Simulation of Summer Groundnut for Yield and Yield Attributing Traits Using CROPGRO-Peanut Model

Field experiments were carried out at College farm, B. A. College of Agriculture, Anand Agricultural University, Anand. The DSSAT v 4.6 CROPGRO-Peanut model was used to predict the phenology of groundnut crop under combinations of three sowing dates and four groundnut cultivars. The model was calibrated with a 2015 dataset of growth and phenological parameters for estimating the genetic coefficients of all four cultivar and was validated with a 2016 dataset of the same parameters. Result found that the model was able to reasonably simulate the pod yield, kernel yield and haulm yield with per cent error ranging (± 10.06) between observed and simulated value for all cultivars under different sowing dates Simulations of yield and yield attributing characters using the calibrated model were found to be quite accurate.

A comprehensive phenotypic and genotypic evaluation of Spanish groundnuts from diverse crosses to identify superior and stable donors for fresh seed dormancy

Breeding high yielding groundnut cultivars with 2–3 weeks of fresh seed dormancy, particularly in Spanish-type cultivars, enhances the sustainability of agriculture in groundnuts. In this context, we conducted a comprehensive phenotypic and genotypic evaluation of advanced breeding lines developed in the genetic background of Spanish types. By employing multi-phenotyping and marker data, we identified PBS 15044, 16004, 16013, 16015, 16016, 16017, 16020, 16021, 16026, 16031, 16035, 16037, 16038, 16039, 16041, and 16042 with 2–3 weeks dormancy (> 90%).The various parametric and non-parametric estimates identified the stable fresh dormant genotypes with one or more superior economic trait. PBS 16021, 15044, 16038, and 16039 identified with high hundred pod weight (HPW) were also reported having high intensity of dormancy (> 90% for up to 3 weeks); PBS 15044, 16016, PBS 16038 and PBS 16039 with high hundred kernel weight (HKW) also reported with up to 3 weeks fresh seed dormancy; and PBS 16013, 16031, and 16038 with up to 3 weeks fresh seed dormancy had high shelling percentage (SP). They can be used to develop lines with the desired level of dormancy, and high yields, by designing appropriate breeding strategies.

Impact of seedbed types and scarification methods on germination percentage and growth performance of Bambara groundnut (Vigna subterranea (L.) Verdc.) in the Sudan Savanna zone of Nigeria

The field experiments were conducted during the 2021 and 2022 wet seasons at The National Institute for Horticultural Research, Bagauda, and Hadejia-Jama’are River Basin Development Authority, Wudil, both in Kano State within the Sudan Savannah agro-ecological zone of Nigeria. The study aimed to find a better seedbed arrangement and the best scarification methods for good germination and growth of Bambara groundnut cultivars. The treatments consisted of four seedbed types (ridge, ridge+earthing, flat, and flat+earthing), four scarification methods (zero, mechanical, acid, and hot water treatments), and three cultivars (cream, black, and speckled). The treatments were factorially combined and laid out in a split-split plot design with three replications. Seedbed was assigned to the main plot, scarification methods were assigned to the subplot, and cultivars were assigned to the sub-subplot. Data collected were days to first emergence, establishment count (%), canopy height (cm), and kernel yield (kg ha-1). The result indicated that mechanically scarified Bambara groundnut seeds resulted in a higher establishment count and uniform canopy height (cm), which led to more dry matter accumulation and a higher kernel yield (kg ha-1).

Bacteria-mediated green synthesis of silver nanoparticles and their antifungal potentials against Aspergillus flavus.

