Phase retrieval for X-ray differential phase contrast radiography with knowledge transfer learning from virtual differential absorption model

Grating-based X-ray phase contrast radiography and computed tomography (CT) are promising modalities for future medical applications. However, the ill-posed phase retrieval problem in X-ray phase contrast imaging has hindered its use for quantitative analysis in biomedical imaging. Deep learning has been proved as an effective tool for image retrieval. However, in practical grating-based X-ray phase contrast imaging system, acquiring the ground truth of phase to form image pairs is challenging, which poses a great obstacle for using deep leaning methods. Transfer learning is widely used to address the problem with knowledge inheritance from similar tasks. In the present research, we propose a virtual differential absorption model and generate a training dataset with differential absorption images and absorption images. The knowledge learned from the training is transferred to phase retrieval with transfer learning techniques. Numerical simulations and experiments both demonstrate its feasibility. Image quality of retrieved phase radiograph and phase CT slices is improved when compared with representative phase retrieval methods. We conclude that this method is helpful in both X-ray 2D and 3D imaging and may find its applications in X-ray phase contrast radiography and X-ray phase CT.

Single-shot grating-based X-ray phase contrast imaging via generative adversarial network

Talbot-Lau interferometry obtains X-ray differential phase contrast (DPC) signals of object by subtracting multiple moiré patterns acquired by phase-stepping (PS) procedure. Due to the need of multiple intensity measurements, the long measuring time is inevitable in the conventional DPC imaging, giving rise to X-ray dose and fluctuations. In this paper, we propose a single-shot X-ray phase contrast imaging (XPCI) method based on deep learning. Specifically, in hardware, we propose to replace the analysis absorption grating with the high-resolution X-ray detector system to avoid the illumination flux loss caused by the analysis grating. In software, we employ a U-net based generative adversarial network (GAN) for solving the phase reconstruction problem with single-shot intensity pattern. By examining the performance on a variety of simulated and experimental datasets, we demonstrate that our approach, in spite of only using single intensity pattern, is able to obtain results with high resolution and image contrast which is competitive with the conventional PS approach, while being less time-consuming and low-dose.

X-ray grating interferometry design for the 4D GRAPH-X system

The 4D GRAPH-X (Dynamic GRAting-based PHase contrast x-ray imaging) project aims at developing a prototype of an x-ray grating-based phase-contrast imaging scanner in a laboratory setting, which is based on the Moirè single-shot acquisition method in order to be optimized for analysing moving objects (in the specific case, a dynamic thorax phantom), that could evolve into a suitable tool for biomedical applications although it can be extended to other application fields. When designing an x-ray Talbot-Lau interferometer, high visibility and sensitivity are two important figures of merit, strictly related to the performance of the system in obtaining high quality phase contrast and dark-field images. Wave field simulations are performed to optimize the setup specifications and construct a high-resolution and high-sensitivity imaging system. In this work, the design of a dynamic imaging setup using a conventional milli-focus x-ray source is presented. Optimization by wave front simulations leads to a symmetric configuration with 5.25 μm pitch at third Talbot order and 45 keV design energy. The simulated visibility is about 22%. Results from GATE based Monte Carlo simulations show a 19% transmission percentage of the incoming beam into the detector after passing through all the gratings and the sample. Such results are promising in view of building a system optimized for dynamic imaging.

Visibility of Polychromatic Grating-Based X-Ray Imaging Systems Based on Wave-Optical Theories and Frequency Domain Analysis

Grating-based X-ray phase-contrast imaging and tomography, applicable with traditional polychromatic X-ray tubes, have shown great potential for future applications of imaging with multimodal information indicating materials and microstructures simultaneously. The parameters and performance of the grating system could be simulated by a wave-optical simulation framework and proved feasible for the design and optimization of both coherent and incoherent gratings systems. However, the simulation involves real-space point-wise calculation of the Fourier transformation, and the direct expression of the relationship of the parameters was absent. In this work, we analyzed the Fourier domain characteristics of the simulated system and the presented visibility of the system of different energies in an analytical form. The derived direct expression which omitted the simulation process was validated with results of both simulation and real experiments and may help future designs, optimizations and studies of the energy-resolved characteristics of the system.

