Platform headspace gas chromatography method for high-throughput determination of residual solvents in pharmaceutical materials

Static headspace capillary gas chromatography (HSGC) has been employed to monitor the level of residual solvents in the pharmaceutical materials. Most of the HSGC methods, however, consume significant amounts of diluents and require considerable amount of sample preparation time. Accordingly, a HSGC method featured with fast turnaround time, and minimal amount of solvent use has been developed for the quantitative analysis of 27 residual solvents frequently used in the development and manufacturing processes of pharmaceutical industry. This HSGC-FID method employs a commercially available fused silica capillary column, a split injection (40:1), and a programmed temperature ramp. It was qualified for specificity, accuracy, repeatability/precision, linearity, LOQ, solution stability, and robustness using two representative sample matrices. The standards, samples and spiked samples were demonstrated to be stable for at least 10 days at room temperature in sealed headspace vials with a recovery of ≥ 93%. The method was also shown to be robust, and its performance was not affected by small changes of carrier gas flow rate, initial oven temperature or the headspace oven temperature. In this new approach, the analytical sample was prepared by dissolving the sample into 1 mL of the diluent and the standard solution was prepared by diluting 1 mL of the custom-made stock into 9 mL of the diluent whereas the traditional approach requires liters of the diluent, making the new approach environmentally friendly, sustainable, economical, agile, error-proofing and thus appropriate for a variety of pharmaceutical applications.

Simultaneous quantitation of water and residual solvents in pharmaceuticals by rapid headspace gas chromatography with thermal conductivity detection (GC-TCD)

In the pharmaceutical industry, an array of analytical testing is performed to demonstrate the safety and efficacy of drug substance and drug products. Among the most critical attributes of release testing are quantitation of residual solvents from the manufacturing process, which pose toxicity concerns, and determination of water content, which can impact potency and shelf life. Residual solvent determination in pharmaceuticals is most commonly performed using headspace capillary gas chromatography (GC) with flame ionization detection (FID), a robust technique that incorporates a mode of detection noteworthy for its sensitivity and wide dynamic range. However, FID responds exclusively to combustible organic species, and does not produce any signal for common gases such as carbon dioxide, ammonia, and notably water. While thermal conductivity detection (TCD) is an alternate, universal mode of detection that has a known response to all GC-appropriate compounds, including water, its use among pharmaceutical companies is uncommon due to the ubiquity of the more sensitive FID and the availability of other techniques for water quantitation such as Karl Fischer titrations (KF). In this work, the use of headspace GC-TCD was successfully demonstrated for the development of a 7.5-minute method for simultaneous quantitation of water, over 25 common residual solvents, and other volatile impurities in small molecule pharmaceutical samples. By carefully controlling sample preparation to minimize the impact of residual water from the diluent, the results for residual solvents and water obtained by this technique were found to be comparable to those of GC-FID and KF, respectively. Headspace GC-TCD improves the throughput of drug testing by greatly reducing the need for KF testing and associated expensive reagents, and helps to conserve samples that are often limited in early stages of development. The technique has desired sensitivity, precision, accuracy and linear dynamic range suitable for pharmaceutical analysis.