The best biocontroller Bacillus subtilis produced silver nanoparticles (AgNPs) with a spherical form and a 62 nm size through green synthesis. Using UV-vis spectroscopy, PSA, and zeta potential analysis, scanning electron microscopy, and Fourier transform infrared spectroscopy, the properties of synthesized silver nanoparticles were determined. Silver nanoparticles were tested for their antifungicidal efficacy against the most virulent isolate of the Aspergillus flavus fungus, JAM-JKB-BHA-GG20, and among the 10 different treatments, the treatment T6 [PDA + 1 ml of NP (19: 1)] + Pathogen was shown to be extremely significant (82.53%). TG-51 and GG-22 were found to be the most sensitive groundnut varieties after 5 and 10 days of LC-MS QTOF infection when 25 different groundnut varieties were screened using the most toxic Aspergillus flavus isolate JAM- JKB-BHA-GG20, respectively. In this research, the most susceptible groundnut cultivar, designated GG-22, was tested. Because less aflatoxin (1651.15 g.kg-1) was observed, treatment T8 (Seed + Pathogen + 2 ml silver nanoparticles) was determined to be much more effective. The treated samples were examined by Inductively Coupled Plasma Mass Spectrometry for the detection of metal ions and the fungicide carbendazim. Ag particles (0.8 g/g-1) and the fungicide carbendazim (0.025 g/g-1) were found during Inductively Coupled Plasma Mass Spectrometry analysis below detectable levels. To protect plants against the invasion of fungal pathogens, environmentally friendly green silver nanoparticle antagonists with antifungal properties were able to prevent the synthesis of mycotoxin by up to 82.53%.

Effects of Different Levels of Gamma Radiation on Growth and Yield Characteristics of Groundnut

One of the key strategy for improving the crops is plant mutation breeding, which generates a large germplasm pool to improve crop characteristics. In this field experiment, the effects of different levels of gamma radiation on the growth and yield characteristics of groundnut were investigated. "Gamma chamber 1200 Cobalt-60" research irradiator was used to irradiate the groundnut seeds. Two experiments were conducted, with the first exposing the seeds to gamma radiation levels ranging from 0 to 100 Gy, and the second exposing them to levels from 0 to 500 Gy. Irradiated seeds were sown in poly bags and kept under shade. After 2 weeks, seedlings were transplanted in an open field using a Randomized Complete Block Design with five replications. Growth and yield parameters were collected, and statistical analysis was carried out using SAS 9.1 software, with treatment means compared using DMRT at 0.05 level. Results from experiment one showed no significant (P ≥ 0.05) differences among treatments. However, in experiment two, treatments showed significant differences in the measured variables. Seeds exposed to 200 Gy had increased shoot fresh weight (123.3%), root fresh weight (69.8%), number of pods (46%), 100-seed weight (47.7%), and total yield (65.6%). The study recommends 200 Gy as a more suitable level of gamma radiation to create desirable characteristics in groundnut. In conclusion, gamma radiation can be an effective tool for inducing mutations in plants for crop improvement. The findings of this radiation study could contribute to the improvement of new cultivars of groundnut with desirable traits, such as increased yield and improved plant growth.

Identification of Donors for Fresh Seed Dormancy and Marker Validation in a Diverse Groundnut Mini-Core Collection

Domestication and extensive selection in the development of modern, high-yielding commercial groundnut cultivars have resulted in the selection of an undesirable trait known as in situ germination, which is also referred to as the pre-harvest sprouting of seeds. This is particularly prevalent in regions where humid weather coincides with the harvest season. Delayed harvesting and pre-sprouting can cause production losses and increase the chances of aflatoxin contamination, thereby impeding the quality and kernel yield. Breeding early maturing groundnut cultivars with 2–3 weeks of fresh seed dormancy, particularly in Spanish-type cultivars, enhances the sustainability of agriculture. In this context, we conducted a comprehensive evaluation of a groundnut mini-core collection, a major resource for genetic diversity, for fresh seed dormancy using an in vitro germination assay for two seasons, viz., rainy 2022 and post-rainy 2022–2023 at ICRISAT (Hyderabad). To enhance the effectiveness and accuracy of traditional breeding methods via the use of markers for marker-assisted selection, we performed molecular screening of the mini-core accessions using two allele-specific markers. The GMFSD1 marker was successfully validated by effectively differentiating dormant and non-dormant genotypes. By employing phenotypic and marker data, we identified a set of accessions, viz., ICG 5827 (Virginia Runner), ICG 11457 (Virginia Runner), ICG 7000 (Virginia Bunch), and ICG 11322 (Virginia Bunch) of sub spp. hypogaea var. hypogaea and ICG 9809 (Spanish Bunch) of sub spp. fastigiata var. vulgaris that exhibited a fresh seed dormancy period ranging from 2 to 3 weeks. These identified accessions hold potential as donors in breeding programs that are designed to address the groundnut production needs in various cropping systems across different countries. The validated marker, particularly GMFSD1, demonstrated considerable potential for facilitating faster breeding of groundnut cultivars with the desired dormancy using marker-assisted selection. This research provides a foundation to expediting groundnut breeding programs and offers opportunities to mitigate pre-harvest sprouting, ultimately improving seed quality and productivity in groundnut-producing regions.