Fourier-based interpretation and noise analysis of the moments of small-angle x-ray scattering in grating-based x-ray phase contrast imaging.

In grating-based x-ray phase contrast imaging, Fourier component analysis (FCA) is usually recognized as a gold standard to retrieve the contrasts including attenuation, phase and dark-field, since it is well-established on wave optics and is of high computational efficiency. Meanwhile, an alternative approach basing on the particle scattering theory is being developed and can provide similar contrasts with FCA by calculating multi-order moments of deconvolved small-angle x-ray scattering, so called as multi-order moment analysis (MMA). Although originated from quite different physics theories, the high consistency between the contrasts retrieved by FCA and MMA implies us that there may be some intrinsic connections between them, which has not been fully revealed to the best of our knowledge. In this work, we present a Fourier-based interpretation of MMA and conclude that the contrasts retrieved by MMA are actually the weighted compositions of Fourier coefficients, which means MMA delivers similar physical information as FCA. Based on the recognized cosine model, we also provide a truncated analytic MMA method, and its computational efficiency can be hundreds of times faster than the original deconvolution-based MMA method. Moreover, a noise analysis for our proposed truncated method is also conducted to further evaluate its performances. The results of numerical simulation and physical experiments support our analyses and conclusions.

Factors Affecting the Spatial Resolution in 2D Grating–Based X-Ray Phase Contrast Imaging

X-ray phase contrast imaging is a promising technique in X-ray biological microscopy, as it improves the contrast of images for materials with low electron density compared to traditional X-ray imaging. The spatial resolution is an important parameter to evaluate the image quality. In this paper, simulation of factors which may affect the spatial resolution in a typical 2D grating–based phase contrast imaging system is conducted. This simulation is based on scalar diffraction theory and the operator theory of imaging. Absorption, differential phase contrast, and dark-field images are retrieved via the Fourier transform method. Furthermore, the limitation of the grating-to-detector distance in the spatial harmonic method is discussed in detail.

Prototype system of noninterferometric phase-contrast computed tomography utilizing medical imaging components

Grating-based x-ray phase-contrast imaging has been demonstrated to provide more information and higher-contrast images for low-Z soft tissues, compared with conventional absorption-based imaging. However, the existing Talbot–Lau phase-contrast devices are operated in either a two- or three-dimensional mode at low energy with a small field of view and long exposure time. This is because of coherence limitations, difficulties in fabricating high aspect ratio gratings, and the slow readout speed of the detector. For preclinical or even clinical applications, a variable x-ray energy, a large field of view, and fast phase-contrast computed tomography (CT) devices are desirable. The noninterferometric grating-based phase-contrast imaging method is a good candidate, as it relaxes requirements on gratings, including grating period and aspect ratio. Based on the noninterferometric imaging principle, we constructed a prototype phase-contrast CT system, at the National Synchrotron Radiation Laboratory of the University of Science and Technology of China, with medical imaging components. This prototype system enables a large field of view and fast phase-contrast CT imaging under medical imaging energies. In this paper, the prototype system and preliminary experimental results are reported, and possible optimization for forthcoming work is also discussed.

Suppression of artifacts in X-ray phase-contrast images retrieved by Fourier transform

Over the last two decades, the grating-based phase-contrast imaging has aroused the interest of a number of researchers. It could provide an access to three complementary signals simultaneously: the conventional absorption contrast, the differential phase contrast related to refraction of incident wave, and the dark-field contrast that relates to ultra small angle scattering in a sample. The grating-based phase-contrast signals have higher contrast sensitivity for some types of soft samples than the absorption signals. Dark-field signals have better diagnostic effects in the detection of lung tumors, pneumothorax and the identification of microcalcifications in breast. There are two main phase retrieval methods in grating-based X-ray phase-contrast imaging, i.e. phase stepping method and Fourier transform method. The phase signals retrieved by phase stepping is high precise and has low noise. But the sample suffers high dose due to at least three exposures. The phase signals retrieved by Fourier transform is low-dose due to the fact that only one image with sample is needed, but it is easily affected by artifacts when the size of the filtering window is too large. However, when the size of the filtering window is too small, the high-frequency information of the phase-contrast image will be lost, and the image will become blurred. A trade-off between definitions of the image and artifacts should be made. Since the phase-contrast signal and the dark-field signal of the sample are modulated by carrier fringes, the frequency spectrum of the detected image consists of many different harmonics. The artifacts in the retrieved signals originate from the spectrum aliasing between primary peak around zero spatial frequency and first-order harmonic peaks. Therefore, the subtraction between two images with phase difference can remove the primary peak, and the artifacts in the phase-contrast signals and dark-field signals will be suppressed. In order to further suppress the artifacts, we increase the frequency of carrier fringes, which results in a larger distance between first-order harmonic peaks in frequency domain. We finally attain artifact-free phase-contrast images and dark-field images while maintaining high definition of the images. The method proposed here is not only applicable to incoherent imaging system, but also to Talbot-Lau interferometer, and it would be useful in fast and low-dose X-ray phase-contrast and dark-field imaging.