Improvement of beer flavour with extruded rice as adjunct

Extruded rice used as adjunct for beer fermentation was prepared using a single-screw extruder. Extrusion pretreatment facilitated the saccharification and gelatinization of the rice starch as well as the formation of a glassy cellular structure, which was evaluated through scanning electron microscopy analysis. The effects of different parameters, including barrel temperature, water content, nozzle diameter and screw speed, as well as their interactions with rice expansion rate, were evaluated by response surface methodology to determine the optimum extrusion conditions. Extruded rice was acquired using the optimum extrusion parameters (water content, barrel temperature, screw speed and nozzle diameter of 22.4%, 103°C, 8 mm and 191.6 rpm, respectively), and the expansion rate reached 275.1%. The effect of the extruded rice adjunct on beer flavour compounds was investigated by comparing flavour compounds with those of traditional rice-adjunct beer through sensitive static headspace–capillary gas chromatography spectrometry. The contents of esters, a highly important flavour group, in extruded-rice-adjunct beer (ethyl acetate, isoamyl acetate, ethyl hexanoate and ethyl caprylate at 11.6, 2.4, 0.2 and 0.3 mg L−1, respectively) were higher than those of the traditional rice-adjunct beer. The contents of high alcohols were lower in the traditional rice-adjunct beer than those in the extruded-rice-adjunct beer, but both levels satisfied the national standard for beer. Gas chromatography–mass spectrometry was employed to identify the flavour compounds of both beer samples through headspace–solid-phase microextraction. Simple Isotope Dilution Headspace-GC-MS Analysis of Naphthalene and p-Dichlorobenzene in Whole Blood and Urine

A very simple and sensitive method for the simultaneous analysis of naphthalene and p-dichlorobenzene in human whole blood and urine by headspace capillary gas chromatography-mass spectrometry (GC-MS) is presented. The advantages of the method were that as much as 1 mL of headspace vapor could be injected into a GC port in the splitless mode, and that the addition of deuterated naphthalene and p-dichlorobenzene as internal standards resulted in much better headspace extraction efficiencies, which resulted in high sensitivity. The detection limits for both naphthalene and p-dichlorobenzene were 1 ng mL(-1) for whole blood and 0.5 ng mL(-1) for urine. Validation data, such as the linearity of calibration curves, reproducibility and recovery rates, were all satisfactory. Using this method, both compounds could actually be detected from whole blood samples of a male volunteer after the inhalation of each gas of the compounds.

Simple analysis of naphthalene in human whole blood and urine by headspace capillary gas chromatography with large-volume injection

A very simple method for analysis of naphthalene in human whole blood and urine by headspace gas chromatography (GC) is presented. It does not require solid-phase microextraction or cryogenic trapping devices, but needs only a conventional capillary GC instrument with flame ionization detection. The advantage of the method is that as much as 5 ml of headspace vapor can be injected into a GC instrument in splitless mode for sensitive detection. After heating a diluted whole blood or urine sample containing naphthalene and 1-methylnaphthalene (internal standard, IS) in a 7.0-ml vial at 80°C for 30 min, 5 ml of the headspace vapor was drawn with a glass syringe and injected into the gas chromatograph. Before injection, the column temperature was set at 50°C to trap the analytes, and then the oven temperature was programmed up to 300°C. Sharp peaks were obtained for both analyte and IS, and only a few impurity peaks appeared, which did not interfere with the test peaks, mainly for whole blood samples. The detection limit (signal-to-noise ratio. 3) were about 0.05 and 0.01 μg/ml for whole blood and urine, respectively. Precision and linearity were also examined to confirm the reliability. Such a simple headspace GC technique with large-volume injection will be applicable to other low-volatility compounds in biological matrices, and will be useful in forensic toxicological analysis.

Analysis of organic volatile impurities in pharmaceutical excipients by static headspace capillary gas chromatography

A systematic approach for the identification and quantification of organic volatile impurities (OVIs) in pharmaceutical excipients is described. Analytical procedures utilizing static headspace capillary gas chromatography coupled with flame-ionization and MS detection techniques were developed for the analysis of toxic ICH class 1 solvents and US Pharmacopeia OVIs at sub-ppm levels, and commonly used organic solvents in a wide range of concentrations. Chromatographic conditions and headspace parameters for the methods were optimized for separation, sensitivity, and speed. The proposed methodologies were demonstrated to be selective, accurate, and reproducible, and were successfully applied to the rapid screening of OVIs in typical excipients.