Prevalence of groundnut dry root rot (Macrophomina phaseolina (Tassi) Goid.) and its pathogenic variability in Southern India.

Macrophomina phaseolina is the most devastating and emerging threat to groundnut production in India. An increase in average temperature and inconsistent rainfalls resulting from changing climatic conditions are strongly believed to aggravate the disease and cause severe yield losses. The present study aims to conduct a holistic survey to assess the prevalence and incidence of dry root rot of groundnut in major groundnut growing regions of Southern India, viz., Andhra Pradesh, Telangana, Karnataka, and Tamil Nadu. Furthermore, the pathogenic variability was determined using different assays such as morphological, cultural, pathogenic, and molecular assays. Results indicate that disease incidence in surveyed locations ranged from 8.06 to 20.61%. Both temperature and rainfall played a major role in increasing the disease incidence. The pathogenic variability of M. phaseolina isolates differed significantly, based on the percent disease incidence induced on cultivars of JL-24 groundnut and K-6 groundnut. Morphological variations in terms of growth pattern, culture color, sclerotia number, and sclerotia size were observed. The molecular characterization of M. phaseolina isolates done by ITS rDNA region using ITS1 and ITS4 primers yielded approximately 600 bp PCR amplicons, sequenced and deposited in GenBank (NCBI). Molecular variability analysis using SSR primers indicated the genetic variation among the isolates collected from different states. The present investigation revealed significant variations in pathogenic variability among isolates of M. phaseolina and these may be considered important in disease management and the development of resistant cultivars against groundnut dry root rot disease.

EFFECT OF LEGUME FODDER ON RUMEN DEGRADATION OF SORGHUM FODDER BY SHEEP

In an agro-pastoral setting where land is a constraint to crop and livestock production, improvement in cereal and legumes for their food value will need to be evaluated for their corresponding improvement for feed. The objective of the study was to examine the effect of legume fodder on degradability of sorghum fodder from improved cereal and legume cultivars. In Sacco rumen degradation technique was used and cultivars of groundnut (samnut23 and samnut26), cowpea (X-IITA) and sorghum (DEKO) were evaluated and examined. Mixed proportions of cereal and legumes as 60:40, 50:50 & 40:60 were ruminally degraded by Yankasa sheep. Results from the experiment unveiled mixture of 50:50 (Deko: samnut26) had better proximate composition, while mixture of 50:50 (Deko:X-IITA) was better in fiber composition. The Potential Degradabilities of both mixtures were 66.94% and 54.46% effectively degraded, respectively. It can be concluded that mixture of 50:50 (Deko: Samnut26) and mixture of 50:50 (Deko:X-IITA) facilitates the effective degradation of sorghum fodder. It is therefore recommended that the cultivars be promoted to farmers and the residue from Deko, haulms from Samnut23 and Samnut26; and X-IITA be fed to sheep at 50:50 for proper residue utilization.

Elucidating Late Leaf Spot Disease Progression and Resistance Components in Different Groundnut (Arachis hypogaea L.) Cultivars towards Phaeoisariopsis personata

Late Leaf Spot (LLS) disease is caused by Phaeoisariopsis personata, a devastating disease that significantly affects groundnut (Arachis hypogaea L.) production worldwide. This research aimed to investigate the disease progress in resistant and susceptible groundnut genotypes under green house conditions with artificial inoculations. The four popularly growing groundnut cultivars in Andhra Pradesh i.e., Kadiri-6 (K6), Dharani, Harithandra and Lepakshi (K-1812) were evaluated in this study. The disease severity scale, percent disease index (PDI), Area under disease progression curve (AUDPC) and the epidemic rate (rate) were estimated by using the linear model. Highest disease severity was observed in K6 (88.19 %) and Dharani (85.19 %) with greater disease progression rate was observed in K6 (0.13) and Dharani (0.12) cultivars. However, the lowest disease progression was observed in Lepakshi (0.04) and Harithandhra (0.08) cultivars. Enhanced resistance to LLS was reported with Harithandhra and Lepakshi cultivars due to slower epidemic rate, longer incubation and latent periods with smaller lesions.