Wave optics simulation of grating-based X-ray phase-contrast imaging using 4D Mouse Whole Body (MOBY) phantom.

Demonstrate realistic simulation of grating-based x-ray phase-contrast imaging (GB-XPCI) using wave optics and the four-dimensional Mouse Whole Body (MOBY) phantom defined with non-uniform rational B-splines (NURBS). We use a full-wave approach, which uses wave optics for x-ray wave propagation from the source to the detector. This forward imaging model can be directly applied to NURBS-defined numerical phantoms such as MOBY. We assign the material properties (attenuation coefficient and electron density) of each model part using the data for adult human tissues. The Poisson noise is added to the simulated images based on the calculated photon flux at each pixel. We simulate the intensity images of the MOBY phantom for eight different grating positions. From the simulated images, we calculate the absorption, differential phase, and normalized visibility contrast images. We also predict how the image quality is affected by different exposure times. GB-XPCI can be simulated with the full-wave approach and a realistic numerical phantom defined with NURBS.

Single spectrum three-material decomposition with grating-based x-ray phase-contrast CT

Grating-based x-ray phase-contrast imaging provides three simultaneous image channels originating from a single image acquisition. While the phase signal provides direct access to the electron density in tomography, there is additional information on sub-resolutional structural information which is called dark-field signal in analogy to optical microscopy. The additional availability of the conventional attenuation image qualifies the method for implementation into existing diagnostic routines. The simultaneous access to the attenuation coefficient and the electron density allows for quantitative two-material discrimination as demonstrated lately for measurements at a quasi-monochromatic compact synchrotron source. Here, we investigate the transfer of the method to conventional polychromatic x-ray sources and the additional inclusion of the dark-field signal for three-material decomposition. We evaluate the future potential of grating-based x-ray phase-contrast CT for quantitative three-material discrimination for the specific case of early stroke diagnosis at conventional polychromatic x-ray sources. Compared to conventional CT, the method has the potential to discriminate coagulated blood directly from contrast agent extravasation within a single CT acquisition. Additionally, the dark-field information allows for the clear identification of hydroxyapatite clusters due to their micro-structure despite a similar attenuation as the applied contrast agent. This information on materials with sub-resolutional microstructures is considered to comprise advantages relevant for various pathologies.

Fourier transform phase retrieval for x-ray phase-contrast imaging based on cascaded Talbot–Lau interferometers

Long exposure time is one of the major limitations in x-ray phase-contrast imaging (XPCI). Therefore, we demonstrate a promising alternative method for grating-based XPCI coupled with cascaded Talbot–Lau interferometers (TLIs). A Fourier analysis of the moire fringe patterns generated by the interferometers was used to obtain the multicontrast images (such as absorption, differential phase-contrast, and normalized visibility-contrast images) with a single exposure. The cascaded configuration with TLI and inverse TLI using large-period absorption gratings was established to verify the effectiveness of the algorithm and three multicontrast images of a polytetrafluoroethylene (PTFE) tube were obtained experimentally. The groove structures in the PTFE can be clearly identified in the differential phase-contrast image. This method shows potential for application of cascaded TLI in medical imaging.