Simple Analysis of Amphetamines in Human Whole Blood and Urine by Headspace Capillary Gas Chromatography with Large Volume Injection

A very simple method for the analysis of methamphetamine and amphetamine in human whole blood and urine by headspace gas chromatography (GC) has been presented. It neither needs solid-phase microextraction nor cryogenic trapping devices, but only a conventional capillary GC instrument with flame ionization detection (FID). The two special points to be mentioned in this method are the in-matrix derivatization of amphetamines for vaporization and the capability of injection of as large as 5 mL of the headspace vapor into a GC instrument in the splitless mode for sensitive detection. After heating a whole blood or urine sample containing amphetamines, α-methylbenzylamine (internal standard, IS) and heptafluoro-n-butyryl chloride under alkaline conditions in a 7.0-mL vial at 90 °C for 20 min, 5 mL of the headspace vapor was drawn with a glass syringe and injected into the gas chromatograph. During injection the column was at 40 °C to trap the analytes, and then the oven temperature was programmed up to 320 °C. Sharp peaks were obtained for each analyte and IS, and only a relatively small number of background impurity peaks for the whole blood and urine samples. The detection limits for each amphetamine were estimated to be 0.1 μg mL−1 for whole blood and 0.03 μg mL−1 for urine. Precision and linearity were also tested to confirm the reliability. Methamphetamine and amphetamine could be determined from whole blood and urine obtained at autopsy in three methamphetamine poisoning cases. The identity of each peak appearing in the gas chromatograms was confirmed by GC/mass spectrometry.

A gas chromatographic method for the indirect determination of hydroxylamine in pharmaceutical preparations: conversion into nitrous oxide

A simple, sensitive, and selective headspace gas-chromatographic method has been developed for measuring hydroxylamine (HA) in a variety of sample matrices including pharmaceutical formulations. This procedure relies on converting HA into nitrous oxide (N 2O), which is a single-step reaction that is carried out directly in a heated headspace vial. The gaseous products are then analyzed by headspace capillary gas chromatography. Several detection strategies were evaluated and electron capture provided the best sensitivity (4 parts-per-billion (ppb)) while the mass selective and thermal conductivity values were higher (14 ppb and 1.4 parts-per-million (ppm), respectively). The method's linear dynamic range spans two to four decades with a run-to-run precision that was better than 5% R.S.D. ( n=7). The reagent concentrations (oxidant, buffer) strongly impact the N 2O signal and the greatest response was obtained for solutions that contained equimolar amounts of reactants. HA was efficiently (98%) recovered from a sample matrix that contained only the active pharmaceutical ingredient (API) but the recovery was lower (83%) when excipients were present.

Sensitive Analysis of Alkyl Alcohols as Decomposition Products of Alkyl Nitrites in Human Whole Blood and Urine by Headspace Capillary GC with Cryogenic Oven Trapping

The abuse of alkyl nitrites is becoming a serious social problem worldwide. In this report, a simple and sensitive method is presented for the determination of n-butyl alcohol, isobutyl alcohol, and isoamyl alcohol as decomposition products of alkyl nitrites in human whole blood and urine samples using capillary gas chromatography (GC) with cryogenic oven trapping. After heating a whole blood or urine sample containing each alkyl alcohol and t-butyl alcohol [the internal standard (IS)] in a 7-mL vial at 55 degrees C for 15 min, 5 mL of the headspace vapor is drawn into a gas-tight syringe and injected into a GC inlet port. The vapor is introduced into an Rtx-BAC2 medium-bore capillary column in the splitless mode at 0 degrees C oven temperature in order to trap the entire analytes, and then the oven temperature is programmed up to 240 degrees C for the GC measurements by flame ionization detection. These conditions give sharp peaks for each compound and the IS and low background noise for whole blood or urine samples. The detection limits of the analytes are 10 ng/mL for whole blood and 5 ng/mL for urine. Linearity and precision are also tested to confirm the reliability of this method. Isobutyl alcohol and methemoglobin could be determined from the whole blood samples of three male volunteers who had sniffed isobutyl nitrite.