Integrating data from asymmetric multi-models can identify drought-resistant groundnut genotypes for drought hot-spot locations

Water/drought stress experiments are frequently conducted under imposed stress or rainout shelters, while natural drought hot-spot investigations are rare. The “drought hot spot” in Anantapur, Andhra Pradesh, India, is appropriate for drought stress evaluation due to its hot, arid environment, limited rainfall, with over 50% rainfall variability. According to reports, 30 out of 200 groundnut cultivars in India are supposed to possess drought-tolerant characteristics. However, these cultivars are yet to be evaluated in areas that are prone to drought. This study tested these drought-tolerant genotypes in naturally drought-prone areas of Anantapur under rainfed conditions from Kharif 2017 to 2019. Pod yield and rainfall-use-efficiency (RUE) were measured for these genotypes. Genotype and genotype*environment interactions affected pod yield and RUE (GEI). The AMMI model exhibits significant season-to-season variability within the same area with environmental vectors > 90° angles. GGE biplot suggested the 2018 wet season for drought-resistant cultivar identification. Kadiri5 and GPBD5 were the most drought-tolerant cultivars for cultivation in Anantapur and adjacent regions. These types could also be used to generate drought-tolerant groundnut variants for drought-prone regions.

Effect of planting method and spacing on the growth parameters of three Bambara groundnut cultivars in two Agro-climatic zones of Sierra Leone

Field trials were conducted in two agro-climatic zones of the country (Lungi and Kabala) to evaluate the effect of planting methods and inter-row spacing on the growth parameters of three Bambara groundnut cultivars. The experiment consisted of a factorial 18 treatment combinations of two planting methods (mound and flat), three inter-row spacing (50 cm x 10 cm, 50 cm x 15 cm, and 50 cm x 20 cm), and three Bambara groundnut cultivars (Lubam1, Lubam 2, and Kabam 1). The study shows that planting Bambara groundnut using mounds produced higher values of the assessed growth parameters compared to planting Bambara groundnut on flat land. The result further shows that the wider inter-row spacing (50 cm x 20 cm) on average recorded the highest value for all the assessed growth parameters. Furthermore, Kabam1 was observed to have recorded the highest value for all the growth parameters in both locations. Thus, for optimum growth parameters to be achieved, it is recommended that Kabam1 should be planted on mound using the inter-row spacing of 50 cm x 20cm.

ATTOCK-2019: A HIGH-YIELDING AND DISEASE-TOLERANT PEANUT (ARACHIS HYPOGAEA L.) CULTIVAR FOR BARANI TRACT OF PUNJAB, PAKISTAN

Attock-2019 (11AK011) is an elite high-yielding, disease-tolerant, bunch-type (decumbent-2) groundnut (Arachis hypogaea L.) cultivar, developed at the Groundnut Research Station, Attock, Punjab, Pakistan. The cultivar resulted from the local crossing between two advanced lines, i.e., 01CG001 and 02CG005. The first cross attempt occurred during the 2005 cropping season at the Groundnut Research Station, Attock, and the selection underwent the pedigree method that continued up to the F6 generation. Attock-2019 underwent evaluation regarding its performance concerning yield, disease resistance, and adaptability to numerous yield trials like preliminary, regular, micro-, and national uniform yield trials, including evaluation at farmer fields and agronomic parameters, during 2012-2018, under the varietal code 11AK011. It out-yielded the check cultivar BARI-2011 in micro-yield trials for two consecutive years in Punjab by producing 19.52% and 17.96% yields higher than the said cultivar. Similarly, at the national level, during the national uniform groundnut yield trials in 2016 and 2017, it surpassed the check cultivars Golden and Pothowar by 30.74% (Golden) and 3.46% (Golden and Pothowar), respectively, in different ecological zones of Pakistan. This genotype is of medium duration, with 20%–25% three-seeded pods per plant. The average number of pods per plant ranged from 85–95. Its shelling percentage is high (70%–72%), with a good 100-seed weight (62-66 g). It also possessed good taste, oil content (40.5%), and protein content (21.6%). The cultivar also showed good performance under drought-stress conditions and tolerance to diseases like the Tikka disease and collar rot of groundnut. Attock-2019 can be sown from 1 April to 15 May under Barani conditions of Punjab, with 30:80:30 NPK kg ha-1 at sowing time and an application of 500 kg ha-1 gypsum at the flowering stage. Attock-2019 gained approval for general cultivation in the Punjab Barani tract during the 2021 in 54th Punjab Seed Council Meeting as the approving authority.