Image Quality Analysis of X-ray Grating Interferometer Using Integrating-bucket Method

Abstract Grating-based x-ray phase-contrast imaging has the potential to enhance image quality and provide inner structure details non-destructively. In this work, using grating-based x-ray phase-contrast imaging system and employing integrating-bucket method, the quantitative expressions of signal-to-noise ratios due to photon statistics and mechanical error are analyzed in detail. Photon statistical noise and mechanical error are the main sources affecting the image noise in x-ray grating interferometry. Integrating-bucket method is a new phase extraction method translated to x-ray grating interferometry; hence, its image quality analysis would be of great importance to get high-quality phase image. The authors’ conclusions provide an alternate method to get high-quality refraction signal using grating interferometer, and hence increases applicability of grating interferometry in preclinical and clinical usage.

A phase-sampling method for an X-ray Talbot-Lau scanner with continuous grating movement

A Talbot-Lau scanner enables fast grating-based X-ray phase-contrast and dark-field imaging of large samples. We present a fast and robust scanning method based on continuous phase-sampling during the usual scan process. For that purpose the source grating is moved back and forth during the whole image acquisition procedure. The scanning method needs no specific detuning of the interferometer. The acquired images are compared to the results of a standard phase-stepping procedure. We show that high quality images are obtained by this continuous phase-sampling scanning method. One main advantage of the method is its independence of the occurring moir'e pattern shape, thus enabling an optimal alignment of the interferometer. Furthermore, the method works with a priori unknown phase-step positions. To our knowledge, this is the first time a grating is moved continuously back and forth while also the sample is in linear motion during a continuous image acquisition in Talbot-Lau imaging.

The trigonometric orthogonality of phase-stepping curves in grating-based x-ray phase-contrast imaging: Integral property and its implications for noise optimization.

Grating-based x-ray phase-contrast imaging (GPCI) is a promising technique for clinical applications as it can provide two newly emerging imaging modalities (differential phase-contrast and dark-field contrast) in addition to the conventional absorption contrast. As far, phase-stepping strategy is the most commonly used approach in GPCI to indirectly acquire differential phase-contrast and dark-field contrast. It is known that the obtained phase-stepping curves (PSCs) have the cosine property and the convolution property, leading to two types of information retrieval approaches in literature: the Fourier component analysis and the multi-order moment analysis. The purpose of this paper is to derive a new property of PSCs and apply the property to noise optimization for information retrieval. Based on the cosine expression of the flat PSC without the sample and the well-established convolution relationship between the flat PSC and the sample PSC, we reveal an important integral property of PSCs: the inner product of PSCs and an arbitrary function contains only zero-order and first-order components in the Fourier series. Furthermore, we apply the property to the direct multi-order moment analysis and propose a set of generalized forms including an optimal one in the presence of noise. To validate the effectiveness of our analysis, we compared the simulated and real experiment results retrieved by the original direct multi-order moment analysis with the ones retrieved by our proposed noise-optimal form. A significant improvement of noise performance by our method is observed and the improvement ratio in differential phase-contrast is consistent with our theoretical calculation (39.2%). In this paper, we reveal a new integral property of the acquired PSCs with and without samples in GPCI, which can be applied to information retrieval approaches like the direct multi-order moment analysis. Then we optimize these approaches to improve the noise performance, offering great potentials of dose reduction in practical applications.

Grating-based x-ray phase contrast imaging has attracted increasing interest in recent decades as multimodal and laboratory source usable method. Specific efforts have been focused on establishing a new extraction method to perform practical applications. In this work, noise properties of multi-combination information of newly established information extraction method, so-called angular signal radiography method, are investigated to provide guidelines for targeted and specific applications. The results show that how multi-combination of images can be used in targeted practical applications to obtain a high-quality image in terms of signal-to-noise ratio. Our conclusions can also hold true for upcoming targeted practical applications such as biomedical imaging, non-destructive imaging, and materials science.

Simple and fast scattering information in X-ray grating-based phase-contrast imaging

X-ray grating-based phase-contrast imaging is a potential imaging method with pre-clinical and clinical applications because it has the possibility of a large field of view and is compatible with X-ray tube sources. However, in X-ray grating-based phase-contrast imaging extraction methods have great importance to retrieve and separate information enabling high acquisition speed, reduced exposure dose and improved mechanical stability. Phase-stepping is a widely used extraction method in X-ray grating-based phase-contrast imaging. However, its discontinuous mode of acquisition leads to worse mechanical stability, exposure dose is generally high, which limits clinical applications significantly. In this work, the integrating bucket method is translated from visible light to X-ray regime, to extract scattering information of the investigated object. Both theoretical derivations with numerical simulation and experiments were performed to confirm the veracity and feasibility of the proposed method. In addition, we present the advantages of the proposed method over the phase-stepping method.