Sensitive determination of styrene and related compounds in human body fluids by headspace capillary gas chromatography with cryogenic oven trapping

A simple and sensitive method is presented for determination of styrene, toluene, ethylbenzene, isopropylbenzene andn-propylbenzene in human body fluids by capillary gas chromatography (GC) with cryogenic oven trapping. After heating a blood or urine sample containing each compound andp-diethylbenzene (internal standard, IS) in a 7.0-mL vial at 60°C for 20 min, 5 mL of headspace vapor was drawn into a glass syringe and injected into a GC. All vapor was introduced into an Rtx-Volatile middle bore capillary column in splitless mode at oven temperature of 20°C to trap entire analytes, and the oven temperature then programmed to 280°C for GC measurements by flame ionization detection. The present conditions gave sharp peaks of each compound and IS, and low background noises for whole blood or urine samples.

Sensitive Determination of n-Hexane and Cyclohexane in Human Body Fluids by Capillary Gas Chromatography with Cryogenic Oven Trapping

A sensitive method was developed for determination of n-hexane and cyclohexane in human body fluids by headspace capillary gas chromatography (GC) with cryogenic oven trapping. Whole blood and urine samples containing n-hexane and cyclohexane were heated in a 7.5 mL vial at 70 degrees C for 15 min, and 5 mL of the headspace vapor was drawn into a glass syringe. All vapor was introduced through an injection port of a GC instrument in the splitless mode into an Rtx-Volatiles middle-bore capillary column at an oven temperature of -40 degrees C for trapping volatile compounds. The oven temperature was programmed to 180 degrees C for GC with flame ionization detection. These conditions gave sharp peaks for both n-hexane and cyclohexane, a good separation of each peak, and low background impurities for whole blood and urine. The extraction efficiencies of n-hexane and cyclohexane were 13.2-30.3% for whole blood and 12.7-20.7% for urine. The coefficients of within-day variation in terms of extraction efficiency of both compounds were 5.0-9.5% for whole blood and 3.8-10.8% for urine; those of day-to-day variation for the compounds were not greater than 16.6%. The regression equations for n-hexane and cyclohexane showed good linearity in the range of 5-500 ng/0.5 mL for whole blood and urine. The detection limits (signal-to-noise ratio = 3) for both compounds were 1.2 and 0.5 ng/0.5 mL for whole blood and urine, respectively. The data on n-hexane or cyclohexane in rat blood after inhalation of each compound are also presented.

Determination of perfluorobutane in rat blood by automatic headspace capillary gas chromatography and selected ion monitoring mass spectrometry.

A new contrast agent (Sonazoid; NC100100) for ultrasound imaging has been developed. It is an aqueous suspension of lipid stabilised perfluorobutane (PFB) gas microbubbles. An automatic headspace capillary gas-chromatographic mass spectrometric method using electron impact ionisation was developed for analysis of Sonazoid PFB in rat blood. The calibration standards were gaseous PFB dissolved in ethanol in the range of 0.5-5000 ng PFB. Fluorotrichloromethane (CFC 11) was used as an internal standard of the method and the MS detector was set to single ion monitoring of the base fragment ions of PFB (m/z 69 and 119) and CFC 11 (m/z 101). The calibration graph, made by plotting the peak area ratios of PFB (m/z 69) to CFC 11(m/z 101) against the amount of PFB, was fitted to a second-order polynomial equation with weighting 1/y2 and found to be reproducible. The limit of quantification of the method was set to 0.4 ng PFB. The between-day variation of the method was below 9.2% relative standard deviation (RSD) and the within-day variation of the method was below 7.6% RSD. The accuracy of the method, as compared to Coulter counter, was estimated by determination of PFB in samples where Sonazoid was added to saline and found to range from 91.5% to 105.2%. PFB, added as Sonazoid, was found to be stable for at least 7 months in rat blood samples when stored at -20 degrees C.

New methods for determination of 2-butoxyethanol, butoxyacetaldehyde and butoxyacetic acid in aqueous systems, with special reference to cell culture conditions.