Morpho-physiological responses and expression analysis of MYB94 transcription factor gene in four groundnut cultivars under drought and salinity stress

Aim: The present study aimed to evaluate the impact of salinity stress on morpho-physiological parameters and relative expression of the MYB94 transcription factor gene among four widely cultivated groundnut cultivars. Methodology: A comparative analysis of morpho-physiological parameters was studied in four groundnut cultivars (Kadiri-Amaravati, Kadiri-Lepakshi, Kadiri-6, and Kadiri-9) subjected to different concentrations of NaCl prepared in half-strength Arnon’s nutrient growth medium. The relative expression analysis of a stress responsive transcription factor gene, MYB94 was studied in four groundnut cultivars subjected to drought and NaCl stress by qRT PCR. Results: NaCl stress resulted in a significant increase in proline content, MDA content in cultivar Kadiri-9 compared to other cultivar studied. On the contrary, plant growth, shoot dry weight, root dry weight, leaf relative water content, cell membrane injury decreased to lesser extent in cultivar Kadiri-9 than Kadiri-Amaravati, Kadiri-Lepakshi, Kadiri-6 under salinity stress. qRT-PCR based relative expression analysis of the MYB94 gene revealed a significant increase of transcript expression levels in cultivar Kadiri-9 compared to other three cultivars under both drought and salinity stress. Interpretation: Groundnut cultivar Kadiri-9 showed better tolerance to NaCl stress as evidenced from enhanced proline content, better relative water content and lesser extent of membrane damage when compared with other groundnut cultivars under given NaCl stress treatments. In addition, relative expression of MYB94 gene was significantly higher due to NaCl stress as compared to drought stress conditions in cultivar Kadiri-9 suggested that the MYB94 could be involved in salt stress tolerance. ​​Proximate Composition, Amino Acid and Fatty Acid Profiles of Eight Cultivars of Groundnut Grown in Nigeria

Background: Groundnut had been established to have great health benefits on account of its biochemical constituents. Methods: Eight cultivars of groundnut grown in Nigeria were analyzed using standard protocols for proximate compositions, amino acid and fatty acid profiles with the sole aim of selecting the most promising cultivars in terms of nutrient composition and oil stability. Result: The results indicated that crude protein was the highest in MK 373 and lesser in Samnut 22. Crude fat ranged from 44.68% in Samnut 21 to 49.98% in Samnut 23. The major amino acids were leucine, glutamic acid and aspartic acid. The cultivars Samnuts 21 and 22 showed higher essential amino acids (EAA), while higher non-essential amino acid (NEAA) was recorded in MK 373. The major fatty acids were palmitic, oleic and linoleic acids. Among the cultivars, Samnut 11 was the only cultivar with better oil quality on account of higher oleic and linoleic O/L and total poly unsaturated fatty acid and total saturated fatty acid (TPUS/TS) ratios that compared well with Codex/WHO standards. The results, in general, have shown that Samnuts 11, 21, 22 and MK 373 are the potentials cultivars that could be selected for breeding programme to improve the quality of groundnut in Nigeria.

Growth and Yield Performance of Four Groundnut Cultivars under Santa Maria, Ilocos Sur Conditions

The study was conducted to determine the growth and yield performance of four groundnut cultivars in the Experimental Farm at Santa Maria, Ilocos Sur. Significant difference between cultivars was also verified. It was laid out in Randomized Complete Block Design and replicated three times. Groundnut cultivars evaluated were Ilocos Pink, Asha and two landraces. All the data gathered were computed, tabulated and analysed using the analysis of variance (ANOVA) of Randomized Complete Block Design. Further test for the significant differences among treatments was subjected to Honest Significant Difference Test. Results showed that the agronomic and economic performances of four peanut varieties significantly different under the Santa Maria, Ilocos Sur condition. In terms of growth performance, Ilocos pink cultivar significantly better than the other cultivars. Ilocos Pink also gave the highest yield performance as manifested by number of seeds per pod, yield per hectare, net income and ROI.

