Biochemical features in rats with different behavioral activity under conditions of emotional stress.

Adaptive capacities of the organism under conditions of emotional stress are determined by individual complex of anti-stress protective mechanisms. We studied the content of coenzyme Q10 and the spectrum of polyunsaturated fatty acids omega-3 and omega-6 in the serum, liver, and brain, and LPO intensity in the serum and liver of behaviorally active and passive rats under conditions of acute emotional stress. The differences in the formation of adaptive responses in animals with various behavioral activities were demonstrated.

Formation and regulation of adaptive response in nematode Caenorhabditis elegans.

All organisms respond to environmental stresses (e.g., heavy metal, heat, UV irradiation, hyperoxia, food limitation, etc.) with coordinated adjustments in order to deal with the consequences and/or injuries caused by the severe stress. The nematode Caenorhabditis elegans often exerts adaptive responses if preconditioned with low concentrations of agents or stressor. In C. elegans, three types of adaptive responses can be formed: hormesis, cross-adaptation, and dietary restriction. Several factors influence the formation of adaptive responses in nematodes, and some mechanisms can explain their response formation. In particular, antioxidation system, heat-shock proteins, metallothioneins, glutathione, signaling transduction, and metabolic signals may play important roles in regulating the formation of adaptive responses. In this paper, we summarize the published evidence demonstrating that several types of adaptive responses have converged in C. elegans and discussed some possible alternative theories explaining the adaptive response control.

The molecular and cellular consequences of the chernobyl accident

In this paper the results of the Chernobyl accident investigation 5-10 and 24 years after are summarized. The genomic instability, adaptive response formation, genome damage and oxidative status have been investigated. The studies were performed on cells in culture, mice, children and adults living in contaminated areas and liquidators. On cells in culture after exposition in the accident zone and culturing thereafter in laboratory conditions the cell proliferative activity decrease; the late cell death, the frequency of cells with micronuclei and giant cells increasing have been observed. In the progeny of exposed cells the enhancement of radiosensitivity has been noticed. So we can suppose that in cultured cells exposition in the zone of the accident the genomic instability is induced which results in many disturbances. At the organism level in mice exposed in the Chernobyl zone the radiosensitivity increase and the decrease of endotheliocytes density in brain tissue has been observed. On the stimulated by PHA blood lymphocytes of children the increase of the frequency of cells with micronuclei more than 2 time have been noticed. In all groups investigated, the decrease of individuals with significant adaptive response was observed. In children and adults inhabitants the increase of radiosensitivity after low dose of irradiation has been noticed. 24-year after the accident it was discovered that in liquidators lymphocytes the frequency of cells with micronuclei, with chromosome type aberrations, with DNA double strand breaks have been increased; the reactive oxygen species (ROS) were decreased in comparison with the control population. We can suppose that genomic instability induced in residents of contaminated regions and liquidators long after the accident results in the genetic apparatus damage, radiosensitivity enhancement, hypoxia that represent risk factors and increase the probability of tumour and non-tumour diseases. The development of these pathological processes may happen in much more remote periods.

Characteristics of the cerebral mechanisms controlling the level of wakefulness, maturity of cognitive functions, and adaptive responses in children with attention deficit hyperactivity disorder and healthy children of the same age

Data on the characteristics of the disturbance of ultraslow cortical-brainstem, limbic-reticular, and suprasegmental mechanisms controlling the wakefulness level (WL) in six- to seven-year-old children with attention deficit hyperactivity disorder (ADHD) and age-matched healthy children (first-year comprehensive primary school pupils) with different types of psychological adaptation have been summarized. An integrated systemic psychophysiological approach has been used to determine the characteristics of (a) the emotional-motivational and cognitive maturities and (b) the psychophysiological mechanisms of the formation of adaptive responses upon tactile interaction with an unfamiliar live object (a dolphin) in the examined groups of healthy and ADHD children.