A fast alignment method for grating-based X-ray phase-contrast imaging systems

The alignment of a grating-based X-ray phase-contrast interferometer is an iterative process that requires numerous steps of changing the distances and angles between the gratings. For each alignment step an image is acquired to evaluate the detected intensity signature in order to optimize the observed moir'e pattern. Thus, a large number of images has to be taken for the alignment procedure. This is not feasible within reasonable time at X-ray sources like X-ray backlighters where the time between two X-ray shots is on the scale of hours. Here, we report on the development of a stable and transportable setup ready-to-use for grating-based X-ray phase-contrast imaging. A comprehensive set of reference images taken at a continuous beam serves as a look-up table which enables the grating alignment within very few alignment steps. Since this method features a fast, reliable and predictable alignment, it is also beneficial for grating-based X-ray phase-contrast imaging systems at common X-ray sources.

Quantitative characterization of ex vivo breast tissue via x-ray phase-contrast tomography.

Grating-based X-ray phase-contrast imaging (GPCI) has received growing interests in recent years due to its high capability of visualizing soft tissue. Breast imaging is one of the most promising candidates for the first clinical application of this imaging modality. In this work, quantitative breast tissue characterization based on GPCI computed tomography (CT) is investigated with a laboratory X-ray tube through a comparison between attenuation-based CT images and phase-contrast CT images. The Hounsfield units (HU) scale was introduced to phase-contrast images due to its wide application in clinical medicine. In this work, instead of water, plastic cylinders composed of polyethylene terephthalate (PET) was treated as the calibration material. An alternative test-retest reliability (TRR) was presented to evaluate the repeatability of GPCI. Comparison between attenuation-based CT imaging and GPCI CT imaging was operated with the use of statistical analysis methods like histograms and receiver operating characteristic (ROC) curves. The determined mean TRR related to cylinders is slightly larger in phase-contrast imaging (0.93) than that in attenuation-based imaging (0.89). With respect to distinguishing breast tissues, the AUC (area under curve) values of ROC curves of phase-contrast images are higher than that of attenuation-based images. An ex vivo study of GPCI shows that it is a stable imaging modality for visualizing the breast tissue with good repeatability, and that it could be of potential for the diagnosis of breast cancer as well.

A comparative study of information retrieval in grating-based x-ray phase-contrast imaging

The cosine-model analysis (CMA) method and the small angle x-ray scattering (SAXS) method are two major types of information retrieval algorithms, commonly utilized in x-ray phase-contrast imaging with a grating interferometer. However, there are significant differences between the two methods in algorithm implementation, and the existing literature has not completely revealed their intrinsic relationship. In this paper, we theoretically derive and experimentally verify the intrinsic connections between CMA and SAXS, and it is seen that SAXS can be interpreted well by the cosine-model assumption of CMA. To validate our analysis of the scattering distribution when applying the cosine model to the convolution used in SAXS, we applied a deconvolution process into CMA before using the Fourier transform to get the three contrasts. Furthermore, the principal component analysis (PCA) is introduced in this work, and two PCA-based retrieval algorithms are presented in order to simplify the iteration process of deconvolution in SAXS or to obtain absorption and dark-field signals instead of the Fourier transform in CMA. Applying a quantitative structural similarity (SSIM) index and a profile analysis to the results of an ex vivo mammography, it is proved that retrieved images via CMA and SAXS are consistent with each other (SSIM values are 1.0000, 0.9845 and 0.9767 respectively), and that the extra deconvolution process applied into CMA shows a good performance and our analytical analysis of the scattering distribution is valid when applying the cosine model to the convolution used in SAXS. Besides, it is concluded that PCA shows almost the same performance with the Fourier transform (SSIM values are 1.0000 for both absorption and dark-field images), and the simplified SAXS-analogous method works well with higher efficiency in computation and better stability relative to the original SAXS, while maintaining the similar level of image quality (SSIM values are 1.0000, 0.9839 and 0.9781 respectively).