EFFECT OF DIETHYL SULPHATE ON M1 ATTRIBUTES AND SDS-PAGE ANALYSIS IN GROUNDNUT (Arachis hypogaea L.)

The present study was carried out to know the effect of diethyl sulphate on groundnut cultivar TMV-7 particularly in M1 generation. The healthy and viable seeds of groundnut cultivar TMV-7 were exposed to different concentrations of diethyl sulphate (5 to 25 mM) and sown in the field. The various growth and morphological parameters like percentage of seed germination, survival percentage plant height, days to flowering, total height at time of harvest, root length and number of lateral roots, number of pods per plant and pod weight were decreased with increasing concentrations of diethyl sulphate. All the characteristics studied were inhibited when compared to control. The lower concentrations such as 5, 10 and 15 mM concentrations enhanced all the parameters. But there was a sudden decrease in 20 and 25 mM. The various concentrations of DES played a different role in the field condition itself. The shoots developed from different concentrations of DES showed variation in their number and height. The protein content also showed decreasing trend with increasing concentrations of DES. There was a steady increase of protein content up to 15 mM DES. After that, there was a drastic reduction in the protein content in 20 and 25 mM. From the diethyl sulphate treated seeds, the embryo axes were collected and subjected to SDS-PAGE analysis. The protein banding pattern also differentiated among various concentrations of DES. The protein band with 77.5 kDa was observed in 5 and 10mM treatments. Like that the 13.5 kDa was present in 10 and 25mM treatments. The 48.25 kDa recorded in 15 and 20 mM treatments.

Discovery of Major Quantitative Trait Loci and Candidate Genes for Fresh Seed Dormancy in Groundnut

Spanish bunch groundnut varieties occupy most of the cultivated area in Asia and Africa, and these varieties lack required 2-3 weeks of fresh seed dormancy (FSD) hampering kernel quality. Genomic breeding can help to improve commercial groundnut cultivars for FSD in a shorter time with greater precision. In this regard, a recombinant inbred line (RIL) population from the cross ICGV 02266 (non-dormant) × ICGV 97045 (dormant) was developed and genotyped with a 5 K mid-density genotyping assay. A linkage map was constructed with 325 SNP loci spanning a total map length of 2335.3 cM and five major QTLs were identified on chromosomes Ah01, Ah11, Ah06, Ah16 and Ah17. Based on differential gene expression using transcriptomic information from dormant (Tifrunner) and non-dormant (ICGV 91114) genotypes, histone deacetylases, histone-lysine N-methyltransferase, cytochrome P450, protein kinases, and ethylene-responsive transcription factor were identified as key regulators involved in the hormonal regulation of dormancy. Six Kompetitive Allele Specific PCR (KASP) markers were successfully validated in the diverse panel including selected RILs of the same population and germplasm lines. These validated KASP markers could facilitate faster breeding of new varieties with desired dormancy using marker-assisted early generation selection.

Altered expression levels of transcripts of GNAC TFs during drought stress in susceptible and tolerant cultivars of groundnut

By modulating gene expression, transcription factors (TFs) play a key role in conferring drought resistance. During drought, NAC, one of the major plant-specific transcription factor families, is found to be substantially up-regulated, indicating that it influences the gene(s) involved in drought tolerance. The goal of this study was to get insight into how the drought-tolerant wild type A. duranensis and the susceptible variety GJG 22 altered their expression levels of GNAC TFs during drought and after re-watering. Physio-biochemical parameters like relative water content electrolytic leakage, chlorophyll content, osmotic potential, and proline content were examined, along with this differential gene expression, with eleven GNAC TF water stress- related genes were evaluated in both types at different stages. In the drought-tolerant cultivars, nine TF(s) were upregulated, whereas, six were up-regulated in GJG 22. Five GNAC TFs in A. duranensis and four in GJG22 were elevated post revival. The influence of drought and revival on physio-biochemical parameters and the degree of expression of GNAC genes, as well as the impact on drought tolerance in plants, is revealed by this finding.