Ethylene glycol ethers, especially 2-ethoxyethanol and 2-butoxyethanol (BE) are frequently used in industry and household as solvents and detergents because of their excellent hydrophilic and lipophilic properties. BE and its oxidation products, butoxyacetaldehyde (BAL) and butoxyacetic acid (BAA), are mainly associated with haemolytic toxicity. No method to determine BAL in aqueous systems (e.g. urine or blood) has been published up to now. BAL was synthesized by dehydration of BE and identified by gas chromatography-mass spectrometry. For determination of BAL and BE with head space-capillary gas chromatography, water and HCl or sodium dihydrogen phosphate were added to the sample. No further extraction or derivatization were necessary. For BAA determination after adding HCl and sodium dihydrogen phosphate the samples were extracted with ethyl acetate and derivatized with 2,2,2-trichloroethanol/HCl. The analytical methods presented here are reliable, sensitive and rapid. The new methods were developed for mammalian cell culture systems, because such in vitro systems are especially useful for metabolic studies and have the advantage of choosing species and organ specificity. In the cell culture experiments presented here it was demonstrated that Opossum kidney cells are able to metabolize BAL to BAA within 24 h. After this interval, in the cells neither BAL nor BAA were accumulated, whereas BAA was found in the cell culture media.

Ultra-sensitive method for determination of ethanol in whole blood by headspace capillary gas chromatography with cryogenic oven trapping

We have established an ultra-sensitive method for determination of ethanol in whole blood by headspace capillary gas chromatography (GC) with cryogenic oven trapping. After heating a blood sample containing ethanol and isobutyl alcohol (internal standard, IS) in a 7.0-ml vial at 55°C for 15 min, 5 ml of the headspace vapor was drawn into a glass syringe and injected into a GC port. All vapor was introduced into an Rtx-BAC2 wide-bore capillary column in the splitless mode at −60°C oven temperature to trap entire analytes, and then the oven temperature was programmed up to 240°C for GC measurements with flame ionization detection. The present method gave sharp peaks of ethanol and IS, and low background noise for whole blood samples. The mean partition into the gaseous phase for ethanol and IS was 3.06±0.733 and 8.33±2.19%, respectively. The calibration curves showed linearity in the range 0.02–5.0 μg/ml whole blood. The detection limit was estimated to be 0.01 μg/ml. The coefficients of intra-day and inter-day variation for spiked ethanol were 8.72 and 9.47%, respectively. Because of the extremely high sensitivity, we could measure low levels of endogenous ethanol in whole blood of subjects without drinking. The concentration of endogenous ethanol measured for 10 subjects under uncontrolled conditions varied from 0 to 0.377 μg/ml (mean, 0.180 μg/ml). Data on the diurnal changes of endogenous ethanol in whole blood of five subjects under strict food control are also presented; they are in accordance with the idea that endogenous blood ethanol is of enteric bacterial origin.

A very thick film open-tubular trap for headspace capillary gas chromatography

An open-tubular trap with a very thick film was studied for “full on-line” headspace capillary gas chromatography, and was used as an enrichment trap for headspace sampling. The preparation of the open-tubular column with up to an 80 μm thick film, by combining both static and dynamic coating, is described in detail. The break-through volume for the open-tubular trap of different film thickness, as the loading capacity for HS sampling, does not increase linearly in proportion to the film thickness. A study of the difference between a packed adsorbent trap and an open-tubular trap shows that HS capillary GC with open-tubular trap loses less of the retention capacity, has better resolution ability and higher recovery. By independently and rapidly heating the open-tubular trap, the GC peaks may have higher resolution. Examples of successful analyses, by the opentubular trap with “full on-line” HS capillary GC, include trace aromatic hydrocarbon (1 ppm-100 ppm) in polybutadiene-styrene rubber and trace alcohol and acrylic esters (10 ppm–1000 ppm) in polyacrylate hydrous emulsion.