Metallothioneins Are Required for Formation of Cross-Adaptation Response to Neurobehavioral Toxicity from Lead and Mercury Exposure in Nematodes

Metallothioneins (MTs) are small, cysteine-rich polypeptides, but the role of MTs in inducing the formation of adaptive response is still largely unknown. We investigated the roles of metallothionein genes (mtl-1 and mtl-2) in the formation of cross-adaptation response to neurobehavioral toxicity from metal exposure in Caenorhabditis elegans. Pre-treatment with mild heat-shock at L2-larva stage effectively prevented the formation of the neurobehavioral defects and the activation of severe stress response in metal exposed nematodes at concentrations of 50 and 100 µM, but pre-treatment with mild heat-shock did not prevent the formation of neurobehavioral defects in 200 µM of metal exposed nematodes. During the formation of cross-adaptation response, the induction of mtl-1 and mtl-2 promoter activity and subsequent GFP gene expression were sharply increased in 50 µM or 100 µM of metal exposed Pmtl-1::GFP and Pmtl-2::GFP transgenic adult animals after mild heat-shock treatment compared with those treated with mild heat-shock or metal exposure alone. Moreover, after pre-treatment with mild heat-shock, no noticeable increase of locomotion behaviors could be observed in metal exposed mtl-1 or mtl-2 mutant nematodes compared to those without mild heat-shock pre-treatment. The defects of adaptive response to neurobehavioral toxicity induced by metal exposure formed in mtl-1 and mtl-2 mutants could be completely rescued by the expression of mtl-1 and mtl-2 with the aid of their native promoters. Furthermore, over-expression of MTL-1 and MTL-2 at the L2-larval stage significantly suppressed the toxicity on locomotion behaviors from metal exposure at all examined concentrations. Therefore, the normal formation of cross-adaptation response to neurobehavioral toxicity induced by metal exposure may need the enough accumulation of MTs protein in animal tissues.

The Involvement of Different Mobile LINE Copies of Blood Plasma and Extrachromosomal DNA of Liver Cells in Systemic Adaptive Response

The aim of this work was to investigate the dynamics of the quantity of LINE in the DNA composition of blood plasma and extrachromosomal DNA of liver cells under conditions of either immobilization or exposure to ionizing radiation or a combination of both factors. It has been established that post-stress changes in the quantity of genetic elements may be interpreted as an activation of the mechanisms of retroposition and intercellular transfer of genetic elements that ensure the formation of adaptive responses at the systemic level. The formation of adaptive responses is expressed through the reduction of the content of the low-molecular-weight fraction of the extracellular DNA.

Effect of sociobiological factors on the formation of adaptive responses in school students during puberty

Analysis of psychological, social, and psychophysiological mechanisms underlying the formation of adaptive responses during puberty demonstrated a significant dependence of adolescent behavioral, autonomic, and endocrine parameters on sociobiological factors that determine the identification and individualization of genetically determined behavioral programs. Comparison of the adaptive capabilities of students from an innovative educational institution and from a vocational school showed that the levels of completeness of their adaptation were different and depended on social support, typological features, and autonomic regulation.

Functioning of malate dehydrogenase system in mesophyll and bundle sheath cells of maize leaves under salt stress conditions

The formation of adaptive response to salt stress in mesophyll and bundle sheath cells of maize (Zea mays L.) leaves was studied at the level of operation of enzyme systems that participate in oxidation of malate. Functioning of four malate dehydrogenases (MDH), the components of this system, was studied and found to maintain malate and pyruvate pools, which are required for operation of the Hatch-Slack cycle and actively used for neutralization of salt treatment. The increase in activity of NAD-MDH was related to salt-induced synthesis of the additional isoform of MDH in mesophyll cells. Such changes in the isozyme pattern were not found in bundle sheath cells.

Mechanisms of human cell protection associated with genetic polymorphism

Systems ensuring protection of human cells against endogenous and exogenous mutagenic factors are considered in terms of genetic polymorphism. Some protection mechanisms are described, including those connected with capturing free radicals, biotransformation of xenobiotics, excision repair of DNA damage (excision of nitrous bases, nucleotides, mismatch repair). A special section is devoted to some issues of using antimutagens in context of genetic polymorphism. The problem of adaptive response is discussed, providing evidence for independence (in some cases) of DNA repair systems and the formation of adaptive response. Some results of the author obtained many years ago but still relevant are presented.

Adaptive response in the reparation-defective gout cells

Adaptive response of fibroblasts from skin biopsy, specimens from patients with gout has been studied; these cells are characterized as reparation-defective during exposure to mutagens N-methyl-N'-nitro-N-nitrosoguanidine and 4-nitroquinolin-N-oxide. Adaptive response formed during exposure to nitrosoguanidine. The data indicate that the formation of adaptive response does not depend on DNA reparation.
