations of ten new 8-alkylaminocaffeine derivatives were investigated. The fragmentation pathways of new polyamine derivatives of caffeine on the basis of low and high-resolution electron ionization (EI) mass spectra were discussed. In the case of new compounds, classical fragmentation of purine skeleton related to the elimination of a neutral molecule CH3N(1)C(2)O from a molecular ion was not observed. Nevertheless, new, interesting fragmentation of described caffeine derivatives was observed. Moreover, heats of formation of odd-electron ions of 8-alkylaminocaffeine derivatives were calculated.

Isolation of squarrosal and squarrosol compounds from methanol root extract of <i>Ruellia squarrosa</i> (Acanthaceae)

Purpose : To identify, characterize and structurally elucidate bioactive compounds from root of Ruellia squarrosa. Methods : One kilogram of crude Ruellia squarrosa root was shade dried for 14 days, ground to a fine powder and subjected to a methanol extraction. The resultant extract underwent column chromatography for further purification. The isolated compounds were subjected to ultraviolet spectroscopy (UV), infra-red (IR), proton nuclear magnetic resonance ( 1 H-NMR), 13 C–NMR and high resolution electron ionization mass spectrometry (HR-EI-MS) for the identification, characterization and structural elucidation of bioactive compounds. The most active compounds were tested for anticancer activities against human prostate cancer cell. Results : Two active compounds, squarrosol and squarrosal, were obtained with half-maximal inhibitory concentration (IC 50 ) of 15.6 and 26.6 μg/mL, respectively, against human prostate cancer cell lines. Squarrasol showed a significantly (p < 0.05) greater inhibition of cell proliferation than the same dose of squarrasal. Conclusion : These findings suggest that extracts of Ruellia squarrosa containing the bioactive compounds, squarrosol and squarrosal, can potentially be developed for the treatment of human prostate cancer.