Study on volatile components in salami by reverse carrier gas headspace gas chromatography–mass spectrometry

Salami are a typical seasoned sausage of Italy; a number of types are produced, according to local traditional recipes. As industrial production has taken place, a number of problems rise in obtaining products similar to the traditional ones. The use of selected microbial starters is permitted by Italian law for some years and at present, micro biological research is engaged in selecting starters similar to the ones isolated from traditional products, with the aim of obtaining organoleptic characteristics close to the ones of traditional recipes. A study was carried out concerning the characterisation of volatile components of salami by headspace capillary gas chromatography–mass spectrometry. As during the sampling step, analytes could reach the analytical column, the carrier gas rate was back flushed in the latter, while a pre column was used as cold trap. Then GC–MS analysis follows. By these techniques, we were able to highlight typical profiles of different salami, as well as monitoring the ripening of a traditional and a starter added salami. Main peaks are of fermentative origin, while also peaks from spices were detected. Ethyl propionate was used as internal standard to be able to normalise the peaks amounts.

Determination of physiological levels of volatile organic compounds in blood using static headspace capillary gas chromatography with serial triple detection.

A static capillary gas chromatographic method using three different detectors [photoionization detector (PID), electron capture detector (ECD) and flame ionization detector (FID)] switched in series is presented for the determination of volatile organic compounds (VOCs) in sub microgram l-1 levels. The method was applied for the analysis of selected environmentally and occupationally relevant non-halogenated and chlorinated aromatic hydrocarbons (e.g., benzene, toluene, xylenes, dichlorobenzenes) as well as chlorinated aliphatic hydrocarbons (e.g., trichloroethene, tetrachloroethene) in blood samples. Detailed investigations, in respect to the figures of merit were carried out. For most of the selected VOCs detection limits (calculated as the three-fold standard deviation of low level calibration standards) in the range from 26 (benzene) to 67 ng l-1 (m/p-xylene) were achieved which are comparable with those reported for dynamic headspace techniques in combination with mass spectrometric detection. For the individual VOCs the within-series precision varied from 4 to 19% and the day-to-day precision from 11 to 28%. Regarding PID as well as FID the calibration graphs for all substances were linear up to at least 10 micrograms l-1 while the ECD response was linear up to concentrations of about 0.6 microgram l-1 for the halogenated compounds. Our method is applicable for the quantitative determination of VOCs in blood in the occupationally as well as in the physiologically relevant (normal) concentration range.

Influence of Heat Treatment on the Volatile Compounds of Milk

In this work dynamic headspace capillary gas chromatography coupled with multivariate statistical techniques was applied to distinguish milks subjected to different heat treatments (pasteurization, direct ultrahigh-temperature method, and “in-bottle” sterilization). Results were obtained by taking into account a reduced, though significant, number of volatile compounds (acetone, 2-butanone, 2-pentanone, 2-heptanone, 3-methylbutanal, pentanal, hexanal, heptanal, dimethyl disulfide, toluene, and limonene). Moreover, the concentration of these volatile compounds, especially aldehydes and ketones, provided discriminant information about storage time and storage temperature in both whole and partially skimmed UHT milk. Complementarity of purge-and-trap and head-space capillary gas chromatographic methods for determination of methyl-tert.-butyl ether in water

Abstract An improved method has been developed for the quantitative determination of traces of methyl-tert.-butyl ether (MTBE) over a large range of concentrations in water samples based on two complementary techniques: head-space capillary gas chromatography and purge-and-trap gas chromatography. The interesting aspect of this study lies in the complementary performances of these two methods, each in its specific range, for the analysis of MTBE in water samples. Head-space gas chromatography was used as a screening technique in routine analyses, while purge-and-trap gas chromatography was used as a confirmation method for the negative results. The analytical precision was good with both methods. Detection limits were 50 μg 1−1 for head-space gas chromatography and 2 μg 1−1 for purge-and-trap gas chromatography, with 90% calculated recovery. Choice of different parameters for the purge-and-trap method is discussed.
